(6) 体液検体及び標準体液検体中の複数の基準miRNAの存在量測定値の代表値が、それぞれ、複数の基準miRNAの存在量測定値の平均値又は中央値である、(1)~(5)のいずれか1項に記載の方法。
(7) 前記測定工程は、体液検体及び標準体液検体中の1又は複数の基準miRNAの存在量測定値を補正することを含み、補正済みの測定値を用いて以後の工程が実施される、(1)~(6)のいずれか1項に記載の方法。
(8) 前記測定工程は、支持体上に固定化された前記1又は複数の基準miRNAを捕捉するためのプローブと、体液検体及び標準体液検体から抽出され標識物質で標識された核酸試料とをそれぞれ接触させてハイブリダイゼーションを行なうことにより、体液検体及び標準体液検体中の当該1又は複数の基準miRNAの存在量を測定することを含む、(1)~(7)のいずれか1項に記載の方法。
(9) 前記測定工程は、体液検体中の前記1又は複数の基準miRNAの存在量の測定と同時に、当該体液検体中の標的miRNAの存在量を測定することを含む、(1)~(8)のいずれか1項に記載の方法。
(10) 前記測定工程は、体液検体中の標的miRNAの存在量測定値を補正することを含む、(9)記載の方法。
(11) 前記測定工程は、支持体上に固定化された、標的miRNAを捕捉するためのプローブ及び前記1又は複数の基準miRNAを捕捉するためのプローブと、体液検体から抽出され標識物質で標識された核酸試料とを接触させてハイブリダイゼーションを行なうことにより、体液検体中の標的miRNA及び当該1又は複数の基準miRNAの存在量をそれぞれ測定することを含む、(9)又は(10)記載の方法。
(12) 前記体液検体が、血液、血清又は血漿である、(1)~(11)のいずれか1項に記載の方法。
(13) 体液検体由来のmiRNAの品質を評価するために、1又は複数のコンピュータに、
体液検体及び標準体液検体より調製された、miRNAを含むRNAサンプルを用いてそれぞれ測定された、配列番号1~12で示される塩基配列からなるmiRNAから選択される1又は複数の基準miRNAの体液検体及び標準体液検体中の存在量測定値を取得する、測定値取得工程;
体液検体中の1又は複数の基準miRNAの存在量測定値又はその代表値を、標準体液検体中の1又は複数の基準miRNAの存在量測定値又はその代表値と比較して、体液検体及び標準体液検体間での1又は複数の基準miRNAの存在量測定値又はその代表値の差又は比を得る、比較工程;及び
前記比較工程で得られた、1又は複数の基準miRNAの存在量測定値又はその代表値の差又は比に基づいて、体液検体由来のmiRNAの品質の良否を判定する、判定工程
を実行させるためのプログラム。
(14) (13)記載のプログラムを記録した、コンピュータ読み取り可能な記録媒体。
(15) 配列番号1~12で示される塩基配列からなるmiRNAから選択される1又は複数の基準miRNAを捕捉するためのプローブが固定化された支持体を含む、miRNA品質評価用チップ。
(16) 標的miRNAを捕捉するためのプローブ及び配列番号1~12で示される塩基配列からなるmiRNAから選択される1又は複数の基準miRNAを捕捉するためのプローブが固定化された支持体を含む、miRNA発現解析用チップ。
本発明では、配列番号1~12で示される塩基配列からなるmiRNAから選択される1又は複数の基準miRNAの体液検体中の存在量、及び、配列番号1~12で示される塩基配列からなるmiRNAから選択される1又は複数の基準miRNAの標準体液検体中の存在量の測定を行う。また、体液検体中の基準miRNAの存在量の測定と同時に、後述する標的miRNAの存在量や補正用標準核酸の存在量の測定を行ってもよい。
本発明において、測定工程で得られた標的miRNA及び基準miRNAの存在量の測定値をそのまま後述する比較工程、判定工程で用いてもよいが、例えば、体液検体中の標的miRNAの発現解析を行う場合には、標的miRNAの測定値及び基準miRNAの測定値の補正を行ない、補正後の測定値を発現量とし、この発現量を用いて比較工程及び判定工程を実施してよい。つまり、測定工程は、標的miRNA及び基準miRNAの測定値を補正する処理を含んでいてもよい。
本発明の比較工程は、前記測定工程で得られた体液検体中の1又は複数の基準miRNAの存在量測定値又はその代表値を、標準体液検体中の1又は複数の基準miRNAの存在量測定値又はその代表値と比較して、両検体中の各基準miRNAの存在量測定値又はその代表値の差又は比を得る工程である。ここで、基準miRNAの存在量測定値又はその代表値の差又は比は、典型的には、次の式で求められる差又は比をいう。

