본 발명은 합성가스 발효 균의 합성가스 발효 시의 대사체 분석을 위한 대사체 샘플링 및 분석 방법에 관한 것으로, 대사체 샘플링을 위한 최적 조건을 확립하고, 선정된 대사체 바이오마커를 사용하여 합성가스 발효 균의 포도당 배양과 합성가스 배양의 구별할 수 있다.
Description
본 발명은 합성가스 발효 균의 합성가스 발효 시의 대사체 분석을 위한 대사체 샘플링 및 분석 방법에 관한 것이다.
미생물에서 대사체 샘플링(metabolome sampling)의 중요성으로 인해 그람 음성 균인 사카로퍼거스 데그라단스, 대장균, 효모를 포함한 다양한 미생물에서 대사체 분석을 위한 대사체 샘플링 방법의 최적화 연구가 진행되고 있으며 이렇게 최적화된 대사체 샘플링 방법을 기반으로 대사체의 변화에 대한 다양한 메커니즘에 관한 연구들이 진행되고 있다(Rabinowitz JD and Kimball E. (2007) Anal Chem vol. 79, pp. 6167-6173; Shin MH et al (2010) Anal Chem vol. 82, pp. 6660-6666; Kim S et al (2013) Anal Chem vol. 85, pp. 2169-2176). 혐기성 균의 대사체 샘플링 방법의 경우 Clostridium
acetobutylicum의 호기성 환경에서 메탄올을 이용한 대사체 추출법이 보고된 바 있지만(Lee SH et al (2014) Biotechnol Bioeng vol. 111, pp. 2528-2536), 탄소를 고정화하는 미생물의 경우엔 대사체 샘플링 방법의 최적화 연구가 보고된 바가 없다.
합성가스 발효 미생물은 일반적인 당류를 기질로 사용할 때의 대사회로와는 완전히 다른 대사회로를 이용하여 에너지를 생성하고 생장하기 때문에(Richter H et al (2016) Energy Environ Sci vol. 9, pp. 2392-2399), 대사물질의 종류 및 양이 크게 차이가 난다. 이러한 대사체는 추출 용매의 성질에 따라서 추출 효율이 크게 달라질 수 있으며, 대사체 프로파일링의 차이를 보일 수 있다(Duportet X et al (2012) Metabolomics vol. 8, pp. 410-421; Canelas AB et al (2009) Anal Chem vol. 81, pp. 7379-7389). 이러한 대사체 프로파일링의 차이는 생물학적 해석이나 메커니즘의 해석을 제한하거나 바꿀 수 있다.
따라서, 합성가스 발효 미생물의 합성가스 배양 시의 대사체 추출 효율을 높이고 재현성 있는 추출을 가능하게 하여, 생물학적 해석이나 메커니즘 해석의 오도를 줄일 수 있는 추출 용매의 최적화가 필요하다.
이에, 본 발명자들은 합성가스 발효 미생물의 특유의 대사체 분석을 위해 합성가스 발효 미생물의 포도당 배양 및 합성가스 배양을 통하여 합성가스 배양 특유의 대사체를 추출 및 정성, 정량하고, 이러한 탄소고정화 미생물의 합성가스 배양에서 대사체를 추출 효율 및 재현성을 기반으로 최대한으로 추출할 수 있는 최적의 대사체 추출 용매를 확립함으로써 본 발명을 완성하게 되었다.
따라서, 본 발명은 합성가스 발효 균의 포도당 배양과 합성가스 배양의 구별용 키트를 제공하는데 그 목적이 있다.
또한, 본 발명은 합성가스 발효 균의 포도당 배양과 합성가스 배양을 구별하기 위한 대사체 차별성을 분석하는 방법을 제공하는데 목적이 있다.
