Abstract
본 발명은 불포화 지방산 결합 CP2c 표적 펩타이드 기반 항암제에 관한 것이다. 본 발명에서는 범용 항암제로서의 효과를 보인 CP2c 표적 펩타이드 선도물질 ACP52C에 불포화 지방산을 연결시켜 항암 효과에 영향이 없이 생체내 안정성을 향상시킴을 확인하였다. 또한 각 종 암세포 및 정상세포에서 암세포 특이적 항암 효력을 확인하였다.

본 발명은 CP2c 표적 펩타이드 기반 항암제에 관한 것이다.
현재까지 많은 단백질/펩타이드성 항암제와 저분자 화합물 기반 항암제가 개발되어 사용되고 있으나, 생체 내에서 암세포에만 선택적으로 작용하지 않고 정상세포에도 영향을 나타내는 심각한 부작용을 유발하며, 약제 내성을 가진 암세포의 출현으로 그 효력이 미미하게 작용하고 있다. 뿐만 아니라 개발 중인 많은 단백질/펩타이드성 항암제가 생체 내에서 짧은 반감기를 가지고 있어서, 이를 극복하기 위한 다양한 연구가 진행 중이다. 이와 같이 전 세계적으로 항암제를 개발하기 위한 노력을 경쟁적으로 하고 있지만, 생체내 안정성, 안전성 및 약제 내성에 대한 문제점으로부터 자유로운 항암제는 아직까지 개발되고 있지 않다. 따라서 생체 내에서 장기간 지속될 수 있는 안정성을 확보하고, 다양한 종류의 암세포만을 선택적으로 제거할 수 있으며, 약제 내성 암세포에도 작용하는 항암제가 개발될 수 있다면 범세계적으로 고부가 가치를 가져올 수 있을 것이다.
본 발명은 생체 내에서 장기간 지속될 수 있는 안정성을 확보하고, 다양한 종류의 암세포만을 선택적으로 제거할 수 있으며, 약제 내성 암세포에도 작용하는 CP2c 표적 펩타이드 기반 항암제를 제공하는 것을 목적으로 한다.
본 발명에 따른 CP2c 표적 펩타이드는 전사인자 CP2c에 결합하여 CP2c를 포함하는 전사인자 복합체 (CP2c homotetramer 및 CP2c/CP2b/PIAS1 heterohexamer) 형성을 억제함으로써 암세포 특이적 세포사를 유도하는 펩타이드를 의미한다. CP2c 표적 펩타이드는 Tyr-Pro-Gln-Arg (서열번호 1)가 항암 효과에 필수적인 아미노산들만으로 구성된 최소 크기의 펩타이드에 해당한다. 따라서, 상기 4개의 아미노산을 필수적으로 포함하며, CP2c 단백질과 상호작용함으로서 항암 효과를 나타내는 펩타이드를 본 발명에 따른 CP2c 표적 펩타이드로서 사용할 수 있다.
본 발명에 따른 CP2c 표적 펩타이드는 상기 서열번호 1의 아미노산 서열을 포함하는 4개 내지 20개의 아미노산으로 이루어진 펩타이드일 수 있다. 바람직하게는 6개의 아미노산(6 aa)'NYPQRP(Asn-Try-Pro-Gln-Arg-Pro, 서열번호 2)'을 포함하는 펩타이드(ACP52)일 수 있다.
본 발명에 따른 CP2c 표적 펩타이드는 세포 투과 활성의 증진을 위해, 세포막 투과 펩타이드(CPP: cell-penetrating peptide)를 결합시켜 사용할 수 있다.
예를 들어, 본 발명에 따른 CP2c 표적 펩타이드는 NYPQRP(Asn-Try-Pro-Gln-Arg- Pro)으로 이루어진 펩타이드(ACP52)와 세포막 투과 펩타이드(CPP: cell-penetrating peptide)로서 9개의 아미노산(9aa) 'CRGDKGPDC(Cys-Arg-Gly-Asp-Lys-Gly-Pro-Asp-Cys, 서열번호 3)'로 이루어진 펩타이드인 iRGD(internalizing RGD)를 포함하는 펩타이드일 수 있다. 본 발명에 따른 CP2c 표적 펩타이드의 일예인 'ACP52C'는 ACP52의 C-말단 Pro에 iRGD가 결합되어 있으며, iRGD의 C-말단에 NH
2(아미드기, amide group)가 결합되어 있는 펩타이드이다.
