Description

細胞の培養方法
 本開示は、塑性流体を用いた細胞の培養方法に関する。
 ヒトiPS(induced pluripotent stem)細胞を用いた再生医療が、CHO(Chinese Hamster Ovary)細胞を用いた抗体医薬生産など、動物細胞を利用した技術の活用が進められている。2次元な平面培養と比べて、細胞を懸濁液状態で培養する3次元培養は単位体積当たりの細胞濃度を高められることで省スペース化や、使用培地量の削減できるなどメリットがある。
 一方で、高い細胞濃度に到達した培養では、細胞の酸素消費速度が大きくなり、培養液中において酸素供給律側が発生する。常温常圧で液中に溶解できる酸素濃度は7-8mg/L程度である。酸素は液中に十分な濃度で保持させることができない栄養成分といえる。
 気相から液相への酸素供給(物質移動)を増大させる方法として、気液接触面の面積を増やすことが挙げられる。微生物のような頑強な細胞の培養では、培地中にスパージャー等を用いて小さく砕かれた泡の通気(バブリング)を行うことができる。しかし、動物細胞一般はシェアストレスへの耐性が十分でなくバブリングが適用できない。
 他にも、泡の発生や、泡と細胞との接触を防ぐような構造を設けた方法がいくつか提案されている。しかしながら、基本的に、動物細胞培養へのバブリングによる通気は実現されていない。
 動物細胞の培養における通気は、主に小型培養槽においては、培養液の気液界面における酸素移動に頼った表面通気法によること、及び、単位体積当たりの気液界面面積を増やすために培地中に疎水性膜チューブを張り巡らす方法によることが一般的である。しかしながら、前者の方法は、培養液量が増加するに従い単位体積当たりの気液界面面積が低下するため、例えば数~数十リットル又はそれ以上などのような、大量の培地体積を用いる培養には適用が難しい。また、後者の方法は、培養槽内が複雑な構造となるため汚染(コンタミネーション)のリスクが高くなることが予想され、実用的でないと考えられる。
 一方、撹拌することなく細胞の浮遊状態を維持して培養できる培地として、特許文献1は、アニオン性官能基を有する高分子化合物又は多糖類を含有する培地組成物を開示する。

 上述したとおり、基本的に、動物細胞培養へのバブリングによる通気は実現されていない。なお、CHO細胞では大型培養槽へのバブリングが行われる事例も知られているが、これはある程度の細胞へのシェアストレスダメージを許容している状態である。再生医療等に用いる細胞の培養は、細胞への物理的ストレスは少ないことが好ましい。また、再生医療用の培地はCHO細胞用の培地に比べて含有するタンパク質の濃度が高く、バブリングによる発泡が問題となる。
 本開示は、一又は複数の実施形態において、物理的ストレスが低減された酸素供給が可能な、細胞の培養方法を提供する。
 本開示は、一態様において、塑性流体である培地中に細胞を配置すること、及び、該培地に気泡を導入することを含む、細胞培養方法に関する。
 本開示にかかる細胞培養方法によれば、一又は複数の実施形態において、細胞に対する物理的ストレスの抑制と、酸素供給の向上とが両立された浮遊細胞培養が可能となる。また、本開示にかかる細胞培養方法によれば、一又は複数の実施形態において、培地体積が大きな浮遊細胞培養においても、細胞に対する物理的ストレスの抑制と、酸素供給の向上との両立が可能となり、物理的ストレスに弱い細胞、例えば、再生医療に使用する細胞や植物細胞など、の大量培養が可能となる。

 本開示は、塑性流体に気泡を導入するという酸素供給方法を組み合わせることで、細胞に対する物理的ストレスの抑制と、酸素供給の向上とが両立された浮遊細胞培養が可能となる、という知見に基づく。
 塑性流体に気泡を導入するという酸素供給方法を組み合わせることで、細胞に対する物理的ストレスの抑制と、酸素供給の向上とが両立された浮遊細胞培養が可能となるメカニズムは、以下にように推測される。
 塑性流体を使用することで、細胞が沈降することなく流体中に保持される。例えば、細胞の浮遊状態が維持され、或いは、細胞を分散させればその分散状態が維持される。これにより、細胞を浮遊させるための撹拌が必要なくなり、物理的ストレスを省くことができる。
 塑性流体中に導入された気泡は、その上昇速度が抑制される。これは、塑性流体における気泡の滞留時間が延長され、ガスホールドアップが増大することを意味する。
 気泡の滞留時間が延長されると、気泡1個当たりの酸素供給量(物質移動)が向上し、通気量を減じることができ、対流や発泡による物理的ストレスが抑制される。
 ガスホールドアップが増大すると、単位培地当たりの見かけの溶存酸素量が増大し、高濃度の細胞培養が可能となる。
 塑性流体中に導入された気泡の移動は、一時的かつ局所的な流動である。気泡の上昇部以外は流動せず、気泡上昇による対流も抑制される。これにより物理的ストレス、例えばシェアストレスなど、が抑制された環境が維持される。
 但し、本開示はこれらのメカニズムに限定して解釈されなくてもよい。
 [塑性流体]
 本開示において、塑性流体とは、流動させるために降伏応力が必要な流体、すなわち、降伏値を持つ流体をいう。塑性流体は、ビンガム流体であってもよく、非ビンガム流体であってもよい。本開示において、塑性流体とは、静置状態において、該塑性流体中の細胞を沈降させずに保持できるものをいう。
 [塑性流体である培地]
 本開示において、塑性流体である培地とは、培地成分と特定化合物とを含有する塑性流体をいい、限定されない一又は複数の実施形態において、液体培地と特定化合物とが混合されて塑性流体となったものが挙げられる。
 前記培地成分及び前記液体培地は、本開示にかかる方法で培養する細胞に応じて通常使用されうる培地成分及び液体培地が使用できる。
 前記特定化合物は、塑性流体ではない液体又は培地を塑性流体とすることができる化合物であり、限定されない一又は複数の実施形態において、WO2014/017513に開示されるものが使用できる。
 [塑性流体とするための特定化合物]
 塑性流体とするための前記特定化合物としては、特に制限されるものではないが、高分子化合物が挙げられ、好ましくはアニオン性の官能基を有する高分子化合物が挙げられる。
 アニオン性の官能基としては、カルボキシ基、スルホ基、リン酸基及びそれらの塩が挙げられ、カルボキシ基またはその塩が好ましい。
 前記高分子化合物は、前記アニオン性の官能基の群より選択される1種又は2種以上を有するものを使用できる。
 塑性流体とするための前記特定化合物の好ましい具体例としては、特に制限されるものではないが、単糖類(例えば、トリオース、テトロース、ペントース、ヘキソース、ヘプトース等)が10個以上重合した多糖類が挙げられ、より好ましくは、アニオン性の官能基を有する酸性多糖類が挙げられる。ここにいう酸性多糖類とは、その構造中にアニオン性の官能基を有すれば特に制限されないが、例えば、ウロン酸(例えば、グルクロン酸、イズロン酸、ガラクツロン酸、マンヌロン酸)を有する多糖類、構造中の一部に硫酸基又はリン酸基を有する多糖類、或いはその両方の構造を持つ多糖類であって、天然から得られる多糖類のみならず、微生物により産生された多糖類、遺伝子工学的に産生された多糖類、或いは酵素を用いて人工的に合成された多糖類も含まれる。より具体的には、ヒアルロン酸、ジェランガム、脱アシル化ジェランガム(以下、DAGという場合もある)、ラムザンガム、ダイユータンガム、キサンタンガム、カラギーナン、ザンタンガム、ヘキスロン酸、フコイダン、ペクチン、ペクチン酸、ペクチニン酸、へパラン硫酸、ヘパリン、へパリチン硫酸、ケラト硫酸、コンドロイチン硫酸、デルタマン硫酸、ラムナン硫酸及びそれらの塩からなる群より1種又は2種以上から構成されるものが例示される。多糖類は、好ましくは、ヒアルロン酸、DAG、ダイユータンガム、キサンタンガム、カラギーナン又はそれらの塩であり、低濃度の使用で細胞又は組織を浮遊させることができ、かつ細胞又は組織の回収のしやすさを考慮すると、より好ましくは、DAGである。ここでいう塩とは、例えば、リチウム、ナトリウム、カリウムといったアルカリ金属の塩、カルシウム、バリウム、マグネシウムといったアルカリ土類金属の塩又はアルミニウム、亜鉛、鋼、鉄、アンモニウム、有機塩基及びアミノ酸等の塩が挙げられる。
 これらの高分子化合物(多糖類等)の重量平均分子量は、好ましくは10,000~50,000,000であり、より好ましくは100,000~20,000,000、更に好ましくは1,000,000~10,000,000である。例えば、当該分子量は、ゲル浸透クロマトグラフィー(GPC)によるフルラン換算で測定できる。さらに、DAGはリン酸化したものを使用することもできる。当該リン酸化は公知の手法で行うことができる。
 前記多糖類は、一又は複数の実施形態において、複数種(好ましくは2種)組み合わせて使用することができる。多糖類の組み合わせの種類は特に限定されないが、好ましくは、当該組合せは少なくともDAG又はその塩を含む。即ち、好適な多糖類の組合せには、DAG又はその塩、及びDAG又はその塩以外の多糖類(例、キサンタンガム、アルギン酸、カラギ-ナン、ダイユータンガム、メチルセルロース、ローカストビーンガム又はそれらの塩)が含まれる。具体的な多糖類の組み合わせとしては、DAGとラムザンガム、DAGとダイユータンガム、DAGとキサンタンガム、DAGとカラギーナン、DAGとザンタンガム、DAGとローカストビーンガム、DAGとκ-カラギーナン、DAGとアルギン酸ナトリウム、DAGとメチルセルロース等が挙げられるが、これらに限定されない。
 塑性流体とするための前記特定化合物の更に好ましい具体例としては、ヒアルロン酸、脱アシル化ジェランガム、ダイユータンガム、カラギーナン及びキサンタンガム及びそれらの塩が挙げられ、好ましい例としては脱アシル化ジェランガムまたはその塩が挙げられる。本開示において脱アシル化ジェランガムとは、1-3結合したグルコース、1-4結合したグルクロン酸、1-4結合したグルコース及び1-4結合したラムノースの4分子の糖を構成単位とする直鎖状の高分子多糖類であり、以下の一般式(I)においてR1、R2が共に水素原子であり、nが2以上の整数で表わされる多糖類である。ただし、R1がグリセリル基を、R2がアセチル基を含んでいてもよいが、アセチル基及びグリセリル基の含有量は、好ましくは10%以下であり、より好ましくは1%以下である。

 前記特定化合物により塑性流体となるメカニズムは様々であるが、脱アシル化ジェランガムの場合について記載すると、脱アシル化ジェランガムは、液体培地と混合した際に、液体培地中の金属イオン(例えば、カルシウムイオン)を取り込み、当該金属イオンを介した不定形な構造体を形成し、細胞及び/又は組織を浮遊させる。脱アシル化ジェランガムから調製される塑性流体である培地の粘度は、一又は複数の実施形態において、8mPa・s以下であり、好ましくは4mPa・s以下であり、細胞または組織の回収のしやすさの点を考慮すると、より好ましくは2mPa・s以下である。しかし、本開示にかかる細胞培養方法における塑性流体である培地の粘度は、特に制限されず、上記値より大きくてもよい。
 塑性流体とするための前記特定化合物は、化学合成法でも得ることができるが、当該化合物が天然物である場合は、当該化合物を含有している各種植物、各種動物、各種微生物から慣用技術を用いて抽出及び分離精製することにより得るのが好適である。その抽出においては、水や超臨界ガスを用いると当該化合物を効率よく抽出できる。例えば、ジェランガムの製造方法としては、発酵培地で生産微生物を培養し、菌体外に生産された粘膜物を通常の精製方法にて回収し、乾燥、粉砕等の工程後、粉末状にすればよい。また、脱アシル化ジェランガムの場合は、粘膜物を回収する際にアルカリ処理を施し、1-3結合したグルコース残基に結合したグリセリル基とアセチル基を脱アシル化した後に回収すればよい。精製方法としては、例えば、液-液抽出、分別沈澱、結品化、各種のイオン交換クロマトグラフィー、セファデックスLH-20等を用いたゲル漏過クロマトグラフィー、活性炭、シリカゲル等による吸着クロマトグラフィーもしくは薄層クロマトグラフィーによる活性物質の吸脱着処理、あるいは逆相カラムを用いた高速液体クロマトグラフィー等を単独あるいは任意の順序に組み合わせ、また反復して用いることにより、不純物を除き精製することができる。ジェランガムの生産微生物の例としてはこれに限定されるものではないが、スフィンゴモナス・エロディア(Sphingomonas elodea)及び当該微生物の遺伝子を改変した微生物が挙げられる。
 脱アシル化ジェランガムの場合、市販のもの、例えば、三品株式会社製「KELCOGEL(シーピー・ケルコ社の登録商標)CGーLA」、三栄源エフ・エフ・アイ株式会社製「ケルコゲル(シーピー・ケルコ社の登録商標)」等を使用することができる。また、ネイティブ型ジェランガムとして、三栄源エフ・エフ・アイ株式会社製「ケルコゲル(シーピー・ケルコ社の登録商標)HT」等を使用することができる。
 [塑性流体である培地中の特定化合物の濃度]
 塑性流体である培地中の前記特定化合物の濃度(質量/容量%、以下単に%で示す)は、特定化合物の種類に依存し、静置状態において培地中の細胞を沈降させずに保持できる塑性流体とすることのできる範囲で適宜設定することができるが、通常0.0005%~1.0%、好ましくは0.001%~0.4%、より好ましくは0.005%~0.1%、さらに好ましくは0.005%~0.05%である。例えば、脱アシル化ジェランガムの場合、0.001%~1.0%、好ましくは0.003%~0.5%、より好ましくは0.005%~0.1%、更に好ましくは0.01%~0.05%、更により好ましくは0.02%~0.05%である。キサンタンガムの場合、0.001%~5.0%、好ましくは0.01%~1.0%、より好ましくは0.05%~0.6%、更により好ましくは0.3%~0.6%である。κ-カラギーナンおよびローカストビーンガム混合系の場合、0.001%~5.0%、好ましくは0.005%~1.0%、より好ましくは0.01%~0.1%、最も好ましくは0.03%~0.05%である。ネイティブ型ジェランガムの場合、0.05%~1.0%、好ましくは0.05%~0.1%である。
 上記多糖類を複数種(好ましくは2種)組み合わせて使用する場合、当該多糖類の濃度は、静置状態において培地中の細胞を沈降させずに保持できる塑性流体とすることのできる範囲で適宜設定することができる。例えば、DAG又はその塩と、DAG又はその塩以外の多糖類との組合せを用いる場合、DAG又はその塩の濃度としては0.005~0.02%、好ましくは0.01~0.02%が例示され、DAG又はその塩以外の多糖類の濃度としては、0005~0.4%、好ましくは0.1~0.4%が例示される。具体的な濃度範囲の組合せとしては、以下が例示される。
DAG又はその塩:0.005~0.02%(好ましくは0.01~0.02%)
DAG以外の多糖類
キサンタンガム:0.1~0.4%
アルギン酸ナトリウム:0.1~0.4%
ロ一カトビーンガム:0.1~0.4%

 なお該濃度は、以下の式で算出できる。

 前記特定化合物は、化学合成法によってさらに別の誘導体に変えることもでき、そのようにして得た当該誘導体も、本開示にかかる細胞培養方法において有効に使用できる。具体的には、脱アシル化ジェランガムの場合、前記一般式(I)で表される化合物のR1及び/又はR2に当たる水酸基を、C1-3アルコキシ基、C1-3アルキルスルホニル基、グルコースあるいはフルクトースなどの単糖残基、スクロース、ラクトースなどのオリゴ糖残基、グリシン、アルギニンなどのアミノ酸残基などに置換した誘導体も本発明に使用できる。また、1-ethyl-3-(3-di-methylaminopropyI)carbodiimide(EDC)等のクロスリンカーを用いて当該化合物を架橋することもできる。
 前記特定化合物或いはその塩は製造条件により任意の結品形として存在することができ、任意の水和物として存在することができるがこれら結品形や水和物及びそれらの混合物も本発明の範囲に含有される。また、アセトン、エタノール、テトラヒドロフランなどの有機溶媒を含む溶媒和物として存在することもあるが、これらの形態はいずれも本開示にかかる細胞培養方法の範囲に含有される。
 本開示にかかる細胞培養方法に使用される特定化合物は、環内或いは環外異性化により生成する互変異性体、幾何異性体、互変異性体若しくは幾何異性体の混合物、又はそれらの混合物の形で存在しもよい。本開示において前記特定化合物は、異性化により生じるか否かに拘わらず、不斉中心を有する場合は、分割された光学異性体或いはそれらを任意の比率で含む混合物の形で存在してよい。
 本開示にかかる細胞培養方法に使用される培地には、金属イオン、例えば2価の金属イオン(カルシウムイオン、マグネシウムイオン、亜鉛イオン、鉄イオンおよび銅イオン等)が存在してもよく、好ましくはカルシウムイオンを含有する。当該金属イオンは、例えばカルシウムイオンとマグネシウムイオン、カルシウムイオンと亜鉛イオン、カルシウムイオンと鉄イオン、カルシウムイオンと銅イオンのように、2種類以上を組み合わせて使用することができる。当業者は適宜その組み合わせを決定することができる。一態様において、培地に金属イオンが含まれることにより、高分子化合物が金属イオンを介して集合し、高分子化合物が三次元ネットワークを形成することにより(例えば、多糖類が金属イオンを介してマイクロゲルを形成することにより)塑性流体が形成される。金属イオンの濃度は、特定化合物が液体培地を、静置状態において培地中の細胞を沈降させずに保持できる塑性流体とすることのできる範囲で、適宜設定することができる。塩濃度は0.1mM~300mMで、好ましくは0.5mM~100mMであるが、これらに限定されない。当該金属イオンは、液体培地と共に前記特定化合物と混合する、あるいは、塩溶液を別途調製しておき、特定化合物と培地成分とを混合した培地に添加してもよい。
