Abstract
本発明は、pKbが3~7の弱塩基性イオン交換体を触媒に用い、脂肪酸エステルと糖類を反応させエステル交換することを特徴とする糖脂肪酸エステルの製造方法を提供する。 本発明によれば、強塩基性陰イオン交換体を用いた場合に生成物の糖脂肪酸エステルの分解が同時進行し、反応の制御が困難であるという問題が解消され、糖脂肪酸エステルを効率的に製造することができる。
Description
本発明は脂肪酸エステルと糖類とのエステル交換反応による糖脂肪酸エステルの製造方法に関する。さらに詳しく言えば、弱塩基性の陰イオン交換体を触媒とする脂肪酸エステルと糖類とのエステル交換反応による糖脂肪酸エステルの製造方法に関する。本発明の方法は、緩和な条件で操作でき、高い反応率で糖脂肪酸エステルを製造することができる。
糖のスクロース(Suc)は、高純度品を安価に入手できることから、化学原料として様々な利用法の提案がなされている。現在、工業的に生産されているスクロース誘導体の一つとして、糖脂肪酸エステルがある。
糖脂肪酸エステルは、親水基の糖と親油基の植物油由来の脂肪酸(FA)がエステル結合した非イオン性の界面活性剤である。糖が有する複数の水酸基(OH基)に結合させる脂肪酸の数によって親水、親油性を変化させることができるため、広範な親水・親油バランス(HLB)値を持つ。糖脂肪酸エステルは乳化作用のほかに、可溶化能、分散能、潤滑能など高い機能性を有している。
また、近年、製薬分野で、糖脂肪酸エステルの錠剤の結合剤としての用途の研究が行われ、医薬品分野での利用も期待されている。

このように、糖脂肪酸エステルは、安価なスクロースと植物油から製造される高付加価値のバイオマス由来化成品であるが、親水性の糖と脂肪酸の混和性が低く合成が困難であることが知られている。
現在、糖脂肪酸エステルは、スクロースと脂肪酸アルキルエステル(例えば、脂肪酸メチルエステル)とから溶媒に両親媒性のジメチルスルホキシド(DMSO)を用い、触媒としてアルカリ触媒を用いた均一相でのエステル交換反応により工業的に製造されている。この方法では、転化率を高めるために副生メタノールを除去する減圧条件が必須であり、脂肪酸メチルと触媒の反応により石鹸を副生すること、均相アルカリ触媒の分離が困難であること等の問題があり、コスト高の要因となっている。このような問題に対処するために、例えば、充填塔と複数個の凝縮器を直列に配置した反応器を用い、第1段凝縮器の凝縮液を充填塔の塔頂に還流する手法が提案されている(特開平6-122694号公報;特許文献1)。
また、糖脂肪酸エステルの製造方法としては、スクロースと脂肪酸の低級アルカノールエステルまたはグリセリド類とをアルカリ金属でイオン交換された陽イオン交換樹脂の存在下でエステル交換反応させる方法が開示されている(特開昭63-179884号公報;特許文献2)。しかしながら、四級アンモニウム基を官能基として有する強塩基性陰イオン交換樹脂あるいは三級アミンを官能基として有する弱塩基性陰イオン交換樹脂を用いた反応では、糖脂肪酸エステルは減圧条件下でわずかに生成するのみで、満足な結果は得られていない。
本発明者等は、先に脂肪酸エステルと糖類とのエステル交換反応を、強塩基性陰イオン交換体を用い糖類を樹脂へ吸着させる工程、及び糖脂肪酸エステルを合成させる工程の2段階からなる工程により行う方法を提案している(WO2018/038103;特許文献3)。この方法は、大気圧下で実施可能であること、石鹸が副生しないこと、及び触媒の分離が容易であること等の利点がある。しかし、強塩基性陰イオン交換体を用いる方法では、生成した糖脂肪酸エステルの分解が同時に進行し、反応の制御が困難な場合があり、製造の歩留まりが低いという問題がある。
