上記反応工程式1に示す方法においては、式(1a)で示される水酸基を有する化合物において、水酸基と式(1b)で示される脱離性官能基を有する化合物と反応させることによりエーテル誘導体(1c)へと誘導することができる。
上記反応工程式3に示す方法においては、式(3a)で示される水酸基を有する化合物において、水酸基を有する化合物又はその活性種と式(3b)で示されるアミン化合物又はその塩を常法により縮合させることにより、カーバメート誘導体(3c)へと誘導することができる。
また、式(3a)で示される水酸基を有する化合物又はその活性種1モルに対し、0.5~100モル、好ましくは0.8~50モルの式(3b)で示されるアミン化合物又はその塩を用い、上記縮合剤を使用する場合、その使用量は式(3a)で示される水酸基を有する化合物又はその塩1モルに対し、0.5~20モル、好ましくは0.8~3モルである。
上記反応工程式4に示す方法においては、工程1において、式(4a)で示される水酸基を有する化合物においては、水酸基をカルボン酸へと酸化することにより式(4b)で示されるカルボン酸誘導体とした後、工程2において、該カルボン酸誘導体(4b)を式(4c)で示されるアミン化合物と縮合させることにより、アミド誘導体(4d)へと誘導することができる。更に工程3において、該アミド誘導体(4d)を脱水させることにより、ニトリル誘導体(4e)へと誘導することができる。
即ち、式(4a)で示される化合物の水酸基をカルボン酸へと酸化する方法としては、アルデヒドを単離して用いる場合も、直接カルボン酸へ酸化する場合もあるが、適当な溶媒中、慣用されている適当な酸化剤を、式(4a)で示される水酸基を有する化合物1モルに対し、0.5~30モル、好ましくは0.8~20モル用いて、-80~150℃程度、好ましくは-80~120℃程度の温度下で、反応させればよい。
本工程は、式(4b)で表される化合物のカルボキシル基と、市販品または、公知の方法によって製造できるアミン化合物(4c)とのアミド化反応により、式(4d)で表されるアミド化合物を得る方法である。
本工程は、式(4d)で表されるアミド化合物を脱水反応により、式(4e)で表されるニトリル化合物を得る方法である。
反応は、使用する塩基や脱水剤によっても異なるが、通常ジクロロメタン、クロロホルム等のハロゲン化炭化水素類、トルエン等の芳香族炭化水素類、テトラヒドロフラン等のエーテル類、酢酸エチル等のエステル類、メタノール、エタノール等のアルコール類、水、アセトニトリル、N,N-ジメチルホルムアミド、N,N-ジメチルアセトアミド、ジメチルスルホキシド、ピリジン等の反応に不活性な溶媒中、-20℃~150℃、好ましくは0℃~100℃で行うことができる。
即ち、式(5a)で示される化合物のケトン基のα位に官能基を導入する方法としては、エノールエーテルを単離して用いる場合も、反応系中で調製して単離することなく反応に用いる場合もあるが、適当な溶媒中、慣用されている適当な塩基を、式(5a)で示されるケトン基を有する化合物1モルに対し、0.5~30モル、好ましくは0.8~10モル用いて、-100~120℃程度、好ましくは-80~100℃程度の温度下で、反応させればよい。
本工程は、式(5b)で表される化合物を脱離反応により、式(5c)で表されるジエノン化合物を得る方法である。適当な溶媒中、慣用されている適当な酸化剤を、式(5b)で示される脱離性官能基を有する化合物1モルに対し、0.5~30モル、好ましくは0.8~10モル用いて、-50~120℃程度、好ましくは-20~100℃程度の温度下で、反応させればよい。
上記反応工程式6に示す方法においては、式(6a)で示されるエノン基を有する化合物において、ハロゲン原子を導入することにより式(6b)で示される化合物へと誘導することができる。ハロゲン化試薬は、例えば、フッ素、塩素、臭素、ヨウ素などを用いる方法、N-クロロスクシンイミド、N-ブロモスクシンイミド、N-ヨードスクシンイミドを用いる方法により行うことができる。
反応に際し、式(6a)で示されるエノン基を有する化合物1モルに対し、0.1~10モル、好ましくは0.2~5モルの炭酸カリウム、4-(N,N-ジメチルアミノ)ピリジン、トリメチルシリルアジド等の試薬存在下反応を行うことにより、反応が円滑に進行する場合がある。
化合物(7b)のL2は、ハロゲン原子であればいずれでもよく、例えば塩素原子、臭素原子、ヨウ素原子等が挙げられる。
R13は水素原子又はC1~C4アルキル基であり、例えば水素原子、メチル基、エチル基、プロピル基、イソプロピル基、ブチル基等が挙げられる。

15-ブロモペンタデカン-1-オ-ル3.07gのクロロホルム30mL溶液にピリジニウム p-トルエンスルホネート251mgと3,4-ジヒドロ-2H-ピラン1.4mLのクロロホルム5mL溶液を加えて室温にて3時間撹拌した。氷冷下、反応溶液に炭酸水素ナトリウム水溶液を加えてクロロホルムにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して残渣(4.1g)を得た。得られた残渣1.17gと3-イソブトキシ-2-メチルシクロヘキサ-2-エン-1オン273mgのテトラヒドロフラン溶液8mLにリチウム金属95mgを加え、70℃にて4時間撹拌した。反応溶液を氷冷したのち、水を加えて室温にて0.5時間撹拌し、酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をメタノール2mLとテトラヒドロフラン4mLに溶解し、p-トルエンスルホン酸 1水和物29mgを加えて室温にて4時間撹拌した。反応溶液に飽和炭酸水素ナトリウム水溶液を加えて、酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物244mgを得た。

15-ブロモペンタデカン-1-オ-ル3.07gのクロロホルム30mL溶液にピリジニウム p-トルエンスルホネート251mgと3,4-ジヒドロ-2H-ピラン1.4mLのクロロホルム5mL溶液を加えて室温にて3時間撹拌した。氷冷下、反応溶液に炭酸水素ナトリウム水溶液を加えてクロロホルムにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して残渣(4.1g)を得た。得られた残渣1.17gと3-イソブトキシ-6,6-ジメチルシクロヘキサ-2-エン-1-オン294mgのテトラヒドロフラン溶液8mLにリチウム金属95mgを加え、70℃にて2時間撹拌した。反応溶液を氷冷したのち、水を加えて室温にて0.5時間撹拌し、酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をメタノール2mLとテトラヒドロフラン4mLに溶解し、p-トルエンスルホン酸 1水和物29mgを加えて室温にて4時間撹拌した。反応溶液に飽和炭酸水素ナトリウム水溶液を加えて、酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物366mgを得た。