본 발명은
팔미트산(palmitic acid), 스테아르산(stearic acid), 아라키드산(arachidic acid), 마르가르산(heptadecanoic acid), 1-모노팔미틴(1-monopalmitin), 알라닌(alanine), N-메틸알라닌(N-methylalanine), 아데노신(adenosine), 글리세롤-1-인산(glycerol-1-phosphate) 및 발린(valine)으로 이루어진 군에서 선택된 하나 이상의 대사체에 대한 정량 장치를 포함하는 합성가스 발효 균의 포도당 배양과 합성가스 배양의 구별용 키트를 제공한다.

호기 조건에서 합성가스 발효 균의 생체시료를 초고속 여과(fast filtration)하고, 여과물을 물로 세척한 다음, 추출 용매로 물, 2-프로판올 및 메탄올의 혼합용매를 사용하여 대사체를 추출하는 대사체 샘플링 단계를 포함하는, 합성가스 발효 균의 포도당 배양과 합성가스 배양을 구별하기 위한 대사체 차별성을 분석하는 방법을 제공한다.

본 발명은 합성가스 발효 균의 대사체 분석을 통한 다양한 메커니즘 연구에 이용될 것으로 기대한다. 또한 각 미생물에 맞는 최적의 대사체 샘플링 방법이 필요함을 입증함으로써 이를 이용하여 다른 미생물의 대사체 샘플링 방법의 최적화에 응용할 수 있다.

도 8은 ROC curve를 이용하여 합성가스 발효 균의 포도당 배양과 합성가스 배양 시의 주요한 10개 대사체를 변수로 포도당 배양과 합성가스 배양을 구분하는 PCA 모델을 검증한 것이다.
본 발명은 팔미트산(palmitic acid), 스테아르산(stearic acid), 아라키드산(arachidic acid), 마르가르산(heptadecanoic acid), 1-모노팔미틴(1-monopalmitin), 알라닌(alanine), N-메틸알라닌(N-methylalanine), 아데노신(adenosine), 글리세롤-1-인산(glycerol-1-phosphate) 및 발린(valine)으로 이루어진 군에서 선택된 하나 이상의 대사체에 대한 정량 장치를 포함하는 합성가스 발효 균의 포도당 배양과 합성가스 배양의 구별용 키트에 관한 것이다.
본 발명자들은 합성가스 발효 균의 포도당 배양과 합성가스 배양의 구별하는 바이오마커를 찾기 위해 호기성 조건에서 초고속 여과 후 물을 이용한 세척 및 순수 메탄올을 사용하여 대사체를 추출하고 GC/TOF MS를 이용하여 포도당 배양 및 합성가스 배양 대사체 프로파일 차이를 비교 분석하고, 이 차이를 이용하여 합성가스 발효 균의 포도당 배양과 합성가스 배양의 구별할 수 있는 바이오마커 발굴 연구를 수행하였다.
그 결과, 아민류, 아미노산류, 지방산류, 유기산류, 인산류, 당류 등으로 구분할 수 있는 82종의 대사체를 동정하였다. 이 중 유기산류, 지방산류, 당류가 가장 많이 검출되었으며, 그 다음으로 아미노산류, 아민류, 인산류 등의 순서로 검출되었다.
합성가스 발효 균의 포도당 배양과 합성가스 배양의 지수기 및 정체기에서 각각 샘플링하여 생체 시료를 비교하였을 때, 부분최소자승판별분석(PLS-DA)을 통해 합성가스 발효 균의 포도당 배양과 합성가스 배양의 대사체 프로파일의 명확한 차이를 확인하였으며, 각각의 대사물질에 대해, PLS-DA 모델의 로딩값을 기준으로 최대 양수값 및 최대 음수값을 갖는 대사물질을 각각 5종씩 선정하고 10종의 대사체를 신규 바이오마커 후보 물질로 선정하였다(표 2). 각각의 대사물질은 합성가스 발효 균의 포도당 배양과 합성가스 배양에서 통계적으로 확연한 차이를 보여, 적절한 후보 생체표지자임을 확인하였다. 또한 HCA를 시행하여, 합성가스 발효 균의 포도당 배양과 합성가스 배양의 개별적인 대사물질의 차이를 보여주고자 하였다, 그 결과 각 배양별로 개별적인 대사물질의 확연한 차이를 확인하였다.
