Abstract

본 발명은 상처 치료용 약학 조성물에 관한 것으로, 보다 상세하게는 벤즈알데히드(benzaldehyde) 유도체를 유효성분으로 포함하는 상처, 특히 창상 치료용 약학 조성물, 상처 개선용 화장료 조성물 또는 상처 개선용 식품 조성물에 관한 것이다. 본 발명에 따른 상기 조성물은 상처, 특히 창상을 입은 피부의 케라틴 생성 세포의 증식 및 이동을 촉진하여 상기 상처를 치료 및/또는 개선하는데 사용될 수 있다.

Description

상처 치료용 약학 조성물
본 발명은 상처 치료용 약학 조성물에 관한 것으로, 보다 상세하게는 벤즈알데히드(benzaldehyde) 유도체에 해당하는 화합물을 유효성분으로 포함하는 상처, 특히 창상의 치료 및/또는 개선용 조성물에 관한 것이다.
피부는 신체를 보호하는 방어막 역할을 수행하고 질환에 대한 신체의 일차 방어선이며, 피부의 가장 외벽에 존재하는 표피(epidermis)가 미생물 침입에 대한 장벽을 제공한다. 따라서 상처, 화상, 찰과상 및 피부에 대한 기타 손상의 치료에서 일차적인 목적은 감염을 예방하기 위한 신속한 봉합(closure) 및 상처 치료이다.
상처 치료는 일반적으로, 염증 (inflammation), 증식(proliferation) 및 재형성(remodelling)의 3가지 단계를 수반하는 복잡한 과정이다. 첫 번째 단계는 지혈(haemostasis)을 달성하기 위한 응고(clotting)와 세균 및 괴사 조직을 파괴하기 위한 호중구의 보충 및 상기 호중구 보충 뒤 대식세포의 보충을 수반한다. 두 번째 단계에서는 내피세포가 상처 부위로 들어가는 혈관신생(angiogenesis)이 발생하고, 이와 동시에 섬유아세포가 상처 부위로 들어가고 육아 조직 (granulation tissue)을 생산하는데 도움을 준다. 상기 육아 조직의 형성은 재상피화(reepithelialisation)가 일어날 수 있도록 하는 역할을 한다. 최종 단계 동안에서는 콜라겐 생산 및 파괴 수준이 균등해지고, 그리고 치료된 상처는 최대 강도를 달성하기 위하여 천천히 변경된다.
상처 치료는 상기 과정 중에서 임의의 한 가지가 시기 적절한 방식으로 기능 하지 않을 때 지연된다. 상기 상처 치료의 지연으로 인하여 만성적인 상처를 유발할 수 있는데, 이러한 상처는 개인에게 중요한 문제일 뿐만 아니라 상기 상처로 인한 치료 및 흉터 제거 등에 있어 비용이 많이 드는 임상적 문제점이 발생할 수 있다. 따라서, 피부 및 표피 조직을 재생시키는 신체의 자연 과정을 가속화시키는 조성물에 대한 필요성이 대두되고 있는 실정이다.
본 발명의 일 목적은 벤즈알데히드(benzaldehyde) 유도체를 유효성분으로 포함하는 상처 치료용 약학 조성물을 제공하는 것이다.
본 발명의 다른 목적은 상기 화합물을 유효성분으로 포함하는 상처 개선용 화장료 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 상기 화합물을 유효성분으로 포함하는 상처 개선용 식품 조성물을 제공하는 것이다.
그러나 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 제한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 당업계에서 통상의 지식을 가진 자에게 명확하게 이해될 수 있을 것이다.
본 발명자들은 상처, 특히 창상을 입은 개체에 벤즈알데히드(benzaldehyde) 유도체를 포함하는 조성물을 투여하는 경우, 상기 개체에서 케라틴 형성 세포의 이동 및 증식을 촉진함으로써 상처를 치료할 수 있음을 확인하여 본 발명을 완성하게 되었다.
단, 본 발명에 있어서 상기 “벤즈알데히드(benzaldehyde)의 유도체”란, 상기 벤즈알데히드 화합물의 작용기를 도입, 치환, 산화, 환원 등에 의해서 모체의 구조와 성질을 대폭적으로 변화시키지 않는 한도에서 변화시킨 화합물을 의미한다.