 [塑性流体である培地の製造方法]
 塑性流体である培地は、一又は複数の実施形態において、前記特定化合物の溶液又は分散液を液体培地に添加することにより製造できる。培地には通常、前記特定化合物がイオンを介して集合、あるいは、前記特定化合物が三次元のネットワークを形成するのに十分な濃度の金属イオンが含まれるので、前記特定化合物の溶液又は分散液を液体培地に添加するのみで、塑性流体である培地を得ることができる。あるいは、前記特定化合物の溶液又は分散液に培地を添加してもよい。さらに、塑性流体である培地は、特定化合物と培地成分とを、水性溶媒(例えばイオン交換水や超純水等を含む水)中で混合して調製することもできる。混合の態様としては、(1)液体培地と前記特定化合物(溶液)とを混合する、(2)液体培地に前記特定化合物(粉末等の固体)を混合する、(3)前記特定化合物の溶液又は分散液に粉末培地を混合する、(4)粉末培地及び前記特定化合物(粉末等の固体)を水性溶媒と混合する、等が挙げられるが、これらに限定されない。塑性流体である培地における特定化合物の分布が不均一になるのを防ぐために、(1)若しくは(4)又は(1)若しくは(3)の態様が好ましい。
 [塑性流体である培地の降伏値]
 本開示にかかる細胞培養方法に用いる塑性流体である培地の降伏値は、静置状態において培地中の細胞を沈降させずに保持できる塑性流体であれば制限されない。本開示にかかる細胞培養方法で使用する塑性流体の降伏値は、細胞を保持する観点から、0.02Pa以上が好ましく、さらに、ガスホールドアップを増大させ培養細胞の生存率を向上させる点から、0.03Pa以上がより好ましく、0.04Pa以上がさらに好ましくい。本開示にかかる細胞培養方法で使用する塑性流体の降伏値は、例えば、0.2Pa以下又は0.1Pa以下である。降伏値は、レオメータで測定される値であり、具体的には実施例に記載の方法で測定される値である。
 [培地]
 本開示にかかる細胞培養方法に使用される培地の培地成分としては、培養対象が動物由来の細胞である場合、動物細胞の培養に用いられる培地であればいずれも用いることができる。このような培地としては、例えば、ダルベッコ改変イーグル培地(Dulbecco’s Modified EagIes’s Medium;DMEM)、ハムF12培地(Ham’s Nutrient Mixture F12)、DMEM/F12培地、マッコイ5AI培地(McCoy’s 5A Medium)、イーグルMEM培地(Eagles’s Minimum Essential Medium;EMEM)、αMEM培地(alpha Modified EagIes’s Minimum Essential Medium;αMEM)、MEM培地(Minimum Essential Medium)、RPMl1640培地、イスコフ改変ダルベッコ培地(Iscove’s Modified Dulbecco’s Medium;IMDM)、MCDB131培地、ウィリアム培地E、IPL41培地、Fischer’s培地、StemPro34(インビトロジェン社製)、X-VIV0 10(ケンプレックス社製)、X-VIVO 15(ケンプレックス社製)、HPGM(ケンプレックス社製)、StemSpan H3000(ステムセルテクノロジー社製)、StemSpanSFEM(ステムセルテクノロジー社製)、StemlinelI(シグマアルドリッチ社製)、QBSF-60(クオリティバイオロジカル社製)、StemProhESCSFM(インビトロジェン社製)、Essential8(登録商標)培地(ギブコ社製)、mTeSR1或いは2培地(ステムセルテクノロジー社製)、リプロFF或いはリプロFF2(リプロセル社製)、PSGrohESC/iPSC培地(システムバイオサイエンス社製)、NutriStem(登録商標)培地(バイオロジカルインダストリーズ社製)、CSTI-7培地(細胞科学研究所社製)、MesenPRORS培地(ギブコ社製)、MF-Medium(登録商標)閏葉系幹細胞増殖培地(東洋紡株式会社製)、Sf-900II(インビトロジェン社製)、Opti-Pro(インビトロジェン社製)、などが挙げられる。
 本開示にかかる細胞培養方法に使用される培地の培地成分としては、培養対象が植物由来の細胞である場合、植物組織培養に通常用いられるムラシゲ・スクーグ(MS)培地、リンズマイヤー・スクーグ(LS)培地、ホワイト培地、ガンボーグB5培地、ニッチェ培地、ヘラー培地、モーレル培地等の基本培地、或いは、これら培地成分を至適濃度に修正した修正培地(例えば、アンモニア態窒素濃度を半分にする等)に、オーキシン類及び必要に応じてサイトカイニン類等の植物生長調節物質(植物ホルモン)を適当な濃度で添加した培地が培地として挙げられる。これらの培地には、必要に応じて、カゼイン分解酵素、コーンスティーフリカー、ビタミン類等をさらに補充することができる。オーキシン類としては、例えば、3-インドール酢酸(IAA)、3-インドール酪酸(IBA)、1-ナフタレン酢酸(NAA)、2,4-ジクロロフエノキシ酢酸(2,4-D)等が挙げられるが、それらに限定されない。オーキシン類は、例えば、約0.1~約10ppmの濃度で培地に添加され得る。サイトカイニン類としては、例えば、カイネチン、ベンジルアデニン(BA)、ゼアチン等が挙げられるが、それらに限定されない。サイトカイニン類は、例えば、約0.1~約10ppmの濃度で培地に添加され得る。
