Abstract
본 발명은 골질환 예방 또는 치료용 약학 조성물에 관한 것으로, 보다 상세하게는 특정 화학식의 화합물을 포함함으로써 파골세포 분화 및/또는 생성을 억제하고 치주염 등을 포함하는 다양한 골질환에 대해 우수한 예방 또는 치료 효과를 나타낼 수 있다.

본 발명은 골질환 예방 또는 치료용 약학 조성물 및 건강기능식품에 관한 것이다.
골조직은 파골세포(osteoclast)에 의한 골흡수와 조골세포(osteoblast)에 의한 골형성 과정이 지속적으로 유지되는 조직이다. 조골세포는 골 형태 발생 단백질 2 (Bone Morphogenetic Protein 2, BMP2) 등의 신호인자에 의해 분화가 촉진된다. 조골세포 분화단계에서 생성되는 RANKL(Receptor Activator of Nuclear factor Kappa-β Ligand) 신호전달물질에 의해 파골세포의 분화가 촉진되며, 분화가 완료되면 세포사멸(apoptosis)이 일어난다. 따라서, 정상적인 골 재생 과정에서 조골세포와 파골세포의 분화 및 활성이 조화롭게 이루어지는 것이 중요하다.
치주염증은 치근단 세균감염에 대한 면역세포의 방어작용에 의한 염증반응으로, 치주염증에 주로 관여하는 호중구는 염증매개물질인 프로스타글란딘을 분비한다. 프로스타글란딘, RANKL 등의 세포신호전달 물질은 골조직을 흡수하는 파골세포를 활성화시켜 염증부위 주변의 치조골의 재흡수를 일으킬 수 있다. 만성치주염 치료를 위해 골재생 촉진제 또는 골흡수 억제제 개발이 요구된다. 뿐만 아니라, 비정상적인 뼈의 흡수를 조절하고, 정상적인 골재생 과정을 회복하기 위해 파골세포 분화 억제제 개발이 요구된다.
본 발명은 골질환 예방 또는 치료용 약학 조성물을 제공하는 것을 목적으로 한다.

약학적으로 허용 가능한 염은 예를 들어 산 부가염, 염기 부가염 또는 금속염일 수 있다.

본 발명 조성물에 포함되는 화학식 1 내지 화학식 4로 표시되는 화합물은 천연으로부터 유래될 수도 있고, 공지의 화학적 합성 방법을 이용하여 합성될 수도 있다.
본 발명의 약학 조성물에 의해 예방 또는 치료될 수 있는 골질환은 조골세포와 파골세포의 활성 불균형에 의해 유발되는 것 일 수 있다.
뼈는 살아 있는 조직이기 때문에 오래된 뼈는 일정하게 파괴되고 다시 새로운 뼈를 만들어내는 재형성 과정을 거친다. 이러한 과정 중에서 파골세포는 오래되어 불필요하게 된 뼈 조직을 파괴하여 칼슘이 혈류로 방출되어 신체기능을 유지할 수 있도록 도와주고, 조골세포는 파괴된 뼈를 다시 재생시키는 역할을 한다. 이 작용은 하루 24시간 계속 일어나며 1년에 성인의 뼈의 약 10 % 내지 30%가 이런 식으로 다시 만들어진다. 따라서 파골세포와 조골세포 간의 균형은 매우 중요하며, 이러한 균형은 여러 호르몬과 기타 몸의 화학 성분 등에 의해 조절된다.
구체적으로, 골질환은 파골세포 과분화 또는 조골세포 활성 감소에 의한 것일 수 있으며, 구체적으로는 골질환은 파골세포 과분화에 의한 것일 수 있다.