이전 연구에서 본 출원인에 의해 종래 개발된 CP2c 표적 펩타이드의 생체 내 안정성을 확보할 목적으로 포화지방산인 팔미토일산과 결합된 CP2c 표적 펩타이드-지방산 컨쥬게이트를 제조하였고, CP2c 표적 펩타이드에 지방산을 결합시키면 혈액 내 알부민과 결합함으로써 생체 내 안정성이 증가되어 높은 치료 효과를 제공할 것으로 예상하였다. 그러나 그 실제 효과는 CP2c 표적 펩타이드를 단독으로 사용하는 경우보다 열등하였고, 이는 팔미토일산은 포화지방산으로 서로 응집하여 micelle 구조를 형성함으로써 알부민과 결합도 못할 뿐만 아니라 세포투과성 펩타이드인 iRGD도 micelle 내부로 들어가게 되어 암세포로의 전달도 함께 억제되었을 것으로 판단하였다. 이와 같은 이와 같은 문제점을 극복할 수 있는 방법으로 micelle을 형성할 수 없는 불포화 지방산을 사용할 수 있을 것이라 판단하고 all-cis-docosa-4,7,10,13,16,19-hexa-enoic acid(DHA)를 ACP52 및 ACP52C에 각각 결합시킨 복합체를 합성하여 다양한 암세포주 및 Hep3B xenograft 생쥐모델에서 항암제로서의 특성을 분석한 결과, 우수한 효과를 확인함으로써 본 발명을 완성하였다.
이에 제한되는 것은 아니나, 상기 불포화 지방산은 C
12 내지 C
22의 지방산일 수 있다. 본 발명의 일 구체예에서, 상기 불포화 지방산은 C
22 불포화 지방산, 예컨데, DHA(all-cis-docosa-4,7,10,13,16,19-hexa-enoic acid)일 수 있다.
본 발명에 따른 CP2c 표적 펩타이드-지방산 컨쥬게이트는 CP2c 표적성을 제공하는 CP2c 표적 펩타이드(예컨대, ACP52)와 CPP(예컨대, iRGD) 및/또는 지방산 사이에 이들을 연결해 주는 링커 펩타이드를 포함할 수 있다. 상기 링커 펩타이드는 당업계에 알려진 아미노산 또는 이들의 조합으로 이루어지는 펩타이드가 제한없이 사용될 수 있다. 구체적으로, 상기 링커 펩타이드는 글리신(Gly, G), 예컨대 Gn(여기서 n은 1 내지 6의 정수)을 포함할 수 있다.

본 발명에 따른 CP2c 표적 펩타이드-지방산 컨쥬게이트 합성시, 상기 글루탐산(Glu, E) 또는 단백질 분해효소 Cathepsin B의 표적서열인 EGLFG로 표시되는 아미노산 서열은 GnKGm 표시되는 링커 펩타이드와 먼저 결합된 후, 지방산과 연결될 수 있지만, 상기 합성 순서에 한정되는 것은 아니다.
한편, 본 발명에서 사용된 CP2c 표적 펩타이드-지방산 컨쥬게이트는 펩타이드의 향상된 안정성, 강화된 약리 특성 (반감기, 흡수성, 역가, 효능 등), 변경된 특이성 (예를 들어, 광범위한 생물학적 활성 스펙트럼), 감소된 항원성을 획득하기 위하여, 펩타이드의 N- 및/또는 C-말단이 수식(modification)되어 있을 수 있다. 바람직하게, 상기 수식은 상기 펩타이드의 N- 및/또는 C-말단에 아세틸기, 플루오레닐 메톡시 카르보닐기, 아미드기, 포르밀기, 미리스틸기, 스테아릴기 또는 폴리에틸렌글리콜(PEG)이 결합된 형태일 수 있으나, 펩타이드의 개질, 특히 펩타이드의 안정성을 향상시킬 수 있는 성분이라면, 제한없이 포함할 수 있다. 본 발명에서 사용되는 용어, "안정성"은 생체 내 단백질 절단효소의 공격으로부터 본 발명의 펩타이드를 보호하는 인 비보 안정성뿐만 아니라, 저장 안정성 (예컨대, 상온 저장 안정성)도 의미한다. 본 발명에 따른 CP2c 표적 펩타이드-지방산 컨쥬게이트에서 펩타이드의 N-말단은 불포화 지방산과 결합되어 있으므로 C-말단만 아미드기로 수식되어 있는 구조일 수 있다.

본 발명의 실시예에서는, 합성된 4종의 CP2c 표적 펩타이드-지방산 컨쥬게이트를 사용하여 간암세포이종이식 (Hep3B), 유방암세포이종이식 (MDA-MB-231) 생쥐모델에서 생체내 안정성, 항암 효력, 종양 전이 억제 효력 및 생리 독성을 분석하였고, 그 결과, 생체 내 안정성, 종양 억제 및 종양 전이 억제 효과가 우수함을 확인하였고, 생체에 대하여 독성이 없는 안전한 물질임을 확인하였다.
이에 따라, 본 발명은 또한, 본 발명에 따른 CP2c 표적 펩타이드-지방산 컨쥬게이트를 유효성분으로 포함하는 암 예방, 개선 또는 치료용 약학 조성물 및 건강기능식품 조성물을 제공한다.