一方、弱塩基性固体触媒は一般に触媒としてではなく医薬品の精製や水処理に用いられている。前記特許文献2には、「陰イオン交換樹脂は全て塩基性を有するが、四級アンモニウム基を官能基として有する塩基性陰イオン交換樹脂あるいは三級アミンを官能基として有する塩基性陰イオン交換樹脂を用いて該反応を行った場合には満足な結果が得られなかった」(第11頁)ことが記載されている。
本発明の目的は、生成物である糖脂肪酸エステルの分解が同時に進行し、反応の制御が困難であること、製造の歩留まりが低くなることなどの上記強塩基性陰イオン交換体を触媒に用いるエステル交換反応の課題を解決した糖脂肪酸エステルの製造方法を提供することにある。
本発明の目的は、生成物である糖脂肪酸エステルの分解が同時に進行し、反応の制御が困難であること、製造の歩留まりが低くなることなどの上記強塩基性陰イオン交換体を触媒に用いるエステル交換反応の課題を解決した糖脂肪酸エステルの製造方法を提供することにある。
本発明者らは、弱塩基性陰イオン交換体について鋭意検討の結果、弱塩基性陰イオン交換体を触媒に用いることで、エステル交換反応能が維持されたまま生成物の分解が抑制され、前記課題が解決できることを見出し、本発明を完成した。
本発明は以下の[1]~[11]の糖脂肪酸エステルの方法に関する。
[1] 弱塩基性固体触媒を用い脂肪酸エステルと糖類をエステル交換反応させることを特徴とする糖脂肪酸エステルの製造方法。
[2] 弱塩基性固体触媒が弱塩基性陰イオン交換体である前項1に記載の糖脂肪酸エステルの製造方法。

本発明は、これまで触媒として利用の報告のなかった弱塩基性陰イオン交換体を触媒に用いて、脂肪酸エステルと糖類をエステル交換反応させる糖脂肪酸エステルの製造方法を提供したものである。
本発明によれば、(1)大気圧下で石鹸を副生することなく、糖脂肪酸エステルを製造することができる。(2)弱塩基性陰イオン交換体を触媒として用いることで、強基性イオン交換体触媒を用いた場合に進行する生成物の分解が抑制される。(3)特に、ポーラス型の弱塩基性陰イオン交換体を用いることによって、強塩基性陰イオン交換体を用いた場合の3倍程度まで糖脂肪酸エステルの収率が増大し、低コストでの製造が可能になる。
本発明によれば、(1)大気圧下で石鹸を副生することなく、糖脂肪酸エステルを製造することができる。(2)弱塩基性陰イオン交換体を触媒として用いることで、強基性イオン交換体触媒を用いた場合に進行する生成物の分解が抑制される。(3)特に、ポーラス型の弱塩基性陰イオン交換体を用いることによって、強塩基性陰イオン交換体を用いた場合の3倍程度まで糖脂肪酸エステルの収率が増大し、低コストでの製造が可能になる。
以下に、本発明の実施形態について詳細に説明する。
本発明の糖脂肪酸エステルの製造方法は、弱塩基性固体触媒を用い脂肪酸エステルと糖類をエステル交換反応させることを特徴とする。

[弱塩基性固体触媒]
本発明で使用する弱塩基性固体触媒の典型例は、弱塩基性陰イオン交換体である(以下の説明では、弱塩基性陰イオン交換体あるいは弱塩基性陰イオン交換樹脂を、それぞれ陰イオン交換体あるいは陰イオン交換樹脂と略記することがある。)。陰イオン交換体の形態は特に限定されず、粒状、膜状、繊維状などが挙げられる。