 本開示にかかる細胞培養方法に使用される培地には、ナトリウム、カリウム、カルシウム、マグネシウム、リン、塩素、各種アミノ酸、各種ビタミン、抗生物質、血清、脂肪酸、糖などを当業者は目的に応じて自由に添加してもよい。動物由来の細胞及び/又は組織培養の際には、当業者は目的に応じてその他の化学成分あるいは生体成分を一種類以上組み合わせて添加することもできる。動物由来の細胞の培地に添加される成分としては、ウシ胎児血清、ヒト血清、ウマ血清、インシュリン、トランスフェリン、ラクトフエリン、コレステロール、エタノールアミン、亜セレン酸ナトリウム、モノチオグリセロール、2-メルカフトエタノール、ウシ血清アルブミン、ピルビン酸ナトリウム、ポリエチレングリコール、各種ビタミン、各種アミノ酸、寒天、アガロース、コラーゲン、メチルセルロース、各種サイトカイン、各種ホルモン、各種増殖因子、各種細胞外マトリックスや各種細胞接着分子などが挙げられる。
 [培養される細胞]
 本開示にかかる細胞培養方法の対象となる細胞は、特に制限されず、懸濁培養可能な細胞が挙げられる。該細胞は、一又は複数の実施形態において、動物由来の細胞及び植物由来の細胞が含まれうる。本開示にかかる細胞培養方法は、細胞に対する物理的ストレスの抑制と、酸素供給の向上とが両立された浮遊細胞培養が可能となるから、物理的ストレスに弱い細胞、例えば、再生医療に使用する細胞や植物細胞なども対象とすることができる。
 本開示にかかる細胞培養方法の対象となる細胞は、一又は複数の実施形態において、細胞塊、スフィア、組織における細胞を含む。よって、本開示にかかる細胞培養方法は、一又は複数の実施形態において、細胞塊、スフィア、組織の培養方法でもある。
 動物由来の細胞には、精子や卵子などの生殖細胞、生体を構成する体細胞、幹細胞、前駆細胞、生体から分離された癌細胞、生体から分離され不死化能を獲得して体外で安定して維持される細胞(細胞株)、生体から分離され人為的に遺伝子改変が成された細胞、生体から分離され人為的に核が交換された細胞等が含まれうる。
 植物由来の細胞には、植物体の各組織から分離した細胞が含まれ、当該細胞から細胞壁を人為的に除いたフロトフラストも含まれる。
 本開示にかかる細胞培養方法の対象となる細胞は、一又は複数の実施形態において、例えば、胚性幹細胞(ES細胞)及び人工多能性幹細胞(iPS細胞)などのような、多能性幹細胞であり、或いは、例えば、間葉系幹細胞、造血幹細胞及び組織幹細胞などのような、多分化能幹細胞が挙げられる。
 [細胞の配置]
 本開示にかかる細胞培養方法は、塑性流体である培地中に細胞を配置することを含む。細胞の配置は、細胞を該培地中に浮遊する状態となるようにすること、或いは、細胞を該培地中に分散した状態となるようにすることを含む。この工程は、一又は複数の実施形態において、以下の方法で行うことができるが、配置方法はこれらに限定されない。まず、前記特定化学物又はその溶液若しくは分散液を混合する前の液体培地に細胞を分散させておくことが挙げられる。次に、前記特定化学物と培地成分と水とが混合されたものに、細胞又はその分散液を混合して分散させることが挙げられる。
 塑性流体である培地に配置された細胞は、沈降が抑制された状態(すなわち、浮遊した状態)が維持される。
 本開示にかかる細胞培養方法において、培養の開示時に培地に配置される細胞は、別途調製又は培養されて得られた細胞単独、細胞塊、スフィア、又は組織の形態であってもよい。
 [気泡の導入]
 本開示にかかる細胞培養方法は、細胞が配置された塑性流体である培地に、気泡を導入し、培養される細胞に酸素を供給することを含む。
 塑性流体に導入された気泡は、塑性流体ではない液体に導入された気泡に比べ、その上昇速度が抑制される。これは、塑性流体における気泡の滞留時間が延長され、ガスホールドアップが増大することを意味する。したがって、気泡の導入は、細胞の増殖に必要な酸素供給ができる範囲で抑制することができる。気泡の導入は連続的でもよく、あるいは、細胞の増殖に必要な酸素供給ができる範囲で間欠的でもよい。
 [気泡]
 導入する気泡のサイズは、発泡による物理ストレスを低減する点及び上昇速度を低減する点から、小さいことが好ましく、マイクロバブル又はサブマイクロバブルが好ましい。気泡の直径としては、例えば、500μm以下、100μm以下、50μm以下、25μm以下、又は10μm以下が挙げられ、そして、0.1μm以上、1μm以上、又は10μm以上が挙げられる。
 [気泡の導入方法]
 気泡の導入方法は、限定されないが、培養用のスパージャーを利用できる。或いは、気泡の導入な可能なノズルを用いてもよい。気泡の導入は、一カ所からでもよく、複数カ所からでもよい。気泡の導入量は、導入箇所と培地体積との関係で適宜調節されうる。
 [撹拌]
 塑性流体中に導入された気泡の移動は、一時的かつ局所的な流動である。気泡の上昇部以外は流動せず、気泡上昇による対流も抑制される。よって、本開示にかかる細胞培養方法は、一又は複数の実施形態において、酸素供給を均一化するために一定周期で培地を撹拌してもよい。該撹拌は、細胞増殖に影響を及ぼさない程度の物理ダメージの範囲の低頻度の撹拌とすることができる。撹拌は連続的でもよく、あるいは、間欠的でもよい。撹拌の頻度は、気泡の導入量や導入箇所と培地体積との関係で適宜調節されうる。
 複数カ所から気泡を導入することで撹拌の工程は省くことも可能である。
 撹拌の方法は特に限定されず、攪拌翼による撹拌、培養容器全体の振とうによる撹拌、又はこれらの組み合わせであってよい。培養容器全体の振とうとしては、培養容器を揺らすこと又は回すことが挙げられる。
 [培地体積]
 本開示にかかる細胞培養方法における培地体積は、限定されないが、数リットル~数千リットル、例えば、3リットル以上、5リットル以上、7リットル以上、9リットル以上、11リットル以上、13リットル以上、又は15リットル以上とすることができる。上限としては、例えば、10000リットル以下が挙げられる。このように、本開示にかかる細胞培養方法によれば、大量の培地体積による細胞培養が可能である。