본 발명에서 상기 암은 항암제, 특히, 본 발명에 따른 CP2c 표적 펩타이드-지방산 컨쥬게이트 기반 항암제에 대한 저항성을 나타내는 내성암도 포함될 수 있다.

본 발명에서 용어 "치료"는 본 발명에 따른 펩타이드 또는 이를 포함하는 약학 조성물의 투여로 암의 증세가 호전되거나 이롭게 변경되는 모든 행위를 의미한다.
본 발명에서, "유효 성분으로 함유"라는 용어는, 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 유효 용량 수준은 환자의 질환의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료 기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 본 발명에 따른 펩타이드 또는 이를 포함하는 약학 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고, 종래의 치료제와 순차적으로 또는 동시에 투여될 수 있으며, 단일 또는 다중 투여될 수 있다. 상기한 요소들을 모두 고려하여 부작용없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 이는 당업자에 의해 용이하게 결정될 수 있다. 본 발명의 약학 조성물의 투여량과 횟수는 치료할 질환, 투여 경로, 환자의 연령, 성별 및 체중 및 질환의 중등도 등의 여러 관련 인자와 함께, 활성성분인 약물의 종류에 따라 결정된다.
따라서, 본 발명에 따른 약학 조성물은 본 발명에 따른 펩타이드가 유효 성분으로 함유되는 한, 약학적으로 허용 가능한 다양한 담체를 포함할 수 있다. 약학적으로 허용되는 담체는 경구 투여용의 경우에는 결합제, 활택제, 붕해제, 부형제, 가용화제, 분산제, 안정화제, 현탁화제, 색소, 향료 등을 사용할 수 있으며, 주사제의 경우에는 완충제, 보존제, 무통화제, 가용화제, 등장화제, 안정화제 등을 혼합하여 사용할 수 있고, 국소 투여용의 경우에는 기제, 부형제, 윤활제, 보존제 등을 사용할 수 있다. 본 발명의 약학 조성물의 제형은 상술한 바와 같은 약학적으로 허용되는 담체와 혼합하여 다양하게 제조될 수 있다. 예를 들어, 경구 투여용의 경우에는 정제, 트로키, 캡슐, 엘릭서, 서스펜션, 시럽, 웨이퍼 등의 형태로 제조할 수 있으며, 주사제의 경우에는 단위 투약 앰플 또는 다수회 투약 형태로 제조할 수 있다. 기타 용액, 현탁액, 정제, 환약, 캡슐, 서방형 제제 등으로 제형화할 수도 있다.
한편, 제제화에 적합한 담체, 부형제 및 희석제의 예로는 락토즈, 덱스트로즈, 수크로즈, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로즈, 폴리비닐피롤리돈, 물, 메틸하이드록시벤조에이트, 프로필하이드록시벤조에이트, 탈크, 마그네슘 스테아레이트 또는 광물유 등이 사용될 수 있다. 또한, 충진제, 항응집제, 윤활제, 습윤제, 향료, 방부제 등을 추가로 포함할 수 있다.
본 발명의 약학적 조성물은 약학적으로 유효한 양으로 투여한다. 본 발명에 있어서 약학적으로 유효한 양은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 유효용량 수준은 환자의 질환의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출비율, 치료기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 본 발명에 다른 약학적 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 순차적 또는 동시에 투여될 수 있으며, 단일 또는 다중 투여될 수 있다. 상기한 요소들을 모두 고려하여 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 이는 당업자에 의해 용이하게 결정될 수 있다.
본 발명에서 용어 "투여"는 임의의 적절한 방법으로 대상에게 소정의 물질, 즉 본 발명에 따른 펩타이드 유도체 또는 이를 포함하는 약학 조성물을 도입하는 것을 의미하고, 이의 투여 경로는 약물이 목적 조직에 도달할 수 있는 한 어떠한 일반적인 경로를 통하여 투여될 수 있다. 예컨대, 투여 경로는 복강 내 투여, 정맥 내 투여, 근육 내 투여, 피하 투여, 피내 투여, 경구 투여, 국소 투여, 비 내 투여, 폐내 투여, 직장 내 투여 등이 될 수 있으나, 이에 제한되지는 않는다. 그러나, 경구 투여 시 펩타이드는 소화가 되기 때문에 경구용 조성물은 활성 약제를 코팅하거나 위에서의 분해로부터 보호되도록 제형화하는 것이 바람직하다. 바람직하게는, 주사제 형태로 투여될 수 있다. 또한, 본 발명에 따른 약학 조성물은 활성 물질이 표적 세포로 이동할 수 있는 임의의 장치에 의해 투여될 수 있다.
