Abstract
에리스로포이에틴 유래 펩티드를 유효성분으로 포함하는 세포증식 촉진용 조성물에 관한 것으로, 종래의 천연 인간 에리스로포이에틴에 비해 간단한 구조를 가지므로 조직-혈관방어막(tissue-blood barrier)의 통과가 용이하고 세포 보호 활성이 우수하고, 세포 증식 부작용이 없으며, 조혈 기능을 개선할 수 있는바, 빈혈 질환의 치료 또는 예방에 이용될 수 있다.

본 출원은 2018년 8월 27일 출원된 대한민국 특허출원 제10-2018-0100580호를 우선권으로 주장하고, 상기 명세서 전체는 본 출원의 참고문헌이다.
에리스로포이에틴 유래 펩티드를 유효성분으로 포함하는 세포증식 촉진용 조성물에 관한 것이다.
살아가는 동안 인체는 인체에 유해한 자극에 끊임없이 노출되고 그 노출에 반응하여 자신의 몸을 보호하게 된다. 유해한 자극에는 저산소증, 감염, 기계적인 자극 등 다양한 자극이 있으며, 이러한 자극에 반응하는 방어 기전은 세포 단위에서부터 존재한다. 자극에 반응하여 나타나는 방어 기전으로써 분비되는 다양한 사이토카인들은 자극에 노출되어 이상이 생긴 세포를 사멸시키거나 정상세포의 사멸을 방지하면서 개체를 보호하는 역할을 한다.
에리스로포이에틴은 분자량이 약 30,000인 당단백질로서 적혈구를 만드는 세포의 분화를 촉진하고 적혈구의 수를 늘려 빈혈의 예방 개선에 효과가 있는 혈액조성 사이토카인이다. 이 단백질은 적혈구 전구세포의 수용체에 접합함으로써 작용을 시작하며 세포 내에 칼슘이온의 증가, DNA 생합성의 증가 및 헤모글로빈 생성자극 등을 유발시킨다. 따라서, 에리스로포이에틴을 신장병 환자의 빈혈, 미성숙아의 빈혈, 갑상선기능 항진결손(hypothyroidism)에 의한 빈혈, 영양실조에 의한 빈혈, 만성신부전에 의한 빈혈, 수술에 따른 빈혈 등, 빈혈을 치료하는 약제로 사용될 수 있다.
세포 배양 또는 합성을 통해 인공 제작된 에리스로포이에틴은 세포보호효과와 세포증식효과에 있어 좋은 활성을 가지지만, 비교적 짧은 반감기를 가지고 있다. 이를 개선하기 위한 다당체화(darbopoetin alfa), 폴리에틸렌글리콜화(Methoxy polyethylene glycol-epoetin beta)를 통해 반감기가 증가된 에리스토포이에틴 변형체가 개발되어 빈혈치료제로 사용되어 왔다. 그러나 이러한 약물들도 뇌조직과 같이 조직·혈관간장벽이 존재하는 일부 조직에서는 약물 전달에 어려움이 있다. 이러한 단점을 보안할 수 있는 대안으로 EPOtris, EPObis, MK-X와 같은 에리스로포이에틴의 일부분을 인공적으로 제작한 크기가 작은 펩티드들이 개발되어왔지만, 이들은 세포증식 효과 없이 조직보호효과만 가지고 있어 신경질환 등에는 효과적이나 세포증식 효과를 요구하는 빈혈이나 다른 질환의 치료제로서는 한계를 가진다는 문제점이 있다.

상기 펩티드는 알파 헬릭스 구조를 형성할 수 있으며, 세포보호 활성을 가질 수 있다. 또한, 상기 펩티드는 조혈모세포, 지방세포, 췌장세포, 근육세포, 혈관세포 또는 피부세포의 증식 또는 분화를 촉진할 수 있다.
본 명세서 내 용어, "조혈모세포"란 조혈줄기세포로도 일컬어지며 자가 증식 및 분화를 통해 상기 조혈작용을 수행하는 세포를 의미한다. 조혈모세포는 각종 혈구의 중간 과정인 전구세포를 거쳐 최종 세포로 성숙하게 되며, 적혈구, 혈소판, 중성구, 호산구, 호염구, 단핵구, T 세포, B세포, 자연살상세포 또는 가지돌기 세포 등의 다양한 혈액 내 세포로 분화하므로, 상기 조혈모세포의 분화에 의하여 적혈구 또는 림프구 세포의 발달이 촉진될 수 있다. 따라서, 일 양상에 따른 조성물은 조혈모세포의 증식 또는 분화를 촉진함으로써 빈혈의 치료제로 이용될 수 있다. 또한, 에리스로포이에틴 수용체를 발현하는 지방조직, 췌장, 근육, 혈관 피부 등과 같은 조직에서 세포 증식 또는 분화를 유도할 수 있는바, 비만 및 당뇨, 근육 재생, 혈관 재생, 피부 재생 등과 같이 세포증식 효과를 필요로 하는 질환의 예방 또는 치료에 이용될 수 있다.