 また、本開示にかかる細胞培養方法によれば、従来の大量培養法では物理ストレスのため培養することが困難である物理的ストレスに弱い細胞(例えば、再生医療に使用する幹細胞や植物細胞など)の大量培養が可能となる。
 塑性流体に気泡を導入するという手法により、細胞に対する物理的ストレスの抑制と、酸素供給の向上との両立が可能となるからである。
 [培養システム]
 本開示にかかる細胞培養方法は、一又は複数の実施形態において、汚染等を予防する点から、閉鎖系培養系、例えば、閉鎖系の培養容器、培養バッグ、バイオリアクタ-、培養装置などで行うことが好ましい。
 本開示は、下記の実施形態に関し得る。
[1] 塑性流体である培地中に細胞を分散した状態で配置すること、及び該培地に気泡を導入することを含む、細胞の培養方法。
[2] 前記細胞を、塑性流体である培地中に分散した状態で配置することを含む、[1]に記載の培養方法。
[3] 前記細胞は、懸濁培養可能な細胞である、[1]又は[2]に記載の培養方法。
[4] 前記細胞は、多能性幹細胞又は多分化能幹細胞である、[1]から[3]のいずれかに記載の培養方法。
[5] 前記塑性流体である培地は、静置状態において、培地中の細胞を沈降させずに保持できる培地である、[1]から[4]のいずれかに記載の培養方法。
[6] 細胞培養容器又は細胞培養バックを用いて培養することを含む、[1]から[5]のいずれかに記載の培養方法。
[7] 気泡の導入は、間欠又は連続で行う、[1]から[6]のいずれかに記載の培養方法。
[8] 気泡が、マイクロバブル又はサブマイクロバブルである、[1]から[7]のいずれかに記載の培養方法。
[9] さらに、培地の攪拌を間欠又は連続で行うことを含む、[1]から[8]のいずれかに記載の培養方法
[10] 前記攪拌は、攪拌翼による撹拌又は培養容器全体の振とうによる撹拌である、[9]に記載の培養方法。
[11] 前記培地の降伏値が、0.03Pa以上である、[1]から[10]のいずれかに記載の培養方法。
 以下、実験例により本開示をさらに詳細に説明するが、これらは例示的なものであって、本開示はこれら実験例に制限されるものではない。
 [材料及び実験方法]
 1.使用細胞・培地、使用試薬及び培養条件
 1.1 使用細胞
 実験にはヒト人工多能性幹細胞(Human induced Pluripotent Stem Cell、以下hiPS細胞という、Tic株,国立成育医療センター)を用いた。このhiPS細胞はiMatrix-511(Nippi社製)コート面上で複数回継代培養を行ったものである。
 1.2 使用培地
 培地にはmTeSR(商標)1(STEMCELL Technologies社製,以下mTeSR1という)を用いた(以下、通常培地という)。
 1.3 使用試薬
 塑性流体とするための化合物として化合物A:脱アシル化ジェランガム(日産化学社製)又は化合物B:キサンタンガム(シグマ-アルドリッチ社製)を用いた。
 1.4 培養条件
 培養はCO2インキュベーター(MCO-17AI、三洋電機社製)で37℃、5%CO2の条件下で行った。
 2.塑性流体培地の調製
 塑性流体とするための化合物A及びB(上述)を通常培地に所定の濃度(質量/容量%、以下単に“%”で表す。)となるように溶解して調製した。
 3.培養
 3.1 スパージングを伴うhiPS細胞の三次元浮遊培養
 iMatrix-511コート面上で前培養したhiPS細胞をPBSで洗浄した。EDTAをPBSで5mMに調整したものに、10μMとなるRock inhibitorを加えたEDTA/PBS溶液を培養面積1cm2あたり55μl添加し室温で10分間静置した。その後TrypLE selectに10μMとなるようRock inhibitorを加えたTrypLE select溶液を培養面積1cm2あたり55μl添加し室温で7分間静置した。通常培地を1cm2あたり55μl添加することで酵素中和を行い、単分散処理されたhiPS細胞を回収した。170×gで室温にて遠心分離を3分間行い、上澄み液を除いたものに再び通常培地を加え再懸濁を行った。これにRock inhibitorを10μMになるよう調整して添加し細胞懸濁液を用意した。これを1.0×105cells/mlになるように調整し、30mlシングルユースバイオリアクターBWV-S03A(Able社製)に播種して24時間培養を行い、細胞集塊形成を行った。その後以下の2条件で120時間まで培養を行った。
 ・培養のコントロール
 コントロールとして、培養24時間後から24時間毎に通常培地に培地交換を行いながら通常培地の懸濁培養を行った。
 ・スパージングを伴う浮遊培養
 培養24時間から24時間毎に、化合物Aを濃度0.02%となるように通常培地に添加した塑性流体培地(塑性流体培地A,以下同じ)に培地交換を行った。培養期間中はCO2 Gas Mixer(トッケン社製)を用い0.01ml/minの通気量でスパージングを行った。塑性流体培地Aを用いた浮遊培養では、間欠的に撹拌翼を30rpmで回転させて撹拌を行った。
 3.2 細胞濃度の測定
 24時間毎に細胞濃度測定を行った。250μl×3回、遠沈管にサンプリングを行った。通常培地で培養したものは170×gで3分間遠心分離を行い、上澄み液を除いた後Accumax(Innovative cell technologies社製)を5ml添加し室温で10分間反応させ単分散処理を行った。塑性流体培地Aで培養をしたものはPBSを添加し5倍希釈を行なった後、170×gで3分間遠心分離を行い、上澄み液を除いた後Accumaxを5ml添加し室温で10分間反応させ単分散処理を行った。その後170×gで再び遠心分離を行って上澄み液を除去し、通常培地を加えて再懸濁しTC20(商標)全自動セルカウンター(Bio-Rad社製)を用いてトリパンブルー色素排除法により生細胞濃度を測定した。
 4.塑性流体培地を用いた影響の観察
