본 발명은 후박 추출물 및 신이 추출물을 유효성분으로 함유하는 치근 및 치주조직 성장 촉진용, 또는 치주염 예방 또는 치료용 약학적 조성물에 관한 것으로, 구체적으로 후박 추출물 및 신이 추출물의 혼합물은 Raw264.7 세포에서 LPS로 유도된 NO(nitric oxide)의 활성을 억제하고, TRAP(Tartrate-resistant acid phosphatase) 활성을 억제하여 파골세포 분화를 억제하였으며, MC3T3 세포에서 ALP(alkaline phosphatase)활성을 증가시킴으로써 조골세포 분화 촉진을 확인하였다. 또한, 사람 치주인대세포에서 우수한 세포 증식률 및 ALP 활성을 나타냄을 확인하였고, 마우스로부터 적출한 치근 및 지주조직 성장이 촉진되는 것을 확인함으로써, 본 발명의 후박 추출물 및 신이 추출물의 혼합물은 치근/치주조직 성장 촉진용 조성물, 치주염의 예방 또는 치료용 약학적 조성물, 치근/치주조직 성장 촉진용 또는 치주염 개선용 의약외품 조성물, 치근/치주조직 성장 촉진용 또는 치주염 개선용 건강기능식품 및 치근/치주조직 성장 촉진용 또는 치주염 개선용 화장료 조성물로써 유용하게 사용될 수 있다.
Description
본 발명은 후박 추출물(Magnolia
officinlis) 및 신이 추출물(Magnolia flower)의 혼합물을 유효성분으로 함유하는 치주조직 성장 촉진용 및 치주염 예방 또는 치료용 약학적 조성물에 관한 것이다.
치주조직은 치은(잇몸), 백악질, 치주인대 및 치조골로 구성된 연조직과 경조직을 모두 포함하는 복잡한 기관이다. 치은은 치아의 지지와 저작과정에 관여하는 단순 결합조직인 반면에, 치주인대는 치아와 치조골 사이에 위치하여 치아를 악골에 부착 지지하며, 저작력과 같은 다양한 방향의 기계적 스트레스에 저항하고, 또한 감각기능을 하는 고도로 분화된 세포성 결합조직이다. 이러한 치주인대는 치조골측에 골모세포(osteoblast)와 파골세포(osteoclast), 치주인대의 중심측에 섬유모세포(fibroblast), Malassez 상피세포(epithelial rests of Malassez), 대식세포, 미분화 간엽세포, 신경 요소 그리고 혈관내피세포; 치근측에 위치하고 있는 백악모세포(cementoblasts) 등의 다양한 세포들로 구성되어 치조골, 치근 및 백악질의 수복과 재생에 관여하는 것으로 알려져 있다.
치주 질환은 세균에 의해 야기되는 치아 지지조직의 염증상태를 말하며, 치은염 및 치주염으로 분리할 수 있다. 발병원인은 불량한 구강 위생상태로 인하여 구강 세균이 치면세균막을 형성하는데 있다. 치면세균막이란 침에 있는 끈끈한 물질을 접착제로 이용하여 세균이 치아 표면에 달라붙은 후 증식한 세균덩어리를 말한다. 치면세균막은 그냥 방치해 두면, 염증이 생겨 가끔 잇몸에서 피가 나고, 구취가 나는 경우가 있으며 이러한 증상을 치은염이라고 한다.
치은염이 더 진행되면, 치아와 잇몸사이의 벌어진 틈이 더 깊어져서 치주낭이 생기고, 여기에 치주질환을 일으키는 세균들이 번식하여 치주염이 발생한다. 치주염이 진행되면 칫솔질과 같은 약한 자극에도 잇몸에서 피가 나기도 하며 붓고, 종종 급성염증으로 변화되어 통증을 유발한다. 이러한 염증은 골을 만드는 기능은 저하시키고, 골을 흡수하는 작용이 높아져서 치조골이 점점 낮아지게 되어 치조골이 파괴되고 결국 치아를 상실하게 된다. 진행된 치주염은 치아 상실을 일으키는데 40세 이후의 성인에 있어서 치아를 뽑게 되는 원인 중 치주염이 차지하는 비율이 40%나 된다는 보고가 있을 정도로 치주염으로 인한 치아상실은 심각한 문제이다.
치주질환에서 치주인대와 치조골의 파괴는 이들 조직의 주성분인 교원질(collagen)의 분해에 의한 것이다. 이에 대한 정확한 기전(mechanism)은 아직 밝혀져 있지는 않지만, 교원질의 분해는 숙주가 분비하는 콜라게나아제와 치주질환 유발 세균으로부터 분비된 중성 단백질 분해 효소에 의한 것으로 알려져 있다. 따라서 콜라게나아제 효소 및 단백질 분해 효소의 활성을 억제함으로써 치주질환의 진행을 억제하려는 연구가 시도되고 있다.
치주질환의 치료는 환자의 개선된 구강위생의 확립, 비외과적 혹은 외과적인 치석제거술, 치근활택술, 치은소파술과 신부착을 응용한 치주조직의 재생술들이 이용되어져 왔다. 그러나 이런 외과적인 치료 방법은 치료를 위해 치과를 가야 하는 번거로움으로 인해 효과적인 치료가 어렵고 또한 병의 예방보다는 병이 어느정도 진행되었을 경우 행하는 치료에 국한되어 있어, 치료를 하지 않을 경우 대부분이 만성으로 진행되는 경우가 대부분이다. 부가적인 치료로 전신적인 항생제의 복용과 국소 서방형 제재가 사용되어 왔으나, 불필요한 부위에도 약물이 너무 많이 전달되어 그로 인한 부작용과 최근 사용되는 항생제에 대한 내성을 나타내는 치주질환균이 분리된 예가 보고되고 있어 심각한 문제점을 안고 있다. 치주질환으로 인하여 치아가 상실될 경우, 보철치료나 치과 임플란트 치료 등으로 인하여 환자의 부담이 급증하게 되며, 특히, 사회경제적 취약 계층에게 더욱 높은 진료 장벽이 될 수 있다. 따라서, 치아 상실에 이르기 전 손상된 조직을 재생할 수 있다면 치아 상실에 따르는 인공치아 또는 보철과 같은 치료의 수요를 줄여 경제적 사회적 부담을 경감시킬 수 있다.
외과적 치료의 한계성과 항생제 사용의 문제점을 보완하고 치주질환 예방 및 치료효과를 향상시키기 위해서는 파괴 및 소실된 치주조직을 원상회복시키는 효능을 지닌 제제의 개발이 절실히 요구되고 있다. 치주조직은 구성 조직 특성이 모두 다르기에 치주질환으로 파괴된 병소를 재생하기 위한 치료는 아직까지 예지성이 있는 결과를 보여주는 것이 많지 않다. 치태, 치석과 같은 염증성 결합조직에 대한 비외과적 혹은 외과적 제거 치료를 통하여 치주 질환은 치유단계에 접어들게 되지만, 상실된 치주 조직은 이러한 전통적인 치료방식을 통해서는 치주 조직 개개 구성요소들의 재생이 아닌 상피조직의 증식으로 불완전한 치유의 결과를 가져오게 된다.
골재형성에는 크게 두 종류의 세포가 관여하는 것으로 알려져 있다. 두 세포 중 하나는 뼈를 생성하는 조골세포(osteoblast)이고, 다른 하나는 뼈를 파괴하는 파골세포(osteoclast)이다. 이들은 서로 다른 세포에서 유래하는데 간엽줄기세포(mesenchymal stem cell)가 분화되어 조골세포(osteoblast), 근원세포(myoblast), 지방세포(adipocyte), 섬유아세포(fibroblast), 연골세포(chondrocyte) 등의 세포로 분화되고, 조혈모세포(hematopoietic stem cell)가 분화되어 파골세포(osteoclast), 대식세포(macrophage), T 세포(T cell), B 세포(B cell), 수지상세포(dentritic cell), 호염구(basophile), 호산구(eosinophile), 호중구(neutrophile), 혈소판(platelet), 적혈구(Red blood cell) 등의 세포로 분화된다. 조골세포 주요 분화인자인 Runx2(Runt-related transcription factor 2), 근원세포 주요 분화인자인 MyoD, 지방세포 주요 분화인자인 PPARγ(peroxisome proliferator-activated receptors γ), 연골세포 주요 분화인자인 Sox9 등은 서로의 발현을 억제조절하며 분화를 유도한다(Yamashita S. et al., Exp Cell Res., 315(13), pp.2231-40, 2009, Charles A. et al., Exp Cell Res., 300(2), pp.406-17, 2004, David V. et al., Endocrinology., 148(5), pp.2553-62, 2007, HillTP. et al., Dev Cell., 8(5), pp.727-38, 2005, Wang H. et al., J Bone Miner Res., 23(6), pp.939-48, 2008, Lengner CJ. et al., J Biol Chem., 280(16), pp.15872-9, 2005). 또한 연골세포 분화의 주요 인자인 Sox9은 조골세포의 성숙과 분화를 억제시키고, 조골세포의 분화인자인 Wnt는 Runx2를 촉진시키나 Sox9은 분해시켜 억제시키며 조골세포의 분화인자인 osterix는 연골세포의 분화를 억제한다(Dy P. et al., Dev Cell., 13;22(3), pp.597-609, 2012, Gunil I. et al., 대한류마티스학회지, 15(1), 2008, Tominaga H. et al., J Bone Miner Metab., 27(1), pp.36-45, 2009).