塩化オキザリル1.1mLの塩化メチレン30mL溶液に-70℃でジメチルスルホキシド2.0mLを加えて15分間撹拌した後、3-(15-ヒドロキシペンタデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン(化合物4、大鵬薬品工業株式会社)3.79gの塩化メチレン10mL溶液を10分間かけて滴下した。ドライアイス-アセトンバスをはずして20分かけて内温-30℃まで昇温した後、再度-70℃まで冷却してトリエチルアミン7.0mLを加えた後、室温下2時間撹拌した。氷冷下、反応溶液に飽和食塩水を加えて酢酸エチルとn-ヘキサンの混合溶媒にて抽出し、1mol/L 塩酸水、飽和重曹水、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物3.04gを得た。

中間体1である15-(2,6,6-トリメチル-3-オキソシクロヘキサ-1-エン-1-イル)ペンタデカナール1.81gのtert-ブチルアルコール6mLと水2mLの混合溶液に氷冷下2-メチル-2-ブテン4mL、リン酸2水素カリウム1.02gを加えた。反応溶液に79%亜塩素酸ナトリウム1.71gの水5mL溶液を滴下し、室温にて1時間撹拌した。氷冷下、反応溶液に1mol/L塩酸水を加えて酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(メタノール/クロロホルム)で精製し、標題化合物1.69gを得た。

3-(15-ヒドロキシペンタデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン(化合物4、大鵬薬品工業株式会社)729mgの塩化メチレン10mL溶液に氷冷下トリエチルアミン0.43mLとクロロ蟻酸4-ニトロフェニル443mgを加えて氷冷下1時間、室温下5時間撹拌した。氷冷下、反応溶液に水を加えて酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物1.03gを得た。

15-ブロモペンタデカン-1-オ-ル3.07gのクロロホルム30mL溶液にピリジニウム p-トルエンスルホネート251mgと3,4-ジヒドロ-2H-ピラン1.35mLを加えて室温にて2時間撹拌した。氷冷下、反応溶液に炭酸水素ナトリウム水溶液を加えてクロロホルムにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をテトラヒドロフラン40mLに溶解し、ナトリウムメトキシド(5M メタノール溶液)10mLとヨウ化ナトリウム250mgを加えて70℃で2時間撹拌した。減圧留去して得られた残渣に氷冷下飽和塩化アンモニウム溶液を加えて酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣にメタノール10mLとテトラヒドロフラン5mLを加えて溶解し、p-トルエンスルホン酸一水和物95mgを加えて室温にて3時間撹拌した。氷冷下、反応溶液に炭酸水素ナトリウム水溶液を加えて、減圧留去して得られた残渣を酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物2.15gを得た。
中間体4である15-メトキシペンタデカン-1-オ-ル2.07gの塩化メチレン25mL溶液に氷冷下四臭化炭素2.92gとトリフェニルホスフィン2.52gを加えて1時間撹拌した。反応溶液に炭酸水素ナトリウム水溶液を加えて酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(クロロホルム/ヘキサン)で精製し、標題化合物2.28gを得た。

4,6,7,8-テトラヒドロ-5H-ベンゾ[d][1,3]ジオキシ-5-ノン1.25gのテトラヒドロフラン40mL溶液に-78℃にてリチウムヘキサメチルジシラザミド(1.3Mテトラヒドロフラン溶液)14.37mLを加え20分間撹拌した。反応溶液にヨウ化メチル1.45mLを加えて同温にて80分間、-45℃にて80分間撹拌した。反応溶液を-78℃に冷却してリチウムヘキサメチルジシラザミド(1.3Mテトラヒドロフラン溶液)3.80mLとヨウ化メチル0.28mLを加えて-45℃にて1時間撹拌した。反応溶液に飽和塩化アンモニウム水溶液を加えて酢酸エチルにて抽出し、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、標題化合物173.1mgを得た。
中間体6である6,6-ジメチル-4,6,7,8-テトラヒドロ-5H-ベンゾ[d][1,3]ジオキシ-5-ノン0.20gと中間体5で得られた1-ブロモ-15-メトキシペンタデカン0.42gのテトラヒドロフラン8.0mL溶液にリチウム金属0.11gを加えて70℃にて85分間撹拌した。反応溶液を氷冷したのち、0.5M塩酸水32mLを加えて室温にて30分間撹拌した。酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、標題化合物276mgを得た。

