ウシパピローマウイルスに起因する牛乳頭腫症を治療等するための組成物及びそれを用いた方法の提供。 下記式(I)で表される化合物又はその製薬上許容される塩を有効成分として含有する、ウシパピローマウイルスに起因する牛乳頭腫症の治療、改善、及び/又は進行抑制用組成物、及び下記式(I)で表される化合物又はその製薬上許容される塩を含有する組成物を、ウシに投与することを含む、ウシパピローマウイルスに起因する牛乳頭腫症の治療、改善、及び/又は進行抑制方法。
Description
本開示は、ウシパピローマウイルス(BPV)に起因する牛乳頭腫症を治療、改善、及び/又は進行抑制するための組成物、並びにそれを用いたウシにおけるBPVに起因する牛乳頭腫症の治療、改善、及び/又は進行抑制方法に関する。
乳頭腫症は、パピローマウイルス(PV)が上皮又は粘膜に感染することによって、感染部位に乳頭腫と呼ばれる良性腫瘍(疣)を形成する疾患である。パピローマウイルスが感染する哺乳動物としては、ヒトをはじめとして、牛、馬、犬、猫及びヤク等が知られている。ヒトを除く哺乳動物のPVとしては、BPVがもっともよく知られている。
BPVは、牛の皮膚や一部の粘膜に感染することによって牛乳頭腫症を引き起こす。牛乳頭腫症は、若齢牛でしばしば発症し発症頻度は非常に高いが、一般には良性であり、多くは自然治癒する場合が多いため、あまり問題にされていなかった。しかしながら、乳頭腫の病変が乳用牛の乳頭部に形成した場合、搾乳の障害になったり、細菌の二次感染を惹起して乳房炎につながったりする場合がある。近年、乳用牛のミルカーを介するなどして乳頭部のパピローマが農場全体に広がるなどといったBPV感染拡大の影響が報告されている。
牛乳頭腫の治療方法としては、外科的に除去する方法、すり潰した疣を無毒化させて注射する方法、飼料に殻付ハトムギを混ぜて摂取させる方法、及び患部に木酢酸を塗布する方法等が試みられている。
本出願人らは、ヒトパピローマウイルス(HPV)に起因するウイルス感染症に対する抗ウイルス剤を提案している(特許文献1)。
上記のとおり、牛乳頭腫症による問題が生じているにもかかわらず、牛乳頭腫症の根本的な治療法は存在しないといえる。したがって、牛乳頭腫症の治療法が望まれている。
本開示は、一態様において、下記式(I)で表される化合物又はその製薬上許容される塩を有効成分として含有する、ウシパピローマウイルスに起因する牛乳頭腫症の治療、改善、及び/又は進行抑制用組成物に関する。

本開示は、一態様において、ウシパピローマウイルスに起因する牛乳頭腫症を治療、改善、及び/又は進行抑制する方法であって、上記式(I)で表される化合物又はその製薬上許容される塩を含有する組成物を、ウシに投与することを含む、牛乳頭腫症の治療、改善、及び/又は進行抑制方法に関する。
本開示は、一態様において、BPVに起因する牛乳頭腫症を治療、改善、及び/又は進行抑制可能な組成物、及びそれを用いた治療、改善、及び/又は進行抑制可能な方法を提供できる。
上記のとおり、PVは、牛、馬、猫、犬及びヒト等といった様々な動物から多くのウイルス型が見つかっており、世界中に広く分布しているウイルスである。PVは多種多様であり、PVが感染する種(宿主動物)によって、PVの遺伝子型及び病態が異なる。さらに、PVは、一般的に種特異性が高く、通常、種を超えての感染が起こらないと考えられている。とりわけ、BPVが感染する対象は牛であり、ヒトがBPVに感染することはない。また、HPVが牛に感染することはない。
BPVは、牛に特異的に感染し、牛の乳頭腫を引き起こすパピローマウイルスである。BPVに起因する牛乳頭腫症の治療法として、外科的除去や病変部の凍結による除去以外に、すり潰した疣を無毒化させて注射する方法、殻付ハトムギの摂取及び患部への木酢酸の塗布等が試みられている。しかしながら、外科的除去の場合、再発率が高く、またその他の方法についても十分な効果が得られているとはいえない。
本開示は、種特異性の高さから、HPVに起因するヒトの子宮頸がんに対するワクチンや治療法が提案されている化合物、すなわち特許文献1に開示されたHPVに対する抗ウイルス剤では、BPVに起因する牛乳頭腫症に対しては治療効果が期待できないと考えられていたところ、BPVに起因する牛乳頭腫症に対して予想外の効果が得られた、との知見に基づく。
本開示は、抗ウイルス作用を示す下記式で表される化合物:N-[5-フルオロ-2-(1-ピペリジニル)フェニル]-4-ピリジンチオアミドを、BPVに起因する牛の乳頭腫(疣)に塗布することにより、既に形成した乳頭腫(疣)の拡大を抑制し、さらには疣の消失又はサイズが縮小できるという優れた効果を発揮しうる、という知見に基づく。
本開示における「乳頭腫症の治療、改善、及び/又は進行抑制」としては、一又は複数の実施形態において、本開示の薬剤(組成物)を対象に投与することによる、既に形成した乳頭腫(疣)のサイズの拡大の抑制、疣の消失、又は疣のサイズの縮小等が挙げられる。
本開示における「BPVに起因する牛乳頭腫症」としては、一又は複数の実施形態において、牛がBPVに感染することによって、感染部位に乳頭腫を形成する疾患をいう。BPVに起因する牛乳頭腫症としては、一又は複数の実施形態において、牛乳頭腫症、皮膚型牛乳頭腫症、乳頭型牛乳頭腫症、及びサルコイド(繊維乳頭症)等が挙げられる。感染部位(検出病変)としては、特に限定されない一又は複数の実施形態において、乳頭部、皮膚、及び消化管(口腔及び食道等)等が挙げられる。