본 발명의 일 구현 예에서는 벤즈알데히드(benzaldehyde) 유도체를 유효성분으로 포함하는, 상처 치료용 약학 조성물을 제공한다.
바람직하게는, 상기 벤즈알데히드 유도체는 하기 화학식 1로 표시되는 4-하이드록시벤즈알데히드(4-Hydroxybenzaldehyde); 하기 화학식 2로 표시되는 4-하이드록시-3-메톡시벤즈알데히드(4-Hydroxy-3-methoxybenzaldehyde); 하기 화학식 3으로 표시되는 4-메틸벤즈알데히드(4-methylbenzaldehyde); 및 하기 화학식 4로 표시되는 4-메톡시벤즈알데히드(4-methoxybenzaldehyde)으로 이루어진 군 중에서 어느 하나로 표시되는 화합물 일 수 있고, 바람직하게는 하기 화학식 1로 표시된 화합물일 수 있으나, 이에 제한되는 것은 아니다:

본 발명의 다른 구현 예에서는 상기 화학식 1로 표시되는 4-하이드록시벤즈알데히드(4-hydroxybenzaldehyde)를 유효성분으로 포함하는 상처 치료용 약학 조성물을 제공한다.
여기서, 본 발명에 따른 상기 화합물은 Src 단백질의 인산화(phosphorylation) 증가를 유도하여, Src 단백질 하위 세포신호전달체계의 활성화로 인해 케라틴 생성 세포의 증식(proliferation) 및 이동을 촉진하여 상처 치료 효과를 부여할 수 있다.
본 발명에 있어서 상기 “Src 단백질”이란, 티로신 키나아제(Tyrosine kinase) 단백질로 원종양 형성 유전자로, 세포의 형태, 이동, 증식 및 생존을 조절하는 중요한 역할을 한다(Res. Commun 324 (2004) 1155e1164.). 상기 Src 단백질은, 또 다른 티로신 키나아제에 해당하는 Focal adhesion kinase (FAK)과 결합하여 Src/FAK 복합체를 형성하고, 세포 부착과 이동에 관여하는 신호 기전을 활성화시키는 역할을 한다(Cell. Mol. Bioeng 4 (2011) 138e147.).
본 발명에 따른 약학 조성물은 혈소판 유래 성장인자(Platelet-derived growth factor; PDGF)를 더 포함하는 경우에는 케라틴 생성 세포의 이동 및 증식을 더욱 촉진하여 상처 치료 효과를 더욱 상승시킬 수 있다.
여기서, 상기 “혈소판 유래 성장인자(Platelet-derived growth factor; PDGF)”란, 혈청에 주로 존재하며, 주로 간엽계 세포 증식을 촉진한다. 상기 혈소판 유래 성장 인자는 혈소판 내지 그 모세포인 골수거핵구에서 분비되며, 외상을 입은 혈관벽에 접착 응집한 혈소판에서 분비되어 혈관벽을 복구하는 작용을 한다(Mayo Clin. Proc. 81 (9): 1241-57.).
본 발명에 있어서, 상기 상처는 창상일 수 있다. 단, 본 발명에 있어서 상기 “창상(Wound)”이란, 외부의 압력에 의하여 조직의 연속성이 끊기거나, 상기 조직의 일부에 결손이 생기는 것을 의미한다. 상기 창상은 찰과상, 타박상, 열상, 칼날에 의한 절창, 자창, 할창, 총창, 폭창 및 교창 일 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 상기 약학 조성물은 추가로 다른 상처 치료용 약물과 병용 투여할 수 있으며, 이를 통해서 일반적인 상처의 치료에 사용될 수 있다.
본 발명에 있어서, 상기 약학 조성물은 캡슐, 정제, 과립, 주사제, 연고제, 분말 또는 음료 형태임을 특징으로 할 수 있으며, 상기 약학 조성물은 인간을 대상으로 하는 것을 특징으로 할 수 있다.