조골세포는 RANKL(Receptor activator of NF-κB ligand)과 이것의 유도 수용체(decoy receptor)인 OPG(Osteoprotegerin)를 생성한다. RANKL이 파골 전구세포(osteoclast progenitor cells) 표면에 있는 수용체인 RANK(Receptor activator of NF-κB)에 결합하면 파골 전구 세포가 거대 다핵 파골세포로 성숙화(maturation)되어 골 흡수(bone resorption)가 일어난다. 그러나 OPG가 RANKL과 결합하면 RANKL과 RANK간 결합이 차단되어 파골세포의 형성이 억제되고 필요 이상의 골 흡수가 일어나지 않게 된다(Theill LE. et al., Annu Rev Immunol., 20, pp.795-823, 2002; Wagner EF. et al., Curr Opin Genet Dev., 11, pp.527-532, 2001). 이러한 파골세포의 활성으로 오래된 뼈의 흡수 또는 파괴가 이루어지며 이는 뼈에 구멍을 내어 적은 양의 칼슘이 혈류로 방출되어 신체기능을 유지하는데 사용된다(William J. et al., Nature., 423, pp.337342, 2003).
한편, 뼈세포에서 생성된 조골세포는 교원질로 구멍을 채우고 칼슘과 인의 침척물(hydroxyapatite)을 덮어서 단단한 새로운 뼈를 만들어 골격을 재건한다(Stains JP. et al., Birth Defects Res C Embryo Today., 75(1), pp.72-80, 2005). 뼈가 파괴되기 시작하여 다시 새로운 뼈로 재형성 되기까지는 약 100일 정도 걸린다(Schwarz EM. et al., Curr Opin Orthop., 11, pp.329-335, 2000). 유아에서는 1년 내에 뼈의 칼슘이 100% 바뀌지만 성인에서는 매년 골격의 약 10~30%가 이런 과정을 통하여 재형성되며, 파골속도와 조골속도가 동일해야만 전과 같은 골밀도를 유지할 수 있다. 이러한 두 과정 즉, 골흡수와 골형성은 서로 밀접하게 연결되어 이루어지고 골의 정상적인 구조를 유지하는 데 중요하다.
염증은 교원질, 칼슘 및 인의 침적물로 단단한 뼈를 만드는 조골세포의 골을 만드는 기능을 저하시키고, 파골세포의 뼈를 파괴 또는 흡수하는 작용이 높아져서 치조골이 점점 낮아지고 파괴되어 결국 치아를 상실하게 된다. 이때, 파골세포는 골수 내의 단핵구/대식세포 계통의 세포로부터 유래하고, 이 단핵전구세포는 혈액 내로 순환되며 골내막층에서 전구세포들이 증식되어 다핵세포를 형성하기 위해 융합된다고 알려져 있으며, 파골세포에는 특징적으로 타르트레이트(tartrate)에 대해 저항성을 나타내는 산성포스파타제인 TRAP을 가지며 이는 다른 골조직 세포와 구별할 수 있는 파골세포의 세포화학적 표지효소로 이용된다고 알려져 있다(대한민국 제 10-2006-0092651호).
치주인대(Periodontal ligament)는 평균 0.2mm 정도 두께의 층이며, 치아에 부착되어 치아를 치조골에 부착하고 지지하는 역할을 한다. 또한 치주인대는 치아가 음식을 씹을 때 치조골에 힘을 분산시켜 완충 작용을 하고, 치아의 감각을 치조골에 전달한다. 치주인대에는 미분화 간엽세포가 존재하고 있기 때문에 전체적인 구조와 기능이 변화되지 않으면서 지속적으로 골개조를 계속할 수 있다. 치주인대 내의 미분화 간엽세포는 치조골쪽으로 이동하여 치조골을 개조시키거나, 치아쪽으로 이동하여 교원섬유를 생산하여 치아를 더욱 단단히 지지하기도 한다(대한민국 제 10-2016-0016127호).
신이(Magnolia flower)는 목련과(Magnoliaceae)에 속하는 목련꽃의 봉우리를 이르는 말로 꽃봉우리가 약간 매운 맛이 나기 때문에 신이(辛夷)라고 한다. 한국에서는 목련과의 백목련(Magnolia denudata Desrousseaux) 또는 동속식물의 꽃봉우리를 말려서 사용하며, 일본에서는 백목련(M. denudata Desr.), 망춘화(M. flower Pamp., 望春花), 일본신이(M. kobus DC., 日本辛夷), 향신이(M. salicifolia Maxim.:香辛夷) 및 무당 옥란(M. sprengeri Pamp.: 武當玉蘭)을 사용하며, 중국에서는 백목련(M. denudata Desr), 망춘화(M. flower Pamp., 望春花) 및 무당옥란(M. sprengeri Pamp., 武當玉蘭)을 사용한다. 신이에 대해 한방에서는 그 성질을 따뜻하다고 보며 두통, 요통 및 비염에 효능이 있고 또한 축농증을 치료해주고 진통제의 약성이 있다고 보고되어 있다.
후박(Magnolia
officinlis
)은 목련과 식물인 후박나무 Magnolia officinalis Rehd. et Wils.의 줄기나 뿌리껍질을 말린 것이다. 봄에 20년이상 자란 나무의 껍질을 벗겨 그늘에서 말리거나 끓는 물에 잠깐 담갔다가 건져서 햇볕에 말린 다음 이것을 다시 증기에 쪄서 원통 모양으로 말아 햇볕에서 말려서 사용한다. 약리 실험에서 항균 작용, 약한 이뇨 작용을 나타낸다는 것이 밝혀졌으며 한방에서는 소화 장애, 구토, 설사, 위장염, 위경련, 기침이 나고 숨이 찬 데, 기관지염, 기관지 천식등에 효과가 있는 것으로 알려져 있다.
미국 출원특허 US 13/739,340호는 골형성(osteogenesis)을 촉진하는 약학적 조성물에 관한 것으로, 목련과 식물의 잎 또는 껍질 등에서 수득한 추출물이 부작용은 없으면서 골밀도 및 골성장을 촉진하는 효과가 있음을 개시하고 있다. 또한, 대한민국 등록특허 10-0204165호는 후박 추출물을 함유하는 골 조직 재생 촉진 조성물에 관한 것으로, 후박 추출물이 절골, 파골, 쇄골 등의 손상된 뼈의 재생을 촉진시키는 효과가 있음을 개시하고 있을 뿐, 후박 추출물 및 신이 추출물의 혼합물의 치주조직 성장 촉진용 및 치주염 치료 기능에 관해서는 보고된 바가 없다.
반면, 치아 형성은 상기 골형성 과정과 달리, 태생 6주에 발생이 시작되면, 표면의 상피가 안으로 함입해 들어오고, 함입한 내부의 결합조직에서 세포들이 증식하기 시작하며, 상피 쪽에서 함입된 세포들이 점점 자라면서 아래에서는 치수의 조직이 형성된다. 상피 쪽 세포들은 나중의 치아의 겉을 싸는 법랑아세포로 분화되고, 내부에서는 상아질을 형성하는 조상아세포와 여러 가지 치수 세포들이 분화된다. 이와 같이, 뼈 형성과 치아 형성은 관여하는 세포 및 기전이 전혀 상이함을 알 수 있다.
이에, 본 연구자들은 치주염으로 인한 치주조직의 손실을 막고, 치주조직의 재생을 촉진할 수 있는 물질을 연구하던 중, 신이 추출물 및 후박 추출물의 혼합물이 파골세포 분화를 억제하고, 조골세포를 활성화 시키며, 치근 및 지주조직 성장을 촉진하는 효능이 있음을 확인하여 본 발명을 완성하였다.
