橙皮苷,为二氢黄酮苷类化合物,广泛分布于很多植物中,具有维持渗透压,增强毛细血管韧性,缩短出血时间,降低胆固醇等作用,常用于心血管系统疾病的辅助治疗。大蓟苷又名柳穿鱼叶苷,来源于菊科植物蓟的干燥地上部分或根,分子式为C28H34O15,可用于治疗衄血、吐血、尿血、便血、崩漏下血、外伤出血等;绿原酸是由咖啡酸(Caffeic acid)与奎尼酸(Quinicacid,1-羟基六氢没食子酸)生成的缩酚酸,是植物体在有氧呼吸过程中经莽草酸途径产生的一种苯丙素类化合物,具有抗菌、抗病毒、增高白血球、保肝利胆、抗肿瘤、降血压、降血脂、清除自由基和兴奋中枢神经系统等作用。
目前,将橙皮苷、大蓟苷、绿原酸组合用于制备具有美白、祛痘、去黑头、抗衰老的护肤品组合物未见有报道。

具有美白、祛痘、去黑头、抗衰老功能的护肤品,由以下重量百分比的原料制成:含橙皮苷、大蓟苷、绿原酸的组合物3%,由重量比为5:1的聚氧乙烯氢化蓖麻油、夏威夷果油混合而成的乳化剂12%,丙二醇15%,黄原胶0.4%,余量为去离子水。其中,含橙皮苷、大蓟苷、绿原酸的组合物中橙皮苷、大蓟苷、绿原酸的重量比为1:0.6:0.2。
该护肤品的制备方法包括以下步骤:
S1.将含橙皮苷、大蓟苷、绿原酸的组合物和由重量比为5:1的聚氧乙烯氢化蓖麻油、夏威夷果油混合而成的乳化剂混合,加热至60℃后搅拌至混合均匀得到A相;
S2.将丙二醇、去离子水混合,加热至60℃后搅拌至混合均匀得到B相;
S3.将A相、B相、黄原胶混合,加热至45℃后搅拌10分钟,然后45℃下均质15分钟出料,得到具有美白、祛痘、去黑头、抗衰老功能的护肤品。
实施例5
具有美白、祛痘、去黑头、抗衰老功能的护肤品,由以下重量百分比的原料制成:含橙皮苷、大蓟苷、绿原酸的组合物4%,由重量比为5:1的聚氧乙烯氢化蓖麻油、夏威夷果油混合而成的乳化剂10%,丙二醇16%,黄原胶0.3%,余量为去离子水。其中,含橙皮苷、大蓟苷、绿原酸的组合物中橙皮苷、大蓟苷、绿原酸的重量比为1:0.6:0.2。
本实施例的制备方法与实施例4相同。
实施例6
具有美白、祛痘、去黑头、抗衰老功能的护肤品,由以下重量百分比的原料制成:含橙皮苷、大蓟苷、绿原酸的组合物2%,由重量比为5:1的聚氧乙烯氢化蓖麻油、夏威夷果油混合而成的乳化剂15%,丙二醇12%,黄原胶0.5%,余量为去离子水。其中,含橙皮苷、大蓟苷、绿原酸的组合物中橙皮苷、大蓟苷、绿原酸的重量比为1:0.6:0.2。
本实施例的制备方法与实施例4相同。
试验例一、抗痤疮、祛痘、去黑头试验:
1.痤疮模型的建立
1.1痤疮丙酸杆菌的培养
将菌种直接接种于生孢梭菌培养基,37℃培养24-48h后观察。使用生理盐水调整痤疮丙酸杆菌浓度为6×10 8/ml。
1.2痤疮模型的建立
取家兔,采用kligman法于每只家兔左耳内侧面耳导管开口处约2cm*2cm范围每日涂抹煤焦油溶液1次,每次0.5-1ml,连续14d,第4d兔耳皮内注射痤疮丙酸杆菌200μl,隔日注射,右耳不做处理,2周后造模成功。
1.3实验分组
将造模成功的35只家兔(雄)随机分为5组:模型组、阳性药物组(盐酸克林霉素乳膏)、组合物1组(实施例1)、组合物2组(实施例2)、组合物3组(实施例3)。另设正常组7只不作处理。
2.给药方案
模型制备成功后,阳性药物组每日给予克林霉素乳膏0.5g,组合物1组、组合物2组、组合物3组每日给予0.5g,每日于相同时间给药,连续2周。
3.检测指标
给药期间,每日观察各组痤疮模型部位表皮组织结构变化,在给药第4周后,在每只兔的左耳涂药处以及右耳皮肤组织活检,制作石蜡切片以备用,在光镜下观察组织学改变。取涂药处皮肤组织制备组织匀浆,检测痤疮相关指标(IL-1α、TNF-a、二氢睾酮)。
4.检测指标
4.1病理组织学切片:
在每只兔的左耳涂药处以及右耳皮肤组织活检,制作石蜡切片以备用,在光镜下观察组织学改变,组织切片在显微镜下观察,显微镜观察结果如表1所示。
病理组织学分级:
①角化程度:与正常组织相同,无痘、无黑头(-);角化物质轻度增厚(+);中度增厚(2+);显著增厚(3+);
②表皮增厚程度:与正常组织相同(-);细胞层数4-5层(+);6-7层(2+);8-9层(3+);
③毛囊内角化物质多少:与正常组织相同(-);角化物质增多,但不致密,且未充满毛囊(+);角化物质致密,但未充满毛囊,或角化物质不致密,但充满毛囊(2+);角化物质致密,充满毛囊(3+);
④毛囊扩张程度:与正常组织相同(-);轻度扩张(+);中度扩张(2+);高度扩张(3+);
⑤炎细胞浸润程度:与正常组织相同(-);局灶性炎细胞浸润,细胞散在(+);局灶性炎细胞浸润,细胞较密集,或出现多个浸润灶(2+);弥漫性炎细胞浸润(3+)。正常组织学表现:表皮组织2-4层,毛囊上皮轻度增厚,真皮层偶见单核炎细胞浸润;
⑥角质栓塞:与正常组织相同,无栓塞(-);角质轻度栓塞(+);中度栓塞(2+);显著栓塞(3+),与人的黑头、青春痘相似。
5.试验结果如表1-3所示:

由表1可知,涂抹本发明实施例1-3后,兔表皮组织、毛囊皮脂腺等显著改善,其中实施例3各项指标均接近正常组,说明本发明具有非常显著的改善痤疮和祛痘、去黑头的作用。

由表1-3可知,本发明所述组合物能显著改善兔痤疮、青春痘和黑头粉刺现象,降低雄性激素二氢睾酮的作用,抑制炎症因子IL-1α、TNF-a的表达,与模型组相比,具有显著性差异。
试验例二、美白试验
1.实验动物
SPF级雄性黄棕色豚鼠,体质量250-270g,许可证号:SCXK(粤)2008-0002,由广州中医药大学实验动物中心提供。
2.实验方法
采用黄棕色豚鼠40只,分成4个组,分别为对照组、组合物1组(实施例1)、组合物2组(实施例2)、组合物3组(实施例3),每组10只。
各组动物背部两侧剃毛成若干1cm×2cm大小的去毛区,给药组动物用棉棒在左、右侧皮肤将给药涂一圈,对照组和模型组动物皮肤涂抹等量的生理盐水,每日2次。28天后,取豚鼠皮取皮肤活组织,用10%福尔马林固定,石蜡包埋、切片,进行观察。组织学观察:硝酸银染色,光镜下观察每份标本的组织学改变,对基底细胞中含黑色素颗粒细胞计数,得出每组10只豚鼠的均值,并进行组间比较。
依据含黑素颗粒细胞的数量将实验组动物的皮肤黑色素含量分为4级(+至++++),分别如下:+少于20个含黑色素颗粒细胞/100个基底细胞;++21-30个含黑色素颗粒细胞/100个基底细胞;+++31-40个含黑色素颗粒细胞/100个基底细胞;++++大于40个含黑色素颗粒细胞/100个基底细胞。
制备豚鼠血液,3000r/min离心20min,取上清,Elisa法测定各组动物血清酪氨酸酶的含量。
3.试验结果如表4-5所示:

由表4-5可知,与空白对照组相比,本发明实施例1-3动物含黑色素颗粒阳性细胞数和皮肤酪氨酸酶含量显著下降,提示本发明所述组合物具有显著的美白效果。
试验例三、抗皮肤氧化衰老实验
1.实验动物
SPF级NIH小鼠,体质量18-22g,雌雄各半,许可证号:SCXK(粤)2008-0002,由广州中医药大学实验动物中心提供。
2.实验材料
超氧化物歧化酶(SOD)、总抗氧化能力(T-AOC)测定试剂,盒,南京建成生物工程研究所;D-半乳糖生化试剂,国药集团化学试剂有限公司。
3.实验方法
昆明种小白鼠50只,分别为对照组、模型组、组合物1组(实施例1)、组合物2组(实施例2)、组合物3组(实施例3),每组10只。对照组每天颈背部注射生理盐水0.3mL,其余6组均给予1g/kg半乳糖0.3mL颈背部皮下注射,连续给药28d建立亚急性衰老模型,同时背部皮肤涂药。
各组皮肤给药剂量如下:对照组:涂抹等量的生理盐水;模型组:涂抹等量的生理盐水;给药组给以相应的药物。6周后测定小鼠背部皮肤组织匀浆中以下指标:超氧化物歧化酶(SOD)活性、氧化性指标丙二醛(MDA)含量。
4.SOD与MDA酶的检测
在试验末次给药1h后,颈椎脱臼处死小鼠,取背部已去毛皮肤各一块。用预冷生理盐水(4度)冲洗,除净血液等杂质,滤纸拭干,切取皮肤、肝脏各约0.5g,手术剪剪碎,用组织研磨器匀浆,以1:9(W/V)加入冰生理盐水,制成10%组织匀浆。匀浆液以3500r·min -1离心15min,取上清液在紫外分光光度计上测定SOD、MDA的OD值,计算其活性或含量。
5.试验结果如表6所示:

由表6可知,与模型组相比,本发明实施例1-3可以显著极显著的提高皮肤SOD活性,降低MDA含量,说明本发明所述组合物可以提高SOD活性、降低MDA含量,具有显著的抗衰老氧化作用。
试验例四、耐寒稳定性试验
取实施例4-6所述样品,并设置对照组1(与实施例4不同的是乳化剂为聚氧乙烯氢化蓖麻油)、对照组2(与实施例4不同的是乳化剂为聚乙二醇),将5组样品在-2℃的恒温箱中放置60天,取出恢复室温后进行观察,结果表明本发明实施例4-6所述样品未出现凝结、混浊现象,具有优异的耐寒稳定性,对照组1出现了轻微的凝结、混浊现象,对照组2凝结、混浊现象的程度比对照组1严重,说明本发明使用的由聚氧乙烯氢化蓖麻油、夏威夷果油复配组成的乳化剂能有效提高护肤品的耐寒稳定性。
上述实施例仅例示性说明本发明的原理及其性能,而非用于限制本发明。任何熟悉此技术的人士皆可在不违背本发明的精神及范畴下,对上述实施例进行修饰或改变。因此,举凡所属技术领域中具有通常知识者在未脱离本发明所揭示的精神与技术思想下所完成的一切等效修饰或改变,仍应由本发明的权利要求所涵盖。

Claims (10)

  1. 一种美白、祛痘、去黑头、抗衰老的组合物,其特征在于,所述组合物由重量比为1:0.6:(0.1-0.3)的橙皮苷、大蓟苷、绿原酸组成。
  2. 根据权利要求1所述的一种美白、祛痘、去黑头、抗衰老的组合物,其特征在于,所述组合物由重量比为1:0.6:0.2的橙皮苷、大蓟苷、绿原酸组成。
  3. 根据权利要求1或2所述的组合物在制备具有美白、祛痘、去黑头、抗衰老功能的护肤品中的用途。
  4. 根据权利要求3所述的用途,其特征在于,所述护肤品被制成膏剂、乳剂、膜剂、溶液剂、凝胶剂或皂剂。
  5. 根据权利要求3所述的用途,其特征在于,所述组合物中橙皮苷、大蓟苷、绿原酸的重量比为1:0.6:0.2。
  6. 一种具有美白、祛痘、去黑头、抗衰老功能的膏剂,其特征在于,由以下重量百分比的原料制成:含橙皮苷、大蓟苷、绿原酸的组合物2-4%,乳化剂10-15%,保湿剂12-18%,增稠剂0.2-0.5%,余量为去离子水。
  7. 根据权利要求5所述的一种具有美白、祛痘、去黑头、抗衰老功能的膏剂,其特征在于,所述含橙皮苷、大蓟苷、绿原酸的组合物中橙皮苷、大蓟苷、绿原酸的重量比为1:0.6:(0.1-0.3)。
  8. 根据权利要求6所述的一种具有美白、祛痘、去黑头、抗衰老功能的膏剂,其特征在于,所述含橙皮苷、大蓟苷、绿原酸的组合物中橙皮苷、大蓟苷、绿原酸的重量比为1:0.6:0.2。
  9. 根据权利要求5所述的一种具有美白、祛痘、去黑头、抗衰老功能的膏剂,其特征在于,所述乳化剂由重量比为5:1的聚氧乙烯氢化蓖麻油、夏威夷果油混合而成,所述保湿剂为丙二醇,增稠剂为黄原胶。
  10. 根据权利要求6至9任意一项所述具有美白、祛痘、去黑头、抗衰老功能的膏剂的制备方法,其特征在于,包括以下步骤:
    S1.将含橙皮苷、大蓟苷、绿原酸的组合物和乳化剂混合,加热至60℃后搅拌至混合均匀得到A相;
    S2.将保湿剂、去离子水混合,加热至60℃后搅拌至混合均匀得到B相;
    S3.将A相、B相、增稠剂混合,加热至45℃后搅拌10分钟,然后45℃下均质15分钟出料,得到具有美白、祛痘、去黑头、抗衰老功能的膏剂。
