Description
本発明は、ホスファローダミン化合物若しくはその塩、並びにそれを用いた蛍光色素に関する。
高い光安定性を有する近赤外蛍光色素の開発は、生命科学研究のみならず、医療分野においても大変興味を持たれている。実際、近赤外蛍光色素の代表格であるインドシアニングリーン(ICG)を用いた外科手術は、近年比較的増えてきており、今後も増え続けることが期待される。しかしながら、ICGは光安定性に乏しく、用途も限定的である。
近赤外蛍光色素は、現在はこのように光安定性に乏しく、用途も限定的であるが、近赤外に強い蛍光を示す化合物の開発は、生体深部の非浸襲イメージングの観点から非常に重要な研究課題である。また、植物類のイメージングにおいても、葉緑体の自家蛍光の波長領域を超える長波長領域に蛍光を有する化合物は極めて有用である。
このような近赤外蛍光色素としては、例えば、特許文献1及び非特許文献1に開示されている。特許文献1で示されている近赤外蛍光色素は、キサンテン骨格の9位の位置に有するリン原子上にP(O-)=O構造を有するホスファローダミン化合物である。また、非特許文献1で示されている近赤外蛍光色素は、キサンテン骨格の9位の位置に有するリン原子上にP(CH3)=O構造を有するホスファローダミン化合物である。このような従来の近赤外蛍光色素は、蛍光極大波長は710nm程度に過ぎなかった。この場合、生体深部透過性の観点からは十分とは言えず、また、葉緑体の自家蛍光も無視できない範囲である。この蛍光極大波長を数十nm長波長化すれば、生体深部透過性をより高めることができるのみならず、葉緑体の自家蛍光の影響を完全に無視できる領域であるため非常に重要である。
上記した特許文献1には、ホスファローダミン骨格の共役系をさらに拡張して左右に芳香環を形成した化合物として、
で表される化合物の蛍光極大波長が732nmであることが記載されている。しかしながら、合成の煩雑さ、水溶性、イメージング時の非特異吸着の起こりやすさ等の観点からは、ホスファローダミン骨格自体を改変することで蛍光極大波長を長波長化できる手法が望まれている。

本発明は、上記のような課題を解決するためになされたものであり、ホスファローダミン骨格の共役系を拡張せずとも、従来の近赤外蛍光色素よりも蛍光極大波長をさらに長波長化することができる化合物を提供することを目的とする。
上記目的を鑑み、鋭意検討した結果、本発明者らは、ホスファローダミン骨格が有するリン原子上にアリール基を結合した構造を有する化合物が、従来の近赤外蛍光色素よりも蛍光極大波長をさらに長波長化することができることを見出した。ホスファローダミン骨格のπ共役を拡張した場合は、蛍光極大波長をさらに長波長化させることが可能である。本発明は、このような知見に基づきさらに研究を重ね、完成させたものである。すなわち、本発明は以下の構成を包含する。

本発明のホスファローダミン化合物は、リン原子上にアリール基を結合した構造を有しているため、ホスファローダミン骨格を拡張せずとも、従来の近赤外蛍光色素よりも蛍光極大波長をさらに長波長化することができる。さらに、ホスファローダミン骨格のπ共役を拡張することにより、蛍光極大波長をさらに長波長化することも可能である。
本発明のホスファローダミン化合物は、R8で示される基の種類によっては、様々なpHにおいて長時間にわたって吸収強度及び蛍光強度を維持することができるため、安定性を特に向上させることもできる。
さらに、本発明のホスファローダミン化合物は、R8で示される基をアミン反応性基又はチオール反応性基とする場合には、タンパク質を標識するタンパク質標識剤として機能することもできる。この場合、市販されているタンパク質標識剤と比較しても、より長時間にわたって蛍光を維持することが可能である。
本明細書において、「(ヘテロ)アリール基」は、アリール基又はヘテロアリール基を意味する。また、本明細書において、「含有」は、「含む(comprise)」、「実質的にのみからなる(consist essentially of)」、及び「のみからなる(consist of)」のいずれも包含する概念である。また、本明細書において、数値範囲を「A~B」で示す場合、A以上B以下を意味する。

上記一般式(1)において、Xは対イオン(アニオン)を示す。対イオン(アニオン)としては、例えば、ハロゲンイオン(フッ素イオン、塩素イオン、臭素イオン、ヨウ素イオン等)、シアンイオン、酢酸イオン、トリフルオロ酢酸イオン等が挙げられる。
上記一般式(1)において、R1~R4で示されるアルキル基としては、直鎖アルキル基及び分岐鎖アルキル基のいずれも採用できる。
直鎖アルキル基としては、炭素数1~6(特に1~4)の直鎖アルキル基が好ましく、具体的には、メチル基、エチル基、n-プロピル基、n-ブチル基、n-ペンチル基、n-ヘキシル基等が挙げられる。
分岐鎖アルキル基としては、炭素数3~6(特に3~5)の分岐鎖アルキル基が好ましく、具体的には、イソプロピル基、イソブチル基、tert-ブチル基、sec-ブチル基、ネオペンチル基、イソヘキシル基、3-メチルペンチル基等が挙げられる。
R1~R4で示されるアルキル基は、置換基を有していてもよい。アルキル基が有していてもよい置換基としては、特に制限はなく、水酸基、ハロゲン原子(フッ素原子、塩素原子、臭素原子、ヨウ素原子等)、後述のアリール基、後述のヘテロアリール基、後述の反応性基等が挙げられる。置換基を有する場合の置換基の数は、特に制限されず、1~6個が好ましく、1~3個がより好ましい。
上記一般式(1)において、R1~R4で示されるアリール基としては、単環アリール基、縮環アリール基及び多環アリール基のいずれも採用でき、例えば、フェニル基、ナフチル基、アントラセニル基、フェナントレニル基、ビフェニル基、ターフェニル基、フルオレニル基、ピレニル基、トリフェニレニル基等のC6-18アリール基(特にC6-14アリール基)が挙げられる。
R1~R4で示されるアリール基は、置換基を有していてもよい。置換基としては、例えば、特に制限はなく、水酸基、ハロゲン原子(フッ素原子、塩素原子、臭素原子、ヨウ素原子等)、上記アルキル基、上記アリール基、後述のヘテロアリール基、後述の反応性基等が挙げられる。置換基を有する場合の置換基の数は、特に制限されず、1~6個が好ましく、1~3個がより好ましい。
上記一般式(1)において、R1~R4で示されるヘテロアリール基としては、単環ヘテロアリール基及び多環ヘテロアリール基のいずれも採用でき、例えば、ピロリジル基、ピロリル基、テトラヒドロチエニル基、チエニル基、オキソラニル基、フラニル基、イミダゾリル基、ピラゾリル基、チアゾリル基、オキサゾリル基、ピペリジル基、ピリジル基、ピラジル基、インドリル基、イソインドリル基、ベンゾイミダゾリル基、キノリル基、イソキノリル基、キノキサリル基等が挙げられる。
R1~R4で示されるヘテロアリール基は、置換基を有していてもよい。置換基としては、例えば、特に制限はなく、水酸基、ハロゲン原子(フッ素原子、塩素原子、臭素原子、ヨウ素原子等)、上記アルキル基、上記アリール基、上記ヘテロアリール基、後述の反応性基等が挙げられる。置換基を有する場合の置換基の数は、特に制限されず、1~6個が好ましく、1~3個がより好ましい。
なかでも、R1~R4としては、置換若しくは非置換単環アルキル基、並びに置換若しくは非置換(ヘテロ)アリール基が好ましく、置換若しくは非置換アルキル基がより好ましい。
上記一般式(1)において、R5で示されるアリール基としては、上記したものを採用できる。置換基の種類及び数も同様である。これにより、蛍光極大波長を長波長化することができ、且つ、細胞膜透過性に優れるために生体深部の非浸襲イメージングがしやすいとともに、葉緑体の自家蛍光を低減した領域で蛍光させることが可能であることから植物類のイメージングもしやすい。
上記一般式(1)において、R6及びR7は水素原子である。R1とR6、及び/又はR3とR7は、互いに結合して置換若しくは非置換アルキレン基を構成してもよい。このようなアルキレン基としては、炭素数1~6(特に炭素数2~4)のアルキレン基が挙げられ、例えば、メチレン基、エチレン基、トリメチレン基、テトラメチレン基等が挙げられる。アルキレン基は、置換基を有していてもよい。置換基としては、例えば、特に制限はなく、水酸基、ハロゲン原子(フッ素原子、塩素原子、臭素原子、ヨウ素原子等)、上記アリール基、上記ヘテロアリール基、後述の反応性基等が挙げられる。置換基を有する場合の置換基の数は、特に制限されず、1~6個が好ましく、1~3個がより好ましい。なお、水溶性をより向上させ、イメージングの際の非特異吸着をより抑制する観点からは、R1とR6、及びR3とR7のいずれも、互いに結合して置換若しくは非置換アルキレン基を構成せず、R6及びR7が水素原子であることが好ましく、蛍光極大波長をより長波長化させる観点からは、R1とR6、及び/又はR3とR7は、互いに結合して置換若しくは非置換アルキレン基を構成することが好ましい。
上記一般式(1)において、R8で示されるアリール基としては、上記したものを採用できる。置換基の種類及び数も同様である。なかでも、R8としては、蛍光極大波長をより長波長化することができ、様々なpHにおいてより長時間にわたって吸収強度及び蛍光強度を維持することができるとともに、耐光性にもより優れる観点から、一般式(2):