본 발명의 목적은 본 발명은 후박(Magnolia
officinlis) 추출물 및 신이(Magnolia flower) 추출물의 혼합물을 유효성분으로 함유하는 치근/치주조직 성장 촉진용 조성물, 치주염 예방 또는 치료용 약학적 조성물, 치근/치주조직 성장 촉진용 또는 치주염 개선용 의약외품 조성물, 치근/치주조직 성장 촉진용 또는 치주염 개선용 건강기능식품 및 치근/치주조직 성장 촉진용 또는 치주염 개선용 화장료 조성물을 제공하는 것이다.
상기 목적을 달성하기 위하여, 본 발명은 후박 추출물 및 신이 추출물의 혼합물을 유효성분으로 함유하는 치근 또는 치주조직 성장 촉진용, 또는 치주염 예방 또는 치료용 약학적 조성물을 제공한다.
또한, 본 발명은 후박 추출물 및 신이 추출물의 혼합물을 유효성분으로 함유하는 치근 또는 치주조직의 성장 촉진용, 또는 치주염의 예방 또는 개선용 의약외품 조성물을 제공한다.
또한, 본 발명은 후박 추출물 및 신이 추출물의 혼합물을 유효성분으로 함유하는 치근 또는 치주조직의 성장 촉진용, 또는 치주염의 개선용 건강기능식품을 제공한다.
아울러, 본 발명은 후박 추출물 및 신이 추출물의 혼합물을 유효성분으로 함유하는 치근 또는 치주조직의 성장 촉진용, 또는 치주염의 개선용 화장료 조성물을 제공한다.

이하, 본 발명을 상세히 설명한다.
본 발명은, 후박 추출물 및 신이 추출물의 혼합물을 유효성분으로 함유하는 치주조직 성장 촉진용, 또는 치주염 예방 또는 치료용 약학적 조성물을 제공한다.
상기 혼합물은 후박 추출물 및 신이 추출물이 1:1 내지 1:50의 질량비로 혼합되는 것이 바람직하며, 1:3 내지 1:30의 질량비로 혼합되는 것이 더욱 바람직하나 이에 한정되지 않는다.
상기 혼합물은 NO(nitric oxide) 활성을 억제하고, TRAP(Tartrate-resistant acid phosphatase) 활성을 억제하며, ALP(alkaline phospatase) 활성을 증가시키는 것이 바람직하나 이에 한정되지 않는다.
본 발명의 구체적인 실시예에서, Raw.264.7 세포에 후박 추출물 또는 신이 추출물 각각의 단독처리, 또는 후박 추출물 및 신이 추출물을 1:3, 1:10 또는 1:30 비율로 혼합한 혼합물을 처리하였을 때, 후박 추출물 및 신이 추출물의 혼합물이 LPS(lipopolysaccharide)에 의해 증가된 NO(nitric oxide) 활성을 억제하며(도 1 참조), RANKL 처리에 의해 증가한 Raw.264.7의 TRAP(Tartrate-resistant acid phosphatase) 활성을 후박 추출물 및 신이 추출물의 혼합물이 효과적으로 낮추는 것을 관찰하여(도 2 참조), 파골세포 분화를 억제하는 효과가 있음을 확인하였으며, 상기 파골세포 분화 억제 효과는 각 추출물의 단독 처리에 비교했을 때, 후박 추출물 및 신이 추출물을 1:10 및 1:30의 농도비로 혼합한 혼합물에서 가장 효과적인 파골세포 분화 억제 효과를 보이는 것을 확인하였다(도 1 및 2 참조).
또한, 본 발명자들은 MC3T3-E1 세포에서 ALP(alkaline phosphatase) 활성을 억제하는 것을 확인하여 조골세포 분화를 촉진하는 효과가 있음을 확인하였고(도 3 참조), 상기 조골세포 분화 촉진 효과는 각 추출물의 단독 처리에 비교했을 때, 후박 추출물 및 신이 추출물을 1:10의 농도비로 혼합한 혼합물에서 가장 효과적인 조골세포 분화 촉진 효과를 보이는 것을 확인하였다.
또한, 본 발명자들은 후박 추출물 및 신이추출물이 사람 치주인대세포 증식및 세포 활성도 증가, 그리고 분화 정도에 미치는 영향을 확인하였으며, 그 결과 후박추출물 또는 신이추출물이 단독으로 존재할 때에 비해 복합물이 1:10의 비율로 존재할 때에 현저하게 높은 세포활성도를 나타내었다(도 4 참조). 이 결과는 후박추출물과 신이추출물을 혼합한 경우에 두 가지 추출물이 함께 작용하여 상승효과를 나타내었기 때문인 것으로 확인되었다.
또한, 본 발명자들은 후박추출물 및 신이추출물 복합물이 ALP 활성을 증가시키는지 여부를 확인하였으며, 그 결과 후박추출물 및 신이추출물의 혼합비율 1:10의 혼합물 처리에서 신이추출물을 단독으로 처리하였을 때보다 ALP 활성이 약 40% 이상 증가된 것을 확인하였다(도 5 참조).
또한, 본 발명자들은 마우스 아래턱 어금니를 후박 추출물 및 신이 추출물을 포함하고 있는 아가로스 비드에 포매한 뒤, 숙주마우스에 이식하여 3주 후, 치근 및 치주조직 성장을 확인한 결과, 대조군에서는 치근 및 뼈의 성장이 거의 관찰되지 않았고, 후박 추출물 또는 신이 추출물 단독 처리시 치근 및 치주조직의 성장이 미미하였으나, 후박 추출물 및 신이 추출물의 혼합물의 처리는 치주조직 성장을 증가시켰으며 혼합물 농도비율 1:10에서 치근성장과 이식된 치아 주위의 치조골 형성이 가장 우수한 것을 확인하였다(도 6 참조).
따라서, 본 발명의 후박 추출물 및 신이 추출물의 혼합물은 치근 또는 치주조직 성장 촉진용 및 치주염 예방 또는 치료용 약학적 조성물로써 유용하게 사용될 수 있다.
본 발명의 조성물은 조성물 총 중량에 대하여 본 발명의 후박 추출물 및 신이 추출물의 혼합물을 0.1 내지 99.9 중량%를 유효성분으로 함유하고, 약제학적으로 허용 가능한 담체, 부형제 또는 희석제를 포함할 수 있다.
약제학적으로 허용 가능한 담체는 식염수, 멸균수, 링거액, 완충 식염수, 덱스트로스 용액, 말토 덱스트린 용액, 글리세롤, 에탄올 및 이들 성분 중 1 성분 이상을 혼합하여 이용할 수 있으며, 필요에 따라 항산화제, 완충액, 정균제 등 다른 통상의 첨가제를 첨가할 수 있다. 또한, 희석제, 분산제, 계면활성제, 결합제 및 윤활제를 부가적으로 첨가하여 수용액, 현탁액, 유탁액 등과 같은 주이용 제형, 산제, 정제, 캡슐제, 환, 과립 또는 주사액제로 제제화 할 수 있다. 더 나아가 당 분야의 적정한 방법으로 또는 Remington's Pharmaceutical Science(Mack Publishing Company, Easton PA, 18th, 1990)에 개시되어 있는 방법을 이용하여 각 질환에 따라 또는 성분에 따라 바람직하게 제제화할 수 있다.
본 발명의 구강질환 예방 및 치료용 약학적 조성물은, 통상적인 방법에 의해 정제, 캅셀제, 산제, 과립제, 현탁제, 유제, 시럽제, 기타 액제로 제형화될 수 있다. 구체적으로 본 발명의 약학적 조성물은 경구 투여용 제형, 예를 들면 정체, 트로치제(troches), 로젠지(lozenge), 수용성 또는 우성현탁액, 조제분말 또는 과립, 에멀젼, 하드 또는 소프트 캡슐, 시럽 또는 엘릭시르제(elixirs)로 제제화된다. 정제 및 캡슐 등의 제형으로 제제하기 위해 락토오스, 사카로오스, 솔비톨, 만니톨, 전분, 아밀로펙틴, 셀룰로오스 또는 젤라틴과 같은 결합제, 디칼슘 포스페이트와 같은 부형제, 옥수수 전분 또는 고구마 전분과 같은 붕해제, 스테아르산 마르네슘, 스테아르산 칼슘, 스테아릴푸마르산 나트륨 또는 폴리에틸렌글리콜 왁스와 같은 윤활유가 함유된다. 캡슐제형의 경우는 상기에서 언급한 물질 이외에도 지방유와 같은 액체 담체를 함유한다.