치주염은 치근단 세균감염에 대한 면역세포의 방어작용에 의한 염증반응이다. 치주염에 주로 관여하는 호중구는 염증매개물질인 프로스타글란딘을 분비한다. 프로스타글란딘 등의 세포신호전달 물질에 의해 골조직을 흡수하는 파골세포가 활성화되어 염증부위 주변의 치조골 소실이 관찰된다. 또한, 만성 치주염은 지속적인 치주근단 부분의 염증 및 치조골 부식을 나타내는 데, 치주염이 악화되어 치아를 살릴 수 없는 경우 발치를 하고 임플란트를 시행한다. 이를 예방하기 위해 치주염 초기단계에서 감염을 일으키는 세포수 감소를 위한 항생제 투여 및 염증매개물질에 의한 파골세포 활성화를 극복할 수 있는 파골세포 억제제를 투여하여 만성치주염의 완화 및 치료를 기대할 수 있다. 본 발명 골질환의 예방 또는 치료용 약학 조성물은 골흡수 억제의 효과를 통해 치주염의 예방 또는 치료에 이용될 수 있다.
본 발명 약학 조성물은 공지된 골질환 치료 물질과 함께 혼합하여 제공될 수도 있다.
본 발명 약학 조성물은 공지된 골질환의 예방 또는 치료 물질과 병용 투여될 수 있다.
용어 "투여"란 적절한 방법으로 개체에게 소정의 물질을 도입하는 것을 의미하며, 용어 "개체"란 골질환이 발병하였거나 발병할 수 있는 인간을 포함한 쥐, 생쥐, 가축 등의 모든 동물을 의미한다. 구체적인 예로, 인간을 포함한 포유동물일 수 있다.
필요에 따라, 본 발명 약학 조성물은 공지의 항골질환 화합물을 추가적으로 포함할 수 있다.
이러한 항골질환 화합물로는 신코닌, 갈색거저리 추출물, 알로에-이모딘 및 오메가-3 지방산, 아르테아뉴인 B, 인돌-2-카르복실레이트 유도체, 유포비아 인자 L1, 스컬캅플라본 유도체, 프락시넬론 등을 들 수 있으나, 이에 제한되는 것은 아니다.
본 발명 약학 조성물은 캡슐, 정제, 과립, 주사제, 연고제, 분말 또는 음료 형태일 수 있다.
본 발명 약학 조성물은 산제, 과립제, 캡슐, 정제, 수성 현탁액 등의 경구형 제형, 외용제, 좌제 및 주사제의 형태로 제형화하여 사용될 수 있다.
본 발명 약학 조성물은 유효성분을 단독으로 포함하거나, 하나 이상의 약학적으로 허용되는 담체, 부형제 또는 희석제를 더 포함할 수 있다.
본 발명 약학 조성물은 약학적으로 허용 가능한 담체를 포함할 수 있다. 약학적으로 허용 가능한 담체는 경구 투여 시에는 결합제, 활탁제, 붕해제, 부형제, 가용화제, 분산제, 안정화제, 현탁화제, 색소, 향료 등일 수 있으며, 주사제의 경우에는 완충제, 보존제, 무통화제, 가용화제, 등장제, 안정화제 등을 혼합하여 사용할 수 있으며, 국소투여용의 경우는 기제, 부형제, 윤활제, 보존제 등을 사용할 수 있다.
본 발명 약학 조성물의 제형은 약학적으로 허용되는 담체와 혼합하여 다양하게 제조될 수 있으며, 예를 들어, 경구 투여시에는 정제, 트로키, 캡슐, 엘릭서(elixir), 서스펜션, 시럽, 웨이퍼 등의 형태로 제조될 수 있으며, 주사제의 경우에는 단위 투약 앰플 또는 다수회 투약 형태로 제조될 수 있다. 또한, 본 발명 약학 조성물의 제형은 용액, 현탁액, 정제, 캡슐, 서방형 제제 등으로 제조될 수 있다.
제제화를 위한 담체, 부형제 및 희석제는 락토즈, 덱스트로즈, 수크로즈, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말디톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로즈, 폴리비닐피롤리돈, 물, 메틸하이드록시벤조에이트, 프로필하이드록시벤조에이트, 탈크, 마그네슘 스테아레이트, 광물유, 충진제, 항응집제, 윤활제, 습윤제, 향료, 유화제 또는 방부제 등일 수 있다.
본 발명 약학 조성물의 투여 경로는 구강, 정맥내, 근육내, 동맥내, 골수내, 경막내, 심장내, 경피, 피하, 복강내, 비강내, 장관, 국소, 설하 또는 직장일 수 있으며, 이에 제한되지 않는다.
