Abstract
본 발명은 지카 바이러스를 베로 세포에서 계대배양하며 선별하고 적응시키는 과정을 통해 확립한 인간용 지카 백신의 제조에 탁월한 생산성 및 우수한 백신 효능을 나타내는 지카 바이러스 변이주 및 이를 포함하여 제조된 지카 백신 조성물에 관한 것이다.
Description
본 발명은 지카 바이러스를 베로 세포에서 계대배양하며 선별하고 적응시키는 과정을 통해 확립한 인간용 지카 백신의 제조에 탁월한 생산성 및 우수한 백신 효능을 나타내는 지카 바이러스 변이주 및 이를 포함하여 제조된 지카 백신 조성물에 관한 것이다.
지카 바이러스는 모기 매개의 Flavivirus의 일종으로, 1947년 우간다 원숭이의 황열을 연구하는 과정에서 최초로 발견되었으며, 이 후
Aedes
africanus 모기에서 발견되었다(Dic GW et al., Trans R Soc Trop Med Hyg, 46:509-20, 1952). 1952년, 우간다와 탄자니아에서 최초의 인체 감염사례가 보고되었으며, 2007년 마이크로네시아 얍(Yap) 섬에서 200명 감염사례가 보고되는 등 지금까지 아프리카, 아메리카, 아시아, 태평양지역 전역에 걸쳐 지카 바이러스 감염사례가 보고되어왔다.
1960년대의 지카 감염 증상은 가벼운 발열을 동반한 독감 증상에 불과하였으나, 2015년 브라질에서 발병한 지카 감염은 길랑-바레(Guillain-Barre) 증후군과의 연관성을 보였으며, 같은 해 지카 감염이 소두증(microcephaly)을 일으킨다는 보고가 잇따랐다. 2007년 이후 총 76개국에서 지카 바이러스의 전이가 보고되었으며, 그 중 6개 국가에서 성관계에 의한 감염사례가 보고되었다.
지카 바이러스는 주로 벡터-유래된, 피복된, 양성-센스, RNA 단일 나선의 특징을 가진 66개 플라비 바이러스과(Flaviviridae) 중의 하나이다 (Chambers, T. J. et al. Ann. Rev. Microbiol, 44:649-88,1990). 지카 바이러스는 총 3,419개의 아미노산을 코딩하는 10,794개의 핵산을 지니고 있으며, 구조적으로는 다른 flavivirus와 마찬가지로 당화된 표피(E)와 막(M)단백질로 구성되어 있다(Kuno G et al., Arch Virol, 152:687-96, 2007).
2015년 브라질에서 발병한 지카 감염 이후, 지카 백신 개발에 많은 연구진이 노력을 쏟고 있으나, 아직까지 치료용 목적으로 환자에게 사용할 수 있는 지카 백신은 개발되지 않았다. 개발 중인 지카 바이러스 백신 후보물질은 약 30개 정도이고, 이들 중 선도그룹의 지카 백신은 임상1상 또는 임상2상에서 안전성과 효능을 평가하고 있다 (Fernandez, E et al., Current Opinion in Virology, 23:59-67, 2017).
지카 백신은 크게 DNA 백신, 불활화 백신, 약독화 백신, mRNA 백신 형태로 개발 중이며, 다른 flavivirus 백신을 개발한 기술에 근간하여 개발되고 있다. 2016년부터 많은 수의 연구그룹이 지카 바이러스에 특이적인 중화항체와 단일클론항체를 개발하고 있고, 최근 'ZIKV-117'이라는 중화항체가 지카 바이러스에 감염된 쥐에서 바이러스의 증식을 억제하고 발병을 줄여주는 결과가 보고되기도 하였다. 하지만, 개발한 항체는 뎅기 바이러스에 cross-reactivity를 가지며, 이로 인해 질병의 antibody-dependent enhancement가 문제가 되고 있다.
한편, 지카 DNA백신은 지카 바이러스의 envelop(E) 또는 premembrane(PrM) 단백질을 발현하는 백신으로, DNA 백신은 생산이 쉽고 안전성이 높다. VRC/NIAID에서 개발하고 있는 VRC705 백신이 대표적이며, 이 DNA 백신은 H/PF/213 지카 바이러스주의 prM-Env를 발현한다. 가장 진척된 DNA백신 개발은 2016년에 쥐와 영장류 실험에서 항체 중화역가와 백신 효능을 검증한 후(Dowd KA et al., Science, 354(6309):237-240, 2016), 2017년 임상 2상에 돌입하였다(NCT03110770).