본 발명의 약학 조성물은 이들로 한정되는 것은 아니지만, 각각 통상의 방법에 따라 산제, 과립제, 캡슐, 정제, 수성 현탁액 등의 경구형 제형, 외용제, 좌제 및 멸균 주사용액의 형태로 제형화하여 사용될 수 있다. 본 발명의 약학 조성물은 약학적으로 허용 가능한 담체를 포함할 수 있다. 상기 약학적으로 허용되는 담체는 경구 투여 시에는 결합제, 활탁제, 붕해제, 부형제, 가용화제, 분산제, 안정화제, 현탁화제, 색소, 향료 등을 사용할 수 있으며, 주사제의 경우에는 완충제, 보존제, 무통화제, 가용화제, 등장제, 안정화제 등을 혼합하여 사용할 수 있으며, 국소투여용의 경우에는 기제, 부형제, 윤활제, 보존제 등을 사용할 수 있다. 본 발명의 약학 조성물의 제형은 상술한 바와 같은 약제학적으로 허용되는 담체와 혼합하여 다양하게 제조될 수 있다. 예를 들어, 경구 투여시에는 정제, 트로키, 캡슐, 엘릭서(elixir), 서스펜션, 시럽, 웨이퍼 등의 형태로 제조할 수 있으며, 주사제의 경우에는 단위 투약 앰플 또는 다수회 투약 형태로 제조할 수 있다. 기타, 용액, 현탁액, 정제, 캡슐, 서방형 제제 등으로 제형할 수 있다.
한편, 제제화에 적합한 담체, 부형제 및 희석제의 예로는, 락토즈, 덱스트로즈, 수크로즈, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말디톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로즈, 폴리비닐피롤리돈, 물, 메틸하이드록시벤조에이트, 프로필하이드록시벤조에이트, 탈크, 마그네슘 스테아레이트 또는 광물유 등이 사용될 수 있다. 또한, 충진제, 항응집제, 윤활제, 습윤제, 향료, 유화제, 방부제 등을 추가로 포함할 수 있다.
본 발명에 따른 약학 조성물의 투여 경로는 이들로 한정되는 것은 아니지만 구강, 정맥내, 근육내, 동맥내, 골수내, 경막내, 심장내, 경피, 피하, 복강내, 비강내, 장관, 국소, 설하 또는 직장이 포함된다. 경구 또는 비경구 투하가 바람직하다. 본원에 사용된 용어 "비경구"는 피하, 피내, 정맥내, 근육내, 관절내, 활액낭내, 흉골내, 경막내, 병소내 및 두개골내 주사 또는 주입기술을 포함한다. 본 발명의 약학 조성물은 또한 직장 투여를 위한 좌제의 형태로 투여될 수 있다.
본 발명의 약학 조성물은 사용된 특정 화합물의 활성, 연령, 체중, 일반적인 건강, 성별, 정식, 투여시간, 투여경로, 배출율, 약물 배합 및 예방 또는 치료될 특정 질환의 중증을 포함한 여러 요인에 따라 다양하게 변할 수 있고, 상기 약학 조성물의 투여량은 환자의 상태, 체중, 질병의 정도, 약무형태, 투여경로 및 기간에 따라 다르지만 당업자에 의해 적절하게 선택될 수 있고, 1일 0.0001 내지 50mg/kg 또는 0.001 내지 50mg/kg으로 투여할 수 있다. 투여는 하루에 한번 투여할 수도 있고, 수회 나누어 투여할 수도 있다. 상기 투여량은 어떠한 면으로든 본 발명의 범위를 한정하는 것은 아니다. 본 발명에 따른 약학 조성물은 환제, 당의정, 캡슐, 액제, 겔, 시럽, 슬러리, 현탁제로 제형될 수 있다.
본 발명의 또 다른 구현 예에서는 벤즈알데히드(benzaldehyde) 유도체를 유효성분으로 포함하는, 상처 개선용 화장료 조성물을 제공한다.