다른 양상은 서열번호 1 내지 28로 구성된 군에서 선택되는 어느 하나 이상의 펩티드를 유효성분으로 포함하는 조혈 기능 개선용 조성물을 제공한다. 상기 펩티드의 구체적인 내용은 전술한 바와 같다. 상기 조성물은 조혈 기능을 개선하여 빈혈, 적혈구 감소증 또는 조혈을 치료 또는 완화할 수 있으며 신체의 전반적인 조혈 활성을 증진시킬 수 있을 뿐만 아니라, 근육, 혈관 및 피부 재생을 촉진하고 비만을 억제함으로써 체력을 증진시키고 건강을 유지하는데 도움을 줄 수 있다.
다른 양상은 서열번호 1 내지 28로 구성된 군에서 선택되는 어느 하나 이상의 펩티드를 유효성분으로 포함하는 빈혈 질환의 예방 또는 치료용 조성물을 제공한다. 또한, 상기 조성물의 약제학적 유효량을 그를 필요로 하는 개체에 투여하거나, 인 비트로(in vitro) 상에서 세포에 접촉시키거나, 인 비보(in
vivo) 상에서 실험동물에 투여하는 단계를 포함하는 빈혈 질환의 예방 또는 치료 방법을 제공한다. 상기 펩티드의 구체적인 내용은 전술한 바와 같다.
본 명세서 내 용어, "빈혈"은 적혈구의 숫자나 적혈구 내의 혈색소가 정상치보다 낮아지는 상태를 의미하는 것으로, 건강한 적혈구를 거의 생산해 내지 못하거나 너무 많은 적혈구가 손실 또는 파괴되어 적혈구가 만들어지는 속도가 이를 따라가지 못하는 상황을 의미한다.
상기 조성물은 조혈줄기세포의 증식 또는 분화를 촉진시킴으로써, 적혈구 또는 림프구 세포의 발달을 촉진시킬 수 있는바, 빈혈 질환의 예방 또는 치료에 이용될 수 있다. 상기 빈혈 질환은 예를 들면, 급성 또는 만성 빈혈, 신장 질환에 의한 빈혈, 신부전에 의한 빈혈, 혈액 질환에 의한 빈혈, 방사선요법-유발 빈혈, 화학요법-유발 빈혈, 수술 과정에 의한 빈혈, 감염에 의한 빈혈, 영양결핍성 빈혈, 비정상적인 적혈구생성, 조산의 초기 빈혈 또는 이들의 혼합 질환일 수 있다.
일 구체예 따른 상기 조성물은 조성물 총 중량에 대하여 0.001 중량% 내지 80 중량%의 서열번호 1 내지 28로 구성된 군에서 선택되는 어느 하나 이상의 펩티드를 포함할 수 있다. 또한, 조성물의 투여 용량은 0.01㎎ 내지 10,000㎎, 0.1㎎ 내지 1000㎎, 1㎎ 내지 100㎎, 0.01㎎ 내지 1000㎎, 0.01㎎ 내지 100㎎, 0.01㎎ 내지 10㎎, 또는 0.01㎎ 내지 1㎎일 수 있다. 다만, 투여량은 제제화 방법, 투여 방식, 환자의 연령, 체중, 성별, 병적 상태, 음식, 투여 시간, 투여 경로, 배설 속도 및 반응 감응성과 같은 요인들에 의해 다양하게 처방될 수 있고, 당업자라면 이러한 요인들을 고려하여 투여량을 적절히 조절할 수 있다. 투여 횟수는 1회 또는 임상적으로 용인 가능한 부작용의 범위 내에서 2회 이상이 가능하고, 투여 부위에 대해서도 1개소 또는 2개소 이상에 투여할 수 있다. 인간 이외의 동물에 대해서도, ㎏당 인간과 동일한 투여량으로 하거나, 또는 예를 들면 목적의 동물과 인간과의 기관(심장 등)의 용적비(예를 들면, 평균값) 등으로 상기의 투여량을 환산한 양을 투여할 수 있다. 가능한 투여 경로에는 경구, 설하, 비경구 (예를 들어, 피하, 근육내, 동맥내, 복강내, 경막내, 또는 정맥내), 직장, 국소 (경피 포함), 흡입, 및 주사, 또는 이식성 장치 또는 물질의 삽입을 포함할 수 있다. 일 구체예에 따른 치료의 대상동물로서는, 인간 및 그 밖의 목적으로 하는 포유동물을 예로 들 수 있고, 구체적으로는 인간, 원숭이, 마우스, 래트, 토끼, 양, 소, 개, 말, 돼지 등이 포함된다.
