ラクトフェリンは、主に哺乳類動物の乳汁中に存在し、好中球、涙、唾液、鼻汁、胆汁、精液などにも見出されている、分子量約80,000の糖タンパク質である。ラクトフェリンは、鉄を結合することから、トランスフェリンファミリーに属する。ラクトフェリンの生理活性としては、抗菌作用(特許文献1)、鉄代謝調節作用、細胞増殖活性化作用、造血作用、抗炎症作用、抗酸化作用、食作用亢進作用、抗ウイルス作用、ビフィズス菌生育促進作用(特許文献2)、抗ガン作用、ガン転移阻止作用、トランスロケーション阻止作用などが知られている。さらに、ラクトフェリンが脂質代謝改善作用、鎮痛・抗ストレス作用、アンチエイジング作用を有することも明らかにされている。このように、ラクトフェリンは、多様な機能を示す生理活性タンパク質であり、健康の維持、回復又は増進のため、医薬品や食品などの用途に使用されることが期待されており、ラクトフェリンを含む食品は既に市販されている。
また、ラクトフェリンはIn vitroにおいてラクトフェリンやその抗菌ペプチドをガン培養細胞株に添加すると、株により効果は異なるが、増殖抑制やアポトーシスの誘導が報告されている(非特許文献1)。マウス等を用いたin vivoにおいても同様に、肺ガンに対してラクトフェリンは治療効果が報告されている(非特許文献2)。一方、大腸ガンモデルにおいては、原発巣への治療効果は見られないものの、転移抑制能があるとの報告もあり、ガン種によっては、その治療効果は異なると考えられる(非特許文献3)。
この他、ラクトフェリンは抗ガン剤としての臨床開発研究もおこなわれていた。非小細胞性肺ガンに対し、経口溶液製剤として開発が行われ、第2相試験ではパクリタキセルとカルボプラチンによる下痢などの副作用を17%程度弱め、全生存期間を122%程度延ばしたが、(非特許文献4)第3相試験の単独治療試験で、効果が得られなかったことから、その後開発は中止されている(非特許文献5)。ガンモデル動物を用いた場合においても、同様に単独での効果が見られない場合が知られている(非特許文献6)。
ラクトフェリン経口製剤のガン治療のメカニズムとしては、腸管吸収の際に生じる樹状細胞、NK細胞の活性化が報告されている。樹状細胞の活性化はナイーブT細胞からエフェクターT細胞への分化を促進させる。現在、ガン免疫療法としてエフェクターT細胞の活性化に関わるPD―1やCTL4を標的とした抗体医薬が開発・製造されているが、奏効率に対して薬価が非常に高く、甲状腺機能異常、下痢、倦怠感、発熱、肝機能異常、間質性肺疾患、発疹等といった副作用も多く、それらに取って代われる治療薬の開発が求められている。
また、ガン治療において、疼痛は患者のQOLを著しく損なう。通常の治療以外にガン発生初期からモルヒネを少量投与する緩和療法を行うと、全生存期間が延長する報告もあり(非特許文献7)、疼痛を制御することも、ガン治療において重要であると考えられる。ガンの疼痛原因は、神経因子疼痛、骨転移疼痛、消化管閉塞、腹部膨満等がある。それらの疼痛の約80%は、適切な投薬により制御できる。治療薬として、アスピリン等の非オピオイド系、モルヒネ等のオピオイド系が利用されている。また、ガン治療時にも抗ガン剤による、末梢も含めた神経系疼痛が生じることがある。
特に白金製剤は投薬を行うと半数以上が副作用として痛覚過敏やアロディニア(疼痛とは感じない刺激を痛みと感じること。水等の低温又は冷たい物に触ると痛みを感じる)を生じ、非オピオイド系や、症状の悪化により、モルヒネの様な強オピオイド薬の服用を必要とされる。しかし、この様な神経因子疼痛はオピオイド系薬剤が効かない場合もあり、治療薬の開発は必要である。
本発明では、ラクトフェリンの抗ガン効果とガン製剤疼痛を確実にするために、ラクトフェリンを注射剤として開発することに着目した。また、抗ガン効果は、ラクトフェリンでは効果が無く、血中安定化させたラクトフェリンでも低いが、白金製剤と血中安定化させたラクトフェリンを併用することで著しく改善ができ、鎮痛効果も劇的に改善することが見られ、臨床的有効性が高まると期待される。
ラクトフェリンは、坐骨神経絞扼による神経障害性疼痛モデルに対して疼痛緩和効果を有することが報告されている(非特許文献8)。また、ラクトフェリンを有効成分とする神経因性疼痛治療用組成物が公知である(特許文献3)。

本発明は、白金系抗ガン剤組成物と併用するための、該組成物の抗ガン活性を増強し、また該組成物に由来する疼痛を低減する、ラクトフェリン活性を有するタンパク質を含む、抗ガン治療補助剤に関するものである。
抗ガン剤には、白金系製剤、アルキル化剤、代謝拮抗剤、ホルモン剤、分子標的薬などの種類が知られており、ガンの種類に応じて治療方法が選択される。これらのうち、白金系抗ガン剤であるオキサロプラチン(Oxaliplatin(OXL))は大腸ガン治療のキードラッグであるが、急性および慢性の末梢神経障害を高頻度で発現する副作用が知られており、(HIGHLIGHTS OF PRESCRIBING INFORMATION These highlights do not include all the information needed to use ELOXATIN safely and effectively. See full prescribing information for ELOXATIN_FDAより抜粋_http://www.accessdata.fda.gov/drugsatfda_docs/label/2009/021492s011,021759s009lbl.pdf)、この好ましくない副作用の程度によっては、投薬の減量、休薬、中止等の処置を余儀なくされる場合がある(非特許文献:エルプラット(登録商標)点滴静注液 医薬品添付文書)。特にオキサロプラチンの投与により特異的に引き起こされる痛覚過敏や末梢神経障害といった副作用があり、また、既存の末梢神経障害に対する疼痛低減剤であっても症状が緩和しないケースも見られる(趙 萌 京都大学学術情報リポジトリ 2014/3/14)。そのため、白金系抗ガン剤に特有の冷感刺激様の末梢神経障害を緩和する剤の開発が望まれていた。
本発明では、マウスを宿主としたマウス肺ガンのモデルを用いて、ラクトフェリン活性を有するタンパク質と白金系抗ガン剤組成物と併用することによる抗ガン活性を評価したところ、ラクトフェリン活性を有するタンパク質単独投与に比べて、白金系抗ガン剤組成物と併用したほうがより強い抗ガン活性があることが認めた。また、本発明では、オキサロプラチン誘発性神経障害性疼痛モデルマウスを作製し、白金系製剤による疼痛緩和効果の検証を行い、鋭意研究の結果、ラクトフェリン活性を有するタンパク質を含む抗ガン治療補助剤により、白金系抗ガン剤に特有の冷感刺激様の疼痛を緩和する剤が提供されることを見いだした。
即ち、本発明は、以下の抗ガン治療補助剤および方法を提供する。
(1) 白金系抗ガン剤組成物と併用するための、該組成物の抗ガン活性を増強し、また該組成物に由来する疼痛を低減する、ラクトフェリン活性を有するタンパク質を含む、抗ガン治療補助剤。
(2) 前記ラクトフェリン活性を有するタンパク質が、天然型ラクトフェリン、遺伝子組換え型ラクトフェリン、遺伝子組換え型ラクトフェリン融合タンパク質であることを特徴とする、上記(1)に記載の抗ガン治療補助剤。
(3) 前記白金系抗ガン剤組成物に由来する疼痛が、体感温度異常である冷感刺激異常に起因する疼痛であることを特徴とする上記(1)または(2)に記載の抗ガン治療補助剤。
(4) 前記ラクトフェリン活性を有するタンパク質が0.1~1,000mg/kg/日、1~500mg/kg/日、または10~100mg/kg/日の投与量で投与される、上記(1)~(3)のいずれかに記載の抗ガン治療補助剤。
(5) 前記白金系抗ガン剤組成物と前記ラクトフェリン活性を有するタンパク質が、逐次投与、併用投与、または同時投与されることを特徴とする、上記(1)~(4)のいずれかに記載の抗ガン治療補助剤。
(6) 前記ラクトフェリン活性を有するタンパク質が経口または非経口で投与されることを特徴とする、上記(1)~(5)のいずれかに記載の抗ガン治療補助剤。
(7) 前記抗ガン治療補助剤が対象とするガンが、大腸ガン、卵巣ガン、精巣ガン、子宮頸ガン、肺ガン、悪性リンパ腫、乳癌、膀胱ガン、前立腺ガン、食道ガン、胃ガン、頭頸部ガン、小細胞肺ガン、非小細胞肺ガンからなる群から選択される1つである、上記(1)~(6)のいずれかに記載の抗ガン治療補助剤。
(8) 前記白金系抗ガン剤組成物が、シスプラチン、オキサリプラチン、カルボプラチン、ネダプラチンからなる群から選択される少なくとも1つである、上記(1)~(7)のいずれかに記載の抗ガン治療補助剤。
(9) 前記ラクトフェリン活性を有するタンパク質が、オキサリプラチンによる疼痛を緩和することを特徴とする、上記(1)~(8)に記載の抗ガン治療補助剤。
(10) 上記(1)~(9)のいずれかに記載の剤を含む、抗ガン治療補助組成物。

本発明により、白金系抗ガン剤組成物と併用するための、該組成物の抗ガン活性を増強し、また該組成物に由来する疼痛を低減する、ラクトフェリン活性を有するタンパク質を含む、抗ガン治療補助剤が提供される。また、本発明では、抗ガン作用の向上と同時に、白金系抗ガン剤の投与量を抑える効果のある剤を提供できることが示される。
以下、本発明を詳細に説明する。以下の実施の形態は、本発明を説明するための例示であり、本発明をこの実施の形態のみに限定する趣旨ではない。本発明は、その要旨を逸脱しない限り、様々な形態で実施をすることができる。 なお、本明細書において引用した全ての文献、および公開公報、特許公報その他の特許文献は、参照として本明細書に組み込むものとする。また、本明細書は、2016年10月28日に出願された本願優先権主張の基礎となる日本国特許出願(特願2016-212076号)の明細書及び図面に記載の内容を包含する。
本発明は、ラクトフェリン活性を有するタンパク質を含む抗ガン治療補助剤に関する。
ラクトフェリン活性を有するタンパク質
本発明で用いるラクトフェリン活性を有するタンパク質は、ヒト及び種々の動物(ウシ、ウマ、ブ夕、ヒツジ、ヤギ、ラクダ等)から得られる天然のラクトフェリン(天然型ラクトフェリン)、またはこれらの天然のラクトフェリンと同一のアミノ酸配列を有するものでもよいし、目的とする活性を有する限りにおいて、一部のアミノ酸が欠失、付加、置換していてもよい。
本発明で用いるラクトフェリン活性を有するタンパク質は、ヒト及び種々の動物(ウシ、ウマ、ブ夕、ヒツジ、ヤギ、ラクダ等)から得られる天然のラクトフェリン(天然型ラクトフェリン)、またはこれらの天然のラクトフェリンと同一のアミノ酸配列を有するものでもよいし、目的とする活性を有する限りにおいて、一部のアミノ酸が欠失、付加、置換していてもよい。
ここで本発明のラクトフェリン活性とは、例えば天然ラクトフェリンが有する鉄キレート(結合)能を意味する。具体的には、例えば特開2016-117717に記載の測定法で測定した、天然ラクトフェリンの鉄結合能を100%とした場合に、本発明の融合タンパク質等は、少なくとも50%以上(例えば約50%~約150%又は約50%~約120%)の鉄結合能を保持しており、好ましい態様においては、本発明のタンパク質は、天然ラクトフェリンの約70%~約100%又はそれ以上(例えば約70%~約150%又は約70%~約120%)、さらには約90%以上に相当する鉄結合能を有する。なお、鉄結合能は、実施例に記載した方法又はそれと同等の方法によって測定する場合、土2%、土5%、土10%、または、土20%程度の誤差がありうる。
ラクトフェリン活性を有するペプチドは、種々のものが公知であり(例えば、「プログラム・抄録集第2回臨床ラクトフェリンシンポジウム2009」、21~27頁(島崎敬一))等を参照し、必要に応じて設計することができる。変異(欠失、付加、置換)を有するラクトフェリンを用いる場合、配列番号1のアミノ酸配列と90%以上、91%以上、92%以上、93%以上、94%以上、95%以上、96%以上、97%以上、98%以上、99%以上、99.1%以上、99.2%以上、99.3%以上、99.4%以上、99.5%以上、99.6%以上、99.7%以上、99.8%以上、または99.9%以上の配列同一性を有するアミノ酸配列を有するタンパク質が挙げられる。上記配列同一性の数値は一般的に大きい程好ましい。また、ラクトフェリン活性を有するタンパク質としては、配列番号1のアミノ酸配列の10%以上、20%以上、30%以上、40%以上、50%以上、60%以上、70%以上、80%以上、または90%以上を占める配列からなるものを用いることができる。ラクトフェリン活性とは、先に述べた鉄キレート能を有するもの、つまり、少なくとも鉄結合能に基づくラクトフェリンの重要な生物活性が保持されていることを意味する。
本願発明のラクトフェリン活性を有するタンパク質は遺伝子組み換え技術を用いて作製してもよい。さらに、本発明のラクトフェリン活性を有するタンパク質と他の機能を有するタンパク質を融合し、融合タンパク質とすることもできる。
本発明のラクトフェリン活性を有するタンパク質の投与量は、疾患の程度、投与経路、患者の年齢、性別、体重などにより適宜選択されるが、例えば、0.1~1000mg/kg/日、好ましくは、1~500mg/kg/日、より好ましくは10~100mg/kg/日の投与量で逐次投与、併用投与または、同時投与することができる。
本発明のラクトフェリン活性を有するタンパク質は、通常、それ自体公知の薬理学的に許容される担体、賦形剤、希釈剤、増量剤、崩壊剤、安定剤、保存剤、緩衝材、乳化剤、芳香剤、着色剤、甘味料、粘ちょう剤、矯味剤、溶解補助剤、その他の添加剤などと混合して錠剤、丸剤、散剤、顆粒剤、座剤、注射剤、点眼剤、液剤、カプセル剤、トローチ剤、エアゾール剤、懸濁剤、乳剤、シロップ剤などの形態にし、経口的または非経口的に投与することができる。本発明のラクトフェリン活性を有するタンパク質は白金系抗ガン剤と同時または逐次投与、併用投与または、同時投与することができる。また、ラクトフェリン活性を有するタンパク質を同時投与を目的として、抗ガン治療補助組成物とすることもできる。
本発明のラクトフェリン活性を有するタンパク質は、抗菌作用、鉄代謝調節作用、細胞増殖活性化作用、造血作用、抗炎症作用、抗酸化作用、食作用亢進作用、抗ウイルス作用、ビフィズス菌生育促進作用、抗ガン作用、ガン転移阻止作用、トランスロケーション阻止作用、脂質代謝改善作用、鎮痛作用、抗ストレス作用などを含む広範な生理活性を有しており、これらの作用によって、生活習慣病(例えば、高コレステロール血症、高脂血症など)、神経因性疼痛、膠原病(シェーグレン症候群によるドライアイ及びドライマウス、リウマチ性関節炎など)、歯周病、C型肝炎などを含む、多くの疾患又は症状の治療(改善を含む)及び予防が可能である。
抗ガン剤
