Abstract
FGF2 활성을 가지며 온도안정성 및 단백질 분해효소 저항성이 향상된 폴리펩타이드가 제공된다. 폴리펩타이드는 서열번호 1에서 28번째 아스파르트산(D)이 글루탐산(E)으로 치환되거나, 78번째 시스테인(C)이 이소류신(I) 또는 류신(L)으로 치환되거나, 96번째 시스테인(C)이 이소류신(I) 또는 트립토판(W)으로 치환되거나 하는 것 중에서 선택된 적어도 하나 이상의 치환을 포함한다.
Description
본 기재는 온도 안정성 및 단백질 분해효소 저항성을 향상시킨 FGF2 폴리펩타이드 및 그 용도에 관한 것이다.
FGF(Fibroblast Growth Factor)는 세포의 성장, 증식, 분화를 조절하는 중요한 역할을 수행하는 인자이다. 인체의 각 조직의 기능을 유지하기 위해 다양한 종류의 FGF들이 생성되며, 이들은 세포의 분화와 증식에 고유의 기능을 수행하고 있다. 그러나, 노화가 진행됨에 따라 피부 등 각 조직에서 FGF들의 농도는 점진적으로 낮아지며, 이에 따라 세포의 재생 및 분열 기능이 약화되어 피부에 주름이 형성되고 탄력이 감소하게 된다.
다양한 FGF 중에서 FGF2(Fibroblast Growth Factor 2)는 주로 155개의 아미노산으로 구성되어 있으며 분자량은 약 18kDa이다. FGF2는 광범위한 유사분열(mitogenic) 및 세포 생존 활성(cell survival activity)을 가지며, 상처 치유, 혈관 생성, 그리고 신경계의 성장에서 강력한 매개체로서의 역할을 한다.
따라서, FGF2는 혈관신생 촉진, 상처 치유 촉진, 연골형성 또는 골형성의 촉진 및 신경발생을 촉진시키는 의약품으로 개발되고 있을 뿐만 아니라 피부 재생, 주름 제거 또는 탄력 증가를 위한 화장품 원료로도 널리 사용되고 있다. 또한 세포를 만능성 상태로 유지하는 기능 때문에 인간 만능성 줄기 세포(PSC) 배양용 배지 등에도 주요인자로 첨가되고 있다.
이와 같이 인체내에서 다양한 기능을 갖는 FGF2는 열역학적으로 안정성이 상피세포성장인자(EGF), 인슐린유사성장인자(IGF) 그리고 혈관내피세포성장인자(VEGF) 등에 비해 크게 떨어지는 것으로 보고되고 있다. 또한 단백질 분해효소에 의해서 절단되기 쉬운 문제도 있다. 따라서 FGF2가 산업적인 용도에 적합하게 사용되기 위해서는 FGF2의 열역학적 안정성 및/또는 단백질 분해효소 저항성이 담보되는 것이 필수적인 조건이다.
본 개시는 온도 안정성 및 단백질 분해효소 저항성을 향상시킨 FGF2 폴리펩타이드를 제공하고자 한다.
본 개시는 온도 안정성 및 단백질 분해효소 저항성을 향상시킨 FGF2 폴리펩타이드를 포함하는 약제학적 또는 화장용 조성물을 제공하고자 한다.
본 개시는 온도 안정성 및 단백질 분해효소 저항성을 향상시킨 FGF2 폴리펩타이드를 포함하는 인간 만능성 줄기 세포 배양 배지를 제공하고자 한다.
실시예들에 따른 온도 안정성을 향상시킨 FGF2 폴리펩타이드는 서열번호 1에서 28번째 아스파르트산(D)이 글루탐산(E)으로 치환되거나, 78번째 시스테인(C)이 류신(L) 또는 이소류신(I)으로 치환되거나, 96번째 시스테인(C)이 트립토판(W) 또는 이소류신(I)으로 치환되는 것 중에서 선택된 적어도 하나 이상의 치환을 포함하고, FGF2 고유의 활성을 가지는 온도안정성을 향상시킨(thermally stable) 폴리펩타이드이다.