바람직하게는, 상기 벤즈알데히드 유도체는 하기 화학식 1로 표시되는 4-하이드록시벤즈알데히드(4-Hydroxybenzaldehyde); 하기 화학식 2로 표시되는 4-하이드록시-3-메톡시벤즈알데히드(4-Hydroxy-3-methoxybenzaldehyde); 하기 화학식 3으로 표시되는 4-메틸벤즈알데히드(4-methylbenzaldehyde); 및 하기 화학식 4로 표시되는 4-메톡시벤즈알데히드(4-methoxybenzaldehyde)으로 이루어진 군 중에서 어느 하나로 표시되는 화합물 일 수 있고, 바람직하게는 하기 화학식 1로 표시된 화합물일 수 있으나, 이에 제한되는 것은 아니다:

본 발명에 따른 화합물을 유효성분으로 포함하는 화장료 조성물은 화장수, 영양로션, 영양에센스, 마사지 크림, 미용목욕물첨가제, 바디로션, 바디밀크, 배스오일, 베이비오일, 베이비파우더, 샤워겔, 샤워크림, 선스크린로션, 선스크린크림, 선탠크림, 스킨로션, 스킨크림, 자외선차단용 화장품, 크렌징밀크, 탈모제화장용, 페이스 및 바디로션, 페이스 및 바디크림, 피부미백크림, 핸드로션, 헤어로션, 화장용크림, 쟈스민오일, 목욕비누, 물비누, 미용비누, 샴푸, 손세정제(핸드클리너), 약용비누비의료용, 크림비누, 페이셜 워시, 전신 세정제, 두피 세정제, 헤어린스, 화장비누, 치아미백용 겔, 치약 등의 형태로 제조될 수 있다. 이를 위해 본 발명의 조성물은 화장료 조성물의 제조에 통상적으로 사용하는 용매나, 적절한 담체, 부형제 또는 희석제를 더 포함할 수 있다.
본 발명의 화장료 조성물 내에 더 추가될 수 있는 용매의 종류는 특별히 한정하지 않으나, 예를 들어, 물, 식염수, DMSO 또는 이들의 조합을 사용할 수 있고, 담체, 부형제 또는 희석제로는 정제수, 오일, 왁스, 지방산, 지방산 알콜, 지방산 에스테르, 계면활성제, 흡습제(humectant), 증점제, 항산화제, 점도 안정화제, 킬레이팅제, 완충제, 저급 알콜 등이 포함되지만, 이에 제한되는 것은 아니다. 또한, 필요에 따라 미백제, 보습제, 비타민, 자외선 차단제, 향수, 염료, 항생제, 항박테리아제, 항진균제를 포함할 수 있다.
상기 오일은 수소화 식물성유, 피마자유, 면실유, 올리브유, 야자인유, 호호바유, 아보카도유가 이용될 수 있으며, 왁스는 밀랍, 경랍, 카르나우바, 칸델릴라, 몬탄, 세레신, 액체 파라핀, 라놀린이 이용될 수 있다.
상기 지방산은 스테아르산, 리놀레산, 리놀렌산, 올레산이 이용될 수 있고, 지방산 알콜로는 세틸 알콜, 옥틸 도데칸올, 올레일 알콜, 판텐올, 라놀린 알콜, 스테아릴 알콜, 헥사데칸올이 이용될 수 있으며, 상기 지방산 에스테르는 이소프로필 미리스테이트, 이소프로필 팔미테이트, 부틸 스테아레이트가 이용될 수 있다. 또한, 상기 계면 활성제로는 당업계에 알려진 양이온 계면활성제, 음이온 계면활성제 및 비 이온성 계면활성제가 사용가능하며 가능한 한 천연물 유래의 계면활성제가 바람직하다.
그 외에도 화장품 분야에서 널리 알려진 흡습제, 증점제, 항산화제 등을 포함할 수 있으며, 이들의 종류와 양은 당업계에 공지된 바에 따른다.
본 발명의 또 다른 구현예 에서는 벤즈알데히드(benzaldehyde) 유도체를 유효성분으로 포함하는, 상처 개선용 식품 조성물을 제공한다.