本発明で使用する弱塩基性陰イオン交換体は、塩基解離定数(pKb)が3~7の範囲のものであり、好ましくは、pKbは5程度である。このような陰イオン交換体は市販品を利用できる。具体例としては、Diaion WA10、WA20、WA21J、WA30(三菱ケミカル(株)製)、Lewatit MP-62、VP OC 1065(ランクセス社製)、Amberlite IRA-478、IRA-68、IRA-96、IRA-98、XE-583、Amberlyst A21、DUOLITE A7、A568、Dowex 66、M-43、Monosphere 66、77、Marathon WBA(ダウ・ケミカル社製)などを挙げることが出来る。
本発明で使用する弱塩基性固体触媒の典型例は、弱塩基性陰イオン交換体である(以下の説明では、弱塩基性陰イオン交換体あるいは弱塩基性陰イオン交換樹脂を、それぞれ陰イオン交換体あるいは陰イオン交換樹脂と略記することがある。)。陰イオン交換体の形態は特に限定されず、粒状、膜状、繊維状などが挙げられる。
本発明で使用する弱塩基性陰イオン交換体は、塩基解離定数(pKb)が3~7の範囲のものであり、好ましくは、pKbは5程度である。このような陰イオン交換体は市販品を利用できる。具体例としては、Diaion WA10、WA20、WA21J、WA30(三菱ケミカル(株)製)、Lewatit MP-62、VP OC 1065(ランクセス社製)、Amberlite IRA-478、IRA-68、IRA-96、IRA-98、XE-583、Amberlyst A21、DUOLITE A7、A568、Dowex 66、M-43、Monosphere 66、77、Marathon WBA(ダウ・ケミカル社製)などを挙げることが出来る。
後述の実施例で使用したWA30、WA20、及びWA21J、及び比較例で用いた強塩基性陰イオン交換樹脂であるPA306S(いずれも三菱ケミカル(株)製)の型式、官能基、特性(塩基性の程度)、塩基解離定数(pKb)、イオン交換容量(mol/m3-resin)、粒子径(mm)、比表面積(m2/g-resin)を表1に示す。
WA30、WA20、及びWA21Jは弱塩基性で、WA20はポーラス型、WA30、WA21Jはハイポーラス型、PA306Sは強塩基性でポーラス型の構造を有する。各樹脂は粒子内部が均一なゲル型を基本とし、ポーラス型はゲル型樹脂に物理的な孔(細孔)を開けた構造を有する樹脂である。ハイポーラス型はポーラス型より細孔量を増やし比表面積を大きくした樹脂である。各樹脂の官能基は、PA306Sはトリメチルアミン型、WA30はジメチルアミン型、WA20、WA21Jはポリアミン型であり、この順に塩基性度が高い。
弱塩基性陰イオン交換樹脂は、購入時(工場出荷時)には水膨潤状態であり、極性の低い脂肪酸エステルは樹脂内部に拡散しにくい。そこで、本発明では反応溶媒で膨潤化させた後に使用する。また、強塩基性樹脂は、工場出荷時は官能基が不活性なCl型で水膨潤の状態にある。そのため、不活性Cl型から活性OH型への官能基置換と溶媒膨潤化を行う活性化処理が必要となる。これらの処理は、本発明者等が構築した樹脂繰り返し利用のための再生処理法に従い行うことができる(化学工学論文集, 42, 30-36 (2016))。
陰イオン交換樹脂は、不溶性担体として樹脂骨格が種々の化学構造を有するものを使用できる。具体的には、例えば、ジビニルベンゼン等で架橋されたポリスチレン、及びポリアクリル酸、架橋ポリ(メタ)アクリル酸エステル、フェノール樹脂等の合成高分子や、セルロース等の天然に生産される多糖類の架橋体等が挙げられる。中でも合成高分子が好ましく、架橋ポリスチレンが更に好ましい。