실시예들에 따른 조성물은 온도안정성 및 단백질 분해효소 저항성을 향상시킨 폴리펩타이드 및 약제학적으로 또는 화장용으로 허용가능한 담체를 포함한다.
실시예들에 따른 인간 만능성 줄기 세포 배양 배지는 온도안정성 및 단백질 분해효소 저항성을 향상시킨 폴리펩타이드를 유효성분으로 포함한다.
실시예들에 따른 FGF2 폴리펩타이드는 제품제조 후 야생형 인간 FGF2 폴리펩타이드와 비교시 향상된 온도안정성 및 단백질 분해효소 저항성을 나타낸다.
온도안정성 및 단백질 분해효소 저항성을 향상시킨 폴리펩타이드는 유통과 보관 과정 중에도 기존의 야생 인간 FGF2 제품과 달리 활성 유지가 가능하다. 따라서, 이를 약제학적 또는 화장용 조성물의 유효성분으로 사용할 수 있다. 또한, 미분화성 증식을 유도하는 활성이 인간 만능성 줄기 세포 배양 배지의 유효성분으로 사용시 야생형 FGF2 대비 오랜 기간 유지될 수 있다는 장점을 가진다.

이하 본 기술분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있도록 실시예들에 대하여 상세히 설명한다. 실시예는 여러가지 상이한 형태로 구현될 수 있으며, 여기에서 설명하는 구체적인 실시예로만 한정되지 않는다.
본 개시에서 사용되는 일부 용어들의 정의가 아래에 달리 정의되지 않은 한, 본원에 사용되는 모든 기술적 및 과학적 용어들은 본 발명이 속하는 기술 분야의 당업자가 통상적으로 이해하는 바와 동일한 의미를 가진다.
본 개시에서 기술된 기법 및 공정들은 일반적으로 통례적인 방법에 따라 수행되며, 이는 본원 전체에 제시된다. 일반적으로, 본 개시에 사용된 명명 및 분자 생물학, 생화학, 분석 화학 및 세포 배양에서의 실험 절차들은 당해 기술 분야에서 널리 공지되어 있으며 통상적으로 사용되는 바와 동일하다.
변이체
본 개시는 부위 특이적인 돌연변이 유발에 의해 열적으로 안정화된 FGF2 폴리펩타이드를 제공한다. 본 개시에서는 생물정보 분석 및 컴퓨터를 이용한 단백질 설계를 통해 기존에 알려져 있지 않던 신규 위치에서 가장 최적의 아미노산을 합리적으로 예측한 후 부위-특이적인 돌연변이 유발에 의해 돌연변이를 제조하였다.
도 1은 야생형 인간 FGF2 폴리펩타이드 서열을 나타낸다.
본 개시에서 용어 '야생형'은 종의 구성원들에서 가장 공통적인 아미노산 서열을 가진 천연형 FGF2를 의미한다. 본 개시에서, 야생형 FGF2는 아미노산 155개 길이의 (서열번호 1, 도 1) 18 kDa 단백질인 인간 FGF2이다.
본 개시에서 '단편'은 FGF2 활성을 가진 FGF2 폴리펩타이드의 기능성 단편을 지칭한다. 또한, 서열번호 1의 서열과 85% 이상의 서열 동일성을 가진 FGF2 폴리펩타이드의 기능성 단편을 지칭한다. FGF2 폴리펩타이드의 단편은 또한 본 발명에 따른 하나 이상의 치환을 적어도 가진다. 적어도 96%, 97%, 98%, 99% 또는 100%의 서열 동일성이 바람직하다. 단편은 온전한 폴리펩타이드 서열 및 구조의 일부로만 구성되는 폴리펩타이드로 의도되며, 변이체의 C-말단 결손 또는 N-말단 결손이 존재할 수 있다. 이러한 기능성 단편은 본 발명에 따른 대상 FGF2 단백질의 세포 결합 영역과 헤파린 결합 세그먼트를 보유할 수 있다.