본 발명 약학 조성물은 경구 또는 비경구로 투여될 수 있으며, 비경구 투여 시 피부 외용 또는 복강내주사, 직장내주사, 피하주사, 정맥주사, 근육내 주사 또는 흉부내 주사 주입방식이 선택될 수 있다.
본 발명의 약학 조성물의 투여량은 환자의 상태 및 체중, 질병의 정도, 약물형태, 투여경로 및 기간에 따라 다르지만, 당업자에 의해 적절하게 선택될 수 있다.
예를 들어, 본 발명 약학 조성물은 1일 0.0001 mg 내지 1000mg/kg 또는 0.001mg 내지 500mg/kg으로 투여될 수 있다. 본 발명 약학 조성물의 투여는 하루에 한번 투여할 수도 있고, 수회 나누어 투여할 수도 있다.
또한, 본 발명은 하기 화학식 1로 표시되는 화합물 또는 이의 식품학적으로 허용 가능한 염을 포함하는 골질환 예방 또는 개선용 건강기능식품을 제공한다:

본 발명 건강기능식품은 담체, 희석제, 부형제 및 첨가제 중 하나 이상을 더 포함하여 정제, 환제, 산제, 과립제, 분말제, 캡슐제 및 액제 제형으로 이루어진 군에서 선택된 하나로 제형될 수 있다.
건강기능식품은 예컨대, 각종 식품류, 분말, 과립, 정제, 캡슐, 시럽제, 음료, 껌, 차, 비타민 복합제, 또는 건강기능성 식품류일 수 있다.
건강기능식품에 포함될 수 있는 첨가제는 천연 탄수화물, 향미제, 영양제, 비타민, 광물(전해질), 풍미제(합성 풍미제, 천연 풍미제 등), 착색제, 충진제(치즈, 초콜렛 등), 팩트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH조절제, 안정화제, 방부제, 산화 방지제, 글리세린, 알콜, 탄산화제 및 과육으로 이루어진 군으로부터 선택될 수 있다.
천연 탄수화물은 예는 모노사카라이드, 예를 들어, 포도당, 과당 등; 디사카라이드, 예를 들어 말토스, 슈크로스 등; 및 폴리사카라이드, 예를 들어 덱스트린, 시클로덱스트린 등과 같은 통상적인 당, 및 자일리톨, 소르비톨, 에리트리톨 등의 당알콜이다. 상기 향미제로서 천연 향미제(타우마틴, 스테비아 추출물(예를 들어 레바우디오시드 A, 글리시르히진등) 및 합성 향미제(사카린, 아스파르탐 등)를 유리하게 사용할 수 있다.
건강기능식품은 여러 가지 영양제, 비타민, 광물(전해질), 합성 풍미제 및 천연 풍미제 등의 풍미제, 착색제 및 중진제(치즈, 초콜릿 등), 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알콜, 탄산 음료에 사용되는 탄산화제, 천연 과일 쥬스 및 야채 음료의 제조를 위한 과육을 등을 더 포함할 수 있다.
담체, 부형제, 희석제 및 첨가제는 이에 한정되는 것은 아니나, 락토즈, 덱스트로즈, 슈크로즈, 솔비톨, 만니톨, 에리스리톨, 전분, 아카시아 고무, 인산칼슘, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리 케이트, 미세결정성 셀룰로즈, 폴리비닐피롤리돈, 셀룰로즈, 폴리비닐피롤리돈, 메틸셀룰로즈, 물, 설탕시럽, 메틸셀룰로즈, 메틸 하이드록시 벤조에이트, 프로필하이드록시 벤조에이트, 활석, 스테아트산 마그네슘 및 미네랄 오일로 이루어진 군에서 선택될 수 있다.
본 발명 건강기능식품을 제제화할 경우에는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제될 수 있다.
이하, 본 발명을 구체적으로 설명하기 위해 아래 실시예로 보다 상세하게 설명한다.
아래 실시예의 35D35는 상기 화학식 2(2-(2-chlorophenoxy)-N-[2-(4-propionyl-1-piperazinyl)phenyl]acetamide)로 표시되는 화합물이고, 35D5는 상기 화학식 3으로 표시되는 화합물이고, 35D3은 N-(2-(4-acetylpiperazin-1-yl)phenyl)-2-(2-chlorophenoxy)acetamide이고, 35D8은 2-(4-chlorophenoxy)-N-(2-(4-methylpiperazin-1-yl)phenyl)acetamide이고, 35D3(또는 PPOA)은 상기 화학식 4로 표시되는 화합물이다.