바람직하게는, 상기 벤즈알데히드 유도체는 하기 화학식 1로 표시되는 4-하이드록시벤즈알데히드(4-Hydroxybenzaldehyde); 하기 화학식 2로 표시되는 4-하이드록시-3-메톡시벤즈알데히드(4-Hydroxy-3-methoxybenzaldehyde); 하기 화학식 3으로 표시되는 4-메틸벤즈알데히드(4-methylbenzaldehyde); 및 하기 화학식 4로 표시되는 4-메톡시벤즈알데히드(4-methoxybenzaldehyde)으로 이루어진 군 중에서 어느 하나로 표시되는 화합물 일 수 있고, 바람직하게는 하기 화학식 1로 표시된 화합물일 수 있으나, 이에 제한되는 것은 아니다:

본 발명의 상기 화합물을 유효성분으로 포함하는 식품 조성물은 각종 식품류, 예를 들어, 음료, 껌, 차, 비타민 복합제, 분말, 과립, 정제, 캡슐, 과자, 떡, 빵 등의 형태로 제조될 수 있다. 본 발명의 식품 조성물은 독성 및 부작용이 거의 없는 식물추출물로 구성된 것이므로 예방 목적으로 장기간 복용 시에도 안심하고 사용할 수 있다.
본 발명의 상기 화합물이 식품 조성물에 포함될 때 그 양은 전체 중량의 0.1 내지 50%의 비율로 첨가할 수 있다.
여기서, 상기 식품 조성물이 음료 형태로 제조되는 경우 지시된 비율로 상기 식품 조성물을 함유하는 것 외에 특별한 제한점은 없으며 통상의 음료와 같이 여러가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 즉, 천연 탄수화물로서 포도당 등의 모노사카라이드, 과당 등의 디사카라이드, 슈크로스 등의 및 폴리사카라이드, 덱스트린, 시클로덱스트린 등과 같은 통상적인 당 및 자일리톨, 소르비톨, 에리트리톨 등의 당알콜 등을 포함할 수 있다. 상기 향미제로서는 천연 향미제(타우마틴, 스테비아 추출물(예를 들어 레바우디오시드 A, 글리시르히진등) 및 합성 향미제(사카린, 아스파르탐 등) 등을 들 수 있다.
그 외 본 발명의 식품 조성물은 여러 가지 영양제, 비타민, 광물(전해질), 합성 풍미제 및 천연 풍미제 등의 풍미제, 착색제, 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알콜, 탄산 음료에 사용되는 탄산화제 등을 함유할 수 있다.
이러한 성분은 독립적으로 또는 조합하여 사용할 수 있다. 이러한 첨가제의 비율은 본 발명의 핵심적인 요소에 해당하지 아니하지만, 본 발명의 식품 조성물 100 중량부 당 0.1 내지 약 50 중량부의 범위에서 선택되는 것이 일반적이다.
본 발명에 따른 조성물은 상처, 특히 창상을 입은 피부의 케라틴 생성 세포의 증식 및 이동을 촉진하여 상기 상처를 치료하는데 효과적으로 사용될 수 있다.

본 발명의 일 구현 예에서는 4-하이드록시벤즈알데히드(4-Hydroxybenzaldehyde)를 유효성분으로 포함하는 상처 치료용 약학 조성물을 제공한다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로서, 본 발명의 요지에 따라 본 발명의 범위가 이들 실시예에 의해 제한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에게 있어서 자명할 것이다.
실시예
[ 실시예 1] 인간 케라티노사이트 세포주( CRL - 2309)의 배양
본 발명에 따른 벤즈알데히드(benzaldehyde)의 유도체의 상처 치료 촉진 효과 확인을 위하여, 인간 케라티노사이트(Human Keratinocyte) 세포주인 CRL-2309를 계대배양 하였다. 상기 CRL-2309는 ATCC에서 구입하여 사용하였다. 상기 세포주는 우태아혈청(FBS; hyclone, logan, UT) 10%를 포함하는 DMEM(Invitrogen Gibco, Grand Island, NY) 영양배지에서 37℃, 5 % CO2 배양기 내에서 배양을 실시하였다.
