Description
バイオサーファクタント(より具体的には、モノアシル型MEL)の生産菌およびそれを用いたモノアシル型MELの製造に関する技術が開示される。
バイオサーファクタントは微生物が生産する天然の界面活性剤であり、生分解性が高く、環境低負荷であり、種々の有益な生理機能を有する。よって、食品工業、化粧品工業、医薬品工業、化学工業、環境工業分野等で使用すれば、環境調和型の社会を実現する上で有意義である。
バイオサーファクタントは、糖脂質系、アシルペプタイド系、リン脂質系、脂肪酸系及び高分子系の5つに分類される。これらの中でも、糖脂質系の界面活性剤が最もよく研究されている。このような糖脂質系のバイオサーファクタントとしては、マンノースにエリスリトールがグリコシド結合したマンノシルエリスリトール(以下、MEとも称す。)に、更に脂肪酸がエステル結合したマンノシルエリスリトールリピッド(以下、MELとも称す。)、並びに、ラムノリピッド、ユスチラジン酸、トレハロースリピッド、及びソホロースリピッド等が知られている。
MELには結合する脂肪酸残基並びにアセチル基の位置及び数等が相違する種々の構造が存在する。図1に、水素原子、アセチル基、及び炭素数3~18の脂肪酸残基をR1~R5で示したMELの構造式を示す。R1及びR2が脂肪酸残基であり、かつR3及びR4がアセチル基である構造物はMEL-A、R3が水素原子でありR4がアセチル基である構造物はMEL-B、R3がアセチル基でR4が水素原子である構造物はMEL-C、R3及びR4が水素原子である構造物はMEL-Dと定義される。マンノースと結合するエリスリトールのヒドロキシメチル基が1位の炭素に由来するか、4位の炭素に由来するかによって、得られるMEの構造は図2(a)、(b)に示すように相違する。図2(a)に示される4-O-β-D-mannopyranosyl-erythritolを糖骨格とするMELを4-O-β-D-MELと称する。シュードザイマ・ツクバエンシス(Pseudozyma tsukubaensis)は、図2(b)に示される1-O-β-D-mannopyranosyl-erythritolを糖骨格とする1-O-β-D-MEL-Bを生産することが知られている。1-O-β-MEL-Bは、4-O-β-MEL-Bと比べて水和性が向上し、ベシクル形成能も高いという特徴を持ち、スキンケア剤などとして有望なバイオ素材である。
炭素源をグルコースのみとして、MEL生産菌を培養することで、図1のMELにおいてR2のみに脂肪酸が結合し、R1、R3及びR4は水素原子であるモノアシル型MEL(1本鎖型MEL)が生産されることが報告されている(非特許文献1)。このモノアシル型MELは、従来のジアシル型MELと比較して、親水性が向上している(非特許文献1)。
MEL生合成経路は既に報告されており、マンノースとエリスリトールを結合する糖転移酵素、脂肪酸を結合するアシル基転移酵素、アセチル基を結合するアセチル基転移酵素の反応によって、MELが細胞内で合成される(非特許文献2)。
本発明者らは、バイオサーファクタントを生産する能力を有する微生物のアシル基転移酵素の遺伝子を欠失させることにより、図1の構造式においてR1だけに脂肪族アシル基が結合したモノアシル型MELが得られることを見出した(特許文献1)。

上記のような現状の下、新たなMELを提供することがひとつの課題である。
斯かる課題を解決すべく鋭意研究を重ねた結果、バイオサーファクタントを生産する能力を有する微生物のトランスポーターの遺伝子を欠失させることにより、図1の構造式においてR1だけに脂肪族アシル基を有し、R4にアセチル基または水酸基が結合したモノアシル型MELが得られることを見出した。これらの知見に基づき、更なる研究と検討を重ねた結果、下記に代表される発明が提供される。

モノアシル型MEL-Bを生産する手段が提供される。
微生物は、モノアシル型MEL-Bを産生することが好ましい。モノアシル型MEL-Bは、上述のとおり、図1に示す構造式において、R1は炭素数2~24の脂肪族アシル基であり、R2は水素原子であり、R3は水素原子であり、R4はアセチル基である。脂肪族アシル基の炭素数は、好ましくは2~24、より好ましくは6~14である。
