Cyclophosphamide exhibits polymorphism and available in two forms i.e. monohydrate form and anhydrous form. Cyclophosphamide monohydrate has a molecular formula of C 7 H 15 Cl 2 N 2 O 2 P.H 2 O and a molecular weight of 279.1 gm/mol. The anhydrous form is less stable than monohydrate form, it easily picks up water when exposed to a relative humidity more than 20% and converted into the monohydrate form at room temperature.

Abstract
La présente invention concerne une formulation injectable stable comprenant de la cyclophosphamide en tant que principe actif et des excipients pharmaceutiquement acceptables, dépourvue de mannitol. La présente invention concerne également des procédés de préparation de compositions pharmaceutiques de cyclophosphamide par un procédé de lyophilisation au moyen d'un solvant et éventuellement d'un agent de solubilisation, le procédé de lyophilisation ne comprenant pas d'étape de réhydratation. La présente invention concerne plus particulièrement la préparation de compositions pharmaceutiques lyophilisées de cyclophosphamide ayant une solubilité et une stabilité améliorées, le procédé de lyophilisation ne comprenant pas d'étape de réhydratation et la composition étant dépourvue de mannitol. La présente invention concerne particulièrement la préparation d'une formulation lyophilisée injectable stable comprenant de la cyclophosphamide en tant que principe actif, dépourvue de solubilisant, ainsi que d'excipients pharmaceutiquement acceptables tels que d'agent de modification de pH et de mannitol.