일 구체예에 따른 약학적 조성물은 약학적으로 허용 가능한 담체 및/또는 첨가물을 포함할 수 있다. 예를 들어, 멸균수, 생리식염수, 관용의 완충제(인산, 구연산, 그 밖의 유기산 등), 안정제, 염, 산화방지제(아스코르브산 등), 계면활성제, 현탁제, 등장화제, 또는 보존제 등을 포함할 수 있다. 국소 투여를 위해, 생체고분자(biopolymer) 등의 유기물, 하이드록시아파타이트 등의 무기물, 구체적으로는 콜라겐 매트릭스, 폴리락트산 중합체 또는 공중합체, 폴리에틸렌글리콜 중합체 또는 공중합체 및 그의 화학적 유도체 등과 조합시키는 것도 포함할 수 있다. 일 구체예에 따른 약학적 조성물이 주사에 적당한 제형으로 조제되는 경우에는, 상기 펩티드가 약학적으로 허용 가능한 담체 중에 용해되어 있거나 또는 용해되어 있는 용액상태로 동결된 것일 수 있다.
일 구체예에 따른 약학적 조성물은 그 투여방법이나 제형에 따라 필요한 경우, 현탁제, 용해보조제, 안정화제, 등장화제, 보존제, 흡착방지제, 계면활성화제, 희석제, 부형제, pH 조정제, 무통화제, 완충제, 환원제, 산화방지제 등을 적절히 포함할 수 있다. 상기에 예시된 것들을 비롯하여 본 발명에 적합한 약학적으로 허용되는 담체 및 제제는 문헌[Remington's Pharmaceutical Sciences, 19th ed., 1995]에 상세히 기재되어 있다. 일 구체예에 따른 약학적 조성물은 당해 발명이 속하는 기술 분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있는 방법에 따라, 약학적으로 허용되는 담체 및/또는 부형제를 이용하여 제제화함으로써 단위 용량 형태로 제조되거나 또는 다용량 용기 내에 내입시켜 제조될 수 있다. 이때 제형은 오일 또는 수성 매질 중의 용액, 현탁액 또는 유화액 형태이거나 분말, 과립, 정제 또는 캡슐 형태일 수 있다.
다른 양상은 서열번호 1 내지 28로 구성된 군에서 선택되는 어느 하나 이상의 펩티드를 유효성분으로 포함하는 빈혈 질환의 예방 또는 개선용 건강기능성 식품 조성물을 제공한다. 상기 펩티드의 구체적인 내용은 전술한 바와 같다.
상기 건강식품 조성물은 상기 펩티드 이외에 다른 식품 또는 식품 성분과 함께 사용될 수 있고, 통상적인 방법에 따라 적절하게 사용될 수 있다. 유효 성분의 혼합양은 사용 목적 (예방, 건강 또는 치료적 처치)에 따라 적합하게 결정될 수 있다. 일반적으로, 건강기능식품의 제조시에 본 명세서의 조성물은 원료에 대하여 15 중량부 이하의 양으로 첨가될 수 있다. 상기 건강식품의 종류에는 특별한 제한은 없다.
다른 양상은 서열번호 1 내지 28로 구성된 군에서 선택되는 어느 하나 이상의 펩티드를 유효성분으로 포함하는 조혈장애의 예방 또는 치료용 약학적 조성물을 제공한다. 또한, 상기 조성물의 약제학적 유효량을 그를 필요로 하는 개체에 투여하거나, 인 비트로(in vitro) 상에서 세포에 접촉시키거나, 인 비보(in
vivo) 상에서 실험동물에 투여하는 단계를 포함하는 조혈장애의 예방 또는 치료 방법을 제공한다.
다른 양상은 서열번호 1 내지 28로 구성된 군에서 선택되는 어느 하나 이상의 펩티드를 유효성분으로 포함하는 조혈장애의 예방 또는 개선용 건강기능성 식품 조성물을 제공한다. 상기 펩티드의 구체적인 내용은 전술한 바와 같다.
본 명세서 내 용어 "조혈장애"는 조혈작용에 결함이 생겨 혈구 생성이 정상적으로 이루어지지 못하는 상태를 의미하며 조혈장애는 다양한 길환을 야기할 수 있다. 구체적으로, 상기 질환은 재생 불량성 빈혈, 악성 림프종 또는 백혈병, 만성 간 장애, 신부전, 중증 감염증, 골수 장애성 혈소판 감소증, 특발성 혈소판 감소성 자반병(ITP), 혈소판 무력증, 림프구감소증, 호중구감소증, 단핵구감소증, 과립구감소증, 골수 형성이상 증후군 또는 골수 증식성 질환일 수 있다.