本発明の方法において、エステル交換反応の原料として用いる脂肪酸エステル及び糖類は、生成物である糖脂肪酸エステルの種類、使用目的等に応じて、当業者に公知の任意の化合物から選択することが出来る。原料化合物は、当業者に公知の任意の方法で調製してもよいし、市販されている各種の脂肪酸エステル及び糖類を用いてもよい。
例えば、炭素原子数1~8、好ましくは、1~5の直鎖または分岐鎖の炭化水素骨格を有する低級アルコールと、例えば、ラウリン酸、ミリスチン酸、パルミチン酸、ステアリン酸、オレイン酸、べヘニン酸、及びエルカ酸等の炭素数1~30の脂肪酸からなる脂肪酸エステルを用いることが出来る。
糖類としては、単糖類、スクロース等の2糖類、及び多糖類から選択される1つまたはそれ以上の糖類を用いることが出来る。
本発明方法において用いる脂肪酸エステルの好ましい具体例としては、例えば、実施例で使用しているオレイン酸メチルや、ラウリン酸メチル、ミリスチン酸メチル、パルミチン酸メチル、ステアリン酸メチル等が挙げられ、糖の具体例としてはスクロース等を挙げることが出来る。
本発明の方法においては、親油性の脂肪酸エステルと親水性の糖類は極性が大きく異なり混和し難いため、両親媒性の、例えばジメチルスルホキシド、ジメチルホルムアミド、エチルメチルケトン、酢酸エチル、2-プロパノール、プロピレングリコール、メタノール、エタノール、及び2-メチル-1-プロパノール等の当業者に公知の溶媒系で陰イオン交換樹脂と反応させることが出来る。
本発明方法においては、実質的に糖類を予め弱塩基性陰イオン交換樹脂に吸着させて用いることが好ましい。例えば、脂肪酸エステルと糖類とのエステル交換反応に先立ち、弱塩基性陰イオン交換樹脂に実質的に糖類を吸着させる工程(吸着工程)、及びその後吸着工程で得られた陰イオン交換樹脂を用いて脂肪酸エステルと糖類とのエステル交換反応により糖脂肪酸エステルを合成する工程(合成工程)の2工程によって実施することが好ましい。
前記の各工程における反応物と陰イオン交換樹脂との接触方式については、バッチ法(回分系)、連続法(流通系)など、当業者に公知の任意の方式で行うことが出来る。装置の形態としては、処理槽を設けたもの、循環系や向流系で樹脂移送するものなどが挙げられる。接触方法としては、流通(イオン交換樹脂の充填層あるいは流動層に通液する方法)、撹拌(撹拌槽を用いる方法)、振とう(振とう型反応器)などが挙げられる。供給原料の導入口、生成物質の回収口が一定のカラム通液型、展開床(エクスパンデッドベッドカラム)の他、回分型を用いることもできる。
なお、バッチ法(回分系)では、反応の進行に伴い反応物濃度が減少するため、糖類濃度を高く維持し続けることが困難な場合には、必要に応じて反応物を追添することができる。
一方、樹脂を充填した反応器を用いる連続法(流通系)は、高い糖類濃度を維持することによって、目的の糖エステル合成反応を優先的に進行させることが出来るので好ましい。

連続法(流通系)で実施する場合は、第一段階として、糖類を含む反応液を陰イオン交換樹脂に供給して糖類の吸着工程(吸着工程)を実施し、次に第二段階として、陰イオン交換樹脂に脂肪酸エステルのみ、または脂肪酸エステルと糖類とを含む反応液を供給してそれらの間のエステル交換反応による糖脂肪酸エステルの合成工程(合成工程)を実施することが好ましい。特に、合成工程中は、触媒である陰イオン交換樹脂内の糖類を高濃度に維持するように脂肪酸エステルと糖類とを含む反応液を供給し、反応液の糖類濃度を高くすることが好ましい。