 4.1 気泡上昇への影響の観察
 CO2 Gas Mixerを用いて塑性流体培地Aと通常培地に0.01ml/minの通気量でスパージングを行い、その様子をビデオカメラ(HDR-CX420,SONY社製)を用いて観察した。
 4.2 細胞集塊への影響の観察
 塑性流体培地Aと通常培地にピペッティングを行い、その様子をビデオカメラを用いて観察した。培地中に含まれる細胞集塊は3.1のコントロールと同手法で120時間培養を行い回収したものを用いた。
 4.3 液流への影響の観察
 CO2 Gas Mixerを用いて塑性流体培地Aと通常培地に0.01ml/minの通気量でスパージングを行い、その様子をビデオカメラを用いて観察した。培地中に含まれる細胞集塊は3.1のコントロールと同手法で120時間培養を行い回収したものを用いた。
 [結果及び考察]
 1.塑性流体の物性
 化合物A又はBを下記表1に示す濃度(質量/容量%)となるように通常培地に溶解した液体組成物を調製し、該液体組成物について下記2点の物性を調べた。その結果を下記表1に示す。
(1)ビーズ保持性
 模擬細胞としてのポリスチレンビーズ(Positive Charged Microcarriers、Corning社製、粒径160 - 200μm、密度1.09 - 1.15 g/cm3)を該液体組成物に懸濁して37℃で静置し、懸濁した時間を始点として24時間後にビーズの様子を観察した。
(2)気泡滞留性
 ピペッティングにて該液体組成物に気泡を封入して37℃で静置し、懸濁した時間を始点として24時間後に気泡の様子を観察した。
(3)降伏値の測定
 レオメータ(商品名MCR302、アントンパール社製)を用い、37℃における該液体組成物の降伏値を測定した。
 下記表1に示す通り、化合物Aを添加した通常培地(液体組成物)では、化合物Aが0.02%以上でビーズを保持(ビーズ沈降を抑制)でき、塑性流体の物性を示した。また、気泡の上昇速度が抑制された結果、化合物Aが0.04%以上で気泡の滞留が認められた。降伏値は、化合物Aが0.03%以上で測定でき、0.03%での降伏値は、0.030Paであった。化合物Aが0.02%以下の液体組成物の降伏値は用いた基材の検出感度以下であった。
 また、下記表1に示す通り、化合物Bを添加した通常培地(液体組成物)では0.3%以上でビーズを保持(ビーズ沈降を抑制)でき、塑性流体の物性を示した。また、気泡の上昇速度が抑制された結果、化合物Bが0.5%以上で気泡の滞留が認められた。降伏値は、化合物Bが0.4%以上で測定でき、0.4%での降伏値は、0.036Paであった。化合物Bが0.3%以下の液体組成物の降伏値は用いた基材の検出感度以下であった。

 2.気泡上昇に対する塑性流体の影響
 深部通気装置を用いて塑性流体培地A(0.02% 化合物A)と通常培地に0.01ml/分の通気量でスパージングを行いその様子を観察した。図1にそれぞれ0.1秒ごとの気泡の上昇を確認しその位置を点で示した。この画像から、気泡の0.1秒ごとの間隔は塑性流体培地Aの方が通常培地より小さく、その上昇が遅いことが確認された。この実験の動画から平均的な気泡上昇速度を計測したものを表2に示す。これらの結果から塑性流体を用いることで気泡の上昇速度を抑え長時間液中に保持しガスホールドアップを上げることが可能であることが示唆される。

 3.細胞集塊に対する塑性流体の影響
 細胞集塊を含む塑性流体培地A及び通常培地にピペッティングを行い、その様子を観察した。図2にそれぞれのピペッティング時、ピペッティング終了時及びその5秒後及び15秒後の画像を示す。この画像から、ピペッティングを行い培地中に均一に拡散した細胞集塊が、通常培地では沈降していくのに対し、塑性流体培地Aでは細胞集塊が培養液中に保持される様子が確認された。このことから塑性流体を用いることで、シェアストレス環境下では培養液は撹拌し液流を発生させることが可能であり、シェアレス(シェアストレスがない又は抑制された)環境下ではピペッティングによって発生した液流はすぐに停止し、細胞集塊を液中に保持可能であることが示唆される。
 4.液流に対する塑性流体の影響
 1つの深部通気装置を用い塑性流体培地Aに0.01ml/minの通気量でスパージングを行い、その様子を観察した。気泡上昇部付近(図3)と、同一容器内の少し離れた位置(図4)での0.00~6.00秒の画像を示す。画像中の丸はその中心に同一の集塊を示している。画像に示すように気泡上昇部付近では細胞集塊も液流に沿って上昇していく様子が確認された。一方少し離れた位置では細胞集塊はほぼ動かず静止状態を保っている様子が確認された。これらのことから気泡上昇部付近のみでしかそれに伴う上昇流は発生せず、さらに少し離れた位置ではニュートン流体では上昇流によって生じる対流も抑制されていることが示唆される。
 5.スパージングを伴う浮遊培養
 これまでの実験により塑性流体を用いるとスパージングを行った際液流が抑えられ対流が生じないことが確認された。そのためスパージングを一カ所から行うだけでは、気泡上昇部付近のみでしか十分な酸素供給が行われず、離れた位置では酸素枯渇が生じることが予測されるため、短時間、低頻度に撹拌を行うことで均一な環境を作り出す必要があると考えられる。そこで塑性流体培地Aでスパージングを行いながら、8時間毎に5分間撹拌を行いhiPS細胞集塊を培養した。なお、短時間、低頻度の撹拌に替えてスパージングの箇所を増やすことでも均一な環境を作り出すことができると考えられる。
 5.1 細胞集塊の形態
 図5にコントロール及び塑性流体培地Aを用いてスパージングを行った浮遊培養の各時間における代表的な画像を示す。この結果、スパージングを伴う浮遊培養でも、コントロールである通常培地の懸濁培養と同様集塊径が増大していく様子が確認された。
 5.2 細胞濃度の経時変化