なかでも、R9及びR10としては、蛍光極大波長をより長波長化することができ、且つ、様々なpHにおいてより長時間にわたって吸収強度及び蛍光強度を維持することができる観点から、いずれも置換若しくは非置換アルキル基、又は置換若しくは非置換アルコキシ基であることが好ましい。また、R9及びR10としては、蛍光極大波長をより長波長化することができ、且つ、様々なpHにおいてより長時間にわたって吸収強度及び蛍光強度を維持することができる観点から、置換若しくは非置換アルキル基、又は置換若しくは非置換アルコキシ基が好ましく、置換若しくは非置換アルコキシ基がより好ましい。特に、R9及びR10がいずれも置換若しくは非置換アルキル基、又は置換若しくは非置換アルコキシ基である場合には、酸性条件、中性条件、アルカリ性条件のいずれにおいても、より長時間にわたって吸収強度及び蛍光強度を維持することができる。R9及びR10の少なくとも1つが水素原子である場合には、アルカリ性条件においては吸収強度及び蛍光強度が時間の経過とともに低減しやすいことと比較して優れた効果を有する。
一般式(2)において、R11で示される反応性基としては、反応性を有する基であれば特に制限されない。R11がカルボキシ基;水酸基;アミノ基;クロロメチル基等のハロゲン化アルキル基;イソシアン基;イソチアシアン基等であるホスファローダミン化合物又はその塩は、当該反応性基と反応させることにより、R11を容易に、タンパク質を標識するための基(アミン反応性基、チオール反応性基等)とすることが可能である。このR11がタンパク質を標識するための基(アミン反応性基、チオール反応性基等)である化合物群も、本発明のホスファローダミン化合物又はその塩の範疇である。
上記アミン反応性基は、標識対象となる化合物が有する置換若しくは非置換アミノ基と反応性を有する基であり、タンパク質(特に抗体)が有する置換若しくは非置換アミノ基と反応することにより、本発明のホスファローダミン化合物又はその塩がタンパク質標識剤(特に抗体標識剤)として機能することができる。
また、上記チオール反応性基は、標識対象となる化合物が有する置換若しくは非置換チオール基と反応性を有する基であり、タンパク質(特に抗体)が有する置換若しくは非置換チオール基と反応することにより、本発明のホスファローダミン化合物又はその塩がタンパク質標識剤(特に抗体標識剤)として機能することができる。この場合、長時間にわたって対象となるタンパク質(特に抗体)を標識する(蛍光させる)ことが可能である。特に、R9及びR10がいずれも置換若しくは非置換アルコキシ基である場合には、水溶性に優れつつも、特に、長時間にわたって対象となるタンパク質(特に抗体)を標識する(蛍光させる)ことが可能である。
このようなアミン反応性基又はチオール反応性基としては、例えば、一般式(3A)~(3E):

本発明のホスファローダミン化合物は、塩の状態で存在することもできる。このような塩としては、塩基付加塩として、例えばナトリウム塩、カリウム塩、カルシウム塩、マグネシウム塩等の金属塩;アンモニウム塩;トリエチルアミン塩等の有機アミン塩等を挙げることができ、酸付加塩として、例えば、塩酸塩、硫酸塩、硝酸塩等の鉱酸塩;p-トルエンスルホン酸塩、メタンスルホン酸塩、マレイン酸塩、シュウ酸塩等の有機酸塩等を挙げることができる。これらのほか、グリシン等のアミノ酸との塩を形成する場合もある。
また、本発明のホスファローダミン化合物は、水和物又は溶媒和物として存在する場合もあるが、これらの物質はいずれも本発明の範囲に包含される。
このような本発明のホスファローダミン化合物又はその塩は、吸収極大波長を680~730nm、特に690~720nmとすることができ、蛍光極大波長を720~750nm、特に730~740nmとすることができる。つまり、ローダミン骨格の共役系を拡張することなく、従来の蛍光色素よりも蛍光極大波長をより長波長化することが可能である。また、従来の蛍光色素と比較しても、様々なpH条件において、より長時間にわたって、吸収強度及び蛍光強度を維持することができる。また、本発明のホスファローダミン化合物又はその塩は、一般式(1)におけるR5が置換若しくは非置換アリール基であることにより、細胞膜透過性に優れているため、生体深部の非浸襲イメージングがしやすく、上記のとおり、R8を選択することにより、生体内のタンパク質(特に抗体)を好適に標識する(蛍光させる)ことができる。また、本発明のホスファローダミン化合物又はその塩は、上記のとおり、蛍光極大波長をより長波長化していることから、葉緑体の自家蛍光をより低減した領域で蛍光させることが可能であることから植物類のイメージングもしやすい。
2.ホスファローダミン化合物又はその塩の製造方法
本発明のホスファローダミン化合物又はその塩は、特に制限されず、例えば、R6及びR7が水素原子であるホスファローダミン化合物(1)は、以下の反応式1:

反応式1及び2において、X1及びX2で示されるハロゲン原子は、フッ素原子、塩素原子、臭素原子及びヨウ素原子をいずれも採用できるが、環化反応の進行のしやすさの観点から、塩素原子、臭素原子、ヨウ素原子等が好ましく、臭素原子がより好ましい。
(2-1)環化反応(化合物(3)→化合物(4))
本工程では、例えば、有機溶媒中で、上記反応式1及び2における化合物(3)と、有機リチウム化合物とを反応させた後に、有機リン化合物を添加し、次いで、過酸化水素を添加することにより、所望の環化反応を進行させることができる。

なお、上記反応式1及び2における化合物(3)は、公知又は市販の化合物を用いることもでき、合成することもできる。合成する場合は、既報にしたがって合成することができる。
有機リチウム化合物としては、特に制限はなく、公知のものが採用でき、例えば、エチルリチウム、n-プロピルリチウム、イソプロピルリチウム、n-ブチルリチウム、sec-ブチルリチウム、tert-ブチルリチウム、n-ペンチルリチウム、n-ヘキシルリチウム等のアルキルリチウム;シクロヘキシルリチウム等のシクロアルキルリチウム;フェニルリチウム等のアリールリチウム等が挙げられる。これらのうち、本工程では、収率、合成の容易さの観点から、アルキルリチウムが好ましく、sec-ブチルリチウムがより好ましい。これら有機リチウム化合物は、単独で用いることもでき、2種以上を組合せて用いることもできる。
有機リン化合物としては、本工程で化合物(4)が得られれば特に制限はなく、公知のものが採用できる。ただし、R5として置換若しくは非置換アリール基を導入することから、ジハロゲン化アリールホスフィンを使用することが好ましく、例えば、ジクロロフェニルホスフィン、ジブロモフェニルホスフィン等が挙げられる。これら有機リン化合物は、単独で用いることもでき、2種以上を組合せて用いることもできる。
上記有機リチウム化合物、有機リン化合物及び過酸化水素の使用量は、特に制限はなく、収率、合成の容易さ等の観点から、化合物(3)1モルに対して、有機リチウム化合物を1.0~10.0モル(特に1.5~5.0モル)、有機リン化合物を0.2~3.0モル(特に0.5~2.0モル)使用することが好ましい。また、過酸化水素は、水溶液である過酸化水素水を過剰量用いることが好ましい。
本工程において使用され得る有機溶媒としては、公知のものを採用することができ、本工程では、例えば、テトラヒドロフラン、ジオキサン等の環状エーテル化合物が好ましい。また、反応条件は、反応が十分に進行する程度が好ましく、例えば、反応雰囲気は不活性ガス雰囲気(窒素ガス雰囲気、アルゴンガス雰囲気等)が好ましく、反応温度は-100~100℃、特に-50~50℃が好ましく、反応時間は5分~12時間、特に10分~6時間が好ましい。
反応終了後は、必要に応じて常法にしたがって精製処理をすることもできる。また、精製処理を施さずに次の工程を行うこともできる。
(2-2)酸化反応(化合物(4)→化合物(5))
本工程では、例えば、有機溶媒中で、上記反応式1及び2における化合物(4)に対して、塩基性触媒及び相間移動触媒の存在下に酸化反応を引き起こすことができる。