또한, 본 발명의 약학적 조성물은 경구 또는 비경구 투여할 수 있으며, 비경구 투여시 피하주사, 정맥주사, 근육내 주사 또는 흉부내 주사 주입방식을 선택하는 것이 바람직하다. 비경구 투여용 제형으로 제제화하기 위해서는 본 발명의 화합물 1과 함께 물에서 혼합하여 현탁액으로 제조하고 이를 앰플 또는 바이알의 단위 투여형으로 제제한다.
본 발명에 따른 유효성분의 투여량은 체내에서 활성성분의 흡수도, 불활성화율 및 배설속도, 환자의 연령, 성별 및 상태, 치료할 질병의 중증 정도에 따라 적절히 선택되나, 경구 투여제의 경우 일반적으로 성인에게 1일에 체중 1 kg당 본 발명의 화합물1을 0.0001 ~ 500 mg의 양으로 1회 내지 수회 나누어 투여할 수 있으며, 0.001 ~ 100 mg의 양으로 투여하는 것이 바람직하다. 본 발명의 조성물은 단독으로, 또는 수술, 방사선 치료, 호르몬 치료, 화학 치료 및 생물학적 반응 조절제를 사용하는 방법들과 병용하여 사용할 수 있다.
또한, 본 발명은 후박 추출물 및 신이 추출물의 혼합물을 유효성분으로 함유하는 치근 또는 치주조직의 성장 촉진용, 또는 치주염의 예방 또는 개선용 의약외품 조성물을 제공한다.
상기 의약외품 조성물은 치약, 구강청정제 또는 구강세정제등의 구강용 조성물 제형인 것이 바람직하나 이에 한정되지 않는다.
상기 혼합물은 후박 추출물 및 신이 추출물이 1:1 내지 1:50의 농도비로 혼합되는 것이 바람직하며, 1:3 내지 1:30의 농도비로 혼합되는 것이 더욱 바람직하나 이에 한정되지 않는다.
상기 혼합물은 NO(nitric oxide) 활성을 억제하고, TRAP(Tartrate-resistant acid phosphatase) 활성을 억제하며, ALP(alkaline phospatase) 활성을 증가시키는 것이 바람직하다.
본 발명의 구체적인 실시예에서, 후박 추출물 및 신이 추출물의 혼합물이 LPS(lipopolysaccharide)에 의해 증가된 NO(nitric oxide) 활성을 억제하며(도 1 참조), RANKL 처리에 의해 증가한 Raw.264.7의 TRAP(Tartrate-resistant acid phosphatase) 활성을 효과적으로 낮추어(도 2 참조), 파골세포 분화를 억제하는 효과가 있음을 확인하였으며, 후박 추출물 및 신이 추출물을 1:10 및 1:30의 농도비로 혼합한 혼합물에서 가장 효과적인 파골세포 분화 억제 효과를 보이는 것을 확인하였다(도 1 및 2 참조).
또한, 본 발명자들은 MC3T3-E1 세포에서 ALP(alkaline phosphatase) 활성을 억제하는 것을 확인하여 조골세포 분화를 촉진하는 효과가 있음을 확인하였고(도 3 참조), 후박 추출물 및 신이 추출물을 1:10의 농도비로 혼합한 혼합물에서 가장 효과적인 조골세포 분화 촉진 효과를 보이는 것을 확인하였다.
또한, 본 발명자들은 후박 추출물 및 신이추출물이 사람 치주인대세포 증식및 세포 활성도 증가, 그리고 분화 정도에 미치는 영향을 확인하였으며, 그 결과 후박추출물 또는 신이추출물이 단독으로 존재할 때에 비해 복합물이 1:10의 비율로 존재할 때에 현저하게 높은 세포활성도를 나타내었다(도 4 참조). 이 결과는 후박추출물과 신이추출물을 혼합한 경우에 두 가지 추출물이 함께 작용하여 상승효과를 나타내었기 때문인 것으로 확인되었다.
또한, 본 발명자들은 후박추출물 및 신이추출물 복합물이 ALP 활성을 증가시키는지 여부를 확인하였으며, 그 결과 후박추출물 및 신이추출물의 혼합비율 1:10의 혼합물 처리에서 신이추출물을 단독으로 처리하였을 때보다 ALP 활성이 약 40% 이상 증가된 것을 확인하였다(도 5 참조).
또한, 본 발명자들은 마우스 아래턱 어금니를 후박 추출물 및 신이 추출물을 포함하고 있는 아가로스 비드에 포매한 뒤, 숙주마우스에 이식하여 3주 후, 치근 및 치주조직 성장을 확인한 결과, 치주조직 성장을 증가시켰으며 혼합물 농도비율 1:10에서 치근성장과 이식된 치아 주위의 치조골 형성이 가장 우수한 것을 확인하였다(도 6 참조).
따라서, 본 발명의 후박 추출물 및 신이 추출물의 혼합물은 치근 또는 치주조직 성장 촉진용 또는 치주염 개선용 의약외품 조성물로써 유용하게 사용될 수 있다.
본 발명에서 제공하는 후박 추출물 및 신이 추출물의 혼합물을 함유하는 의약외품 조성물은 그 제형에 있어 특별히 한정되지 않고 통상의 제형을 가질 수 있으며, 구체적으로는 치약, 구강세정제 또는 구강청정제 등의 제형을 가질 수 있다. 본 발명에서 제공하는 구강 조성물은 그 제형에 따라 제제화에 필요한 각종의 기제와 첨가물을 함유할 수 있으며, 이들 성분의 종류와 양은 당업자에 의해 용이하게 선정될 수 있다. 예를 들면, 구강용 조성물의 제형이 치약류인 경우, 연마제, 습윤제, 기포제, 결합제, 감미제, pH조절제, 방부제, 약효성분, 향료, 증백제, 색소, 용제 등을 첨가하여 제조할 수 있다.
또한, 본 발명은 후박 추출물 및 신이 추출물의 혼합물을 유효성분으로 함유하는 치근 또는 치주조직의 성장 촉진용, 또는 치주염의 개선용 건강기능식품을 제공한다.
상기 혼합물은 후박 추출물 및 신이 추출물이 1:1 내지 1:50의 농도비로 혼합되는 것이 바람직하며, 1:3 내지 1:30의 농도비로 혼합되는 것이 더욱 바람직하나 이에 한정되지 않는다.
상기 혼합물은 NO(nitric oxide) 활성을 억제하고, TRAP(Tartrate-resistant acid phosphatase) 활성을 억제하며, ALP(alkaline phospatase) 활성을 증가시키는 것이 바람직하나 이에 한정되지 않는다.
본 발명의 구체적인 실시예에서, 후박 추출물 및 신이 추출물의 혼합물이 LPS(lipopolysaccharide)에 의해 증가된 NO(nitric oxide) 활성을 억제하며(도 1 참조), RANKL 처리에 의해 증가한 Raw.264.7의 TRAP(Tartrate-resistant acid phosphatase) 활성을 효과적으로 낮추어(도 2 참조), 파골세포 분화를 억제하는 효과가 있음을 확인하였으며, 후박 추출물 및 신이 추출물을 1:10 및 1:30의 농도비로 혼합한 혼합물에서 가장 효과적인 파골세포 분화 억제 효과를 보이는 것을 확인하였다(도 1 및 2 참조).
또한, 본 발명자들은 MC3T3-E1 세포에서 ALP(alkaline phosphatase) 활성을 억제하는 것을 확인하여 조골세포 분화를 촉진하는 효과가 있음을 확인하였고(도 3 참조), 후박 추출물 및 신이 추출물을 1:10의 농도비로 혼합한 혼합물에서 가장 효과적인 조골세포 분화 촉진 효과를 보이는 것을 확인하였다.
또한, 본 발명자들은 후박 추출물 및 신이추출물이 사람 치주인대세포 증식및 세포 활성도 증가, 그리고 분화 정도에 미치는 영향을 확인하였으며, 그 결과 후박추출물 또는 신이추출물이 단독으로 존재할 때에 비해 복합물이 1:10의 비율로 존재할 때에 현저하게 높은 세포활성도를 나타내었다(도 4 참조). 이 결과는 후박추출물과 신이추출물을 혼합한 경우에 두 가지 추출물이 함께 작용하여 상승효과를 나타내었기 때문인 것으로 확인되었다.