合成工程で使用する反応液中の糖類濃度(脂肪酸エステルに対する糖類のモル濃度比)は1:0.5~1:5の範囲であることが好ましく、1:0.5~1:3の範囲であることがより好ましい。糖類のモル濃度比が0.5未満の場合、陰イオン交換樹脂の触媒活性が消失してしまい、その後、糖類が陰イオン交換樹脂に取り込まれても脂肪酸エステルとの間でエステル交換反応が効率的に生じないことがある。一方、糖類のモル濃度比が5を超える場合は、反応液の濃度が高くなり過ぎ、またコスト高となって生産性の点で好ましくない。また、糖類を過剰に入れすぎると、製品から糖類を除去する操作が必要となるため生産性が悪くなる。
本発明方法において所望の目的を達成するためには、上記の2工程の操作を任意の生産プロセスと組み合わせてもよい。例えば、目的生成物である糖脂肪酸エステルをその他の生成物であるアルコール及びその他の不純物等から分離・精製する工程を更に含んでもよい。すなわち、本発明の弱塩基性陰イオン交換体を触媒として用いる糖脂肪酸エステルの製造方法で脂肪酸エステルと糖類とのエステル交換反応を実施した後に得られる組成物は、溶媒中に生成物(糖脂肪酸エステル)と、未反応の反応物(糖類や脂肪酸エステル)とが含まれた組成物、あるいはアルコール等が含まれた組成物である。これら組成物から生成物である糖脂肪酸エステルを取得し精製するためには、抽出工程と水洗工程を所望の回数繰り返し行い、抽出液から抽出溶媒を留去後乾燥して、溶媒を含まない生成物を含む混合物を得ることができる。この生成物を含む混合物から、公知の任意の手段、例えば、蒸留工程、膜分離工程、分取液体クロマト分離工程等を組み込んで高純度の糖脂肪酸エステルを得ることができる。
本発明の方法におけるその他の条件や手段、例えば、各反応物のモル濃度及びモル比、陰イオン交換樹脂の使用量、陰イオン交換樹脂と反応物との量比、反応系の温度・圧力、及び反応時間等は、使用する各反応物及び陰イオン交換樹脂の種類等を考慮して、公知の任意のものから適宜選択することができる。例えば、各工程の反応時間は反応温度、陰イオン交換樹脂の使用量等に応じて適宜設定することが出来る。反応は通常、20~100℃、好ましくは、30~70℃で実施する。反応圧力は特に制限ない。常圧下で実施するのが操作上簡便であるが、必要に応じて1~10気圧程度に加圧、あるいは、0.002~1気圧程度に減圧してもよい。
以下に糖脂肪酸エステル合成の実施例及び比較例を挙げて説明するが、本発明は下記の記載に限定されるものではない。
反応原料:
脂肪酸エステルとしてオレイン酸メチル(OAMeと略記することがある。)〈富士フイルム和光純薬(株)製〉、及び糖としてスクロース(Sucと略記することがある。)(富士フイルム和光純薬(株)製)を用いた。
溶媒:工業的製造法と同じジメチルスルホキシド(DMSO)を用いた。
触媒:市販(三菱ケミカル(株)製)の4種の多孔性樹脂を用いた。各樹脂の型、官能基(functionality)、特性(塩基性の程度)、pKb、イオン交換容量(mol/m3)、粒子径(mm)、及び比表面積(m2/g)は先の表1に示した通りである。
脂肪酸エステルとしてオレイン酸メチル(OAMeと略記することがある。)〈富士フイルム和光純薬(株)製〉、及び糖としてスクロース(Sucと略記することがある。)(富士フイルム和光純薬(株)製)を用いた。
溶媒:工業的製造法と同じジメチルスルホキシド(DMSO)を用いた。
触媒:市販(三菱ケミカル(株)製)の4種の多孔性樹脂を用いた。各樹脂の型、官能基(functionality)、特性(塩基性の程度)、pKb、イオン交換容量(mol/m3)、粒子径(mm)、及び比表面積(m2/g)は先の表1に示した通りである。
触媒の前処理:
弱塩基性陰イオン交換樹脂は、反応溶媒で膨潤化させてから使用した。また、強塩基性樹脂は、特許文献3の実施例に準じて、活性OH型へ官能基置換し溶媒膨潤化を行う活性化処理を行ってから使用した。