또한, 상기 후보 대상물질을 이용해 합성가스 발효 균의 포도당 배양과 합성가스 배양을 구분하기 위하여 PCA를 이용한 모델을 만들었으며, 10종의 대사체를 이용해 생성한 모델이 각 배양 조건의 대사체를 완전히 구분한 결과를 보였다. 또한 이 모델에 ROC curve를 적용하여 검증을 시행하였다. 그 결과 높은 통계학적 유의미성을 띠어 합성가스 발효 균의 포도당 배양과 합성가스 배양 시의 대사체 구분에 적절함을 검증할 수 있었다.
본 명세서에서 용어 "합성가스 발효 균"은 클로스트리디움 카르복시디보란스(Clostridium
carboxidivorans)을 의미하며, 구체적으로 클로스트리디움 카르복시디보란스 P7(Clostridium
carboxidivorans P7)을 포함한다.
특히, 포도당 배양에서의 합성가스 발효 균은 대사체 중에서 알라닌(alanine), N-메틸알라닌(N-methylalanine), 아데노신(adenosine), 글리세롤-1-인산(glycerol-1-phosphate) 및 발린(valine)은 증가하는 경향을, 팔미트산(palmitic acid), 스테아르산(stearic acid), 아라크산(arachidic acid), 마르가르산(heptadecanoic acid) 및 1-모노팔미틴(1-monopalmitin)은 감소하는 경향을 나타낸다.
또한, 합성가스 배양에서의 합성가스 발효 균은 대사체 중에서 팔미트산(palmitic acid), 스테아르산(stearic acid), 아라키드산(arachidic acid), 마르가르산(heptadecanoic acid) 및 1-모노팔미틴(1-monopalmitin)은 증가하는 경향을, 알라닌(alanine), N-메틸알라닌(N-methylalanine), 아데노신(adenosine), 글리세롤-1-인산(glycerol-1-phosphate) 및 발린(valine)은 감소 경향을 나타낸다.
상기 증가 또는 감소 경향이란 대사체 농도의 증가 또는 감소를 의미하는 것으로, 용어 “대사체 농도의 증가”는 합성가스 배양 시의 합성가스 발효 균에서 포도당 배양 대비 대사체 농도가 측정 가능할 정도로 유의하게 증가된 것을 의미하며, 본 명세서에서, 용어 “대사체 농도의 감소”는 합성가스 배양 시의 합성가스 발효 균에서 포도당 배양 대비 대사체 농도가 측정 가능할 정도로 유의하게 감소된 것을 뜻한다.
본 발명의 키트에 포함된 정량 장치는 크로마토그래피/질량분석기일 수 있다.
본 발명에서 이용되는 크로마토그래피는 가스 크로마토그래피(Gas Chromatography), 액체-고체 크로마토그래피(Liquid-Solid Chromatography, LSC), 종이 크로마토그래피(Paper Chromatography, PC), 박층 크로마토그래피(Thin-Layer Chromatography, TLC), 기체-고체 크로마토그래피(Gas-Solid Chromatography, GSC), 액체-액체 크로마토그래피(Liquid-Liquid Chromatography, LLC), 포말 크로마토그래피(Foam Chromatography, FC), 유화 크로마토그래피(Emulsion Chromatography, EC), 기체-액체 크로마토그래피(Gas-Liquid Chromatography, GLC), 이온 크로마토그래피(Ion Chromatography, IC), 겔 여과 크로마토그래피(Gel Filtration Chromatograhy, GFC) 또는 겔 투과 크로마토그래피(Gel Permeation Chromatography, GPC)를 포함하나, 이에 제한되지 않고 당업계에서 통상적으로 사용되는 모든 정량용 크로마토그래피를 사용할 수 있다. 바람직하게는, 본 발명에서 이용되는 크로마토그래피는 GC/TOF MS(gas chromatography/time-of-flight mass spectrometry) 분석기기일 수 있다.