상기한 바와 같이, 일 양상에 따른 에리스로포이에틴 유래 펩티드는 세포의 증식 또는 분화를 촉진하고, 손상된 세포에 대하여 세포 보호 효과를 가질 뿐만 아니라 조혈 효과를 나타냄으로써 빈혈 질환, 조혈장애를 야기하는 다양한 질환의 치료 또는 예방에 이용될 수 있다. 뿐만 아니라, 상기 펩티드는 에리스로포이에틴 수용체를 발현하는 지방조직, 췌장, 근육, 혈관 피부 등과 같은 조직에서 세포 증식 또는 분화를 유도할 수 있는바, 비만 및 당뇨, 근육 재생, 혈관 재생, 피부 재생 등과 같이 세포증식 효과를 필요로 하는 질환의 예방 또는 치료에 이용될 수 있다.
일 양상에 따른 조성물은 에리스로포이에틴 단백질 서열로부터 유래된 펩티드를 포함하는 것으로서, 종래의 천연 인간 에리스로포이에틴에 비해 간단한 구조를 가지므로 조직-혈관방어막(tissue-blood barrier)의 통과가 용이하고 세포 보호 활성이 우수하고, 세포 증식 부작용이 없으며, 조혈 기능을 개선할 수 있는바, 빈혈 질환의 치료 또는 예방에 이용될 수 있다. 뿐만 아니라, 생산비용이 적게 들어 경제적이므로 생체 조직으로의 수송이 용이하면서 세포증식 효과가 있는 에리스로포이에틴의 대체 물질로 활용될 수 있다.

이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 하기 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
[실시예]
실시예 1. 에리스로포이에틴 유래 펩티드의 합성
본 발명의 에리스로포이에틴 유래 펩티드는 종래 공지된 고상 합성(Solid Phase Peptide Synthesis)기술에 따라 단량체로 합성하였다(Peptron, Daejeon, Korea).
구체적으로, 천연 에리스로포이에틴 수용체의 타겟 지역(site 2)의 서열 중 중요한 아미노산 서열(Arg103, Ser104, Leu105, Leu108 및 Arg110)에 결합할 수 있는 에리스로포이에틴 유래 펩티드를 합성하였으며, 상기 펩티드 각각의 구체적인 특성을 확인하였다. 상기 합성된 펩티드의 농도를 측정하기 위해 액체크로마토그래피/질량 선택 검출기(liquid chromatography/mass selective detector)(HP 1100 series)를 이용하였다. 순도의 측정은 고성능 액체크로마토그래피(SHIMADZU prominence HPLC) 분석에 의해 수행되었다(> 95% purity). 에리스로포이에틴 유래 펩티드를 하기 표 1에 나타냈다.

실시예 2. 일부 서열을 이용한 에리스로포이에틴 유래 펩티드(1)
시퀀스의 변화 실험을 위해 에리스로포이에틴과 그 수용체의 결합 모형은 이미 알려진 결합 구조(Protein Data Bank ID: 1EER)를 바탕으로 하였다. 알려진 아미노산의 특성을 바탕으로 에리스로포이에틴 유래 펩티드의 아미노산을 치환 하였다. 아미노산 곁사슬의 극성에 따라 4종(① 비극성 혹은 소수성, ② 중성, ③ 음전하, ④ 양전하)으로 분류된다. 이렇게 비극성(소수성), 중성, 음전하, 또는 양전하를 띄는 아미노산의 정보를 이용하여, 기존의 아미노산 서열을 치환하여 각 특성의 변화를 유도하였다.
ML1 펩티드의 일부 서열을 변화시킨 펩티드 및 이들의 특징을 하기 표 3 및 표 4에 나타냈다.

실시예 3. 일부 서열을 이용한 에리스로포이에틴 유래 펩티드(2)
상기 실시예 2와 동일한 ML1 기본 서열을 이용하여 펩티드의 일부 아미노산을 치환하였다. 이때, 기존의 에리스로포이에틴과 그 수용체의 결합 모형을 바탕으로 치환된 아미노산이 기존의 결합 형태(단백질 간 거리 또는 단백질 구조)를 해치지 않도록 하였다. 도 8은 아미노산 서열 치환의 예시를 나타낸 것으로, 알라닌(Ala)을 아르기닌(Arg)으로 치환하는 경우 기존의 결합을 방해하게 되므로 세린(Ser)으로 치환해야 결합 방해를 없앨 수 있다.
ML1 펩티드의 전하를 변화시킨 펩티드 및 이들의 특징을 하기 표 5 및 표 6에 나타냈다.

상기 ML1, ML1-C3, ML1-H2의 서열을 이용하여 다음의 표 8과 같이 효율성과 안정성을 보완하기 위해 핵심 부위를 제외한 양 말단의 아미노산을 2개씩을 제거한 펩티드를 추가 제작하였다.
추가 제작된 MLP, MLP-C, MLP-H 펩티드의 서열 및 이들의 특징을 하기 표 8 및 표 9에 나타냈다.