反応条件:
弱塩基性樹脂を用いた合成(実施例1~3)では、前処理後の樹脂をSuc0.66 mol/dm3を含むDMSO溶液に接触させ、十分にSucを吸着させた。次に、樹脂を、Suc0.30 mol/dm3とOAMe0.10mol/dm3を含む反応液に、湿質量で20質量%となるように投入し、60℃で十分に振盪して反応を行った。
強塩基性樹脂を用いた合成(比較例1)は、特許文献3の実施例に準じて行った。
弱塩基性樹脂を用いた合成(実施例1~3)では、前処理後の樹脂をSuc0.66 mol/dm3を含むDMSO溶液に接触させ、十分にSucを吸着させた。次に、樹脂を、Suc0.30 mol/dm3とOAMe0.10mol/dm3を含む反応液に、湿質量で20質量%となるように投入し、60℃で十分に振盪して反応を行った。
強塩基性樹脂を用いた合成(比較例1)は、特許文献3の実施例に準じて行った。
各実験では、生成物OASuc濃度を蒸発光散乱検出器を備えた液体クロマトグラフィを用いて測定した。また、反応前後での物質収支を確認するために、Suc濃度は蒸発光散乱検出器を備えた液体クロマトグラフィで、OAMe濃度は紫外可視分光検出器を備えた液体クロマトグラフィを用いて測定した。
[結果]
図1に実施例1~3と比較例1における生成物のOASuc濃度の経時変化を示す。強塩基性イオン交換樹脂PA306Sを用いた比較例1(△)では、反応開始後OASuc濃度が増加し、最大濃度に達した後、徐々に減少した。これは、生成したOASucが分解したためと考えられる。一方、弱塩基性イオン交換樹脂を用いた実施例1~3では、いずれの場合もOASuc濃度が緩やかに増加した後一定となった。OASuc濃度の最大値は、WA21J(■)< WA30(◆)< WA20(●)の順に高くなった。最も高かったWA20では、強塩基性イオン交換樹脂PA306Sを用いた比較例1での最大濃度の2.8倍となった。弱塩基性イオン交換樹脂を用いた場合、反応前後での物質収支を確認したところ、生成物OASucの分解は生じていないことが明らかとなった。
図1に実施例1~3と比較例1における生成物のOASuc濃度の経時変化を示す。強塩基性イオン交換樹脂PA306Sを用いた比較例1(△)では、反応開始後OASuc濃度が増加し、最大濃度に達した後、徐々に減少した。これは、生成したOASucが分解したためと考えられる。一方、弱塩基性イオン交換樹脂を用いた実施例1~3では、いずれの場合もOASuc濃度が緩やかに増加した後一定となった。OASuc濃度の最大値は、WA21J(■)< WA30(◆)< WA20(●)の順に高くなった。最も高かったWA20では、強塩基性イオン交換樹脂PA306Sを用いた比較例1での最大濃度の2.8倍となった。弱塩基性イオン交換樹脂を用いた場合、反応前後での物質収支を確認したところ、生成物OASucの分解は生じていないことが明らかとなった。

- 弱塩基性固体触媒を用い脂肪酸エステルと糖類をエステル交換反応させることを特徴とする糖脂肪酸エステルの製造方法。

- 前記弱塩基性陰イオン交換体のpKbが3~7である請求項2に記載の糖脂肪酸エステルの方法。
- 前記弱塩基性陰イオン交換体が、ポーラス型の弱塩基性陰イオン交換樹脂である請求項2または3に記載の糖脂肪酸エステルの製造方法。
- 糖類を弱塩基性固体触媒へ吸着させる工程(吸着工程)、及び糖類と脂肪酸エステルとのエステル交換反応により糖脂肪酸エステルを合成する工程(合成工程)を含む請求項1~4のいずれかに記載の糖脂肪酸エステルの製造方法。