本発明の判定工程は、比較工程で得られた体液検体中の1又は複数の基準miRNAの存在量測定値又はその代表値と、前記標準体液検体中の1又は複数の基準miRNAの存在量測定値又はその代表値との差又は比に基づいて、体液検体に含まれるmiRNAの品質の良否を判定する工程である。miRNAの品質の良否の判定は、体液検体と標準体液検体に含まれる1又は複数の基準miRNAの存在量測定値又はその代表値の差又は比について、品質の良否を判定するための基準とする閾値を予め設定し、その閾値を超えるか否かによって品質の良否(良又は不良)を判定することができる。すなわち、上記式I又は式IIで得られる基準miRNAの存在量測定値又はその代表値の差又は比が、予め任意に定めた閾値を超える場合に、体液検体に含まれるmiRNAの品質が良であると判定し、当該基準miRNAの存在量測定値又はその代表値の差又は比が閾値以下である場合に、体液検体に含まれるmiRNAの品質を不良であると判定することができる。あるいは、上記式I'又は式II'で得られる差又は比を用いる場合には、当該差又は比が予め任意に定めた基準とする閾値を下回る場合に、体液検体に含まれるmiRNAの品質が良であると判定し、当該差又は比が該閾値以上である場合に、体液検体に含まれるmiRNAの品質を不良であると判定することができる。

あるいは、式5Aに示すように、体液検体中の基準miRNAの存在量測定値eと非分解内因性miRNAの存在量測定値cとの差(存在量差e-c)、及び、標準体液検体中の基準miRNAの存在量測定値Eと非分解内因性miRNAの存在量測定値Cとの差(存在量差E-C)を求め、これら2つの各存在量差の比が閾値t5を上回る場合に、体液検体に含まれるmiRNAの品質を良と判定することができる。この場合の閾値t4及びt5は、好ましくは0.7であり、より好ましくは0.8である。
式4A、式5Aは、存在量測定値の比に基づく判定に該当する。

あるいは、式7Aに示すように、体液検体中の基準miRNAの存在量測定値eと非分解内因性miRNAの存在量測定値cとの差(存在量差e-c)、及び、標準体液検体中の基準miRNAの存在量測定値Eと非分解内因性miRNAの存在量測定値Cとの差(存在量差E-C)を求め、これら2つの各存在量差の差が閾値t7を上回る場合に、体液検体に含まれるmiRNAの品質を良と判定することができる。閾値t6及びt7は、例えば-50以上0以下の範囲で設定することができ、例えば0であり得る。
式6A、式7Aは、存在量測定値の差に基づく判定に該当する。

あるいは、閾値t9として、標準体液検体中の基準miRNAの存在量測定値Eと非分解内因性miRNAの存在量測定値Cとの差(存在量差E-C)を採用してもよく、その場合は、式9Aに示すように、体液検体中の基準miRNAの存在量測定値eと非分解内因性miRNAの存在量測定値cとの差(存在量差e-c)が閾値t9、すなわち標準体液検体中の基準miRNAの存在量測定値Eと非分解内因性miRNAの存在量測定値Cとの差(存在量差E-C)を上回る場合に、体液検体に含まれるmiRNAの品質を良と判定することができる。これは、式7Aを採用する場合において、閾値t7を0とする場合に相当する。従って、式9Aは式7Aの一態様であり、存在量測定値の差に基づく判定に該当する。

体液検体及び標準体液検体より調製された、miRNAを含むRNAサンプルを用いてそれぞれ測定された、配列番号1~12で示される塩基配列からなるmiRNAから選択される1又は複数の基準miRNAの体液検体及び標準体液検体中の存在量測定値を取得する、測定値取得工程;
体液検体中の1又は複数の基準miRNAの存在量測定値又はその代表値を、標準体液検体中の1又は複数の基準miRNAの存在量測定値又はその代表値と比較して、体液検体及び標準体液検体間での1又は複数の基準miRNAの存在量測定値又はその代表値の差又は比を得る、比較工程;及び
前記比較工程で得られた、1又は複数の基準miRNAの存在量測定値又はその代表値の差又は比に基づいて、体液検体由来のmiRNAの品質の良否を判定する、判定工程
を実行させるための(すなわち、1又は複数のコンピュータに上記各工程を実行させる命令を含む)プログラム、及び該プログラムを記録したコンピュータ読み取り可能な記録媒体を提供する。
(DNAマイクロアレイ)
東レ株式会社製の“3D-Gene” human miRNA oligo chip(miRBase release 21対応)を用いて以下の実験を行なった。