세포독성 평가를 위한 MTT 분석
전구파골세포(preosteoclast)에 35D35 또는 35D3(=PPOA)(1~10uM)를 처리한 후 세포생존능을 평가했다. 96 well plate에 well당 1x10
4개의 세포를 넣고, 오직 MCSF(30ng/mL)와 PPOA만을 제시된 농도 조건하에서 날짜별로 배양했다. 이 후 세포생존능 분석 시약(DoGen, EZ-3000)을 배지 용량의 1/10 로 처리한 후 37℃에 30분간 배양하고 450nM에서 확인했다. 35D35 화합물 및 PPOA를 이용해 세포독성을 확인한 결과, 10uM 농도를 처리한 경우에도 세포사멸 효과를 나타내지 않았다(도 3 및 도 15참조). 즉, 세포독성은 없는 것으로 확인되었다.
파골세포 분화억제 효과 확인
1. 화학식 2 및 3으로 표시되는 화합물의 파골세포 분화억제 효과
1) 마우스 골수세포의 분리 및 파골세포의 분화
10주령 암컷 마우스의 골수(bone marrow)를 관골(hip bone), 넙다리뼈 (femur) 및 정강뼈(tibia)에서 분리하여 대식세포콜로니자극인자 (Macrophage Colony Stimulating Factor, M-CSF, PeproTech, 315-02, 30ng/ml)가 포함된 α-MEM 배지 (Gibco, 12561-056, 10% fetal bovine serum, 1% penicillin/streptomycin)에서 3 일 동안 배양하여 단핵세포(monocyte)만을 분리하였다. 분리한 단핵세포에 RANKL(Receptor Activator of Nuclear factor kappa B ligand, PeproTech, 315-11, 50ng/ml)을 처리하여 파골세포 분화를 유도하였다. RANKL 처리시 각 화합물(35D35, 35D5, 35D3, 35D8)들을 처리하여 3일 또는 5일 동안 배양해 파골세포 분화 정도를 확인하였다.
2) TRAP 염색 및 분석을 통한 파골세포 분화억제능 확인
마우스 골수에서 분리한 단핵세포 l x 10
4개를 96 well plate에 넣고 RANKL과 각 화합물(35D35, 35D5, 35D3, 35D8)을 함께 처리하여 각 화합물이 파골세포 분화능에 미치는 영향을 확인하였다. TRAP activity assay kit (Cosmo Bio, PMC-AK04F-COS)를 사용하여 파골세포 분화시 발현이 증가하는 Tartrate-resistant acid phosphatase(TRAP)의 활성을 확인하였다. TRAP staining으로 파골세포 분화 정도를 확인한 결과, 다른 유도체 대비 35D35, 35D5 및 35D3 화합물이 보다 우수한 파골세포 분화억제 효과를 나타내었다(도 1 참조). 염색 후 현미경으로 파골세포 분화 시 형성되는 다핵 세포를 확인하였으며, 각 well을 사진으로 찍어 이를 Image J software를 이용하여 전체 면적당 분화면적비율을 나타내었다. 그 결과 35D35 화합물은 0.5uM의 농도에서 효과적인 파골세포 분화 억제능을 나타내었다(도 2 참조).
또한, 파골세포 생성억제능이 우수한 35D35 화합물이 파골세포 분화 시기 별 미치는 영향을 확인하였다. 구체적으로, BMM을 5개 그룹(Ctrl, Period I 내지 IV)으로 나누었고, 30ng/mL M-CSF 와 50ng/mL RANKL를 함유하는 배양배지에서 4일동안 배양하였다. Period I 내지 IV 그룹의 BMM을 각각 다른 날짜에 24시간 동안 1 μM 35D35 에 노출시켰다. 4일 후 각 그룹의 셀을 고정하였고, TRAP 염색을 수행한 후 파골세포 존재를 확인하였다. 그 결과, 35D35 화합물을 처리한 그룹에서 초기 24시간 동안 파골세포 분화가 억제됨을 확인할 수 있었다(도 4 참조, "M+R"은 M-CSF+RANKL 처리를 의미한다.).
