Abstract
본 발명은 체내에서 항원으로써 항-RANKL 항체의 생성을 유도하여 파골세포의 생성을 억제할 수 있고, RANK에 결합하면서도 파골세포의 분화를 유도하지 않는 RANKL 단백질의 돌연변이체 및 이를 유효성분을 포함하는, 골대사 질환을 예방 또는 치료하기 위한 조성물의 제공에 관한 것이다.
Description
RANKL의 돌연변이체, 및 이를 포함하는 골다공증 예방 또는 치료용 약학 조성물에 관한 것이다.
골다공증은 골량과 골질이 감소하여 골밀도(Bone mineral density : BMD)가 2.5 이하이거나 T-score(일반적인 성인의 평균 골질량과의 표준편차)가 -2.5이하인 경우로 뼈가 약해져 골절이 일어나기 쉬운 상태를 말한다. 골밀도가 과다하게 감소하게 되면 작은 충격에도 쉽게 골절이 생기게 된다. 골다공증은 그 증세 자체보다는 뼈의 약화에 따라 초래되는 각종 골절, 특히 대퇴골 골절 또는 척추골절 등으로 장기간 활동을 제한하여 건강한 생활을 영위할 수 없고, 결과적으로 노인층 사망의 15%에 대한 원인이 되는 것으로 알려져 있다.
인간의 뼈는 조골세포(osteoblast), 골세포(osteocyte), 파골세포(osteoclast)로 이루어져 있다. 그 중 조골세포는 증식기, 골 기질 형성기, 석회화기를 거쳐 골조직을 형성하는 역할을 수행한다. 또한 파골세포는 골을 흡수하는 역할을 수행한다. 성장이 끝난 성인의 뼈는 파골세포에 의해 오래된 뼈는 제거하고 조골세포에 의해 새로운 뼈로 대체하는 골 흡수와 생성을 지속적으로 반복 재생하면서 골재형성 과정이 일어난다. 예를 들어 조골세포는 NF-κB의 수용체 활성물질 리간드(receptor activator of Nuclear factor-kappa B ligand : RANKL) 및 그의 교란 수용체인 오스테로프로테게린(osteoprotegerin : OPG)과 같은 물질의 분비를 통해 골흡수를 담당하는 파골세포의 분화를 조절함으로써 체내의 골대사의 항상성을 유지한다. 상기 골대사의 항상성이 특정 원인에 의해 무너지면 골다공증, 골형성 장애 또는 골절 등과 같은 골대사 질환이 발생하게 된다.
상기한 바와 같이, 파골세포는 뼈 관련 질환에서 골 흡수에 관여하며, Denosumab과 같은 항-사이토카인 항체가 골다공증 치료에 있어서 효과적으로 알려져 있다. 그러나, 고비용의 제조원가 및 항체의 다중 투여로 인한 면역원성의 발생은 이러한 항-사이토카인 면역 치료에 있어서 큰 문제로 남아 있다. 이러한 면역요법의 내성과 항-사이토카인 항체 사용 요법의 문제를 극복하기 위해 본 발명은 항-사이토카인 항체를 유도하기 위한 면역원으로서 RANKL의 돌연변이체를 골다공증에 적용하기 위한 면역요법을 제안한다.

용어 'RANKL 단백질'은 NF-κB의 수용체 활성물질 리간드(receptor activator of Nuclear factor-kappa B ligand : RANKL)를 의미할 수 있다. RANKL은 유형 II 막 단백질로 알려져 있으며 종양 괴사 인자(Tumor ncerosis factor : TNF) 슈퍼 패밀리의 구성원이다. RANKL은 괴사 조절 유전자이며 Id4, Id2 및 cyclin D1의 단백질 수준을 조정하여 세포 증식을 조절할 수 있다. RANKL은 골격근, 흉선, 간장, 결장, 소장, 부신 동맥, 조골 세포, 유선 상피 세포, 전립선 및 췌장을 비롯한 여러 조직 및 기관에서 발현될 수 있다.
NF-κB는 염증반응 조절, 면역체계 조절, 세포고사(apoptosis), 세포증식, 상피세포의 분화 등에 관여하는 단백질군이다. NF-κB는 다양한 유전자들의 발현을 조절하며 세포내의 신호전달 체계의 중심축을 이룰 수 있다. RANKL가 NF-κB의 수용체 활성물질(Receptor activator of nuclear factor kappa-B : RANK)에 결합하면, RANK가 활성화되어, NF-κB, 미토겐-활성 단백질 키나제(Mitogen-activated protein kinase:MAPK), 활성화 단백질 1(AP-1) 및 활성화된 T 세포의 핵인자(NFATc1)를 단계적으로 활성화시킬 수 있다.
‘돌연변이체’란 돌연변이가 일어난 개체 내지 단백질을 의미할 수 있다. ‘돌연변이’란 유전정보가 기록된 DNA분자가 원본과 달라지는 것을 의미할 수 있다. 돌연변이가 일어나면 그 유전자에 의해 생산되는 단백질에 변화가 생기고, 이는 유전형질의 변화를 불러올 수 있다.
돌연변이의 종류는 그것이 작용하는 방식과 규모에 따라 구분될 수 있다. 돌연변이에는 뉴클레오타이드 수준에서 일어나는 돌연변이로 하나의 뉴클레오타이드가 변환되어 나타나는 ‘점(point) 돌연변이’, 뉴클레오타이드의 일부가 원래의 염기 서열 사이로 삽입되어 일어나는 돌연 변이인 ‘삽입 돌연변이’, 원래 있던 뉴클레오타이드의 일부가 사라져 버린 돌연변이인 ‘결실 돌연변이’, 및 염색체 수준에서 일어나는 돌연변이인 ‘유전자 중복’, ‘유전자 결실’, ‘염색체 역위’, ‘간질성 결실’, ‘염색체 전위’, 및 ‘이형접합 소실’ 등이 있을 수 있다.
상기 RANKL 단백질은 아미노산 서열 중 하나 이상의 아미노산이 치환된 돌연변이체일 수 있다.