본 발명에 따른 펩타이드는 건강기능식품 조성물에 유효성분으로서 함유되는 것도 가능한 바, 본 발명에 따른 식품 조성물은 항암 기능이 있는 것으로 알려진 공지의 활성 성분과 함께 혼합하여 조성물 형태로 제조할 수 있으며, 식품학적으로 허용가능한 식품보조첨가제를 추가로 포함할 수 있다. 본 발명의 건강기능식품 조성물은 기능성 식품 (functional food), 영양 보조제 (nutritional supplement), 건강식품 (health food) 및 식품 첨가제(food additives) 등의 모든 형태의 식품에 대한 조성물을 포함한다.
상기 유형의 식품 조성물은 당 업계에 공지된 통상적인 방법에 따라 다양한 형태로 제조할 수 있다. 예를 들면, 건강식품으로는 정제, 환약, 분말, 캡슐, 껌, 비타민 복합체, 쥬스 및 드링크 등의 다양한 형태로 제조될 수 있으며, 본 발명에 따른 펩타이드를 유효성분으로 하는 식품 조성물을 과립화, 캡슐화 또는 분말화하여 섭취할 수 있다. 본 발명의 식품 조성물은 식품 제조 시에 통상적으로 첨가되는 성분을 포함할 수 있으며, 예를 들어, 단백질, 탄수화물, 지방, 영양소 및 조미제를 포함할 수 있다. 예컨대, 드링크제로 제조되는 경우에는 본 발명의 펩타이드 이외에 구연산, 액상과당, 설탕, 포도당, 초산, 사과산, 과즙, 대추 추출액, 감초 추출액 등을 추가로 포함시킬 수 있다. 또한, 식품보조첨가제로서, 당업계에 통상적인 식품첨가제, 예를 들어 향미제, 풍미제, 착색제, 충진제, 안정화제 등을 포함시킬 수 있다. 또한, 통상의 음료와 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수도 있다. 천연 탄수화물의 구체적인 예로는, 예를 들어, 포도당, 과당 등의 모노사카라이드, 말토스, 슈크로스 등의 디사카라이드, 덱스트린, 시클로덱스트린 등의 폴리사카라이드 등과 같은 통상적인 당 및 자일리톨, 소르비톨, 에리트리톨 등의 당알콜이다. 상술한 것 이외의 향미제로서 천연 향미제 (타우마틴, 스테비아 추출물 (예를 들어, 레바우디오시드 A, 글리시르히진 등)) 및 합성 향미제(사카린, 아스파르탐 등)를 포함시킬 수도 있다.

이하, 실시예를 통해서 본 발명을 더욱 상세하게 설명하기로 하되, 하기 실시예는 본 발명의 이해를 돕기 위한 것일 뿐, 본 발명의 범위를 제한하는 것은 아니다.
본 발명에서는 범용 항암제로서의 효과를 보인 전사인자 CP2c 표적 펩타이드 ACP52C의 생체내 안정성을 향상시킨 최종후보물질을 동정하기 위해, CP2c 표적 펩타이드 선도물질 ACP52C에 지방산을 연결시켜 생체내 안정성 및 항암 효력을 보이는지를 암세포이종이식 생쥐 모델에서 확인하였다. 또한 각 종 암세포 및 정상세포에서 최종항암후보물질의 암세포 특이적 항암 효력을 확인하였다.

전사인자 CP2c는 다양한 암에서 과발현되고 있다고 알려져 있으며, 미국 연구팀의 연구에 의하면 간암 세포주에서 CP2c의 발현을 억제하면 성장이 억제되며, CP2c를 과발현할 경우 암의 악성화와 전이가 일어나는 것으로 보고된 바 있다(Grant et al., Antiproliferative small-molecule inhibitors of transcription factor LSF reveal oncogene addiction to LSF in hepatocellular carcinoma.

본 발명자들은 전사인자 CP2c (Tfcp2, LSF, LBP1, UBP1 등으로도 알려짐)와 결합하는 펩타이드들을 파지 디스플레이(phage display) 방법으로 동정하였고 (Kang et al., Identification and characterization of four novel peptide motifs that recognize distinct regions of the transcription factor CP2.
FEBS 2005, 272:1265-1277), 이들 중 1종의 펩타이드(PEP #5, 서열번호 1)는 CP2c 전사인자 복합체 형성을 억제함으로써 DNA 결합을 방해함을 확인하였다. 특히 PEP #5의 카복실 말단 6개 아미노산으로 이루어진 PEP #5-2(서열번호 2)도 PEP #5와 동일한 효과를 나타냄을 확인하였고 이를 CP2c 표적 펩타이드로 선별하였다(도 1). PEP #5-2는 자체적으로 세포 내로 들어가지 못하므로, 세포 침투성을 향상시키기 위하여 세포막 투과 펩타이드(CPP: cell-penetrating peptide)로서 9개의 아미노산(9aa) 'CRGDKGPDC(Cys-Arg-Gly-Asp-Lys-Gly-Pro-Asp-Cys, 서열번호 3)'으로 이루어진 펩타이드인 iRGD(internalizing RGD)를 상기 CP2c 표적 펩타이드에 결합시킨 형태의 ACP52C 선도물질을 합성하였다 (도 7).