スピロ[2,5]オクタン-4,6-ジオン0.50gのシクロヘキサン6.5mL溶液に、イソブタノール0.54gとp-トルエンスルホン酸一水和物31mgを加えて70℃にて6時間撹拌した。反応溶液にモレキュラーシーブス4A1.0gを加え、16時間加熱撹拌した。室温まで放冷したのち、酢酸エチルで不溶物を濾別して飽和炭酸水素ナトリウム水溶液を加えた。有機層を水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、標題化合物0.51gを得た。
スピロ[2,5]オクタン-4,6-ジオン1.0gと水酸化カリウム2.0gをメタノール10mL、水20mLに溶解して70℃に加温し、ヨウ化メチル2mLを40分毎に4回加えて撹拌した。室温まで冷却して減圧留去して得られた残渣に飽和塩化アンモニウム水溶液を加えて塩化メチレンで抽出し、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、標題化合物0.3gを得た。
中間体9である5-メチルスピロ[2,5]オクタン-4,6-ジオン200mgをトルエンに溶解し、イソブタノール2.0当量とp-トルエンスルホン酸一水和物0.043当量を加えて70℃にて1日間撹拌した。反応溶液にモレキュラーシーブス4A400mgを加え、120℃で1日間撹拌した。室温まで放冷したのち、酢酸エチルで不溶物を濾別して飽和炭酸水素ナトリウム水溶液を加えた。有機層を水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、標題化合物73.8mgを得た。
3-イソブトキシ-2-メチルシクロペンタ-2-エン-1-オン1.00gのテトラヒドロフラン50mL溶液に-78℃にてリチウムヘキサメチルジシラザミド(1.3Mテトラヒドロフラン溶液)9.60mLを加え10分間撹拌した。反応溶液にヨウ化メチル0.78mLを加えて同温にて10分間、-45℃にて3時間撹拌した。反応溶液を-78℃に冷却してリチウムヘキサメチルジシラザミド(1.3Mテトラヒドロフラン溶液)1.20mLとヨウ化メチル0.10mLを加えて同温にて40分間、-45℃にて1時間撹拌した。氷冷下、反応溶液に飽和塩化アンモニウム水溶液を加えて酢酸エチルにて抽出し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、標題化合物419mgを得た。
中間体2である15-(2,6,6-トリメチル-3-オキソシクロヘキサ-1-エン-1-イル)ペンタデカノイックアシッド379mgのアセトニトリル5mL、N,N-ジメチルホルムアミド1mL溶液に1-ヒドロキシベンゾトリアゾール1水和物168mg、1-(3-ジメチルアミノプロピル)-3-エチルカルボジイミド塩酸塩230mg、メタノール0.4mLを加え、室温にて1時間撹拌した。反応溶液にトリエチルアミン0.21mL溶液を加え、室温にて16時間撹拌した。氷冷下、反応溶液に炭酸水素ナトリウム溶液を加えて、酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して、標題化合物を386mg得た。
中間体12であるメチル 15-(2,6,6-トリメチル-3-オキソシクロヘキサ-1-エン-1-イル)ペンタデカノエート118mgのテトラヒドロフラン2mL、メタノール1mL溶液に氷冷下水素化ホウ素ナトリウム34mgを加えて2時間撹拌した。反応溶液に飽和塩化アンモニウム水溶液を加えて、酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残留物を塩化メチレン3mLに溶解し、トリエチルアミン126μL、tert-ブチルジメチルクロロシラン90mg、4-ジメチルアミノピリジン7mgを加えて、室温にて2時間撹拌した。反応液に水を加えて酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物116mgを得た。
中間体13であるメチル 15-3-((tert-ブチルジメチルシリル)オキシ)-2,6,6-トリメチルシクロヘキサ-1-エン-1-イル)ペンタデカノエート108mgのテトラヒドロフラン2mL溶液に氷冷下0.96mol/L メチルマグネシウムブロミドのテトラヒドロフラン溶液2.2mLを加えて1時間撹拌した。反応溶液に飽和塩化アンモニウム水溶液を加えて、酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物89mgを得た。
中間体14である16-3-((tert-ブチルジメチルシリル)オキシ)-2,6,6-トリメチルシクロヘキサ-1-エン-1-イル)-2-メチルメチルヘキサデカ-2-ノール509mgのテトラヒドロフラン10mL溶液に氷冷下55%水素化ナトリウム130mgを加え室温にて1時間撹拌した。反応溶液にヨウ化エメチル125μLを加えて室温下2日間撹拌した。氷冷下、反応溶液に飽和塩化アンモニウム溶液を加えて酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(クロロホルム/ヘキサン)で精製し、標題化合物435mgを得た。
3-(15-ヒドロキシペンタデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン(化合物4、大鵬薬品工業株式会社)3.64gの塩化メチレン40mL溶液に氷冷下四臭化炭素3.65gとトリフェニルホスフィン3.15gを加えて3時間撹拌した。反応溶液に炭酸水素ナトリウム水溶液を加えて酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣にジエチルエ-テル:ヘキサン(1:1)の混合溶液を加えて不溶物を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/クロロホルム)で精製し、標題化合物4.18gを得た。

リチウムジイソプロピルアミド(1Mヘキサン・テトラヒドロフラン溶液)120mLをテトラヒドロフラン400mLに加え、ドライアイス-アセトンバスにて3-イソブトキシ-2-メチルシクロペンタ-2-エン-1-オン20.0gのテトラヒドロフラン100mL溶液を加え、さらにヨウ化メチル0.78mLを加え10分間撹拌した。反応溶液にヨウ化メチル7.36mLを加えて20℃まで昇温して10分間撹拌した。反応溶液をドライアイス-アセトンバスで冷却してリチウムジイソプロピルアミド(1Mヘキサン・テトラヒドロフラン溶液)120mLとヨウ化メチル7.36mLを加えて15℃まで昇温して15分間撹拌した。氷冷下、反応溶液に水を加えてジイソプロピルエーテルにて抽出し、10%食塩水、飽和食塩水で洗浄して無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、標題化合物14.6gを得た。

12-ブロモドデカン-1-オ-ル3.81gのクロロホルム39.6mL 溶液にピリジニウム p-トルエンスルホネート361mgと3,4-ジヒドロ-2H-ピラン1.97mLを加えて室温にて1時間撹拌したのち、p-トルエンスルホネート180mgと3,4-ジヒドロ-2H-ピラン985μLを加えて室温にて30分間撹拌した。氷冷下、反応溶液に飽和炭酸水素ナトリウム水溶液を加えてクロロホルムにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して残渣を得た。
リチウム金属249mgにテトラヒドロフラン10.8mLを加え、40℃に加温した。上記で得られた残渣と中間体17である3-イソブトキシ-2,5,5-トリメチルシクロペンタ-2-エン-1-オン1.41gのテトラヒドロフラン溶液10.8mLを滴下し、50℃にて2時間撹拌した。反応溶液を氷冷したのち、水7mLを加えて室温にて30分間撹拌した。セライトで不溶物を濾別したのち、酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をメタノール14.4mLとテトラヒドロフラン10.8mLに溶解し、p-トルエンスルホン酸 1水和物137mgを加えて室温にて49分間撹拌した。氷冷下、飽和炭酸水素ナトリウム水溶液を加えて、減圧下溶媒留去したのち、酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、標題化合物1.88gを得た。

14-ブロモテトラデカン-1-オ-ル1.99gのクロロホルム20.8mL 溶液にピリジニウム p-トルエンスルホネート171mgと3,4-ジヒドロ-2H-ピラン931μLを加えて室温にて45分間撹拌したのち、p-トルエンスルホネート85mgと3,4-ジヒドロ-2H-ピラン466μLを加えて室温にて30分間撹拌した。氷冷下、反応溶液に飽和炭酸水素ナトリウム水溶液を加えてクロロホルムにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して残渣を得た。
リチウム金属131mgにテトラヒドロフラン5.66mLを加え、40℃に加温した。上記で得られた残渣と中間体17である3-イソブトキシ-2,5,5-トリメチルシクロペンタ-2-エン-1-オン740mgのテトラヒドロフラン溶液5.66mLを滴下し、50℃にて2時間撹拌した。反応溶液を氷冷したのち、水3.7mLを加えて室温にて撹拌した。セライトで不溶物を濾別したのち、酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をメタノール7.55mLとテトラヒドロフラン3.77mLに溶解し、p-トルエンスルホン酸 1水和物72mgを加えて室温にて1時間撹拌した。氷冷下、飽和炭酸水素ナトリウム水溶液を加えて、減圧下溶媒留去したのち、酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、標題化合物832mgを得た。

リチウム金属249mgにテトラヒドロフラン10.8mLを加え、40℃に加温した。上記で得られた残渣と中間体17である3-イソブトキシ-2,5,5-トリメチルシクロペンタ-2-エン-1-オン1.41gのテトラヒドロフラン溶液10.8mLを滴下し、50℃にて2時間撹拌した。反応溶液を氷冷したのち、水7.19mLを加えて室温にて撹拌した。セライトで不溶物を濾別したのち、酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をメタノール14.4mLとテトラヒドロフラン7.19mLに溶解し、p-トルエンスルホン酸 1水和物137mgを加えて室温にて1時間撹拌した。氷冷下、飽和炭酸水素ナトリウム水溶液を加えて、減圧下溶媒留去したのち、酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、標題化合物2.08gを得た。