다른 일 양상은 서열번호 1의 아미노산 서열을 포함하는 RANKL 단백질 N-말단에서, 180번째의 리신(Lys)이 아르기닌(Arg)으로, 189번째의 아스파르산(Asp)이 이소루신(Ile), 루신(Leu) 및 아스파라긴(Asn) 중 어느 하나로, 190번째의 아르기닌(Arg)이 리신(Lys)으로, 223번째의 히스티딘(His)이 페닐알라닌(Phe) 또는 티로신(Try)으로, 및 224번째의 히스티딘(His)이 페닐알라닌(Phe) 또는 티로신(Try)으로의 치환으로 이루어진 군으로부터 선택된 하나 이상의 치환을 갖는 돌연변이체를 제공한다.

서열번호 1은 마우스(Mus musculus)의 RANKL 단백질(mRANKL)의 아미노산 서열이다. 상기 RANKL 단백질의 아미노산 치환 부위는 RANK의 수용체와 결합하는 부위와 관련될 수 있다.
서열번호 2는 인간(Homo sapiens)의 RANKL 단백질(hRANKL)의 아미노산 서열이다. 인간의 RANKL 단백질의 아미노산 서열은 전장 317로, 전장 316인 마우스의 아미노산 서열보다 1개의 아미노산을 추가로 포함하고 있고, 치환되는 RANKL 부위는 마우스의 아미노산 서열보다 한자리 뒤에 위치할 수 있다. 상기 치환되는 부위의 아미노산은 서로 동일할 수 있다.
상기 돌연변이체는 개체에 투여되어 항체를 생성하는 것일 수 있다.
따라서, 본 명세서는 RANKL 단백질의 돌연변이체 의해서 생성된 항체를 추가적으로 제공한다.
항체(immunoglobulin)는 항원과 특이적 결합을 하여 항원-항체 반응을 일으키는 물질일 수 있다. 항체는 폴리클로날 항체, 모노클로날 항체, 미니바디(minibody), 도메인 항체, 이중특이적 항체, 항체 모방체, 키메라 항체, 항체 접합체(conjugate), 인간항체 또는 인간화 항체이거나 이의 단편 중 어느 하나일 수 있다.
상기 돌연변이체는 개체에 투여된 후, 개체 내 항-RANKL 항체(anti-RANKL antibody)의 형성을 유도하는 것일 수 있다. 즉, 상기 돌연변이체는 능동면역을 위한 면역원으로서 작용하는 물질인 항원(antigen)일 수 있다. 생성된 항체는 개체 내 RANKL과 항원-항체반응을 할 수 있다. 항체에 의해 개체 내 RANKL의 농도가 감소하면, RANKL과 RANK의 결합이 줄어들어, 파골세포(osteoclast)의 분화(differentiation)를 억제할 수 있다.

일 구체예에서, 상기 RANKL의 돌연변이체는 RANKL 단백질의 아미노산을 암호화하는 핵산에 점 돌연변이가 발생한 것일 수 있다. ‘점(point) 돌연변이’는 뉴클레오타이드 수준에서 일어나는 돌연변이로, 하나의 뉴클레오타이드가 변환되어 DNA 전사 단계에서 특정 단백질의 생성을 막거나 변형시킬 수 있다. 점 돌연변이는, 변이된 코돈이 기존의 코돈과 같은 아미노산 형성을 지시하는 ‘불현성 돌연변이’, 변이된 코돈이 다른 아미노산 형성을 지시하는 ‘미스센스 돌연변이’, 및 변이된 코돈으로 인해 아미노산 형성이 중단되거나 생략되는 ‘넌센스 돌연변이’와 같은 효과를 나타낼 수 있다.
상기 RANKL 돌연변이체를 암호화하는 핵산 분자는 서열번호 3 또는 4의 뉴클레오티드 서열을 포함하는 것일 수 있다.
서얼번호 3의 뉴클레오티드 서열의 경우, 기존 마우스의 RANKL을 암호화하는 핵산의 뉴클레오티드 서열 중, 676번째 내지 677 번째의 뉴클레오티드가 AA에서 CG으로, 674번째 내지 679번째의 뉴클레오티드가 GATCGA에서 ATCAAG로, 803번째, 804번째 및 806번째의 뉴클레오티드가 CAC에서 TTT로의 치환으로 이루어지는 군으로부터 선택된 하나 이상의 치환일 수 있다.
서얼번호 4의 뉴클레오티드 서열의 경우, 기존 인간의 RANKL을 암호화하는 핵산의 뉴클레오티드 서열 중, 669번째 내지 670 번째의 뉴클레오티드가 AA에서 CG으로, 696번째 내지 671번째의 뉴클레오티드가 GATCG에서 ATCAA로, 798번째, 799번째 및 801번째의 뉴클레오티드가 CAC에서 TTT 로의 치환으로 이루어지는 군으로부터 선택된 하나 이상의 치환일 수 있다.
일 구체예에서 상기 RANKL 단백질을 암호화하는 핵산의 점돌연변이는 메가프라이머(megaprimer)에 의해 유도될 수 있다. 메가프라이머 방법은 PCR 방법의 일종으로 돌연변이를 유도하고자 하는 위치가 단백질의 중간 부분에 있어서 한 번에 돌연변이를 완성할 수 없는 경우, 2단계에 걸쳐 PCR을 진행하여 돌연변이를 완성할 때 사용하는 방법이다. 구체적으로, 유발하고자 하는 뉴클레오티드를 포함한 정방향 프라이머(primer)와 단백질 C-말단의 뉴클레오티드를 포함하는 역방향 프라이머를 이용하여 PCR증폭을 유도하면 정방향 프라이머 보다 앞 부분에 있는 N-말단에 대한 정보가 없는 DNA를 얻게 될 수 있다. 그 다음 단계에서 메가프라이머를 역방향 프라이머로 사용하고 단백질의 N-말단을 포함하는 올리고 DNA를 정방향 프라이머로 사용하여 PCR을 진행하면 원하는 위치에 돌연변이가 유도된 전장 단백질을 암호화하는 DNA를 얻을 수 있다.