[실시예 1] ACP52C의 항암 효과 확인
상기 선별된 펩타이드는 CP2c에 결합하여 CP2c를 포함하는 전사인자 복합체(CP2c homotetramer 및 CP2c/CP2b/PIAS1 heterohexamer) 형성을 억제함으로써 간접적으로 CP2c의 DNA 결합을 방해한다. 상기 합성된 ACP52C를 대상으로 다양한 암종에서 항암 효력을 분석한 결과 암세포 특이적으로 성장억제 및 세포사 효능을 나타냄을 확인할 수 있었다(도 2). ACP52C는 MDA-MB-231과 HepG2 세포주에서 콜로니 형성능도 효율적으로 억제하며, CP2c를 과발현 시킬 경우 더욱 효율적으로 콜로니 형성능이 억제되는 것으로 보아 ACP52C의 효능은 CP2c 발현양 의존적임을 확인하였다(도 3). 본 결과는 대부분의 암세포주에서 CP2c의 발현은 정상세포에 비해 50배 이상 높게 나타나며, ACP52C가 암세포 특이적으로 작용하는 현상과 일맥상통한다.
ACP52C 처리에 따른 세포 주기 변화를 FACS로 분석한 결과 non tumorigenetic breast epithelial 세포주인 MCF10A에서는 효과를 보이지 않은 반면, MDA-MB-231에서는 G2/M기에서 세포주기 멈춤이 일어나며 subG1 세포가 증가함을 확인하였다. 또한, ACP52C에 의한 G2/M기 멈춤 현상은 p53, CHK1, CDC25c axis를 따라 Cdc2(CDK1) 및 CyclinB1의 발현감소에 기인함을 MCF7에서 검증하였다. 정확한 분석을 위해 double thymidine block 방법을 통해 G1/S기에 세포주기를 동기화한 다음 ACP52C를 처리하여 세포주기의 변화를 FACS로 분석한 결과, 시간경과에 따라 G2/M기 멈춤과 polyploid가 발생함을 확인하였다. 한편 nocodazole 처리에 따른 G2/M기에 동기화시킨 세포주에 ACP52C를 처리할 경우 시간 경과에 따라 G2/M기 멈춤과 polyploid 외에도 subG1 세포가 증가함을 확인하였다(도 4). 따라서 ACP52C처리에 의한 세포사 유도는 순차적으로 G2/M 세포주기 멈춤, polyploid 형성 및 SubG1 세포 형성을 통해 일어날 것으로 추정할 수 있었다.
대조군 및 ACP52 처리한 세포주에서 시간경과에 따른 apoptosis관련 단백질의 발현을 western blot으로 확인한 결과, ACP52C 처리에 따른 세포사 유도는 anti-apoptotic 표지 단백질의 발현 감소와 함께 pro-apoptotic 단백질의 발현 증가 및 caspase의 활성화로 이어지는 apoptosis에 의함을 확인하였다(도 5A, B). 전자현미경으로 ACP52C처리에 따른 세포의 변화를 분석한 결과, MCF10A에서는 큰 변화가 없는 반면, MDA-MB-231에서는 세포핵이 응축되는 등의 apoptosis의 특징을 관찰하였다(도 5C). ACP52C가 apoptosis를 통해 세포사를 유도한다는 사실은 AnnexinV/PI 염색법으로도 확인하였다(도 5D). 따라서 ACP52C는 apoptosis 과정을 통해 세포사 효능을 나타낼 것으로 추정할 수 있었다.
암세포 특이적으로 세포사를 유도하는 ACP52C의 안정성 및 안전성을 확인하기 위해 다음과 같은 방법을 사용하였다. 혈청(in vitro)과 혈액(in vivo) 내에서 ACP52C의 half life를 HPLC를 통해서 분석한 결과, in vitro에서는 24시간까지 전혀 분해되지 않았지만 in vivo에서는 2시간 만에 절반 수준으로 감소하였다(도 6A, 6B). ACP52C는 pH 7.4에서 8시간동안 사람 적혈구에 처리하여도 hemolysis를 일으키지 않았다(도 6C). 한편 Cy5 표지된 펩타이드(Cy5-ACP52C)를 Hep3B 세포주 이식생쥐에 tail vein으로 주입한 후 시간경과에 따라 live imaging을 실시한 결과 5일 뒤에도 신장과 간 및 tumor에서 펩타이드가 관찰되었지만, 주입한 대부분의 ACP52는 24시간 이내에 사라지는 것으로 보아 단백질분해효소에 의한 분해보다는 renal secretion되는 것으로 추정되며, 체내에 존재하는 ACP52C의 반감기는 약 7.95시간으로 측정되었다(도 6D). 실제 ACP52C는 15개 아미노산으로 구성되어 있으므로 신장에서 renal secretion이 용이할 것으로 판단하였다.
[제조예 2] CP2c 표적 펩타이드-포화 지방산 컨쥬게이트의 합성
상기 실시예 1의 결과를 종합해 보면, ACP52C는 정상세포에는 거의 영향을 나타내지 않지만 암세포 특이적으로 세포사를 일으킴을 확인하였다. 또한 세포사를 유도하는 작용기전도 확인하였다. 그러나 항암제 후보물질이 되려면 생체내 안정성이 확보되어야 할 것이므로 이를 극복하기 위한 방안으로 알부민 친화성 지방산 결합 펩타이드를 활용하기로 하였다. 알부민은 혈액 내에 다량 존재하는 안정한 단백질로서 지방산과 잘 결합하므로 약물을 지방산과 결합시켜놓을 경우 알부민과 결합함으로서 약물의 renal secretion을 저해할 수 있다(Sleep et al., Albumin as a versatile platform for durug half-life extension.

ChemMedChem 2016, 11: 2474-2495). 또한 당뇨병 치료제로 임상에 활용되고 있는 liraglutide도 혈액내 안정성을 확보하기 위하여 C16 팔미토일산을 펩타이드에 결합시킨 바 있다. 이러한 사실에 근거하여 C16 팔미토일산을 ACP52C의 N-말단에 결합시킨 펩타이드 C-16-ACP52C(ACP52CG)를 합성하였다(도 8A). 또한, 지방산에 결합된 펩타이드가 암 세포로 들어갈 경우 지방산에 의해 핵으로의 이동 혹은 CP2c와의 결합이 저해될 수도 있을 것이므로, 암 조직 주위에 전달된 지방산 결합 펩타이드에서 지방산을 제거할 필요가 있으므로 암 조직 주위에 cathepsin B peptidase 활성이 높다는 사실에 근거하여 암조직에서 지방산이 ACP52C로부터 용이하게 제거할 목적으로 cathepsin B 작용 서열 GFLG를 지방산과 ACP52C 사이에 결합시킨 펩타이드 C16-GFLG-ACP52Cn(ACP52CK)도 함께 합성하였다(도 8B).
[실시예 2] CP2c 표적 펩타이드-지방산 컨쥬게이트의 in vitro 항암 효과 확인
ACP52CG 및 ACP52CK가 ACP52C와 유사하게 암세포 특이적 세포사를 유도하는지를 확인하기 위하여 다양한 암세포주를 대상으로 MTT [(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] assay를 수행한 결과 2가지 지방산 결합 펩타이드는 ACP52C와 유사한 GI
50 값(50% 성장억제 농도)을 나타내었다(도 9). 이의 예시로서, 유방암 세포주(MDA-MB-231), 정상 hMSC(human mesenchymal stem cells, LT18), non-tumorigenic 폐 세포주(BEAS-2B)에 각 약물을 농도 별로 처리한 후 48시간 및 96시간에 측정한 세포 수, GI50값을 구하는 그래프 및 대표적인 세포사진을 도 10에 나타내었다. MDA-MB-231를 포함한 여러 암세포나 BEAS-2B에서 ACP52CG와 ACP52CK 모두 양성 대조군인 ACP52C와 유사한 효능을 나타낸 반면, hMSC 세포주(LT18)에서는 ACP52C보다 오히려 ACP52CG와 ACP52CK의 세포 독성이 더 적음을 확인하였다. 한편 ACP52CK를 catehpsin B와 함께 처리하더라도 더 우수한 효과를 나타내지 않는 것으로 보아 세포내로 들어간 지방산 결합 ACP52C에서 지방산은 ACP5-2의 항암활성에 간섭하지 않음을 확인하였다.
[실시예 3] 다양한 암 세포주 이식 생쥐모델에서 CP2c 표적 펩타이드-지방산 컨쥬게이트 처리에 따른 종양 성장 억제 효과 및 일반 생리 독성 확인
In vivo에서 ACP52CG 및 ACP52CK의 항암효능을 확인하기 위하여 Hep3B xenograft 생쥐 모델을 대상으로 음성 대조군(saline; mock), 양성 대조군(sorafenib, 10 mg/kg 및 ACP52C, 2 mg/kg), 그리고 실험군(ACP52CK, 5 mg/kg 및 ACP52CG, 5 mg/kg)으로 나누어 각각 3일 간격으로 총 5회 꼬리 정맥을 통해 주입하였다. ACP52CG와 ACP52CK 모두 종양크기 및 무게에서 음성 대조군인 mock 그룹에 비해서는 효능을 나타냈지만 sorafenib과 유사한 수준이고, 양성 대조군인 ACP52C에 비해 유의미하게 항암 효과가 저해되었다(도 11).