[실험예]
에리스로포이에틴 유래 펩티드의 에리스로포이에틴 수용체(EPOR)에 대한 결합 친화도(Binding affinity) 확인
상기 실시예 1 내지 3에서 제작한 에리스로포이에틴 유래 펩티드가 타겟 지역을 가지는 에리스로포이에틴 수용체에 결합하여 작용할 수 있는지 확인하고자 SPR(Surface plasmon resonance) 기법을 이용하여 결합 강도를 확인하였다. SPR 기법은 광학적 원리를 이용하여 특정한 표지 없이 실시간으로 생체분자(biomolecules) 사이의 상호작용을 측정하는 것으로 두 분자 간의 친화력과 동역학(kinetics), 즉 Ka (association rate) 및 Kd (dissociation rate)를 분석하는 시스템이다.
구체적으로, 실시간 SPR 분석은 Reichert SPR Biosensor SR 7500C 장비(Reichert Inc., NY, USA)를 사용하여 수행하였다. 수용체 마우스 EPOR 키메라 단백질(Soluble mouse EPOR chimera protein)(R&D Systems, Minneapolis, MN, USA)은 제조사의 메뉴얼에 따라 아민 커플링 키트(BR-1000-50, GE Healthcare, USA)를 이용한 아민 커플링 절차에 의해 카르복시 메틸화된 덱스트란 메트릭스가 코팅된 칩(BR-1005-39, Pharmacia Biosensor AB) 상에 공유결합시켰다. 본 발명의 5, 2.5 및 1.25 μM의 각각의 펩티드 샘플 및 혼합된(scrambled) 펩티드는 5 ㎕/minute의 유속으로 흘렸으며 실험을 독립적으로 2회 반복 수행하였다. 신호의 정상화(normalize)를 위해 DMSO를 5 ㎕/minute으로 흘렸고, 각각의 결합 사이클 이후, 센서 칩은 25 mM의 아세트산을 20 ㎕/minute로 주입함으로써 재생하였다.
그 결과, 도 2에 나타난 바와 같이, 일 구체예의 에리스로포이에틴 유래 펩티드 농도에 따라 결과값이 증가함으로 타겟 지역을 가지는 에리스로포이에틴 수용체에 결합함으로써 작용이 가능함을 확인하였다. 또한, 표 10 및 11에 나타난 바와 같이, 일 구체예의 에리스로포이에틴 유래 펩티드는 기존에 알려진 결합 친화도(~1 uM)와 유사하게 나타남을 확인하였다.

에리스로포이에틴 유래 펩티드의 2차 알파 헬릭스 형성 확인
상기 실시예 2~3에서 합성한 에리스로포이에틴 유래 펩티드가 천연 에리스로포이에틴과 같이 안정된 알파 헬릭스(나선)를 형성하는지 알아보았다.
그 결과, 도 3에 나타난 바와 같이, 상기 실시예 2~3에서 합성한 에리스로포이에틴 유래 펩티드는 천연 에리스로포이에틴과 같은 안정된 2차 알파 헬릭스를 형성하는 것을 확인하였다.
에리스로포이에틴 유래 펩티드의 세포보호 효과 확인(1)
상기 실시예 1 내지 3에서 제작한 에리스로포이에틴 유래 펩티드가 세포 보호 효과를 나타내는지 확인하기 위해 과산화수소(H2O2)로 활성 산소 증가를 유도한 스트레스 상황에서 세포 생존력(cell viability)을 확인하였다.
구체적으로, 세포 생존력을 평가하기 위해, MTS 분석(CellTiter 96 Aqueous One Solution Cell Proliferation Assay, Promega, Madison, WI, USA)을 사용하였다. PC12 세포를 96 웰 플레이트(웰 당 5x104 세포)에 접종하였고, 150 μM 과산화수소(H2O2)로 활성 산소 증가를 유도하였다. 이후, 양성 대조군으로써 NGF(nerve growth factor)를 25 ng/㎖ 첨가하였고, 에리스로포이에틴 화합물 1 IU/ml, 실시예 1의 펩티드를 각각 0.25, 1, 2 또는 4 pM, 실시예 2 및 3의 펩티드의 경우는 0.25, 1, 2, 10 또는 100 pM, 실시예 4의 펩티드의 경우는 0.1, 1, 50 pM 또는 0.5 nM을 첨가한 후, MTS 용액 20 ㎕를 각 웰에 첨가하여 3시간 두었다. 초기 세포수(0 시간) 및 48시간 후 세포수를 측정하였다. 세포 감소에 의해 생성된 세포 내 용해성 포르마잔은 490 nm의 파장에서 VERSA MAX를 이용하여 각 96 웰 플레이트의 흡광도를 기록함으로써 결정되었다.
그 결과, 도 4에 나타난 바와 같이, 에리스로포이에틴 유래 펩티드는 활성 산소 증가로 인한 세포 사멸로부터 세포를 보호하는 것을 확인하였다. 이는 천연 에리스로포이에틴 화합물 처리에 의한 세포 보호 효과와 비슷한 결과인 것을 확인할 수 있다.