[ 실시예 2] 인간 케라티노사이트 세포주( CRL -2309) 이동 능력 확인
상기 실시예 1의 세포주에서 본 발명에 따른 화합물에 의한 세포 이동 촉진 효과를 알아보기 위해, 세포 스크래치 분석을 수행하였다.
세포 스크래치 분석 방법은 상기 실시예 1의 인간 케라티노사이트 세포를 12웰에 분주하고, 세포 분포 비율(confluency)이 100%가 될 때까지 배양한 뒤에 24시간 동안 serum이 없는 상태에 노출 시켰다. 상기 세포에 음성 대조군 DMSO, 양성 대조군 혈소판 유래 성장인자(Platelet-derived growth factor; PDGF) 5μM, 실험군 으로 4-하이드록시벤즈알데히드(4-HBA) 1μM 단독 및 혈소판 유래 성장인자와 4-하이드록시벤즈알데히드(combine)를 혼합하여 처리하였다. 각각의 군에 상기 시약을 처리한 뒤 멸균된 파이펫 팁을 이용하여 물리적인 스크래치를 형성한뒤, 24시간 동안 12시간 간격으로 상기 세포의 스크래치 면적을 현미경을 이용하여 촬영하여, 그 결과를 도 1 및 2에 나타내었다.
도 1 및 2에서 보는 바와 같이, 양성 대조군인 PDGF에서 세포 이동이 촉진되는 조건하에서 4-하이드록시벤즈알데히드(4-HBA)를 처리한 군 역시 음성대조군(DMSO)에 비하여 세포 이동이 속도가 20% 증가된 것을 확인할 수 있었다. 또한, 혈소판 유래 성장인자 및 4-하이드록시벤즈알데히드(combine)를 혼합하여 처리한 군에서는 상기 4-HBA 군에 비하여 세포 이동 속도가 13% 증가되어 있는 것을 확인할 수 있었다.
[ 실시예 3] 인간 케라티노사이트 세포주( CRL -2309) 침투 능력 확인
상기 실시예 1의 세포주에서 본 발명에 따른 화합물에 의한 세포 이동 촉진 효과를 알아보기 위하여, 마트리겔 침투 챔버를 이용한 침투 분석(invasion assay)을 수행하였다.
마트리겔 침투 분석(matrigel invasion assay)은 Biocoat matrigel invasion chamber (Corining, 354480)를 이용하여 상기 제품의 제조사에서 제공하는 가이드라인에 따라 실험을 진행하였다. 키모 어트렉턴트 웰에 상기 실시예 1의 세포를 분주하고, 24시간 동안 무혈청 상태에 노출 시켰다. 그 뒤, 상기 실시예 2와 같은 조건으로 DMSO, 혈소판 유래 성장인자(PDGF), 4-하이드록시벤즈알데히드(4-HBA) 단독, 및 혈소판 유래 성장인자와 4-하이드록시벤즈알데히드(combine) 혼합한 화합물을 각각 처리하고 상기 세포를 24시간 배양한 뒤, 마트리겔 막 위쪽의 세포를 면섬유를 이용하여 닦아내고, 상기 마트리겔을 4%의 파라포르말린으로 고정시킨 후 0.2% 크리스탈 바이올렛으로 염색하여 사진 촬영하여, 그 결과를 도 3 및 4에 나타내었다.
도 3 및 4에서 보는 바와 같이, 음성 대조군(DMSO)에 비하여 4-하이드록시벤즈알데히드(4-HBA)에서 침투한 세포의 수가 2.5배 증가하였다. 또한, 혈소판 유래 성장인자와 4-하이드록시벤즈알데히드(combine)를 혼합하여 처리한 군에서 음성 대조군에 비해 침투한 세포의 수가 3배 증가한 것을 확인할 수 있었다.
상기 결과를 통하여 세포수준에서 4-하이드록시벤즈알데히드(4-HBA)가 케라틴 형성 세포(CRL-2309)의 이동이 촉진을 유도하는 것을 알 수 있다. 또한, 혈소판 유래 성장인자 및 4-하이드록시벤즈알데히드를 혼합하여 처리하는 경우에 케라틴 형성 세포의 이동이 더욱 촉진되는 것을 알 수 있다.