위 결과들로부터, 35D35, 35D5, D35D3 화합물은 우수한 파골세포 분화 억제효과를 나타내며, 특히 파골세포의 분화 초기에 영향을 미친다는 것을 확인하였다.
3) 골흡수 저해능 평가
분화된 파골세포의 활성을 확인하기 위해 F-actin ring 형성능 (formation)을 측정하였다. 구체적으로, 화합물을 포함(35D3: 6μM, 35D5 및 35D35: 2μM)하거나 포함하지 않은 12mm 커버 글라스에 BMM을 시딩하고, M-CSF와 LANKL를 처리하였다. 대조군에서 파골세포가 형성된 후, 4.0% 파라포름알데히드에서 15분간 파골세포를 고정한 후, 5% FBS에서 배양하여 약 60분 동안 블로킹하였다. PBS를 사용하여 세척한 후 각 웰에 로다민이 컨쥬게이션된 팔로이딘(rhodamine-conjugated phalloidin)(1:40)을 첨가하여 F-액틴 벨트를 시각화하였다. 20분 후, DAPI(1:5000) 용액을 5분 동안 세포와 함께 배양하였다. PBS로 3회 세척한 후 형광 현미경을 사용하여 세포들을 관찰하였다. 그 결과, 35D35, 35D5 및 35D3은 우수한 RANKL-유도 F-actin 벨트 형성 억제 효과를 나타내었다(도 5 참조, scale bar=200μm, "Con" = M-CSF+RANKL 처리군).
또한, 우수한 F-actin 벨트 형성 억제 효과를 나타내는 35D35 화합물을 다양한 농도로 처리하여 F-actin ring 형성능을 확인한 결과, 35D35 0.5uM을 처리한 세포에서 F-actin ring 형성능이 50%이상 감소하는 것을 확인하였다(도 6 참조, scale bar=200μm, "Con" = M-CSF+RANKL 처리군, "M" = M-CSF 처리군). F-actin 크기는 Image J를 이용하여 측정하였다.
아울러, Bone resorption assay kit(Cosmo Bio,CSR-BRA)를 사용하여 35D35의 골 흡수능을 측정하였다. 구체적으로, BMM을 플루오레세아민(fluoresceinamine) 표지된 칼슘인산염 플레이트에 시딩하고 35D35를 다양한 용량으로 처리하였다. 6일 후, 형광판독기를 사용하여 485nm와 535nm의 흡수 파장과 방출 파장에서 형광 강도를 측정하였다. Image J를 이용하여 재흡수 Pit area를 계산하였다. 그 결과, 화합물을 처리하지 않은 대조군 세포(Ctrl)에 비해 35D35 0.5uM을 처리한 세포에서 골 흡수능이 대략 10% 저하 되는 것을 확인할 수 있었다(도 7 참조, scale bar=200μm, "Con" = M-CSF+RANKL 처리군, "M" = M-CSF 처리군).
4) 파골세포 분화 관련 전사인자의 발현감소 확인
파골세포 특이적 유전자의 발현 정도를 확인하기 위해, 35D35(1μM) 및 35D3(6μM) 화합물이 처리된 세포에서 RT-PCR로 mRNA 발현량을 측정하였다. RNA extraction kit를 이용해 파골세포로 분화한 세포로부터 RNA를 분리하고 spectrophotometer로 정량한 0.5ug의 RNA를 역전사 효소(Takara, RR037 A)를 이용해 cDNA를 합성하였다. QuantStudio 3 real- time PCR system(Applied Biosystems)으로 합성된 cDNA를 이용해 qRT-PCR을 수행하였다. cell lysates를 처리한 후, 항-Ctsk 항체로 면역블롯팅을 수행하였다. 로딩 컨트롤로 β-actin을 사용하였다.