本発明の抗ガン剤は、一般的なガンに有効なものが含まれ、副腎皮質ガン、肛門ガン、胆管ガン、膀胱ガン、乳ガン、子宮頚ガン、慢性リンパ球性白血病、慢性骨髄性白血病、大腸ガン、子宮内膜ガン、食道ガン、ユーイングガン、胆嚢ガン、ホジキン病、下咽頭ガン、喉頭ガン、口唇口腔ガン、肝臓ガン、非小細胞肺ガン、非ホジキンリンパ腫、黒色腫、中皮腫、多発性骨髄腫、卵巣ガン、膵臓ガン、前立腺ガン、胃ガン、睾丸ガン、甲状腺ガン等のガンに有効な組成物が含まれる。本発明の制ガン剤は少なくとも、大腸ガン、卵巣ガン、精巣ガン、子宮頸ガン、肺ガン、悪性リンパ腫、乳ガン、膀胱ガン、前立腺ガン、食道ガン、胃ガン、頭頸部ガン、小細胞肺ガン、非小細胞肺ガンの治療に好適な薬剤が含まれる。
本発明の抗ガン剤は、一般的なガンに有効なものが含まれ、副腎皮質ガン、肛門ガン、胆管ガン、膀胱ガン、乳ガン、子宮頚ガン、慢性リンパ球性白血病、慢性骨髄性白血病、大腸ガン、子宮内膜ガン、食道ガン、ユーイングガン、胆嚢ガン、ホジキン病、下咽頭ガン、喉頭ガン、口唇口腔ガン、肝臓ガン、非小細胞肺ガン、非ホジキンリンパ腫、黒色腫、中皮腫、多発性骨髄腫、卵巣ガン、膵臓ガン、前立腺ガン、胃ガン、睾丸ガン、甲状腺ガン等のガンに有効な組成物が含まれる。本発明の制ガン剤は少なくとも、大腸ガン、卵巣ガン、精巣ガン、子宮頸ガン、肺ガン、悪性リンパ腫、乳ガン、膀胱ガン、前立腺ガン、食道ガン、胃ガン、頭頸部ガン、小細胞肺ガン、非小細胞肺ガンの治療に好適な薬剤が含まれる。
白金系抗ガン剤組成物
本発明の抗ガン治療補助剤に併用される抗ガン剤組成物としては、アルキル化剤、白金系抗ガン剤、代謝拮抗剤、抗ガン性抗生物質、抗ガン性植物アルカロイド等が挙げられる。これらのうち、白金系抗ガン剤組成物は、白金の錯体からなる抗ガン剤であり、DNAと共有結合して鎖を架橋させ、DNA合成を阻害し、または転写を阻害するものである。白金系抗ガン剤組成物としては、シスプラチン、オキサリプラチン、カルボプラチン、ネダプラチン等を用いることができる。一実施形態において、白金系抗ガン剤組成物はオキサリプラチンである。一実施形態として、白金系抗ガン剤組成物は、パクリタキセル等の他の抗ガン剤と、逐次投与、併用投与または同時投与をする。また一実施形態において、白金系抗ガン剤組成物は、イリノテカンおよび/またはフルオロウラシルと逐次投与、併用投与または同時投与をすることができる。
本発明の抗ガン治療補助剤に併用される抗ガン剤組成物としては、アルキル化剤、白金系抗ガン剤、代謝拮抗剤、抗ガン性抗生物質、抗ガン性植物アルカロイド等が挙げられる。これらのうち、白金系抗ガン剤組成物は、白金の錯体からなる抗ガン剤であり、DNAと共有結合して鎖を架橋させ、DNA合成を阻害し、または転写を阻害するものである。白金系抗ガン剤組成物としては、シスプラチン、オキサリプラチン、カルボプラチン、ネダプラチン等を用いることができる。一実施形態において、白金系抗ガン剤組成物はオキサリプラチンである。一実施形態として、白金系抗ガン剤組成物は、パクリタキセル等の他の抗ガン剤と、逐次投与、併用投与または同時投与をする。また一実施形態において、白金系抗ガン剤組成物は、イリノテカンおよび/またはフルオロウラシルと逐次投与、併用投与または同時投与をすることができる。
治療補助剤
本明細書における「抗ガン治療補助剤」とは、逐次投与、併用投与、または同時投与された抗ガン剤の作用を安定化、増強または持続させるか、および/あるいは前記抗ガン剤の副作用、特に疼痛を減少、消滅または軽減する作用を呈する1つ又はそれ以上の剤を意味する。
本明細書における「抗ガン治療補助剤」とは、逐次投与、併用投与、または同時投与された抗ガン剤の作用を安定化、増強または持続させるか、および/あるいは前記抗ガン剤の副作用、特に疼痛を減少、消滅または軽減する作用を呈する1つ又はそれ以上の剤を意味する。
抗ガン剤組成物に由来する疼痛
抗ガン剤組成物に由来する末梢神経障害としては、刺痛や焼けるような痛み等の疼痛、四肢末端のしびれ、灼熱感等の知覚異常、体感温度異常である冷感刺激異常に起因する疼痛、感覚消失・感覚麻痺や違和感等の感覚異常、知覚性運動失調、筋力の低下等が挙げられる(WO2012/105476)。