 図6に細胞濃度の経時変化を示す。この結果から、塑性流体培地Aであればスパージングを行っても動物細胞であるhiPS細胞が通常培地の懸濁培養と同様に増殖可能であることが確認された。
 以上の結果からより大きなスケールのバイオリアクタでもスパージングによる酸素供給が可能となることが示唆された。一方、通常培地の懸濁培養の場合(すなわち、表面通気による酸素供給のみの場合)、培養液量が増加するに従い単位体積当たりの気液界面面積が低下するため、例えば数~数十リットル又はそれ以上などのような大量の培地体積を用いる培養では酸素供給が間に合わないと考えられる。
 6.異なる降伏値条件下における酸素供給と細胞増殖の確認
 0.02%及び0.05%となるように化合物Aを通常培地に溶解し、降伏値が異なる2つの塑性流体培地を調製した(上記表1)。各溶液に対しピペッティング操作にて不均一径の気泡を液中に封入後、120時間の前培養をした細胞集塊を播種密度4.0 x 105 cells/mLで懸濁し、30 mlシングルユースバイオリアクター(同上)を用いて37℃インキュベーター内で培養を行った。培養6時間毎に細胞密度を測定し,時間経過における細胞密度の推移を培養24時間まで測定した。その結果を図7及び8に示す。
 図7に時間経過における細胞集塊の様子を示す。また、図8に時間経過における細胞密度の様子を示す。
 気泡が液中に保持されない濃度条件(化合物A 0.02%)においては、時間経過とともに細胞集塊径が減少する傾向と細胞集塊の輪郭が滑らかな形状から歪な形状へと変化する傾向が確認された。また、細胞密度も時間経過とともに減少した。一方で気泡の液中保持が確認される濃度条件(化合物A 0.05%)下では,細胞集塊径及び細胞密度は維持される傾向がみられた。細胞密度の結果から培養24時間後における生存率αを算出した結果を下記表3に示す。

 気泡が保持されない濃度条件下(化合物A 0.02%)においては細胞密度及び生存率が減少していく傾向を示したのに対し、気泡が保持される濃度条件下(化合物A 0.05%)においては,初期の細胞密度増加とその後の維持が確認された。以上のことにより、塑性流体培地を用いることにより、液中に気泡を保持しそれによる酸素供給が可能であることが確認された。
 7.酸素供給がスパージングに限定される条件下での塑性流体浮遊培養
 図9に示すような、液中に供給される気泡(スパージング)からのみ酸素供給が行われる装置を作成した。同装置は、装置上部から継続的に窒素を通気することにより液上部のスペースを窒素置換し,下部の1つのノズルから液中に供給される気泡からのみ酸素供給が行われる。同装置によれば、気泡通気を伴う細胞培養を行うことで,気泡通気(スパージング)における酸素供給下での細胞培養を達成できる。
 下記に示す実験概要にて実験を行った。二次元平面上で前培養したhiPS細胞 (D2) を5 mM EDTA/PBS、TrypLE(商標)select処理により培養面から剥離・単分散処理を行った.10 mM Rock inhibitor Y-27632に調整したStemFit(商標)AK02Nにより細胞懸濁液を播種密度 1.0 × 105 cells/mLに調整しSingle use bioreactor 30 mLに播種し48 h前培養を行った。48 h後に細胞集塊を回収し,化合物A含有塑性培地に細胞集塊を播種した。スパージングを伴う条件、及び、コントロールとしてスパージングを行わない条件の2条件で培養を行った。両条件とも培養液上部は窒素ガス通気を行い常に置換した条件で培養を行った。培養24 h後に細胞密度測定を行った。その結果を図10及び11に示す。

  :化合物A:0.05 %
 図10に示すとおり、スパージングをしないコントロール条件と比較し,スパージングを伴う条件では培養24 h後において高い細胞密度を示した。また、図11に示すとおり、培養24 hまでの比増殖速度μapp (h-1) はスパージングを伴う条件ではコントロールと比較して高い値を示し,三次元静置培養でのみかけの比増殖速度と同程度の値を示した。これらの結果から、コントロール条件では細胞の増殖に酸素が不足し増殖速度が低下していることが想定された。塑性流体培地を用いれば、スパージングによるhiPS細胞培養系への酸素供給を可能であることが確認された。またその際三次元静置培養と同程度の比増殖速度を示すことから、スパージングによるシェアストレスは、塑性流体では抑制されていることが確認された。
 よって、本開示にかかる細胞培養方法によれば、細胞に対する物理的ストレスの抑制と、酸素供給の向上とが両立された浮遊細胞培養が可能となる。そして、本開示にかかる細胞培養方法によれば、例えば数~数十リットル又はそれ以上などのような大量の培地体積を用いる浮遊細胞培養においても、細胞に対する物理的ストレスの抑制と、酸素供給の向上との両立が可能となり、物理的ストレスに弱い細胞、例えば、再生医療に使用する細胞や植物細胞などの大量培養が可能となると考えられる。

Claims (8)

  1.  塑性流体である培地中に細胞を配置すること、及び
     該培地に気泡を導入することを含む、
     細胞の培養方法。
  2.  前記細胞は、懸濁培養可能な細胞である、請求項1に記載の培養方法。
  3.  前記細胞は、多能性幹細胞又は多分化能幹細胞である、請求項1又は2に記載の培養方法。
  4.  前記塑性流体である培地は、静置状態において、培地中の細胞を沈降させずに保持できる培地である、請求項1から3のいずれかに記載の培養方法。
  5.  細胞培養容器又は細胞培養バックを用いて培養することを含む、請求項1から4のいずれかに記載の培養方法。
  6.  気泡の導入は、間欠又は連続で行う、請求項1から5のいずれかに記載の培養方法。
  7.  気泡が、マイクロバブル又はサブマイクロバブルである、請求項1から6のいずれかに記載の培養方法。
  8.  前記培地の降伏値が、0.03Pa以上である、請求項1から7のいずれかに記載の培養方法。
