Abstract
Description
また、ドラッグデリバリーをはじめとする分野では、タンパク質、ペプチド又は低分子化合物を細胞内へ導入し機能させる技術として、これらの分子に膜透過性ペプチド(Cell Penetrating Peptide:CPP)を結合することが行われている。
しかし、PNAやCPPを結合したPNAをアジュバントとして利用することは知られていない。
1)細胞膜透過性ペプチドが結合したペプチド核酸からなるアジュバント。
2)細胞膜透過性ペプチドが結合したペプチド核酸及び薬学的に許容される担体を含有するアジュバント組成物。
3)1)のアジュバント及び抗原を含有するワクチン組成物。
4)アジュバントを製造するための、細胞膜透過性ペプチドが結合したペプチド核酸の使用。
5)細胞膜透過性ペプチドが結合したペプチド核酸のアジュバントとしての使用。
6)細胞膜透過性ペプチドが結合したペプチド核酸をアジュバントとして使用する方法。
本発明において、「細胞膜透過性ペプチドが結合したペプチド核酸」とは、細胞膜透過性ペプチドのN若しくはC末端にペプチド核酸が結合した分子を意味し、本明細書においては、Mitsumata-Immunostimulatory Peptide nucleic acid(MIP)とも称す。
好ましくは、TAT由来ペプチド(配列番号1で示されるアミノ酸配列からなるペプチド)が挙げられる。
下記式(1)に、3~12merのペプチド核酸の構造を示す。
なお、ペプチド核酸は、例えばFmoc型PNAモノマーユニット等を用い、当技術分野で公知の固相ペプチド合成法を利用して合成することができる。
すなわち、MIPは抗原を注射若しくは経粘膜投与する場合のいずれにおいても血中若しくは粘膜にける抗体誘導能を高めるアジュバント活性を有しており、よって、MIPはアジュバントとなり、またMIPは薬学上許容される担体を含む組成物はアジュバント組成物になり得る。また、MIPはアジュバント又はアジュバント組成物を製造するために使用できる。
ここで、「粘膜」とは脊椎動物において、消化器、呼吸器、泌尿生殖器、眼等、特に外通性の内腔器官の内壁をいう。したがって、経粘膜投与としては、例えば、経口投与、鼻腔投与(経鼻投与)、口腔投与、膣内投与、上気道投与、肺胞投与、点眼投与等が挙げられるがそれらに限定されない。
本発明のアジュバント又はアジュバント組成物の投与量は、投与対象、投与方法、投与形態、抗原の種類に応じて適宜決定することができる。
これらの製剤には、通常使用されている吸収促進剤、界面活性剤、保存剤、安定化剤、防湿剤、溶解補助剤等を必要に応じて添加することができる。
<CPP及びMIP>
CPPとしてCPP01(TAT由来ペプチド(配列番号1)、MIPとして前記式(2)で示したMIP01及び式(3)で示したMIP03を株式会社ペプチド研究所に合成を委託した。
インフルエンザHAワクチン「生研」のA/H1N1亜型(A/Singapore/GP1908/2015株)及びB/Victoria系統(B/Maryland/15/2016株)の各原液をスプリット抗原(SV)とし、0.3mL当りに各株のヘムアグルチニンが1μgとなるよう各スプリット抗原を混合し、そこへHIVのTAT由来の13アミノ酸からなる細胞膜透過性ペプチド若しくはMIP01が1μgとなるよう添加した。また、対照としてアジュバントを添加しない抗原のみの投与液も併せて調製した。
実施例1と同様にインフルエンザHAワクチン「生研」のA/H1N1亜型(A/Singapore/GP1908/2015株)及びB/Victoria系統(B/Maryland/15/2016株)の各原液をスプリット抗原とし、10μL当りに各株のヘムアグルチニンが1μgとなるよう各スプリット抗原を混合し、そこへCPP01若しくはMIP01が10μgとなるよう添加した。また、対照としてアジュバントを添加しない抗原のみの投与液も併せて調製した。
MIP03の自然免疫活性化能をin vitroで評価した。
上記の通り、NFκBの活性化及びIL-1βの高い誘導能を有するため、MIP03はC-type Lectin ReceptorやNod-Like Receptorを介した自然免疫系の活性化を促している可能性が高く、その結果、抗体応答に関連するTh1/Th2サイトカインの誘導能を示すと考えられる。したがって、MIPは、自然免疫の活性化能を有するアジュバントである。
Claims (12)
- 細胞膜透過性ペプチドが結合したペプチド核酸からなるアジュバント。
- 細胞膜透過性ペプチドが、配列番号1~7で示されるアミノ酸配列からなるペプチドから選ばれる請求項1に記載のアジュバント。
- 細胞膜透過性ペプチドが、配列番号1で示されるアミノ酸配列からなるペプチドである請求項1に記載のアジュバント。
- 式(1)において、nが1~8の整数である請求項4に記載のアジュバント。
- 式(1)において、Rが-CONH2である請求項4又は5に記載のアジュバント。
- 式(1)において、Baseがいずれもグアニン又はシトシンである請求項4~6のいずれか1項に記載のアジュバント。
- 細胞膜透過性ペプチドが結合したペプチド核酸及び薬学的に許容される担体を含有するアジュバント組成物。
- 請求項1~7のいずれか1項に記載のアジュバント及び抗原を含有するワクチン組成物。
- アジュバントを製造するための、細胞膜透過性ペプチドが結合したペプチド核酸の使用。
- 細胞膜透過性ペプチドが結合したペプチド核酸のアジュバントとしての使用。
- 細胞膜透過性ペプチドが結合したペプチド核酸をアジュバントとして使用する方法。