ACP52CG와 ACP52CK의 항암효과가 ACP52C보다 좋지 않은 이유로 다음과 같은 가설을 수립하였다. 포화지방산인 팔미토일산이 서로 응집하여 미셀(micelle) 구조를 형성하면 알부민과 결합을 못할 뿐만 아니라 세포투과성 펩타이드인 iRGD가 미셀 내부로 들어가게 되어 암세포로의 전달도 억제될 것으로 추측하였다. 이와 같은 문제점을 극복하기 위하여 미셀을 형성할 수 없는 불포화 지방산인 오메가 3을 이용하였다. 탄소사슬의 끝에서 세 번째 탄소에서 이중 결합이 있는 불포화 지방산 오메가 3은 자연에서 쉽게 발견되는 물질로 여러 식자재에 다량 함유되어 있고 건강한 삶을 위해 권장되는 식품중 하나이며 DHA(Docosahexaenoic acid), EPA(Elcosapentaenoic acid), ALA(Alpha-linolenic acid) 등이 여기에 해당된다(도 12A). 불포화 지방산은 여러 가지 경로를 통해 쉽게 세포막을 통과하는 성질이 밝혀져 있고, 세포 내에 들어가서 다양한 신호전달계를 조절한다고 알려졌 있다(Seitz and Ojima, Drug conjugates with polyunsaturated fatty acids.

J Cardiol 2016, 67: 22-27)(도 12B). 특히, DHA는 생화학 전구물질 및 에너지 원천으로 사용될 수 있어 인체에 무해하고, 세포막의 인지질에 손쉽게 결합하여 통과하거나 세포막 구조 단백질 및 유체로부터 신호 전달계를 조절한다는 보고가 있다(Sauer et al., Uptake of plasma lipids by tissue-isolated hepatomas 7288CTC and 7777 in vivo.

[제조예 3] CP2c 표적 펩타이드-DHA 컨쥬게이트의 합성
우선 DHA 자체의 효과를 확인하기 위해서 유방암 세포주(MDA-MB-231), 간암 세포주(Hep3B, HepG2), 정상 hMSC 세포주(LA211) 및 non-tumorigenic 폐 세포주(BEAS-2B)에 DHA를 농도 별로 처리한 뒤 세포 성장을 MTT assay로 분석한 결과, 정상 세포주에는 전혀 영향이 없었으나, 암세포들과 BEAS-2B에서는 20 μM 이상의 농도에서 세포 독성을 나타내었다(도 13). 그러나 암세포 특이적 세포사를 유도하는 ACP52C의 유효농도는 10 μM 이하이므로, DHA를 ACP52C에 결합하여 사용할 경우 DHA 자체가 항암효과를 나타내는지를 확인하기 위하여 10 μM 이하의 DHA를 처리하여 Hep3B와 MDA-MB-231 세포주에서 세포사 유도 여부를 분석한 결과, 전혀 항암효과를 나타내지 않았다(도 14). 한편, 2 μM의 ACP52C를 1~3 μM의 DHA와 병용하여 정상 및 암세포주에 처리하여 ACP52C의 세포사에 미치는 영향을 분석한 결과, 정상 hMSC 세포주(LA211)와 non-tumorigenic 폐 세포주(BEAS-2B)에서는 전혀 영향이 없었지만 유방암 세포주(MDA-MB-231)에서는 DHA와 ACP52C를 조합한 혼합물의 경우 오히려 ACP52C를 단독으로 처리한 것보다 효능이 떨어지는 양상을 보였다(도 15). 따라서 DHA는 고농도에서는 항암효과를 나타낼 수 있으나, ACP52C의 안정성 확보를 위한 농도에서는 DHA 자체의 항암효과는 없을 것으로 추정하였다.
이에 따라, DHA를 직접 ACP52에 접합한 2종의 펩타이드를 합성하였다. DHA는 자체적으로 세포막 투과성을 지니므로 ACP52에 DHA만을 접합한 DHA-ACP52 및 DHA 및 세포투과성 펩타이드 iRGD를 모두 결합한 DHA-ACP52C를 합성하였다(도 16).