モノアシル型MEL-Bを生産する微生物の種類は特に制限されない。一実施形態においてモノアシル型MEL-B生産微生物は、シュードザイマ属、モイジオマイセス属、ウスチラゴ属、スポリソリウムに属、Melanopsichium属、又はクルツマノマイセス属に属することが好ましい。好ましいシュードザイマ属微生物は、Pseudozyma antarctica(Moesziomyces antarcticus)、Pseudozyma parantarctica、Pseudozyma rugulosa、 Pseudozyma siamensis、Pseudozyma shanxiensis、Pseudozyma crassa、 Pseudozyma churashimaensis、Pseudozyma aphidis(Moesziomyces aphidis)、Pseudozyma hubeiensis、及びPseudozyma tsukubaensisである。好ましいモイジオマイセス属微生物はMoesziomyces antarcticus、 Moesziomyces aphidisである。好ましいウスチラゴ属微生物は、Ustilago hordei及びUstilago maydisである。好ましいSporisorium属微生物は、Sporisorium reilianum及びSporisorium scitamineumである。好ましいMelanopsichium属微生物は、Melanopsichium pennsylvanicumである。好ましいクルツマノマイセス属微生物は、Kurtzmanomyces sp. I-11である。好適な一実施形態において、MEL産生微生物は、シュードザイマ属微生物であり、より好ましくはPseudozyma tsukubaensisに属する微生物であり、更に具体的には、Pseudozyma tsukubaensis 1E5(JCM16987株)、NBRC1940(ATCC24555、CBS422.96、CBS6389、DBVPG6988、PYCC4855、JCM10324、MUCL29894、NCYC1510、NRRLY-7792)である。Pseudozyma tsukubaensisに属する微生物は、1-O-β-MEL-Bを選択的に生産することが知られている。
一実施形態において、モノアシル型MEL生産微生物は、従来型のMELを生産する微生物に変異を加えることによって得ることができる。ここで従来型のMELとはジアシル型MELである。変異の種類は特に制限されないが、好ましくはMEL生産微生物が有するトランスポーターをコードする遺伝子を破壊する変異であることが好ましい。遺伝子の破壊とは、遺伝子がコードするタンパク質(例えば、トランスポーター)が機能しなくなることを意味し、その態様は特に制限されない。一実施形態において、MEL産生微生物が有するトランスポーターをコードする遺伝子を破壊することによって、モノアシル型MELを生産する微生物を得ることができる。MEL生産微生物は、一般的に、トランスポーター(MMF1)を有する。
遺伝子の破壊は任意の手法で行うことができる。例えば、遺伝子の破壊は、当該遺伝子の塩基配列に変異を導入する方法、当該遺伝子の発現制御領域(プロモーター等)を破壊又は欠失させる方法、又は当該遺伝子の転写産物の翻訳を阻害する方法によって実施することができる。これらは、例えば、相同組換え法、トランスポゾン法、トランスジーン法、転写後遺伝子サイレンシング法、RNAi法、ナンセンス仲介減衰(Nonsense mediated decay, NMD)法、リボザイム法、アンチセンス法、miRNA(micro-RNA)法、siRNA(small interfering RNA)法等を用いて行うことができる。