白金系抗ガン剤組成物に由来する疼痛としては、手、足や口唇周囲部の感覚異常又は知覚不全(末梢神経に伴う疼痛、体感温度異常である冷感刺激異常に起因する疼痛等が知られており、これらのうち冷感刺激異常に起因する疼痛は、オピオイド系鎮痛剤が効きにくいという特徴が有る。本発明において、ラクトフェリン活性を有するタンパク質は、神経因性疼痛の緩和のみならず、少なくとも白金系抗ガン剤により生じる体感温度異常である冷感刺激異常に起因する疼痛を低減する。
抗ガン剤組成物に由来する末梢神経障害としては、刺痛や焼けるような痛み等の疼痛、四肢末端のしびれ、灼熱感等の知覚異常、体感温度異常である冷感刺激異常に起因する疼痛、感覚消失・感覚麻痺や違和感等の感覚異常、知覚性運動失調、筋力の低下等が挙げられる(WO2012/105476)。
白金系抗ガン剤組成物に由来する疼痛としては、手、足や口唇周囲部の感覚異常又は知覚不全(末梢神経に伴う疼痛、体感温度異常である冷感刺激異常に起因する疼痛等が知られており、これらのうち冷感刺激異常に起因する疼痛は、オピオイド系鎮痛剤が効きにくいという特徴が有る。本発明において、ラクトフェリン活性を有するタンパク質は、神経因性疼痛の緩和のみならず、少なくとも白金系抗ガン剤により生じる体感温度異常である冷感刺激異常に起因する疼痛を低減する。
抗ガン活性
本発明の抗ガン活性は、ガン細胞移植モデルであるXenograftモデル、Syngenicモデルを用いた移植固形ガン細胞の体積や重量の増減、または増殖率、アポトーシスの割合などの指標をもとに測定することができる。本発明において「抗ガン活性を増強する」とは、ガン移植モデルの条件で、ラクトフェリン活性を有するタンパク質と白金系抗ガン剤を投与した際に、固形ガン細胞体積がコントロール又は、対照薬より体積が抑制、減少したことを意味する。
本発明の抗ガン活性は、ガン細胞移植モデルであるXenograftモデル、Syngenicモデルを用いた移植固形ガン細胞の体積や重量の増減、または増殖率、アポトーシスの割合などの指標をもとに測定することができる。本発明において「抗ガン活性を増強する」とは、ガン移植モデルの条件で、ラクトフェリン活性を有するタンパク質と白金系抗ガン剤を投与した際に、固形ガン細胞体積がコントロール又は、対照薬より体積が抑制、減少したことを意味する。
疼痛の低減

オキサリプラチン誘発性神経障害性疼痛に対するヒトラクトフェリン(hLf)の予防効果
オキサリプラチン誘発性神経障害性疼痛に対するhLfの予防効果を評価した、Von-Frey testの結果を図1に示す。マウスの肢に太さの異なるフィラメントをフットパットに押し当て、50%反応閾値を計測した。ここで、Pre 測定の後、hLf 100 mg/kg, ip投与し、その2時間後にオキサリプラチン10 mg/kg, ip(1回のみ投与)を行った。 *P<0.05 vs. OXL+salineである。

マウス肺ガンに対するラクトフェリン類の活性評価試験
6~8週齢のC57BL/6マウス(体重 18~22g)を用いて、マウスのガンであるルイス肺ガンに対する効果を、ヒト組換えラクトフェリン(以下rhLFと記載)およびヒト組換えラクトフェリンとヒトIgGのFc領域との融合体(以下rhLF-Fcと記載)について評価した。
6~8週齢のC57BL/6マウス(体重 18~22g)を用いて、マウスのガンであるルイス肺ガンに対する効果を、ヒト組換えラクトフェリン(以下rhLFと記載)およびヒト組換えラクトフェリンとヒトIgGのFc領域との融合体(以下rhLF-Fcと記載)について評価した。