에리스로포이에틴 유래 펩티드의 세포 보호 효과 확인(2)
상기 실시예 4에서 제작한 에리스로포이에틴 유래 펩티드가 세포 보호 효과를 나타내는지 확인하기 위해 과산화수소(H2O2)로 활성 산소 증가를 유도한 스트레스 상황에서 미토콘드리아의 활성(activity)을 확인하였다.
구체적으로, PC12 세포 또는 인간 SH-SY5Y 세포를 96 웰 플레이트(웰 당 5x104 세포)에 접종하였고, 150 μM 과산화수소(H2O2)로 활성 산소 증가를 유도하였다. 이후, 양성 대조군으로써 NGF(nerve growth factor)를 25 ng/㎖ 첨가하였고, 에리스로포이에틴 화합물 1 IU/㎖, 실시예 4의 펩티드 0.1, 1, 50 pM 또는 0.5 nM을 첨가하였다.
미토콘드리아의 활성이 억제되면 미토콘드라아 막전위의 이상으로 인한 미토콘드리아 팽윤, 활성산소종, 또는 자유 라디칼 등에 의한 산화적 스트레스로 인한 기능이상, 유전적 요인으로 인한 기능이상 및 미토콘드리아의 에너지 생성을 위한 산화적 인산화 기능의 결함으로 인한 기능이상이 발생한다. 따라서 미토콘드리아 막 전위를 측정함으로 활성 측정을 할 수 있게 된다. 미토콘드리아에 TMRM(tetramethylrhodamine methyl ester) 염색을 하였으며, 미토콘드리아의 막 전압에 비례하여 TMRM 염색 수준이 증가하므로 microplate reader(excitation, 485 ㎚; emission, 535 ㎚)를 통해 TMRM 염색수준을 측정함으로써 세포 내 미토콘드리아의 막 전압을 측정하였다.
그 결과, 도 5에 나타난 바와 같이, 에리스로포이에틴 유래 펩티드는 활성 산소 증가로 인한 미토콘드리아 활성 저해를 억제하는 것을 확인하였다. 이는 천연 에리스로포이에틴 화합물 처리에 의한 효과와 비슷한 결과인 것을 확인할 수 있다.
에리스로포이에틴 유래 펩티드의 세포 증식 억제 효과 확인
상기 실시예 4에서 제작한 3개의 펩티드(ML1-L2, ML1-K2 및 ML1-R2)의 세포 증식과 같은 부작용을 확인하였다.
구체적으로, 세포 증식 정도를 확인하기 위해, MTS 분석(CellTiter 96 Aqueous One Solution Cell Proliferation Assay, Promega, Madison, WI, USA)을 사용하였다. PC12 세포를 96 웰 플레이트(웰 당 5x104 세포)에 접종하였고, 음성 대조군으로써 Scr(scrambled peptide)를 1 pM, 에리스로포이에틴 화합물 0.5, 1 또는 10 IU/ml, 실시예 4의 펩티드 1, 10 pM, 0.5 nM을 첨가한 후, MTS 용액 20 ㎕를 각 웰에 첨가하여 3시간 두었다. 초기 세포수(0 시간) 및 48시간 후 세포수를 측정하였다. 세포 감소에 의해 생성된 세포 내 용해성 포르마잔은 490 nm의 파장에서 VERSA MAX를 이용하여 각 96 웰 플레이트의 흡광도를 기록함으로써 결정되었다.
그 결과, 도 6에 나타난 바와 같이, 같이 펩티드 모두 세포증식률이 대조군과 유사하였으며, 세포증식 부작용이 없는 것을 확인하였다.
에리스로포이에틴 유래 펩티드의 세포 증식 억제 및 증강 효과 확인
상기 실시예 1 내지 3에서 제작한 펩티드의 세포 증식과 같은 부작용을 확인하기 위하여, 세포 생존율을 MTT assay 법에 따라 평가하였다.
구체적으로, PC12 세포주를 10% 우태아 혈청 (FBS, Hyclone, UT, USA)과 100 unit/ml 페니실린, 100 ㎍/㎖ 스트렙토마이신 (Hyclone, UT, USA)을 첨가한 DMEM (Dulbecco's Modified Eagle's Medium) 배양액 (Hyclone, USA)과 RPMI1640 배양액 (Hyclone, UT, USA)으로 5% CO₂가 공급되는 배양기에서 37℃조건으로 배양하였다. PC12 세포주를 5Х104 cells/㎖의 밀도로 96-웰 배양 플레이트에 분주하고, 24시간 동안 37℃, 5% CO2 조건으로 배양하였다. 이후, 상기 세포에 실시예 1 내지 3에서 제작한 펩티드를 10 ng/㎖의 농도로 제조하여 처리하여 24시간 동안 배양하였다. 이후, 5 ㎎/㎖ 농도의 MTT(3-[4,5-dimethyl-thiazol]-2,5-diphenyl-tetrazolium bromide) 시약을 20 ㎕씩 첨가하여 2시간 동안 반응시켰다. 반응 후 200 ㎕의 DMSO (dimethyl sulfoxide, Duksan, Gyeonggi-do, Korea)를 첨가하여 형성된 포마젠 (formazan)을 모두 녹인 뒤 마이크로플레이트 리더기 (microplate reader, Molecular Devices, CA, USA)를 이용하여 570㎚에서 흡광도를 측정하였다.