塩基性触媒としては、特に制限はなく、例えば、ナトリウムメトキシド、ナトリウムエトキシド、カリウムtert-ブトキシド等のアルカリ金属アルコキシド;水酸化リチウム、水酸化ナトリウム、水酸化カリウム、水酸化セシウム等のアルカリ金属水酸化物;炭酸リチウム、炭酸ナトリウム、炭酸カリウム、炭酸セシウム等のアルカリ金属炭酸塩等が挙げられ、収率、合成の容易さ等の観点から、アルカリ金属水酸化物及びアルカリ金属炭酸塩が好ましく、水酸化ナトリウム及び炭酸カリウムがより好ましい。特に、炭酸カリウムを用いて1回目の酸化反応を起こした後に、残存する原料に対して、水酸化ナトリウムを用いて2回目の酸化反応を起こすことが好ましい。
相間移動触媒としては、特に制限はなく、例えば、テトラメチルアンモニウムクロリド、テトラメチルアンモニウムブロミド、テトラメチルアンモニウムヨージド、テトラメチルアンモニウムヒドロキシド、テトラエチルアンモニウムクロリド、テトラエチルアンモニウムブロミド、テトラエチルアンモニウムヒドロキシド、テトラプロピルアンモニウムブロミド、テトラブチルアンモニウムクロリド、テトラブチルアンモニウムブロミド(TBAB)、テトラブチルアンモニウムヨージド、テトラブチルアンモニウムヒドロキシド等の4級アンモニウム塩;メチルトリブチルホスホニウムヨージド、テトラブチルホスホニウムクロリド、テトラブチルホスホニウムブロミド等の4級ホスホニウム塩;ジベンゾ18クラウン6、ベンゾ15クラウン5、18クラウン6、ジアザ18クラウン6、21クラウン7等のクラウンエーテル等が挙げられ、収率、合成の容易さ等の観点から、4級アンモニウム塩が好ましく、テトラブチルアンモニウムブロミド(TBAB)がより好ましい。
上記塩基性触媒及び相間移動触媒の使用量は、特に制限はなく、収率、合成の容易さ等の観点から、化合物(4)1モルに対して、塩基性触媒を0.5~10.0モル(特に1.0~5.0モル)、相間移動触媒を0.01~0.10モル(特に0.02~0.05モル)使用することが好ましい。複数回酸化反応させる場合は、各酸化反応ごとに、上記使用量を使用することが好ましい。
酸化反応は、酸化剤を使用することもできるし、酸化剤を使用しないこともできる。
酸化剤を使用する場合、特に制限はなく、過マンガン酸塩(過マンガン酸カリウム等)等を使用することができる。この場合、上記酸化剤の使用量は、特に制限はなく、収率等の観点から、化合物(4)1モルに対して、1~10モル(特に2~5モル)使用することが好ましい。
酸化剤を使用しない場合、反応雰囲気を酸素を含む雰囲気(酸素雰囲気、空気雰囲気等)とすることにより、効率的に酸化反応を進行させることができる。
本工程において使用され得る有機溶媒としては、公知のものを採用することができ、本工程では、例えば、テトラヒドロフラン、ジオキサン等の環状エーテル化合物;ジメチルスルホキシド(DMSO)等が好ましい。なお、上記環化工程と同じ溶媒を使用することもできる。また、反応条件は、反応が十分に進行する程度が好ましく、例えば、反応雰囲気は酸素を含む雰囲気(酸素雰囲気、空気雰囲気等)が好ましく、反応温度は-50~150℃、特に0~100℃が好ましく、反応時間は5分~72時間、特に10分~48時間が好ましい。
反応終了後は、必要に応じて常法にしたがって精製処理をすることもできる。また、精製処理を施さずに次の工程を行うこともできる。
(2-3)求核付加反応(化合物(5)→化合物(1))
本工程では、例えば、有機溶媒中で、有機リチウム化合物及び酸化剤の存在下に、上記反応式1における化合物(5)と所望のハロゲン化芳香族化合物とを反応させることで、求核付加反応を引き起こすことができる。なお、本工程では、有機リチウム化合物及び所望のハロゲン化芳香族化合物の代わりに、所望のグリニャール試薬を使用することによっても、同様の反応を進行することができる。グリニャール試薬は、あらかじめ合成することもでき、上記ハロゲン化芳香族化合物を用いて、常法により系中で合成することもできる。
本工程では、例えば、有機溶媒中で、有機リチウム化合物及び酸化剤の存在下に、上記反応式1における化合物(5)と所望のハロゲン化芳香族化合物とを反応させることで、求核付加反応を引き起こすことができる。なお、本工程では、有機リチウム化合物及び所望のハロゲン化芳香族化合物の代わりに、所望のグリニャール試薬を使用することによっても、同様の反応を進行することができる。グリニャール試薬は、あらかじめ合成することもでき、上記ハロゲン化芳香族化合物を用いて、常法により系中で合成することもできる。
この際使用できるハロゲン化芳香族化合物は、例えば、一般式(6):

酸化剤としては、特に制限はなく、塩化水素(塩酸)等を使用することができる。
上記有機リチウム化合物及びハロゲン化芳香族化合物の使用量は、特に制限はなく、収率等の観点から、化合物(5)1モルに対して、有機リチウム化合物を1~10モル(特に2~5モル)、ハロゲン化芳香族化合物を1~10モル(特に2~5モル)使用することが好ましい。また、酸化剤は水溶液として過剰量使用することが好ましい。
本工程において使用され得る有機溶媒としては、公知のものを採用でき、本工程では、例えば、テトラヒドロフラン、ジオキサン等の環状エーテル化合物が好ましい。なお、上記環化工程と同じ溶媒を使用することもできる。また、反応条件は、反応が十分に進行する程度とすることができ、例えば、反応雰囲気は不活性ガス雰囲気(窒素ガス雰囲気、アルゴンガス雰囲気等)が好ましく、反応温度は-50~100℃、特に0~50℃が好ましく、反応時間は5分~10時間、特に10分~5時間が好ましい。
反応終了後は、必要に応じて常法にしたがって精製処理をすることにより、本発明のホスファローダミン化合物を得ることができる。
(2-4)求核付加反応及び酸化反応(化合物(5)→化合物(1A))
本工程では、例えば、有機溶媒中で、有機リチウム化合物及び酸化剤の存在下に、上記反応式1における化合物(5)と所望のハロゲン化芳香族化合物とを反応させることで、求核付加反応を引き起こし、さらに、別途酸化剤を用いて酸化反応を起こすことで、化合物(1A)を得ることができる。なお、本工程では、上記(2-3)とは異なり、有機リチウム化合物及び所望のハロゲン化芳香族化合物の代わりに、所望のグリニャール試薬を使用した場合には、ほとんど反応が進行しない。
本工程では、例えば、有機溶媒中で、有機リチウム化合物及び酸化剤の存在下に、上記反応式1における化合物(5)と所望のハロゲン化芳香族化合物とを反応させることで、求核付加反応を引き起こし、さらに、別途酸化剤を用いて酸化反応を起こすことで、化合物(1A)を得ることができる。なお、本工程では、上記(2-3)とは異なり、有機リチウム化合物及び所望のハロゲン化芳香族化合物の代わりに、所望のグリニャール試薬を使用した場合には、ほとんど反応が進行しない。