상기 돌연변이체는 재조합 단백질의 형태일 수 있다. 용어 '재조합 단백질'은 유전자 재조합(genetic recombination) 방법을 이용하여 특정 유전자를 벡터(vector)에 삽입하여 만든 새로운 DNA인 '재조합 DNA'를 인위적으로 세포 내에 발현시켜 얻어진 단백질을 의미한다. '유전자 재조합' 방법이란 임의의 생물의 DNA 단편을 다른 DNA 분자에 결합시키는 기술을 의미한다. 유전학적으로 대개의 경우 형질전환, 형질도입, 접합 (교배), 세포융합 등을 통해 유전자 재조합이 이루어질 수 있다.

상기 RANKL 단백질의 돌연변이체는 서열번호 1 또는 서열번호 2로 표시되는 RANKL 단백질의 아미노산 서열이 치환된 것일 수 있다. 치환되는 부위는 상술한 부위와 동일하다.
상기 골대사 질환은 골다공증, 골형성장애 및 골절로 이루어진 군으로부터 선택된 하나 이상인 것일 수 있다. 상기 골다공증은 RANKL이 RANK와 결합하여 파골세포(osteoclast)의 분화를 유도하여 발생되는 것일 수 있다.
파골세포란 뼈를 파괴하는 세포로서, 신체의 필요에 따라 뼈에서 칼슘을 빼내야 할 때 뼈는 파골세포에 의해 파괴될 수 있다. 혈액 속에 칼슘이 부족해서 뼈의 칼슘을 혈액에 충당해야 하거나, 미세한 금이 가거나 흠집이 생긴 뼈, 오래된 뼈를 새 뼈로 바꾸어야 할 때, 파골 세포는 뼈를 파괴할 수 있다. 이러한 파골세포와 뼈를 생성하는 조골세포의 불균형은 골다공증과 같은 골대사질환을 유발할 수 있다.
용어 "분화"는 세포가 분열 증식하여 성장하는 동안에 서로 구조나 기능이 특수화되는 현상, 즉 생물의 세포, 조직 등이 각각에게 주어진 일을 수행하기 위하여 형태나 기능이 변해가는 것을 의미한다.
상기 파골세포의 분화 및 활성화는 RANKL에 의해 조절될 수 있다. 파골세포는 파골 전구세포에서 RANKL에 의한 RANK의 활성화가 TNF 수용체 관련 인자의 자극과 NF-κB, 미토겐-활성 단백질 키나제(Mitogen-activated protein kinase : MAPK), 활성화 단백질 1(AP-1) 및 활성화된 T 세포의 핵인자(NFATc1)를 단계적으로 활성화하여, 다핵 골흡수성 파골세포로 형성된다.
상기 돌연변이체는 개체 내에서 RANK 수용체의 길항제(antagonist)인 것일 수 있다. 길항제란 수용체의 활성부위에 결합하여 효현제(agonist)의 작용을 억제하는 분자이다. 길항제는 수용체길항제와 비수용체길항제로 분류될 수 있다. 수용체 길항제는 수용체의 활성부위나 알로스테릭 부위에 결합하여, 수용체와 효현제의 결합을 억제한다. 이와 달리 비수용체 길항제는 효현제에 의하여 유발되는 반응을 억제할 수 있는 능력을 가지고 있다.
또한, 수용체 길항제는 효현제의 작용을 저해하는 경쟁적 길항제와, 수용체의 수에 영향을 미치거나 수용체와 비가역적으로 결합하여 효현제의 작용을 저해하는 비경쟁적 길항제가 있다.
일 구체예에서, 상기 길항제는 경쟁적 길항제를 포함할 수 있다. 경쟁적 길항제는 수용체의 활성부위와 가역적으로 결합한 후, 효현제와 달리 수용체의 활성화에 필요한 구조변화를 안정화시키지 않는다. 이에 따라, 수용체는 불활성인 상태를 유지하고, 효현제와의 결합이 차단된다.
상기 돌연변이체는 경쟁적 길항제로서, RANKL과 마찬가지로 RANK와 결합할 수 있지만, 결합한 후에도 파골세포의 분화를 유도하지 않을 수 있다. 상기 돌연변이체는 면역원으로써 작용하지만, RANK에 결합하여도 어떠한 생리활성을 일으키지 않을 수 있다. 따라서, 파골세포의 분화가 유도되지 않음에 따라, 상기 조성물은 RANKL로 발현으로 인한 골대사 질환의 예방 또는 치료용 효과를 낼 수 있다.
다른 양상은 상기 약학 조성물의 치료학적 유효량을 개체에 투여하는 단계를 포함하는 골대사 질환을 치료 또는 예방하는 방법을 제공한다.
상기 개체는 인간을 포함할 수 있다.
골대사 질환은 상기한 바와 같이 골다공증, 골형성장애 및 골절로 이루어진 군으로부터 선택된 하나 이상인 것일 수 있다. 골다공증은 상기한 바와 같이 RANKL이 RANK와 결합하여 파골세포의 생성이 유도되어 발생하는 것일 수 있다.
일 구체예 따른 약학적 조성물의 투여량(유효량)은 0.01mg 내지 10,000mg, 0.1mg 내지 1000mg, 1mg 내지 100mg, 0.01mg 내지 1000mg, 0.01mg 내지 100mg, 0.01mg 내지 10mg, 또는 0.01mg 내지 1mg일 수 있다. 다만, 투여량은 제제화 방법, 투여 방식, 환자의 연령, 체중, 성별, 병적 상태, 음식, 투여 시간, 투여 경로, 배설 속도 및 반응 감응성과 같은 요인들에 의해 다양하게 처방될 수 있고, 당업자라면 이러한 요인들을 고려하여 투여량을 적절히 조절할 수 있다. 투여 횟수는 1회 또는 임상적으로 용인가능한 부작용의 범위 내에서 2회 이상이 가능하고, 투여 부위에 대해서도 1개소 또는 2개소 이상에 투여할 수 있다. 인간 이외의 동물에 대해서도, kg당 인간과 동일한 투여량으로 하거나, 또는 예를 들면 목적의 동물과 인간과의 기관(심장 등)의 용적비(예를 들면, 평균값) 등으로 상기의 투여량을 환산한 양을 투여할 수 있다. 가능한 투여 경로에는 경구, 설하, 비경구 (예를 들어, 피하, 근육내, 동맥내, 복강내, 경막내, 또는 정맥내), 직장, 국소 (경피 포함), 흡입, 및 주사, 또는 이식성 장치 또는 물질의 삽입을 포함할 수 있다. 일 구체예에 따른 치료의 대상동물로서는, 인간 및 그 밖의 목적으로 하는 포유동물을 예로 들 수 있고, 구체적으로는 인간, 원숭이, 마우스, 래트, 토끼, 양, 소, 개, 말, 돼지 등이 포함된다.
