- 액상 검체를 수용하는 검체패드;상기 검체패드의 일측에 연결되며 표지물질 및 표적물질 유사체를 수용하는 접합체패드;상기 접합체패드의 일측에 연결되며 검사선, 제1 대조선 및 제2 대조선을 포함하는 검출패드; 및상기 검출패드의 일측에 연결되며 반응 후 잔존하는 액상을 수용하는 흡습패드; 를 포함하고,상기 검사선은 제1 리간드를 포함하며, 상기 제1 리간드는 표적물질 및 제1 접합체와 반응할 수 있고,상기 제1 대조선은 제2 접합체와 반응하는 제2-1 리간드를 포함하며, 상기 제2 대조선은 제2 접합체와 반응하는 제2-2 리간드를 포함하되, 상기 제2-1 리간드와 상기 제2-2 리간드의 농도는 서로 다른 것을 특징으로 하는, 면역 크로마토그래피용 스트립.
- 제1 항에 있어서, 검체 내 표적물질의 농도 측정범위(dynamic range)는 5 mg/ml 내지 140 mg/ml 인 면역 크로마토그래피용 스트립.
- 제1 항에 있어서,상기 제1 접합체는 상기 표적물질의 유사체와 표지물질의 결합으로 이루어지고,상기 제1 접합체와 상기 검체 내 표적물질은 경쟁적으로 검사선의 제1 리간드와 반응하며, 상기 제1 접합체가 상기 검사선의 제1 리간드와 반응하는 경우 제1 복합체를 형성하는 것을 특징으로 하는 면역 크로마토그래피용 스트립.
- 제3 항에 있어서,상기 제1 복합체는 검사선에 포획되어 소정의 신호를 나타내고,상기 신호를 이용하여 상기 검체 내 표적물질의 양을 측정하는 면역 크로마토그래피용 스트립.
- 제4 항에 있어서, 상기 검체 내 표적물질의 농도가 증가할수록 상기 검사선의 신호 세기는 감소하는 면역 크로마토그래피용 스트립.
- 제1 항에 있어서, 상기 제1 리간드, 상기 제2-1 리간드 및 상기 제2-2 리간드는 단백질, 항원, 항체, DNA, RNA, PNA 및 압타머로 이루어진 군에서 선택된 어느 하나로 이루어지며,상기 제1 리간드는 상기 제2-1 리간드 및 상기 제2-2 리간드와 상이한 리간드로 이루어지는 면역 크로마토그래피용 스트립.
- 제1 항에 있어서, 상기 표지물질은 금콜로이드 (colloidal gold), 라텍스 입자, 유색 폴리스티렌 미세입자, 효소, 형광성 염료, 전도성 고분자, 발광 물질 및 자성입자로 이루어진 군에서 선택된 적어도 어느 하나를 포함하는 면역 크로마토그래피용 스트립.
- 제1 항 내지 제7 항 중 어느 한 항의 면역 크로마토그래피용 스트립이 케이스 내에 고정되는 키트로서,상기 케이스는 상부 케이스 및 하부 케이스를 포함하며,상기 하부 케이스는 스트립 지지부을 포함하고,상기 상부 케이스는 검체 투입구; 및 검사선, 제1 대조선 및 제2 대조선과 상응하는 위치에 결과 확인창; 을 포함하는 진단 키트.
- 제8 항에 있어서, 상기 지지부는니트로셀룰로스, 나일론, 피브이디에프 (PVDF), 유리 및 플라스틱으로 이루어진 군에서 선택된 적어도 어느 하나로 이루어진 진단 키트.
- 검체 투입구에 검체를 포함하는 소정의 액체를 주입하는 단계;검체패드에서 상기 액체를 수용하여 이동상을 형성하는 단계;상기 이동상이 접합체 패드로 전개되며 제1 접합체를 포함하는 단계;상기 제1 접합체를 포함하는 이동상이 검사선으로 전개되면서 상기 제1 접합체 및 상기 검체 내 표적물질이 경쟁적으로 제1 리간드와 결합하는 단계;상기 제1 접합체가 상기 제1 리간드와 결합하여 제1 복합체를 형성하고, 형성된 제1 복합체가 표지물질에 따른 제1 신호를 나타내는 단계;상기 이동상이 제2-1 리간드가 고정된 제1 대조선 및 제2-2 리간드가 고정된 제2 대조선으로 전개되는 단계;농도가 서로 다른 상기 제2-1 리간드 및 제2-2 리간드에 제2 접합체가 결합되어 제1 대조선 및 제2 대조선에서 제2 신호를 나타내는 단계; 및상기 제1 신호 및 제2 신호에 따라 상기 검체 내 표적물질의 양을 분석하는 단계; 를 포함하는, 면역 크로마토그래피 분석 방법.