有機リチウム化合物としては、上記(2-3)と同様のものを使用することができる。好ましい具体例も同様である。
酸化剤としては、特に制限はなく、求核付加反応及び酸化反応ともに、塩化水素(塩酸)等を使用することができる。
上記有機リチウム化合物及びハロゲン化芳香族化合物の使用量は、特に制限はなく、収率等の観点から、化合物(5)1モルに対して、有機リチウム化合物を1~10モル(特に2~5モル)、ハロゲン化芳香族化合物を1~10モル(特に2~5モル)使用することが好ましい。また、酸化剤は求核付加反応及び酸化反応ともに、水溶液として過剰量使用することが好ましい。
本工程において使用され得る有機溶媒としては、公知のものを採用でき、本工程では、例えば、テトラヒドロフラン、ジオキサン等の環状エーテル化合物が好ましい。なお、上記環化工程と同じ溶媒を使用することもできる。また、反応条件は、反応が十分に進行する程度とすることができ、例えば、反応雰囲気は不活性ガス雰囲気(窒素ガス雰囲気、アルゴンガス雰囲気等)が好ましく、反応温度は-50~100℃、特に0~50℃が好ましく、反応時間は5分~10時間、特に10分~5時間が好ましい。
反応終了後は、必要に応じて常法にしたがって精製処理をすることもできる。また、精製処理を施さずに次の工程を行うこともできる。
(2-5)縮合反応(化合物(1A)→化合物(1B))
R11として反応性基を導入する場合、常法により、化合物(1A)のカルボキシ基を所望の反応性基に置換することができる。この際、例えば、カルボジイミド系縮合剤(N,N’-ジシクロヘキシルカルボジイミド、1-(3-ジメチルアミノプロピル)-3-エチルカルボジイミド塩酸塩、ジイソプロピルカルボジイミド)、イミダゾール系縮合剤(カルボニルジイミダゾール、2-クロロ-1,3-ジメチルイミダゾリニウムクロリド)、トリアジン系縮合剤(4-(4,6-ジメトキシ-1,3,5-トリアジン-2-イル)-4-メチルモルホリニウムクロリド)、ホスホニウム系縮合剤(ベンゾトリアゾール-1-イルオキシ-トリスジメチルアミノホスホニウム塩、ヘキサフルオロリン酸(ベンゾトリアゾール-1-イルオキシ)トリピロリジノホスホニウム)、ウロニウム系縮合剤({{[(1-シアノ-2-エトキシ-2-オキソエチリデン)アミノ]オキシ}-4-モルホリノメチレン}ジメチルアンモニウムヘキサフルオロリン酸塩、O-(ベンゾトリアゾール-1-イル)-N,N,N’,N’-テトラメチルウロニウムヘキサフルオロリン酸塩、O-(7-アザベンゾトリアゾール-1-イル)-N,N,N’,N’-テトラメチルウロニウムヘキサフルオロリン酸塩、N,N,N’,N’-テトラメチル-O-(N-スクシンイミジル)ウロニウムテトラフルオロボレート(TSTU))等の縮合剤を使用することができる。例えば、TSTUを使用した場合には、アミン反応性基としてスクシンイミド基を導入し、タンパク質標識剤(特に抗体標識剤)として機能することができる。
R11として反応性基を導入する場合、常法により、化合物(1A)のカルボキシ基を所望の反応性基に置換することができる。この際、例えば、カルボジイミド系縮合剤(N,N’-ジシクロヘキシルカルボジイミド、1-(3-ジメチルアミノプロピル)-3-エチルカルボジイミド塩酸塩、ジイソプロピルカルボジイミド)、イミダゾール系縮合剤(カルボニルジイミダゾール、2-クロロ-1,3-ジメチルイミダゾリニウムクロリド)、トリアジン系縮合剤(4-(4,6-ジメトキシ-1,3,5-トリアジン-2-イル)-4-メチルモルホリニウムクロリド)、ホスホニウム系縮合剤(ベンゾトリアゾール-1-イルオキシ-トリスジメチルアミノホスホニウム塩、ヘキサフルオロリン酸(ベンゾトリアゾール-1-イルオキシ)トリピロリジノホスホニウム)、ウロニウム系縮合剤({{[(1-シアノ-2-エトキシ-2-オキソエチリデン)アミノ]オキシ}-4-モルホリノメチレン}ジメチルアンモニウムヘキサフルオロリン酸塩、O-(ベンゾトリアゾール-1-イル)-N,N,N’,N’-テトラメチルウロニウムヘキサフルオロリン酸塩、O-(7-アザベンゾトリアゾール-1-イル)-N,N,N’,N’-テトラメチルウロニウムヘキサフルオロリン酸塩、N,N,N’,N’-テトラメチル-O-(N-スクシンイミジル)ウロニウムテトラフルオロボレート(TSTU))等の縮合剤を使用することができる。例えば、TSTUを使用した場合には、アミン反応性基としてスクシンイミド基を導入し、タンパク質標識剤(特に抗体標識剤)として機能することができる。
この際の他の添加剤、反応条件等は常法により行うことができる。
このようにして、一般式(1)で表されるホスファローダミン化合物を得ることができ、必要に応じて通常の単離及び精製工程を経て使用することもできる。
なお、上記では、本発明のホスファローダミン化合物の一態様の合成方法の一例について記載したが、この製造方法に限定されることはなく、様々な合成方法で合成することができる。
3.蛍光色素及びタンパク質標識剤
本発明の蛍光色素は、上記の本発明のホスファローダミン化合物又はその塩からなる。