일 구체예에 따른 약학적 조성물은 약학적으로 허용가능한 담체 및/또는 첨가물을 포함할 수 있다. 예를 들어, 멸균수, 생리식염수, 관용의 완충제(인산, 구연산, 그 밖의 유기산 등), 안정제, 염, 산화방지제(아스코르브산 등), 계면활성제, 현탁제, 등장화제, 또는 보존제 등을 포함할 수 있다. 국소 투여를 위해, 생체고분자(biopolymer) 등의 유기물, 하이드록시아파타이트 등의 무기물, 구체적으로는 콜라겐 매트릭스, 폴리락트산 중합체 또는 공중합체, 폴리에틸렌글리콜 중합체 또는 공중합체 및 그의 화학적 유도체 등과 조합시키는 것도 포함할 수 있다. 일 구체예에 따른 약학적 조성물이 주사에 적당한 제형으로 조제되는 경우에는, 상기 RANKL 돌연변이체 또는 돌연변이체 의해 생성된 항체가 약학적으로 허용가능한 담체 중에 용해되어 있거나 또는 용해되어 있는 용액상태로 동결된 것일 수 있다.
일 구체예에 따른 약학적 조성물은 그 투여방법이나 제형에 따라 필요한 경우, 현탁제, 용해보조제, 안정화제, 등장화제, 보존제, 흡착방지제, 계면활성화제, 희석제, 부형제, pH 조정제, 무통화제, 완충제, 환원제, 산화방지제 등을 적절히 포함할 수 있다. 상기에 예시된 것들을 비롯하여 본 발명에 적합한 약학적으로 허용되는 담체 및 제제는 문헌[Remington's Pharmaceutical Sciences, 19th ed., 1995]에 상세히 기재되어 있다. 일 구체예에 따른 약학적 조성물은 당해 발명이 속하는 기술 분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있는 방법에 따라, 약학적으로 허용되는 담체 및/또는 부형제를 이용하여 제제화함으로써 단위 용량 형태로 제조되거나 또는 다용량 용기 내에 내입시켜 제조될 수 있다. 이때 제형은 오일 또는 수성 매질 중의 용액, 현탁액 또는 유화액 형태이거나 분말, 과립, 정제 또는 캡슐 형태일 수 있다.
일 양상에 따른 RANKL의 돌연변이체는 항-RANKL 항체를 생성시키는 면역원, RANK 수용체에 경쟁적 길항제로 작용함으로써, 효과적으로 골대사 질환을 예방하거나 치료할 수 있는 효과가 있다.
도 1a는 마우스 RANKL의 잔기 158 내지 316을 포함하는 전장 158 아미노산 표적 영역을 나타낸다.
도 1b는 클로닝된 유전자를 pET30a 벡터 다중 클로닝 부위 중 NdeI 및 XhoI 제한 효소 부위에 삽입한 것을 나타낸다.
도 2a는 IPTG 유도 하에 대장균에 의해 생성된 RANKL의 단계별 정제에서의 SDS-PAGE를 나타낸다(1(단계) : 유도 전, 2 : 유도, 3: 상층액, 4: 세포 파편, 5: 칼럼 흐름, 6: 세척, 7: 2차 세척, 8: 제 1 용출, 9: 제 2 용출, 10: 제 3 용출).
도 2b는 wtRANKL 주입이 TRAP-양성 다핵 세포의 수를 증가시켰고, mtRANKL 주입은 TRAP 양성 세포의 감소를 이끌었음을 보여준다. TRAP-양성 세포를 광학 현미경(100Х배율)하에 영상화하였고, 눈금은 20 μm를 나타낸다.
도 2c는 wtRANKL 주입 마우스와 mtRANKL 주입 마우스의 혈청에서, 파골세포(TRAP-양성 세포)를 계수한 것을 나타낸다(p < 0.05).
도 3a는 마이크로-CT 3 차원 이미지를 통해, PBS, wtRANKL 및 mtRANKL 처리 마우스 경골을 관심용적(VOI) 분석으로 나타낸 해면골의 구조를 나타낸다. 근위 경골 스캔은 성장판 수준에서 근접하게 개진되었고, 해상도는 세 공간 차원 모두에서 19 μm이다.
도 3b는 PBS 처리 마우스, wtRANKL 처리 마우스, 및 mtRANKL 처리 마우스에서 골 미네랄 밀도(bone mineral denisity : BMD), 골 부피, 골 부피(%), 및 해면 두께의 수치를 나타낸다(p<0.05).

이하 본 발명을 실시예를 통하여 보다 상세하게 설명한다. 그러나, 이들 실시예는 본 발명을 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.
참고예 1. RANKL의 제조, 돌연변이의 도입 및 배양
1.1. RANKL의 제조
RANKL을 발현하는 마우스의 MC3T3-E1 세포(한국 세포주 은행, 서울)로부터 RANKL cDNA를 복제하는데 사용될 RNA를 추출하였다. 추출된 RNA는 아가로스 겔 전기 영동에 의해 확인하였다. AccuPower RT PreMix Kit(Bioneer, Daejeon, Korea)를 사용하여 제조사의 지시에 따라 cDNA를 준비한다. 반응 혼합물은 Taq 중합효소 완충액, 10 mM의 dNTPs, 25 mM의 MgCl2, 10 μM의 프라이머(RANKL-K158: 5'-CAT ATG AAG CCT GAG GCC CAG CCA TT-3', RANKL-D316 : 5'-CTC GAG GTC TAT GTC CTG AAC TTT GAA AGC C-3'), 2.5 U의 KOD DNA 중합 효소(EMD Millipore, Billerica, MA, USA) 및 2 μL의 RANKL 유전자 구조체 주형을 포함하고, 상기 반응 혼합물에서 RANKL 단편의 증폭 및 복제를 수행하였다.