[실시예 4] CP2c 표적 펩타이드-DHA 컨쥬게이트의 in vitro 항암 효과 확인
정상세포주 및 암세포주에서 DHA 결합 펩타이드를 처리 한 뒤 세포 성장을 MTT assay로 확인한 결과, 정상 세포주와 non-tumorigenic BEAS-2B 세포주에서는 DHA-ACP52와 DHA-ACP52C 모두 전혀 독성을 나타내지 않았다. 유방암(MDA-MB-231) 및 간암(HepG2) 세포주에서는 DHA-ACP52의 GI
50는 5.7~6.95 μM 수준으로 ACP52C의 2 μM보다 오히려 효능이 감소한 반면, DHA-ACP52C의 GI
50는 1.43~2.4 μM 수준으로 유사한 효능을 보였다(도 17). 또한 DHA-ACP52C 처리에 따른 세포 내 apoptosis 및 세포 주기 멈춤 관련 인자들의 단백질 발현은 ACP52C 처리에 따른 변화와 유사한 양상을 나타내었다(도 18, 도 19). 따라서 DHA 자체가 세포막을 통해 세포 내로 들어가는 것으로 알려져 있으나 DHA-ACP52의 항암효과가 나타나지 않는 것으로 보아 DHA는 iRGD 대비 세포 투과성은 미미할 것으로 추정하였다.
[실시예 5] 다양한 암 세포주 이식 생쥐모델에서 CP2c 표적 펩타이드-DHA 컨쥬게이트 처리에 따른 종양 성장 억제 효과 및 일반 생리 독성 확인
세포주 수준에서 검증된 CP2c 표적 펩타이드-DHA 컨쥬게이트의 효능을 Hep3B xenograft 생쥐 모델을 대상으로 in vivo에서 항암 효과를 확인하였다. 암이 형성된 생쥐에 음성 대조군으로 saline(mock), 양성 대조군으로 sorafenib(10 mg/kg), 실험군으로 DHA-ACP52(1.65 mg/kg) 및 DHA-ACP52C(2.79 mg/kg)을 각각 3일 간격으로 총 6회 꼬리 정맥을 통해 주입하였다(도 20A). 실험 기간 동안 3일에 한 번씩 생쥐의 종양 크기를 측정한 결과 sorafenib 혹은 DHA-ACP52C를 처리한 군들이 mock 혹은 DHA-ACP52를 처리한 군보다 암의 성장을 효과적으로 저해함을 확인하였다(도 20B). 실험 종료 후 생쥐들을 희생시켜 암을 적출하고 실제 암의 크기 및 모양, 무게를 분석한 결과, sorafenib은 mock에 비해 유의미하게 항암 효능을 보여준 반면, DHA-ACP52의 경우에는 일정한 효과를 나타내지 못하였으며 DHA-ACP52C의 경우에는 sorafenib보다 우수한 항암 효능을 보였다(도 20C, D). 한편 DHA-ACP52C 처리 생쥐는 무게, 혈액학적 분석 및 조직학적 분석에서 대조군을 포함하는 서로 다른 처리군과 유사한 것으로 보아 DHA-ACP52C는 생체 내 정상세포에는 부작용을 나타내지 않을 것으로 추정할 수 있었다(도 21). 한편, 암이 형성된 생쥐에 음성 대조군saline(mock), 양성대조군 ACP52C(2 mg/kg), 실험군 DHA-ACP52(1.65 mg/kg) 및 DHA-ACP52C(2.79 mg/kg)을 각각 5일 간격으로 총 6회 꼬리 정맥을 통해 주입하여 항암효과를 분석하였다(도 22A). 실험 기간 동안 3일에 한 번씩 생쥐의 종양 크기를 측정한 결과 DHA-ACP52C는 ACP52C보다 우수하게 작용하였지만, DHA-ACP52는 음성대조군과 그 결과가 유사하였다(도 22B). 실험 종료 후 생쥐들을 희생시켜 암을 적출하여 실제 암의 크기 및 모양, 무게를 분석한 결과, DHA-ACP52C만이 유의미한 항암 효능을 나타내었다(도 22C, D). 한편 DHA-ACP52C 처리 생쥐는 무게, 혈액학적 분석 및 조직학적 분석에서 대조군을 포함하는 서로 다른 처리군과 유사한 것으로 보아 DHA-ACP52C는 생체 내 정상세포에는 부작용을 나타내지 않을 것으로 추정할 수 있었다(도 23).
상기의 결과를 종합하면 DHA-ACP52C는 세포주 수준에서 종래의 ACP52C와 유사한 암세포 성장 억제 효과를 나타낸 반면, Hep3B xenograft 생쥐 모델에서는 ACP52C에 비해 우수한 항암 효과를 나타내었다. 특히, ACP52C는 3일 간격으로 주입할 경우 유의미한 항암효과를 보였으나 5일 간격 주입 시에는 유의미한 항암 효과를 보이지 않은 반면(도 11, 도 20), DHA-ACP52C는 3일 및 5일 간격으로 주입한 경우 모두에서 뚜렷한 항암 효과를 나타내었다(도 20, 도 22). 따라서 DHA-ACP52C에서 DHA는 ACP52C의 안정성을 향상시키지만 정상 세포 및 정상 조직에서는 부작용을 나타내지 않는 것으로 확인되었다.