[ 실시예 4] 인간 케라티노사이트 세포주( CRL -2309) 세포신호전달
본 발명의 상기 실시예 2 및 3의 세포 이동 촉진 효과가 어떤 세포신호전달 체계에 의해 유도되는지 확인하기 위하여, 단백질 발현 및 mRNA 발현 수준을 웨스턴 블롯 분석(Western blot assay) 및 실시간 중합효소 연쇄반응(real-time PCR)을 각각 수행하였다.
상기 분석을 위하여 상기 실시예 2와 같은 조건으로 DMSO 및 4-하이드록시벤즈알데히드(4-HBA)를 처리하고 90분 동안 배양한 세포주를 이용하였다.
웨스턴 블롯 분석을 위한 단백질 추출은, 상기 세포주 각각을 수득(harvest)하고, 상기 수득된 세포주를 PBS로 세척 후 단백질 분해 억제제가 포함되어 있는 세포 용해 버퍼(cell lysis buffer)를 이용하여 20분 동안 얼음에서 보관하여 용해시켰다. 상기 용해된 용액을 원심분리기를 이용하여 13000rpm에서 전체 용액을 분획한 뒤, 단백질이 포함된 상등액만을 추출하였다. 상기 추출된 단백질은 브래드포드(Bradford) 방법에 의해 정량화 하였다. 정량화를 통해 동일 농도의 단백질을 8% SDS-폴리아크릴아미드(polyacrylamide) 겔 전기영동을 수행하여 분리한 뒤, PVDF(Millipore, MA, USA) 막에 옮겼다. 단백질이 옮겨진 막은 비 특이적 결합을 줄이기 위하여 5% 탈지유(non-fat milk)가 포함된 TBS-T(Tris-buffered saline/0.05% Tween-20)용액으로 상온에서 1시간 동안 차단(blocking)한 후, 1차 항체 4℃ 조건에서 24시간 동안 반응시킨 뒤, 2차 항체를 2시간 동안 상온에서 반응시켜 시각화 하였다.
웨스턴 블롯 분석은 p-FAK, p-Src, Src, p-Akt, Akt, p-Erk, Erk, (Cell signaling) 및 단백질 발현의 정량적 비교를 위한 항존 유전자(housekeeping gene)인 β-actin 마커를 이용하여 수행하였고, 그 결과는 도 5에 나타내었다.
또한, 상기 실시간 중합효소 연쇄반응(real-time PCR)을 위한 mRNA 추출은, 상기 세포주 각각에 TRizol을 처리하고, 통상의 방법에 의해 mRNA를 수득하였다. 상기 mRNA는 나노드롭(Nano-drop)을 이용하여 정량화 한 뒤, 1μg의 mRNA를 이용하여 역전사 효소 중합반응에 의해 cDNA를 합성하였다. 상기 합성된 cDNA는 Human eNOS, TNFα, Keratin 6 및 Keratin 17 프라이머를 이용하여 95에서 15분간 변성 후 95(30s), 60℃(30s)에서 40사이클의 조건하에서 실시간 중합효소 연쇄반응을 수행하여, 그 결과를 도 6에 나타내었다.
도 5에서 보는 바와 같이, 케라틴 형성 세포(CRL-2309)의 증식과 이동에 관여하는 대표적 인자인 Src의 인산화가 음성대조군(DMSO)에 비하여 4-하이드록시벤즈알데히드를 처리한 군에서 증가되어 있는 것을 확인하였다.
또한, 도 6에서 보는 바와 같이, Src의 하위 활성화 인자로 알려져 있는 eNOS뿐만 아니라, 상피화를 통한 창상 치료에 중요한 역할을 한다고 알려져 있는 TNFα, Keratin 6 및 Keratin 17의 mRNA 발현이 음성대조군에 비하여 4-하이드록시벤즈알데히드를 처리한 군에서 증가되어 있는 것을 확인하였다.
상기 결과를 통하여 4-하이드록시벤즈알데히드에 의한 상처 치료 효과는 Src 인산화로 인해 하위 세포신호전달의 활성을 통해 유도되는 것을 알 수 있다.