그 결과, 도 7에 나타난 바와 같이, 펩티드 대부분의 세포증식률이 대조군과 유사하였으나, ML1-C2 및 ML1-C3의 경우, 천연 에리스로포이에틴과 비슷한 수준의 세포 증식 효과가 있는 것을 확인할 수 있었다. 즉, 상기 펩티드는 천연 에리스로포이에틴과 유사한 세포 증식 효과를 가짐으로써 대체 물질로 활용될 수 있다.
에리스로포이에틴 유래 펩티드의 세포 보호 및 증식 효과 비교
상기 실시예 3에서 제작한 7개의 펩티드(ML1, ML1-C1, ML1-C2, ML1-C3, ML1-H1, ML1-H2, ML1-H3)의 세포 보호 및 증식 효과를 EPO와 통계적으로 비교하였다.
구체적으로, 도 4와 도 7에서의 효과를 EPO에 의한 효과를 기준으로 보정하여 정량화한 후, K평균 알고리즘(K-means clustering algorithm)을 통해 클러스터링을 수행하였다.
그 결과, 도 9에 나타난 바와 같이, ML1-C1, ML1-C2와 ML1-C3는 세포 보호 및 효과 면에서 EPO와 비슷한 효과를 가진 것으로 클러스터링 되었다. 반면, ML1, ML1-H1, ML1-H2, ML1-H3은 세포 보호 효과면에서는 EPO 그룹들과 큰 차이를 보이지 않으나, 세포 증식 효과 면에서는 큰 차이를 보이는 것으로 분석되었다.
에리스로포이에틴 유래 펩티드의 in vivo 조혈 효과 확인
상기 실시예 5에서 제작한 펩티드 MLP, MLP-C, MLP-H의 생쥐 생체 내에서 조혈 활성을 측정하였다.
구체적으로, 제작한 펩티드 또는 음성 대조군으로써 Scr(scrambled peptide)를 0 ㎍/㎏, 1 ㎍/㎏, 100 ㎍/㎏을 14일 동안 매일 반복적으로 생쥐에 복강 주사를 통해 투여하였다. 이후, 체중 검사 및 혈액 검사를 수행하였다.
그 결과, 도 10에 나타난 바와 같이, 모든 펩티드는 생쥐에 대하여 유의적인 체중 변화를 나타내지 않는 것을 확인할 수 있었다. 또한, 도 11a에 나타난 바와 같이 MLP-C를 처리한 생쥐에서만 혈액 중 적혈구가 차지하고 있는 용적의 비중인 헤마토크릿(hematocrit)이 투여 용량에 따라 유의미하게 증가하였으며, 도 11c에 나타난 바와 같이, 100 ㎍/㎏를 투여한 생쥐의 경우, 적혈구 수와 혈색소(헤모글로빈)의 함량, 평균적혈구혈색소농도(MCHC) 평균적혈구혈색소(MCH)가 유의미하게 증가됨을 확인할 수 있었다. 반면, 도 11d에 나타난 바와 같이, 혈소판 수치는 크게 변화하지 않는 것을 확인할 수 있었다. 즉, 일 양상에 따른 펩티드는 생체 내에서 조혈작용을 유도할 수 있는바, 조혈 기능을 개선할 수 있을 뿐만 아니라, 조혈장애의 예방 또는 치료에 이용될 수 있다.
에리스로포이에틴 유래 펩티드의 에리스로포이에틴 수용체(EPOR)에 대한 활성화 조절 확인
상기 실시예 5에서 제작한 펩티드 중 MLP-C, MLP-H에 의한 에리스로포이에틴 수용체의 활성화 정도를 천연 에리스로포이에틴과 비교하였다.
구체적으로, 녹색형광단백질을 퓨전(fusion)한 에리스로포이에틴 수용체를 발현할 수 있는 DNA 플라스미드를 HEK293 세포주에 유전자 도입하고 녹색형광단백질을 퓨전한 에리스로포이에틴을 안정적으로 발현하는 stable 세포주를 제작하였다. 양성 대조군으로서 EPO, MLP-C 또는 MLP-H를 처리한 후 0분, 30분, 60분, 90분에 샘플링하여 웨스턴블랏을 수행한 후 에리스로포이에틴 수용체의 활성도를 ERK1/2의 인산화 정도를 통해 확인하였다.