中間体20である3-(15-ヒドロキシペンタデシル)-2,4,4-トリメチルシクロペンタ-2-エン-1-オン1.00gの塩化メチレン7.98mL溶液にジメチルスルホキシド3.18mLを加えて氷冷下、トリエチルアミン3.18mL、ピリジン三酸化硫黄錯体1.82gを加えた後、室温下30分間撹拌した。氷冷下、反応溶液に水を加えて酢酸エチルとn-ヘプタンの混合溶媒にて抽出し、0.1mol/L 塩酸水、飽和塩化アンモニウム水溶液、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、標題化合物530mgを得た。
中間体21である15-(2,5,5-トリメチル-3-オキソシクロペンタ-1-エン-1-イル)ペンタデカナール530mgのtert-ブチルアルコール1.83mLと水0.609mLの混合溶液に氷冷下2-メチル-2-ブテン1.22mL、リン酸2水素カリウム310mgを加えた。反応溶液に80%亜塩素酸ナトリウム515mgの水1.52mL溶液を滴下し、室温にて1時間撹拌した。氷冷下、反応溶液に1mol/L塩酸水を加えて酢酸エチルにて抽出し、飽和チオ硫酸ナトリウム水溶液と飽和食塩水の混液、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(メタノール/クロロホルム)で精製し、標題化合物505mgを得た。

3-(12-ヒドロキシドデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン(化合物1)161mgのテトラヒドロフラン2.0mL溶液に氷冷下tert-ブトキシナトリウム72mgを加え30分間撹拌した。反応溶液にヨウ化メチル62μLを加えて氷冷下0.5時間、室温下2時間撹拌した。氷冷下、反応溶液に飽和塩化アンモニウム溶液を加えて酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物114mgを得た。
3-(12-ヒドロキシドデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン(化合物1)161mgのテトラヒドロフラン2.0mL溶液に氷冷下55%水素化ナトリウム43mgを加え30分間撹拌した。反応溶液にヨウ化エチル80μLを加えて氷冷下1時間、室温下24時間撹拌した。氷冷下、反応溶液に飽和塩化アンモニウム溶液を加えて酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物57mgを得た。
3-(12-ヒドロキシドデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン(化合物1)161mgの2-ヨ-ドプロパン0.57mL溶液に酸化銀232mgを加えて室温下7日間撹拌した。反応溶液をtert-ブチルメチルエーテルで希釈し、セライトで不溶物を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物106mgを得た。

中間体2である15-(2,6,6-トリメチル-3-オキソシクロヘキサ-1-エン-1-イル)ペンタデカノイックアシッド717mgのテトラヒドロフラン15mL溶液に氷冷下トリエチルアミン0.98mLとクロロ炭酸エチル0.59mLを加え30分間撹拌した。反応溶液に重水素化ホウ素ナトリウム544mgの重水3.0mL溶液を加えて同温にて0.5時間撹拌した。氷冷下、反応溶液に飽和塩化アンモニウム溶液を加えて酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、3-(15-ヒドロキシペンタデシル-15,15-d2)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン548mgを得た。得られた3-(15-ヒドロキシペンタデシル-15,15-d2)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン183mgのテトラヒドロフラン3.0mL溶液に氷冷下55%水素化ナトリウム43mgを加え30分間撹拌した。反応溶液にヨウ化メチル93μLを加えて氷冷下1時間、室温下3日間撹拌した。氷冷下、反応溶液に飽和塩化アンモニウム溶液を加えて酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物100mgを得た。