본 발명의 대사체는 가스 크로마토그래피에서 각 성분들이 분리되며, TOF MS를 거쳐 얻어진 정보를 이용하여 정확한 분자량 정보뿐만 아니라 구조 정보(elemental composition)를 통해 구성 성분을 확인한다.
본 발명은 또한 합성가스 발효 균의 포도당 배양과 합성가스 배양을 구별하기 위한 대사체 차별성을 분석하는 방법을 포함한다.
상기 대사체 차별성을 분석하는 방법은 포도당 배양 시의 지수기 및 정체기와 합성가스 배양 시의 지수기 및 정체기 간의 별성을 분석하는 방법으로써, 우선, 퀜칭 과정과 대사체 추출 과정을 포함하는 대사체 샘플링 단계를 거친다.
대사체 샘플링은 호기 조건에서 혐기성 균의 생체시료를 호기성 조건 하에서 초고속 여과(fast filtration)하고, 여과물을 물로 세척한 다음, 추출 용매로 아세토니트릴과 물의 혼합 용매, 아세토니트릴, 메탄올 및 물의 혼합 용매, 순수 메탄올, 또는 물, 2-프로판올 및 메탄올의 혼합 용매를 사용하여 대사체를 추출하는 과정이다. 이때, 세척에 사용되는 물은 혐기성 균액 1 부피비에 대하여, 3 부피부 내지 7 부피부로 사용하는 것이 바람직하며, 추출용매로는 물, 2-프로판올 및 메탄올의 혼합 용매가 추출 효율 및 재현성 면에서 바람직하다. 특히, 물: 2-프로판올 : 메탄올 = 2:2:5 (v/v/v)로 혼합된 혼합 용매를 사용하는 것이 보다 바람직하다.
본 발명의 일 구체예에 따르면, 아민류, 아미노산류, 당 및 당 알코올류, 지방산류, 인산류, 유기산류 등을 포함한 86개의 대사체가 동정되었다.
상기 대사체 샘플링 단계에서 추출된 대사체에 대해서는 다음의 분석 단계를 거친다:
추출된 대사체를 GC/TOF MS(gas chromatography/time-of-flight mass spectrometry) 분석기기로 분석하는 단계;
GC/TOF MS 분석 결과를 통계처리 가능한 수치로 변환하는 단계; 및
변환된 수치를 이용하여 통계학적으로 상기 두 생체시료군의 차별성을 검증하는 단계를 더 포함한다.
다음으로, 대사체의 프로파일링 차이를 비교하기 위해 부분최소제곱회귀법(Partial least squares discriminant analysis: PLS-DA)을 수행하여 두 생체시료군 간의 유의적인 차이를 나타내는 대사체 바이오마커를 선정하고, 분석 및 검증한다.
일 구현예로서, 본 발명의 분석 방법은 GC/TOF MS 분석 결과를 통계처리 가능한 수치로 변환하는 단계는 총 분석시간을 단위시간 간격으로 나누어 단위시간 동안 나타난 크로마토그램 피크의 면적 또는 높이 중 가장 큰 수치를 단위시간 동안의 대표값으로 정한다.
변환된 수치를 이용하여 통계학적으로 상기 두 생체시료군의 차별성을 검증하는 단계는 부분최소제곱회귀법(Partial least squares discriminant analysis: PLS-DA)을 수행하여 두 생체시료군 간의 유의적인 차이를 나타내는 대사체 바이오마커를 분석 및 검증한다.
대사체 바이오마커는 합성가스 발효 균의 포도당 배양과 합성가스 배양을 구별한다.