불활화 지카 백신 개발은 Walter Reed Army Institute of Research(WRAIR)에서 착수하였으며, WNV, JEV 등과 같은 다른 Flavivirus의 불활화 백신 개발 플랫폼을 적용하였다. 이 불활화 지카 백신은 PRVABC59(Puerto Rico) 지카 바이러스주를 이용하여 개발한 것으로, 베로 세포에서 배양하고 포르말린으로 불활성화한 백신이다. WRAIR의 불활화 지카 백신은 Balb/c 마우스에서 1ug 접종 4주 후 중화항체가 PRNT
50=15의 값을 나타냈으며(Rafael A. Larocca at al., Nature, 536(7617):474-478, 2016) non-human primate(NHP)에서 추가적인 백신의 효능평가를 마친 후 2016년에 첫 임상시험에 돌입하였다. Bharat Biotech에서 MR766 지카 바이러스를 이용하여 개발 중인 불활화 백신은 Balb/c 마우스에 5ug씩 2회 접종 시 약 PRNT
50=200의 값이 나타났으며(US20170014502A1), 현재 임상 1상을 진행 중이다. 상기 백신 개발에서 플라크 감소 중화 시험(PRNT)은 백신의 효능을 평가하는 시험 방법으로, flavivirus의 혈청진단 표준 평가방법이다. 지금까지의 백신개발연구 사례와 WHO 보고에서 백신의 혈청학적 양성효능을 나타내는 기준으로 PRNT
50=10 이상을 제시하고 있으며, flavivirus에 속하는 뎅기 바이러스, 황열 바이러스, 일본뇌염 바이러스에 대한 백신 개발 역시 이 기준을 따르고 있다(WHO, 2007; J. Flipse and JM. Smit, PLoS neglected tropical diseases, 9(6):e0003749, 2015; XF Li et al., Nature communications, 9(1):673, 2018).
DNA백신과 불활화 백신 외에도 Valera Moderna에서 개발하고 있는 mRNA 백신(mRNA-1325), Themis Bioscience에서 개발 중인 재조합 약독화 백신(MV-Zika), SEEK와 NIH에서 공동개발 중인 단백질 서브유닛 백신(AGS-v) 등이 임상 1상 단계에서 활발히 연구개발되고 있다.
다양한 지카 백신 플랫폼 중 가장 유력한 후보인 불활화 지카 백신은 DNA 백신에 비해 생산이 오래 걸리고 복잡하지만, DNA백신보다 높은 중화항체 역가를 보여 우수한 백신으로 여겨진다. 특히 같은 Flavivirus에 속하는 황열 바이러스, 일본뇌염 바이러스 등에 대한 불활화 백신을 성공적으로 개발한 기술을 기반으로 하기 때문에 가장 유력한 백신 후보라고 여겨진다. 이러한 불활화 지카 백신 개발을 위해서는 사람 백신의 생산세포 기질로서 인정받아온 베로 세포 또는 사람 이배체 세포와 같은 표준 세포주에서 효율적으로 지카 백신을 생산하는 기술이 필요하다.
베로 세포는 원숭이 신장으로부터 유도된 형질전환 비-종양유도성 세포이다. 베로 세포는 큰 규모의 세포 배양에 보다 쉽게 적응하며 형질전환 세포로서 무한한 수명을 지닌다는 점에서 다른 표준세포주보다 백신 생산에 더욱 유리하다. 특히, 일본 뇌염 백신 생산에 있어 베로 세포주의 효율성이 검증되어 있기 때문에, 같은 Flavivirus에 속하는 지카 바이러스의 백신 개발에 있어서 베로 세포의 잠재적 효용성이 높다.

본 발명은 상기의 문제점을 해결하고, 상기의 필요성에 의하여 안출된 것으로서, 본 발명의 목적은 인간용 지카 백신의 제조에 탁월한 생산성 및 우수한 백신 효능을 나타내는 지카 바이러스 변이주를 확보하는 것이다.
본 발명의 다른 목적은 상기 변이주를 이용한 안전하고 효능이 탁월한 불활화 지카 백신을 개발하는 것이다.
상기의 목적을 달성하기 위하여 본 발명은 베로 세포에서 계대 배양하여 베로 세포에 선별 적응시킨 지카 바이러스 변이주를 제공한다.
본 발명의 일 구현예에 있어서, 상기 변이주는 바이러스 역가가 베로 세포에서 1x10
8 PFU/ml 이상인 것이 바람직하며, 상기 변이주는 기탁번호 KCTC 13551BP(이하, 본 발명에서는 'GMZ-002'로 기재한다)인 것이 바람직하며, 상기 변이주는 원형 지카 바이러스 균주인 Aedes.sp/MEX/MEX_2-81/2016(bei resources, NR-50280)에서 37번 아미노산이 Leu으로 변이되고, 196번 아미노산이 Asp로 변이된 것을 특징으로 한다.
또한 본 발명은 상기 본 발명의 지카 바이러스 변이주를 유효성분으로 포함하는 지카 바이러스 백신 조성물을 제공한다.
본 발명의 일 구현예에 있어서, 상기 변이주는 불활화한 것이 바람직하며, 상기 조성물은 약제학적으로 허용되는 첨가제를 추가로 포함하는 것이 바람직하나 이에 한정되지 아니한다.
본 발명의 다른 구현예에 있어서, 상기 조성물은 주사 또는 점막성 경로에 의해 투여되는 것이 바람직하며, 상기 주사 경로는 피하, 피내, 또는 근육내로 이루어지는 것이 바람직하며,
상기 점막성 경로는 경구, 구강, 혀 밑, 비강내 또는 직장으로 이루어지는 것이 바람직하나 이에 한정되지 아니한다.
이하 본 발명을 설명한다.
본 발명은 지카 백신의 제조에 우수한 성질을 나타내는 지카 바이러스 변이주에 관한 것이다. 본 발명의 바이러스는 계대배양하며 높은 증식성이 나타나도록 선별하여 적응시킨 바이러스이며 사람 백신의 제조를 위한 세포 기질로써 세계보건기구(WHO)에 의해 승인된 연속 세포주 베로(Vero)에서 증식할 수 있다. 따라서, 본 발명의 바이러스는 안전하고 높은 생산성을 가진 불활화 백신 제조를 위해 사용될 수 있다. 또한, 본 발명은 안전하고 효능이 우수한 불활화 지카 백신 조성물에 관한 것이다.
본 발명자들은 안전성이 검증된 세포주에서 장기간 적응하여 높은 증식성과 면역원성을 가지는 신규 지카 바이러스를 개발하고자 집중적인 연구를 수행한 결과, 베로 세포에 지카 바이러스를 계대배양하고 선별과 적응을 반복하였을 때 높은 생산성과 효능을 가진 뛰어난 지카 백신의 제조에 적합한 지카 바이러스 변이주를 얻을 수 있음을 발견하였다. 특정하여 예를 들면, BEI Resource에서 분양 받은 원형 지카 바이러스주(NR-50280, Aedes.sp/MEX/MEX_2-81/2016)의 혈청 요구량을 낮추어가며 계대배양을 수행하고 계대별 선별 적응 과정을 거쳐 베로 세포주에서 상업적 생산이 가능한 지카 바이러스주(GMZ-002)를 얻었다.
본 발명은 베로 세포주에서 선별 적응시켜 확립한 지카 바이러스 변이주를 제공한다. 본 발명의 지카 바이러스는 2018년 O7월 17일에 대한민국 전라북도 정읍시 입신길 181 소재한 한국생명공학연구원(KRIBB) 생물자원센터(KCTC)에 기탁하고 기탁번호 KCTC 13551BP를 부여받았다.
또한, 본 발명은 베로 세포주에 선별 적응시킨 지카 바이러스 변이주를 포함하는 지카 백신 조성물을 제공한다.
본원에서는 본 발명을 대표하여 지카 바이러스 GMZ-002 변이주에 관하여 기술할 것이다.
본 발명에서 원형 바이러스로 사용된 지카 바이러스는 BEI Resources에서 분양받은 Aedes.sp/MEX/MEX_2-81/2016 (이하 MEX 2-81) 바이러스주이다. BEI Resources에서 보존되어온 MEX 2-81 바이러스주는 2016년 멕시코 치아파스 주의 모기에서 분리된 원형 바이러스 주를 Cercopithecus aethiops 신장 상피세포(베로 76, clone E6)에서 배양하여 분리한 것이다.
MEX 2-81 바이러스는 Flaviviridae에 속하는 바이러스의 하나이며, 단일가닥 양성-센스 RNA를 가진다. RNA 유전체의 크기는 약 11 Kb이며, 구조 단백질로 캡시드 단백질, 막 단백질, 외피 단백질을 포함하고, 비구조 단백질로는 NS1, 2a, 2b, 3, 4a, 4b, 5 등을 포함하는 것으로 알려져 있다.
본 발명에서, 지카 바이러스를 37℃ 온도에서 베로 세포에서 45회 이상 계대배양하여 적응시키고 베로 세포에서 형성된 플라크(plaque) 수를 기준으로 바이러스의 증식을 모니터하면서 배양된 바이러스를 선별함으로써 본 발명의 지카 바이러스 GMZ-002를 수득하였다. 이러한 선별 적응으로부터 확립된 본 발명의 지카 바이러스 변이주 GMZ-002는 베로 세포 배양물의 상등액 1ml당 1x10
8 pfu 이상의 높은 바이러스 역가를 나타내어 원형주 대비 100배 이상 향상된 증식성을 보이는 동시에 회수를 위한 배양 시간이 크게 감소된 특징을 갖는다. 또한, 보충제로써 혈청이 요구되지 않는 것을 특징으로 하며, 이는 효율적인 비용으로 백신을 대량 생산할 수 있는 상업상 실시 가능한 특징이 된다.
본 발명의 지카 바이러스 변이주 GMZ-002는 베로 세포에서 45계대 이상 계대적응 과정 동안 바이러스 역가 및 플라크의 형태에 변동이 없었다는 점으로 미루어 보아, 본 발명의 바이러스가 베로 세포 계대 중에 안정한 표현형 특성을 유지하고 있으며, 백신 제조에 안정적으로 사용될 수 있음을 시사한다.
본 발명의 바이러스 GMZ-002의 생물학적 특징과 관련된 분자적 기초를 파악하기 위해 본 바이러스의 물리화학적 성질을 분석하였다. cDNA 클로닝 및 서열분석으로 본 발명 바이러스의 게놈의 염기서열을 분석한 결과, 원형의 지카 바이러스와 달리 GMZ-002는 막(M) 단백질 유전자의 염기서열 109번째 티민이 시토신으로 치환되어 있으며, 외피(E) 단백질 유전자의 염기 서열 586번째 구아닌이 아데닌으로 치환되어 있었다. 그에 따라 그 유전 정보에 상응하는 막(M) 단백질 아미노산 37번째 서열이 페닐알라닌에서 류신으로, 외피(E) 단백질 아미노산 글리신에서 아스파르트산으로 변환되어 있음을 알 수 있었다. 해당 유전자 변이로 인한 아미노산 변환은 베로 세포에서 바이러스의 30계대 이상 계속 유지되었다.
본 발명의 지카 바이러스 GMZ-002는 베로 세포에서 증식된다. 세포배양 플라스크 내부 표면에 성장한 베로 세포를 GMZ-002 바이러스로 감염시키고 배양한다. 배양한 바이러스는 다중 회수 방법으로 회수하며, 회수 시점은 감염의 MOI에 따라 감염 후 2일째 또는 3일째 수행하고 새로운 배지를 그 배양물에 재공급한다. 이 배양물을 2일간 배양한 후 배양물 상등액을 다시 회수한다. 회수는 감염 후 7일까지 3회에 걸쳐 반복할 수 있으며 바이러스 역가는 1x10
8 pfu/ml 이상으로 유지되었다.
회수한 배양물 상등액은 정제할 때까지 4℃에서 단기간 보존할 수 있다. 회수한 배양물은 3200xg의 중속으로 15분간 원심분리한 후, 상층액을 다시 회수한다. 회수한 상층액은 농축할 때까지 4℃에서 단기간 보존할 수 있다. 농축 방법으로서, 폴리에틸렌 글리콜(PEG) 8000을 배양액에 10%까지 용해시키고 침전물을 트리스-완충 염수(Tris-HCl, pH7.4)와 같은 적당한 완충액에 용해시킨다. DNA 또는 RNA를 제거하기 위해 프로타민 설페이트를 0.2mg/ml 농도로 첨가한 후 4℃에서 2시간 동안 바이러스 농축액과 혼합한 후, 10000xg의 중속으로 5분간 원심분리하여 상층액을 회수한다. 바이러스를 좀더 정제하기 위하여 밀도 구배 초원심분리를 15-60% 연속 또는 다층 수크로즈 농도구배 상에서 수행한다. 수크로즈 구배를 분별한 후 각 분획 별 바이러스 역가를 검정한다. 바이러스 양성 분획을 검정하는 방법에는 플라크 검정, 폴리아크릴아미드 겔 전기영동 및 웨스턴 블로팅 항원검정이 포함 된다. 고순도의 바이러스 정제물 확보를 위해 높은 바이러스 역가와 낮은 불순물 수준을 기준으로 바이러스 정제물 분획을 확보하였다. 1L의 감염된 배양액으로부터 지카 바이러스 정제 수율은 약 0.7밀리그램인 것으로 나타났다.
본 발명은 또한 지카 불활화 백신의 제조방법에 관한 것이다. 본 발명의 불활화 지카 백신은 강화된 면역원성을 지니고 질병에 대해 보다 큰 보호능을 제공할 것으로 기대된다. 본 발명의 정제된 불활화 지카 백신은 베로 세포에서 배양한 지카 바이러스 변이주를 정제하였으며, 그 과정이 치료용 백신을 위한 개발요건을 충족한다는 점에서 상당한 이점을 지닌다.
또 다른 관점에서, 본 발명은 지카 바이러스를 불활화시켜 항원성은 보존하면서 감염성을 파괴하는 방법을 제공한다. 본 발명의 방법은 유효량의 포름알데하이드를 첨가하여 바이러스가 불활화되는 조건에서 배양하는 것이다. 바이러스의 불활화 과정은 바이러스 배양액 수득 후 정제 과정 전에 이루어지거나 또는 정제 과정이 끝난 후 이루어질 수 있다. 구체적으로 설명하면, 수득한 바이러스 배양 상등액에 포름알데하이드를 첨가한 후 37℃ 또는 4℃에서 배양한다. 바이러스의 항원성을 상실하지 않고 바이러스의 감염성을 완전히 파괴하기 위해서는 37℃에서는 적어도 2일, 4℃에서는 4일이 걸린다. 불활화가 완료되면, 소듐바이설페이트로 포름알데하이드를 중화한 후, 상기 본문에 서술한 방법으로 바이러스 정제를 수행한다. 바이러스의 우선 불활화는 취급자의 안전성을 향상시킬 수 있는 방법으로 권장되나, 불활화 과정의 순서에 한정 되지는 않는다. 또한, 효과적인 불활화제의 예로는 포름알데하이드를 들 수 있지만 이로서 한정되는 것은 아니다. 일반적으로, 불활화는 화학적 수단 또는 물리적 수단에 의해 달성될 수 있다. 화학적 불활화는 예를 들면 효소, β프로피온락톤, 에틸렌이민 또는 이의 유도체, 또는 트윈, 트리톤, 나트륨 데옥시콜레이트 및 설포헤타인과 같은 유기 용매로 바이러스를 처리함으로써 달성될 수 있으며, 필요할 경우 후에 중화될 수 있다. 물리적 불활화는 바람직하게는 바이러스에 UV, X선 또는 감마선과 같이 고에너지의 방사선을 쪼여 달성될 수 있다.
본 발명의 지카 백신은 주사제, 즉 액제 또는 현탁제로서 제조된다. 탄수화물 (솔비톨, 만니톨, 전분, 수크로즈, 덱스트란, 글루코즈 등), 단백질 (알부민, 카세인 등), 단백질 함유제 (소혈청, 탈지유 등) 및 완충액 (알카리 금속 포스페이트)과 같은 안정화제를 첨가할 수 있다. 안정화제를 첨가한 후 제제는 동결건조시키고 진공 또는 질소하에 보존할 수 있다. 필요한 경우, 부형제 작용을 나타내는 하나 이상의 화합물을 첨가할 수 있다. 이러한 목적에 적합한 화합물로서는 예들 들면 수산화알루미늄, 인산알루미늄 또는 산화 알루미늄, 광유 (예, Bayol, Marcol 52) 및 사포닌이 있다. 또한, 필요한 경우, 트윈 및 스판과 같은 하나 이상의 유화제가 바이러스 물질에 첨가될 수 있다.
부형제의 유효성은 부형제에 흡착되는 백신 중의 불활성 바이러스를 실험 동물에 투여하여 발생되는 바이러스에 대한 중화항체의 양을 측정하여 결정할 수 있다. 효과적인 부형제의 예로는 수산화알루미늄이 포함되나 이로써 한정되는 것은 아니다. 백신의 효능은 백신을 마우스에 접종한 후 면역된 마우스로부터 채취한 혈청으로 야생형 독성주 바이러스를 중화시키는 정도를 검사하는 플라크 감소 중화 시험(PRNT) 또는 면역된 마우스에 독성주 바이러스를 주사하여 마우스의 생존률을 검사하는 직접도전법에 의해 결정되었다. 그 결과 본 발명의 지카 백신은 중화항체를 발생하는 효능과 면역된 마우스의 보호능에 있어서 상업화 가능한 우수한 성능을 가지고 있음이 밝혀졌다.
본 발명을 통하여 알 수 있는 바와 같이, 본 발명의 지카 바이러스 변이주는 우수한 생산성과 면역원성을 가져 인간 불활화 지카 백신 제조에 매우 유용하게 사용될 수 있다.
도 1은 베로 세포에서 지카 바이러스를 계대배양하며 높은 증식성을 보이는 균주를 선별하여 적응시키는 과정을 나타낸다. 1계대 지카 바이러스는 베로 세포 단층에 지카 바이러스 MEK 2-81을 0.01 MOI로 접종한 후 3일 경과하여 회수한 것이다. 지카 바이러스의 역가는 베로 세포 단층에 플라크 검정을 실시하여 측정하였다. 실시예 1에 기술된 바와 같이 연속적으로 바이러스를 계대하고 역가를 토대로 선별하여 46계대까지 실시하였다.
도 2는 베로 세포에서 계대배양 및 선별 적응한 지카 바이러스 변이주의 뉴클레오타이드 서열 변이를 분석한 크로마토그램 결과이다. 지카 바이러스 주 MEX 2-81과 지카 바이러스 변이주 GMZ-002의 RNA를 추출하여 지카 바이러스의 prME 서열을 3500xL Dx 유전자 분석기(Applied Biosystems)로 분석하였다. 본 발명에서 지카 바이러스 주 MEX 2-81의 서열은 진 뱅크의 LOCUS:KX446950, 10796 bp ss-RNA linear VRL 18-NOV-2016, DEFINITION:Zika virus strain ZIKV/Aedes.sp/MEX/MEX_2-81/2016, complete genome., ACCESSION: KX446950, VERSION: KX446950.2, SOURCE: Zika virus 서열임.
도 3은 수크로즈 농도구배 초원심분리에 의해 정제된 지카 바이러스를 SDS-PAGE 및 웨스턴 블롯 분석한 결과이다. 2.7ml의 농축된 배양물 상등액을 5ml의 15~60% 수크로즈 농도구배층 위에 적층한 후 200,000g, 4℃에서 3시간 동안 원심분리하였다. 원심분리 후 상층에서부터 0.7ml씩 샘플을 수거하여 각 분획을 SDS-PAGE와 웨스턴 블롯으로 분석하였다. SDS-PAGE로 분리된 단백질은 쿠마시블루로 염색하여 가시화하였고(패널 A), 웨스턴 블롯 분석에서는 항 지카 바이러스 외피단백질(E) 단일클론항체와 반응시켜 가시화하였다(패널 B). 레인 1~9는 초원심분리 후 상층부터 회수한 분획물 1~9번이며, 레인 10은 기염색 단백질 표준물( 상표명: GangnamStain)이다. 영문자 E는 외피 단백질을, 영문자 C는 캡시트 단백질을, 영문자 M은 막 단백질을 가리킨다.
도 4는 불활화 지카 바이러스 변이주를 접종한 마우스에서의 항체생성 역가를 나타낸다. 지카 바이러스주 GMZ-002를 정제한 후 불활화하여 PIV를 생산하였으며, PIV를 0.2ug, 1ug, 5ug 만큼 4주령 Balb/c 마우스의 피하에 접종하였다. 한편, 면역화 유도를 보조하기 위해 PIV와 부형제를 혼합하여 Balb/c 마우스에 동량 접종하였다. ELISA 분석을 위해 접종한 1ug PIV를 coating antigen으로 이용하였고, 혈청을 3-fold로 희석하여 end-point titer 분석하였다. End-point titer 분석은 흡광도 cut-off 0.2를 기준으로 분석하였다.
이하 비한정적인 실시예를 통하여 본 발명을 더욱 상세하게 설명한다. 단 하기 실시예는 본 발명을 예시하기 위한 의도로 기재한 것으로 본 발명의 범위는 하기 실시예에 의하여 제한되는 것으로 해석되지 아니한다.

지카
바이러스의 선별 적응
지카 바이러스를 베로 세포에서 연속계대하기 위해 지카 바이러스 MEX 2-81을 사용하였다. 배양 플라스크에서 배양한 베로 세포에 지카 바이러스 MEX 2-81을 세포당 0.01 MOI로 접종하였다. 감염된 베로 세포는 2% 소태혈청(FBS)를 첨가한 이글 최소 필수 배지 알파(MEMa)로 구성된 영양배지 조건에서 배양되었으며, 약 37℃ 온도와 약 5% CO
2 대기 조성 조건에서 성장시켰다. 세포병변효과를 현미경으로 관찰하고, 플라크 검정법을 통해 배양물 내 바이러스 항원의 역가를 모니터하였다. 배양물이 최고의 바이러스 역가를 나타낸 시점에 바이러스를 회수하고 원심분리로 분리하였다. 연속 계대를 위해 가장 높은 역가를 보인 배양물의 바이러스를 선별하여 베로 세포에 재감염 시켰으며, FBS 농도를 0%까지 단계별로 낮춰가며 연속적인 바이러스 감염, 역가측정 및 플라크 검정을 병행하며 45 계대를 선별 적응 실시하였다. 도1에서 볼 수 있듯이 베로 세포에서 1계대 시 바이러스 역가는 배양물 상등액 1ml 당 약 7x10
5 pfu 였으며, 이후로 선별 계대를 계속할수록 역가가 상승하였고, 14계대 이후에는 1계대 대비 100배 이상 증가하여 배양물 상등액 1ml 당 1x10
8 pfu 이상의 역가를 나타내었다. 이로써 바이러스 생산 수율이 증가하고, 최적 배양시간도 감소하여 바이러스 회수를 위한 최적 시간은 1계대 시 5일에서, 14계대 시 3~4일로 단축되었으며 혈청을 첨가하지 않고 높은 수율로 배양되었다. 이후 지카 바이러스는 베로 세포에서 45계대 동안 향상된 바이러스 생산 수율이 유지되었다. (도면 1)

2:지카
바이러스 변이주 성상 확인 : 외피 유전자의 서열분석
베로 세포에서 선별 적응을 반복하는 동안 지카 바이러스 변이주의 생물학적, 물리화학적 특징을 파악하기 위하여 주요 에피토프를 보유한 외피 유전자를 포함하여, pr 유전자와 M 유전자에 대한 유전자 서열을 분석하여 원형 바이러스인 지카 바이러스 MEX 2-81 주의 유전자 서열과 비교하였다. 45계대를 거치는 동안 지카 바이러스 변이주의 유전자 서열을 분석한 결과, pr 유전자 서열은 완전히 보존되어 있는 한편, 막 단백질 유전자(M)와 외피 단백질 유전자(E)의 뉴클레오타이드 서열은 원형의 MEX 2-81 바이러스주의 서열과 상이하였다. 이의 결과는 하기 표 1과 도면 2에 요약되어 있다.
지카 바이러스 변이주의 아미노산 서열은 37(M)과 196(E) 2개 위치에서 MEX 2-81 바이러스의 공지된 서열과 다르다. 막 단백질 유전자(M)의 109번째 뉴클레오타이드와 외피 단백질 유전자(E)의 586번째 뉴클레오타이드 변화는 지카 바이러스 변이주에서 2개의 아미노산 차이를 유발하였다. 12계대 시점에서 최초 유전자 변이가 일어난 후 지카 바이러스 변이주를 30계대 이상 추가로 계대배양한 경우, 유전자 변이 양상은 안정적으로 유지되었으며, 더 이상 뉴클레오타이드의 변화는 일어나지 않았다. 따라서 지카 바이러스 최종 변이주와 모 바이러스 MEX 2-81 사이에는 2개의 뚜렷한 뉴클레오타이드와 2개의 아미노산 변화가 있다. 또한, 지카 바이러스 최종 변이주에서 확인된 2개의 뉴클레오타이드 변이는 WRAIR에서 개발중인 지카 백신의 원형주 PRVABC59를 포함하여 2017년까지 보고된 총 105 종류의 지카 바이러스 변이종에서 발견되지 않은 고유한 유전적 변이의 형태이다.

배양 및 정제
베로 세포에서 45계대 선별 적응시킨 지카 바이러스 변이주를 종균으로 준비하고 -80℃ 조건에서 동결보관 하였다. 최소 필수배지 알파(MEMa, Gibco)에서 소태혈청(FBS)를 요구하지 않도록 적응시킨 베로 세포를 성장시켰다. 배양한 베로 세포 단층 배양물에 종균 바이러스를 세포당 0.01 MOI로 감염시켰다. 37℃, 5% CO
2 대기조건에서 2시간 동안 바이러스를 흡착시킨 후, 혈청이 없는 MEM을 재공급한 다음 37℃에서 배양하였다. 감염 후 3일째, 감염된 베로 세포 배양물에서 회수한 지카 바이러스의 역가는 1x10
8 pfu/ml 이상이었다. 모은 바이러스 배양액을 3200g에서 15분간 원심분리하고, 상등액을 분리하였다. 바이러스 배양물 상등액은 10% PEG8000으로 침전시켜 농축시켰다. PEG에 의해 침전된 바이러스는 10,000g, 4℃ 조건에서 1시간 동안 원심분리하여 수거하고, PBS 또는 TNE(50mM Tris-HCl, pH 7.4, 1mM EDTA, 150mM NaCl) 완충액으로 재현탁시켰다. 농축한 바이러스는 수크로즈 구배상에서 초원심 분리하여 정제하였다. 15%~60% 농도구배의 수크로즈 층 위에 바이러스 농축액을 올린 후 200,000g, 4℃ 에서 3시간 동안 초원심분리를 수행하였다. 초원심분리 후 각각의 분획을 SDS가 함유된 폴리아크릴아미드 겔상에서 전기영동(SDS-PAGE) 하였다. 뉴클레오캡시드 단백질(C, 13,500Da), 막단백질(M, 8,700Da) 및 외피단백질(E, 55,000Da) 밴드가 SDS-PAGE에서 나타났다(도면 3-A). 또한 초원심분리 후 각각의 분획에 대해 웨스턴 블롯팅을 수행하였으며, 마우스 항-지카 바이러스 외피단백질(E) 단일클론항체를 이용하여 분석한 결과, 외피 항원(E)이 검출되었다(도면 3-B). 바이러스 양성분획을 모으고, BCA 방법에 의해 단백질 농도를 정량하였다. PEG8000 침전과 수크로즈 농도구배를 통해 농축 및 정제된 바이러스의 정제수율을 정제 단계별로 분석하였다(표 2).

불활화
정제된 바이러스는 불활화된 백신 제조를 위해 포름알데하이드로 불활화시켰다. 불활화는 0.05% 포름알데하이드로 37℃ 또는 4℃에서 수행하였으며, 플라크 역가측정 방법을 통해 포름알데하이드 처리기간에 따른 불활화 여부를 검정하였다. 플라크 검정 결과에서 음성으로 판정된 바이러스 정제물을 대상으로 베로 세포 단층에서 계대한 후 재차 플라크 검정을 실시함으로써 낮은 수준의 바이러스의 활성여부까지 검정하였다. 지카 바이러스가 0.05% 포름알데하이드 처리 후 완전히 불활화되기까지 37℃에서는 2일, 4℃에서는 4일이 걸리는 것으로 나타났다 (표 3). 최대 안전성을 확보하기 위하여 지카 바이러스 정제물은 37℃에서 4일 이상 또는 4℃에서 7일 이상 불활화시켰다. 불활화 후 샘플에 잔존한 포름알데하이드는 1%의 소듐바이설파이트를 첨가하여 중화하였으며, 동시에 PBS로 투석하였다.

정제물(PIV)의
마우스에서의 면역원성 평가
PIV의 면역원성을 마우스에서 검정하기 위해, 30마리의 4주령 Balb/c 마우스에 2주 간격으로 부형제 없이 또는 부형제와 함께 3회 피하주사하여 면역하였다. PIV의 면역원성을 향상시키기 위한 부형제는 접종 용량 100ul에 0.1% Alum(수산화알루미늄)과 2ug MPL(mono-phosphoryl lipid A)이 포함되도록 하였다. 3차 접종 2주 후 각 그룹의 마우스로부터 심장채혈 하고, ELISA 방법으로 혈청 내 항체역가를 측정하였다. 96웰 ELISA 플레이트에 PIV를 웰당 0.1ug씩 4℃에서 16시간 이상 흡착시킨 후 1X TBS-T로 세척하였다. 이후 0.5% 카제인으로 1시간 동안 블로킹한 후 TBS-T로 세척하였다. 1:100으로 희석한 마우스 혈청을 웰에 첨가하고 37℃에서 1시간 반응시킨 뒤 PBS-T로 3회 세척 후, 1:5000으로 희석한 Goat anti-mouse IgG HRP를 100ul 분주하여 37℃에서 1시간 반응하였다. 최종 발색에 앞서 chromogen 용액 25ul과 TMB(Substrate) 50ul를 혼합하여 각 웰에 반응시켰다. 음성 비교군에서 발색이 나타나는 시점에서 2M 황산용액 25ul를 첨가하여 반응을 중지시키고, 450nm 파장에서 흡광도를 측정하였다. ELISA 측정 결과, 모든 농도의 PIV 접종군에서 부형제 첨가 여부와 무관하게 높은 항체 생성을 확인하였다.