中間体2である15-(2,6,6-トリメチル-3-オキソシクロヘキサ-1-エン-1-イル)ペンタデカノイックアシッド379mgのアセトニトリル5mL溶液に1-ヒドロキシベンゾトリアゾール1水和物168mg、1-(3-ジメチルアミノプロピル)-3-エチルカルボジイミド塩酸塩230mgを加え、室温にて15分間撹拌した。反応溶液に28%アンモニア水0.5mLを加え、室温にて18時間撹拌した。氷冷下、反応溶液に炭酸水素ナトリウム溶液を加えて、析出した固体を濾取して減圧乾燥することで、標題化合物299mgを得た。
中間体2である15-(2,6,6-トリメチル-3-オキソシクロヘキサ-1-エン-1-イル)ペンタデカノイックアシッド379mgのアセトニトリル5mL溶液に1-ヒドロキシベンゾトリアゾール1水和物168mg、1-(3-ジメチルアミノプロピル)-3-エチルカルボジイミド塩酸塩230mgを加え、室温にて15分間撹拌した。反応溶液に2mol/L メチルアミン-テトラヒドロフラン溶液1mLを加え、室温にて14時間撹拌した。氷冷下、反応溶液に炭酸水素ナトリウム溶液を加えて、析出した固体を濾取して減圧乾燥することで、標題化合物340mgを得た。
実施例22で得られた15-(2,6,6-トリメチル-3-オキソシクロヘキサ-1-エン-1-イル)ペンタデカナミド113mgの塩化メチレン2mL溶液にBurgess試薬108mgを加え2時間撹拌した。反応溶液を減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物104mgを得た。
中間体3である4-ニトロフェニル(15-(2,6,6-トリメチル-3-オキソシクロヘキサ-1-エン-1-イル)ペンタデシル)カーボネート265mgのテトラヒドロフラン5mL溶液に28%アンモニア水0.5mLを加え、室温にて1時間撹拌した。反応溶液に飽和炭酸水素ナトリウム水溶液を加えて、酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物169mgを得た。
中間体3である4-ニトロフェニル(15-(2,6,6-トリメチル-3-オキソシクロヘキサ-1-エン-1-イル)ペンタデシル)カーボネート159mgのテトラヒドロフラン3mL溶液に氷冷下2mol/L ジメチルアミン-テトラヒドロフラン溶液0.2mLを加え、室温にて3時間撹拌した。氷冷下、反応溶液に飽和塩化アンモニウム水溶液を加えて、酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物113mgを得た。
3-エトキシシクロヘキサ-2-エン-1オン1.40gと中間体5である1-ブロモ-15-メトキシペンタデカン3.86gのテトラヒドロフラン50mL溶液にリチウム金属694mgを加えて70℃にて22時間撹拌した。反応溶液を氷冷したのち、0.5M塩酸水を加えて室温にて30分間撹拌した。酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物2.10gを得た。
3-(15-ヒドロキシペンタデシル)-2-メチルシクロヘキサ-2-エン-1-オン(化合物7)101mgのテトラヒドロフラン2.0mL溶液にtert-ブトキシナトリウム43mg、ヨウ化メチル34μLを加えて、室温下44時間撹拌した。氷冷下、反応溶液に飽和塩化アンモニウム溶液を加えて酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物38mgを得た。
3-メトキシ-6,6-ジメチルシクロヘキサ-2-エン-1オン154mgと中間体5である1-ブロモ-15-メトキシペンタデカン642mgのテトラヒドロフラン6mL溶液にリチウム金属80mgを加えて70℃にて8時間撹拌した。反応溶液を氷冷したのち、0.5M塩酸水を加えて室温にて30分間撹拌した。酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物224mgを得た。
実施例29で得られた3-(15-メトキシペンタデシル)-4,4-ジメチルシクロヘキサ-2-エン-1-オン729mgのクロロホルム5mL溶液に氷冷下トリメチルシリルアジド0.63mLを加えて、3時間撹拌した。反応溶液に氷冷下ヨウ素2.03gとピリジン2mLのクロロホルム2mL溶液を加えて室温にて3日間撹拌した。10%亜硫酸ナトリウム水溶液を加えて酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物693mgを得た。
実施例14で得られた3-(15-メトキシペンタデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン200mgのテトラヒドロフラン4.0mL溶液に氷冷下リチウムヘキサメチルジシラザミド(1.3Mテトラヒドロフラン溶液)609μLを加え30分間撹拌した。反応溶液にクロロトリメチルシラン100μLを加えて室温下1時間撹拌した。反応溶液に炭酸水素ナトリウム水溶液を加えて酢酸エチルにて抽出し、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をテトラヒドロフラン5.0mLに溶解し、-78℃にてリチウムヘキサメチルジシラザミド(1.3Mテトラヒドロフラン溶液)609μLを加え30分間撹拌した。反応溶液にフェニルセレニルクロリド102mgのテトラヒドロフラン5.0mL溶液を滴下して同温にて30分間撹拌した後、室温まで昇温した。反応溶液に水を加えて酢酸エチルにて抽出し、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製して得られた3-(15-メトキシペンタデシル)-2,4,4-トリメチル-6-(フェニルセレニル)シクロヘキサ-2-エン-1-オン155mgの塩化メチレン16mL溶液にm-クロロ過安息香酸71.5mgを加えて室温下2時間撹拌した。反応溶液にチオ硫酸ナトリウム水溶液を加えて塩化メチレンにて抽出し、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物65.2mgを得た。
中間体7である2-(ヒドロキシメチル)-3-(15-メトキシペンタデシル)-4,4-ジメチルシクロヘキサ-2-エン-1-オン60mgの塩化メチレン1.0mL溶液に氷冷下1,8-ビス(ジメチルアミノ)ナフタレン212mgとトリメチルオキソニウム テトラフルオロボレート146mgを加えて室温にて1時間撹拌した。反応溶液に1M塩酸水を加えて中和し、酢酸エチルにて抽出し、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、標題化合物50mgを得た。
中間体8である6-イソブトキシスピロ[2,5]オクタ-5-エン-4-オン0.2gと中間体5である1-ブロモ-15-メトキシペンタデカン0.5gのテトラヒドロフラン8mL溶液にリチウム金属0.1gを加えて70℃にて3時間撹拌した。反応溶液を氷冷したのち、0.5M塩酸水を加えて室温にて30分間撹拌した。酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、標題化合物0.36gを得た。
中間体10である6-イソブトキシ-5-メチルスピロ[2,5]オクタ-5-エン-4-オン73.8mgと中間体5である1-ブロモ-15-メトキシペンタデカン1.2当量のテトラヒドロフラン溶液にリチウム金属14当量を加えて70℃にて4時間撹拌した。反応溶液を氷冷したのち、0.5M塩酸水を加えて室温にて30分間撹拌した。酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、標題化合物77.9mgを得た。
3-イソブトキシ-2-メチルシクロペンタ-2-エン-1-オン63.6mgと中間体5である1-ブロモ-15-メトキシペンタデカン232.1mgのテトラヒドロフラン3.0mL溶液にリチウム金属38.3mgを加えて70℃にて4時間撹拌した。反応溶液を氷冷したのち、0.5M塩酸水を加えて室温にて30分間撹拌した。酢酸エチルにて抽出し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、標題化合物89.0mgを得た。
中間体11である3-イソブトキシ-2,4,5-トリメチルシクロペンタ-2-エン-1-オン75.6mgと中間体5である1-ブロモ-15-メトキシペンタデカン230mgのテトラヒドロフラン3.0mL溶液にリチウム金属35.5mgを加えて70℃にて3.5時間撹拌した。反応溶液を氷冷したのち、0.5M塩酸水を加えて室温にて20分間撹拌した。酢酸エチルにて抽出し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、標題化合物112.5mgを得た。
中間体17である3-イソブトキシ-2,5,5-トリメチルシクロペンタ-2-エン-1-オン981mgと中間体5である1-ブロモ-15-メトキシペンタデカン2.89gのテトラヒドロフラン15mL溶液にリチウム金属156mgを加えて70℃にて5時間撹拌した。反応溶液を氷冷したのち、1M塩酸水を加えて室温にて30分間撹拌した。酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物1.68gを得た。
3-(12-ヒドロキシドデシル)-2,4,4-トリメチルシクロペンタ-2-エン-1-オン(中間体18)300mgのテトラヒドロフラン3.9mL溶液に氷冷下tert-ブトキシナトリウム140mgを加え30分間撹拌した。反応溶液にヨウ化メチル122μLを加えて氷冷下0.5時間、室温下1時間撹拌した。氷冷下、反応溶液に飽和塩化アンモニウム溶液を加えて酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、標題化合物199mgを得た。
3-(12-ヒドロキシドデシル)-2,4,4-トリメチルシクロペンタ-2-エン-1-オン(中間体18)500mgのテトラヒドロフラン8.09mL溶液に氷冷下55%水素化ナトリウム141mgを加え30分間撹拌した。反応溶液にヨウ化プロピル323μLを加えて氷冷下1時間、室温下44時間撹拌した。氷冷下、反応溶液に飽和塩化アンモニウム溶液を加えて酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、標題化合物119mgを得た。