또한, 본 발명자들은 후박추출물 및 신이추출물 복합물이 ALP 활성을 증가시키는지 여부를 확인하였으며, 그 결과 후박추출물 및 신이추출물의 혼합비율 1:10의 혼합물 처리에서 신이추출물을 단독으로 처리하였을 때보다 ALP 활성이 약 40% 이상 증가된 것을 확인하였다(도 5 참조).
또한, 본 발명자들은 마우스 아래턱 어금니를 후박 추출물 및 신이 추출물을 포함하고 있는 아가로스 비드에 포매한 뒤, 숙주마우스에 이식하여 3주 후, 치근 및 치주조직 성장을 확인한 결과, 치주조직 성장을 증가시켰으며 혼합물 농도비율 1:10에서 치근성장과 이식된 치아 주위의 치조골 형성이 가장 우수한 것을 확인하였다(도 6 참조).
따라서, 본 발명의 후박 추출물 및 신이 추출물의 혼합물은 치근 또는 치주조직 성장 촉진용 또는 치주염 개선용 건강기능식품으로써 유용하게 사용될 수 있다.
본 발명의 건강기능식품은 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 상술한 천연 탄수화물은 포도당, 과당과 같은 모노사카라이드, 말토스, 슈크로스와 같은 디사카라이드 및 덱스트린, 사이클로덱스트린과 같은 폴리사카라이드, 자일리톨, 소르비톨, 에리트리톨 등의 당알콜이다. 감미제로서는 타우마틴, 스테비아 추출물과 같은 천연 감미제나 사카린, 아스파르탐과 같은 합성 감미제 등을 사용할 수 있다. 상기 천연 탄수화물의 비율은 본 발명의 건강기능식품 100 중량부당 0.01 ~ 0.04 중량부, 구체적으로는 약 0.02 ~ 0.03 중량부 범위에서 선택할 수 있다.
상기 외에 본 발명의 건강기능식품은 여러 가지 영양제, 비타민, 전해질, 풍미제, 착색제, 펙트산 및 그의 염, 알긴산 및 그의 염, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알콜, 탄산 음료에 사용되는 탄산화제 등을 함유할 수 있다. 이러한 성분은 독립적으로 또는 조합하여 사용할 수 있다. 이러한 첨가제의 비율은 크게 중요하진 않지만 본 발명의 건강식품 100 중량부 당 0.01 ~ 0.1 중량부의 범위에서 선택되는 것이 일반적이다.
상기 식품의 종류에는 특별한 제한은 없다. 상기 물질을 첨가할 수 있는 식품의 예로는 육류, 소세지, 빵, 쵸코렛, 캔디류, 스넥류, 과자류, 피자, 라면, 기타 면류, 껌류, 아이스크림류를 포함한 낙농제품, 각종 스프, 음료수, 차, 드링크제, 알콜 음료 및 비타민 복합제 등이 있으며, 통상적인 의미에서의 건강식품을 모두 포함한다.
아울러, 본 발명은 후박 추출물 및 신이 추출물의 혼합물을 유효성분으로 함유하는 치근 또는 치주조직의 성장 촉진용, 또는 치주염의 개선용 화장료 조성물을 제공한다.
상기 혼합물은 후박 추출물 및 신이 추출물이 1:1 내지 1:50의 농도비로 혼합되는 것이 바람직하며, 1:3 내지 1:30의 농도비로 혼합되는 것이 더욱 바람직하나 이에 한정되지 않는다.
상기 혼합물은 NO(nitric oxide) 활성을 억제하고, TRAP(Tartrate-resistant acid phosphatase) 활성을 억제하며, ALP(alkaline phospatase) 활성을 증가시키는 것이 바람직하나 이에 한정되지 않는다.

또한, 본 발명자들은 MC3T3-E1 세포에서 ALP(alkaline phosphatase) 활성을 억제하는 것을 확인하여 조골세포 분화를 촉진하는 효과가 있음을 확인하였고(도 3 참조), 후박 추출물 및 신이 추출물을 1:10의 농도비로 혼합한 혼합물에서 가장 효과적인 조골세포 분화 촉진 효과를 보이는 것을 확인하였다.
또한, 본 발명자들은 후박 추출물 및 신이추출물이 사람 치주인대세포 증식및 세포 활성도 증가, 그리고 분화 정도에 미치는 영향을 확인하였으며, 그 결과 후박추출물 또는 신이추출물이 단독으로 존재할 때에 비해 복합물이 1:10의 비율로 존재할 때에 현저하게 높은 세포활성도를 나타내었다(도 4 참조). 이 결과는 후박추출물과 신이추출물을 혼합한 경우에 두 가지 추출물이 함께 작용하여 상승효과를 나타내었기 때문인 것으로 확인되었다.
또한, 본 발명자들은 후박추출물 및 신이추출물 복합물이 ALP 활성을 증가시키는지 여부를 확인하였으며, 그 결과 후박추출물 및 신이추출물의 혼합비율 1:10의 혼합물 처리에서 신이추출물을 단독으로 처리하였을 때보다 ALP 활성이 약 40% 이상 증가된 것을 확인하였다(도 5 참조).
또한, 본 발명자들은 마우스 아래턱 어금니를 후박 추출물 및 신이 추출물을 포함하고 있는 아가로스 비드에 포매한 뒤, 숙주마우스에 이식하여 3주 후, 치근 및 치주조직 성장을 확인한 결과, 치주조직 성장을 증가시켰으며 혼합물 농도비율 1:10에서 치근성장과 이식된 치아 주위의 치조골 형성이 가장 우수한 것을 확인하였다(도 6 참조).
따라서, 본 발명의 후박 추출물 및 신이 추출물의 혼합물은 치근 또는 치주조직 성장 촉진용 또는 치주염 개선용 화장료 조성물로써 유용하게 사용될 수 있다.
상기 화장료 조성물은 로션, 연고, 겔, 크림, 패치 또는 분무제 등이 있으나 여기에 국한되는 것은 아니다. 본 발명의 후박 추출물 및 신이 추출물의 혼합물을 함유하는 화장료 조성물을 제조함에 있어서, 통상적으로 함유되는 화장료 조성물에 본 발명의 후박 추출물 및 신이 추출물의 혼합물이 3 내지 30 중량부, 바람직하게는 5 또는 20 중량부로 첨가할 수 있다.
본 발명의 화장료 조성물에는 본 발명의 후박 추출물 및 신이 추출물의 혼합물에 추가로 지방 물질, 유기 용매, 용해제, 농축제 및 겔화제, 연화제, 항산화제, 현탁화제, 안정화제, 발포제(foaming agent), 방향제, 계면활성제, 물, 이온형 또는 비이온형 유화제, 충전제, 금속이온봉쇄제 및 킬레이트화제, 보존제, 비타민, 차단제, 습윤화제, 필수 오일, 염료, 안료, 친수성 또는 친유성 활성제, 지질 소낭 또는 화장료 조성물에 통상적으로 사용되는 임의의 다른 성분과 같은 피부 과학 분야에서 통상적으로 사용되는 보조제를 함유할 수 있다. 또한 상기 성분들은 피부 과학 분야에서 일반적으로 사용되는 양으로 도입될 수 있다.
이하, 본 발명을 실시예 및 실험예에 의해서 상세히 설명한다.
단 하기 실시예 및 실험예는 본 발명을 예시하기 위한 것일 뿐, 본 발명이 하기 실험예에 의해서 한정되는 것은 아니다.

실시예
1> 신이 추출물의 제조
본 발명자들은 중국산 신이(일본 오사카 천해당에서 구입) 꽃봉오리를 이의 중량 10배에 해당하는 에탄올을 첨가하여 3-7일 동안 추출하였다. 이후, 추출액을 여과하여 불용성 성분인 잔사를 제거하였고, 여과된 추출액은 감압농축기를 이용하여 농축하였다.
농축된 추출액을 동결건조하여 고상의 신이 추출물 300 g을 수득하였다.

실시예
2> 후박 추출물의 제조
본 발명자들은 일본 오사카 천해당에서 구입한 후박나무 껍질(1 kg)을 이의 중량 10배에 해당하는 에탄올을 첨가하여 3-7일 동안 추출하였다. 이후, 추출액을 여과하여 불용성 성분인 잔사를 제거하였고, 여과된 추출액은 감압농축기를 이용하여 농축하였다.
농축된 추출액을 동결건조하여 고상의 후박추출물 200 g을 수득하였다.

