Abstract

미토콘드리아는 세포내에서 에너지를 조절하는 주요 세포소기관으로서, 세포사멸 관련 기작 또한 조절하기에 세포 생존에 주요한 역할을 한다. 이 때, 뇌는 인체 내에서 가장 에너지를 많이 필요로하는 조직으로서 미토콘드리아의 기능에 크게 영향을 받으며, 미토콘드리아 기능이상에 따른 활성산소(ROS, Reactive oxygen specices)의 증가는 뇌 기능에 이상을 가져오는 동시에 자가포식반응을 조절하여 퇴행성 뇌질환과 밀접한 관련을 가진다. 아울러 미토콘드리아의 기능성 약화 및 사멸이 퇴행성 뇌신경계 질환 환자들의 공통적인 초기증상으로서 밝혀진 이후 (Hirai, K. et al., J Neurosci, 21:3017-3023, 2001), 이와 관련된 세포사멸 및 자가포식반응 또한 알쯔하이머, 파킨슨씨, 헌팅턴병 등의 주요 기전으로서 여러 문헌을 통해 밝혀지고 있다 (Martinez-Vicente, M. et al., Nat Neurosci, 13:567-576, 2010).
한편, ATPase inhibitory factor 1 (IF1)은 84개 아미노산으로 이루어진 9.6-kDa basic 단백질이고, ATP5IF1 유전자에 의해 암호화된다. IF1은 미토콘드리아 내에서 ATP synthesis 합성 및 분해에 관여하는 F1Fo ATP synthase (multi-subunit, membrane-bound assembly)에 결합하게 된다. IF1은 심허혈 등과 같은 hypoxia시에 F1Fo ATP synthase에 결합, ATP 분해를 저해함으로써 ATP depletion을 막게 되고, 따라서 세포사멸을 억제하는 것으로 알려져 있다. 여러 문헌을 통해 미토콘드리아 기능저하를 병태생리로 하는 질환인 암이나 알쯔하이머병 등에서 IF1 과발현 (+F1Fo ATP synthase 발현 저하)이 관찰되지만, IF1을 외부적으로 주입하여 기능을 관찰한 보고는 없다.
본 발명자들은 한국인 남녀에서 나이가 많을수록 혈중 IF1의 농도가 통계적으로 유의하게 낮음을 관찰한 바 있으며, 이는 뇌 관련 질환이 다양하게 나타나는 노인층에서 IF1의 역할이 더욱 중요함을 보여주는 것이다.
이에, 본 발명자들은 부작용이 없고 효과가 우수하여 기존 치료제의 한계점을 보완할 수 있는 퇴행성 신경질환 치료제를 개발하고자 예의 노력한 결과, IF1 재조합 단백질 투여에 의해 세포내 Akt 신호전달경로가 조절되며, 혈중 세로토닌 (serotonin) 및 장내 GLP-1 (glucagon like peptide-1)의 농도가 증가하므로 퇴행성 신경질환 치료 효과가 있을 뿐만 아니라, 신경세포의 세포사멸 및 자가포식반응을 억제하는 동시에 미토콘드리아 기능성을 회복시켜 퇴행성 신경질환 동물모델의 기억력이 개선되는 것을 확인하고, 본 발명을 완성하게 되었다.
발명의 요약
본 발명의 목적은 Akt 경로 활성화, 혈중 세로토닌 증가 및 장내 GLP-1 분비 증가를 통해 신경가소성을 증가시키고, 뇌신경 퇴행을 억제하는 효과를 나타내며, 신경세포의 세포사멸 및 자가포식반응을 억제하며 미토콘드리아의 기능성을 회복시켜 퇴행성 신경질환 동물모델의 기억력과 운동 및 척수기능이 개선되는 효과를 가지는 IF1 (ATPase inhibitory factor 1)를 유효성분으로 함유하는 퇴행성 신경질환의 예방 또는 치료용 약학 조성물 및 식품을 제공하는데 있다.
상기 목적을 달성하기 위하여, 본 발명은 IF1 (ATPase inhibitory factor 1)을 유효성분으로 함유하는 퇴행성 신경질환의 예방 또는 치료용 약학 조성물을 제공한다.
본 발명은 또한, IF1 (ATPase inhibitory factor 1)을 유효성분으로 함유하는 퇴행성 신경질환의 예방 또는 개선용 식품을 제공한다.

발명의 상세한 설명 및 바람직한 구현예
다른 식으로 정의되지 않는 한, 본 명세서에서 사용된 모든 기술적 및 과학적 용어들은 본 발명이 속하는 기술 분야에서 숙련된 전문가에 의해서 통상적으로 이해되는 것과 동일한 의미를 갖는다. 일반적으로, 본 명세서에서 사용된 명명법은 본 기술 분야에서 잘 알려져 있고 통상적으로 사용되는 것이다.