RT-PCR 수행결과, 35D35를 처리하지 않은 대조군(Ctrl)에 비해 35D35를 처리한 그룹에서 파골세포 특이적 유전자인 AcP5(TRAP), NFATc1, DC-STAMP, ATP6v0d2, MMP9 및 CTSK의 mRNA 발현양이 감소하였다(도 8 참조, "M" = M-CSF 처리군). 도 8의 그래프는 Day 0에서 컨트롤의 발현량으로 전사 정도를 정규화한 것이다. 특히, 35D3 화합물을 처리한 경우에 비해 35D35 화합물을 처리한 경우 Ctsk 단백질 발현을 낮은 농도에서도 강하게 억제하는 것을 확인하였다(도 9 참조,). 이를 통해 35D3 화합물에 비해 35D35는 보다 적은 양으로도 파골세포의 분화를 억제함으로써 효과적인 골 대사 억제제로서 기능할 수 있을 것이다.
2. 화학식 4로 표시되는 화합물의 파골세포 분화억제 효과
1) 마우스 골수 세포의 분리 및 파골세포의 분화
12주령 암컷 마우스의 골수(bone marrow)를 관골(hip bone), 넙다리뼈(femur) 및 정강뼈(tibia)에서 분리하여 대식세포 콜로니자극인자 (Macrophage Colony Stimulating Factor, M-CSF, PeproTech, cat# 315-02, 30ng/ml)가 포함된 α-MEM 배지(Gibco, 12561-056, 10% fetal bovine serum, 1% penicillin/streptomycin)에서 3일 동안 배양하여 단핵세포(monocyte)만을 분리하였다. 분리한 단핵세포에 RANKL(PeproTech, cat# 315-11, 50ng/ml)을 처리하여 파골세포 분화를 유도하였다. 여기에 35D3(PPOA)을 함께 처리하여 3일 또는 5일 동안 배양하였다.
2) TRAP 염색 및 분석을 통한 파골세포 분화억제능 확인
마우스 골수에서 분리한 단핵세포 1x10
4 개를 96 well plate에 넣고 RANKL과 함께 처리하여 35D3(PPOA)(3μM)가 파골세포 분화능에 미치는 영향을 확인했다. 파골세포 분화시 발현이 증가하는 Tartrate-resistant acid phosphatase(TRAP)의 활성을 TRAP activity assay kit(Sigma-Aldrich, 387A-1kt)를 사용하여 확인하였으며, 이는 도 16에 나타냈다.
염색 후 현미경으로 파골세포 분화시 형성되는 다핵 세포를 현미경으로 확인하고, well당 12 장의 이미지를 찍어 다시 하나의 well 이미지로 합친 뒤 이를 Image J software를 이용하여 전체 면적 당 분화면적비율을 확인하였다. 전체 면적 당 분화면적비율을 도 17A에 나타나내었고, TRAP 양성 세포 (핵 번호> 3)의 면적을 DMSO (100 %)와 비교하여 상대 값으로 제시하였다.
또한 분화된 파골세포수의 개수를 세어 확인했고, 이를 도 17B에 나타내었고, 분화된 파골세포 안에 내재된 세포핵의 개수가 3-5개, 6-10개, 11-20개, 21개 이상인 파골세포를 구분하여 도 17C에 나타내었다.
도 16 및 도 17을 참조하면, 본 발명 화합물을 처리시에 정상적으로 파골세포를 분화시킨 그룹에 비해 분화면적, 파골세포의 수, 거대다핵세포 모두 유의하게 줄어든 것을 확인할 수 있었다. 이들 결과를 통해 35D3(PPOA)의 파골세포 분화 억제능을 확인할 수 있다.
3) 액틴-고리 형성 억제 및 골 흡수 저해 효과 확인
분화된 파골세포의 활성을 측정하기 위해 F-actin 염색을 통해 다핵 세포로의 분화능을 확인하였다. L-Lysine이 코팅된 유리(Corning, 354085)를 넣은 24 well plate에 마우스 골수 세포를 well당 5x10
4개를 넣고 동시에 35D3(PPOA) 6uM을 처리해 4 내지 5일동안 배양하였다. 분화된 세포를 4% formaldehyde를 이용해 10분간 고정하고, PBS로 세척하였다. 항원-항체 반응을 위해 0.1% Triton-PBS에 20분간 반응 시킨 후, 1% BSA로 1시간 동안 반응시켰다. F-actin 항체(Thermo Fisher Scientific, A12379)를 1시간 동안 반응 시킨 후 세척하였다. DAPI (4’,6-Diamidino-2-Phenylindole, Dilactate, Thermo Fisher Scientific, D3571) 염색을 통해 세포 핵을 염색한 후 현미경으로 사진을 찍었다. 그 결과, 35D3(PPOA) 처리 유무에 따라 F-actin의 크기가 다른 것을 확인할 수 있었다(도 18을 참조). 이를 통해 35D3(PPOA)가 다핵 세포로의 분화를 억제할 수 있음을 알 수 있었다.