열 사이클은 a) 95 ℃에서 5 분간의 초기 변성, b) 95 ℃에서 30 초 간 변성, c) 55 ℃에서 30 초간 프라이머의 결합(annealing), d) 70 ℃에서 30 초간 변성으로, 총 40 사이클을 반복했다. RANKL 서열은 도 1a에서 나타낸, 잔기 158 내지 316을 포함하는 전장 158 아미노산 표적 영역을 코딩하였다.
1.2. 돌연변이 도입 및 형질전환
마우스의 RANKL(mRANKL)의 아미노산 서열 위치 180, 189-190 및 223-224에 치환이 이루어진 아미노산을 암호환하는 핵산의 뉴클레오티드 서열은 서열번호 3에 나타내었다. 유전자의 돌연변이는 메가프라이머(megaprimer)를 통하여 도입하였다. 구체적으로, 아미노산 서열 위치 180번째 리신을 아르기닌, 189번째 아스파르산을 이소루신, 190번째 아르기닌을 리신, 223번째 히스티딘을 페닐알라닌, 그리고 224번째 히스티딘을 티로신으로 치환하였다.
인간의 RANKL 단백질(hRANKL)의 경우, 전장은 317로, 전장 316인 마우스의 아미노산 서열보다 1개의 아미노산을 추가로 포함하고 있고, 치환되는 RANKL 부위는 상기 마우스의 아미노산 서열보다 한자리 뒤에 위치한다. 상기 치환되는 부위의 아미노산은 서로 동일하다. 이러한 치환된 아미노산을 암호화하는 핵산의 뉴클레티드 서열은 서열번호 4에 나타내었다.
수득된 PCR 생성물을 복제하기 위해, 상기 생성물을 도 1b에서 나타낸 pET-30a 벡터(Novagen, Madison, WI, USA) NdeI/XhoI 부위에 클로닝하였다. RANKL 단편을 pET30a로 서브 클로닝한 후, RANKL 및 6xHis 태그의 순차적 번역을 위해 정확한 서열을 가지고 있음을 확인하였다. PCR 생성물의 형질전환은 전기천공(5 msec, 12.5 kV/cm)에 의해 E. coli BL21-CodonPlus(DE3)-RIPL Novagen)을 통하여 수행하였다. 모든 서열 분석은 Vector NTI Advance 9.1.0(Invitrogen, Carlsbad, CA, USA)의 프로그램을 사용하였다.
1.3. 배양
재조합 플라스미드를 함유하는 단일 콜로니를 카나마이신(50㎍/mL)이 보충된 LB 배지 20㎖에 접종하고, 200rpm에서 24 시간 동안 교반하면서 37 ℃에서 배양하였다. 이 배양액 10㎖를 50μg/mL의 카나마이신을 포함하는 1L의 LB 배지를 함유하는 삼각 플라스크에 접종하였다. OD600 값이 약 1.0에 도달할 때까지 180 rpm으로 격렬하게 진탕하면서 37 C°에서 배양하였다. 이어서, 이소프로필 β-D-1-티오갈락토피라노사이드(IPTG)를 0, 0.2, 0.4, 0.6, 0.8 및 1.0 mM의 농도로 첨가하여 단백질 발현을 유도하고, 6 시간 동안 배양하였다. 유도 후, 배양물을 5600 Υg 에서 4 ℃, 20분동안 원심분리하였고, 세포 펠렛은 -20℃에서 보관하였다.
참고예 2. mtRANKL 정제
배양물을 원심분리한 후, 펠렛화된 세포를 10 mL의 용해 완충액(ysis buffer)(20 mM 인산 나트륨, 500 mM NaCl, 10 mM 이미다졸, pH 7.4)에 재현탁시켰다. 세포 현탁액에 0.1 mg/mL의 리소자임과 0.1 mM의 페닐메틸술포닐 플루오라이드 albiochem, La Jolla, CA, USA)를 보충하고 아이스(ice)에서 1 시간 동안 배양하였다. 이어서, 글리세롤(20 % v/v, Carlo Erba, France)을 세포 현탁액에 첨가하였다. 세포를 초음파 처리하고 15,000 Υg으로 4 ℃에서 10 분간 원심분리하였다. 상층액을 0.2 μm의 여과지에 통과시키고, 결합 완충액(20 mM 소듐 포스페이트, 500 mM NaCl, pH 7.4, 10 mM 이미다졸, 5 mM DTT, pH 7.4)으로 평형화된 Ni2+ 친화성 크로마토그래피 HisTrap FF 컬럼(1 mL, GE Healthcare Life Science, Piscataway, NJ, USA)에 고정하였다. 이어서, 상기 컬럼을 20 mM 이미다졸로 보충된 결합 완충액을 사용하여 세척하였다. 세척 후, 단백질을 용출 완충액(Qiagen)으로 용출시켰다. 용출된 단백질을 10,000 MW Slide-A-Lyzer 투석 카세트(Thermo Fisher Scientific, Waltham, MA, USA)에서 투석완충액(인산 완충 식염수[phosphate-buffered saline:PBS] 중 글리세롤 20 % v/v)으로 투석하였다. 정제된 단백질을 진공 농축시켰다(Savant Instruments, Holbrook, NY, USA). 마지막으로, 단백질을 소듐 도데실술페이트 폴리아크릴아미드 겔 전기영동(SDS-PAGE)으로 분석하고, 단백질 농도를 계산하여 브래드포드(bradford) 분석법으로 결정하였다. 내독소 제거를 위해 크로마토그래피를 위한 초기 세척 후에 추가적으로 세척 단계를 도입했다. 이 단계는 수지 상(resin bed)의 80 배 부피에서 W1-TX114(0.1% 트리톤 X-114를 갖는 W1-TX100) 또는 1 % 소듐 데옥시콜레이트(완충액 W1-DOC)를 사용하고, 25 ℃에서 W1-TX100 및 W1-DOC, 또는 4 ℃에서 W1-TX114으로 수행된다. 각 세척 단계의 용출액을 내독소 정량을 위해 수집했다.
