실시예 5: 피험자들의 개선도 평가Example 5: Evaluation of improvement in subjects
현호색 추출물 처리구과 무처리구에게 실험 참여 전후의 냄새 변화에 대한 설문조사를 실시하였다. 설문조사는 공통적으로 개인 신상, 도포방법의 준수여부, 액취 고민 여부에 대해 조사하였다. 그리고 현호색 추출물 처리구에 대해서는 도포 전후의 액취 정도, 및 개선 여부에 대해 조사하였고, 무처리구에 대해서는 실험참여 전후의 액취 정도에 대해 조사하였다. 액취의 정도는 '매우 나쁨(5점)', '나쁨(4점)', '보통(3점)', '좋음(2점)', '매우 좋음(1점)' 5단계로 구별하였고, 개선 여부는 긍정과 부정 2단계로 평가하였다. 액취 정도 및 개선 여부는 본인의 주관적 평가를 기준으로 하였다. 설문조사 결과는 하기 표 1 및 표 2에 나타내었다. 현호색 추출물 처리구 8명은 모두 도포 방법을 준수하였으며 특별한 부작용이 없다고 답변하였다. 상기 표 1에 따르면, 현호색 추출물을 도포하기 전에는 액취의 정도가 '매우 나쁨'으로 답변한 피험자는 1명이고, '나쁨'으로 답변한 피험자는 5명, '보통'으로 답변한 피험자가 2명이었다. 현호색 처리구 8명 중 7명이 현호색 추출물 사용에 의해 액취가 개선되었다고 답변하였다. 실시예 6: 액취증이 개선된 피험자의 미생물 군집의 변화 확인Example 6: Confirmation of changes in the microbial community of subjects with improved bromhidrosis
상기 실시예 1에서 준비한 미생물의 군집의 변화를 분석하였다. PowerSoil DNA Pro Kit (QIAGEN, Germany)를 이용하여 미생물의 total DNA를 추출하고, 16S rRNA V4-V5 영역(515F: 5′-GTGCCAGCMGCCGCGG-3′, 907R: 5′-CCGTCAATTCMTTTRAGTTT-3′)을 PCR로 증폭하였다. 증폭 산물을 Illumina MiSeq 장비를 사용하여 서열을 분석하였다. 서열 데이터는 QIIME version 1.9.1를 통해 OTUs picking을 진행하였고, Greengenes database에 등재되어 있는 표준 염기서열과 97% 이상의 상동성을 기준으로 박테리아를 분류하였다.Changes in the community of microorganisms prepared in Example 1 were analyzed. Total DNA of microorganisms was extracted using PowerSoil DNA Pro Kit (QIAGEN, Germany), and 16S rRNA V4-V5 region (515F: 5′-GTGCCAGCMGCCGCGG-3′, 907R: 5′-CCGTCAATTCMTTTRAGTTT-3′) was subjected to PCR amplified. Amplification products were sequenced using Illumina MiSeq equipment. For sequence data, OTUs picking was performed through QIIME version 1.9.1, and bacteria were classified based on 97% or more homology with the standard nucleotide sequence registered in the Greengenes database.
도 4는 실험에 참여한 16명의 피험자의 미생물 군집 변화를 주좌표분석 (Principal Coordinate Analysis; PCoA) 방법을 통해 나타낸 것이다. 도 4에 따르면, 현호색 추출물 처리구는 현호색 추출물 사용 전과 후의 미생물 군집의 차이가 뚜렷하였다. 구체적으로 현호색 추출물 사용 전후에 좌표상 위치가 크게 변화하였고(도 4의 붉은색 원과 파란색 원 참조), 좌표상 위치들을 종합한 면적의 크기 및 위치 또한 크게 변화하였다(도 4의 붉은색 면적과 파란색 면적 참조). 유사성 분석 (Analysis of similarities; ANOSIM) 결과, 현호색 추출물 사용 전과 사용 후의 미생물 군집은 통계적으로 유의한 차이를 보였다. 반면 무처리군 8명의 미생물 군집은 실험 참여 전후의 좌표상 위치 및 면적의 차이가 크지 않고, 유사성 분석 결과에서도 통계적으로 유의한 차이를 나타내지 않았다. Figure 4 shows the changes in the microbial community of 16 subjects participating in the experiment through the principal coordinate analysis (PCoA) method. 도 5는 현호색 추출물 처리구 8명과 무처리구 8명 각각의 미생물 군집 비율의 평균값을 나타낸 것이다. 도 5에 따르면 현호색 추출물 처리구는 현호색 추출물 사용에 의해 미생물 군집 내에서 액취를 유발하는 코리네박테리움속 및 아나에로코커스속 미생물이 차지하는 비율이 크게 감소하였다. 반면 프로피오니박테리움속(Propionibacterium), 세라티아속(Serratia), 크리세오박테리움속(Chryseobacterium), 라이시니바실러스속(Lysinibacillus) 균주의 비율은 증가하였다. 반면 무처리구 8명은 미생물 군집 내 미생물의 비율이 크게 변화하지 않았다. 다만, 현호색 추출물의 사용에 의해 코리네박테리움속 및 아나에로코커스속 2 종의 미생물이 감소하였더라도, 이러한 결과가 현호색 추출물의 상기 코리네박테리움속 및 아나에로코커스속 2 종의 미생물에 대한 직접적인 항균 또는 활성 억제 효과에 의한 것이라고 단정할 수는 없다. 현호색 추출물이 다른 미생물의 군집을 증가시킴으로써 코리네박테리움속 및 아나에로코커스속 미생물을 감소시킨 것일 수도 있고, 또는 현호색 추출물이 코리네박테리움속 및 아나에로코커스속 미생물과 상호 작용할 수 있는 제3의 미생물의 활성을 변화시킴으로써 간접적으로 코리네박테리움속 및 아나에로코커스속 미생물을 감소시킨 것일 수도 있기 때문이다. 상기 실험결과를 종합하면 현호색 추출물은 액취 물질을 생산하는 코리네박테리움속 및 아나에로코커스속 세균의 군집 내 비율을 감소시킬 수 있으므로 액취증 개선 효과를 가질 수 있는 것으로 판단된다.