3-(14-ヒドロキシテトラデシル)-2,4,4-トリメチルシクロペンタ-2-エン-1-オン(中間体19)250mgのテトラヒドロフラン2.98mL溶液に氷冷下tert-ブトキシナトリウム107mgを加え30分間撹拌した。反応溶液にヨウ化メチル122μLを加えて氷冷下30分間、室温下3時間撹拌した。氷冷下、反応溶液に飽和塩化アンモニウム溶液を加えて酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、標題化合物174mgを得た。
3-(14-ヒドロキシテトラデシル)-2,4,4-トリメチルシクロペンタ-2-エン-1-オン(中間体19)500mgのテトラヒドロフラン5.95mL溶液に氷冷下55%水素化ナトリウム130mgを加え30分間撹拌した。反応溶液にヨウ化エチル246μLを加えて氷冷下1時間、室温下21時間撹拌した。氷冷下、反応溶液に飽和塩化アンモニウム溶液を加えて酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、標題化合物274mgを得た。
中間体22である15-(2,5,5-トリメチル-3-オキソシクロペンタ-1-エン-1-イル)ペンタデカノイックアシッド250mgのアセトニトリル3.44mL溶液に1-ヒドロキシベンゾトリアゾール1水和物115mg、1-(3-ジメチルアミノプロピル)-3-エチルカルボジイミド塩酸塩158mgを加え、室温にて16分間撹拌した。反応溶液に28%アンモニア水343μLを加え、室温にて17時間撹拌し、28%アンモニア水170μLを追加し、室温にて4時間撹拌し、28%アンモニア水343μLを追加し、室温にて20時間撹拌した。氷冷下、反応溶液に飽和炭酸水素ナトリウム溶液を加えて、減圧下溶媒留去したのち、酢酸エチル、水を添加して分液した。有機層にヘプタンを添加して10%食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(メタノール/クロロホルム)で精製し、標題化合物146mgを得た。
実施例42で得られた15-(2,5,5-トリメチル-3-オキソシクロペンタ-1-エン-1-イル)ペンタデカナミド73mgの塩化メチレン2mL溶液にBurgess試薬71mgを加え2時間撹拌した。反応溶液を減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物51mgを得た。
中間体14であるtert-ブチル((3-(15-メトキシ-15-メチルヘキサデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-イル)オキシ)ジメチルシラン366mgのテトラヒドロフラン5mL溶液に1mol/L フッ化テトラブチルアンモニウムのテトラヒドロフラン溶液1.4mLを加えて室温にて2日間撹拌した。反応溶液を減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物272mgを得た。
参考例1で得られた3-(15-メトキシ-15-メチルヘキサデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オ-ル204mgのクロロホルム10mL溶液に活性二酸化マンガン435mgを加えて4日間撹拌した。不溶物をセライトで濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物199mgを得た。
3-(15-ヒドロキシペンタデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン(化合物4、大鵬薬品工業株式会社)365mg、トリフルオロメタンスルホン酸銀514mg、N-フルオロ-N'-(クロロメチル)トリエチレンジアミンビス(テトラフルオロボラート)531mg、フッ化カリウム174mgに酢酸エチル5mLを加え、室温にて2-フルオロピリジン172μL、(トリフルオロメチル)トリメチルシラン295μLを加えて25時間撹拌した。反応溶液を酢酸エチルで希釈し、セライトで不溶物を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物212mgを得た。
3-(15-ヒドロキシペンタデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン(化合物4、大鵬薬品工業株式会社)182mgのN,N-ジメチルホルムアミド1mL溶液にイミダゾール102mg、tert-ブチルジメチルクロロシラン151mg を加え、室温にて2時間撹拌した。反応溶液に水を加えて酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物226mgを得た。
3-(15-ヒドロキシペンタデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン(化合物4、大鵬薬品工業株式会社)182mgとフェノール52mgのテトラヒドロフラン4mL溶液に氷冷下トリフェニルホスフィン144mgとジイソプロピルアゾジカルボキシレート0.12mLを加え1時間、室温にて22時間撹拌した。反応溶液を酢酸エチルにて希釈し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物125mgを得た。
3-(15-ヒドロキシペンタデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン(化合物4、大鵬薬品工業株式会社)182mgのテトラヒドロフラン3mL溶液に氷冷下55%水素化ナトリウム55mgを加え30分間撹拌した。反応溶液にベンジルブロミド178μLを加えて氷冷下1時間、室温下3日間撹拌した。氷冷下、反応溶液に飽和塩化アンモニウム溶液を加えて酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物107mgを得た。
3-(15-ヒドロキシペンタデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン(化合物4、大鵬薬品工業株式会社)1.82gの塩化メチレン20mL溶液に氷冷下トリエチルアミン1.05mL、ベンゾイルクロリド0.64mLを加え、室温にて2日間撹拌した。氷冷下、反応溶液に水を加えて酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物2.34gを得た。
中間体2である15-(2,6,6-トリメチル-3-オキソシクロヘキサ-1-エン-1-イル)ペンタデカノイックアシッド379mgのアセトニトリル5mL溶液に1-ヒドロキシベンゾトリアゾール1水和物168mg、1-(3-ジメチルアミノプロピル)-3-エチルカルボジイミド塩酸塩230mgを加え、室温にて15分間撹拌した。反応溶液に2mol/L ジメチルアミン-テトラヒドロフラン溶液1mLを加え、室温にて14時間撹拌した。氷冷下、反応溶液に炭酸水素ナトリウム溶液を加えて、酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/クロロホルム)で精製し、標題化合物338mgを得た。
中間体3である4-ニトロフェニル(15-(2,6,6-トリメチル-3-オキソシクロヘキサ-1-エン-1-イル)ペンタデシル)カーボネート265mgのテトラヒドロフラン5mL溶液に氷冷下2mol/L メチルアミン-テトラヒドロフラン溶液0.3mLを加え、室温にて1時間撹拌した。氷冷下、反応溶液に飽和塩化アンモニウム水溶液を加えて、酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物177mgを得た。
中間体3である4-ニトロフェニル(15-(2,6,6-トリメチル-3-オキソシクロヘキサ-1-エン-1-イル)ペンタデシル)カーボネート159mgのテトラヒドロフラン3mL溶液に氷冷下2mol/L エチルアミン-テトラヒドロフラン溶液0.2mLを加え、室温にて3時間撹拌した。氷冷下、反応溶液に飽和塩化アンモニウム水溶液を加えて、酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物101mgを得た。
中間体3である4-ニトロフェニル(15-(2,6,6-トリメチル-3-オキソシクロヘキサ-1-エン-1-イル)ペンタデシル)カーボネート159mgのテトラヒドロフラン3mL溶液にエタノール175μL と1,8-ジアザビシクロ[5.4.0]ウンデカ-7エン67μLを加え、室温にて22時間撹拌した。氷冷下、反応溶液に飽和塩化アンモニウム水溶液を加えて、酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物36mgを得た。
中間体16である3-(15-ブロモペンタデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン214mgのテトラヒドロフラン5mL溶液に2mol/L メチルアミン-テトラヒドロフラン溶液1mLを加え、室温にて4日間撹拌した。反応溶液に水を加えて、酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をNHシルカゲルカラムクロマトグラフィー(酢酸エチル/クロロホルム)で精製し、標題化合物98mgを得た。
中間体16である3-(15-ブロモペンタデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン214mgのテトラヒドロフラン5mL溶液に2mol/L ジメチルアミン-テトラヒドロフラン溶液1mLを加え、室温にて3日間撹拌した。反応溶液に水を加えて、酢酸エチルにて抽出し、水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(メタノール/クロロホルム)で精製し、標題化合物162mgを得た。
4,4-ジメチルシクロヘキサン-1,3-ジオン1.0gのエタノール20mL溶液に水酸化カリウム1当量、水8mL、ヨウ化エチル1.5当量を加えて70℃で1日間撹拌した。反応溶液を減圧乾燥して酢酸エチルで濾過したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(アセトン/ヘプタン)で精製し、2-エチル-4,4-ジメチルシクロヘキサン-1,3-ジオン221mgを得た。得られた2-エチル-4,4-ジメチルシクロヘキサン-1,3-ジオン221mgをトルエンに溶解し、イソブタノール2.0当量とp-トルエンスルホン酸一水和物0.043当量を加えて70℃にて1日間撹拌した。反応溶液にモレキュラーシーブス4A2.0当量を加え、120℃で1日間撹拌した。室温まで放冷したのち、酢酸エチルで不溶物を濾別して0.5mol/L水酸化ナトリウム水溶液を加えた。有機層を水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、2-エチル-3-イソブトキシ-6,6-ジメチルシクロヘキサ-2-エン-1-オン143.8mgを得た。得られた2-エチル-3-イソブトキシ-6,6-ジメチルシクロヘキサ-2-エン-1-オン143.8mgと参考例5で得られた1-ブロモ-15-メトキシペンタデカン1.2当量のテトラヒドロフラン溶液にリチウム金属14当量を加えて70℃にて4時間撹拌した。反応溶液を氷冷したのち、0.5mol/L塩酸水を加えて室温にて30分間撹拌した。酢酸エチルにて抽出し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、標題化合物84.6mgを得た。
4,4-ジメチルシクロヘキサン-1,3-ジオン3.0gのエタノール24mL溶液に水酸化カリウム1当量、水24mL、ヨウ化プロピル1.5当量を加えて70℃で1日間撹拌した。反応溶液を減圧乾燥して酢酸エチルで濾過したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(アセトン/ヘプタン)で精製し、4,4-ジメチル-2-プロピルシクロヘキサン-1,3-ジオン868.5mgを得た。得られた4,4-ジメチル-2-プロピルシクロヘキサン-1,3-ジオン868.5mgをトルエンに溶解し、イソブタノール2.0当量とp-トルエンスルホン酸一水和物0.043当量を加えて70℃にて1日間撹拌した。反応溶液にモレキュラーシーブス4A2.0当量を加え、120℃で1日間撹拌した。室温まで放冷したのち、酢酸エチルで不溶物を濾別して0.5mol/L水酸化ナトリウム水溶液を加えた。有機層を水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、3-イソブトキシ-6,6-ジメチル-2-プロピルシクロヘキサ-2-エン-1-オン723mgを得た。得られた3-イソブトキシ-6,6-ジメチル-2-プロピルシクロヘキサ-2-エン-1-オン723mgと参考例5で得られた1-ブロモ-15-メトキシペンタデカン1.2当量のテトラヒドロフラン溶液にリチウム金属14当量を加えて70℃にて4時間撹拌した。反応溶液を氷冷したのち、0.5mol/L 塩酸水を加えて室温にて30分間撹拌した。酢酸エチルにて抽出し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、標題化合物78.1mgを得た。
4,4-ジメチルシクロヘキサン-1,3-ジオン3.0gのエタノール44mL溶液に水酸化カリウム1当量、水24mL、ヨウ化イソプロピル1.5当量を加えて70℃で1日間撹拌した。反応溶液を減圧乾燥して酢酸エチルで濾過したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(アセトン/ヘプタン)で精製し、2-イソプロピル-4,4-ジメチルシクロヘキサン-1,3-ジオン168.5mgを得た。得られた2-イソプロピル-4,4-ジメチルシクロヘキサン-1,3-ジオン168.5mgをトルエンに溶解し、イソブタノール2.0当量とp-トルエンスルホン酸一水和物0.043当量を加えて70℃にて1日間撹拌した。反応溶液にモレキュラーシーブス4A2.0当量を加え、120℃で1日間撹拌した。室温まで放冷したのち、酢酸エチルで不溶物を濾別して0.5mol/L水酸化ナトリウム水溶液を加えた。有機層を水、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、3-イソブトキシ-2-イソプロピル-6,6-ジメチルシクロヘキサ-2-エン-1-オン70.2mgを得た。得られた3-イソブトキシ-2-イソプロピル-6,6-ジメチルシクロヘキサ-2-エン-1-オン70.2mgと参考例5で得られた1-ブロモ-15-メトキシペンタデカン1.2当量のテトラヒドロフラン溶液にリチウム金属14当量を加えて70℃にて4時間撹拌した。反応溶液を氷冷したのち、0.5mol/L塩酸水を加えて室温にて30分間撹拌した。酢酸エチルにて抽出し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘプタン)で精製し、標題化合物28.5mgを得た。
実施例14で得られた3-(15-メトキシペンタデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン100mgのエタノール5mL溶液にパラジウム炭素10mgを加え、水素雰囲気下4時間撹拌した。反応溶液をセライトで濾過したのち、減圧留去することで、標題化合物68.9mgを得た。
アセトニトリル1mL、トリフルオロ酢酸1mL、酢酸1mLの混合溶液に氷冷下にて水素化ホウ素ナトリウム240mgを加えた。次いで実施例14で得られた3-(15-メトキシペンタデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン200mgの塩化メチレン0.4mL溶液を加えて氷冷下2時間撹拌した。反応溶液に飽和炭酸水素ナトリウム水溶液を加えて塩化メチレンにて抽出し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(クロロホルム/ヘキサン)で精製し、標題化合物138mgを得た。
実施例14で得られた3-(15-メトキシペンタデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン150mgのメタノール3mL溶液に氷冷下塩化セリウム7水和物148mg、水素化ホウ素ナトリウム15mgを加え30分間撹拌したのち、室温で撹拌した。反応溶液に飽和塩化アンモニウム水溶液を加えて酢酸エチルにて抽出して無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物130mgを得た。
参考例19で得られた3-(15-メトキシペンタデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オ-ル200mgのテトラヒドロフラン4mL溶液にフタルイミド154mg、トリフェニルホスフィン275mgを加え、氷冷下ジイソプロピルアゾジカルボキシレート204μLを加え、室温にて終夜撹拌した。反応溶液を減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製したのち、エタノール4mL、ヒドラジン1水和物107μLを加えて終夜加熱撹拌した。反応溶液を室温まで放冷したのち、析出物を濾別した濾液を減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン)で精製し、標題化合物69.1mgを得た。
実施例14で得られた3-(15-メトキシペンタデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン500mgのエタノール8mL溶液に水4mL、ヒドロキシアミン塩酸塩550mg、炭酸カリウム1.09gを加えマイクロウェーブ装置で70℃、2時間30分間撹拌した。反応溶液に水を加えて酢酸エチルにて抽出し、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をNHシルカゲルカラムクロマトグラフィー(酢酸エチル/塩化メチレン)で精製し、標題化合物361mgを得た。
実施例14で得られた3-(15-メトキシペンタデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン300mgのエタノール3mL溶液にO-メチルヒドロキシアミン塩酸塩198mg、炭酸カリウム329mgを加えマイクロウェーブ装置で70℃、2時間30分間撹拌した。反応溶液に水を加えて酢酸エチルにて抽出し、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥した。乾燥剤を濾別したのち、減圧留去して得られた残渣をシルカゲルカラムクロマトグラフィー(塩化メチレン/ヘキサン)で精製し、標題化合物151mgを得た。
化合物を必要量秤量し、0.5%ヒドロキシプロピルメチルセルロースにて薬剤懸濁液を調製した。各群のラットの数は2匹とし、それぞれの薬剤懸濁液を経口ゾンデを用いて雄性ラット(Crl:CD(SD))に経口投与した(本化合物の投与量は10mg/kg)。各採血ポイント(投与後0.5、1、2、4、8及び24時間)にて注射筒及び注射針を用いて頚静脈より採血した(抗凝固剤はヘパリンナトリウム)。採取した血液は遠心分離(13000rpm、2分、4℃)により血漿を調製し、除タンパク処理を行った後に血漿中の化合物濃度をLC/MS/MS(LCMS-8040(shimadzu)またはAPI4000(AB SCIEX)、LC:30-A,20-Aシリーズ (shimadzu)またはWaters Acquity (waters))にて測定した。
糖尿病モデルにおける化合物の効果を検討するため、Saitohらの方法を参考に(European Journal of Pharmacology 501(2004)143-149)シストメトリー法による膀胱機能を解析した。
神経成長促進作用評価のため、Topalliらの方法を参考に(Brain Research 1030 (2004) 116-124)neurite bearing assay(有突起細胞割合)及びneurite length assay(神経突起長)を解析した。
Claims (15)
- 下記一般式(I)
R1は、-CH2OR4、-CH2OCON(R5)2、-CONHR6、又はシアノ基であり、
R2は、同一又は相異なって、水素原子、若しくはメチル基であるか、又は2個のR2が隣接する炭素原子と共にシクロプロピル基を形成しても良く、
R3は、水素原子、メチル基、-CH2OR7、又はハロゲン原子であり、
R4は、CmH2m+1であり、
R5は、同一又は相異なって、水素原子、又はメチル基であり、
R6は、水素原子、又はメチル基であり、
R7は、C1-C3アルキル基であり、
mは、1~14の整数であり、
nは、1~15の整数であり、
ただし、
R4が直鎖状のアルキル基である場合、mとnの合計は、12~17の整数であり、R4が分岐鎖状のアルキル基である場合、前記分岐鎖状のアルキル基の最も長い直鎖の炭素数をmbとし、mbとnの合計は、12~17の整数である。)で表される化合物又はその塩。 - 下記一般式(I)