[ 실시예 5] 상처 유도 마우스 모델에서 상처 치료 효과
4주된 C57BL/6 수컷 마우스를 마취상태에서 등 부위에 가로, 세로 각각 1cm 상처를 유발한 후 0,3,6 및 9일째 되는 때 대조군 CMC 겔(gel) 5%, 4-하이드록시벤즈알데히드(4-HBA) 0.7mM, 혈소판 유래 성장인자(PDGF) 3.3mM 및 혈소판 유래 성장인자(PDGF) 및 4-하이드록시벤즈알데히드를 혼합(combine)하여 상처 부위에 도포하였다. 상기 실험 마우스의 상처 부위를 0,3,6 및 9일 째 같은 높이에 설치한 카메라를 이용하여 상처 부위를 촬영하여 상처 부위가 닫히는 정도를 측정하였고, 상기 실험군별로 8개의 조직 슬라이드를 준비하여 H&E 염색을 한 뒤, 그 결과를 도 7 및 8에 나타내었다.
또한, 상기 마우스의 분리된 조직에 상피화 과정이 진행되고 있는 세포의 마커로 알려져 있는 keratin 17 항체를 이용하여 통상의 방법에 의해 형광면역염색을 수행한 뒤, 그 결과를 도 9에 나타내었다.
도 7에서 보는 바와 같이, 대조군인 CMC 겔만 도포한 군에 비하여 혈소판 유래 성장인자(PDGF)를 도포한 군에서 8.1% 더 빠르게 상처가 닫히는 것을 확인 하였다. 또한, 상기 혈소판 유래 성장인자와 동일한 조건 하에서 상기 대조군에 비해 4-하이드록시벤즈알데히드(4-HBA)를 도포한 군에서 13% 더 빠르게 닫히는 것을 확인할 수 있었으며, 혈소판 유래 성장인자(PDGF) 및 4-하이드록시벤즈알데히드를 혼합하여 도포한 군 (Combine)에서는 대조군에 비하여 19% 더 빠르게 상처가 닫히는 것을 확인할 수 있었다.
도 8 및 도 9에서 보는 바와 같이, 대조군인 CMC 겔만 도포한 군에 비하여 혈소판 유래 성장인자(PDGF)를 도포한 군 및 4-하이드록시벤즈알데히드(4-HBA)를 도포한 군에서 케라틴 층이 더 빠르게 이동한 것을 확인할 수 있었으며, 혈소판 유래 성장인자(PDGF) 및 4-하이드록시벤즈알데히드를 혼합하여 도포한 군(Combine)에서는 상기 케라틴 층의 이동이 각각 도포한 군에 비하여 더 빠르게 이동하는 것을 확인할 수 있었다.
상기 결과를 통하여 4-하이드록시벤즈알데히드(4-HBA) 단독 및, 혈소판 유래 성장인자(PDGF) 및 4-하이드록시벤즈알데히드를 혼합하여 도포하는 경우 케라틴 층의 이동을 촉진함으로 인해 상처 치료 효과를 나타내는 것을 알 수 있다.
이상에서 본 발명에 대하여 상세하게 설명하였지만 본 발명의 권리범위는 이에 한정되는 것은 아니고, 청구범위에 기재된 본 발명의 기술적 사상을 벗어나지 않는 범위 내에서 다양한 수정 및 변형이 가능하다는 것은 당 기술분야의 통상의 지식을 가진 자에게는 자명할 것이다.
본 발명은 상처 치료용 약학 조성물에 관한 것으로, 보다 상세하게는 벤즈알데히드(benzaldehyde) 유도체를 유효성분으로 포함하는 상처, 특히 창상 치료용 약학 조성물, 상처 개선용 화장료 조성물 또는 상처 개선용 식품 조성물에 관한 것으로, 상처, 특히 창상을 입은 피부의 케라틴 생성 세포의 증식 및 이동을 촉진하여 상기 상처를 치료 및/또는 개선하는 분야에 널리 활용될 수 있다.

  1. 하기 화학식 1 내지 4 중 어느 하나로 표시되는 화합물을 유효성분으로 포함하는 상처 치료용 약학 조성물:
