Abstract
本発明は、ドレブリンの細胞内イメージングに応用可能な、ドレブリンと特異的に結合するペプチド及びマーカータンパク質の提供、さらにそのマーカータンパク質を用いたドレブリンの検出方法を提供することを課題とする。ドレブリンに特異的に結合するアミノ酸配列を有するペプチド及びそのペプチドを含む各種マーカータンパク質、さらに、そのマーカータンパク質を利用したドレブリンの検出方法を提供することにより、本発明に係るペプチドをドレブリンの細胞内イメージングに応用可能にする。
Description
本発明は、ドレブリンに特異的に結合するペプチド、及びそのペプチドを用いたドレブリンの検出方法に関する。本願は、2018年5月8日に出願された日本国特許出願第2018-089929号に対し優先権を主張し、その内容をここに援用する。
ドレブリンは、脳に多く存在するシナプスタンパク質であり、神経細胞の発達にとって重要な役割を担っているタンパク質である。ドレブリンには成熟型アイソフォームのAタイプ(ドレブリンA)と幼弱型アイソフォームのEタイプ(ドレブリンE)が存在する。神経細胞の発達に伴い、ドレブリンEの発現が減少し、ドレブリンAが発現する。このドレブリンAの発現が正常に行われることにより、神経細胞において樹状突起スパインの形成が促進され、神経回路が正常に形成される。
発明者らは、アルツハイマー病等の痴呆性疾患において、樹状突起スパインのドレブリンが広範囲に消失していることを報告している(非特許文献1)。また、ドレブリンの定量的な解析から、ドレブリンの集積量と認知能力との相関が認められている(非特許文献2)。つまり、ドレブリンは神経科学分野において、非常に有用な生体内物質であることがわかっている。
さらに、近年では、ドレブリンは神経科学分野のみならず、癌や感染症分野にまで広く関与していることがわかっており(非特許文献3、4)、それらの分野の疾患においても、疾患の初期発生過程から疾患の進行状態を把握するためのバイオマーカーとしての役割への期待が高まっている。
これまでに、抗ドレブリン抗体を用いて、ドレブリンを特異的に検出し定量することについては既に行われており、そのようなドレブリンを特異的に検出し定量する方法としては、組織切片染色法やウェスタンブロット法、ELISA(Enzyme-Linked ImmunoSorbent Assay)法等が用いられている(特許文献1)。

しかしながら、これまでドレブリンに特異的に結合できるペプチドのアミノ酸配列はわかっておらず、既存の市販されている抗ドレブリン抗体はIgG抗体であり、約160kDaの分子量を有する。そのため、分子量の大きさに起因して、抗体の細胞内移行性が悪く、ドレブリンの細胞内イメージングが困難であるという問題点があり、既存の抗ドレブリン抗体のみを用いた場合において、ドレブリンの動態を探るのには限界があった。
また、既存の抗ドレブリン抗体は、組織切片染色法やウェスタンブロット法、ELISA法といった検出方法に利用できるが、これらの検出方法では生体から採取した試料中のドレブリンを検出及び定量することに留まっており、細胞内におけるドレブリンの動態を探ることはできなかった。
ここで、細胞内で発現させることができるドレブリン結合ペプチドを用いることができれば、そのペプチドを用いた細胞内イメージングによりドレブリンの細胞内動態を探ることが可能となる。その結果、シナプスの作動メカニズムを解明することができ、神経細胞の成熟におけるドレブリンの役割を詳細に知ることができる。そしてそれは、神経科学分野における神経難病の病態解明のみならず、癌及び感染症の分野においてもそれらの病態解明に寄与することが可能である。
かかる状況に鑑みて、本発明は、ドレブリンと特異的に結合できるアミノ酸配列を有するペプチドであって、ドレブリンの細胞内イメージングにも応用可能であるペプチドを提供すること、及び当該ペプチドをコードするポリヌクレオチドを含むベクターを提供すること、さらにそのペプチドを用いたドレブリンの検出方法を提供することを課題とする。
本発明者らは、上記課題を解決するためにVHH抗体作成法を用いて、鋭意研究を行った結果、細胞内イメージングに応用可能である、ドレブリンに特異的に結合するアミノ酸配列を有するペプチドを同定することに成功した。さらに、そのペプチドを利用することで、ドレブリンを効率よく検出できることを見いだし、発明を完成させた。
すなわち、本発明は以下の事項により特定されるとおりのものである。
〔1〕以下の(1)若しくは(2)に記載のアミノ酸配列を含むことを特徴とするドレブリン結合ペプチド又はその機能性断片。
(1)配列番号16~24のいずれかで示されるアミノ酸配列;
(2)配列番号16~24のいずれかで示されるアミノ酸配列において、1若しくは数個のアミノ酸が挿入、欠失、置換、及び/又は付加されたアミノ酸配列;
〔2〕マーカータンパク質及び/若しくはペプチドタグを融合したことを特徴とする上記〔1〕に記載のペプチド又はその機能性断片。
〔3〕マーカータンパク質が蛍光タンパク質であり、ペプチドタグがFlagであることを特徴とする上記〔2〕に記載のペプチド又はその機能性断片。
〔4〕検出可能な標識を付加したことを特徴とする上記〔1〕~〔3〕のいずれかに記載のペプチド又はその機能性断片。
〔5〕標識が、酵素、蛍光物質、ジゴキシゲニン、若しくはビオチンであることを特徴とする上記〔4〕に記載のペプチド又はその機能性断片。
〔6〕細胞内移行ペプチドを付加したことを特徴とする上記〔1〕~〔5〕のいずれかに記載のペプチド又はその機能性断片。

本発明により、ドレブリンに特異的に結合できるペプチド、当該ペプチドと蛍光タンパク質等のマーカータンパク質やペプチドタグを含む融合ペプチド、及びそれらのペプチドや融合タンパク質を利用したドレブリンの検出方法が提供される。
本発明に係るペプチドはシングルドメイン構造であるため、マーカータンパク質、ペプチドタグ、及び抗体のFc領域等の他の分子を結合させることが容易に行える。
本発明に係るペプチドは低分子量であり、細胞内で効率よく発現させることができるので、ドレブリンの細胞内イメージングにも応用可能である。
本発明に係るペプチドはシングルドメイン構造であるため、マーカータンパク質、ペプチドタグ、及び抗体のFc領域等の他の分子を結合させることが容易に行える。
本発明に係るペプチドは低分子量であり、細胞内で効率よく発現させることができるので、ドレブリンの細胞内イメージングにも応用可能である。
本発明の第一の態様は、ドレブリンに特異的に結合できるペプチド(以下、「ドレブリン結合ペプチド」ともいう)であって、以下の(1)若しくは(2)から選択されるペプチド又はその機能性断片である。
(1)配列番号16~24のいずれかで示されるアミノ酸配列を含むペプチド;
(2)配列番号16~24のいずれかで示されるアミノ酸配列において、1若しくは数個のアミノ酸が挿入、欠失、置換、及び/又は付加されたアミノ酸配列を含むペプチド;
ここで、機能性断片とは、ドレブリン結合能を失わない範囲で、ペプチドのN末端側及び/又はC末端側のアミノ酸残基を除去して得られたペプチドをいう。
(1)配列番号16~24のいずれかで示されるアミノ酸配列を含むペプチド;
(2)配列番号16~24のいずれかで示されるアミノ酸配列において、1若しくは数個のアミノ酸が挿入、欠失、置換、及び/又は付加されたアミノ酸配列を含むペプチド;
ここで、機能性断片とは、ドレブリン結合能を失わない範囲で、ペプチドのN末端側及び/又はC末端側のアミノ酸残基を除去して得られたペプチドをいう。
ここで、1若しくは数個とは、1~10個、好ましくは1~5個、より好ましくは1~3個である。1若しくは数個のアミノ酸が挿入、欠失、置換、及び/又は付加されたアミノ酸配列を含むペプチドとは、そのペプチドがドレブリンに特異的に結合できる機能を有する限り、特に限定されない。
ここで、これらの挿入、欠失、置換、及び/又は付加等のアミノ酸変異は天然に生じる変異でもよいし、人工的に導入される変異でもよい。
例えば、アミノ酸変異は部位特異的変異導入法や、Error prone PCR法等のランダム変異導入法により導入することができる。また、変異導入された配列を有するペプチドがドレブリン結合能を有していることは、ELISA、ウェスタンブロットやアフィニティクロマトグラフィー等の方法で確認できる。変異が導入される部位は抗体におけるフレームワーク領域(FR)に相当する領域でもよいし、相補性決定領域(以下、CDRという)に相当する領域でもよい。CDRに相当する領域における変異が結合性を増加させる場合もある。

また、前記アミノ酸配列は、ペプチドがドレブリンと特異的に結合できる限り、前記配列番号16~24のいずれかで示されるアミノ酸配列と80%、好ましくは90%以上、より好ましくは95%以上、さらに好ましくは98%以上の同一性を有するものであってよい。
本発明の第二の態様は、前記第一の態様のペプチドに、マーカータンパク質及び/又はペプチドタグを融合させたペプチドである。これらは前記第一の態様のペプチドのN末端側に融合させてもよいし、C末端側に融合させてもよい。
前記マーカータンパク質としては、特に限定されないが、例えば、アルカリフォスファターゼ(以下、APという)、西洋わさび過酸化酵素(以下、HRPという)等の酵素、抗体のFc領域、緑色蛍光タンパク質(GFP)等の蛍光タンパク質を挙げることができる。また、前記ペプチドタグとしては、特に限定されないが、例えば、HA(ヘマグルチニン)、Flag、Myc等のエピトープタグ、グルタチオンS-トランスフェラーゼ(GST)、ヒスチジン(His)、アビジン、V5(GKPIPNPLLGLDST:配列番号5)のペプチドタグが挙げられる。
上記マーカータンパク質及び/又はペプチドタグの融合は、制限酵素やPCR(Polymerase Chain Reaction)等を利用した通常の遺伝子組み換え技術により行うことができる。なお、上記マーカータンパク質及び/又はペプチドタグは前記第一の態様のペプチドに直接融合させてもよいが、融合タンパク質においてドレブリン結合能が維持される限り、他のアミノ酸配列(例えば、1~10アミノ酸又は1~5アミノ酸)を介して融合させてもよい。
本発明の第三の態様に係るペプチドは、前記マーカータンパク質が蛍光タンパク質であり、前記ペプチドタグがFlagである、前記ペプチドである。
本発明の第四の態様に係るペプチドは、前記第一の態様乃至第三の態様のペプチドに検出可能な標識を付加したペプチドである。
上記標識としては、特に限定されないが、例えば、酵素(ペルオキシダーゼ、アルカリフォスファターゼ、ルシフェラーゼ又はβガラクトシダーゼ等)、蛍光物質(フルオレセイン、ルシフェラーゼ等)、ジゴキシゲニン、ビオチンが挙げられる。
すなわち、本発明の第五の態様に係るペプチドは、前記標識が、酵素、蛍光物質、ジゴキシゲニン又はビオチンである、前記ペプチドである。
本発明の第六の態様は、前記第一の態様乃至第五の態様のペプチドにIgG-Fc領域を融合させた抗体である。具体的には、ヒンジドメイン、CH2ドメイン及びCH3ドメインをこの順で含む哺乳類(好ましくはヒト)IgG-Fc領域のヒンジドメインに、前記第一の態様又は第二の態様のペプチドをそのC末端を介して融合させた抗体のことである。
また、前記抗体は二量体化させることも可能である。

本発明の第七の態様に係るポリヌクレオチドとしては、配列番号16~24の何れかのアミノ酸配列、又は、配列番号16~24のいずれかで示されるアミノ酸配列において、1若しくは数個のアミノ酸が挿入、欠失、置換、及び/又は付加されたアミノ酸配列、をコードする塩基配列を含むポリヌクレオチドが好ましく、例えば、配列番号8~11のいずれかで示される塩基配列を含むポリヌクレオチド、又は、配列番号8~11のいずれかで示される塩基配列の相補配列を有するポリヌクレオチドとストリンジェントな条件下でハイブリダイズするポリヌクレオチドが挙げられる。
ここで、ストリンジェントな条件としては、例えば、サザンブロット法におけるハイブリダイゼーション後の洗浄に使用される条件が挙げられ、具体的には、0.1×SSCの塩濃度及び摂氏60度の温度で洗浄を行う条件が挙げられる。

本発明の第八の態様は、前記第七の態様に係るポリヌクレオチドを含むベクターである。ベクターとしては、プラスミドベクター、ファージベクター、ウイルスベクター等が例示され、前記ポリヌクレオチドを導入する宿主の種類によって適宜選択される。ベクターはヒト細胞等の哺乳動物で発現させるためのベクターであってもよいし、大腸菌等の原核細胞や酵母等の真核細胞で発現させるためのベクターであってもよい。

本発明の第九の態様は、前記第七の態様に係るポリヌクレオチド又は前記第八の態様に係るベクターが導入された細胞である。細胞は、真核細胞でもよいし、原核細胞でもよいが、内在性のドレブリンを発現する細胞が好ましく、内在性のドレブリンを発現する、神経細胞等の哺乳動物細胞が好ましい。
ここで、前記第七の態様に係るポリヌクレオチド又は前記第八の態様に係るベクターの細胞内への導入は、公知のマイクロインジェクション法、又はリポフェクション法やエレクトロポレーション法等の形質転換法によって行うことができる。
本発明の第十の態様は、前記第一乃至第五の態様に係るペプチド、又は第六の態様に係る抗体を用いた、被検試料中のドレブリンの検出方法である。この検出方法は、前記ペプチド又は抗体を前記被検試料に添加又は導入する工程を含む。
検出対象のドレブリンは哺乳動物由来のドレブリンが挙げられ、ヒト由来のドレブリンであることが好ましい。
被検試料はドレブリンを含む限り特に限定されないが、哺乳動物由来の被検対象が挙げられ、ヒト由来であることが好ましい。例えば、脳組織の生検試料、神経細胞、脳脊髄液、血液、血漿、血清、組織液、リンパ液、体腔液、尿、又はそれらのタンパク質抽出液が挙げられる。
前記ペプチド又は抗体を前記被検試料に添加する態様について、例を挙げて説明する。
例えば、免疫組織染色法により検出を行う態様として、前記ペプチド又は抗体を含む溶液を適当な倍率に希釈し、被検試料である組織切片や固定化培養細胞に添加する態様が挙げられる。
また、ウェスタンブロット法により検出を行う態様として、前記被検試料であるタンパク質抽出液を用いてゲル電気泳動等でタンパク質を分離し、分離されたタンパク質に対して前記ペプチド又は抗体を含む溶液を適当な倍率に希釈して添加する態様が挙げられる。
例えば、免疫組織染色法により検出を行う態様として、前記ペプチド又は抗体を含む溶液を適当な倍率に希釈し、被検試料である組織切片や固定化培養細胞に添加する態様が挙げられる。
また、ウェスタンブロット法により検出を行う態様として、前記被検試料であるタンパク質抽出液を用いてゲル電気泳動等でタンパク質を分離し、分離されたタンパク質に対して前記ペプチド又は抗体を含む溶液を適当な倍率に希釈して添加する態様が挙げられる。
前記ペプチド又は抗体を前記被検試料である細胞に導入する態様について、例を挙げて説明する。
前記ペプチド又は抗体を細胞内に導入するには、マイクロインジェクション等で直接細胞内に注入してもよいが、前記ペプチド又は抗体をコードするポリヌクレオチドを、ベクター等を用いて細胞内に導入し、前記ペプチド又は抗体を細胞内で発現させることによって導入してもよい。発現は一過的な発現でも恒常的な発現でもよい。
前記ペプチド又は抗体を細胞内に導入するには、マイクロインジェクション等で直接細胞内に注入してもよいが、前記ペプチド又は抗体をコードするポリヌクレオチドを、ベクター等を用いて細胞内に導入し、前記ペプチド又は抗体を細胞内で発現させることによって導入してもよい。発現は一過的な発現でも恒常的な発現でもよい。
また、一態様において、前記ペプチド又は抗体は、低分子量の一本鎖ペプチドであるため、細胞内移行ペプチドを用いて細胞内に導入してもよい。細胞内移行ペプチドとしては、HIV-1 Tat蛋白質のアミノ酸配列48-60位に対応するペプチド配列(Tatペプチド)やオリゴアルギニン、ショウジョウバエのアンテナペディア蛋白質由来ペプチド(penetratin)、神経ペプチドgalaninとハチ毒mastoparanのキメラペプチドであるtransportan、分泌シグナルペプチド由来ペプチドMTS(Mitochondrial targeting signal)等が挙げられる。細胞内移行ペプチドは、ドレブリン結合ペプチドのN末端又はC末端にペプチド結合により結合してもよく、非共有結合により複合体を形成させてもよい。
本発明の第十一の態様は、本発明の第十の態様に係る被検試料中のドレブリンの検出方法において、さらに被検試料中のドレブリンに結合したペプチド又は抗体を発色又は発光により検出する工程を含む、方法である。
この態様において、ドレブリンの検出は、例えば、以下のようにして行うことができる。
前記ペプチドが蛍光タンパク質と融合されている場合は、前記ペプチドが被検試料中のドレブリンと結合することで、被検試料中のドレブリンの量に依存した蛍光が発せられるので、当該蛍光強度に基づいて被検試料中のドレブリンを検出し、定量することができる。特に、前記ペプチドを細胞内に導入する態様においては、蛍光タンパク質と融合されている前記ペプチドを用いることで細胞内のドレブリンの挙動を検出することができる。
前記ペプチドが蛍光タンパク質と融合されている場合は、前記ペプチドが被検試料中のドレブリンと結合することで、被検試料中のドレブリンの量に依存した蛍光が発せられるので、当該蛍光強度に基づいて被検試料中のドレブリンを検出し、定量することができる。特に、前記ペプチドを細胞内に導入する態様においては、蛍光タンパク質と融合されている前記ペプチドを用いることで細胞内のドレブリンの挙動を検出することができる。
また、前記ペプチドがFlag等のペプチドタグと融合されている場合は、前記ペプチドを被検試料中のドレブリンと結合させた後、当該ペプチドタグに対する抗体であって、発色物質や発光物質又はそれらの物質を生じさせる酵素等で標識された抗体を反応させることで、被検試料中のドレブリンを検出し、定量することができる。
また、前記ペプチドがIgG-Fc領域を融合させた抗体である場合は、前記抗体を被検試料中のドレブリンと結合させた後、当該Fc領域に対する抗体であって、発色物質や発光物質又はそれらの物質を生じさせる酵素等で標識された抗体を反応させることで、被検試料中のドレブリンを検出し、定量することができる。
なお、前記ペプチド自体が発色物質や発光物質又はそれらの物質を生じさせる酵素等で標識されていてもよい。
定量の際は、例えば、予め既知の濃度の試料で検量線(標準曲線)を作成しておき、測定値を検量線に照合して試料中のドレブリン濃度を算出することができる。

本発明の第十二の態様は、本発明の第十一の態様に係る被検試料中のドレブリンの検出方法において、前記検出方法が蛍光に基づく画像解析法である、方法である。
この態様においては、前記ペプチド又は抗体が蛍光タンパク質との融合タンパク質として細胞内に導入されるので、前記ペプチド又は抗体が細胞内の内在性ドレブリンと結合することで、蛍光タンパク質が発する蛍光を画像解析することにより内在性ドレブリンの挙動をリアルタイムでモニターすることができる。内在性ドレブリンの挙動としては、ドレブリンの細胞内局在性、細胞内での量の変動等が挙げられる。内在性ドレブリンの挙動をリアルタイムでモニターすることにより、シナプスの作動メカニズムを解明することができ、神経細胞の成熟におけるドレブリンの役割を詳細に知ることができる。
この態様においては、前記ペプチド又は抗体が蛍光タンパク質との融合タンパク質として細胞内に導入されるので、前記ペプチド又は抗体が細胞内の内在性ドレブリンと結合することで、蛍光タンパク質が発する蛍光を画像解析することにより内在性ドレブリンの挙動をリアルタイムでモニターすることができる。内在性ドレブリンの挙動としては、ドレブリンの細胞内局在性、細胞内での量の変動等が挙げられる。内在性ドレブリンの挙動をリアルタイムでモニターすることにより、シナプスの作動メカニズムを解明することができ、神経細胞の成熟におけるドレブリンの役割を詳細に知ることができる。
蛍光に基づく画像解析法は特に制限されないが、蛍光顕微鏡等を用いた画像解析が挙げられる。健常人由来の神経細胞と神経疾患患者由来の神経細胞において、内在性ドレブリンの挙動を蛍光に基づく画像解析により比較することで、神経疾患におけるドレブリンの関与を調べることができる。また、細胞に薬剤を加えて、内在性ドレブリンの挙動を蛍光に基いて画像解析することで、神経疾患に対する薬剤の評価等を行うこともできる。
以下実施例により、本発明をより詳細に説明するが、本発明の範囲が実施例のみに限定されないことは言うまでもない。
1.ドレブリン結合ペプチドの単離
(A)VHH抗体提示ファージライブラリーの調製
実施例2及び3に用いたドレブリン結合ペプチドを以下の手順で作成した。
市販されているプラスミドpUC119(例えば、タカラバイオ株式会社より入手可能)由来のVector1に、制限酵素SfiI切断サイトを挿入し、制限酵素SfiIによりVector1を処理し、ベクターの断片を得た。制限酵素SfiI切断サイトは、GGCCCAGCCGGCC(配列番号6)又はGGCCTCTGCGGCC(配列番号7)により表されるDNA配列からなる。
(A)VHH抗体提示ファージライブラリーの調製
実施例2及び3に用いたドレブリン結合ペプチドを以下の手順で作成した。
市販されているプラスミドpUC119(例えば、タカラバイオ株式会社より入手可能)由来のVector1に、制限酵素SfiI切断サイトを挿入し、制限酵素SfiIによりVector1を処理し、ベクターの断片を得た。制限酵素SfiI切断サイトは、GGCCCAGCCGGCC(配列番号6)又はGGCCTCTGCGGCC(配列番号7)により表されるDNA配列からなる。
同様に、PCR法によって獲得したラクダのVHH抗体遺伝子ライブラリも制限酵素SfiIにより処理し、VHH抗体の遺伝子断片を得た。
制限酵素処理したVector1及びVHH抗体の遺伝子断片を、1:2の割合で混合し、ライゲーション試薬(東洋紡株式会社より入手、商品名:Ligation High ver.2、以下同じ)を注入した。この混合液を摂氏16度の温度で2時間静置することで、VHH抗体の遺伝子断片をVector1にライゲーションした。
このVHH抗体の遺伝子断片をライゲーションしたVector1を用いて、大腸菌(タカラバイオ株式会社より入手、商品名:HST02)にトランスフェクションした。
次いで、このトランスフェクションした大腸菌を、100μg/mLの濃度を有するアンピシリン含有2YT培地で15時間培養した。このようにして、VHH抗体の遺伝子ライブラリに含まれる遺伝子断片由来のペプチドを提示するファージのライブラリを得た。
(B)ドレブリン抗原の固定化
ドレブリン(群馬大学神経薬理学分野から入手)をリン酸緩衝生理食塩水(以下、PBSという)と混合し、ドレブリン溶液を調製した。ドレブリンの濃度は1μg/mLであった。3mLのドレブリン溶液を、イムノチューブ(Nunc社より購入)に注入し、ドレブリン溶液をイムノチューブ内で摂氏4度の温度で一晩静置することで、イムノチューブ内部にドレブリンを固定した。
ドレブリン(群馬大学神経薬理学分野から入手)をリン酸緩衝生理食塩水(以下、PBSという)と混合し、ドレブリン溶液を調製した。ドレブリンの濃度は1μg/mLであった。3mLのドレブリン溶液を、イムノチューブ(Nunc社より購入)に注入し、ドレブリン溶液をイムノチューブ内で摂氏4度の温度で一晩静置することで、イムノチューブ内部にドレブリンを固定した。
次いで、イムノチューブ内部を、PBSを用いて3回洗浄した。
3%スキムミルク(和光純薬株式会社より入手)含有PBSをイムノチューブに満たすことで、イムノチューブ内部のブロッキング処理を行った。
イムノチューブを、室温にて1時間静置し、イムノチューブの内部を、PBSを用いて3回洗浄した。
(C)パニング
VHH抗体を提示するファージのライブラリ(濃度:およそ10E+11/mL)を、3mLの3%スキムミルク含有PBSと混合し、混合液を調製した。この混合液を、ドレブリン抗原を固定化したイムノチューブに注入した。

イムノチューブにパラフィルムからなる蓋を取り付け、ローテーターを用いて10分間転倒しながら回転した。
イムノチューブを、室温にて30分間静置した。
イムノチューブ内部を、0.05%Tween20含有PBS(以下、「PBST」という)で10回洗浄した。
ドレブリン抗原に結合したVHH抗体を提示するファージを抽出するために、1mLの100mMトリメチルアミン溶液をイムノチューブに注入した。
イムノチューブにパラフィルムからなる蓋を取り付け、ローテーターを用いて、10分間転倒しながら回転した。
溶液を中和するため、溶液を1mLの0.5M Tris-HCl(pH:6.8)を注入したチューブに移した。再度、1mLの100mMトリメチルアミン溶液を用いたファージの抽出を繰り返し、3mLの抽出液を得た。
3mLの抽出液を、12mLの大腸菌TG-1と混合した。混合液を、摂氏30度の温度で1時間静置した。
コロニーを計数するため、大腸菌TG-1を含有する15mLの混合液15μLを、2YTAG培地(Bacto Tryptone 1.6%、Bacto Yeast Extract 1.0%、NaCl 0.5%、Glucose 2.0%、Ampicillin 0.01%)を含むアガープレート(10mL/プレート)に播種した。摂氏30度の温度で一晩静置し、コロニー数を計数することで抽出液に含まれていたファージのタイターを測定した。
残りの混合液を遠心分離した。上澄み液を捨て、沈殿物を小プレート3枚(5mL/プレート)の2YTAG培地に播種した。これらの3枚のプレートを、摂氏30度の温度で一晩静置した。このようにして、1回目のパニングを行った。
1回目のパニングの手順と全く同様に、2回目、3回目とパニングを繰り返し行った。このようにして、VHH抗体を提示するモノクローナルファージを精製した。
3回目のパニングの後、大腸菌のコロニーを爪楊枝でピックアップした。ピックアップした1つのコロニーを、200μLの2YTAG培地を注入した96穴平底プレートの1つのウェルに置き、この操作を繰り返した。
ウェル内の溶液を、摂氏30度の温度で160rpmの回転数で撹拌した。
50μLの増殖した大腸菌を含む溶液を回収し、この溶液をプレート上で50μLの2YTAG培地と混合した。2YTAG培地には、感染多重度(MOI)が20になるようなヘルパーファージを含有させた。そして、溶液を、摂氏30度の温度で45分間静置した。
2YTAG培地を含むプレートを、4000rpmにて5分間遠心分離し、上清を捨てた。大腸菌を含む沈殿物を、200μLの2YTAG培地と混合した。混合液を、摂氏30度の温度で一晩160rpmの回転数で撹拌した。
その後、混合液は、4000rpmにて5分間遠心分離し、大腸菌を含む上清を回収した。
(D)ELISAによるファージ提示VHH抗体及び抗原の定性評価
96ウェルプレート(Thermo Fisher Scientific社より購入、商品名:Maxisorp(登録商標))の各ウェルに、1μg/mLの濃度を有するドレブリン溶液を抗原として50μLずつ添加した。96ウェルプレートを、摂氏4度の温度で一晩静置し、ドレブリン抗原を各ウェルに固定した。
96ウェルプレート(Thermo Fisher Scientific社より購入、商品名:Maxisorp(登録商標))の各ウェルに、1μg/mLの濃度を有するドレブリン溶液を抗原として50μLずつ添加した。96ウェルプレートを、摂氏4度の温度で一晩静置し、ドレブリン抗原を各ウェルに固定した。
各ウェルを、PBSで3回洗浄した。次いで、3%スキムミルク(和光純薬株式会社より入手)含有PBSを各ウェルに注入し(200μL/ウェル)、96ウェルプレートを、室温で1時間放置することで、各ウェルのブロッキング処理を行った。その後、各ウェルを、PBSで3回洗浄した。
VHH抗体を提示するモノクローナルファージ(M13バクテリオファージ)を、各ウェルに注入し(50μL/ウェル)、96ウェルプレートを1時間静置した。このようにして、ファージをドレブリン抗原と反応させた後、各ウェルを、 PBSTで5回洗浄し、抗M13抗体(GEヘルスケア社より入手、型番:27942101、10000倍希釈)を各ウェルに添加した(50μL/ウェル)。次いで、各ウェルをPBSTで6回洗浄した。
各ウェルに発色剤(和光純薬工業社より入手、型番:155-02161)を注入し(50μL/ウェル)、96ウェルプレートを10分間静置することで、発色剤を抗体と反応させた。反応後、硫酸水溶液(1規定)を50μL/ウェルの濃度で各ウェルに添加し、反応を停止させ、490nmの波長での溶液の吸光度を測定した。
良好な吸光度測定の結果を有する4つのウェルを選択し、それらのウェル上のファージに含まれるDNA配列を解析した。DNA配列の解析結果は以下の通りであり、4つのDNA配列を見いだすことができた。

市販されているpET22b(+)(Merck Millipore株式会社より入手可能)由来のプラスミドVector2に、制限酵素SfiI切断サイトと3xFlagタグを挿入し、Vector2を制限酵素SfiIで処理した。制限酵素SfiI切断サイトは、GGCCCAGCCGGCC(配列番号6)若しくはGGCCTCTGCGGCC(配列番号7)により表されるDNA配列からなる。
市販されているpET22b(+)(Merck Millipore株式会社より入手可能)由来のプラスミドVector2に、制限酵素SfiI切断サイトと3xFlagタグを挿入し、Vector2を制限酵素SfiIで処理した。制限酵素SfiI切断サイトは、GGCCCAGCCGGCC(配列番号6)若しくはGGCCTCTGCGGCC(配列番号7)により表されるDNA配列からなる。