本発明の蛍光色素は、ローダミンのキサンテン骨格の9位の位置にP(R5)=O構造を有しているため、ホスファローダミン骨格を拡張せずとも、従来の近赤外蛍光色素よりも蛍光極大波長をさらに長波長化することができる。具体的には、吸収極大波長を680~730nm、特に690~720nmとすることができ、蛍光極大波長を720~750nm、特に730~740nmとすることができる。また、このように、本発明の蛍光色素は、蛍光極大波長をより長波長化していることから、葉緑体の自家蛍光をより低減した領域で蛍光させることが可能であることから植物類のイメージングもしやすい。
また、本発明の蛍光色素は、また、従来の蛍光色素と比較しても、様々なpH条件において、より長時間にわたって、吸収強度及び蛍光強度を維持することができる。特に、R8として、R9及びR10がいずれも置換若しくは非置換アルキル基、又は置換若しくは非置換アルコキシ基である一般式(2)で表される基を採用することにより、蛍光極大波長をより長波長化することができ、且つ、様々なpHにおいてより長時間にわたって吸収強度及び蛍光強度を維持することができる。
本発明の蛍光色素は、一般式(1)におけるR5が置換若しくは非置換アリール基であるため、リン原子上にP(O-)=O構造を有する場合と異なり、アニオン化しない。一方、細胞膜は、細胞の内外を隔てる生体膜であり、特異的なチャンネルによってイオン等の低分子を透過させたり、受容体を介して細胞外からのシグナルを受け取ったりする機能等を有しており、この細胞膜を構成する主要成分はリン脂質であり、アニオン性が高い構造であることが知られている。R5がアニオン化しやすい構造である場合には、アニオン性の高い細胞膜を透過させることは困難であるため、本発明の蛍光色素は、アニオン化しないことにより、細胞膜を透過しやすい蛍光色素である。このため、本発明の蛍光色素は、生体深部の非浸襲イメージングがしやすい。
さらに、本発明のホスファローダミン化合物又はその塩において、R8で示される基をアミン反応性基又はチオール反応性基とする場合には、タンパク質(特に抗体)が有する置換若しくは非置換アミノ基、チオール基等と反応させることができる。このため、本発明のホスファローダミン化合物又はその塩を、タンパク質(特に抗体)を標識する(蛍光させる)ためのタンパク質標識剤(特に抗体標識剤)として用いることが可能である。この際、本発明のホスファローダミン化合物又はその塩は、上記のとおり、蛍光極大波長をより長波長化していることから、生体へのダメージを大幅に低減することができる。また、本発明のホスファローダミオン化合物又はその塩を用いた場合には、対象となるタンパク質(特に抗体)を従来よりも長時間にわたって蛍光させることが可能である。
本発明のホスファローダミン化合物又はその塩をタンパク質標識剤(特に抗体標識剤)として使用する場合、その対象となるタンパク質(特に抗体)としては、例えば、アビジン、ストレプトアビジン、アネキシンV、抗IgG抗体、抗IgM抗体、抗CD3抗体、抗CD4抗体、抗CD20抗体、抗CD25抗体、抗CD43抗体、抗CD44抗体、抗CD68抗体、抗IFN-γ抗体、抗TNF-α抗体、抗Ly-6G抗体、抗Ku70抗体、抗IL-4抗体、抗IL-17抗体、抗IL-31抗体、抗Notch1抗体、抗Notch3抗体、抗FOXBP3抗体、抗Ki-67抗体、抗HLA-A2抗体、抗α-チューブリン抗体、抗カテプシン-D抗体、抗アンジオテンシン抗体、抗COX1抗体、抗GLUT1抗体、抗AKT1/2/3抗体、抗Apg3抗体、抗βカテニン抗体、抗CDK5抗体、抗CEA抗体、抗HER2抗体、等が挙げられる。
本発明のホスファローダミン化合物又はその塩のうち、R8で示される基をアミン反応性基又はチオール反応性基とした化合物をタンパク質標識剤(特に抗体標識剤)に用いる場合は、本発明のタンパク質標識剤(特に抗体標識剤)は、本発明のホスファローダミン化合物又はその塩を含有しているが、有機溶媒中に溶解させて溶液とすることが好ましい。具体的には、本発明のホスファローダミン化合物の濃度は1×10-8~1×10-4 mol/Lが好ましく、1×10-7~1×10-5 mol/Lがより好ましい。このように、本発明では、従来の蛍光色素と比較し、ホスファローダミン化合物の含有量を低く抑えることができる。
本発明の蛍光色素(ホスファローダミン化合物又はその塩)を、本発明のタンパク質標識剤(特に抗体標識剤)を含有する溶液とする場合、使用し得る有機溶媒としては、特に制限はなく、極性溶媒及び非極性溶媒のいずれも使用できる。
極性溶媒としては、例えば、エーテル化合物(テトラヒドロフラン、アニソール、1,4-ジオキサン、シクロペンチルメチルエーテル等)、アルコール(メタノール、エタノール、アリルアルコール等)、エステル化合物(酢酸エチル等)、ケトン(アセトン等)、ハロゲン化炭化水素(ジクロロメタン、クロロホルム)、ジメチルスルホキシド、アミド系溶媒(N,N-ジメチルホルムアミド、ジメチルアセトアミド、1,3-ジメチル-2-イミダゾリジノン、N-メチルピロリドン等)等が挙げられる。
非極性溶媒としては、例えば、ペンタン、ヘキサン、シクロヘキサン、ヘプタン等の脂肪族有機溶媒;ベンゼン、トルエン、キシレン、メシチレン等の芳香族溶媒等が挙げられる。
本発明のタンパク質標識剤(特に抗体標識剤)は、上記のとおり、吸収極大波長を680~730nm、特に690~720nmとし、蛍光極大波長を720~750nm、特に730~740nmとしつつも、生理的条件下でもより十分に高い蛍光量子収率としつつ、細胞中に投入する観点から、pHは4~11程度が好ましく、4.5~8.0程度がより好ましい。本発明のタンパク質検出剤(特に抗体検出剤)のpHを調整するために、緩衝剤(ヘペス緩衝剤、トリス緩衝剤、トリシン-水酸化ナトリウム緩衝剤、リン酸系緩衝剤、リン酸緩衝生理食塩水等)等を使用してもよい。
4.タンパク質標識キット及びタンパク質標識方法
本発明のタンパク質標識キットは、本発明のタンパク質標識剤(特に抗体標識剤)を含有する。その他、必要に応じて、タンパク質(特に抗体)の標識に用いる際に使用するバッファー、培地、使用説明書等を含有することもできる。本発明のタンパク質標識キットを用いれば、タンパク質(特に抗体)を容易に標識することが可能である。使用できるバッファー、培地等は、従来から使用されている公知のものを採用することができる。
本発明のタンパク質標識キットは、本発明のタンパク質標識剤(特に抗体標識剤)を含有する。その他、必要に応じて、タンパク質(特に抗体)の標識に用いる際に使用するバッファー、培地、使用説明書等を含有することもできる。本発明のタンパク質標識キットを用いれば、タンパク質(特に抗体)を容易に標識することが可能である。使用できるバッファー、培地等は、従来から使用されている公知のものを採用することができる。
また、本発明のタンパク質標識方法によれば、本発明のタンパク質標識剤と、タンパク質(特に抗体)とを反応させることにより、タンパク質(特に抗体)を容易に標識することが可能である。
この反応は、本発明のタンパク質標識剤(特に抗体標識剤)と、タンパク質(特に抗体)とを接触させることにより行うことができる。接触方法は特に制限されず、例えば、タンパク質(特に抗体)を含有する溶液中に、本発明のタンパク質標識剤(特に抗体標識剤)を含有する溶液を添加する方法が挙げられる。また、本発明のタンパク質標識剤(特に抗体標識剤)を含むバッファー又は培地に、タンパク質(特に抗体)を移す方法も挙げられる。各溶液中のタンパク質(特に抗体)及び本発明のタンパク質標識剤(特に抗体標識剤)の分量は特に制限されず、常法にしたがうことができる。
実施例に基づいて、本発明を具体的に説明するが、本発明は、これらのみに限定されるものではない。
1H NMRスペクトル及び13C{1H} NMRスペクトルは、JEOL AL-400 spectrometer(1H: 400 MHz、13C: 100 MHz)、JEOL JNM-ECS400(1H: 400 MHz、13C: 100 MHz)、又はJEOL A-600 spectrometer(1H: 600 MHz、13C: 150 MHz)を用いて、溶媒としてCDCl3又はCD3OD中で測定した。1H NMRスペクトルの化学シフトは、内部標準(CHCl3 δ 7.26、CD3OD δ 5.30)として溶媒の残留プロトンを用いてδppmで表記した。13C NMRスペクトルの化学シフトは、内部標準(CDCl3 δ 77.16、CD3OD δ 49.0)として溶媒のシグナルを用いて報告した。マススペクトルは、Thermo Fisher Scientific Exactiveによるエレクトロスプレーイオン化(ESI)法により測定した。薄層クロマトグラフィー(TLC)は、シリカゲル60F254(Merck)を0.25 mmの厚さで塗布した板を用いて行った。カラムクロマトグラフィーは、中性シリカゲルPSQ100B(富士シリシア化学(株))又はシリカゲル60(関東化学(株))を用いて行った。分取リサイクルHPLCには、逆相カラム(YMC-Actus Triart C18)を備えたYMC LC-Forte/Rを用いて行った。特に制限のない限り、全ての反応は窒素雰囲気下で行った。特に制限のない限り、市販の溶媒及び試薬は、さらに精製することなく使用した。無水テトラヒドロフラン(THF)及びCH2Cl2は関東化学(株)から購入し、Glass Contour Solvent Systemsで精製した。ビス(2-ブロモ-4-ジエチルアミノフェニル)メタン(化合物1)及び4-ブロモ-3,5-ジメチルベンズアルデヒドは既報にしたがい合成した(Chem. Commun. 2012, 48, 8781-8783、Chem. Sci. 2012, 3, 1938-1944)。
合成例1:3,7-ビス(ジエチルアミノ)-5-フェニル-10H-アクリドホスフィン-10-オン5-オキシド(化合物2)
[式中、Etはエチル基を示す。s-BuLiはsec-ブチルリチウムを示す。Phはフェニル基を示す。TBABはテトラブチルアンモニウムブロミドを示す。DMSOはジメチルスルホキシドを示す。THFはテトラヒドロフランを示す。以下同様である。]
無水テトラヒドロフラン(THF; 50mL)中のビス(2-ブロモ-4-ジエチルアミノフェニル)メタン(化合物1; 5.15g, 11.0mmol)の溶液に、sec-ブチルリチウム(シクロヘキサン中の0.99M溶液, 22.2mL, 22mmol)を-78℃で30分間かけて添加し、得られた混合物を-78℃で1.5時間撹拌した。次いで、P,P-ジクロロフェニルホスフィン(1.49mL, 11.0mmol)をゆっくりと30分以上かけて添加し、混合物を一晩で室温まで昇温した。次いで、混合物を0℃まで冷却し、35%過酸化水素水(5mL)を添加した。得られた混合物を0℃で1時間撹拌し、飽和Na2SO3水溶液(35mL)を添加して反応をクエンチした。混合物をクロロホルムで4回抽出し、水及び飽和食塩水で洗浄し、無水Na2SO4で乾燥した。溶媒を蒸発させ、油状の残渣を50mLのジメチルスルホキシド(DMSO)に溶解させ、K2CO3粉末(4.56g, 33mmol)及びテトラブチルアンモニウムブロミド(TBAB; 89mg, 0.275mmol)を添加した。空気雰囲気下、室温で、得られた懸濁液を一晩激しく撹拌した。次いで、再度、K2CO3粉末(3.04g, 22mmol)及びTBAB(89mg, 0.275mmol)を添加し、懸濁液をさらに50℃で24時間撹拌した。混合物を水(150mL)で希釈し、冷ました。得られた黄色沈殿をろ過し、水で洗浄した。得られた固体をジクロロメタン(DCM)に溶解させ、層分離した。水層をDCMで5回抽出し、合わせた有機層を水で2回洗浄し、無水Na2SO4で乾燥した。溶媒を真空下に除去した後、残渣(5.35g)をトルエンから再結晶し、山吹色固体として化合物2を得た(2.18g, 44%)。再結晶により得られたろ液には、まだ化合物2と未酸化の前駆体2-Hが残存している。ろ液を真空下に濃縮し、残渣を60mLのTHFに溶解させた。NaOH粉末(720 mg)及びTBAB(97mg, 0.30mmol)をこの溶液に添加し、得られた懸濁液を、室温で空気雰囲気下に2時間激しく撹拌し、酸化を完了した。混合物を水で希釈し、DCMで4回抽出し、水で洗浄し、無水Na2SO4で乾燥した。溶媒を蒸発させた後に、生成物をシリカゲルカラムクロマトグラフィー(90/10 to 85/15 DCM/アセトン)により精製し、次いで、トルエン/ヘキサンから再結晶させ、化合物2を得た(836mg, 17%)。合計3.02 g(6.76mmol, 61%)の化合物2を山吹色結晶性固体として得た。
無水テトラヒドロフラン(THF; 50mL)中のビス(2-ブロモ-4-ジエチルアミノフェニル)メタン(化合物1; 5.15g, 11.0mmol)の溶液に、sec-ブチルリチウム(シクロヘキサン中の0.99M溶液, 22.2mL, 22mmol)を-78℃で30分間かけて添加し、得られた混合物を-78℃で1.5時間撹拌した。次いで、P,P-ジクロロフェニルホスフィン(1.49mL, 11.0mmol)をゆっくりと30分以上かけて添加し、混合物を一晩で室温まで昇温した。次いで、混合物を0℃まで冷却し、35%過酸化水素水(5mL)を添加した。得られた混合物を0℃で1時間撹拌し、飽和Na2SO3水溶液(35mL)を添加して反応をクエンチした。混合物をクロロホルムで4回抽出し、水及び飽和食塩水で洗浄し、無水Na2SO4で乾燥した。溶媒を蒸発させ、油状の残渣を50mLのジメチルスルホキシド(DMSO)に溶解させ、K2CO3粉末(4.56g, 33mmol)及びテトラブチルアンモニウムブロミド(TBAB; 89mg, 0.275mmol)を添加した。空気雰囲気下、室温で、得られた懸濁液を一晩激しく撹拌した。次いで、再度、K2CO3粉末(3.04g, 22mmol)及びTBAB(89mg, 0.275mmol)を添加し、懸濁液をさらに50℃で24時間撹拌した。混合物を水(150mL)で希釈し、冷ました。得られた黄色沈殿をろ過し、水で洗浄した。得られた固体をジクロロメタン(DCM)に溶解させ、層分離した。水層をDCMで5回抽出し、合わせた有機層を水で2回洗浄し、無水Na2SO4で乾燥した。溶媒を真空下に除去した後、残渣(5.35g)をトルエンから再結晶し、山吹色固体として化合物2を得た(2.18g, 44%)。再結晶により得られたろ液には、まだ化合物2と未酸化の前駆体2-Hが残存している。ろ液を真空下に濃縮し、残渣を60mLのTHFに溶解させた。NaOH粉末(720 mg)及びTBAB(97mg, 0.30mmol)をこの溶液に添加し、得られた懸濁液を、室温で空気雰囲気下に2時間激しく撹拌し、酸化を完了した。混合物を水で希釈し、DCMで4回抽出し、水で洗浄し、無水Na2SO4で乾燥した。溶媒を蒸発させた後に、生成物をシリカゲルカラムクロマトグラフィー(90/10 to 85/15 DCM/アセトン)により精製し、次いで、トルエン/ヘキサンから再結晶させ、化合物2を得た(836mg, 17%)。合計3.02 g(6.76mmol, 61%)の化合物2を山吹色結晶性固体として得た。