그 결과, 도 12에 나타난 바와 같이, EPO와 MLP-C는 30분에 최대 활성을 나타내고 이후 감소하는 반면, MLP-H는 30분 이후에도 활성화가 지속되었음을 확인하였다. 즉, MLP-C는 에리스로포이에틴 수용체에 대하여 짧지만 강한 활성화를 나타내는 반면, MLP-H는 약하지만 장시간 동안의 활성화를 나타내는 바, 일 양상에 따른 에리스로포이에틴 유래 펩티드는 에리스로포이에틴 수용체의 활성화를 조절할 수 있다. 따라서, 세포 증식 효과를 필요로 하는 빈혈 등과 같은 질환의 치료제로 이용될 수 있다.
Claims (19)
- 서열번호 26 내지 28로 구성된 군으로부터 선택되는 어느 하나 이상의 서열로 기재되는 펩티드.
- 청구항 1에 있어서, 상기 펩티드는 에리스로포이에틴(erythropoietin) 단백질 서열로부터 유래된 것인 펩티드.
- 서열번호 1 내지 25로 구성된 군에서 선택되는 어느 하나 이상의 펩티드를 유효성분으로 포함하는 세포증식 또는 분화 촉진용 조성물.
- 청구항 3에 있어서, 상기 펩티드는 에리스로포이에틴 수용체에 결합하는 것인 조성물.
- 청구항 3에 있어서, 상기 펩티드는 알파 헬릭스 구조를 형성하는 것인 조성물.
- 청구항 3에 있어서, 상기 펩티드는 세포보호 활성을 갖는 것인 조성물.
- 청구항 3에 있어서, 상기 세포는 조혈모세포, 지방세포, 췌장세포, 근육세포, 혈관세포 또는 피부세포인 것인 조성물.
- 서열번호 1 내지 25로 구성된 군에서 선택되는 어느 하나 이상의 펩티드를 유효성분으로 포함하는 조혈 기능 개선용 조성물.
- 서열번호 1 내지 25로 구성된 군에서 선택되는 어느 하나 이상의 펩티드를 유효성분으로 포함하는 빈혈 질환의 예방 또는 치료용 약학적 조성물.
- 청구항 9에 있어서, 상기 조성물은 조혈줄기세포의 증식을 촉진시키는 것인 조성물.
- 청구항 9에 있어서, 급성 또는 만성 빈혈, 신장 질환에 의한 빈혈, 신부전에 의한 빈혈, 혈액 질환에 의한 빈혈, 방사선요법-유발 빈혈, 화학요법-유발 빈혈, 수술 과정에 의한 빈혈, 감염에 의한 빈혈, 영양결핍성 빈혈, 비정상적인 적혈구생성, 조산의 초기 빈혈 및 이들의 혼합 질환로 구성된 군에서 선택된 것인 조성물.
- 서열번호 1 내지 25로 구성된 군에서 선택되는 어느 하나 이상의 펩티드를 유효성분으로 포함하는 빈혈 질환의 예방 또는 개선용 건강기능성 식품 조성물.
- 서열번호 1 내지 25로 구성된 군에서 선택되는 어느 하나 이상의 펩티드를 유효성분으로 포함하는 조혈장애의 예방 또는 치료용 약학적 조성물.
- 청구항 13에 있어서, 조혈장애는 재생 불량성 빈혈, 악성 림프종 또는 백혈병, 만성 간 장애, 신부전, 중증 감염증, 골수 장애성 혈소판 감소증, 특발성 혈소판 감소성 자반병(ITP), 혈소판 무력증, 림프구감소증, 호중구감소증, 단핵구감소증, 과립구감소증, 골수 형성이상 증후군 및 골수 증식성 질환으로 구성된 군에서 선택되는 것인 조성물.
- 서열번호 1 내지 25로 구성된 군에서 선택되는 어느 하나 이상의 펩티드를 유효성분으로 포함하는 조혈장애의 예방 또는 개선용 건강기능성 식품 조성물.
- 서열번호 1 내지 28로 구성된 군에서 선택되는 어느 하나 이상의 펩티드를 포함하는 조성물의 치료학적 유효량을 그를 필요로 하는 개체에 투여하는 단계를 포함하는 빈혈 질환을 치료하는 방법.
- 서열번호 1 내지 28로 구성된 군에서 선택되는 어느 하나 이상의 펩티드를 포함하는 조성물의 치료학적 유효량을 그를 필요로 하는 개체에 투여하는 단계를 포함하는 조혈장애를 치료하는 방법.
- 빈혈 질환의 치료를 위한 약제의 제조를 위하여 서열번호 1 내지 28로 구성된 군에서 선택되는 어느 하나 이상의 펩티드를 포함하는 조성물의 용도.