本開示は、一態様において、BPVに起因する牛乳頭腫症を治療、改善、及び/又は進行抑制するための組成物であって、該組成物は、下記式(I)で表される化合物又はその製薬上許容される塩を有効成分として含有する。式(I)で表される化合物は、一又は複数の実施形態において、抗DNAウイルス作用及び/又は抗RNAウイルス作用を有しうる。
式(I)において、Rは、酸素原子又は硫黄原子を示す。

本開示における式(I)で表される化合物としては、一又は複数の実施形態において、下記式で表される化合物:N-[5-フルオロ-2-(1-ピペリジニル)フェニル]-4-ピリジンチオアミド(化合物A)及びN-[5-フルオロ-2-(1-ピペリジニル)フェニル]-4-ピリジンカルボキサミド(化合物B)が挙げられる。
式(I)で表される化合物は、一又は複数の実施形態において、公知の製造方法やWO2009/020198を参照して製造することができる。

本開示の組成物中の有効成分である式(I)で表される化合物の含有量は、一又は複数の実施形態において、0.01重量%~50重量%、又は0.1重量%~20重量%である。
本開示の組成物の投与量は、剤型及び投与経路に応じ、適宜選択できる。局所投与の場合の本開示の組成物の投与量は、疣の大きさ及び範囲等に応じて適宜設定でき、一又は複数の実施形態において、組成物中の有効成分の量が、約0.01mg/疣~40mg/疣となるように投与することが挙げられる。例えば、これに限定されないが、本開示の組成物を乳頭に塗布して用いる場合、組成物中の有効成分の塗布量は、乳頭表面の面積に対して、0.01mg/cm2~40mg/cm2、好ましくは0.1mg/cm2~20mg/cm2、より好ましくは0.3mg/cm2~6mg/cm2で投与することが挙げられる。本開示の組成物の投与量としては、経口投与又は注射の場合、一又は複数の実施形態において、組成物中の有効成分の量が約0.01mg/kg(体重)~100mg/kg(体重)となるように投与することが挙げられる。
本開示の組成物を、乳頭に局所投与する場合、或いは、乳頭に塗布して投与する場合、本開示の組成物が投与又は塗布された乳頭全体、或いは投与又は塗布された部分を被覆するのが好ましい。被覆することにより、患部への有効成分の投与又は浸透がより効果的になる。乳頭部分の被覆は、乳頭を被覆できるものであれば特に制限なく用いることができるが、例えば、乳頭被覆容器又は被覆材を用いることができる。乳頭被覆容器としては、これに限定されないが、例えば、弾力性の材質で作られた乳頭の形状を有した容器(例えば、サック)を挙げることができる。弾力性の材質としては、一般に用いられている弾力性のある高分子素材であれば特に制限なく用いることができるが、好ましくは乳頭に対する刺激が少ない素材を挙げることができる。また、患部がある乳頭の状態を良好に保つために、本開示の組成物が漏出しないが通気性のある素材が好ましい。これに限定されないが、例えば、天然ゴム素材、シリコン素材、及びラテックス素材等を挙げることができる。乳頭被覆材としては、乳頭に塗布した後、皮膜を形成するものであれば特に制限なく用いることができるが、好ましくは乳頭に対する刺激が少ない成分からなる被覆材である。例えばこれに限定されないが、乳頭などの皮膚に塗布することにより被膜を形成することができる天然の成分からなる又はそのような天然の成分の混合物からなる組成物(被膜剤)を挙げることができる。これらの被覆材は、本開示の組成物が投与又は塗布された乳頭に、塗布又はスプレーすることにより、そこに被膜を形成できる。形成される被膜は、本開示の組成物が漏出しないが通気性のあることが好ましい。
また、一又は複数の実施形態において、本開示の組成物を予め乳頭保護容器又は被覆材に塗布し、それで乳頭を覆うことによって、本開示の組成物をウシに投与してもよい。