合成例2:2-(4-ブロモ-3,5-ジメチルフェニル)-4,4-ジメチル-2-オキサゾリン(化合物3)
化合物3は、以下の通り、4-ブロモ-3,5-ジメチルベンズアルデヒドから、Gloriusらにより報告されたオキサゾリン合成法(Synthesis, 2006, 2996-3002)を修正した方法により得た。

窒素雰囲気下、4-ブロモ-3,5-ジメトキシ安息香酸(5.22g, 20.0mmol)、N,N’-ジシクロヘキシルカルボジイミド(DCC; 5.36g, 26.0mmol)、及びN,N-ジメチル-4-アミノピリジン(DMAP; 244mg, 2.00mmol)を60mLのジクロロメタンに溶解させた。ここに、ジクロロメタン(20mL)中のtert-ブチルアルコール(3.85g, 52.0mmol)の溶液を添加し、得られた混合物を室温で3日間撹拌した。白色沈殿をろ過により除去し、ろ液を減圧下に濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/DCM 3/2 to 1/2)で精製し、白色固体として化合物4を得た(3.34g, 10.5mmol, 53%)。

実施例1:ホスファローダミン化合物PR1
無水テトラヒドロフラン(THF; 3mL)中の2-ブロモトルエン(90μL, 0.75mmol)の溶液に、sec-ブチルリチウム(シクロヘキサン中の0.99M溶液, 0.91mL, 0.90mmol)を、-78℃でゆっくりと添加し、得られた混合物を同じ温度で1時間撹拌した。ここに、テトラヒドロフラン(THF; 3mL)中の合成例1で得た化合物2(112mg, 0.251mmol)の溶液を20分以上かけて滴下し、混合物を1.5時間かけて室温まで昇温した。次いで、ここに20mLの1M塩酸を添加し、撹拌を30分間継続した。この間に、混合物の色彩が淡黄色から深緑色に変化した。混合物を飽和食塩水で希釈し、DCMで5回抽出した。有機層を合わせ、Na2SO4で乾燥した。乾燥剤をろ過し、溶媒を減圧下に除去した。生成物をシリカゲルカラムクロマトグラフィー(DCM/メタノール 95/5 to 85/15)で精製した。溶離液を減圧下に除去した後、生成物のクロロホルム溶液にジエチルエーテルを少しずつ添加して再結晶させた。1H NMR分析の結果から、得られた緑色結晶性粉末(89mg)は、化合物PR1とCHCl3のモル比1:2の共結晶であり、真空下の乾燥では溶媒は除去できなかった。固体をメタノール(約3mL)中に溶解させ、溶媒を蒸発させて乾燥させ、残渣を真空下に乾燥し、深緑色固体として化合物PR1を得た(69mg, 0.124mmol, 49%)。

実施例2:ホスファローダミン化合物PR2
2-ブロモ-1,3-ジメチルベンゼン(200μL, 1.50mmol)を、無水テトラヒドロフラン(THF; 5mL)中の削り状マグネシウム(38.3mg, 1.58mmol)の溶液に添加した。1,2-ジブロモエタン(15μL, 0.17mmol)を添加して金属-ハロゲン交換反応を起こし、得られた混合物を室温で16時間撹拌した(マグネシウム金属は完全に消費した)。得られたグリニャール試薬の溶液に、無水テトラヒドロフラン(THF; 10mL)中の合成例1で得た化合物2(223mg, 0.50mmol)の溶液を添加し、反応混合物を室温で2時間、還流(70℃)下に20時間撹拌した。混合物を室温まで冷却した後、20mLの2M塩酸を添加し、得られた乳濁液を1時間撹拌した。混合物をクロロホルム及び水で希釈し、層分離した。水層をクロロホルムで5回抽出し、合わせた有機層を2M塩酸及び水で洗浄し、無水Na2SO4で乾燥した。乾燥剤をろ過し、溶媒を減圧下に除去した。生成物をシリカゲルカラムクロマトグラフィー(DCM/メタノール 9/1)で精製し、生成物のDCM溶液にジエチルエーテルをゆっくりと添加して再結晶させた。結晶性溶媒(DCM, ジエチルエーテル及び水)を除去するため、得られた茶色粉末をメタノールに溶解させ、溶媒を蒸発させて乾燥させた。残渣を真空下に乾燥し、深緑色固体として化合物PR2を得た(57mg, 0.10mmol, 20%)。

無水テトラヒドロフラン(THF; 8mL)中の合成例2で得た化合物3(436mg, 1.55mmol)の溶液に、sec-ブチルリチウム(シクロヘキサン中の0.99M溶液, 1.59mL, 1.57mmol)を-78℃でゆっくりと添加し、得られた混合物を同じ温度で30分間撹拌した。ここに、無水テトラヒドロフラン(THF; 10mL)中の合成例1で得た化合物2(223mg, 0.50mmol)の溶液を20分以上かけて滴下し、混合物を2時間かけて室温まで昇温した。次いで、ここに20mLの2M塩酸を添加し、撹拌を30分間継続した。混合物を飽和食塩水で希釈し、DCMで5回抽出した。有機層を合わせ、飽和食塩水で洗浄し、無水Na2SO4で乾燥した。乾燥剤をろ過し、溶媒を減圧下に除去した。残渣を50mLの6M塩酸に溶解させ、80℃で6時間撹拌した。混合物を飽和食塩水で希釈し、DCMで抽出した。合わせた有機層を飽和食塩水で洗浄し、無水Na2SO4で乾燥した。ろ過し、溶媒を減圧下に除去した後に、生成物をシリカゲルカラムクロマトグラフィー(DCM/メタノール/HCOOH 88/12/0.5 to 80/20/1)で単離した。生成物を含む留分を2M塩酸で2回洗浄し、無水Na2SO4で乾燥した。乾燥剤をろ過して除去し、溶媒を蒸発させた。生成物のDCM溶液にジエチルエーテルをゆっくりと添加して再結晶させた。生成物を真空下に乾燥させ、緑色粉末として化合物PR4を得た(47mg, 0.076mmol, 15%)。

実施例5:ホスファローダミン化合物PR5
無水テトラヒドロフラン(THF; 10mL)中の合成例3で得た化合物4(856mg, 2.70mmol)の溶液に、tert-ブチルリチウム(n-ペンタン中の1.64M溶液, 3.30mL, 5.41mmol)を-78℃でゆっくりと添加し、得られた混合物を同じ温度で1時間撹拌した。ここに、無水テトラヒドロフラン(THF; 20mL)中の合成例1で得た化合物2(402mg, 0.90mmol)の溶液を20分以上かけて滴下し、混合物を一晩かけて室温まで昇温した。次いで、ここに30mLの2M塩酸を添加し、撹拌を1時間継続した。混合物を飽和食塩水で希釈し、DCMで5回抽出した。有機層を合わせ、希釈塩酸で洗浄し、無水Na2SO4で乾燥した。乾燥剤をろ過し、溶媒を減圧下に除去した。残渣を10mLのトリフルオロ酢酸に溶解させ、室温で2時間撹拌した。混合物を100mLの水に注ぎ、130mLの1M NaOH水溶液を添加して中和した。得られた溶液をDCMで5回抽出した。合わせた有機層を2M HClで2回洗浄し、無水Na2SO4で乾燥した。ろ過し、溶媒を減圧下に除去した後に、生成物をシリカゲルカラムクロマトグラフィー(DCM/メタノール/HCOOH 85/15/0.2)で単離した。溶離液を蒸発させ、生成物をDCMに溶解させ、2M HClで3回洗浄し、無水Na2SO4で乾燥した。乾燥剤をろ過して除去し、溶媒を蒸発させた。生成物のDCM溶液にジエチルエーテルをゆっくりと添加して再結晶させた。生成物を真空下に乾燥させ、茶色粉末として化合物PR5を得た(62mg, 0.096mmol, 11%)。