健常ヒト3名よりそれぞれ採血し、血清を調製した。得られた血清を300μLずつ1名につき6本に分注し、うち5本を4℃に設定した冷蔵庫に静置した。1名につき1本はただちに-80℃に設定した冷凍庫に収容した(0時間)。冷蔵庫に静置した血清検体について、6時間、24時間、48時間、72時間および168時間後にそれぞれ回収して、-80℃に設定した冷凍庫に収容した。-80℃の冷凍庫に収容した検体は、下記に述べるRNA抽出作業まで、そのまま静置した。
上記のように調製され、冷凍庫に静置された血清を同時に溶解し、血清検体に含まれるRNA(以下、検体RNAという。)を抽出した。抽出には、“3D-Gene” RNA extraction reagent from liquid sample kit(東レ社)を用いた。
上記のようにして得られた各血清検体のmiRNA発現量を比較し、静置時間に依存して発現量(検体中存在量)が変動する程度の大きいmiRNAを抽出することで、基準miRNAの選択を行った。
健常ヒト1名より採血し、血清検体を調製した。得られた血清を300μL分注し、ただちに-80℃に設定した冷凍庫に収容した。RNAの抽出には、“3D-Gene” RNA extraction reagent from liquid sample kit(東レ社)を用いた。
品質の判定に用いる基準miRNAとして、hsa-miR-125b-1-3p(配列番号2)の代わりに、hsa-miR-125b-1-3p(配列番号2)とhsa-miR-6798-5p(配列番号9)の2種を用いることに変更し、これ以外は実施例2と同様にして、血清検体由来及び標準体液検体由来のこれら2種のmiRNAの発現量を測定した。
血清検体を、血清を調製後、4℃で静置168時間経過後のものに変更し、これ以外は実施例3と同様にして、血清検体由来及び標準体液検体由来の2種のmiRNA(hsa-miR-125b-1-3p(配列番号2)とhsa-miR-6798-5p(配列番号9))の発現量を測定した。2種のmiRNAの発現量の平均値を代表値とし、代表値から発現量比を求めた。
血清検体を、血清を調製後、4℃で静置72時間経過後のものに変更し、これ以外は実施例3と同様にして、血清検体由来及び標準体液検体由来の2種のmiRNA(hsa-miR-125b-1-3p(配列番号2)とhsa-miR-6798-5p(配列番号9))の発現量を測定した。hsa-miR-125b-1-3pの発現量比とhsa-miR-6798-5pの発現量比をそれぞれ求め、発現量の比較を行なった。
本発明のmiRNAの品質の評価方法を、従来の品質評価方法である電気泳動による方法と比較するため、上記実施例4、5で使用した血清検体を用いて電気泳動による品質の評価を実施した。
品質の判定に用いる基準miRNAとして、hsa-miR-125b-1-3p(配列番号2)の代わりに、hsa-miR-149-3p(配列番号25)を用いることに変更し、また、血清検体を、血清調製後に4℃で静置168時間経過後のものに変更し、これら以外は実施例2と同様にして、血清検体由来及び標準体液検体由来のこれら2種のmiRNAの発現量を測定した。ここで、基準miRNAとして用いたhsa-miR-149-3pは、上記実施例1において、静置時間の経過によっても発現量が変動(低下)せず最も安定していたmiRNAの1つである。
実施例2と同様にして、品質の判定に用いる基準miRNAとして、hsa-miR-125b-1-3p(配列番号2)を用いることに加えて、RNAの分解に依存しない非分解内因性miRNAであるhsa-miR-4463(配列番号26)も利用して、血清検体由来及び標準体液検体由来のこれら2種のmiRNAの発現量を測定した。血清検体と標準体液検体のそれぞれについて、hsa-miR-125b-1-3pの発現量をhsa-miR-4463の発現量で割り返した発現量比を求めた後、血清検体由来の発現量比を標準体液検体由来の発現量比で割り返して発現量比の比を求め、閾値と比較した。
Claims (16)
- 体液検体由来のmiRNAの品質を評価する方法であって、
体液検体及び標準体液検体より調製された、miRNAを含むRNAサンプルを用いて、配列番号1~12で示される塩基配列からなるmiRNAから選択される1又は複数の基準miRNAの、体液検体及び標準体液検体中の存在量をそれぞれ測定する、測定工程;
体液検体中の1又は複数の基準miRNAの存在量測定値又はその代表値を、標準体液検体中の1又は複数の基準miRNAの存在量測定値又はその代表値と比較して、体液検体及び標準体液検体間での1又は複数の基準miRNAの存在量測定値又はその代表値の差又は比を得る、比較工程;及び