ATPase inhibitory factor 1 (이하 IF1)은 84개 아미노산으로 이루어진 9.6-kDa basic 단백질이고, ATP5IF1 유전자에 의해 암호화된다. ATPase는 F0, F1 domain과 central, pheripheral stalk로 구성되며 여러 subunit으로 세분화된다. IF1은 ATPase의 기능을 방해하는 주요한 단백질로서 인체 내에서 자연적으로 생성되어 미토콘드리아에 의한 ATP 생성 및 분해를 조절하는 타겟으로서 많은 연구가 이루어져 있다 (Campanella et al., Cell Metab, 8:13-25, 2008). IF1의 inhibition 작용은 ATPase의 α subunit, β subunit 부위에 결합함으로써 회전운동을 방해하여 시작된다. IF1이 세포막 (plasma membrane)에 위치하는 F1-ATPase subunit과 결합하고, 이때 F1-ATPase 활성 조절로 ATP 가수분해가 억제됨으로 인해 세포외액의 ATP (extracellular ATP, exATP) 증가를 유발하며, exATP 는 세포막의 퓨린성 수용체와 반응한 후 퓨린성 신호전달을 통해 여러 가지 유용한 세포내 반응을 유발하게 된다.
하지만, IF1의 퇴행성 신경질환 관련 기작은 전혀 알려진 바가 없다.
이에, 본 발명에서는 마우스 IF1 전체 mRNA 서열 (NCBI No. NM_007512.3)을 바탕으로 GST-tag를 포함한 IF1의 DNA data를 클로닝을 통하여 재조합 단백질 (서열번호 1)을 생산하고, 재조합 IF1을 비만 마우스 모델에 투여하여 IF1에 의한 뇌조직내 Akt 신호전달경로 활성화, 혈중 세로토닌 및 장내 GLP-1의 농도 증가의 효과를 확인하였다. 또한, 파킨스병 및 치매 동물모델에서 IF1에 의한 척수기능 회복, 운동기능의 개선과 기억력 및 인지능력 개선 효과를 확인하였으며, 이는 마우스 일차배양세포 및 신경세포주에서 미토콘드리아 기능성의 회복과 신경세포 사멸 및 손상 억제을 추가로 확인하였다. 즉, ATPase inhibitory factor 1 (IF1)의 퇴행성 신경질환 예방 및 치료 효과를 입증하였다.
따라서, 본 발명은 일관점에서 IF1 (ATPase inhibitory factor 1)을 유효성분으로 함유하는 퇴행성 신경질환의 예방 또는 치료용 약학 조성물에 관한 것이다.
본 발명의 용어 "퇴행성 신경질환"은 신경세포가 퇴화하고, 기능을 잃고, 그리고 사멸하는 경우 나타나는 증상들과 관련된 질환을 의미하며, 알츠하이머, 파킨슨병, 크로이츠펠트-야콥병, 헌팅톤병, 다발성경화증, 루게릭병, 치매, 척수근위측증, 근위축성측색경화증, 니만-픽병, 뇌졸중, 뇌허혈 및 뇌출혈로 구성된 군에서 선택되는 것을 특징으로 할 수 있다.
IF1의 치료 효과는 다양한 원인에 의하여 발병되는 퇴행성 신경질환에 동등하게 적용될 수 있다.
본 발명의 약학 조성물은 상기 기재한 유효성분 이외에 약학적으로 허용 가능한 담체, 부형제 또는 희석제를 추가로 포함할 수 있다. 본 발명에서, 상기 약학 조성물에 포함될 수 있는 담체, 부형제 및 희석제로는 락토즈, 덱스트로즈, 수크로스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유를 들 수 있다. 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제된다.
본 발명의 약학 조성물은 각각 통상의 방법에 따라 산제, 과립제, 정제, 캡슐제, 현탁액, 에멀젼, 시럽, 에어로졸 등의 경구형 제형, 외용제, 좌제 또는 멸균 주사용액의 형태로 제형화하여 사용할 수 있다. 상세하게는, 제형화할 경우 통상 사용하는 충진제, 중량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제될 수 있다.
경구투여를 위한 고형 제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형제제는 적어도 하나 이상의 부형제 예를 들면, 전분, 칼슘카보네이트(calcium carbonate), 수크로스(sucrose) 또는 락토오스(lactose), 젤라틴 등을 섞어 조제된다. 또한 단순한 부형제 이외에 마그네슘 스티레이트, 탈크 같은 윤활제들도 사용된다.
경구를 위한 액상 제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는 데 흔히 사용되는 단순희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조 제제, 좌제가 포함된다. 비수성용제, 현탁제로는 프로필렌글리콜(propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다.
본 발명의 약학 조성물은 목적하는 방법에 따라 경구 투여하거나 비경구 투여 (예를 들어, 정맥 내, 피하, 복강내 또는 국소에 적용)할 수 있다.
의약 조성물은 멸균 주사용 수성 또는 유성 현탁액으로서 멸균 주사용 제제의 형태일 수 있다. 이 현탁액은 적합한 분산제 또는 습윤제(예, 트윈 80) 및 현탁화제를 사용하여 본 분야에 공지된 기술에 따라 제형될 수 있다. 멸균 주사용 제제는 또한 무독성의 비경구적으로 허용되는 희석제 또는 용매중의 멸균 주사용액 또는 현탁액(예, 1,3-부탄디올중의 용액)일 수 있다. 허용적으로 사용될 수 있는 비히클 및 용매로는 만니톨, 물, 링겔 용액 및 등장성 염화나트륨 용액이 있다. 또한, 멸균 불휘발성 오일이 통상적으로 용매 또는 현탁화 매질로서 사용된다. 이러한 목적을 위해, 합성 모노 또는 디글리세라이드를 포함하여 자극성이 적은 어떠한 불휘발성 오일도 사용할 수 있다. 올레산 및 이의 글리세라이드 유도체와 같은 지방산이 약제학적으로 허용되는 천연오일(예, 올리브유 또는 피마자유), 특히 이들의 폴리옥시에틸화된 것과 마찬가지로 주사 제제에 유용하다.