재조합 단백질 샘플을 2x 용해 완충액(0.5 M Tris-HCl, pH 6.8, 0.5 %(/v) 브로모 페놀블루, 10 %(v/v) 글리세롤, 2 %(v/v) SDS 및 10 %(v/v) β-메르캅토 에탄올)에 1:1(v/v) 비율로 첨가하고 5 분간 끓였다. 그런 다음 단백질을 전기 영동으로 분석하였다. 분리 후, 겔을 코마씨(comassie) 브릴리언트 블루 G-250로 염색하였다. 재조합 단백질의 순도 및 회수율을 결정하기 위해, 고정된 양의 단백질로 로딩된 염색된 겔을 디지털 스캐너(EPSON, USA)를 사용하여 300 dpi로 이미지화하였다.
도 2a에서 SDS-PAGE 는 유도된 샘플의 전체 세포 단백질 추출물에서 약 19kDa 크기의 밴드를 나타내었다.
참고예 3. TRAP 분석
4 주된 수컷 BALB/c 마우스를 Orient Bio(광주, 한국)에서 구입하여 조선대학교 동물관리위원회(IACUC2017-A0002)의 승인을 받은 동물시설에서 보관하였다. 마우스에서 골수 세포를 획득한 후, 골수 단핵세포를 96-웰(well) 플레이트에(1 Υ 104 cells/well)로 시딩(seed)하고, 다양한 농도의 화합물의 부재 또는 존재 하에 RANKL(50 ng/mL)로 자극하기 전 밤새 M-CSF(100 ng/mL)와 함께 배양하였다. 상기 배지는 2 일마다 교체하였다. 6일 후, 세포를 4% 파라 포름 알데하이드로 고정시키고, 0.1 % Triton X-100에서 투과시킨 후, PBS로 세척하였다. 그리고 TRAP 활성(Sigma-Aldrich, St. Louis, MO, USA)을 염색하였다. 5 개 이상의 핵을 함유하는 TRAP-양성 다핵세포를 파골세포로서 계수하였다.
참고예 4. 웨스턴 블랏
단백질을 SDS-PAGE로 분리하고 니트로 셀룰로오스 멤브레인(Bio-Rad)에 전기 침투시켰다. TBST [10 mM Tris(pH 7.5), 150 mM NaCl, 0.1 %(vol/vol) Tween 20] 중 5 %(중량/부피) 무지방 분유로 막을 차단 한 후, 차단 용액 중 1 차 항체에 대한 각 마우스 혈청을 조사하였다. 상기 막을 트리스 완충 식염수로 3 회 세척하였다. 당근과산화효소(horse peroxidase:HRP)-결합 2 차 항체를 1 %(wt/vol) 무지방 분유에서 TBST로 1 : 5000 희석 하였다. 막은 ECL_system(Amersham Pharmacia Biotech)을 사용하였다.
참고예 5. ELISA(Enzyme-linked immunosorbent assay)
시판되는 효소 면역 측정법 키트(R & D Systems, MN, USA)를 사용하여 제조자의 프로토콜에 따라 마우스의 혈청의 RANKL 양을 측정하였다. 비색 마이크로 플레이트 판독기(BioTek, Winooski, USA)에서 흡광도(450 nm)를 측정하였다. 이어서, 570 nm에서의 판독 값으로부터 상기 흡광도의 판독 값을 뺐다.
참고예 6. 마이크로-CT(Micro-computed tomography: Micro-CT)
각 실험의 대상이 된 마우스의 오른쪽 대퇴골을 절개하고 CT 영상을 광주 한국기초과학연구소에 위치한 Quantum GX μCT imaging system(PerkinElmer, Hopkinton, MA, USA)을 사용하여 수행하였다. X 선원(X-ray source)은 시야를 45mm로, 90kV 및 88mA로 설정하였다(보셀[voxel] 크기, 90㎛; 스캐닝 시간, 14 분). CT 이미지는 Quantum GX 3D Viewer 소프트웨어를 사용하였다. 스캔 후, 이미지 분할은 Analyze nalyzeDirect, Overland Park, KS, USA)를 사용하여 수행되었다. 간단히 말해, 볼륨 편집 도구를 사용하여 반자동 및 수동 도구(예 : 개체 추출, 영역 확장 및 반대자 분리 기호)를 사용하여 다리를 분할하였다. 이어서 다리의 3D 렌더링을 생성하고 골밀도(BMD), 골 부피(BV), 골 부피 백분율(%) 및 해면 두께를 ROI 도구를 사용하여 계산했다.
참고예 7. 통계적 분석
통계적 분석을 위해 양측의 쌍으로 된 Student t 검정을 사용했다. P<0.05은 통계적으로 유의하다고 간주하였다. 데이터는 다르게 명시되지 않는 한 평균±표준 편차(SD)로 표시하였다. 데이터는 Windows 용 SPSS 버전 20.0 소프트웨어 프로그램(SPSS, Chicago, IL, USA)을 사용하여 분석하였다. Windows 용 GraphPad Prism 버전 6.00 소프트웨어 프로그램(GraphPad, La Jolla, CA, USA)을 시험관 및 생체 내 실험에서 데이터를 분석하는데 사용하였으며, 평균 ± SD로 데이터를 표시한다.
실시예 1. 체외(in vitro) 파골세포 생성 분석
파골세포 형성에 대한 mtRANKL의 영향을 평가하기 위해, RANKL 처리된 1 차 골수 유래 대식세포(BMM)에서 참고예 2에 기술한 방법으로 TRAP 분석을 수행하였고, 그 결과는 도 2b, 2c에 나타내었다.
도 2b 및 도 2c에서 나타내고 있는 바와 같이, BMM은 성숙한 TRAP 양성 다핵 파골세포로 분화하였고, 그 수는 mtRANKL로 처리한 대조군 세포에서보다 야생형 RANKL (wtRANKL)로 처리한 세포에서 유의하게 높았다. 따라서, mtRANKL은 wtRANKL과 달리 파골세포의 분화를 유도하지 않는 것을 알 수 있었다.
실시예 2. mtRANKL의 골 파괴 분석
mtRANKL의 골 파괴 효과 여부를 조사하기 위해, 4 주된 수컷 BALB/c 마우스 Orient Bio)를 광조절 환경(2 시간의 명암주기)에서 최대 4 마리를 수용할 수 있는 우리에서 오토-클레이브된 물과 전체 및 분말 설치류 식이요법(Purina, St. Louis, MO, USA)으로 제공하였다. 6 주부터 9 마리의 마우스를 3개의 그룹으로 무작위로 나누어, 각 그룹은 야생형 RANKL(Amgen, Thousand Oaks, CA, USA), 상기 참고예 2에서 정제된 mtRANKL, 및 식염수를 주 3 회 피하 주사(1.0 mg/kg)로 주입하였다. 경부 해면골을 참고예 6에서 기술한 마이크로-CT 분석 방법으로 검사하였다. 그 결과는 도 3a과 도 3b에 나타내었다.
도 3a에서 나타낸 바와 같이, wtRANKL에 의해 주입된 마우스는 경미한 골다공증을 보였다. BMD, 골 부피(BV), 골 부피의 백분율(%), 및 해면 두께가 현저하게 감소했다. 그러나 도 3b에서 나타낸 바와 같이, mtRANKL이 주입된 마우스의 해면골에서는 유의한 변화가 나타나지 않았다. 따라서 mtRANKL은 파골세포 분화에 영향을 미치지 않는 것으로 판단된다.
실시예 3. mtRANKL 면역화의 효과 분석
mtRANKL은 파골세포-자극 활성이 없으므로, mtRANKL로 면역화하면 외인성 RANKL의 파골세포-자극 활성을 억제하는 항-RANKL 항체가 유도될 수 있다. 이러한 가능성을 입증하기 위해, wtRANKL 유도된 마우스를 골다공증 동물 모델로 사용하여 골다공증에 대한 mtRANKL 단백질 면역화의 치료 효과를 조사하였다.
3.1. mtRANKL 면역화
17 주 된 수컷 BALB/c 마우스를 3 그룹으로 균등하게 나누었다. PBS를 복강내 주사하는 음성대조군 그룹, wtRANKL만을 주입하는 그룹, 52 일 동안 mtRANKL로 면역화한 후, wtRANKL을 주입하는 그룹으로 나누었다. mtRANKL은 수산화 알루미늄 보조제와 혼합하여 주입하였다. 면역화 과정은 wtRANKL 주입 52일 전 mtRANKL의 1차 접종을 시작하여, 39일 전 2차 접종, 14일 전 3차 접종, 총 3 회에 걸친 접종으로 진행됬다. mtRANKL의 양은 마우스 당 매회 0.2 mg이었다.
52일 후, 음성대조군에는 PBS를, 나머지 두 그룹에는 wtRANKL을 주입하였다. 그 다음날 wtRANKL을 1회 더 주입하였다. wtRANK은 피하 주사(2.0 ㎎/㎏)하였다. 항-RANKL 항체의 역가를 결정하기 위해 혈청을 수집했다. 그 후 경골을 추출한 후, 경골의 접착성 연조직을 제거했다.
3.2. 항체의 생성 결과
mtRANKL의 면역화로 인해 항체가 생성되었는지 알아보기 위해 수집된 혈청을 대상으로, 참고예 4의 웨스턴 블랏을 시행하였고, 그 결과를 도 4a에 나타내었다.
도 4a에 나타낸 바와 같이 mtRANKL로 면역화를 진행한 마우스의 혈청에서 항체가 검출되었다. 반면, 면역화를 진행하지 않은 마우스(mock)의 경우, 아무런 항체도 검출되지 않았다.
또한, 참고예 5에서 기술한 방법으로 ELISA 분석을 시행하였고, 그 결과는 도 4b에 나타내었다.
도 4b에 나타낸 바와 같이, 면역화를 하지 않은 마우스에 wtRANKL을 투여할 경우, 혈액내 RANKL의 수치가 상승하였다. 반면, mtRANKL로 면역화한 마우스에 wtRANKL을 투여할 경우 그렇지 않은 마우스에 비해 혈액내 RANKL이 제거되었다. 이에 따라, mtRANKL의 면역화로 인해, 마우스 체내에 항-RANKL 항체가 형성되었음을 알 수 있다.
3.3. 면역화에 따른 골 파괴 분석 결과
mtRANKL의 면역화 후, RANKL을 주입할 경우, RANKL의 골 파괴 효과 여부를 알기 위해 참고예 6의 방법으로 마이크로-CT 분석을 진행하였다. 근위 경골의 대표적인 3 차원 이미지와 그래프를 도 5a, 5b에 나타내었다.
도 5a에서 나타내는 바와 같이, wtRANKL 주입 마우스의 해면골은 PBS로 처리한 마우스와 비교하여, 더욱 구멍이 많아지고, 해면골 조직을 형성하는 막대는 더 얇아졌다. 그리고 경미한 골다공증을 보였다. 그러나 PBS로 처리한 마우스와 마찬가지로 mtRANKL로 면역화한 wtRANKL 주입 마우스의 해면골은 wtRANKL 주입 마우스보다 섬유질이 두껍고 밀도가 더 높았으며 PBS 처리 마우스와 유사한 상태였다.
도 5b에서 나타낸 바와 같이, PBS를 처리한 마우스의 경골 BMD(821.1 ± 14.8 mg/cm3, P <0.05)와 mtRANKL 면역화한 wtRANKL 주입 마우스의 경골 BMD(795.8 ± 11.9 mg/cm3, P <0.05)는 wtRANKL 주입 마우스의 경골 BMD(738.0 ± 19.1 mg/cm3 , P <0.05)에 비해 상당히 높게 나타났다.