각 군별 최종 혈청의 항체 역가를 비교하고자 마우스 혈청을 1/100 희석 후 3배씩 단계별로 희석하였다. 450nm 파장에서 흡광도 0.2 이상의 값이 연속 발색된 구간을 수치화하여 End-point titer 평가를 수행하였다. 도면 6과 같이 End-point titer는 PIV 접종 농도와 비례하여 증가하였으며, 면역증강 부형제 미첨가군에 비해 첨가군에서 더욱 높은 반응역가가 나타났다.

상기 획득한 마우스 혈청 내 중화항체가를 측정하기 위해 플라크 감소 중화항체 검사(PRNT)를 실시하였다. 10% FBS를 함유한 α-MEM에서 베로 세포를 배양하였으며, 56℃에서 30분간 불활화한 마우스 혈청과 지카 바이러스를 2% FBS를 함유한 α-MEM에서 동량 혼합하였다. 혼합물은 37℃에서 30분간 반응시킨 후 베로 세포에 분주하였으며, 37℃에서 2시간 동안 세포에 흡착되도록 배양하였다. 이후 접종물을 제거한 후 1.4% 메틸셀룰로오즈(methylcellulose)와 5% FBS를 함유한 배지를 베로 세포 위에 분주한 후 37℃에서 4일 동안 흔들림에 주의하여 배양하였다. 배양 종료 후 배양액을 제거한 뒤 포르말린(formalin)과 크리스탈 바이올렛(crystal violet)을 사용하여 고정 및 염색하여 형성된 플라크 수를 분석하였다. 최종적으로 플라크 수가 50% 감소하는 혈청 희석배수를 PRNT
50으로 결정하였다. 표 4 와 같이, PIV 용량이 증가함에 따라 중화항체 역가가 높게 나타났으며, Alum과 MPL을 함유한 PIV를 접종한 마우스 그룹에서 보다 높은 중화항체 역가가 나타났다.

마우스의 보호 효능 평가
PIV의 생체 내 항 지카 바이러스 보호 효능을 검증하기 위해 마우스에서 직접도전법을 수행하였다. 일반적인 성체 마우스는 지카 바이러스에 대한 감수성이 없기 때문에, IFNAR-1 항체를 마우스에 선행 접종하여 IFNAR-1을 블락킹해 지카 바이러스에 대한 감수성을 부여하는 방법을 사용하였다. (Richner, J. M. et al., Cell, 168(6):1114-1125, 2017) 생후 8주된 C57BL/6 마우스에 PIV 200ng 또는 1ug을 Alum+MPL 부형제 없이 또는 함께 2주 간격으로 총 3회 피하주사하여 면역화 접종하였다. 동일한 주령의 대조군 그룹에 PBS를 Alum+MPL 부형제와 함께 피하주사하여 음성대조군으로 설정하였다. 마지막 접종 2주일 후 마우스에 InVivoMAb anti-mouse IFNAR-1(Bxcell)을 2mg 복강주사하고, 24시간 후 야생형 지카 바이러스주(MEX 2-81)를 10
6 pfu 피하주사하였다. 공격접종 시점 이후 3주간 마우스의 생존율을 24시간 간격으로 모니터하였다. 표 5에서 볼 수 있듯이, PIV 200ng 또는 1ug 단독으로 면역화된 마우스는 각각 80% 또는 100% 보호된 것으로 나타났다. Alum+MPL과 혼합된 PIV 200ng 또는 1ug으로 면역화된 마우스는 모두 100% 보호되었다. 한편 음성대조군의 마우스는 도전 후 15일 시점에서 모두 사망하였다. 본 마우스 직접도전법의 결과는 GMZ-002 바이러스주를 불화화시킨 백신(PIV)이 우수한 보호 효능을 나타내었음을 시사한다.

Claims (10)
- 베로 세포에서 계대 배양하여 베로 세포에 선별 적응시킨 지카 바이러스 변이주.
- 제1항에 있어서, 상기 변이주는 바이러스 역가가 베로 세포에서 1x10 8 PFU/ml 이상인 지카 바이러스 변이주.
- 제1항 또는 제2항에 있어서, 상기 변이주는 기탁번호 KCTC 13551BP인 것을 특징으로 하는 지카 바이러스 변이주.
- 제3항에 있어서, 상기 변이주는 원형 지카 바이러스 균주인 Aedes.sp/MEX/MEX_2-81/2016(bei resources, NR-50280)에서 37번 아미노산이 Leu으로 변이되고, 196번 아미노산이 Asp로 변이된 것을 특징으로 하는 지카 바이러스 변이주.
- 제1항의 지카 바이러스 변이주를 유효성분으로 포함하는 지카 바이러스 백신 조성물.
- 제5항에 있어서, 상기 변이주는 불활화한 것을 특징으로 하는 지카 바이러스 백신 조성물.
- 제5항에 있어서, 상기 조성물은 약제학적으로 허용되는 첨가제를 추가로 포함하는 것을 특징으로 하는 지카 바이러스 백신 조성물.
- 제5항에 있어서, 상기 조성물은 주사 또는 점막성 경로에 의해 투여되는 것을 특징으로 하는 지카 바이러스 백신 조성물.
- 제8항에 있어서, 상기 주사 경로는 피하, 피내, 또는 근육내로 이루어지는 특징으로 하는 지카 바이러스 백신 조성물.