본 발명의 용어 "예방"이란, 본 발명에 따른 약학적 조성물의 투여로 퇴행성 신경질환의 발병을 억제 또는 지연시키는 모든 행위를 의미한다.
본 발명의 용어 "치료"란, 본 발명의 약학 조성물을 투여함으로써, 퇴행성 신경질환의 증세가 호전되거나 이롭게 변경시키는 모든 행위를 의미한다.
상기 퇴행성 신경질환 치료는 퇴행성 신경질환이 발생할 수 있는 임의의 포유 동물에 적용이 가능하며, 그 예로 인간 및 영장류뿐만 아니라, 소, 돼지, 양, 말, 개 및 고양이 등 가축을 제한없이 포함하나, 바람직하게는 인간일 수 있다.
본 발명에서 "투여"는 어떠한 적절한 방법으로 환자에게 소정의 물질을 도입하는 것을 의미하며, 상기 조성물들의 투여 경로는 약물이 목적 조직에 도달할 수 있는 한 어떠한 일반적인 경로를 통하여 투여될 수 있다. 복강내 투여, 정맥내 투여, 근육내 투여, 피하 투여, 피내 투여, 경구 투여, 국소 투여, 비강내 투여, 폐내 투여, 직장 내 투여 등이 될 수 있으나, 이에 제한되지는 않는다.
상기 본 발명의 약학 조성물은 약학적으로 유효한 양으로 투여될 수 있는데, 본 발명의 용어 "약제학적으로 유효한 양"이란 의학적 치료 또는 예방에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료 또는 예방하기에 충분한 양을 의미하며, 유효 용량 수준은 질환의 중증도, 약물의 활성, 환자의 연령, 체중, 건강, 성별, 환자의 약물에 대한 민감도, 사용된 본 발명의 조성물의 투여 시간, 투여 경로 및 배출 비율 치료기간, 사용된 본 발명의 조성물과 배합 또는 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다.
본 발명의 약학 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 순차적 또는 동시에 투여될 수 있다. 그리고 단일 또는 다중 투여될 수 있다. 상기 요소를 모두 고려하여 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하다.
본 발명의 약학조성물의 투여량은 사용목적, 질환의 중독도, 환자의 연령, 체중, 성별, 기왕력, 또는 유효성분으로서 사용되는 물질의 종류 등을 고려하여 당업자가 결정할 수 있다. 예를 들어, 본 발명의 약학 조성물을 사람을 포함하는 포유동물에 하루 동안 10 내지 100 ㎎/㎏, 보다 바람직하게는 10 내지 30 ㎎/㎏으로 투여할 수 있고, 본 발명의 조성물의 투여빈도는 특별히 이에 제한되지 않으나, 1일 1회 내지 3회 투여하거나 또는 용량을 분할하여 수회 투여할 수 있다.

IF1의 치료 효과는 다양한 원인에 의하여 발병되는 퇴행성 신경질환에 동등하게 적용될 수 있다.
본 발명의 식품 조성물을 식품 첨가물로 사용할 경우, 상기 조성물을 그대로 첨가하거나 다른 식품 또는 식품 성분과 함께 사용될 수 있고, 통상적인 방법에 따라 적절하게 사용될 수 있다. 일반적으로, 식품 또는 음료의 제조 시에 본 발명의 조성물은 원료에 대하여 15중량% 이하, 바람직하게는 10중량% 이하의 양으로 첨가된다. 그러나 건강 및 위생을 목적으로 하거나 또는 건강 조절을 목적으로 하는 장기간의 섭취의 경우에는 상기 범위 이하일 수 있으며, 안전성 면에서 아무런 문제가 없기 때문에 유효성분은 상기 범위 이상의 양으로도 사용될 수 있다.
본 발명의 식품은, 기능성 식품(functional food), 영양 보조제(nutritional supplement), 건강식품(health food) 및 식품 첨가제(food additives) 등의 모든 형태로 제조할 수 있다. 예를 들면, 건강식품으로는 본 발명의 조성물을 차, 쥬스 및 드링크의 형태로 제조하여 음용하도록 하거나, 과립화, 캡슐화 및 분말화하여 섭취할 수 있다. 또한, 기능성 식품으로는 음료(알콜성 음료 포함), 과실 및 그의 가공식품(예: 과일통조림, 병조림, 잼, 마말레이드 등), 어류, 육류 및 그 가공식품(예: 햄, 소시지, 콘비프 등), 빵류 및 면류(예: 우동, 메밀국수, 라면, 스파게티, 마카로니 등), 과즙, 각종 드링크, 쿠키, 엿, 유제품(예: 버터, 치츠 등), 식용식물유지, 마가린, 식물성 단백질, 레토르트 식품, 냉동식품, 각종 조미료(예: 된장, 간장, 소스 등) 등에 본 발명의 조성물을 첨가하여 제조할 수 있다.
상기 건강 기능식품 또한, 식품조성물로써 기능성 식품, 영양보조제, 건강식품, 식품 첨가제 등의 다양한 형태를 포함하는 것이며, 당업계에 공지된 통상적인 방법에 따라 다양한 형태, 예컨대, 앞서 언급한 본 발명의 조성물을 차, 쥬스, 드링크의 형태로 제조하거나, 과립화, 캡슐화, 분말화 하거나, 이러한 화합물 또는 추출물을 음료, 과실 및 가공식품, 어유, 육류 및 그 가공식품, 빵류, 면류, 조미료 등 각종 식품에 첨가하여 제조함으로써 제공될 수 있다.