乳頭被覆容器及び被覆材の一実施形態を図1に示す。図1(a)は、ウシの乳頭を、円柱状の袋(指サック形状)の乳頭被覆容器で覆った様子を示す図である。図1(b)~(d)は、パッド(パッチ又はシート)状の乳頭被覆材の形状の一例を示す。乳頭被覆材の形状は、四角形(図1(b))であってもよいし、アンカー形状(図1(c))であってもよいし、H型形状(図1(d))であってもよい。乳頭被覆材は、一又は複数の実施形態において、図1(b)に示すように、ウシの乳頭の先端に、乳頭被覆材の中心部を当接させ、その状態で乳頭全体を覆うように貼付することにより、乳頭を被覆することができる。
本開示の組成物は、乳頭被覆容器又は被覆材とともに用いるのが好ましく、組成物を乳頭に投与又は塗布した後に乳頭被覆容器又は被覆材で乳頭を覆うことにより、有効成分の投与や浸透がより有効となる。乳頭被覆容器又は被覆材とともに用いられることを特徴とする本開示の組成物もまた本開示に含まれる。本開示はさらに、本開示の組成物と乳頭被覆容器又は被覆材との組合せも含む。このような組合せはセット又はキットとすることもできる。本開示の組成物はまた、予め乳頭被覆容器の内側に塗布し、容器を乳頭に被せることによっても牛に投与できる。このような本開示の組成物が内側に塗布された乳頭被覆容器もまた本開示に含まれる。
本開示の組成物は、ウシにおけるBPVに起因する牛乳頭腫症の治療、改善、及び/又は進行抑制に使用できる。乳頭腫の病変が乳頭である場合、一又は複数の実施形態において、病変である乳頭に本開示の組成物を塗布することによって、乳頭に形成した乳頭腫の拡大の抑制、乳頭腫の縮小、及び/又は乳頭腫の消失を生じさせうる。よって、一又は複数の実施形態において、本開示の組成物を乳用牛に投与することにより、乳頭に乳頭腫が形成されることに伴う搾乳の障害を防止することができうる。本開示の組成物は、病変が乳頭である場合、一又は複数の実施形態において、乳頭腫(疣)が発生した乳頭全体に塗布してもよいし、乳頭に発生した乳頭腫(疣)のみに塗布してもよい。
但し、投与経路はこれらに限定されるものではなく、経口投与であってもよいし、注射によって投与してもよい。

[乳頭腫症の治療、改善、及び/又は進行抑制方法]
本開示は、その他の態様において、ウシにおけるBPVに起因する牛乳頭腫症の治療、改善、及び/又は進行抑制方法に関する。本開示の方法は、上記式(I)で表される化合物又はその製薬上許容される塩を含有する組成物を、対象であるウシに投与することを含む。

本開示は以下の一又は複数の実施形態に関しうる。

以下、実施例により本開示をさらに詳細に説明するが、これらは例示的なものであって、本開示はこれら実施例に制限されるものではない。なお、本開示中に引用された文献のその全体は、本開示の一部として組み入れられる。
製造例1:
N-[5-フルオロ-2-(1-ピペリジニル)フェニル]-4-ピリジンチオアミド(化合物A)の製造
下記化学式で表される化合物Aを、WO2009/020198の参考例11を参照して製造した。

製造例2:化合物Aを含有する軟膏剤の調製
化合物A 120mgを、あらかじめ60℃に加温して溶解した軟膏基剤(白色ワセリン)3880mgに溶解し、60℃で均一になるまで撹拌することにより軟膏剤(化合物A:3重量%)を調製した。

[乳頭腫発症乳牛に対する治療効果確認試験]
前部乳頭の両方に乳頭腫を有する雌性乳用牛(4頭)において、前部乳頭の片方の乳頭の乳頭腫(疣)に、化合物Aを含有する軟膏剤を1日1回塗布した(試験区)。塗布後の乳頭は外界からの汚染の防止等のため、ドレッシングテープ(Navis、プロシェアドレッシングロール、No.8-5965-01)を巻き付けた。軟膏剤は、乳頭の表面に、表面積1cm2あたり、約0.01g~約0.2g(有効成分約0.3mg~約6mg相当量)塗布した。塗布(治療)は5投薬2休薬で4週間継続して行った。1週間に一度、患部画像を撮影した。塗布開始から4週間経過後での患部(疣)の状態に基づき薬効を評価した。
前部乳頭の両方に乳頭腫を有する雌性乳用牛(4頭)において、前部乳頭の片方の乳頭の乳頭腫(疣)に、化合物Aを含有する軟膏剤を1日1回塗布した(試験区)。塗布後の乳頭は外界からの汚染の防止等のため、ドレッシングテープ(Navis、プロシェアドレッシングロール、No.8-5965-01)を巻き付けた。軟膏剤は、乳頭の表面に、表面積1cm2あたり、約0.01g~約0.2g(有効成分約0.3mg~約6mg相当量)塗布した。塗布(治療)は5投薬2休薬で4週間継続して行った。1週間に一度、患部画像を撮影した。塗布開始から4週間経過後での患部(疣)の状態に基づき薬効を評価した。
コントロールとして、化合物Aを含有しない軟膏基剤を、試験区としていないもう片方の乳頭の疣に塗布した(対照区)。
[評価方法1]
4週間の投薬後の疣の変化(縮小/消失、変化なし、及び拡大/発生)に対し、下記の3段階評価のスコアを与えた。なお、「変化なし」は、拡大抑制と効果なしが含まれうる。「発生」は、4週間の投薬中に新たに発生した疣をいう。
縮小/消失=+1、変化なし=0、拡大/発生=-1
その結果を下記表1に示す。
4週間の投薬後の疣の変化(縮小/消失、変化なし、及び拡大/発生)に対し、下記の3段階評価のスコアを与えた。なお、「変化なし」は、拡大抑制と効果なしが含まれうる。「発生」は、4週間の投薬中に新たに発生した疣をいう。
縮小/消失=+1、変化なし=0、拡大/発生=-1
その結果を下記表1に示す。
上記表1において、試験区の合算スコアは+1であり、対照区の合算スコアは-8であるから、試験区での化合物Aを含有する軟膏剤の効果が認められた。
上記表2において、個体ごとで評価すると、4例中3例が有効であり、化合物Aを含有する軟膏剤の効果が認められた。

なお、4週間の投薬後、同様の条件でさらに4週間投薬を継続して行った。その結果、試験区においてはさらに4個の疣が縮小した(うち2個は消失)。
[評価方法2]
小さい疣のみを対象とした場合、疣の拡大や縮小が顕著に表れ、化合物Aの効果が判定易いと考えられる。このため、上記確認試験における試験区及び対照区から軟膏剤の塗布前の大きさが5mm未満の疣のみを抽出し、縮小/消失した疣の割合を算出した。
その結果、解析対象の疣(試験区14個、対象区15個)において、試験区では縮小/消失した疣の割合が28.6%であるに対し、対照区では0%であった。よって、化合物Aを含有する軟膏剤による、ウシに発生した乳頭腫に対する明確な有効性が確認できた。
小さい疣のみを対象とした場合、疣の拡大や縮小が顕著に表れ、化合物Aの効果が判定易いと考えられる。このため、上記確認試験における試験区及び対照区から軟膏剤の塗布前の大きさが5mm未満の疣のみを抽出し、縮小/消失した疣の割合を算出した。
その結果、解析対象の疣(試験区14個、対象区15個)において、試験区では縮小/消失した疣の割合が28.6%であるに対し、対照区では0%であった。よって、化合物Aを含有する軟膏剤による、ウシに発生した乳頭腫に対する明確な有効性が確認できた。
Claims (13)
- 前記乳頭腫症が、乳頭型牛乳頭腫症である、請求項1又は2に記載の組成物。
- 乳用牛の乳頭に塗布するための局所用組成物である、請求項1から3のいずれかに記載の組成物。
- 前記化合物又はその製薬上許容される塩が、0.01mg/cm2~40mg/cm2にて乳頭腫が存在する乳頭に塗布される請求項4に記載の組成物。
- 乳頭を覆うことができる乳頭被覆容器又は被覆材とともに用いられ、前記組成物を乳頭に塗布した後に該乳頭被覆容器又は被覆材によって乳頭が被覆されるようして用いられる請求項4又は5に記載の組成物。
- 前記乳頭被覆容器又は被覆材が、乳頭を覆う容器又は乳頭に塗布することにより皮膜を形成することができる被覆材である請求項6に記載の組成物。
- 前記ウシが乳用牛であって、前記乳頭腫症が乳頭型牛乳頭腫症であり、
前記乳用牛の乳頭に、前記組成物を塗布することを含む、請求項8又は9に記載の方法。 - 前記化合物又はその製薬上許容される塩が、0.01mg/cm2~40mg/cm2にて乳頭腫が存在する乳頭に塗布される請求項10に記載の方法。
- 前記化合物又はその製薬上許容される塩を乳頭に塗布した後に、塗布した部位を乳頭被覆容器又は被覆材によって被覆することをさらに含む請求項10又は11に記載の方法。
- 前記頭被覆容器又は被覆材が、乳頭を覆う容器又は乳頭に塗布することにより皮膜を形成することができる被覆材である請求項12に記載の方法。