본 개시에서 '서열 동일성'은 동일한 아미노산 잔기가 전술한 바와 같은 본 발명에 따른 FGF2 폴리펩타이드에서 발견되는 것을 의미한다. FGF2 폴리펩타이드의 아미노산 서열의 명시된 연속 세그먼트를 정렬하여 기준 분자에 해당되는 특정 아미노산 서열과 비교하였을 때, 야생형 인간 FGF2 폴리펩타이드가 기준으로서 사용된다. 서열 동일성의 %는, 양쪽 서열에서 동일한 아미노산 잔기가 존재하는 위치의 수를 측정함으로써 일치된 위치의 수를 산정하고, 이를 기준 분자와 비교하는 세그먼트의 전체 위치 개수로 나누고, 이에 100을 곱하여, 서열 동일성의 %를 산출함으로써, 계산한다. 서열 정렬 방법은 당해 기술 분야에 잘 공지되어 있다. 본원에 사용되는 기준 서열은 본 발명에 따른 특정 대응되는 인간 야생형 FGF2 단백질을 지칭한다. 예를 들어, 마우스, 랫, 토끼, 영장류, 돼지, 개, 소, 말 및 인간 등의 포유류 종의 경우, FGF2는 고도로 보존되어 있으며, 광범위한 종들에서 85% 이상의 서열 동일성을 나타낸다. 바람직하게는 서열 동일성은 적어도 96%, 97%, 98%, 또는 99% 이상 또는 100%이다. 당해 기술 분야의 당업자라면, 본 발명에 따른 FGF2 단백질의 전장에서 나머지 15% 이하의 아미노산은 예를 들어 FGF2 종의 다른 소스를 이용하거나 또는 당해 기술 분야에 일반적으로 공지된 적합한 비-FGF2 펩타이드 서열 또는 태그의 부가로 인해, 가변적일 수 있음을, 이해할 것이다. 야생형 FGF2에 대해 85% 이상의 동일성을 가진 본 발명의 구현예에 따른 FGF2 단백질은, FGF 패밀리의 다른 멤버들은 일반적으로 매우 낮은 서열 동일성을 가지므로, 비슷한 FGF2 이외의 다른 단백질을 포함할 가능성은 낮다.
본 발명자는 야생형 인간 FGF2에서 28번째 위치 또는 137번째 위치가 FGF2 폴리펩타이드의 열적 안정성 및/또는 단백질 분해효소 저항성과 연관된 위치임을 확인하였다. 그리고, FGF2의 표면에 노출되어 있는 시스테인 중 78번째 위치 또는 96번째 위치의 시스테인이 열적 안정성 및/또는 단백질 분해효소 저항성과 연관된 위치임을 확인하였다.
열적 안정성 및/또는 단백질 분해효소 저항성과 연관된 위치에서 가장 적절한 아미노산으로 바꾸는 것 발명자의 노력(inventive step)을 요하는 것이다.
본 발명자는 28번째 위치의 아스파르트산(D)를 글루탐산(E)으로 치환함으로써 열적 안정성을 향상시킬 수 있음을 확인하였다.
또한, 137번째 위치의 세린(S)을 프롤린(P)으로 치환함으로써 열적 안정성을 보다 더 향상시킬 수 있음을 확인하였다.
또한, 28번째 위치와 137번째 위치를 함께 치환함으로써 열적 안정성을 보다 더 향상시킬 수 있음을 확인하였다.
78번째 위치와 96번째 위치의 경우에는 SDM((http://marid.bioc.cam.ac.uk, University of Cambridge)와 Discovery studio 2019(BIOVIA)을 통해 19개의 돌연변이 후 안정화 비교를 진행하여 확인하였다.