前記比較工程で得られた、1又は複数の基準miRNAの存在量測定値又はその代表値の差又は比に基づいて、体液検体由来のmiRNAの品質の良否を判定する、判定工程
を含む、前記方法。 - 前記比較工程は、1の基準miRNAの存在量測定値の差若しくは比、複数の基準miRNAそれぞれの存在量測定値の差若しくは比、又は複数の基準miRNAの存在量測定値の代表値の差若しくは比を得る工程である、請求項1記載の方法。
- 前記判定工程は、前記1若しくは複数の基準miRNAの存在量測定値又はその代表値の差又は比を、基準として予め定められた閾値と対比することを含む、請求項1又は2記載の方法。
- 前記比較工程は、体液検体中の存在量測定値又はその代表値から標準体液検体中の存在量測定値又はその代表値を減算して差を求めること、又は、体液検体中の存在量測定値又はその代表値を標準体液検体中の存在量測定値又はその代表値で除算して比を求めることを含む、請求項1~3のいずれか1項に記載の方法。
- 前記判定工程は、前記1又は複数の基準miRNAの存在量測定値又はその代表値の差又は比を、基準として予め定められた閾値と対比することを含み、該差又は比が該閾値を超える場合に体液検体由来のmiRNAの品質を良と判定する、請求項4記載の方法。
- 体液検体及び標準体液検体中の複数の基準miRNAの存在量測定値の代表値が、それぞれ、複数の基準miRNAの存在量測定値の平均値又は中央値である、請求項1~5のいずれか1項に記載の方法。
- 前記測定工程は、体液検体及び標準体液検体中の1又は複数の基準miRNAの存在量測定値を補正することを含み、補正済みの測定値を用いて以後の工程が実施される、請求項1~6のいずれか1項に記載の方法。
- 前記測定工程は、支持体上に固定化された前記1又は複数の基準miRNAを捕捉するためのプローブと、体液検体及び標準体液検体から抽出され標識物質で標識された核酸試料とをそれぞれ接触させてハイブリダイゼーションを行なうことにより、体液検体及び標準体液検体中の当該1又は複数の基準miRNAの存在量を測定することを含む、請求項1~7のいずれか1項に記載の方法。
- 前記測定工程は、体液検体中の前記1又は複数の基準miRNAの存在量の測定と同時に、当該体液検体中の標的miRNAの存在量を測定することを含む、請求項1~8のいずれか1項に記載の方法。
- 前記測定工程は、体液検体中の標的miRNAの存在量測定値を補正することを含む、請求項9記載の方法。
- 前記測定工程は、支持体上に固定化された、標的miRNAを捕捉するためのプローブ及び前記1又は複数の基準miRNAを捕捉するためのプローブと、体液検体から抽出され標識物質で標識された核酸試料とを接触させてハイブリダイゼーションを行なうことにより、体液検体中の標的miRNA及び当該1又は複数の基準miRNAの存在量をそれぞれ測定することを含む、請求項9又は10記載の方法。
- 前記体液検体が、血液、血清又は血漿である、請求項1~11のいずれか1項に記載の方法。
- 体液検体由来のmiRNAの品質を評価するために、1又は複数のコンピュータに、
体液検体及び標準体液検体より調製された、miRNAを含むRNAサンプルを用いてそれぞれ測定された、配列番号1~12で示される塩基配列からなるmiRNAから選択される1又は複数の基準miRNAの体液検体及び標準体液検体中の存在量測定値を取得する、測定値取得工程;
体液検体中の1又は複数の基準miRNAの存在量測定値又はその代表値を、標準体液検体中の1又は複数の基準miRNAの存在量測定値又はその代表値と比較して、体液検体及び標準体液検体間での1又は複数の基準miRNAの存在量測定値又はその代表値の差又は比を得る、比較工程;及び
前記比較工程で得られた、1又は複数の基準miRNAの存在量測定値又はその代表値の差又は比に基づいて、体液検体由来のmiRNAの品質の良否を判定する、判定工程
を実行させるためのプログラム。 - 請求項13記載のプログラムを記録した、コンピュータ読み取り可能な記録媒体。
- 配列番号1~12で示される塩基配列からなるmiRNAから選択される1又は複数の基準miRNAを捕捉するためのプローブが固定化された支持体を含む、miRNA品質評価用チップ。
- 標的miRNAを捕捉するためのプローブ及び配列番号1~12で示される塩基配列からなるmiRNAから選択される1又は複数の基準miRNAを捕捉するためのプローブが固定化された支持体を含む、miRNA発現解析用チップ。