一実施形態において、遺伝子の破壊は、相同組換え法を用いて行うことが好ましい。相同組換え法による遺伝子を破壊する手法は公知である。例えば、相同組換え法による標的遺伝子の破壊は、標的遺伝子のORF中に、薬剤耐性又は栄養要求性を相補する遺伝子等の選択マーカー遺伝子を挿入した遺伝子カセットを作成し、それを適当なベクター(例えば、プラスミド)に組み込んで、宿主微生物(例えば、従来型MEL生産微生物)に導入し、相同組換えによって標的遺伝子中にマーカー遺伝子を挿入する方法である。標的遺伝子が破壊された微生物は、上記マーカー遺伝子の発現により選択することができる。
相同組換え法で使用するマーカー遺伝子は、遺伝子工学において、通常使用される形質転換体の選択マーカー遺伝子を用いることができる。例えば、ハイグロマイシン、ゼオシン、カナマイシン、クロラムフェニコール、及びG418等の薬剤に耐性を付与する遺伝子、ウラシル合成酵素、ロイシン合成酵素、アデニン合成酵素、及びリシン合成酵素等の栄養要求性を相補する遺伝子が挙げられる。
一実施形態において、標的遺伝子は、MMF1遺伝子であることが好ましい。代表的なMMF1遺伝子の例は次のとおりである。配列番号1は、Pseudozyma antaractica T34株由来のトランスポーター(PaMMF1)をコードする塩基配列である。配列番号2は、Pseudozyma antaractica JCM10317株由来のトランスポーター(PaMMF1)をコードする塩基配列である。配列番号3は、Pseudozyma hubeiensis SY62株由来のトランスポーター(PhMMF1)をコードする塩基配列である。配列番号4は、Pseudozyma tsukubaensis NBRC1940株由来のトランスポーター(PtMMF1)をコードする塩基配列である。配列番号5は、Pseudozyma tsukubaensis1E5株由来のトランスポーター(PtMMF1)をコードする塩基配列である。配列番号6は、Pseudozyma aphidis DSM70725株由来のトランスポーター(MMF1)をコードする塩基配列である。これらの配列情報に基づいてトランスポーターの遺伝子を破壊するためのベクターを構築することができる。尚、P.antarctica T-34は、「Moesziomyces antarcticus T-34」とも称される。P. aphidisは、「Moesziomyces aphidis」とも称される。
遺伝子破壊に使用するベクターの種類は任意であり、宿主の種類に応じて適宜選択できる。シュードザイマ属を宿主とする場合のベクターとしては、例えば、pUXV1 ATCC 77463、pUXV2 ATCC 77464、pUXV5 ATCC 77468、pUXV6 ATCC 77469、pUXV7 ATCC 77470、pUXV8 ATCC 77471、pUXV3 ATCC 77465、pU2X1 ATCC 77466、pU2X2 ATCC 77467、pTA2、pUXV1-neo、pPAX1-neo、pPAA1-neo(Appl Microbiol Biotechnol (2016) 100:3207-3217)、pUC_neo及び pUCT_neo等を例示することができる。
宿主細胞へのベクターの導入は任意の手法で行うことができ、宿主細胞及びベクターの種類等に応じて適宜選択できる。ベクターの導入は、例えば、エレクトロポレーション、リン酸カルシウム共沈降法、リポフェクション、マイクロインジェクション、及び酢酸リチウム法等によって実施することができる。
微生物は、モノアシル型MEL-Bに加えてモノアシル型MEL-Dを産生することが好ましい。このような微生物も上述のトランスポーター遺伝子破壊によって得ることができる。一実施形態において、微生物は、モノアシル型MEL-B、モノアシル型MEL-D、及びジアシル型MEL-Bを産生することが好ましい。このような微生物も上述のトランスポーター遺伝子破壊によって得ることができる。
上述の微生物を用いたモノアシル型MEL-B、モノアシル型MEL-D、及び/又はジアシル型MEL-Bの生産は任意の方法で行うことができる。例えば、上述の微生物の培養に適した培地で培養することによってモノアシル型MEL-B、モノアシル型MEL-D、及び/又はジアシル型MEL-Bを生産することができる。そのような培地としては、特に制限されないが、例えば、炭素原料にグルコース、ショ糖、廃糖蜜などの糖質を用いることが望ましい。糖質に加えて、もしくは置き換えて、油脂類などを炭素源として用いることもできる。油脂類の種類は特に制限されず、例えば、植物油脂、或いは、脂肪酸又はそのエステル類を添加することが好ましい。