실시예
3> 후박 추출물 및 신이 추출물의 혼합물 제조
본 발명자들은 상기 <실시예 1> 및 <실시예 2>에서 얻은 분말 형태의 신이 추출물 및 후박 추출물을 DMSO에 용해하여 (후박추출물 0.5, 1, 3 mg/ml, 신이추출물 1.5, 3, 5, 10, 15 ,30 mg/ml)의 농도로 제조하였다.
하기 <실험예 1>에 사용하기 위한 후박 추출물 및 신이 추출물의 혼합물 제조를 위해, 상기 제조된 후박 추출물 및 신이 추출물의 농도비를 1:3(후박 추출물 1 ㎍/ml + 신이 추출물 3 ㎍/ml), 1:10(후박 추출물 1 ㎍/ml + 신이 추출물 10 ㎍/ml) 또는 1:30(후박 추출물 1 ㎍/ml + 신이 추출물 30 ㎍/ml)으로 제조하였다.
또한, 하기 <실험예 2>에 사용하기 위한 후박 추출물 및 신이 추출물의 혼합물 제조를 위해, 후박 추출물 및 신이 추출물의 농도비를 1:3(후박 추출물 0.5 ㎍/ml + 신이 추출물 1.5 ㎍/ml), 1:10(후박 추출물 0.5 ㎍/ml + 신이 추출물 5 ㎍/ml) 또는 1:30(후박 추출물 0.5 ㎍/ml + 신이 추출물 15 ㎍/ml)으로 제조하였다.
또한, 하기 <실험예 3>에 사용하기 위한 후박 추출물 및 신이 추출물의 혼합물 제조를 위해, 후박 추출물 및 신이 추출물의 농도비를 1:1(후박 추출물 3 ㎍/ml+신이 추출물 3 ㎍/ml), 1:3(후박 추출물 3 ㎍/ml+신이 추출물 10 ㎍/ml) 또는 1:10(후박 추출물 3 ㎍/ml+신이 추출물 30 ㎍/ml)으로 제조하였다.

실험예
1> 후박 추출물 및 신이 추출물의 혼합물이 nitric oxide(NO) 생성에 미치는 영향 확인
<1-1> 후박 추출물 및 신이 추출물 처리에 따른 세포 생존율 확인
본 발명자들은 후박 추출물 및 신이 추출물의 혼합물의 세포독성을 확인하고자 상기 <실시예 1> 내지 <실시예 3>에서 제조한 후박 추출물 또는 신이 추출물, 그리고 이의 혼합물을 Raw 264.7 세포에 하기와 같이 처리하는 실험을 수행하였다.
구체적으로, Raw 264.7 세포를 96웰 배양접시 3 X 10³세포/웰의 농도로 접종한 후, 37℃, 5% CO₂에서 4시간 배양했다. 여기에 0.01 ㎍/ml의 LPS를 처리하고, 상기 <실시예 2>에서 얻은 1 mg/ml 농도의 후박 추출물 (배지 50 ㎕에 1 ㎍/ml의 농도가 되도록 0.05 ㎕), 상기 <실시예 1>에서 얻은 3, 10, 30 mg/ml 농도의 신이 추출물을 각각 (신이 3, 10, 30 ㎍/ml의 신이추출물의 농도가 되도록 배지 50 ul에 각각 0.05 ul로), 또는 상기 <실시예 3>에서 제조한 후박 추출물 및 신이 추출물 1:3, 1:10 및 1:30의 농도비로 혼합한 혼합물 (배지 50 ㎕에 후박 및 신이추출물 0.05 ㎕)를 처리한 후 4일 동안 배양하였다. Cell Counting kit(Dojindo)를 10 ㎕/웰 처리한 후 microplate reader를 이용하여 450/620 nm에서 흡광도를 측정하고 대조군의 흡광도와 비교하여 세포생존율을 백분율로 환산하였다.
그 결과, 후박 및 신이 추출물 처리군은 대조군과 세포 생존율의 유의한 차이를 보이지 않아, 세포독성이 없음을 확인하였다.

본 발명자들은 후박 추출물 및 신이 추출물의 혼합물이 NO 생성에 미치는 영향을 확인하고자 하기와 같은 실험을 하였다.
구체적으로, Raw 264.7 세포를 96웰 배양접시에 3 X 10³세포/웰의 농도로 접종한 후 37℃, 5% CO₂에서 4시간 동안 배양하고, 0.01 ㎍/ml의 LPS를 처리하고, 상기 <실시예 2>에서 얻은 1 mg/ml 농도의 후박 추출물 (배지 50 ul에 1 ㎍/ml의 농도가 되도록 0.05 ul)ml, 상기 <실시예 1>에서 얻은 3, 10, 30 mg/ml 농도의 신이 추출물을 각각 (신이 3, 10, 30 ㎍/ml의 신이추출물의 농도가 되도록 배지 50 ul에 각각 0.05 ul로), 또는 상기 <실시예 3>에서 제조한 후박 추출물 및 신이 추출물 1:3, 1:10 및 1:30의 농도비로 혼합한 혼합물 (배지 50 ㎕에 후박 및 신이추출물 0.05 ㎕)를 처리한 후 24시간 동안 배양하였다. 배양액 50 ㎕를 취하여 같은 양의 Griess Reagent(Promega)를 넣어주고 10분간 상온에 반응시킨 후 ELISA 분석기로 540 nm에서 흡광도를 측정하였다.
그 결과, 도 1에 나타낸 바와 같이 후박 추출물 및 신이 추출물의 1:30의 농도 비율로 처리한 세포에서 LPS에 의해 증가한 NO의 생성 억제 활성이 가장 큰 것을 확인하였다. 후박 추출물 1 ㎍/ml 및 신이 추출물 3 및 10 ㎍/ml의 농도에서는 LPS에 의해 증가한 NO를 억제하지 못하였고, 신이 추출물 30 ㎍/ml 처리시 NO 활성이 약 30% 억제하는 것을 확인하였다.
또한, 후박 추출물 및 신이 추출물의 혼합 처리시 NO 활성이 유의적으로 억제되었으며, 후박 및 신이 추출물 1:3 비율 혼합물 처리군은 신이 추출물 3 ㎍/ml 처리에 비하여 약 10%, 후박 및 신이 추출물 1:30 비율 혼합물 처리군은 신이 추출물 30 ㎍/ml에 비하여 약 20%의 NO 활성 억제 효과를 보였지만, 후박 및 신이 추출물 1:10 비율 혼합물 처리군은 신이 추출물 10 ㎍/ml 처리군에 비하여 약 50%의 NO 활성 억제 효과를 보여, 농도비율 1:10의 혼합 효과가 가장 크며, 10 ㎍/ml 농도의 신이 추출물이 포함된 후박 추출물 및 신이 추출물의 혼합물의 제조가 경제성이 있을 것으로 판단된다.

실험예
2> 후박 추출물 및 신이 추출물의 혼합물의 파골세포 분화 억제 효과 확인
<2-1> 후박 추출물 및 신이 추출물 처리에 따른 세포생존율 확인
본 발명자들은 후박 추출물 및 신이 추출물의 혼합물의 세포독성을 확인하고자 상기 <실시예 1> 내지 <실시예 3>에서 제조한 후박 추출물 또는 신이 추출물, 그리고 이의 혼합물을 Raw 264.7 세포에 하기와 같이 처리하는 실험을 수행하였다.
구체적으로, Raw 264.7 세포를 96 웰 배양접시 3 X 10³세포/웰의 농도로 접종한 후, 37℃, 5% CO₂에서 4시간 배양했다. 여기에 50 mg/ml의 RANKL을 (배지 50ul에 50 ㎍/ml의 농도가 되도록 0.05 ul)로 처리하고, 상기 <실시예 2>에서 얻은 0.5 mg/ml 농도의 후박 추출물, 상기 <실시예 1>에서 얻은 1.5, 5, 15 mg/ml 농도의 신이 추출물을 각각 (0.5 ㎍/ml농도의 후박, 1.5, 5, 15 ㎍/ml의 신이추출물의 농도가 되도록 배지 50 ul에 각각 0.05 ul로) 처리 또는 상기 <실시예 3>에서 제조한 후박 추출물 및 신이 추출물 1:3, 1:10 및 1:30의 농도비로 혼합한 혼합물(배지 50 ㎕에 후박 및 신이추출물 0.05 ㎕)를 처리한 후 4일 동안 배양하였다. Cell Counting kit(Dojindo)를 10 ㎕/웰 처리한 후 microplate reader를 이용하여 450/620 nm에서 흡광도를 측정하고 대조군의 흡광도와 비교하여 세포생존율을 백분율로 환산하였다.
그 결과, 후박 및 신이 추출물 처리군은 대조군과 세포 생존율의 유의한 차이를 보이지 않아, 세포독성이 없음을 확인하였다.
<2-2> 후박 추출물 및 신이 추출물의 혼합물의 TRAP(

-resistant acid phosphatase) 억제 활성 확인
본 발명자들은 후박 추출물 및 신이 추출물의 혼합물이 파골세포 분화에 미치는 효과를 확인하기 위하여, Raw 264.7 세포에 RANKL을 처리하여 파골세포 분화를 유도하고, 후박 추출물, 신이 추출물을 또는 후박 추출물 및 신이 추출물의 혼합물을 처리한 후, 파골세포 특이 유전자인 TRAP(Tartrate-resistant acid phosphatase)활성을 측정하였다.
구체적으로, Raw 264.7 세포를 96 웰 배양접시 3 X 10³세포/웰의 농도로 접종한 후 37℃, 5% CO₂에서 4시간 배양한 후, RANKL을 (배지 50 ㎕에 50 ㎍/ml의 농도가 되도록 0.05 ul)로 처리하고, 상기 <실시예 2>에서 얻은 0.5 mg/ml 농도의 후박 추출물, 상기 <실시예 1>에서 얻은 1.5, 5, 15 mg/ml 농도의 신이 추출물을 각각 (0.5 ㎍/ml농도의 후박, 1.5, 5, 15 ㎍/ml의 신이추출물의 농도가 되도록 배지 50 ul에 각각 0.05 ul로) 처리, 또는 상기 <실시예 3>에서 제조한 후박 추출물 및 신이 추출물 1:3, 1:10 및 1:30의 농도비로 혼합한 혼합물 (배지 50 ㎕에 후박 및 신이추출물 0.05 ㎕)를 처리한 후 4일 동안 배양하였다. 배지를 제거하고 10% 포르말린용액을 각 웰당 50 ㎕씩 첨가하여 25분 동안 고정시켰다. 10% 포르말린용액을 제거한 후 100 ㎕의 100% 에탄올로 세척한 뒤, 1일 동안 plate를 건조시킨 후 p-Nitrophenyl phosphate를 1.36 mg/ml의 농도로 함유한 TRAP assay 완충액을 100㎕/웰씩 처리하였다. 1시간 동안 반응시킨 뒤 반응액 85 ㎕를 취하여 동량의 0.1N NaOH와 혼합하여 새로운 배양 접시에서 반응시킨 뒤, 405/620 nm에서 흡광도를 측정하였다.
그 결과, 도 2에 나타낸 바와 같이 후박 추출물 0.5 ㎍/ml 및 신이 추출물 1.5 및 5 ㎍/ml의 농도에서는 TRAP 억제 활성을 나타내지 않았으나, 15 ㎍/ml의 신이 추출물 처리시 약 20%의 TRAP 억제효능을 나타내었다. 그러나 후박 추출물과 신이 추출물을 1:3, 1:10, 1:30의 농도비율로 혼합하여 처리하였을 때 신이 추출물 15 ㎍/ml 단독 처리보다도 TRAP 억제 효과가 더 뛰어난 것을 확인하였다(도 2).
후박 추출물 및 신이 추출물의 1:3 비율 혼합물 처리군은 신이 추출물 1.5 ㎍/ml 처리에 비하여 약 25%, 후박 추출물 및 신이 추출물의 1:30 비율 혼합물 처리군은 신이 추출물 30 ㎍/ml에 비하여 약 30%의 TRAP 활성 억제 효과를 보여 주었으나, 후박 추출물 및 신이 추출물의 1:10 비율 혼합물 처리군은 신이 추출물 5 ㎍/ml 처리군에 비하여 약 50%의 TRAP 활성 억제 효과를 보여주어 농도비율 1:10의 혼합 효과가 가장 크며, 10 ㎍/ml 농도의 신이 추출물이 포함된 후박 추출물 및 신이 추출물의 혼합물의 제조가 경제성이 있을 것으로 판단된다.
또한, 후박 추출물 0.5 ㎍/ml 또는 신이 추출물 1.5 또는 5 ㎍/ml의 단독 처리에 의하여 TRAP 활성이 억제되지 않았으나, 후박 추출물 및 신이 추출물의 1:3(후박 0.5 ㎍/ml+신이 1.5 ㎍/ml) 또는 1:10(후박 0.5 ㎍/ml+신이 5 ㎍/ml) 혼합 처리에 의하여 TRAP 활성이 억제되었고, 1:30의 비율이 가장 큰 파골세포 분화 억제 효과를 나타내었으며, 후박 및 신이 추출물의 혼합처리가 파골세포 억제에 대해 시너지효과를 가지는 것을 확인하였다. 특히, 1:10 혼합처리에 의해 약 50%의 TRAP 활성 억제효과를 보이며, 1:30 비율의 혼합처리와 크게 차이나지 않아, 후박 추출물 및 신이 추출물의 혼합비율 1:10 에서 혼합으로 인한 파골세포 분화 억제 효과가 뛰어남을 확인하였다(도 2).

실험예
3> 후박 추출물 및 신이 추출물의 혼합물의 조골세포 분화 유도 효과 확인
후박 추출물 및 신이 추출물의 혼합물의 조골세포 분화 유도 효과를 확인하기 위해 MC3T3-E1 세포에서 조골세포 분화의 표지자인 alkialine phosphatase(ALP) 활성을 측정하였다.
구체적으로, MC3T3-E1 세포를 96웰 배양접시에 5 X 10³세포/웰 농도로 접종한 후 37℃, 5% CO₂에서 2일간 배양하였다. 상기 <실시예 2>에서 얻은 3 mg/ml 농도의 후박 추출물, 상기 <실시예 1>에서 얻은 3, 10, 30 mg/ml 농도의 신이 추출물, 또는 상기 <실시예 3>에서 제조한 후박 추출물 및 신이 추출물 1:1, 1:3 및 1:10의 농도비로 혼합한 혼합물을 각각 50 ㎕/웰로 처리한 후 분화유도인자를 포함한 배양배지를 150 ㎕/웰으로 배지를 교환한 후 4일간 배양하였다. 배지를 제거하고 100㎕의 100% 메탄올로 세척한 후 1일 동안 배양접시를 건조한 뒤 p-Nitrophenyl phosphate를 2.47 mg/ml의 농도로 함유한 ALP assay buffer를 100 ㎕/웰으로 처리하여 1시간 반응시킨 후 microplate reader를 이용하여 405/620 nm에서 흡광도를 측정하였다.
그 결과, 도 3에 나타낸 바와 같이 후박 추출물 3 ㎍/ml 또는 신이 추출물 3, 10, 30 ㎍/ml을 단독으로 처리하였을 때 신이 추출물 30 ㎍/ml에서 대조군에 비해 ALP 활성이 약 20% 증가하였고, 후박 추출물 및 신이 추출물의 혼합물을 처리하였을 때, 혼합비율 1:10의 후박 추출물 및 신이 추출물 혼합물 처리에서 신이 추출물을 단독으로 처리(30 ㎍/ml)하였을 때 보다 ALP 활성이 약 50% 이상 증가되었다. 따라서, 후박 추출물 및 신이 추출물의 혼합물 중 특히 1:10 비율의 혼합물은 조골세포의 분화를 촉진하는 것을 확인하였다(도 3).

본 발명자들은 상기 <실시예 1> 내지 <실시예 >에서 수득한 후박 추출물 및 신이추출물이 사람 치주인대세포 증식 및 세포 활성도 증가 그리고 분화 정도에 미치는 영향을 알아보기 위해, 하기와 같은 실험들을 실시하였다.