このようにしてライゲーションしたプラスミドVector2を用いて、大腸菌(Competent Cell BL21 (DE3) ,New England Biolabsより入手)にトランスフェクションした。
次いで、大腸菌をプレート上に100μg/mLの濃度を有するアンピシリン含有2YT(2YTA)培地で15時間培養して、ドレブリン結合ペプチドを発現する大腸菌株を得た。
2YTAG培地を含むアガープレートに形成されたコロニーの中から、1つのコロニーを選択し、爪楊枝でピックアップした。ピックアップしたコロニーを、2mLの2YTA培地中で、600nmの波長での混合液の吸光度が2.0になるまで200rpmで振とうしながら摂氏37度の温度で培養した。
さらに、2mLの培養液を200mLの2YTA培地に混合した。600nmの波長での混合液の吸光度が0.8になるまで、混合液を200rpmで振とうしながら摂氏37度の温度で培養した。
吸光度が0.8になった後、イソプロピルチオガラクトシド溶液を混合液に添加した。イソプロピルチオガラクトシド溶液の最終濃度は10μMであった。混合液に含有される大腸菌を、摂氏25度の温度で15時間培養した。培養した大腸菌を回収するため、混合液を8000rpmで5分間遠心した。
回収した大腸菌を、45mLのPBSに混合し、ボルテックスミキサーを用いて撹拌した。このようにして、大腸菌を洗浄し、混合液に含有される大腸菌を、超音波を用いて破砕した。大腸菌を含む破砕液を、8000rpmで5分間遠心し、上清を回収した。回収した上清を、0.8μmフィルタを用いて濾過した。
濾液を、Ni Sepharose 6 Fast Flow(GE Healthcare社より入手)を用いて推奨プロトコルに従って精製した。精製は45mLの濾液に対して、3mLの溶出液を用いた。溶出液に含有される緩衝液を、透析によってPBSに置換した。置換は3mLの溶出液に対して、1.5LのPBSを用いた。このようにして、ドレブリン結合ペプチドを含有する溶液を得た。
このようにして得られた溶液に含有されるドレブリン結合ペプチドを、吸光度計(島津製作所より入手、商品名:BioSpec-nano)を用いて、280nmでの波長での吸収測定値に基づいて定量した。
(F)ドレブリン結合ペプチド-ヒトIgG Fc融合抗体の発現
市販されているpcDNA3.1(+)(例えば、Thermo Fisher Scientific株式会社より入手可能)由来のプラスミドVector3に、制限酵素SfiI切断サイトとヒトIgG-Fc領域を挿入し、Vector3を制限酵素SfiIにより処理した。制限酵素SfiI切断サイトは、GGCCCAGCCGGCC(配列番号6)若しくはGGCCTCTGCGGCC(配列番号7)により表されるDNA配列からなる。また、このヒトIgG-Fc領域は、N末端側がヒンジ領域の一部をなしており、このヒンジ領域の一部がVHHと結合する。

このようにしてライゲーションしたプラスミドVector3を用いて、ヒト培養細胞株(HEK293T)にトランスフェクションした。
トランスフェクションは、遺伝子導入試薬(HilyMax、同人化学社製)を用いて推奨プロトコルに従って行われた。培養液中に、ドレブリン結合ペプチド-ヒトIgG Fc融合抗体を産生した。このようにして、ドレブリン結合ペプチド-ヒトIgG Fc融合抗体を含有する溶液を得た。
2.配列番号1のアミノ酸配列を有するドレブリン結合ペプチド-ヒトIgG Fc融合抗体を用いたウェスタンブロット法によるドレブリン検出

#1ペプチドは配列番号1のアミノ酸配列を有するドレブリン結合ペプチドに3つのFlag配列を融合して発現させたものであり、2次抗体としてHRP標識抗Flag抗体を用いることにより検出できる。#1Fc抗体は配列番号1のアミノ酸配列を有するドレブリン結合ペプチドにヒト免疫グロブリンGのFc領域が融合してあり二量体を形成している。#1Fc抗体は、2次抗体としてHRP標識抗ヒト免疫グロブリンG抗体を用いることによって検出できる。
抗Myc抗体及びM2F6がドレブリンに特異的に結合することは既に証明されており、ドレブリンの分子量が140kDaであることが分かった(図1の第1レーン及び第2レーンを参照)。そして、#1ペプチド及び#1Fc抗体を一次抗体としてMyc-drebrin Aと反応させた結果、140kDaの位置でバンドを確認できたため、#1ペプチド及び#1Fc抗体がドレブリンに特異的に結合することが分かった。
さらに、ドレブリンノックアウトマウス(以下、KO。図1同じ)、野生型マウス(以下、WT。図1同じ)の大脳皮質抽出物を用いてウェスタンブロット法を行った。そうしたところ、#1Fc抗体を一次抗体として反応させた結果、M2F6を一次抗体として反応させた場合と同様に、WTの140kDaの位置にのみバンドが現れることが確認できた。また、KOにおいては同位置にバンドが確認できなかったため、#1Fc抗体が特異的に野生型マウスのドレブリンを認識していることが分かった(第5~8レーン)。
3.配列番号2~4により表されるアミノ酸配列を有するドレブリン結合ペプチドを用いたウェスタンブロット法によるドレブリン検出