78번째 위치의 경우 SDM 분석결과에서는 류신(L)으로 돌연변이시킬 경우에 예측 값(Predicted pseudo △G) 0.33으로 가장 높게 나왔으며, Discovery studio에서는 이소류신(I)으로 돌연변이시킬 경우에 돌연변이 에너지 변화 값(kcal/mol)이 -1.20으로 가장 안정한 것으로 예측되었다.
96번째 위치의 경우 SDM 분석결과에서는 이소류신(L)으로 돌연변이시킬 경우에 예측 값(Predicted pseudo △G) 0.19으로 가장 높게 나왔으며, Discovery studio에서는 트립토판(W)으로 돌연변이시킬 경우에 돌연변이 에너지 변화 값(kcal/mol)이 -0.39으로 가장 안정한 것으로 예측되었다.
US9169309, US2017-0291931, EP3380508, US20180319857 등에서는 78번째의 시스테인을 세린(S), 티로신(Y)으로 치환하거나, 96번째의 시스테인을 세린(S), 티로신(Y), 트레오닌(T) 또는 아스파라긴(N) 등으로 치환한 내용이 개시되어 있다. 이들 치환된 아미노산의 경우에는 대다수가 친수성이며 전하가 없는 아미노산인 반면, 본 개시에서는 치환된 아미노산은 소수성 아미노산으로 종래의 기술로부터 용이하게 예측할 수 없는 범주로 볼 수 있다.
따라서, 본 개시에서 가능한 변이체는 아래 표 1에 개시된 다양한 변이체중 어느 하나일 수 있다.

본 개시는 조작된 대상 FGF2의 특징 규명, 단백질에서의 치환 효과 입증, 인간 PSC 배양에서 단백질의 이용 방법, 및 인간 PSC를 미분화된 상태로 배양하는데 적합한 본원에 기술된 하나 이상의 내열성 FGF2 단백질을 포함하는 배지를 제공한다. 본원에 제공된 실시예들에서 사용되는 인간 배아 줄기 세포 (ESC)는 의사에 의한 사전동의 하에 수득된 배반포기 배아로부터 유래되었다. 29-41회 계대의 특징이 잘 규명된 인간 ESC 세포주 (Adewumi, et al. 2007, Nat Biotechnol 25, 803-816) CCTL14 (Centre of Cell Therapy Line)를 사용하였다. 인간 유도된 만능성 줄기 세포 (iPSC)에서와 같이, 야마나카의 칵테일 (Yamanaka's cocktail) 및 센다이 바이러스 형질감염 (Sendai virus transfection)에 의한 피부 섬유모세포의 리프로그래밍을 이용하여 유래된 AM13 세포주는 34-41회 계대 상태를 사용하였다 (Kruta et al. 2014, Stem Cells and Development 23, 2443-2454).
이하, 본 발명의 이해를 돕기 위하여 바람직한 실험예들을 제시하나, 하기 실험예들은 본 발명을 예시하는 것일 뿐 본 발명의 범위가 하기 실시예에 한정되는 것은 아니다.
실험예 1: pQE80 벡터를 이용한 돌연변이체들의 구축, 정제 및 열적 안정성 분석
FGF2의 1개 위치의 돌연변이(S137P)와 2개 위치의 돌연변이(D28E, S137P)를 합성하고, His-Tag를 가진 pQE80L 벡터에 서브클로닝하였다. FGF2가 삽입된 재조합 벡터를 Rosetta(DE3)pLysS 세포에 형질전환하여 발현하였다.
10ml LB media(Ambrothia) (0.25g 사용)에 접종하고 암피실린(Ampicillin, 50mg/ml)을 10ul을 첨가 후 37℃에 전 배양하였다.