또한, Bone resorption assay kit (Cosmo Bio,CSR-BRA)를 사용하여 골 흡수능을 확인하였다. 마우스 골수 세포를 30 ng/Ml의 M-CSF를 함유한 bone resorption assay plate 48 (2 × 10
4 cells/well) 에 시딩하였다. 24 시간 후, 성숙한 파골 세포가 형성 될 때까지 마우스 골수 세포에 M-CSF 30 ng/mL 및 RANKL 100 ng/mL 자극하에 6μM 35D3(PPOA)를 처리하거나 처리하지 않았다. 다음날 세포에서 배양 상등액을 96-well black polypropylene micro-well plate (Thermo Fisher Scientific Nunc, Waltham, MD, USA)에 수거하였다. 0.1 N NaOH 50μL를 혼합 한 후, fluorescence plate reader (Molecular Devices, San Jose, CA, USA - model: SpectraMax i3x)로 형광 강도를 측정 하였고; excitation 및 emission wavelenghts는 각각 485 nm 및 535 nm였다. 흡수 영역은 ImageJ software (https://imagej.nih.gov/ij)를 사용하여 10 배로 촬영 한 well 당 무작위로 선택된 10 개의 사진을 고려하여 계산하였다.
도 19A는 광학 현미경을 사용하여 골 흡수 영역을 시각화한 것을 나타낸다. 도 19B는 485nm의 excitation wavelength 및 535nm의 emission wavelength에서 측정된 형광 강도를 나타낸다. 도 19C는 골 흡수 pits area를 계산한 결과를 나타낸다. 골 흡수 면적의 비율은 3, 6, 10 μM의 35D3(PPOA)에 노출 된 후에 유의한 감소를 보였다.
조골세포 분화 여부 효과 확인
생후 3 일령의 C57BL/6J mouse calvaria에서 세포를 채취하여 96 well plate 에 well 당 4x10
3 cell을 시딩한 후 분화 유도인자인 hBMP2를 100ng/ml 처리하였다. 분화 유도인자 처리와 동시에 35D35 화합물을 처리하여 7일간 배양한 후 ALP 염색을 통해 분화 영향성을 평가하였다. 구체적인 실험 방법 및 결과는 아래와 같다.
1) 마우스 두개골 세포의 분리 및 조골세포의 분화
마우스의 두개골에서 분리한 전구세포 (precursor cells)에 hBMP2 (Bone morphogenic protein 2, Sino biological, 10426- HNAE, 100ng/ml)를 처리하여 조골세포로의 분화를 유도하였다. 조골세포 특이적 단백질인 alkaline phosphatase(ALP) 염색을 통해 분화된 조골세포를 구분할 수 있다. 전구세포에 hBMP2를 처리할 때 35D35 화합물 각각을 함께 처리하며, 2~3 일마다 배지를 갈아주면서 7 일간 배양하였다.
2) 조골세포의 분화능 분석을 위한 ALP(alkaine phosphatase) 염색
조골세포 전구세포 4x10
3개를 96 well plate에 넣고 hBMP2를 처리하여 조골세포의 분화를 유도하였다. 이 때 35D35를 처리하여 7 일 동안 배양하였다.