一実施形態において、培地に植物油脂を添加することが好ましい。植物油脂の種類は特に制限されず、目的とするMELの種類等に応じて適宜選択することができる。例えば、大豆油、オリーブ油、ナタネ油、紅花油、ゴマ油、パームオイル、ひまわり油、ココナッツ油、カカオバター、及びひまし油等を挙げることができる。脂肪酸としては、例えば、カプリル酸、カプリン酸、ラウリン酸、ミリスチン酸、パルミチン酸、ステアリン酸、オレイン酸、リノール酸、リノレン酸、アラキドン酸、ベヘン酸、及びネルボン酸等を挙げることができる。一実施形態において、好ましい脂肪酸は、オレイン酸である。
一実施形態において、炭素源としてグルコースのみを含む培地でモノアシル型MEL-B、モノアシル型MEL-D、及び/又はジアシル型MEL-Bを生産する微生物を培養することができる。窒素源としては、有機窒素源と無機窒素源を組み合わせて用いることができる。例えば、有機窒素源として、酵母エキス、麦芽エキス、ペプトン、ポリペプトン、コーンスティープリカー、カザミノ酸、及び尿素から成る群より選択される一種類もしくは二種類以上を組み合わせて用いることができる。
無機窒素源としては、硝酸ナトリウム、硝酸カリウム、硝酸アンモニウム、硫酸アンモニウム、及びアンモニアから成る群より選択される一種類もしくは二種類以上を組み合わせて用いることができる。別の実施形態において、脂肪酸及びグリセリンを添加した培地で上述微生物を培養することを含む、モノアシル型MEL-B、モノアシル型MEL-D、及び/又はジアシル型MEL-Bを製造する方法が提供される。
脂肪酸及び油脂類の量は、特に制限されないが、例えば、各々培地中の濃度が0.1~40容量%となるように添加することができる。
微生物の培養条件は特に制限されない。例えば、pH5~8、好ましくはpH6、温度20~35℃、好ましくは22~28℃の条件で3~7日間培養することができる。
上述の微生物が産生したモノアシル型MEL-B、モノアシル型MEL-D、及び/又はジアシル型MEL-Bの抽出は、任意の手法で行うことができる。例えば、培養液又は菌体破砕液を遠心分離し、上清を回収し、上清に適当な抽出溶媒を添加し、抽出溶媒層を回収し、必要に応じて更なる精製を行って、モノアシル型MELを得ることができる。一実施形態において、モノアシル型MELの抽出に用いる抽出溶媒は、酢酸エチル、メタノール、エタノール及び、アセトン及びこれらの混合物からなる群より選択される一種以上であることが好ましい。
上述の微生物を培養して培養物(又はその抽出物)は、モノアシル型MEL-Bを含む組成物、モノアシル型MEL-B及びモノアシル型MEL-Dを含む組成物、又はモノアシル型MEL-B、モノアシル型MEL-D及びジアシル型MEL-Bを含む組成物であることが好ましい。一実施形態において、MEL含有組成物は、モノアシル型MEL-B、モノアシル型MEL-D及びジアシル型MEL-Bを含むことが好ましく、全MELに占める各MELの割合は、モノアシル型MEL-Bが約50質量%、モノアシル型MEL-Dが約17質量%、ジアシル型MEL-Bが約33質量%であることが好ましい。このようなMEL含有組成物(MEL混合物)は、より高濃度で水系溶媒に溶解することが可能である。
以下、実施例により本発明についてさらに詳細に説明するが、本発明はこれらに制限されるものではない。
材料
・使用菌体
シュードザイマ・ツクバエンシス(Pseudozyma tsukubaensis)1E5株(JCM16987)
・培地
YM培地:脱イオン水1Lに、酵母エキス3g、麦芽エキス3g、ペプトン5g、グルコース10gを溶かして調製した。必要に応じて、アガー20gを添加し寒天培地とした。
グリセロール添加YM培地:脱イオン水1Lに、酵母エキス3g、麦芽エキス3g、ペプトン5g、グルコース10g、グリセロール50gを溶かして調製した。
MEL生産培地:脱イオン水1Lに、酵母エキス5g、硝酸ナトリウム3g、リン酸二水素カリウム0.3g、硫酸マグネシウム・七水和物0.3g、グリセロール20gを溶かして調製した。5-FOA寒天培地:脱イオン水1Lに、Yeast nitrogen base w/o AA 1.7g、硫酸アンモニウム5g、グルコース20g、ウラシル0.5g、5-FOA 2g、アガー20gを溶かして調整した。
・使用菌体
シュードザイマ・ツクバエンシス(Pseudozyma tsukubaensis)1E5株(JCM16987)

MEL生産培地:脱イオン水1Lに、酵母エキス5g、硝酸ナトリウム3g、リン酸二水素カリウム0.3g、硫酸マグネシウム・七水和物0.3g、グリセロール20gを溶かして調製した。5-FOA寒天培地:脱イオン水1Lに、Yeast nitrogen base w/o AA 1.7g、硫酸アンモニウム5g、グルコース20g、ウラシル0.5g、5-FOA 2g、アガー20gを溶かして調整した。
1.ウラシル要求性変異株の取得
Pseudozyma tsukubaensis 1E5株を1白金耳分をYM培地2mlに植菌し、25℃、180rpmの条件で24時間振とう培養した。培養液をシャーレに広げ、UV灯(Panasonic社製GL15殺菌灯)から45cmの位置にプレートを配置した。UVを照射し、培養液を0.2ml回収した。回収した培養液を25℃で3時間インキュベートし、5-FOA寒天培地に塗布植菌した。植菌したプレートを25℃で10日間インキュベートし、コロニーを生育させた。5-FOA寒天培地に生育したコロニーをYM寒天培地につまようじで植え継ぎ、菌体の一部をコロニーPCR及びシーケンス解析に供した。PCRのプライマーには下記の塩基配列を有するものを用いた。

シーケンス解析の結果、PtURA5遺伝子に変異が挿入されていることを確認した。尚、得られたウラシル要求性変異体の栄養要求性試験を行い、ウラシル要求性を有していることを確認した。また、MEL生産能を維持していることも確認した。
Pseudozyma tsukubaensis 1E5株を1白金耳分をYM培地2mlに植菌し、25℃、180rpmの条件で24時間振とう培養した。培養液をシャーレに広げ、UV灯(Panasonic社製GL15殺菌灯)から45cmの位置にプレートを配置した。UVを照射し、培養液を0.2ml回収した。回収した培養液を25℃で3時間インキュベートし、5-FOA寒天培地に塗布植菌した。植菌したプレートを25℃で10日間インキュベートし、コロニーを生育させた。5-FOA寒天培地に生育したコロニーをYM寒天培地につまようじで植え継ぎ、菌体の一部をコロニーPCR及びシーケンス解析に供した。PCRのプライマーには下記の塩基配列を有するものを用いた。

2.PtMMF1遺伝子の破壊株の取得
2-1.PtMMF1ベクターの構築
Pseudozyma tsukubaensis 1E5株のゲノムDNAをテンプレートとしたPCRにてPtMMF1遺伝子領域(遺伝子の上流・下流2kbを含む)を増幅した。PCRのプライマーには下記の塩基配列を有するものを用いた。

Pseudozyma tsukubaensis 1E5株のゲノムDNAをテンプレートとしたPCRにてPtURA5遺伝子領域(遺伝子の上流1kbおよび下流0.5kbを含む)の増幅を行い、PtURA5遺伝子断片を取得した。PCRのプライマーには下記の塩基配列を有するものを用いた。

2-3.形質転換体の調製
上記2-2.で得られたPtMMF1破壊ベクターを制限酵素KpnIおよびXbaIで処理したものを用いて線状化した後、エレクトロポレーション法にて上記1.で得たウラシル要求性変異株に形質転換した。形質転換体の選別には、ウラシル要求性消失を使用した。目的のDNA断片が相同組み換えによって目的のゲノム位置に挿入されたことは、コロニーPCRおよびシーケンス解析にて確認した。PCRのプライマーには下記の塩基配列を有するものを用いた。

図4は、形質転換体のコロニーPCRの結果を示す。図中ΔPtMMF1はPtMMF1破壊株を、矢印はプライマーを示す。また、図中(a)はコロニーPCRに用いたプライマーの結合位置、(b)はプライマーペアAによるコロニーPCRの結果、(c)はプライマーペアBによるコロニーPCRの結果、(d)はプライマーペアCによるコロニーPCRの結果を示す。コロニーPCRの結果、PtMMF1遺伝子を破壊した変異株5株を取得した。
3.PtMMF1遺伝子破壊株の生産物の評価
3-1.PtMMF1遺伝子破壊株の培養
PtMMF1遺伝子破壊株5株をそれぞれグリセロール添加YM培地2mLで25℃、250rpmで2日間振とう培養し、前培養液を得た。次いで、前培養液1mLをMEL生産培地に6%オリーブ油を添加した培地20mLに接種し、25℃、250rpmで7日間振とう培養した。
3-1.PtMMF1遺伝子破壊株の培養
PtMMF1遺伝子破壊株5株をそれぞれグリセロール添加YM培地2mLで25℃、250rpmで2日間振とう培養し、前培養液を得た。次いで、前培養液1mLをMEL生産培地に6%オリーブ油を添加した培地20mLに接種し、25℃、250rpmで7日間振とう培養した。
3-2.PtMMF1遺伝子破壊株の培養物の抽出
上記3-1で得た菌体培養液を3,000rpmで遠心し、培養上清を回収した。回収した上清は-20℃で凍結した後、凍結乾燥を行った。乾燥後の上清にアセトンを添加し、ボルテックスで攪拌した後一晩室温で静置した。静置後、アセトン層を回収し、0.45μmのフィルターでろ過したものをアセトン抽出物とした。
上記3-1で得た菌体培養液を3,000rpmで遠心し、培養上清を回収した。回収した上清は-20℃で凍結した後、凍結乾燥を行った。乾燥後の上清にアセトンを添加し、ボルテックスで攪拌した後一晩室温で静置した。静置後、アセトン層を回収し、0.45μmのフィルターでろ過したものをアセトン抽出物とした。
3-3.抽出物中のMELの評価
各アセトン抽出物に含まれるMELは、薄層クロマトグラフィ(TLC)で分析した。展開相の組成は、クロロホルム:メタノール:12%アンモニア水=55:25:2で行った。展開後のTLCプレートに2%アンスロン硫酸試薬を噴霧し、95℃、5分間加熱してMELのスポットを検出した。
各アセトン抽出物に含まれるMELは、薄層クロマトグラフィ(TLC)で分析した。展開相の組成は、クロロホルム:メタノール:12%アンモニア水=55:25:2で行った。展開後のTLCプレートに2%アンスロン硫酸試薬を噴霧し、95℃、5分間加熱してMELのスポットを検出した。
PtMMF1遺伝子破壊株を培養した結果を図5に示す。図中ΔPtMAT1はPtMAT1破壊株を、ΔPtMAC2はPtMAC2破壊株を、ΔPtMMF1はPtMMF1破壊株を示す。分析の結果、PtMMF1遺伝子破壊株ではジアシル型MEL-Bの生産量が減少し、代わりにモノアシル型MEL-Dと構造未知の糖脂質の生産が優勢となっていることが確認された。
4.構造未知の糖脂質の分析
4-1.高速液体クロマトグラフィー-質量分析計(LC-MS)による分析
PtMMF1遺伝子破壊株の培養物をアセトン抽出したサンプルをLC-MSで分析した。分析の条件は下記の通りである。

LC-MS分析の結果を図6に示す。図6に示される通り、PtMMF1破壊株が生産する構造未知のMELの質量数は、モノアシル型MEL-B(カプリル酸付加体)のギ酸イオン付加体と一致することが確認された。
4-2.核磁気共鳴分光法(NMR)による分析
アセトン抽出物をシリカゲルカラム(ワコーシルC-200、富士フイルム和光純薬社製)を用いたオープンカラムに供し、構造未知のMELを精製した。溶媒として、クロロホルムおよびアセトンを用いた。

シリカゲルカラムで精製した構造未知のMEL50mgを1mL d4-メタノールに溶解し、Bruker AVANCE 400(400 MHz)で解析を行った。1H-NMRおよび、2次元NMR(COSY)の結果を図7(a)(b)に示す。図7に示される通り、アセチル基のプロトンに由来するシグナルが2.10ppm付近に1本検出され、かつマンノース1位のシグナル(4.75ppm)との積分比が1:3であることから、構造未知のMELはアセチル基がひとつ結合したモノアセチル型MELであることが判明した。また、-COCH
2 -のシグナル(2.43ppm)がマンノース1位のシグナル(4.75ppm)と積分比1:2であることから、構造未知のMELは脂肪酸鎖が1本結合したモノアシル型MELであることが判明した。
図7に示される通り、マンノース6位のシグナルが4.23-4.48ppmに低磁場シフトしていること、加えて、PtMMF1破壊株の親株であるP. tsukubaensis 1E5株が、マンノース6位にアセチル基が結合したMEL-Bを生産することから、構造未知のMELはMEL-Bであると考えられる。また、マンノース2位のシグナルが5.38-5.39ppmに低磁場シフトしていることに加え、P. tsukubaensis 1E5株が、マンノース2位および3位に脂肪酸鎖が結合したMEL-Bを生産することから、脂肪酸鎖はマンノース2位に結合していると考えられる。以上の結果から、PtMMF1破壊株が生産する構造未知のMELは、マンノース2位に脂肪酸鎖が1本結合したモノアシル型MEL-Bであることが判明した。
5.PtMMF1破壊株が生産するMEL混合物の表面張力の測定
PtMMF1破壊株の菌体培養液をアセトン抽出して得られた、モノアシル型MEL-Bを主成分、モノアシル型MEL-D、およびジアシル型MEL-B、ジアシル型MEL-Dを副成分として含むMEL混合物の界面活性能の評価を行った。アセトン抽出物をシリカゲルカラム(ワコーシルC-200、富士フイルム和光純薬社製)を用いたオープンカラムに供し、抽出物中の残存油脂および残存脂肪酸を除去した。表面張力の測定は、協和界面科学株式会社製のDropmaster DMo-501を用いて行い、種々の濃度のMEL混合物の表面張力を測定した。
PtMMF1破壊株の菌体培養液をアセトン抽出して得られた、モノアシル型MEL-Bを主成分、モノアシル型MEL-D、およびジアシル型MEL-B、ジアシル型MEL-Dを副成分として含むMEL混合物の界面活性能の評価を行った。アセトン抽出物をシリカゲルカラム(ワコーシルC-200、富士フイルム和光純薬社製)を用いたオープンカラムに供し、抽出物中の残存油脂および残存脂肪酸を除去した。表面張力の測定は、協和界面科学株式会社製のDropmaster DMo-501を用いて行い、種々の濃度のMEL混合物の表面張力を測定した。
図8に示すように、表面張力の値はMEL混合物の濃度の上昇に伴って減少し、臨海ミセル濃度(CMC)に達すると一定となった。なお、図8より、MEL混合物のCMCは4.0mg/mLであった。MEL混合物の主成分であるモノアシル型MEL-B(カプリル酸付加体)の分子量452から計算すると、CMC=8.88x10-3 Mと換算できる。また、γCMCは33.1mN/mであった。
親株が生産するジアシル型MEL-Bの場合、CMCが3.1x10-6 Mであることが知られている(T. Fukuokaら、Carbohydrate Research., 351 (2012) 81-86)。PtMMF1破壊株が生産するMEL混合物のCMCはジアシル型MEL-Bよりも1000倍以上高く、高濃度でも単一MEL分子として水に均一に分散し、水溶性が向上し、水に速やかに溶解可能である。すなわち、PtMMF1破壊株が生産するMEL混合物は、水系での使いやすさに優れている。
図6に示す結果から、PtMMF1破壊株が生産するMELは、約33質量%がジアシル型MEL-Bであり、約50質量%がモノアシル型MEL-Bであり、約17質量%がモノアシル型MEL-Dであると推定される。
Claims (6)
- モノアシル型MEL-Bを産生するシュードザイマ属微生物。
- トランスポーター(PtMMF1)をコードする遺伝子が欠損されている、請求項1に記載の微生物。
- 更に、モノアシル型MEL-Dを産生する、請求項1又は2に記載の微生物。
- 微生物がシュードザイマ・ツクバエンシスである、請求項1~3のいずれかに記載の微生物。
- 請求項1~4のいずれかに記載の微生物を用いてモノアシル型MEL-Bを製造する方法。
- 請求項1~4のいずれかに記載の微生物を培養することによって得られるMEL含有組成物。