본 발명의 건강음료 조성물은 통상의 음료와 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 상술한 천연 탄수화물은 포도당, 과당과 같은 모노사카라이드, 말토스, 슈크로스와 같은 디사카라이드, 및 덱스트린, 사이클로덱스트린과 같은 천연 감미제나, 사카린, 아스파르탐과 같은 합성 감미제 등을 사용할 수 있다. 상기 천연 탄수화물의 비율은 당업자의 선택에 의해 적절하게 결정될 수 있다.
상기 외에 본 발명의 조성물은 여러 가지 영양제, 비타민, 전해질, 풍미제, 착색제, 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알콜, 탄산 음료에 사용되는 탄산화제 등을 함유할 수 있다. 그 밖에 본 발명의 조성물은 천연 과일쥬스, 과일쥬스 음료 및 야채 음료의 제조를 위한 과육을 함유할 수 있다. 이러한 성분은 독립적으로 또는 조합하여 사용할 수 있다. 이러한 첨가제의 비율 또한 당업자에 의해 적절히 선택될 수 있다.
실시예
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 요지에 따라 본 발명의 범위가 이들 실시예에 의해 제한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.
실시예 1: 비만 동물모델
비만 마우스 모델로는 C57BL/6J 수컷을 사용하였으며, JOONGAH BIO에서 5주령의 마우스 40마리를 분양받아 1주간 보통식이를 제공하며 적응기간을 거쳤다. 사육환경으로는 18~24℃, 50~60%의 습도를 항시 유지하였으며, 조명을 통해 일주기를 조절하고, 적응기간 및 실험기간 모두 자유급식을 이행하였다.
1주일 적응기간이 끝난 후, 3개의 그룹으로 나누어 각각 다음 같은 조건으로 6주간 사육하였다.
그룹 1: 정상식이(ND) 대조군 (n=10)
그룹 2: 고지방식이(HFD) 대조군 (n=15)
그룹 3: 고지방식이(HFD) 실험군 (n=15)
1주일 적응기간이 끝난 마우스들은 고지방식이로 6주 동안 비만을 유도한 다음, IF1을 투여하였다.
실시예 2: 비만 동물모델에서 IF1 투여에 따른 Akt 경로 활성화
적응기간 1주 이후 6주 동안 지속적인 비만을 유도한 마우스에 IF1 (5mg/kg BW)을 이후 4주 동안 주당 4회 복강주사 하였다. IF1은 GST-tag를 포함한 IF1의 DNA data를 클로닝을 통하여 분리, 정제하여 재조합 단백질을 생산하였다.
실시예 1의 마우스 3가지 그룹에 IF1를 투여하는 조건은 다음과 같다 (표2).
IF1 투여 4주까지 모든 실험 및 사육기간이 모두 종료된 마우스는 해부를 위해 16시간 절식시키고, 다음날 isopropanol을 이용한 호흡마취를 진행하였다. 마취 여부는 다리부분의 반사작용을 통해 검증하였으며, 마취가 이루어지면 인슐린 시린지 (syringe)를 통해 채혈하였다. 채혈이 완료된 마우스는 해부를 통해 whole brain과 intestine을 분리하여 빠르게 PBS에 담가 표면에 묻은 이물질을 제거하였다.
적출한 whole brain은 lysis buffer tissue homogenate를 이용하여 단백질 발현의 정량을 위해 웨스턴블랏을 수행하였다. 각 샘플의 단백질량을 동일하게 정량하여 SDS-PAGE를 시행하였다. 각각의 gel을 nitrocellulose membrane (Immobilon-P Transfer Membranes, Millipore Corporation, Billerica, Massachusetts)로 Bio-rad transfer 기구를 사용하여 전이시켰다. Blocking 후, 각 membrane을 5% BSA buffer (TBS-T, BSA, 0.02% Na-azaid)로 1차, 2차 항체 반응 후 FluorChem system (Alpha Innotech, U.S.A)을 사용하여 Akt 단백질의 발현 및 인산화 정도를 사진 찍고 정량화하였다.
모든 자료는 SPSS-PC+ 통계 package를 사용하여 처리하였다. 각 항목에 따라 백분율과 평균치±표준오차(SE)를 구하고, 군별 유의성 검증을 위해 1차원 분산분석(ANOVA)을 수행하여, 던컨의 다중검정 (Duncan's multiple range test)을 이용하여 유의수준 5% 안에서 검증하였다.
그 결과, HFD 대조군에 비하여 IF1을 투여한 그룹의 뇌조직 내 Akt 인산화 경로가 활성화되어 있고 통계적으로 유의성을 보였다 (도 1).
실시예 3: 비만 동물모델에서 IF1 투여에 따른 혈중 세로토닌 증가
실시예 2에서 채혈로 얻어진 혈액은 즉시 8000rpm으로 10분간 4℃에서 원심분리하여 혈장을 분리하였다. 그 다음 ELISA 분석을 통해 혈중 세로토닌 농도를 분석하였다.