조골세포 분화능 분석을 위해 조골세포 초기 분화단계에서 발현이 증가하는 ALP의 활성을 확인하기 위해 분화 유도 7 일 후 배지를 버리고, HBSS (Hanks’ Balanced Salt Solution, welgene)로 한 번 세척한 후 차가운 70% 에탄올을 넣고 1 시간 동안 세포를 고정하였다. 에탄올을 버리고 HBSS로 세척한 후 NBT solution(Sigma, B1911)을 well 당 l00μl 넣고 15분 동안 염색하였다. 물로 2번 세척하여 남아있는 염색약을 제거하였다. ALP 염색 후 그 발색 정도를 Image J software를 이용하여 측정한 뒤 그래프로 나타내어 비교하였다. 그 결과 35D35는 조골세포에 대해 영향을 미치지 않는 것을 확인하였다 (도 10 참조). 도 10은 ALP 염색을 통해 35D35 화합물에 의한 조골세포 분화능을 확인한 결과를 나타낸다. 도 10A는 ALP 염색을 통해 분화된 조골세포를 확인한 결과를 나타내며, 도 10B는 ALP 강도 그래프를 나타낸다("Con" = BMP2 단독 처리군),
생체 내 효과 확인
8주령 암컷 마우스에서 난소적제술을 시행해 에스트로겐 부족으로 인한 골다공증 동물 모델(OVX, ovariectomized mouse)을 제작하였다. 골다공증 마우스 모델에 화합물(35D35, 35D5, 35D3)을 2일에 한 번씩 복강 내 주사 하였다. 4주 후에 마우스의 정강뼈(tibia)를 마이크로 컴퓨터 단층촬영으로 골 밀도를 분석하였다. 그 결과, 35D3 화합물을 주사한 OVX 모델에서는 대조군(OVX) 보다 높은 골 밀도와 뼈 볼륨이 관측 되지 않은 반면, 35D35 및 35D5 화합물을 주사한 OVX 모델에서 대조군(OVX) 보다 높은 골 밀도와 뼈 볼륨이 관측 되었다(도 11 내지 14참조).
도 11 내지 14는 마우스 모델의 정강뼈를 마이크로 컴퓨터 단층 촬영 결과와(Sham: 난소가 제거되지 않은 대조군), 마이크로 컴퓨터 단층촬영 결과를 분석한 그래프를 나타낸다(* P <0.05, ** P <0.01, and *** P < 0.001 vs. the control group, OVX. BMD: trabecular bone mineral density).
전술한 실험 결과들을 통해, 본 발명 화합물(35D35, 35D5, 35D3)은 우수한 파골세포의 분화 억제 효과를 나타낼 수 있는 반면, 조골세포 분화에는 영향을 주지 않는 것을 확인하였다. 또한, 분자생물학적 실험을 통해 화합물이 NFATc1를 포함하는 파골세포 관련 유전자의 발현을 저해함으로써 파골세포 관련 유전자의 발현을 저해하는 것을 예측할 수 있었다. 아울러, 난소적출 골다공증 마우스 모델에 화합물을 처리(35D35, 35D5)한 경우 우수한 골 흡수능 억제를 나타냈다. 이처럼, 본 발명 화합물은 우수한 파골세포 분화억제 효과를 나타냄으로써 효과적인 골 대사 억제제로서 기능할 수 있을 것이다.
Claims (10)
- 청구항 1에 있어서, 상기 R 1은 메틸, 에틸 또는 페닐인, 골질환 예방 또는 치료용 약학 조성물.
- 청구항 1에 있어서, 상기 R 1은 C1 내지 C3의 알킬이고, 상기 R 2는 할로이고, 상기 R 3은 H이거나; R 1은 페닐이고, 상기 R 2는 H 이고, 상기 R 3은 할로인, 골질환 예방 또는 치료용 약학 조성물.
- 청구항 1에 있어서, 상기 골질환은 골절, 골다공증, 류마티스성 관절염, 치주염, 파제트병, 골연화증, 골감소증, 골위축, 골관절염 및 무혈성대퇴골괴사로 이루어진 군에서 선택된 적어도 하나인, 골질환 예방 또는 치료용 약학 조성물.
- 청구항 6에 있어서, 상기 R 1은 메틸, 에틸 또는 페닐인, 골질환 예방 또는 개선용 건강기능식품.
- 청구항 6에 있어서, 상기 R 1은 C1 내지 C3의 알킬이고, 상기 R 2는 할로이고, 상기 R 3은 H이거나; R 1은 페닐이고, 상기 R 2는 H 이고, 상기 R 3은 할로인, 골질환 예방 또는 개선용 건강기능식품.