R4は、メチル基、エチル基、n-プロピル基、又はイソプロピル基であり、

R4がメチル基である場合、nは、11~14の整数であり、
R4がエチル基、又はイソプロピル基である場合、nは、11~13の整数であり、
R4がn-プロピル基である場合、nは、11又は12であり、
R6は、水素原子、又はメチル基である。)で表される化合物又はその塩。 - 一般式(I)中、
Xは、-CH2-、-CH2-CH2-、又は-CH=CH-である、請求項1~3のいずれか1項記載の化合物又はその塩。 - 一般式(I)中、

R3が水素原子である場合、R2は水素原子であり、
R3がメチル基、-CH2OCH3、又はヨウ素原子である場合、R2は同一又は相異なって、水素原子、又はメチル基であるか、又は2個のR2が隣接する炭素原子と共にシクロプロピル基を形成しても良く、
R4は、メチル基、エチル基、n-プロピル基、又はイソプロピル基であり、
ただし、
R4がメチル基である場合、nは、11~14の整数であり、
R4がエチル基、又はイソプロピル基である場合、nは、11~13の整数であり、
R4がn-プロピル基である場合、nは、11又は12である、請求項1~4のいずれか1項記載の化合物又はその塩。 - 一般式(I)中、
Xは、-CH2-、又は-CH2-CH2-であり、
R1は、-CH2OR4、又は-CONH2であり、
R2は、メチル基であり、
R3は、メチル基、又は-CH2OCH3であり、

R4がエチル基、又はイソプロピル基である場合、nは、11~13の整数である、請求項1~5のいずれか1項記載の化合物又はその塩。 - 一般式(I)中、
Xは、-CH2-CH2-である、請求項1~6のいずれか1項記載の化合物又はその塩。 - 化合物が、次の(1)~(10)のいずれかに記載の化合物である、請求項1~7のいずれか1項記載の化合物又はその塩。
(1)3-(12-メトキシドデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン、
(2)3-(12-エトキシドデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン、
(3)3-(12-イソプロポキシドデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン、
(4)3-(13-メトキシトリデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン、
(5)3-(13-エトキシトリデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン、
(6)3-(13-イソプロポキシトリデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン、
(7)3-(14-メトキシテトラデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン、
(8)3-(14-エトキシテトラデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン、
(9)3-(14-イソプロポキシテトラデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン、
(10)3-(15-メトキシペンタデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オン。 - 化合物が、3-(15-メトキシペンタデシル)-2,4,4-トリメチルシクロヘキサ-2-エン-1-オンである、請求項1~8のいずれか1項記載の化合物又はその塩。
- 請求項1~9のいずれか1項記載の化合物又はその塩を有効成分として含有する神経突起成長剤。
- 請求項1~9のいずれか1項記載の化合物又はその塩を有効成分として含有する神経変性疾患治療剤。
- 請求項1~9のいずれか1項記載の化合物又はその塩を有効成分として含有する筋萎縮性側索硬化症治療剤。
- 請求項1~9のいずれか1項記載の化合物又はその塩を有効成分として含有する疼痛治療剤。
- 請求項1~9のいずれか1項記載の化合物又はその塩を有効成分として含有する下部尿路障害治療剤。
- 請求項1~9のいずれか1項記載の化合物又はその塩と、薬学的担体とを含有する医薬組成物。