모든 자료는 SPSS-PC+ 통계 package를 사용하여 처리하였다. 각 항목에 따라 백분율과 평균치±표준오차(SE)를 구하고, 군별 유의성 검증을 위해 1차원 분산분석(ANOVA)을 수행하여, 던컨의 다중검정 (Duncan's multiple range test)을 이용하여 유의수준 5% 안에서 검증하였다.
그 결과, HFD 대조군에 비하여 IF1을 투여한 그룹의 혈중 세로토닌 농도가 통계적으로 유의하게 증가되어 있음을 확인하였다 (도 2).
실시예 4: 비만 동물모델에서 IF1 투여에 따른 장내 GLP-1 분비 증가
실시예 2에서 분리한 intestine은 homogenate를 이용하여 GLP-1 (glucagon like peptide-1) ELISA 분석을 수행하였다.
모든 자료는 SPSS-PC+ 통계 package를 사용하여 처리하였다. 각 항목에 따라 백분율과 평균치±표준오차(SE)를 구하고, 군별 유의성 검증을 위해 1차원 분산분석(ANOVA)을 수행하여, 던컨의 다중검정 (Duncan's multiple range test)을 이용하여 유의수준 5% 안에서 검증하였다.
그 결과, HFD 대조군에 비하여 IF1을 투여한 그룹 동물의 장조직내 GLP-1 분비가 통계적으로 유의하게 증가되어 있음을 확인하였다 (도 3). 이는 IF1 처리가 생체내에서 GLP-1 minetics로 활용됨을 보여주는 것으로, 알쯔하이머, 파킨슨 질환과 뇌졸중 등 기타 퇴행성 신경질환의 치료제로 활용될 수 있음을 보여주는 것이다.
실시예 5: 퇴행성 신경계질환 동물모델
5-1: 파킨슨병 동물 모델
파킨슨 질병 동물 모델의 경우 20주령의 암컷 C57BL/6 트랜스제닉 마우스를 식품의약품안전평가원에서 분양받아 실험에 사용하였다 (등록번호 18-NIFDS-M-NE-014). Tg 마우스는 C57BL/6-Tg(NSE-hαSyn)Korl 모델로 파킨슨 질환의 원인물질인 알파 시누클레인 (alpha synuclein)을 뇌조직에서만 과발현하도록 변형시킨 마우스이다. 마우스는 실험 전 1주간의 적응기간을 거친 후, 18~24℃, 50~60%의 온도 및 습도가 유지된 상태에서 자유급식을 통해 사육하였다.
적응기간이 끝난 후, 2가지 그룹으로 나뉘어 각각 다음의 조건으로 사육하였다.
그룹 1: GST 대조군 (n=5)
그룹 2: IF1 실험군 (n=5)
5-2: 치매 동물 모델
치매 질병 동물 모델의 경우 25주령의 암/수 C57BL/6-Tg(NSE-hAPPsw)Korl 마우스 20마리를 식품의약품안전평가원에서 분양받아 실험에 사용하였다 (등록번호 18-NIFDS-M-NE-004). 상기 동물은 아밀로이드 전구 단백질의 아미노산 잔기에 변형을 주고(Lys670 -> Asn, Met671 -> Leu) NSE 프로모터를 결합한 뇌 특이적 치매 질병 모델이다. 1주간의 적응기간 이후, 18~24℃, 50~60%의 온도 및 습도가 유지된 상태에서 자유급식을 통해 사육하였다.
적응기간이 끝난 후, 4가지 그룹으로 나뉘어 각각 다음의 조건으로 사육하였다.
그룹 1: GST 암컷 대조군 (n=2)
그룹 2: IF1 암컷 실험군 (n=3)
그룹 3: GST 수컷 대조군 (n=3)
그룹 4: IF1 수컷 실험군 (n=4)
5-3: IF1 투여
적응기간 1주 이후, 나누어진 그룹에 따라 각 마우스에 단백질을 총 9주간 투여하였다. 단백질의 투여는 인슐린 시린지 (syringe)로 주 3회 복강투여 하였으며, 1회당 각 GST(7.5mg/kg BW), IF1(7.5mg/kg BW) 농도로 총 1주간 각 22.5mg/kg BW의 단백질을 사용하였다. IF1은 GST-tag를 포함하며, 각 단백질은 pGEX vector를 이용한 유전자 재조합과, 친화성 크로마토그래피 방법을 통해 분리, 정제하여 사용하였다.
실시예 6: 행동실험
6-1 : Hanging test & pole test
파킨슨 질병 동물 모델을 대상으로 근력을 측정하여, 근육의 기능회복 및 강화를 확인하였다. 근력의 측정은 Hanging test를 통해 진행하였으며, pole test를 통해 근육 기능 및 균형의 개선을 확인하였다.
Hanging test는 1cm x 1cm 크기의 철망으로 이루어진 장치에 마우스가 매달린 후, 떨어지기 까지의 시간을 측정하였으며, impulse는 무게를 곱하여 보정한 지표이다.
그 결과 hanging time과 holding impulse 모두 IF1 처리 그룹에서 유의하게 증가함을 확인할 수 있었다 (도 4).
Pole test는 1cm의 직경을 가진 55cm길이의 봉에 거친 천을 표면에 덧씌워 실험에 사용하였다. 각 마우스는 천장을 바라본 상태로 봉의 윗부분에 매달린 상태에서 실험에 참여한다. 이때, 마우스가 아래로 향하기까지의 시간 (turn time) 및 바닥에 닿을 때까지의 시간 (descent time)을 측정하여 총 시간을 구해 비교한다. 실험은 1회 총 5번의 trial로 이루어 지며, 총 2회 진행한 값의 평균을 내어 비교하였다.