対数増殖期のルイス肺ガン細胞(3x105個)を、マウスの右下方脇腹の皮下に移植した。ガンのサイズが約76mm3になった時点(ガン移植後5日目)から治療を開始した。実験の概要を以下に示した。グループあたり10匹のマウスを用いた。
薬剤は、リン酸バッファー(pH7)に溶解し、10ml/kgの用量で投与した。ガンの大きさは、週2回長径と短径を測定し計算で求めた。抗ガン効果については、ガン増殖阻害率(TGIと表す)を、TGI=(1‐T/C)x 100%式、(Tは治療グループの平均ガン増殖率%であり、Cは対照グループの平均ガン増殖率%)で計算して求めた。実験中のガン増殖率の変化を図4のグラフに示した。図4に示したように、ガン移植後12日目の観察において、rhLF-Fcは10 mg/kg投与した時、TGIは33.5%であった(対照グループに対するP値:0.01)。一方、rhLFについては、有意な抗ガン活性は認められなかった。
マウス肺ガンに対するrhLF-Fcとシスプラチンの併用による抗ガン活性評価試験

血中サイトカインの測定
実施例2の実験終了時に、グループ1(G1:ビークルコントロール)、グループ2(G2:シスプラチン5 mg/kg)、グループ5(G5:rhLF-Fc 30mg/kg)、およびグループ6(G6:シスプラチン5mg/kg+ rhLF-Fc 30mg/kg)のマウスの血液を回収し、TNFα(図6)IL-2(図7)、IFN-γ(図8)、の測定を行った。その結果、rhLF-Fc単独およびシスプラチンとの併用グループにおいては、対照グループと比較して、IL-2およびIFN-γの値が上昇する傾向が認められた。本実験は、マウスのガンをマウスに移植した同種性の実験であり、rhLF-Fcによる、宿主のIL-2およびIFN-γレベルの上昇による宿主の免疫活性の増強が抗ガン活性として現れたと考えることができる。

Claims (10)
- 白金系抗ガン剤組成物と併用するための、該組成物の抗ガン活性を増強し、また該組成物に由来する疼痛を低減する、ラクトフェリン活性を有するタンパク質を含む、抗ガン治療補助剤。
- 前記ラクトフェリン活性を有するタンパク質が、天然型ラクトフェリン、遺伝子組換え型ラクトフェリン、遺伝子組換え型ラクトフェリン融合タンパク質であることを特徴とする、請求項1に記載の抗ガン治療補助剤。
- 前記白金系抗ガン剤組成物に由来する疼痛が、体感温度異常である冷感刺激異常に起因する疼痛であることを特徴とする請求項1または2に記載の抗ガン治療補助剤。
- 前記ラクトフェリン活性を有するタンパク質が0.1~1,000mg/kg/日、1~500mg/kg/日、または10~100mg/kg/日の投与量で投与される、請求項1~3のいずれかに記載の抗ガン治療補助剤。
- 前記白金系抗ガン剤組成物と前記ラクトフェリン活性を有するタンパク質が、逐次投与、併用投与、または、同時投与されることを特徴とする、請求項1~4のいずれかに記載の抗ガン治療補助剤。
- 前記ラクトフェリン活性を有するタンパク質が経口または非経口で投与されることを特徴とする、請求項1~5のいずれかに記載の抗ガン治療補助剤。
- 前記抗ガン治療補助剤が対象とするガンが、大腸ガン、卵巣ガン、精巣ガン、子宮頸ガン、肺ガン、悪性リンパ腫、乳癌、膀胱ガン、前立腺ガン、食道ガン、胃ガン、頭頸部ガン、小細胞肺ガン、非小細胞肺ガンからなる群から選択される1つである、請求項1~6のいずれかに記載の抗ガン治療補助剤。
- 前記白金系抗ガン剤組成物が、シスプラチン、オキサリプラチン、カルボプラチン、ネダプラチンからなる群から選択される少なくとも1つである、請求項1~7のいずれかに記載の抗ガン治療補助剤。
- 前記ラクトフェリン活性を有するタンパク質が、オキサリプラチンによる疼痛を緩和することを特徴とする、請求項1~8に記載の抗ガン治療補助剤。
- 請求項1~9のいずれかに記載の剤を含む、抗ガン治療補助組成物。