実施例6:ホスファローダミンNHSエステルPR4-NHS
無水ジメチルホルムアミド(DMF; 3mL)中の、実施例4で得た化合物PR4(24mg, 0.039mmol)及びN,N,N’,N’-テトラメチル-O-(N-スクシンイミジル)ウロニウムテトラフルオロボレート(TSTU; 23.5mg, 0.078mmol)の溶液に、N,N-ジイソプロピルエチルアミン(27μL, 0.156mmol)を添加し、室温で混合物を18時間撹拌した。反応混合物を5%クエン酸水溶液で希釈し、DCMで4回抽出した。合わせた有機層を飽和食塩水で2回洗浄し、無水Na2SO4で乾燥した。乾燥剤をろ過し、ろ液を減圧下に濃縮した。残存する生成物のDMF溶液に、2mLのDCMを添加し、次いで、ジエチルエーテルをゆっくりと添加して生成物を沈殿させた。ろ過により固体を集め、2mLの水に溶解させた。残存する溶液を液体窒素中で凍結してフリーズドライし、緑色粉末として化合物PR4-NHSを得た(24.5mg, 0.034mmol, 87%)。

実施例7:ホスファローダミンNHSエステルPR5-NHS
無水ジメチルホルムアミド(DMF; 3mL)中の、実施例5で得た化合物PR5(35mg, 0.054mmol)及びN,N,N’,N’-テトラメチル-O-(N-スクシンイミジル)ウロニウムテトラフルオロボレート(TSTU; 32.5mg, 0.108mmol)の溶液に、N,N-ジイソプロピルエチルアミン(38μL, 0.218mmol)を添加し、室温で混合物を16時間撹拌した。反応混合物をDCMで希釈し、5%クエン酸水溶液で2回洗浄し、飽和食塩水で2回洗浄し、無水Na2SO4で乾燥した。乾燥剤をろ過し、ろ液を減圧下に濃縮した。生成物を水に溶解させ、得られた溶液を液体窒素中で凍結してフリーズドライし、緑色粉末として化合物PR5-NHSを得たことを確認した。

合成例4(キサントン化合物6)
窒素雰囲気下、無水テトラヒドロフラン(THF; 10mL)中の化合物5(960mg, 2.20mmol)の溶液に、sec-ブチルリチウム(シクロヘキサン及びn-ヘキサン中の1.00M溶液, 5.10mL, 5.10mmol)を-78℃でゆっくりと添加し、得られた混合物を-78℃で1.5時間撹拌した。次いで、P,P-ジクロロフェニルホスフィン(0.33mL, 2.4mmol)をゆっくりと30分以上かけて添加し、混合物を一晩で室温まで昇温した。次いで、混合物を0℃まで冷却し、35%過酸化水素水(1mL)を添加した。得られた混合物を0℃で1時間撹拌し、水及びジクロロメタンで希釈した。層を分離し、水層をジクロロメタンで4回抽出した。合わせた有機層をブラインで洗浄し、無水Na2SO4で乾燥させた。溶媒を蒸発させ、残渣を30mLのテトラヒドロフラン(THF)に溶解させた。NaOH粉末(264mg, 6.60mmol)及びテトラブチルアンモニウムブロミド(TBAB; 35mg, 0.11mmol)を添加し、空気雰囲気下、室温で、得られた懸濁液を4時間激しく撹拌した。混合物をロータリーエバポレーターで約10mLの体積まで濃縮した。次いで、混合物を水で希釈し、ジクロロメタン(DCM)で4回抽出し、ブラインで洗浄し、無水Na2SO4で乾燥させた。溶媒を蒸発させた後、生成物をシリカゲルカラムクロマトグラフィー(シリカ, DCM/アセトン 90/10 to 80/20)により精製した。溶媒を真空下に蒸発させ、キサントン化合物6を山吹色固体として得た(249mg, 0.601mmol, 27%)。

実施例8:ホスファローダミン化合物PR6
窒素雰囲気下、無水テトラヒドロフラン(THF; 5mL)中の2-ブロモ-1,3-ジメトキシベンゼン(217mg, 1.00mmol)の溶液に、sec-ブチルリチウム(シクロヘキサン中の1.00M溶液, 1.10mL, 1.10mmol)を、-78℃でゆっくりと添加し、得られた混合物を同じ温度で1.5時間撹拌した。ここに、テトラヒドロフラン(THF; 10mL)中の合成例4で得たキサントン化合物6(83mg, 0.20mmol)の溶液を30分以上かけて滴下した。室温まで昇温し、室温で混合物を一晩撹拌した。次いで、ここに15mLの1M塩酸を添加し、撹拌を1時間継続した。得られた混合物を水で希釈し、ジエチルエーテルで2回洗浄した。水層をジクロロメタン(DCM)で4回抽出した。ジクロロメタン(DCM)層を合わせ、ブラインで洗浄し、Na2SO4で乾燥させた。乾燥剤をろ過し、溶媒を減圧下に除去した。生成物をシリカゲルカラムクロマトグラフィー(DCM/メタノール 85/15)で精製した。溶媒を除去し、ホスファローダミン化合物PR6を深緑色固体として得た(18mg, 0.032mmol, 16%)。

試験例1:光物理特性
PR1(実施例1)、PR2(実施例2)、PR4(実施例4)及びPR6(実施例8)の吸収スペクトル及び蛍光スペクトルを測定した。具体的には、吸収スペクトルは、PBSバッファー水溶液(pH7.4)に各化合物を10-6M溶解させた試料溶液を用いて、Shimadzu UV-3150 spectrometerにより、解像度0.2nmで測定した。また、蛍光スペクトルは、解像度1nmのHitachi F-4500 spectrometerで、PBSバッファー水溶液(pH7.4)に各化合物を10-6M溶解させた試料溶液を用いて測定した。絶対蛍光量子収率は、Hamamatsu photonics PMA-11で測定した。結果を図1及び表1に示す。図1において、破線が吸収スペクトル、実線が蛍光スペクトルである。この結果、PR1、PR2及びPR4は、類似した光物理特性を有していた。また、PR6のように、ホスファローダミン骨格のπ共役を拡張することによって、吸収極大波長及び蛍光極大波長を70~90nm程度長波長化することが可能であった。さらに、これら全ての化合物は水溶性である。このため、DMSOを共溶媒として添加することなく、測定試料を作製することができた。
PR1(実施例1)、PR2(実施例2)、PR4(実施例4)及びPR6(実施例8)の吸収スペクトル及び蛍光スペクトルを測定した。具体的には、吸収スペクトルは、PBSバッファー水溶液(pH7.4)に各化合物を10-6M溶解させた試料溶液を用いて、Shimadzu UV-3150 spectrometerにより、解像度0.2nmで測定した。また、蛍光スペクトルは、解像度1nmのHitachi F-4500 spectrometerで、PBSバッファー水溶液(pH7.4)に各化合物を10-6M溶解させた試料溶液を用いて測定した。絶対蛍光量子収率は、Hamamatsu photonics PMA-11で測定した。結果を図1及び表1に示す。図1において、破線が吸収スペクトル、実線が蛍光スペクトルである。この結果、PR1、PR2及びPR4は、類似した光物理特性を有していた。また、PR6のように、ホスファローダミン骨格のπ共役を拡張することによって、吸収極大波長及び蛍光極大波長を70~90nm程度長波長化することが可能であった。さらに、これら全ての化合物は水溶性である。このため、DMSOを共溶媒として添加することなく、測定試料を作製することができた。
試験例2:蛍光のpH依存性の評価
PR1(実施例1)、PR2(実施例2)及びPR4(実施例4)の吸収スペクトル及び蛍光スペクトルを、様々なpHのバッファー水溶液中で測定した。pH3~6の調整にはクエン酸/Na2HPO4バッファー水溶液を用い、pH7~8の調整にはNa2HPO4/NaH2PO4バッファー水溶液を用い、pH9~11の調整にはNa2CO3/NaHCO3バッファー水溶液を用いた。その他、吸収スペクトル及び蛍光スペクトルの測定方法は試験例1と同様に行った。
PR1(実施例1)、PR2(実施例2)及びPR4(実施例4)の吸収スペクトル及び蛍光スペクトルを、様々なpHのバッファー水溶液中で測定した。pH3~6の調整にはクエン酸/Na2HPO4バッファー水溶液を用い、pH7~8の調整にはNa2HPO4/NaH2PO4バッファー水溶液を用い、pH9~11の調整にはNa2CO3/NaHCO3バッファー水溶液を用いた。その他、吸収スペクトル及び蛍光スペクトルの測定方法は試験例1と同様に行った。
様々なpHにおける各化合物の吸収スペクトル及び蛍光スペクトルを図2に示す。図2において、実線はpH4.17、破線はpH7.43、一点鎖線はpH10.30の結果である。この結果、立体的に保護されたPR2(実施例2)及びPR4(実施例4)については、吸収強度及び蛍光強度に対するpHの影響は無視できた。一方、PR1(実施例1)については、OH-アニオンによる9位の炭素原子への求核攻撃により、塩基性下での吸収強度及び蛍光強度は低減し、無色アルコールを形成した。
次に、様々なpHにおけるPR1(実施例1)、PR2(実施例2)及びPR4(実施例4)の時間の経過に伴う吸収強度の安定性を図3及び表2に示す。図3及び表2において、安定性は、吸収強度の維持率(A/A0; 所定時間経過後の吸収強度と、初期吸収強度との比)により示している。図2において、実線はpH4.17、破線はpH7.43、一点鎖線はpH10.30の結果である。表2では、PR1(実施例1)及びPR2(実施例2)について、図3の結果から、十分(無限大)に時間が経過した際の吸収強度の維持率を示している。図3及び表2の結果から、実施例1のPR1を使用した場合は、アルカリ性領域では時間経過とともに吸収強度が低下したのに対し、実施例2のPR2及び実施例4のPR4を使用した場合は、いずれのpHにおいても時間経過による吸収強度の低下は著しく小さかった。
さらに、pH10におけるPR1(実施例1)、PR2(実施例2)、PR3(実施例3)及びPR4(実施例4)の時間の経過に伴う吸収強度の安定性を図4に示す。図4において、安定性は、吸収強度の維持率(A/A0; 所定時間経過後の吸収強度と、初期吸収強度との比)により示している。なお、図4において、実線は実施例1のPR1、破線は実施例2のPR2、二点鎖線は実施例3のPR3、一点鎖線は実施例4のPR4の結果を示す。
この結果、立体的に保護されたPR2(実施例2)、PR3(実施例3)及びPR4(実施例4)については、時間の経過による吸収強度の減少を低減することができた。一方、PR1(実施例1)については、時間経過により吸収強度は低減した。
試験例3:ホスファローダミンNHSエステルと抗体との接合
1mgのヤギ抗マウス抗体IgG(TCI, 1mg/vial, 防腐剤: 0.07% NaN3)を約0.5mLのPBSバッファー溶液(pH7.4)に溶解させ、さらに、PBSバッファー溶液(pH7.4)で希釈して全量を1.0mLとし、抗体溶液を得た。
1mgのヤギ抗マウス抗体IgG(TCI, 1mg/vial, 防腐剤: 0.07% NaN3)を約0.5mLのPBSバッファー溶液(pH7.4)に溶解させ、さらに、PBSバッファー溶液(pH7.4)で希釈して全量を1.0mLとし、抗体溶液を得た。
1mLのバイアルに、PR4-NHS(実施例6; 0.010~0.100mg, 8~40当量)又はPR5-NHS(実施例7; 0.010~0.100mg, 8~40当量)を水(30μL)に溶解させ、上記抗体溶液(500μL)で希釈した。次いで、27μLのカーボネートバッファー溶液(pH9.0, 1M NaOHでpH9.0に調整した0.2M重曹溶液)を添加してpHを8.0~8.5に上昇させ、バイアルを密封し、得られた溶液を手作業で混合した(やさしくシェイクした)。バイアルをアルミニウムホイウムで覆い、室温で1時間放置した。この間、数回やさしくシェイクした。次いで、得られた混合物を、溶離液としてPBSバッファー溶液(pH7.4)を用いたSephadex G-25カラム(Sephadexゲル約4g, 使用前にPBSバッファー溶液(pH7.4)30 mLで安定化させた)でろ過した。最初に色づけられたフラクション(非常に淡い緑色)を集め、約3~4mLの接合溶液を得、UV-Vis分光計及びPL量子収率分光計で測定した。標識化度(DOL)はThermoFisher Scientific社のAlexa Fluor(登録商標)488 Microscale Protein Labeling Kitのマニュアル(https://tools.thermofisher.com/content/sfs/manuals/mp30006.pdf)の「Determination of Degree of Labeling (DOL)」に記載の計算方法に準拠して、A280及びA494の代わりに、A280及びA710を用いて計算した。
結果を図5に示す。図5では、絶対量子収率Φを、DOLに対してプロットした。この結果、DOLが約3.5の場合が最も明るいことが理解できる。
試験例4:蛍光イメージング
蛍光色素を接合した抗体の退色を以下のように評価した。測定用試料としては、PR4-NHS(実施例6; DOL3.5, 27μg)、PR5-NHS(実施例7; DOL3.1, 20μg)、Cy5.5(比較例1; DOL2.3, 30μg)により接合したヤギ抗マウス抗体IgGを使用した。300Wキセノンランプを使用し、フィルターを使用せず、385~750 nmの波長域で吸収強度を測定し、初期吸収強度が約0.1となるように調整した。各化合物の吸収強度及び蛍光強度について、時間経過にともなう維持率と、吸収スペクトル及び蛍光スペクトルとを図6に示す。図6の上図において、実線はPR4-NHS(実施例6; DOL3.5)、破線はPR5-NHS(実施例7; DOL3.1, 20μg)、一点鎖線はCy5.5(比較例1; DOL2.3)を使用した結果である。さらに、PR4-NHS(実施例6)、PR5-NHS(実施例7)及びCy5.5(比較例1)で標識したヤギ抗マウス抗体IgGを用いて10分間共焦点蛍光イメージングを行った結果を図7に示し、照射領域の平均蛍光強度を図8に示す。図8は細胞を染色した状態での蛍光強度の時間推移を評価した結果である。図8において、実線は(実施例6; DOL3.5, 27μg)、破線はPR5-NHS(実施例7; DOL3.1, 20μg)、一点鎖線はCy5.5(比較例1; DOL2.3, 30μg)を用いた結果である。この結果、細胞を染色した状態においても、本発明のホスファローダミン化合物は、従来の蛍光色素であるCy5.5と比較して、長時間にわたって抗体を蛍光させることができた。

試験例5:植物イメージング
PR5-NHSで標識したオリゴアルギニン(8量体)を用いて、ヒメツリガネゴケ(学名:Physcomitrella patens)の原糸体の蛍光イメージングを行った。
PR5-NHSで標識したオリゴアルギニン(8量体)を用いて、ヒメツリガネゴケ(学名:Physcomitrella patens)の原糸体の蛍光イメージングを行った。
オリゴアルギニンとPR5-NHSの接合を以下のように実施した。担持樹脂として2-クロロトリチルポリスチレンを用いた一般的なFmoc(9-フルオレニルメトキシカルボニル)固相合成法により、8個のアルギニンが連結したオリゴアルギニン(8量体)のペプチド延伸反応を行った。なお、脱Fmoc化はピペリジンのDMF溶液を用い、保護アミノ酸にはFmoc-Arg(Pbf)-OH(N-α-(9-フルオレニルメトキシカルボニル)-N-ω-(2,2,4,6,7-ペンタメチルジヒドロベンゾフラン-5-スルホニル)-L-アルギニン)を用い、縮合剤としてHCTU(O-(6-クロロ-1H-ベンゾトリアゾール-1-イル)-N,N,N’,N’-テトラメチルウロニウムヘキサフルオロホスフェート)を用いた。オリゴアルギニン(8量体)に対して2等量のPR5-NHSのDMF溶液を室温で1時間作用させることで末端アミノ基を蛍光標識し、引き続く樹脂からの切り出しと脱保護を過剰のトリフルオロ酢酸存在を作用させて実施した。粗生成物を逆相HPLC(固定相:Phenomenex Jupiter C18;移動相:0.1% TFA(トリフルオロ酢酸)を含むアセトニトリルと水の混合溶媒)を用いて精製し、得られたフラクションを凍結乾燥することでPR5-NHSで標識したオリゴアルギニン(8量体)を得た。
蛍光イメージングには、蛍光顕微鏡IX-71(Olympus)を使用し汎用の水銀ランプで励起した。蛍光フィルターは励起フィルター、ダイクロイックフィルター、バリアフィルターの順に、PR5-NHS用にはFF01-710/13、FF740-dio1、FF01-785/62(Semrock)を用い、葉緑体内のクロロフィル用にはG365、FT396、LP420(Olympus)を用いた。
結果を図9に示す。画像(a)において観察されるのがPR5-NHSのシグナルであり、画像(b)において粒状に観察されるのが葉緑体内のクロロフィルのシグナルであり、画像(c)は(a)及び(b)の重ね合わせ画像である。画像(a)においてクロロフィルのシグナルが検出されていないことから、本蛍光色素を用いることで、クロロフィルの存在下であっても蛍光色素由来のシグナルのみを特異的に検出できることが示された。