전배양액 10ml과 암피실린(Ampicillin, 50mg/ml) 1ml을 1L LB media(ambrothia)(25g 사용)에 접종하여 37℃에서 본 배양하였다. OD600의 수치가 0.6일 때 4℃냉장고에서 배양액을 10분간 냉각시켜준 후 베타-디-1-티오갈락토피라노사이드 (β-D-l-thiogalactopyranoside; IPTG) 0.5mM을 첨가하여 20℃에서 20시간 발현을 유도한 대장균 세포를 얻었다.
발현된 pQE80_FGF2는 용해완충용액(20mM Tris pH 8.0, 200mM NaCl, 3mM DTT)에 녹여서 초음파 처리 후 13000 r.p.m에 30분 동안 원심분리를 수행한 다음 정제하였다.
원심분리 후, 최적으로 용해된 상층액은 Heparin 비즈가 있는 컬럼에 주입하였다. 컬럼에 주입된 pQE80_FGF2 단백질 부피의 3배 되는 첫 번째 세척(wash) 완충용액(20mM Tris pH 8.0, 200mM NaCl, 3mM DTT)과 두 번째 세척(wash) 완충용액 (20mM Tris pH 8.0, 500mM NaCl, 3mM DTT)으로 씻고, 60ml 용리완충용액(elution buffer), 20mM Tris pH 8.0, 1800mM NaCl, 3mM DTT)으로 용리하여 1차 정제하였다.
마지막으로 pQE80_FGF2 단백질 분획은 HiLoadTM 16/60 Superdex 75(Amersham Biosciences) 컬럼과 1X PBS 버퍼(137mM NaCl, 2.7mM KCl, 10mM Na2HPO4, 2mM KH2PO4, pH 7.4), (WELGENE)를 이용한 젤 여과 방법에 의해 정제되었다.
정제된 FGF2 단백질 들은 1X PBS 버퍼를 기본 사용하여 0.5mg/ml 농도로 37℃에서 0, 2, 4, 6일 동안 반응하였으며 코마쉬 블루 염색시약으로 염색하여 15% SDS-PAGE 전기영동을 실시하였다. 도 2에 그 결과가 예시되어 있다.
도 2에 예시되어 있는 바와 같이, 15% SDS-PAGE를 통해 확인된 FGF2 폴리펩타이드 밴드들로부터 두 변이 폴리펩타이드 (pQE80_hFGF2(S137P), pQE80_hFGF2 (D28E, S137P)) 모두 변이되지 않은 hFGF2 (야생형) 폴리펩타이드 보다 SDS-PAGE상 안정성이 개선되었음을 알 수 있다.
실험예 2: pET17b 벡터를 이용한 돌연변이체들의 구축, 정제 및 열적 안정성 분석
FGF2의 1개 위치의 돌연변이(S137P), 2개 위치의 돌연변이(D28E, S137P), 4개 위치의 돌연변이((D28E, C78L, C96I, S137P), (D28E, C78L, C96W, S137P), (D28E, C78I, C96I, S137P), (D28E, C78I, C96W, S137P))를 합성하고, His를 가진 pET17b 벡터에 서브클로닝하였다. FGF2가 삽입된 재조합 벡터를 Rosetta(DE3)pLysS 세포에 형질전환하여 발현하였다.

4개 위치 돌연 변이체들은 친화성 크로마토그래피 방법에서 완충용액과 용리완충용액 모두에서 3mM DTT를 포함하지 않도록 제조한 용액을 사용하여 정제하였다.
마지막으로 pET17b_FGF2 단백질 분획은 HiLoadTM 16/60 Superdex 75(Amersham Biosciences) 컬럼과 1X PBS 버퍼(137mM NaCl, 2.7mM KCl, 10mM Na2HPO4, 2mM KH2PO4, pH 7.4), (WELGENE)를 이용한 젤 여과 방법에 의해 정제되었다.
37℃안정성 실험
정제된 FGF2 단백질 들은 1X PBS 버퍼를 기본 사용하여 0.5mg/ml 농도로 37℃에서 0, 3, 6, 9일 동안 반응하였으며 코마쉬 블루 염색시약으로 염색하여 15% SDS-PAGE 전기영동을 실시하였다. 도 3에 그 결과가 예시되어 있다.
도 3에 예시되어 있는 바와 같이, 15% SDS-PAGE를 통해 확인된 FGF2 폴리펩타이드 밴드들로부터 변이체들의 열적 안정성이 향상되었음을 알 수 있다.
밀도 측정은 imageJ 프로그램(Wanyne Rasband)을 사용하여 SDS-PAGE 젤의 밀도를 측정하였다. 그 결과가 아래 표 2에 기재되어 있다.

단위 %표 2의 결과로부터 4개 위치의 변이체들이 야생형 hFGF2 보다 열적 안정성이 향상되었음을 알 수 있다. 특히, △9N-hFGF2(D28E, C78L, C96I, S137P), △9N-hFGF2(D28E, C78L, C96W, S137P), △9N-hFGF2(D28E, C78I, C96I, S137P)의 경우에 다른 변이체들 대비 열적 안정성이 상대적으로 더 우수함을 알 수 있다. 45℃ 안정성 실험정제된 FGF2 단백질 들은 1X PBS 버퍼를 기본 사용하여 0.5mg/ml 농도로 45℃에서 0, 1, 2, 3, 4, 5, 6일 동안 반응하였으며 코마쉬 블루 염색시약으로 염색하여 15% SDS-PAGE 전기영동을 실시하였다. 도 4에 그 결과가 예시되어 있다.
도 4에 예시되어 있는 바와 같이, 15% SDS-PAGE를 통해 확인된 FGF2 폴리펩타이드 밴드들로부터 변이체들의 열적 안정성이 향상되었음을 알 수 있다.

표 3의 결과로부터 모든 변이체들이 야생형 hFGF2 보다 열적 안정성이 향상되었음을 알 수 있다. 특히, △9N-hFGF2(D28E, C78L, C96I, S137P), △9N-hFGF2(D28E, C78I, C96I, S137P)의 경우에 다른 변이체들 대비 열적 안정성이 상대적으로 더 우수함을 알 수 있다. 실험예 3: pQE80 벡터를 이용한 돌연변이체들의 세포증식능 확인실험예 1과 동일한 방법으로 제조한 돌연변이체들에 대해서 BALB3T3 세포를 사용하였고, 10% bovine serum을 포함한 DMEM 배지에서 배양 및 유지하였다. FGF2에 의한 세포증식활성을 확인하기 위해서 세포는 인슐린(insulin) 10 ug/ml, 덱사메타손(dexamethasone) 1 uM, 트랜스페린(transferrin) 10 ug/ml, 아셀렌산나트륨(sodium selenite) 10 ng/ml, 오브알부민(ovalbumin) 100 ug/ml, 피브로넥틴(fibronectin) 5 ug/ml을 포함하는 F12/DMEM 배지에서 배양하였다.
96 웰 플레이트에 세포수 0.5 x 104 /well 로 배양하였고, 헤파린(heparin) (10 ug/ml) 과 함께 FGF2 (0.3 ng/ml)로 42시간 처리하였다. 세포수 증가는 WST-8 [2-(2-methoxy-4-nitrophenyl)-3-(4-nitrophenyl)-5-(2,4-disulfophenyl)-2H-tetrazolium, monosodium salt]를 사용하여, 전자 매개체(electron mediator)와 세포내 탈수소효소(dehydrogenases)에 의해 형성되는 WST-8 포르마잔(formazan) 생성 정도를 측정함으로써 확인하였다. WST-8 포르마잔 생성 정도는 흡광도 (450 nm)을 통하여 확인할 수 있다. 실험은 3번 반복하였고, '평균 ± 표준편차' 방식으로 표현하였다. FGF2 단백질들은 37℃에서 0, 2, 4, 6일 동안 각각 보관한 후, 세포증식활성 변화를 확인하였다.
그 결과가 도 5에 예시되어 있다. 도 5를 참조하면, hFGF2는 37℃ 보관 후 2일 조건에서 활성 감소가 관찰되었고, 37℃ 보관 4일 이후로는 37℃ 보관 0일 단백질의 60% 수준의 활성만이 관찰되었다. 반면, hFGF2 S137P 변이체는 37℃ 보관 2일 조건에서 hFGF2 보다 더 낮아진 활성이 관찰되었고, 37℃ 보관 4일 조건에서는 37℃ 보관 0일 단백질의 60% 수준의 활성만이 관찰되었다. 반면, hFGF2 D28E+S137P 변이체는 37℃ 보관 날짜가 길어질수록 활성 감소가 나타나기는 하나, hFGF2와 hFGF2 S137P 변이체보다는 활성 감소가 훨씬 적게 나타남을 알 수 있다.
실험예 4: pET17b 벡터를 이용한 돌연변이체들의 세포증식능 확인
실험예 2와 동일한 방법으로 제조한 돌연변이체들에 대해서 BALB3T3 세포를 사용하였고, 10% bovine serum을 포함한 DMEM 배지에서 배양 및 유지하였다. FGF2에 의한 세포증식활성을 확인하기 위해서 세포는 인슐린(insulin) 10 ug/ml, 덱사메타손(dexamethasone) 1 uM, 트랜스페린(transferrin) 10 ug/ml, 아셀렌산나트륨(sodium selenite) 10 ng/ml, 오브알부민(ovalbumin) 100 ug/ml, 피브로넥틴(fibronectin) 5 ug/ml 을 포함하는 F12/DMEM 배지에서 배양하였다.

그 결과가 도 6에 예시되어 있다. 도 6을 참조하면, △9_hFGF2 (야생형)은 37℃ 보관 3일 조건에서 활성이 감소되어 있었고, 37℃ 보관 6일 이후로는 활성을 거의 잃어버리는 결과를 확인하였다. △9_hFGF2 D28E+S137P 변이체는 야생형 단백질 보다 오래 활성을 가지는 결과를 보였으며, 37℃ 9일 보관 후로는 활성이 감소하는 결과를 나타내었다. 이들과 다르게, △9_hFGF2 D28E + C78L + C96I + S137P 변이단백질과 △9_hFGF2 D28E + C78I + C96I + S137P 변이체는 37℃ 12일 보관 조건까지도 활성에 감소가 관찰되지 않음을 확인할 수 있었다. 따라서 실시예들에 따른 변이체들이 37℃에서는 단백질의 활성이 안정적으로 유지됨을 확인할 수 있었다.
FGF2 단백질들은 45℃에서 0, 1, 2, 3, 4, 5, 6, 7일동안 각각 보관한 후, 세포증식활성 변화를 확인하였다. 그 결과가 도 7에 예시되어 있다. 도 7을 참조하면, △9_hFGF2 (야생형)은 45℃ 보관 1일 조건에서부터 활성이 급격히 감소하였음을 확인하였고, 45℃ 보관 2일 조건에서부터 활성을 거의 잃어버리는 결과를 확인하였다. △9_hFGF2 D28E+S137P 변이단백질은 야생형 단백질 보다 오랫동안 활성이 관찰되는 결과를 보였으나, 45℃ 2일 보관 이후로 활성을 떨어지는 결과를 나타내었다. 이들과 다르게, △9_hFGF2 D28E + C78L + C96I + S137P 변이단백질과 △9_hFGF2 D28E + C78I + C96I + S137P 변이단백질의 활성이 45℃에서 보관날짜에 따라 조금씩 감소하기는 하지만, 야생형보다는 오랫동안 활성이 유지되며, 45℃ 6일 보관에서도 여전히 2배 이상의 증식 활성능이 유지되고 있음을 확인할 수 있었다.
실험예 5: pET17b 벡터를 이용한 돌연변이체들의 단백질 분해효소 절단에 대한 저항성 확인
실험예 2와 동일한 방법으로 제조한 △9_hFGF2 (야생형)과 △9_hFGF2 D28E + C78L + C96I + S137P 변이단백질의 단백질 분해효소 절단에 대한 저항성을 측정하였다.
도 8 및 도 9는 각각 △9_hFGF2 (야생형)과 △9_hFGF2 D28E + C78L + C96I + S137P 변이단백질의 SDS-PAGE를 측정한 결과를 나타낸다. 도 8 및 도 9에서 1번은 단백질 분해효소 처리 후 인큐베이션 없이 바로 측정한 경우를, 2번은 단백질 분해효소 처리 후 37 ℃에서 3시간 인큐베이션한 후 측정한 경우를, 3 내지 14번은 FGF2 0.25mg/mL에 대해서 12가지의 서로 다른 종류의 단백질 분해효소를 각각 0.0025mg/mL 처리한 후 37℃ 에서 3시간 인큐베이션한 후에 측정한 결과를 나타낸다.
각 밴드의 밀도 측정은 imageJ 프로그램(Wanyne Rasband)을 사용하여 SDS-PAGE 젤의 밀도를 측정하였다. 그 결과가 아래 표 4에 기재되어 있다.

도 8 및 도 9의 결과와 표 4의 내용으로부터 △9_hFGF2 (야생형) 보다 △9_hFGF2 D28E + C78L + C96I + S137P 변이단백질이 단백질 분해효소에 대한 저항성이 전반적으로 향상됨을 알 수 있다. 특히, α-Chymotrypsin (α-C), Trypsin (TR), Subtilisin (SU), Proteinase K (P-K), Clostripain (Endoproteinase-Arg-C) (CL), Thermolysin (TH), Actinase (A-E) 에 대한 저항성이 상대적으로 많이 향상되었음을 알 수 있다. 상기에서는 다양한 실시예들에 대하여 설명하였지만, 권리범위는 이에 의해 한정되는 것이 아니다. 구현되는 형태는 발명의 상세한 설명 및 첨부한 도면의 범위 안에서 여러 가지로 변형하여 실시하는 것이 가능하고 이 또한 권리 범위에 속하는 것은 당연하다.
화장품 또는 의약품 기술 분야에 이용가능하다.
Claims (8)
- 서열번호 1에서 28번째 아스파르트산(D)이 글루탐산(E)으로 치환되거나,78번째 시스테인(C)이 이소류신(I) 또는 류신(L)으로 치환되거나,96번째 시스테인(C)이 이소류신(I) 또는 트립토판(W)으로 치환되거나 하는 것 중에서 선택된 적어도 하나 이상의 치환을 포함하고,FGF2 활성을 가지는 온도안정성이 향상된(thermally stable) 폴리펩타이드.
- 제1 항에 있어서,137번째 세린(S)이 프롤린(P)으로 더 치환된 온도안정성이 향상된 폴리펩타이드.
- 제2 항에 있어서,상기 치환은 2개, 3개 또는 4개인 온도안정성이 향상된 폴리펩타이드.
- 제3 항에 있어서,상기 28번째 아스파르트산(D)이 글루탐산(E)으로 치환되고,상기 78번째 시스테인(C)이 이소류신(I) 또는 류신(L)으로 치환되고,상기 96번째 시스테인(C)이 이소류신(I)으로 치환되고,상기 137번째 세린(S)이 프롤린(P)으로 치환된 온도안정성이 향상된 폴리펩타이드.

- 제1 항 내지 제6 항 중 어느 한 항에 따른 온도안정성이 향상된 폴리펩타이드; 및약제학적으로 또는 화장용으로 허용가능한 담체를 포함하는 조성물.