#2ペプチド~#4ペプチドは、それぞれ配列番号2、3、4のアミノ酸配列を有するドレブリン結合ペプチドに3つのFlag配列を融合して発現させたものであり、2次抗体としてHRP標識抗Flag抗体を用いることにより検出できる。#2Fc抗体~#4Fc抗体は、それぞれ配列番号2、3、4のアミノ酸配列を有するドレブリン結合ペプチドにヒト免疫グロブリンGのFc領域が融合してあり二量体を形成している。#2Fc抗体~#4Fc抗体は、2次抗体としてHRP標識抗ヒト免疫グロブリンG抗体を用いることによって検出できる。
抗Myc抗体及びM2F6がドレブリンに特異的に結合することは既に証明されており、図2よりドレブリンの分子量が140kDaであることが分かった(図2の第1レーン及び第2レーンを参照)。そして、Myc-drebrin Aを#2ペプチド~#4ペプチド及び#2Fc抗体~#4Fc抗体を一次抗体として反応させた結果、140kDaの位置でバンドを確認できたため、#2ペプチド~#4ペプチド及び#2Fc抗体~#4Fc抗体がドレブリンに特異的に結合することが分かった。
4.ドレブリン結合ペプチドを用いた海馬神経細胞の免疫蛍光染色
固定後の初代培養海馬神経細胞にドレブリン結合ペプチド(#4Fc)を特異的に結合させ、ヒト抗体を認識する二次抗体(蛍光標識:画像上では白で表示)を使って本ペプチドの分布を同定した(図3左)。蛍光がスパインに局在していることから本ペプチドがドレブリンに特異的に結合していることがわかる。このことよりドレブリンの局在の検出法として、現在市販のマウスモノクローナル抗体M2F6(図3右)と比べて、同等であることがわかる。
固定後の初代培養海馬神経細胞にドレブリン結合ペプチド(#4Fc)を特異的に結合させ、ヒト抗体を認識する二次抗体(蛍光標識:画像上では白で表示)を使って本ペプチドの分布を同定した(図3左)。蛍光がスパインに局在していることから本ペプチドがドレブリンに特異的に結合していることがわかる。このことよりドレブリンの局在の検出法として、現在市販のマウスモノクローナル抗体M2F6(図3右)と比べて、同等であることがわかる。
5.GFP・Fc-融合ドレブリン結合ペプチドによる、ドレブリンの神経細胞内イメージング
初代培養海馬神経細胞に、N末端にGFPを融合したGFP・Fc-融合ドレブリン結合ペプチド(GFP-#4Fc)を強制発現させた。図4の蛍光画像により、発現したペプチドの集積部位(Aの矢印)とマウスモノクローナル抗体M2F6により明らかとされた内在性ドレブリンの集積部位(Bの矢印)が一致していることがわかる。
初代培養海馬神経細胞に、N末端にGFPを融合したGFP・Fc-融合ドレブリン結合ペプチド(GFP-#4Fc)を強制発現させた。図4の蛍光画像により、発現したペプチドの集積部位(Aの矢印)とマウスモノクローナル抗体M2F6により明らかとされた内在性ドレブリンの集積部位(Bの矢印)が一致していることがわかる。
なお、GFP・Fc-融合ドレブリン結合ペプチドの強制発現は、以下のようにして作製したpEGFP-C1-#4Fcを初代培養海馬神経細胞に導入することにより行った。
pEGFP-C1ベクター(GenBank Accession #: U55763 Clontech社より入手)のマルチクローニングサイトに存在するBglII、EcoRIサイトを制限酵素で切断後、アガロースゲルで電気泳動を行い、クローニングベクターの精製を行った。また、当該ペプチド#4Fcをコードする、配列番号11で示される配列を含む核酸フラグメントをBglII、EcoRIサイトを有するプライマーを用いたPCRで増幅し、同様に制限酵素処理及びアガロースゲル電気泳動を行い、挿入核酸の精製を行った。上記のクローニングベクターと挿入核酸のライゲーションを行い、目的のGFP融合ペプチド発現ベクターを作製した。
pEGFP-C1ベクター(GenBank Accession #: U55763 Clontech社より入手)のマルチクローニングサイトに存在するBglII、EcoRIサイトを制限酵素で切断後、アガロースゲルで電気泳動を行い、クローニングベクターの精製を行った。また、当該ペプチド#4Fcをコードする、配列番号11で示される配列を含む核酸フラグメントをBglII、EcoRIサイトを有するプライマーを用いたPCRで増幅し、同様に制限酵素処理及びアガロースゲル電気泳動を行い、挿入核酸の精製を行った。上記のクローニングベクターと挿入核酸のライゲーションを行い、目的のGFP融合ペプチド発現ベクターを作製した。
6.ランダム変異ライブラリによるドレブリン結合ペプチドの作製
実施例1で得られたドレブリン結合ペプチド遺伝子に対するランダム変異により、ドレブリン結合ペプチドのスクリーニングを行った。