구체적으로, 건강한 사람의 교정치료 목적으로 발거한 소구치의 치주인대 부분을 큐렛(curret)을 이용하여 무균적으로 긁어 배양접시에 올려놓은 후 10% FBS(Fetal bovine serum)을 첨가한 배양액을 넣어 배양하였다. 3일 간격으로 배양액을 교환해 주면서 5계대 까지 배양하여 실험에 사용하였다. 배양시 습도는 95%, 온도 37C, 5% CO2를 계속 공급하여 배양하였다. 치주인대세포 증식효과를 확인하기 위해 96 well plate에 3X103 세포/웰 접종한 후 2일간 배양하여 100% Conflency 시켰다. 2일 후 100% Confluency 된 세포에 상기 <실시예 2>에서 얻은 1 mg/ml 농도의 후박추출물, 상기 <실시예 1>에서 얻은 1, 3, 10 mg/ml의 농도의 신이추출물(1 ㎍/ml의 농도의 후박, 1, 3, 10 ㎍/ml의 신이추출물의 농도가 되도록 배지 50 ㎕에 각각 0.05 ㎕로 처리)또는 <상기예 3>에서 제조한 후박추출물 및 신이추출물 1:1, 1:3 및 1:10의 농도비로 혼합한 혼합물(50 ㎕)을 넣어 총 200 ㎕가 되게 하여 4일간 배양하였다. 배양이 끝난 후 PBS에 용해한 MTT(methyl thiazol-2-YL-2,5-diphenyl tetraolium bromide, sigma)용액 20 ㎕를 각 well에 넣고 4시간 동안 배양한 후 MTT 용액을 제거하고 formazon 결정을 용해시키기 위해 각 well에 100 ㎕씩 첨가하였다. Plate를 잘 흔든 후 microplate reader를 이용하여 405/620 nm에서 흡광도를 측정하였다.
그 결과, 도 4에 나타낸 바와 같이 후박추출물 또는 신이추출물이 단독으로 존재할 때에 비해 복합물이 1:10의 비율로 존재할 때에 현저하게 높은 세포활성도를 나타내었다(도 4). 이 결과는 후박추출물과 신이추출물을 혼합한 경우에 두 가지 추출물이 함께 작용하여 상승효과를 나타내었기 때문인 것으로 확인되었다.

ALP activity 확인
ALP(Alkaline phosphatase, ALP)는 치주인대세포 분화의 초기 지표로 알려져 있다. 따라서, 본 발명자들은 후박 및 신이추출물 복합물이 ALP 활성을 증가시키는지 여부를 평가하였다.
구체적으로, 배양된 세포를 96 well plate 각 well에 3X103 cells/well의 농도로 접종한 후 37C, 5% CO2에서 2일간 배양하였다. 100% Confluency 된 세포에 상기 <실시예 2>에서 얻은 1 mg/ml 농도의 후박추출물, 상기 <실시예 1>에서 얻은 1, 3, 10 mg/ml의 농도의 신이추출물(1 ㎍/ml의 농도의 후박, 1, 3, 10 ㎍/ml의 신이추출물의 농도가 되도록 배지 50 ㎕에 각각 0.05 ㎕로 처리) 또는 <상기예 3>에서 제조한 후박추출물 및 신이추출물 1:1, 1:3 및 1:10의 농도비로 혼합한 혼합물(50 ㎕)과 분화유도인자((10 mM beta-glycerophosphate(Sigma), 50 ㎍/ml L-ascorbic acid(Sigma))를 포함한 배양배지 150 ㎕/well 배지를 교환한 후 4일간 배양하였다. 배지를 제거하고 100 ㎕의 100% 메탄올로 세척한 후 1일 동안 배양접시를 건조한 뒤 p-Nitrophenyl phosphate를 2.47 mg/ml의 농도로 함유한 ALP assay buffer를 100 ㎕/well로 처리하여 1시간 반응시킨 후 microplate reader를 이용하여 405/620 nm에서 흡광도를 측정하였다.
그 결과, 도 5에 나타낸 바와 같이 후박 추출물 1 ㎍/ml 또는 신이추출물 1, 3, 10 ㎍/ml을 단독으로 처리하였을 때 신이추출물 10 ㎍/ml에서 대조군에 비해 ALP 활성이 약 10% 증가하였고, 후박추출물 및 신이추출물의 혼합물을 처리하였을 때, 혼합비율 1:10의 후박추출물 및 신이추출물 혼합물 처리에서 신이추출물을 단독으로 처리하였을 때보다 ALP 활성이 약 40% 이상 증가된 것을 확인하였다(도 5). 따라서, 후박추출물 및 신이추출물의 혼합물 중 특히 1:10의 비율의 혼합물은 치주인대세포의 분화를 촉진하는 것을 확인하였다.

실험예
5> 후박 추출물 및 신이 추출물의 혼합물의 치근 및 치주조직 성장 촉진 효과 확인
후박 추출물 및 신이 추출물의 혼합물의 치근 및 치주조직 성장 촉진 효과를 확인하기 위하여 후박 추출물, 신이 추출물 또는 후박 추출물 및 신이 추출물의 혼합물을 마우스로부터 적출한 치아의 치수에 하기와 같이 처리하는 실험을 수행하였다.
구체적으로, 1 ml의 PBS에 5 mg/ml 후박 추출물 (5 ㎍/ml 되도록 1 ㎕ 첨가) 및 1 ml의 PBS에 15, 50 mg/ml 신이 추출물(15, 50 ㎍/ml 되도록 1ul), 그리고 후박 추출물 및 신이 추출물의 혼합 비율이 1:3 또는 1:10의 비율이 되도록 PBS에 희석하여 준비하였다. 그리고 4개의 아가로스 비드(Affi-Gel Blue Gel, #153-7301, Bio-Rad Laboratories Inc.)를 PBS로 세척한 뒤 상기 후박 추출물, 신이 추출물 또는 후박 추출물 및 신이 추출물의 혼합물이 비드에 흡수되도록 최소 60분 동안 반응시켰다. 대조군으로 다른 4개의 비드는 PBS에만 반응시켰다. 그 후, 5일령의 C57BL/6 마우스(대한바이오링크)를 희생시켜 아래턱(Lower jaws)에서 어금니(Molar)를 적출하고, 적출한 치아의 치수를 후박 추출물, 신이 추출물 또는 후박 추출물 및 신이 추출물의 혼합물을 포함하고 있는 아가로스 비드에 포매(Embedding)하였다. 대조군으로는 적출한 치아의 치수를 PBS를 포함한 아가로스 비드에 포매하였다. 아가로스 비드는 8주령의 C57BL/6 숙주마우스 신장의 캡슐에 각각 이식(Graft)하였고, 숙주 마우스는 12시간 명암주기, 실온, 자유섭식 조건에서 사육하였고, 이식 3주 후에 희생시켜 신장을 꺼내고 치아를 관찰하였다. 상기 치아의 3D 이미지는 마이크로 CT로 촬영하였다.
그 결과, 도 6에 나타낸 바와 같이 대조군(PBS)에서는 치근(dental root) 및 뼈의 성장이 거의 관찰되지 않았고, 후박 추출물 또는 신이 추출물 단독 처리시 치근 및 치주조직의 성장이 미미하였으나, 후박 추출물 및 신이 추출물의 혼합물을 처리하였을 경우 치주조직 성장이 증가되었으며, 1:3 비율에 비하여 1:10의 비율의 농도에서 치근 성장과 이식된 치아 주위에 치조골이 새롭게 형성이 더욱 증가되어 있는 것을 관찰하였다. 따라서, 후박 추출물 또는 신이 추출물 단독 처리보다 상기 두 추출물을 혼합하여 처리하였을 때, 치근 및 치주조직 형성 촉진 효과가 뛰어나며, 특히, 후박 추출물:신이 추출물의 혼합 비율이 1:10에서 치근 및 치주조직 형성 촉진 효과가 가장 우수한 것을 확인하였다.
이하, 본 발명의 후박 추출물 및 신이 추출물의 혼합물을 활성성분으로 함유하는 약학적 조성물, 의약외품 조성물, 및 화장료 조성물을 위한 제조예를 예시한다.

Claims (9)
- 후박 추출물 및 신이 추출물의 혼합물을 유효성분으로 함유하는 치근 또는 치주조직의 성장 촉진용, 또는 치주염의 예방 또는 치료용 약학적 조성물.
- 제 1항에 있어서, 상기 혼합물은 후박 추출물 및 신이 추출물이 1:1 내지 1:50의 질량비로 혼합되는 것을 특징으로 하는 조성물.
- 제 1항에 있어서, 상기 혼합물은 NO(nitric oxide) 활성 및 TRAP(Tartrate-resistant acid phosphatase) 활성을 억제하며, ALP(alkaline phospatase) 활성을 증가시키는 것을 특징으로 하는 조성물.
- 후박 추출물 및 신이 추출물의 혼합물을 유효성분으로 함유하는 치근 또는 치주조직의 성장 촉진용, 또는 치주염의 예방 또는 개선용 의약외품 조성물.