대사체 바이오마커는 팔미트산(palmitic acid), 스테아르산(stearic acid), 아라키드산(arachidic acid), 마르가르산(heptadecanoic acid), 1-모노팔미틴(1-monopalmitin), 알라닌(alanine), N-메틸알라닌(N-methylalanine), 아데노신(adenosine), 글리세롤-1-인산(glycerol-1-phosphate) 및 발린(valine)으로 구성된다.
부분최소제곱회귀법(Partial least squares discriminant analysis: PLS-DA)의 로딩 값이 양수인 것은 대사체 바이오마커의 증가 경향을, 로딩 값이 음수인 것은 대사체 바이오마커의 감소 경향을 나타낸다.
부분최소제곱회귀법의 로딩 값이 양수인 것은 대사체 바이오마커의 증가 경향을, 로딩 값이 음수인 것은 대사체 바이오마커의 감소 경향을 나타내는 것으로 판정한다. 본 발명의 일 구현예에 따르면, 합성가스 발효 균의 포도당 배양과 합성가스 배양 시의 대사체를 구별하기 위한 바이오마커로, 팔미트산(palmitic acid), 스테아르산(stearic acid), 아라키드산(arachidic acid), 마르가르산(heptadecanoic acid), 1-모노팔미틴(1-monopalmitin), 알라닌(alanine), N-메틸알라닌(N-methylalanine), 아데노신(adenosine), 글리세롤-1-인산(glycerol-1-phosphate) 및 발린(valine)을 사용할 수 있다.
상기 바이오마커들은 합성가스 배양 시의 합성가스 발효 균에서, 팔미트산(palmitic acid), 스테아르산(stearic acid), 아라키드산(arachidic acid), 마르가르산(heptadecanoic acid) 및 1-모노팔미틴(1-monopalmitin)은 증가하는 경향을, 알라닌(alanine), N-메틸알라닌(N-methylalanine), 아데노신(adenosine), 글리세롤-1-인산(glycerol-1-phosphate) 및 발린(valine)은 감소 경향을 나타낼 수 있다.
이하, 본 발명에 따르는 실시예를 통하여 본 발명을 보다 상세히 설명하나, 본 발명의 범위가 하기 제시된 실시예에 의해 제한되는 것은 아니다.
[실시예]
참조예 1: 균주, 배지 및 배양 조건
C. carboxidivorans P7을 2x 효모 추출물, tryptone 및 포도당 (YTG) 배지(표 1)에서 배양하여 종 배양액을 수득하였다. 세포는 OD600이 2.0-2.5에 도달할 때 수확되었다 (지수 중간 단계로 간주). 세포를 변형된 P7 배지(표 1)로 2회 세척하고 주 배양물로서 25 mL 혈청 병에서 5 mL의 변형된 P7 배지로 접종하였다 (10%, v/v). CO 발효를 위해 기체 혼합물 (10% H2, 70% CO, 20% CO2, v/v/v, Air Korea, Seoul, Korea)을 사용하여 혈청 병의 헤드 스페이스에 1.5 bar를 가하고 세포를 37 ℃ 및 200 rpm에서 배양하였다. 포도당 발효를 위해 25 mL 혈청 병에서 5 mL의 변형된 P2 배지(MP2)를 주 배양에 사용하였다 (Phillips JR, Atiyeh HK, Tanner RS, Torres JR, Saxena J, Wilkins MR, Huhnke RL. 2015. Butanol and hexanol production in Clostridium
carboxidivorans syngas fermentation: medium development and culture techniques. Bioresour Technol 190:114-121.). 세포를 포도당 배지에 접종하고 37 ℃에서 200 rpm으로 진탕 배양하였다. 모든 배지는 아르곤 (99.9%, w/w)으로 퍼징한 후 혐기성으로 준비하였다.
하기 실시예에서 사용된 합성가스 발효 균주는 클로스트리디움 카르복시디보란스 P7(Clostridium
carboxidivorans P7)이다.

프로파일링
합성가스 발효 균의 포도당 배양과 합성가스 배양의 지수기 및 정체기에서 각각 균액 2 ml를 샘플링하여 대기 중에서 초고속 여과 방법을 수행하고, 증류수 10 ml를 이용하여 세척하였다. 기존에 많이 사용되고 있는 순수 메탄올 10 ml을 추출 용매로 이용하여 여과물과 혼합 및 액체질소에 얼려서 대사활성을 정지시켰다. 이후 얼음 위에서 혼합액을 녹인 후 5분간 초음파 처리, 3분간 볼텍싱 한 후에 16,100 g, 4 ℃에서 10분간 원심분리 후 상층액을 완전 건조하여 GC/TOF MS로 분석하였다.
합성가스 발효 균의 포도당 배양과 합성가스 배양 시의 대사체를 메탄올을 이용하여 추출 및 GC/TOF MS 분석한 데이터를 프로세싱하였을 때, 아민류, 아미노산류, 당 및 당 알코올류, 지방산류, 인산류, 유기산류 등을 포함한 82개의 대사체를 동정하였다(표 2).
합성가스 발효 균의 포도당 배양과 합성가스 배양 시의 대사체 프로파일링 차이를 비교하기 위하여 82개의 대사체를 이용해 PLS-DA 및 HCA를 실시하였다.
PLS-DA의 결과에 따르면 지수기 및 정체기에 관계없이, 포도당 배양 시에는 t[1] 축을 기준으로 양수의 값을, 합성가스 배양 시에는 음수의 값을 나타내서 대사체 프로파일이 확연히 다름을 보였다(도 1, 표 3).
또한, HCA로 개별적인 대사물질의 증감을 살펴본 결과, 포도당 배양 시에는 HCA 상단의 대사체가, 합성가스 발효 시에는 HCA 중, 하단의 대사체가 높은 인텐시티를 보여서 확연히 다름을 보였다(도 2). 따라서 대사체 프로파일링의 차이가 크게 나타남을 확인하였다. 이를 통하여 합성가스 발효 균은 합성가스 배양 시 특유의 대사체 패턴을 가짐을 확인하였다.

합성가스 발효 균주의 포도당 배양 및 합성가스 배양에서 차이를 나타내는 대표적 대사물질을 선정하기 위하여 실시예 1로부터 나온 PLS-DA 분석 모델을 이용하여 모델의 양수의 로딩값 및 음수의 로딩값에서 최대의 값을 갖는 5개의 대사체를 각각 계산하고 총 대사체 10개를 선정하였다(표 4).

합성가스 발효 균의 합성가스 배양 시의 지수기 및 정체기에서 대사체 샘플을 얻기 위하여 호기 조건에서 2 mL의 균액을 샘플링 후 초고속 여과 방법을 사용한 뒤 10 mL의 물로 세척하여 합성가스 발효 균의 각 단계에서 대사체를 얻은 뒤 50ACN(acetonitrile-water=1:1, v/v), AMW(acetonitrile-methanol-water=2:1:1, v/v/v), PM(pure methanol), WiPM(water-2-propanol-methanol=2:2:5, v/v/v)의 용매를 각각 10 ml씩 추출 용매로 이용하여 여과물과 혼합 및 액체질소에 얼려서 대사활성을 정지시켰다. 이후 얼음 위에서 혼합액을 녹인 후 5분간 초음파 처리, 3분간 볼텍싱 한 후에 16,100 g, 4 ℃에서 10분간 원심분리 후 상층액을 완전 건조하여 GC/TOF MS로 분석하였다. GC/TOF-MS로 분석하여 추출 효율을 비교 분석하였다.
합성가스 발효 균의 합성가스 배양 시의 대사체를 4종류의 서로 다른 추출용매를 이용하여 추출 및 GC/TOF MS 분석한 데이터를 프로세싱하였을 때, 아민류, 아미노산류, 당 및 당 알코올류, 지방산류, 인산류, 유기산류 등을 포함한 86개의 대사체를 동정하였다(표 5).
도 3, 도 4 및 표 6에 나타난 바와 같이, 각 단계에서 모두 추출 용매에 따라서 대사체 프로파일링의 차이가 나는 것을 확인할 수 있었으며, 추출 효율 또한 다름을 확인할 수 있었다. 합성가스 발효 균의 지수기 및 정체기에서 정성 및 상대적으로 정량 분석된 피크 인텐시티가 WiPM에서 가장 높아, 종합적인 대사체의 추출 효율이 WiPM에서 가장 높음을 볼 수 있었다 (도 5).
또한 합성가스 발효 균의 지수기 및 정체기에서 추출 용매에 따른 재현성을 살펴보면, %CV 값이 WiPM에서 모두 최저의 수치를 기록하여, 재현성이 제일 높음을 알 수 있었다(도 6). 이를 통하여 합성가스 발효 균의 대사체 분석을 위한 대사체 추출 시에 추출 효율 및 재현성에 기반한 최적 용매로 WiPM을 선정하였다.

실시예
4: 주요
대사체
선정 및 이를 이용하여 포도당 배양과 합성가스 배양 구별 검증
실시예 2로부터 선정된 합성가스 발효 균주의 포도당 배양 및 합성가스 배양에서 차이를 나타내는 대표적 10개의 대사체를 이용하여 합성가스 발효 균주의 포도당 배양과 합성가스 배양을 완전히 구분하기 위하여 PCA 모델을 생성하였다(도 7). 10개의 대사체로 생성된 PCA 모델은 PC1 축을 기반으로 합성가스 배양 시의 대사체 프로파일은 양수를, 포도당 배양 시의 대사체 프로파일은 음수를 띠며 완전히 구분되었다 (도 7A). 10개의 대사체가 PCA 모델에 어떻게 관여하는지 보여주기 위하여 loading plot을 이용하여 나타내었다 (도 7B). 또한, 이러한 구분 모델이 통계학적으로 유의미한지 살펴보기 위하여 ROC curve를 적용하였다(도 8). 그 결과 sensitivity 100%, specificity 100%, AUC 1.000으로 10개의 대사체로 생성된 합성가스 발효 균주의 포도당 배양과 합성가스 배양 구분을 위한 PCA 모델이 통계학적으로 매우 유의미함을 검증할 수 있었다.
Claims (12)
- 팔미트산(palmitic acid), 스테아르산(stearic acid), 아라키드산(arachidic acid), 마르가르산(heptadecanoic acid), 1-모노팔미틴(1-monopalmitin), 알라닌(alanine), N-메틸알라닌(N-methylalanine), 아데노신(adenosine), 글리세롤-1-인산(glycerol-1-phosphate) 및 발린(valine)으로 이루어진 군에서 선택된 하나 이상의 대사체에 대한 정량 장치를 포함하는 합성가스 발효 균의 포도당 배양과 합성가스 배양의 구별용 키트.
- 제 1 항에 있어서,정량 장치는 GC/TOF MS(gas chromatography/time-of-flight mass spectrometry) 분석기기인 키트.
- 제 1 항에 있어서,포도당 배양에서의 합성가스 발효 균은 대사체 중에서 알라닌(alanine), N-메틸알라닌(N-methylalanine), 아데노신(adenosine), 글리세롤-1-인산(glycerol-1-phosphate) 및 발린(valine)은 증가하는 경향을, 팔미트산(palmitic acid), 스테아르산(stearic acid), 아라키드산(arachidic acid), 마르가르산(heptadecanoic acid) 및 1-모노팔미틴(1-monopalmitin)은 감소하는 경향을 나타내는 키트.
- 제 1 항에 있어서,합성가스 배양에서의 합성가스 발효 균은 대사체 중에서 팔미트산(palmitic acid), 스테아르산(stearic acid), 아라키드산(arachidic acid), 마르가르산(heptadecanoic acid) 및 1-모노팔미틴(1-monopalmitin)은 증가하는 경향을, 알라닌(alanine), N-메틸알라닌(N-methylalanine), 아데노신(adenosine), 글리세롤-1-인산(glycerol-1-phosphate) 및 발린(valine)은 감소 경향을 나타내는 키트.
- 제 1 항에 있어서,혐기성 발효 균은 클로스트리디움 카르복시디보란스(Clostridium carboxidivorans)인 키트.
- 합성가스 발효 균의 포도당 배양과 합성가스 배양을 구별하기 위한 대사체 차별성을 분석하는 방법으로서,호기 조건에서 합성가스 발효 균의 생체시료를 초고속 여과(fast filtration)하고, 여과물을 물로 세척한 다음, 추출 용매로 물, 2-프로판올 및 메탄올의 혼합 용매를 사용하여 대사체를 추출하는 대사체 샘플링 단계를 포함하는, 합성가스 발효 균의 포도당 배양과 합성가스 배양을 구별하기 위한 대사체 차별성을 분석하는 방법.
- 제 6 항에 있어서,상기 분석 방법은 추출된 대사체를 GC/TOF MS(gas chromatography/time-of-flight mass spectrometry) 분석기기로 분석하는 단계; GC/TOF MS 분석 결과를 통계처리 가능한 수치로 변환하는 단계; 및 변환된 수치를 이용하여 통계학적으로 합성가스 발효 균의 포도당 배양과 합성가스 배양을 구별하기 위한 두 생체시료군의 차별성을 검증하는 단계를 더 포함하는, 합성가스 발효 균의 포도당 배양과 합성가스 배양을 구별하기 위한 대사체 차별성을 분석하는 방법.
- 제 6 항에 있어서,GC/TOF MS 분석 결과를 통계처리 가능한 수치로 변환하는 단계는 총 분석시간을 단위시간 간격으로 나누어 단위시간 동안 나타난 크로마토그램 피크의 면적 또는 높이 중 가장 큰 수치를 단위시간 동안의 대표값으로 정하는 것인, 합성가스 발효 균의 포도당 배양과 합성가스 배양을 구별하기 위한 대사체 차별성을 분석하는 방법.
- 제 6 항에 있어서,변환된 수치를 이용하여 통계학적으로 합성가스 발효 균의 포도당 배양과 합성가스 배양을 구별하기 위한 두 생체시료군의 차별성을 검증하는 단계는 부분최소제곱회귀법(Partial least squares discriminant analysis: PLS-DA)을 수행하여 두 생체시료군 간의 유의적인 차이를 나타내는 대사체 바이오마커를 분석 및 검증하는 것인, 합성가스 발효 균의 포도당 배양과 합성가스 배양을 구별하기 위한 대사체 차별성을 분석하는 방법.
- 제 9 항에 있어서,부분최소제곱회귀법(Partial least squares discriminant analysis: PLS-DA)의 로딩 값이 양수인 것은 대사체 바이오마커의 증가 경향을, 로딩 값이 음수인 것은 대사체 바이오마커의 감소 경향을 나타내는 것인, 합성가스 발효 균의 포도당 배양과 합성가스 배양을 구별하기 위한 대사체 차별성을 분석하는 방법.
- 제 9 항에 있어서,대사체 바이오마커는 팔미트산(palmitic acid), 스테아르산(stearic acid), 아라키드산(arachidic acid), 마르가르산(heptadecanoic acid), 1-모노팔미틴(1-monopalmitin), 알라닌(alanine), N-메틸알라닌(N-methylalanine), 아데노신(adenosine), 글리세롤-1-인산(glycerol-1-phosphate) 및 발린(valine)으로 구성된 것인, 합성가스 발효 균의 포도당 배양과 합성가스 배양을 구별하기 위한 대사체 차별성을 분석하는 방법.