그 결과, IF1을 처리한 그룹에서 유의한 차이를 보여 근육 기능 및 균형의 개선 효과를 확인하였다 (도 5).
6-2 : Limb clasping test
척수기능의 회복 및 강화를 확인하기 위해, limb clasping test를 진행하였다. 실험자에 의해 각 마우스는 꼬리를 통해 거꾸로 매달리게 되며, 카메라를 통해 10초간 공중에서 마우스의 움직임을 기록한다. 실험이 모두 끝난 후, 2명의 실험원이 표 3 (Limb clasping 점수 측정표)의 측정기준에 의거해 각각 점수를 기록하였다. 이때, 마우스별로 점수를 비교하여 1점 이상 차이가 나는 개체는 다시 점수를 매겨 결과를 낸다.
그 결과, 대조군에 비해 IF1 처리 그룹에서 55.5% 수준의 낮은 점수를 기록하였다. 이를 통해 파킨슨 모델에서 IF1에 의한 척수기능의 회복 및 강화를 확인할 수 있었다.
6-3 : Y maze test
Y maze 실험을 통해, 치매 모델에서 공간기억력 개선효과를 확인하였다. 실험은 3개의 동일한 길이의 팔로 이루어진 Y자 모양의 미로 장치를 통해 시행하였다. 각 실험의 전후에 장치는 70% 알코올과 증류수를 통해 세척하여 이물질을 제거하였으며, 고정된 카메라를 통해 전 실험과정을 기록하였다. 실험을 위해 마우스는 미로장치의 중앙을 바라보는 자세로 옮겨주며, 8분간 자유롭게 이동하게 된다. 이때, 실험자는 멀리 떨어진 곳에 위치하여 마우스가 실험자를 인식하지 못하게 하여 진행하였다. 모든 실험이 종료된 이후, 각 방향의 팔에 들어간 횟수와 자발적 교차행동량 (spontaneous alteration) 점수를 측정하여 그 정도를 비교하였다.
그 결과, IF1 그룹에서 유의한 차이를 보였고, 치매모델에서 IF1의 공간기억력 개선효과를 확인하였다 (도 6).
6-4 : Novel object recognition test (신물질탐색 시험)
치매모델을 대상으로, NORT (Novel object recognition test, 신물질탐색 시험)을 진행하여 단기기억력의 개선효과를 확인하였다. NORT 실험은 천장이 개방된 55 x 55 x 30 cm의 불투명한 장치에서 영상녹화와 함께 진행하였다. 본 실험 하루 전, 각 마우스는 30분간 실험장치에서 적응기를 가지게 된다. 본 실험의 경우 두가지 단계로 나뉘며, 첫 번째 단계(T1)의 경우 두 개의 같은 물체를 장치 정중앙 기준 북동, 남서방향에 각각 고정시켜 10분간 탐색시킨다. 실험 종료 두시간 후, 두 번째 단계(T2)를 진행한다. T2에서는 T1에서 사용한 물체 및 이와 비슷한 복잡도(크기, 모양, 색감, 밝기)를 지닌 새로운 물체를 이용한다. 마찬가지로 10분간 각 마우스는 자유롭게 물체를 탐색하게 되며, 실험 종료 후 영상분석을 통해 그룹간 차이를 비교한다.
그 결과, IF1 처리그룹에서 암/수컷 모두 새로운 물체에 대한 변별력 지수 (discrimination index) 와 인식력 지수 (recognition index)에서 유의한 증가를 보여 단기기억력의 개선 효과를 확인할 수 있었다 (도 7 및 도 8).
6-5 : Morris water maze test
치매모델에서 IF1의 장기 기억능력 개선효과를 확인하기 위해, morris water maze test (수중미로 실험)를 진행하였다. Morris water maze test는 원형의 수조에 물을 채운 후, 이보다 2cm가량 높은 platform을 정 가운데에 설치하여 진행하였다. 각 마우스는 실험을 위해 platform에 옮겨져 20초간 방치한다. 이때, 마우스가 수조로 뛰어들거나 점프하는 행동을 보일 시 다시 platform으로 위치시켰다. 20초가 지난 후, 무작위 방향의 수조 벽을 향해 꼬리부터 마우스를 입수시킨다. 마우스가 platform에 올라온 시점에 실험을 종료하며, 1분 내에 성공하지 못할 시 platform에 즉시 올려주어 다시 20초간 방치하였다. 이를 하루 3번씩 4일간 반복하여, 마우스가 platform을 기억하도록 훈련하였다. 본 실험 당일, 수조의 물에 탈지분유를 넣는 동시에 platform보다 수위를 2cm가량 높여 마우스가 platform을 보지 못하게 조성하였다. 실험은 훈련과 마찬가지로 벽으로부터 시작하여 보이지 않는 platform에 올라가는 시점에 종료하며, 1분간 성공하지 못한 개체는 최대값인 1분으로 기록하였다.
그 결과, IF1 처리시, 암컷에서 43.3% 감소된 시간을 나타내 장기 기억능력 개선 효과를 보였다.