또한, PBS-처리 마우스 및 mtRANKL 면역화한 wtRANKL 주입 마우스의 골 부피 및 골 부피 백분율의 값은 유사한 수준으로 유지되었고, wtRANKL 주입 마우스의 값은 감소하였다. 이에 따라, mtRANKL의 면역화로 마우스 체내에 항-RANKL 항체가 형성되었고, 이로 인해 파골세포 분화의 유도가 감소하였음을 알 수 있다.
실시예 4. OVX(ovariectomy) 실험 분석
4.1. 면역화 및 OVX 시술
17 주 된 수컷 BALB/c 마우스를 mtRANKL로 면역화 후 OVX시술을 하는 치료 그룹, OVX 시술만 진행하는 그룹, 어떠한 처리도 진행하지 않는 음성대조군 그룹으로 균등하게 나누었다.
mtRANKL 면역화 그룹의 경우, OVX 시술 52일 전 1차 접종을 시작하여, 시술 39일 전 2차 접종, 시술 14일 전 3차 접종, 총 3 회에 걸쳐 mtRANKL을 접종하였다. 면역화에 사용된 mtRANKL은 수산화 알루미늄 보조제와 혼합하여 주입하였다. mtRANKL의 양은 마우스 당 매회 0.2 mg이었다.
1차 접종을 시작으로 52일 후, OVX 시술을 진행하였다. 시술 후 70일 째, 혈청을 수집하고, 경골을 추출한 후, 경골의 접착성 연조직을 제거했다.
4.2. 효과 분석 결과
난소를 제거한 마우스(OVX)에 mtRANKL 면역화 효과를 분석하기 위해, 참고예 6에 따른 마이크로-CT 분석을 진행하였다. 근위 경골의 대표적인 3 차원 이미지와 BMD, 골 부피, 골 부피의 백분율 및 해면 두께의 수치를 표시한 그래프를 각각 도 6a와 도 6b에 나타내었다.
도 6a에서 나타내는 바와 같이, OVX 시술 마우스의 해면골은 OVX 시술을 하지 않은 비교하여, 경미한 골다공증을 보였다. 그러나 mtRANKL로 면역화 후 OVX를 시술한 마우스의 해면골은 OVX 시술만 진행한 마우스보다 섬유질이 두껍고 밀도가 더 높았으며 음성대조군(-)의 마우스와 유사한 상태였다.
도 6b에서 나타낸 바와 같이, mtRANKL 면역화 후, OVX를 시술한 마우스의 경골 BMD와 음성대조군의 BMD는 OVX 시술 마우스의 경골 BMD에 비해 상당히 높게 나타났다. 마찬가지로, 전체 부피에 대한 골 부피, 골 부피 백분율(%), 및 해면 두께에서도 mtRANKL 면역화한 마우스 그룹은 음성대조군과 유사한 값을 가졌다. 해당 결과는 mtRANKL의 면역화가 난소를 절제한 OVX 마우스에서도 파골세포 분화의 유도를 차단할 수 있음을 보여준다.
Claims (13)
- 서열번호 1의 아미노산 서열을 포함하는 NF-κB의 수용체 활성물질 리간드(receptor activator of Nuclear factor-kappa B ligand : RANKL) 단백질 N-말단에서, 180번째의 리신(Lys)이 아르기닌(Arg)으로, 189번째의 아스파르산(Asp)이 이소루신(Ile), 루신(Leu), 및 아스파라긴(Asn) 중 어느 하나로, 190번째의 아르기닌(Arg)이 리신(Lys)으로, 223번째의 히스티딘(His)이 페닐알라닌(Phe) 또는 티로신(Try)으로, 및 224번째의 히스티딘(His)이 페닐알라닌(Phe) 또는 티로신(Try)으로의 치환으로 이루어진 군으로부터 선택된 하나 이상의 치환을 갖는 돌연변이체, 또는서열번호 2의 아미노산 서열을 포함하는 RANKL 단백질 N-말단에서, 181번째의 리신(Lys)이 아르기닌(Arg)으로, 190번째의 아스파르산(Asp)이 이소루신(Ile), 루신(Leu), 및 아스파라긴(Asn) 중 어느 하나로, 191번째의 아르기닌(Arg)이 리신(Lys)으로, 224번째의 히스티딘(His)이 페닐알라닌(Phe) 또는 티로신(Try)으로, 및 225번째의 히스티딘(His)이 페닐알라닌(Phe) 또는 티로신(Try)으로의 치환으로 이루어진 군으로부터 선택된 하나 이상의 치환을 갖는 돌연변이체.
- 청구항 1에 있어서, 상기 돌연변이체는 개체 투여되어 항체를 생성하는것인 돌연변이체.
- 청구항 1의 돌연변이체에 의해서 생성된 항체.
- 청구항 1의 돌연변이체를 암호화하는 핵산 분자.
- 청구항 4에 이어서, 상기 핵산 분자는 서열번호 3 또는 4의 뉴클레오티드 서열을 갖는 것인 핵산 분자.
- 청구항 4 또는 5의 핵산 분자를 포함하는 벡터.
- 청구항 6의 벡터를 포함하는 숙주세포.
- 청구항 1의 돌연변이체 또는 청구항 3의 항체를 유효성분으로 포함하는, 골대사 질환을 예방 또는 치료하기 위한 약학 조성물.
- 청구항 8에 있어서, 상기 골대사 질환은 인간의 골다공증, 골형성장애 및 골절로 이루어진 군으로부터 선택된 하나 이상인 것인 약학 조성물.
- 청구항 9에 있어서, 상기 골다공증은 RANKL이 NF-κB의 수용체 활성물질(receptor activator of Nuclear factor-kappa B : RANK)에 결합하여 파골세포(osteoclast)의 분화를 유도하여 발생되는 것인 약학 조성물.
- 청구항 8에 있어서, 상기 돌연변이체는 개체 내 투여되어 항체를 생성하는 것인 약학 조성물.
- 청구항 8에 있어서, 상기 돌연변이체는 개체 내에서 RANK 수용체의 길항제인 것인 약학 조성물.
- 청구항 8에 있어서, 상기 돌연변이체는 개체 내 투여되어 파골세포의 분화를 일으키지 않는 것인 약학 조성물.