(A)ランダム変異ライブラリの構築
配列番号19(配列番号4から、C末端側Fcヒンジ領域を除去したもの)のドレブリン結合ペプチドをコードする配列番号11の遺伝子に対してError Prone PCRを行ってランダム変異を挿入し、ファージミドベクターへと連結させた。作製したファージミドベクターを大腸菌へと形質転換することで約1.0×108の多様性を有するランダム変異ライブラリを構築した。
配列番号19(配列番号4から、C末端側Fcヒンジ領域を除去したもの)のドレブリン結合ペプチドをコードする配列番号11の遺伝子に対してError Prone PCRを行ってランダム変異を挿入し、ファージミドベクターへと連結させた。作製したファージミドベクターを大腸菌へと形質転換することで約1.0×108の多様性を有するランダム変異ライブラリを構築した。
(B)ランダム変異ライブラリを用いたドレブリン結合ペプチドのスクリーニング
実施例1(C)に記載の方法により、2ラウンドのパニングを行った。単離した86クローンの結合活性を配列番号19のドレブリン結合ペプチドと比較するため、実施例1(D)に記載の方法によりELISAを行ったところ、ほぼ全てのクローンで結合活性の上昇が確認できた。特に結合活性の高かった10クローン(Z01~Z10)の結合活性を図5に示す。10クローンのうち、Z02~Z10の9クローンは独立していた。
実施例1(C)に記載の方法により、2ラウンドのパニングを行った。単離した86クローンの結合活性を配列番号19のドレブリン結合ペプチドと比較するため、実施例1(D)に記載の方法によりELISAを行ったところ、ほぼ全てのクローンで結合活性の上昇が確認できた。特に結合活性の高かった10クローン(Z01~Z10)の結合活性を図5に示す。10クローンのうち、Z02~Z10の9クローンは独立していた。
(C)ランダム変異により得られたドレブリン結合ペプチド-ヒトIgG Fc融合抗体の、ELISA法によるドレブリン結合活性評価
実施例1(F)に記載の方法により、Z02~Z10を有するドレブリン結合ペプチド-ヒトIgG Fc融合抗体発現ベクターを作製した。作製した発現ベクターをHEK293T細胞にトランスフェクションし、ドレブリン結合ペプチド-ヒトIgG Fc融合抗体を一過性に培養上清中に発現させ、ELISAで結合活性を評価した。
実施例1(F)に記載の方法により、Z02~Z10を有するドレブリン結合ペプチド-ヒトIgG Fc融合抗体発現ベクターを作製した。作製した発現ベクターをHEK293T細胞にトランスフェクションし、ドレブリン結合ペプチド-ヒトIgG Fc融合抗体を一過性に培養上清中に発現させ、ELISAで結合活性を評価した。
結果を図6に示す。オリジナルである配列番号19のアミノ酸配列を含むペプチドの結合活性に比べて、ランダム変異クローンはどれも結合活性が低く、上記実施例6(B)とは異なる結果が得られた。この理由は明らかではなく、ランダム変異クローンは大腸菌内での発現に適した変異が加えられて生産性が高まり、VHH提示ファージにおけるVHHの提示量が増加した結果、実施例6(B)のELISAでは結合活性が高く測定されたのみであって、ペプチドの結合活性自体は低下した可能性がある。一方で、ランダム変異クローンは、Fc融合抗体にすると結合活性が低下するが、ペプチドの結合活性自体は上がっている可能性もあるため、ランダム変異クローン全てについてさらなる解析を行った。なお、Z03、Z07は、cDNAを単離することができなかった。
7.ドレブリン結合ペプチドによる、ドレブリンの神経細胞内イメージング
上記実施例6(C)で得られた各クローンについて、pEGFP-N1ベクター(GenBank Accession #: U55762 Clontech社より入手)に組み入れてC末端側にGFPを融合したコンストラクトを作製し、実施例5と同様の方法により初代培養海馬神経細胞にGFP・Fc-融合ドレブリン結合ペプチドを発現させ、蛍光画像を取得した。また、ヒトのIgG抗体のFc領域と融合していないドレブリン結合ペプチドについても、同様の方法でC末端側にGFPを融合したコンストラクトを作製し、実施例5と同様の方法により初代培養海馬神経細胞にGFP-融合ドレブリン結合ペプチドを発現させ、蛍光画像を取得した。
上記実施例6(C)で得られた各クローンについて、pEGFP-N1ベクター(GenBank Accession #: U55762 Clontech社より入手)に組み入れてC末端側にGFPを融合したコンストラクトを作製し、実施例5と同様の方法により初代培養海馬神経細胞にGFP・Fc-融合ドレブリン結合ペプチドを発現させ、蛍光画像を取得した。また、ヒトのIgG抗体のFc領域と融合していないドレブリン結合ペプチドについても、同様の方法でC末端側にGFPを融合したコンストラクトを作製し、実施例5と同様の方法により初代培養海馬神経細胞にGFP-融合ドレブリン結合ペプチドを発現させ、蛍光画像を取得した。
結果を図7に示す。図7Aは、GFP・Fc-融合ドレブリン結合ペプチドによりドレブリンの細胞内イメージングを行った結果を示す。Z04、Z10以外のクローンにおいて、GFP・Fc-融合ドレブリン結合ペプチドが細胞内に発現し、親クローンである配列番号19と同程度に、内在性ドレブリンの集積部位を検出することができた。
また、Z02、Z06について、ヒトのIgG抗体のFc領域と融合していない、GFP-融合ドレブリン結合ペプチドを用いて同様の試験を行ったところ、Fc領域を融合した場合と同程度に、内在性ドレブリンの集積部位を検出することができた(図7B)。
8.取得クローンのアミノ酸配列
実施例7で、内在性ドレブリンの集積部位を検出することができた5クローンのアミノ酸配列を決定した。得られた配列を以下に示す。なお、下線は配列番号19からの変異箇所である。

本発明のドレブリン結合ペプチドの用途は、ドレブリンの検出や定量のみに限られない。例えばGFPを付加したドレブリン結合ペプチドを作成すれば、ドレブリンの細胞内イメージングが可能となる。これにより、神経科学分野のみならず癌や感染症の分野においても種々の疾患の病態解明に寄与することが可能となるため、産業上非常に有用である。
Claims (14)
- 以下の(1)若しくは(2)に記載のアミノ酸配列を含むことを特徴とするドレブリン結合ペプチド又はその機能性断片。
(1)配列番号16~24のいずれかで示されるアミノ酸配列;
(2)配列番号16~24のいずれかで示されるアミノ酸配列において、1若しくは数個のアミノ酸が挿入、欠失、置換、及び/又は付加されたアミノ酸配列; - マーカータンパク質及び/若しくはペプチドタグを融合したことを特徴とする請求項1に記載のペプチド又はその機能性断片。
- マーカータンパク質が蛍光タンパク質であり、ペプチドタグがFlagであることを特徴とする請求項2に記載のペプチド又はその機能性断片。
- 検出可能な標識を付加したことを特徴とする請求項1~3のいずれかに記載のペプチド又はその機能性断片。
- 標識が、酵素、蛍光物質、ジゴキシゲニン、若しくはビオチンであることを特徴とする請求項4に記載のペプチド又はその機能性断片。
- 細胞内移行ペプチドを付加したことを特徴とする請求項1~5のいずれかに記載のペプチド又はその機能性断片。
- 請求項1~6のいずれかに記載のペプチド又はその機能性断片に、IgG-Fc領域を融合させたことを特徴とする抗体。
- 請求項1~6のいずれかに記載のペプチド又はその機能性断片、或いは請求項7に記載の抗体をコードするポリヌクレオチド。
- 配列番号8~11のいずれかで示される塩基配列を含むか、又は配列番号8~11のいずれかで示される塩基配列の相補配列を有するポリヌクレオチドとストリンジェントな条件下でハイブリダイズすることを特徴とする請求項7に記載のポリヌクレオチド。
- 請求項8又は9に記載のポリヌクレオチドを含むベクター。
- 請求項8又は9に記載のポリヌクレオチド、或いは請求項10に記載のベクターが導入されたことを特徴とする、請求項1~6のいずれかに記載のペプチド又はその機能性断片、或いは請求項7に記載の抗体を発現する細胞。
- 請求項1~6のいずれかに記載のペプチド又はその機能性断片、或いは請求項7に記載の抗体を用いた、被検試料中のドレブリンの検出方法であって、前記ペプチド又は抗体を前記被検試料に添加又は導入する工程を含む方法。
- 請求項1~6のいずれかに記載のペプチド又はその機能性断片、或いは請求項7に記載の抗体を、発色又は発光により検出する工程をさらに含むことを特徴とする請求項12に記載の方法。
- 検出する工程が、発光に基づく画像解析法であることを特徴とする請求項13に記載の方法。