실시예 7: 신경세포주에서 IF1의 기능
7-1 : IF1 처리에 의한 도파민성 신경 세포사멸의 억제
파킨슨병에서 특이적으로 사멸하는 도파민성 신경세포의 사멸과정에 미치는 IF1의 기능을 확인하기 위하여 도파민성 신경세포주인 MN9D에 신경독소 (paraquat)를 처리한 모델을 사용하였다. 24 well의 각 well 마다 40000개의 세포를 깔고 3일이 지난 후에 신경독소 (paraquat)를 2NM, 5nM, 10Nm의 세가지 다른 농도의 IF1과 함께 또는 단독으로 16시간 처리하였다. 처리 후 살아있는 세포를 양을 측정하여 세포의 사멸정도의 변화를 평가하는 MTT 분석법을 사용하였다.
그 결과, IF1은 신경독소 (paraquat)에 의한 세포사멸을 억제하였으며, 가장 높은 농도인 10nM처리 하였을 때 세포의 생존력이 가장 높은 것으로 나타났다 (도 9).
7-2: IF1 처리에 의한 세포막 파괴 억제
PI (Propidium iodide)는 세포막을 투과하지 못하는 색소로써 세포막이 손상되는 necrotic 또는 후기 apoptotic cell death에 의한 세포의 사멸을 평가하기 위해 사용된다.
따라서, 도파민성 신경세포주에 신경독소 (paraquat)를 처리한 모델에 다양한 농도의 IF1 (5nM, 10nM)을 처리하였다.
그 결과, PI에 의한 염색 정도에 차이를 보이지 않아 IF1이 세포막을 파괴하지 않는 것을 관찰하였다. 동시에 신경독소 (paraquat)를 처리한 세포에서는 세포사멸이 진행되면서 세포막의 투과도가 증가하여 PI에 의해 염색되는 세포의 수가 증가하는 것이 관찰되었다. 반면에 신경독소 (paraquat)와 IF1을 함께 처리했을 때에는 신경독소 (paraquat) 단독처리에 비해 PI로 염색되는 세포의 수가 현저히 감소하였으며, 이를 통해 IF1에 의해 세포사멸이 억제됨을 확인할 수 있었다 (도 10).
7-3: 세포 자가포식 (autophagy)의 측정
IF1이 신경계 세포에 미치는 영향을 알아보고자, 자가포식 (autophagy)의 활성을 나타내는 마커인 Lc3b를 형광 단백질로 표지한 GFP-Lc3b를 도파민성 신경세포주인 MN9D세포에 형질 주입하고, 신경독소 (paraquat)를 단독 또는 IF1와 함께 처리하여 자가포식의 변화를 분석하였다.
그 결과, 대조군에 비하여 신경독소 (paraquat)를 처리한 군에서는 Lc3b 단백질이 뭉쳐서 나타나는 반점과 같은 형태가 증가되는 것을 확인할 수 있었으며, 이는 비정상적으로 증가된 자가포식을 나타낸다고 볼 수 있다. 한편 신경독소와 IF1을 함께 처리한 군에서는 신경독소 단독처리 군에 비하여 Lc3b 반점이 현저히 감소하는 것을 확인할 수 있었다. 이를 통해 IF1가 신경독소에 의해 형성된 자가포식의 활성화를 억제하는 기능을 가지는 것으로 유추할 수 있다 (도 11).
7-4: 세포 아폽토시스 (apoptosis)의 측정
IF1이 신경계 세포 아폽토시스 (apoptosis)에 미치는 영향을 알아보고자, 아폽토시스를 일으키는데 주요한 효소인 caspase-3의 염색을 진행하였다. 24 well에 PDL로 코팅된 유리를 놓고 그 위에 well당 30000개씩 도파민성 신경세포주인 MN9D 세포를 배양한 후 3일이 지났을 때 신경독소를 단독 또는 IF1과 함께 처리하고 16시간이 지나면 1% 파라포름알데하이드로 고정하였다. 고정시킨 세포를 BSA로 blocking한 후에 caspase-3 항체로 형광면역반응을 시켰으며 핵은 Topro3로 염색하였다.
그 결과, 신경독소를 처리한 세포군에서는 거의 대부분의 세포가 형광을 표지하는 것에 반해서 신경독소와 IF1을 함께 처리한 세포군에서는 형광표지가 현저하게 감소한 것를 확인할 수 있었다. 따라서 신경독소에 의해 일어나는 아폽토시스 과정을 매개하는 caspase-3의 활성이 IF1을 처리함으로써 감소됨을 알 수 있다 (도 12).
실시예 8: 일차배양 세포에서 IF1의 기능
8-1: 마우스 일차배양 중뇌세포에서 IF1의 세포사멸 억제
마우스의 중뇌 세포에서 IF1이 미치는 영향을 알아보고자 다음과 같이 실험을 진행하였다.
임신 후 14일 지난 마우스 태아의 중뇌를 해부하여 일차 세포를 배양한 후에 신경독소 (paraquat)를 단독 또는 IF1과 함께 처리하고 고정하였다. 그 다음 세포를 도파민성 신경세포의 마커인 tyrosine hydroxylase (TH)에 대한 항체로 형광염색한 후 분석하였다.
TH에 염색된 세포의 수를 정량한 결과, 대조군에 비해 신경독소 (paraquat)를 처리한 군에서 그 수가 감소하는 것으로 나타났다. 이는 도파민성 신경세포가 사멸한 것임을 알 수 있다. 한편 신경독소와 IF1을 함께 처리하면 TH에 염색된 세포의 수가 회복되므로, 도파민성 신경세포의 사멸을 IF1이 억제하는 것을 일차배양 세포에서도 확인할 수 있었다 (도 13).
8-2: 마우스 일차배양 중뇌세포에서 IF1의 신경돌기 손상 억제
실시예 8-1과 동일하게 마우스의 중뇌의 도파민성 일차배양 세포를 배양한 후에 신경독소 (paraquat)를 단독 또는 IF1과 함께 처리하였다.
그 다음 일차배양 세포에서 신경돌기(neurite)를 조사한 결과, 신경독소 (paraquat)를 처리하면 가장 긴 돌기의 길이가 대조군에 비해 현저히 짧아짐을 확인하였다. 하지만 IF1을 신경독소와 함께 처리한 군에서는 신경돌기의 길이가 회복됨을 알 수 있었다. 이것으로부터 IF1이 도파민성 신경세포의 신경돌기 손상을 억제하는 기능을 나타냄을 알 수 있었다 (도 14).
8-3: 알츠하이머 마우스의 일차배양 해마세포에서 IF1의 기능 측정
알츠하이머 마우스의 해마에서 IF1이 미치는 영향을 알아보고자 다음과 같이 실험을 진행하였다.
임신 후 18-21일 지난 마우스 태아의 해마를 해부하여 일차 세포를 배양한 후에 IF1을 처리하고 고정하였다. 그 다음 세포에 미토콘드리아를 염색하는 mitotracker를 사용하여 형광염색한 후 분석하였다.
그 결과, 5xFAD 알츠하이머 모델 마우스의 일차배양 해마 세포의 미토콘드리아 평균 길이가 대조군에 비해 감소한 것을 알 수 있었다. 여기에 IF1을 처리하면 5xFAD 모델 마우스의 해마세포에서 줄어든 미토콘드리아의 길이가 대조군과 비슷한 수준으로 늘어나는 것을 확인하였으며, 이로부터 5xFAD 모델 마우스의 미토콘드리아 이상을 IF1이 회복시키는 것을 알 수 있었다 (도 15).
본 발명에 따른 IF1은 비만이 유발된 마우스에서 부작용 없이 세포내 Akt 신호전달경로 활성화, 혈중 세로토닌 및 장내 GLP-1의 농도 증가의 효과를 나타낼 뿐만 아니라, 신경세포의 세포사멸 및 자가포식 반응의 억제와 동시에 미토콘드리아의 기능을 회복시켜 퇴행성 신경질환 동물모델의 기억력과 운동 및 척수기능이 개선되므로, 치매, 파킨슨병, 헌팅톤, 우울증, 알쯔하이머병, 뇌졸중과 심혈관질환 등과 같은 여러 노인성, 퇴행성 신경질환 치료제로 매우 유용하다.
이상으로 본 발명 내용의 특정한 부분을 상세히 기술하였는 바, 당업계의 통상의 지식을 가진 자에게 있어서, 이러한 구체적 기술은 단지 바람직한 실시양태일 뿐이며, 이에 의해 본 발명의 범위가 제한되는 것이 아닌 점은 명백할 것이다. 따라서 본 발명의 실질적인 범위는 첨부된 청구항들과 그것들의 등가물에 의하여 정의된다고 할 것이다.

Claims (8)
- IF1 (ATPase inhibitory factor 1)을 유효성분으로 함유하는 퇴행성 신경질환의 예방 또는 치료용 약학 조성물.
- 제1항에 있어서, 상기 퇴행성 신경질환은 알츠하이머, 파킨슨병, 크로이츠펠트-야콥병, 헌팅톤병, 다발성경화증, 루게릭병, 치매, 척수근위측증, 근위축성측색경화증, 니만-픽병, 뇌졸중, 뇌허혈 및 뇌출혈로 구성된 군에서 선택되는 것을 특징으로 하는 약학 조성물.
- 제1항에 있어서, 상기 IF1은 신경세포의 세포사멸, 세포막 파괴, 자가포식 활성화, 아폽토시스, 신경돌기 손상 또는 미토콘드리아 이상을 억제하는 것을 특징으로 하는 약학 조성물.
- 제1항에 있어서, 약학적으로 허용 가능한 담체, 부형제 또는 희석제를 추가로 포함하는 것을 특징으로 하는 약학 조성물.
- IF1 (ATPase inhibitory factor 1)을 유효성분으로 함유하는 퇴행성 신경질환의 예방 또는 개선용 식품.
- 제5항에 있어서, 상기 퇴행성 신경질환은 알츠하이머, 파킨슨병, 크로이츠펠트-야콥병, 헌팅톤병, 다발성경화증, 루게릭병, 치매, 척수근위측증, 근위축성측색경화증, 니만-픽병, 뇌졸중, 뇌허혈 및 뇌출혈로 구성된 군에서 선택되는 것을 특징으로 하는 식품.
- 제5항에 있어서, 상기 IF1은 신경세포의 세포사멸, 세포막 파괴, 자가포식 활성화, 아폽토시스, 신경돌기 손상 또는 미토콘드리아 이상을 억제하는 것을 특징으로 하는 식품.
- 제5항에 있어서, 식품학적으로 허용 가능한 식품보조 첨가제를 추가로 포함하는 것을 특징으로 하는 식품.
