Abstract
Description
[1]Dedicator of cytokinesis 2(DOCK2)によるRac活性化の制御剤を有効成分として含有する、免疫制御剤。
[2]DOCK2によるRac活性化の前記制御剤が、下記式(1)で表される化合物、その薬学的に許容される塩、又はそれらの溶媒和物である、[1]に記載の免疫制御剤。
[3]DOCK2によるRac活性化の前記制御剤が、コレステロール3-硫酸又はその塩に対する特異的結合物質である、[1]に記載の免疫制御剤。

本実施形態の免疫制御剤は、DOCK2によるRac活性化の制御剤(DOCK2制御剤)を有効成分として含有する。本明細書において、Rac活性化の制御とは、Rac活性化の阻害、Rac活性化の促進又はRac活性化の維持を意味する。また、例えば、「Racの活性化の阻害」とは、Racの活性化を完全に阻害すること及びRacの活性化を一部阻害すること(Racの活性化を抑制すること)を意味する。本実施形態の免疫制御剤としては、例えば、DOCK2のDHR-2ドメインに対する特異的結合物質、Racに対する特異的結合物質、コレステロール3-硫酸に対する特異的結合物質、コレステロール3-硫酸の合成や分解を担う酵素の阻害剤等であって、DOCK2によるRacの活性化を制御するものが挙げられる。特異的結合物質については後述する。
1実施形態において、DOCK2によるRac活性化の制御剤は、下記式(1)で表される化合物、その薬学的に許容される塩、又はそれらの溶媒和物である。すなわち、1実施形態において、本発明は、下記式(1)で表される化合物、その薬学的に許容される塩、又はそれらの溶媒和物(以下、「コレステロール3-硫酸又はその誘導体」という場合がある。)を有効成分として含有する免疫制御剤を提供する。式(1)中、R1は、直鎖状であっても分岐鎖状であっても環状であってもよく、置換されていてもよい炭素数3~12のアルキル基又は置換されていてもよい炭素数6~12の芳香族基を表す。
1実施形態において、DOCK2によるRac活性化の制御剤は、コレステロール3-硫酸又はその塩に対する特異的結合物質である。すなわち、1実施形態において、本発明は、コレステロール3-硫酸又はその塩に対する特異的結合物質を有効成分として含有する免疫制御剤を提供する。コレステロール3-硫酸の塩としては、上記式(1)で表される化合物の塩と同様のものが挙げられる。
1実施形態において、DOCK2によるRac活性化の制御剤は、配列番号1に記載のアミノ酸配列からなるタンパク質の発現ベクター、又は配列番号1に記載のアミノ酸配列において1若しくは数個のアミノ酸が欠失、置換若しくは付加されたアミノ酸配列からなり、かつコレステロール3-硫酸又はその塩に対する結合活性を有するタンパク質の発現ベクターである。すなわち、1実施形態において、本発明は、配列番号1に記載のアミノ酸配列からなるタンパク質の発現ベクター、又は配列番号1に記載のアミノ酸配列において1若しくは数個のアミノ酸が欠失、置換若しくは付加されたアミノ酸配列からなり、かつコレステロール3-硫酸又はその塩に対する結合活性を有するタンパク質の発現ベクターを有効成分として含有する免疫制御剤を提供する。

上述した免疫制御剤は、それ自体を投与してもよいし、薬学的に許容される担体と混合した医薬組成物として製剤化したものを投与してもよい。
1実施形態において、本発明は、コレステロール-3硫酸若しくはその誘導体、コレステロール-3硫酸若しくはその誘導体に対する特異的結合物質、配列番号1に記載のアミノ酸配列からなるタンパク質若しくはその変異体の発現ベクター、SULT2B1bタンパク質若しくはその変異体、SULT2B1bタンパク質若しくはその変異体の発現ベクター、SULT2B1bタンパク質の発現阻害物質、SULT2B1bタンパク質の阻害剤、STSタンパク質若しくはその変異体の発現ベクター、STSタンパク質の発現阻害物質、又はSTSタンパク質の阻害剤の有効量を、治療を必要とする患者又は患畜に投与する工程を備える免疫制御方法を提供する。
(コレステロール3-硫酸によるグアニンヌクレオチド交換因子(GEF)活性の阻害効果の選択性の検討)
図1は、主な生体内ステロイドの構造と生合成経路を示す図である。コレステロール3-硫酸(図中、Cholesterol sulfateと示す。)は、SULT2B1b酵素の働きによって、コレステロールの3位の炭素に硫酸基が付与されて生合成される。一方、STS酵素の働きによって、コレステロール3-硫酸の硫酸基が除去され、コレステロールが生成される。
(コレステロール3-硫酸によるDOCK2の活性阻害の検討)
DOCK2のDHR-2ドメインのBローブ及びCローブからなるタンパク質によるRac活性化に対する各種ステロイド化合物の効果を検討した。
(ELISA法によるDOCK2のDHR-2ドメインとコレステロール3-硫酸との特異的結合の検出)
DOCK2のDHR-2ドメインのBローブ及びCローブからなるタンパク質と、各種ステロイド化合物との結合をELISA法により検討した。ステロイド化合物としては、コレステロール3-硫酸、コレステロール3-酢酸、コレステロール、プレグネノロン3-硫酸、エストロン3-硫酸、エストリオール3-硫酸、エストラジオール3-硫酸、デヒドロエピアンドロステロン、デヒドロエピアンドロステロン3-硫酸を用いた。
(DOCK2のDHR-2ドメインとRacとの結合に対するコレステロール3-硫酸の影響)
DOCK2のDHR-2ドメインとRacとの結合に対する、コレステロール3-硫酸の影響をGST-Racビーズを用いたPull downアッセイにより検討した。
(トランスウェルを用いたリンパ球の遊走アッセイ1)
T細胞の遊走に対する、コレステロール3-硫酸の影響を検討した。また、対照として、DOCK2ノックアウトマウス由来のT細胞を用いた実験も行った。より具体的には、まず、マウス脾臓細胞(1×107個/mL)を、所定濃度のコレステロール3-硫酸又はDMSOのみ(対照)の存在下で、0.5%BSA含有RPMI-1640(トランスウェル培地)中で37℃、1時間培養した。全てのサンプルにおいてDMSOの最終濃度が0.2%になるように調整した。
(トランスウェルを用いたリンパ球の遊走アッセイ2)
T細胞の遊走に対する、各種ステロイド化合物の影響を検討した。ステロイド化合物としては、コレステロール3-硫酸、コレステロール、コレステロール3-酢酸、プレグネノロン3-硫酸、エストロン3-硫酸、エストリオール3-硫酸、エストラジオール3-硫酸、デヒドロエピアンドロステロン、デヒドロエピアンドロステロン3-硫酸を用いた。
(好中球の遊走アッセイ1)
好中球の遊走速度に対するコレステロール3-硫酸の影響を検討した。より具体的には、マウス骨髄から単離した好中球を、所定濃度のコレステロール3-硫酸、又はDMSO単独(溶媒コントロール)の存在下で、20mM HEPES-NaOH(pH7.3)-0.1%BSA含有RPMI-1640(Taxiscan培地)中で室温、1時間インキュベーションした。全てのサンプルにおいてDMSOの最終濃度が0.2%になるように調整した。
(好中球の遊走アッセイ2)
好中球の遊走速度に対するコレステロール3-硫酸の影響を、細胞の前処理でなく、化学遊走因子(fMLP)と同じウェルに添加した場合についても検討した。より具体的には、マウス骨髄由来好中球をEZ-TAXIScanチャンバー(Effector Cell Institute社)の一方のウェルにロードし、260μm離れたもう一方のウェルにTaxiscan培地で3μMに調製したfMLP溶液に、150μMになるように調製したコレステロール3-硫酸、デヒドロエピアンドロステロン3-硫酸又はDMSO(対照、最終濃度3%)を1μL添加し、好中球を遊走させた。位相差顕微鏡画像を30秒間隔で40分間撮影し、上記と同様の方法で解析した。
(ストローマ細胞上でのリンパ球の遊走アッセイ)
コレステロール3-硫酸の産生酵素であるSult2B1bを過剰発現したストローマ細胞上でのT細胞の遊走性を検討した。より具体的には、マウスの二次リンパ組織を摘出し、細切れにした後、1mg/mLコラゲナーゼD(ロシュ・ダイアグノスティックス社)、0.8mg/mLディスパーゼ(ロシュ・ダイアグノスティックス社)及び0.1mg/mL DNase Iを添加したDMEM培地に懸濁し、37℃で45分間処理してストローマ細胞を分離した。
(マウスの各組織におけるSult2B1bタンパク質の発現の検討)
Sult2B1bタンパク質は、Sult2B1aタンパク質と同じ遺伝子から形成されるが、RNAスプライシングの結果、N末端領域のアミノ酸配列が異なっている。コレステロール3-硫酸の産生にはSult2B1bタンパク質の発現が必須である。そこで、Sult2B1bタンパク質に特異的な抗体を作製し、マウスの各組織におけるSult2B1bタンパク質の発現をウエスタンブロット法により検討した。
(マウスの各組織におけるコレステロール-3硫酸の存在量の検討)
野生型マウス(+/+)及びSult2B1b遺伝子のノックアウトマウス(-/-)の各組織及び涙におけるコレステロール-3硫酸(C3S)を、質量分析により定量した(n≧7)。具体的には、重水素で標識したコレステロール-3硫酸を内部標準として各組織に添加し、ホモジナイズした。続いて、各組織の懸濁液を限外濾過チューブで限外濾過し、ろ液をLC-MS/MSに供して直接解析した。コレステロール-3硫酸の絶対量は、内部標準のピーク面積に対するコレステロール-3硫酸のピーク面積の比に基づいて算出した。図15(a)及び(b)は、コレステロール-3硫酸の定量結果を示すグラフである。
(眼の炎症誘導におけるコレステロール-3硫酸の役割の検討1)
Sult2B1b遺伝子のヘテロマウス(+/-)及びノックアウトマウス(-/-)を用いたUV誘導性角膜炎モデルを検討した結果、いずれのマウスにおいても角膜上皮の萎縮が観察された。そこで、各マウスの眼球切片をヘマトキシリン・エオジン(HE)染色し、光学顕微鏡で観察した。
(眼の炎症誘導におけるコレステロール-3硫酸の役割の検討2)
Sult2B1b遺伝子のヘテロマウス(+/-)及びノックアウトマウス(-/-)をブタクサ花粉で感作し、その10日及び12日後に同抗原を眼内に投与することで誘導される結膜炎モデルを検討した。各マウスの眼球切片をヘマトキシリン・エオジン(HE)染色し、光学顕微鏡で観察した。
(コレステロール3-硫酸の検出1)
質量分析顕微鏡を用いて妊娠期マウス胎児中のコレステロール3-硫酸の分布を検出した。より具体的には、野生型妊娠マウスの胎児(E8.5)を母体より摘出し、凍結した後に新鮮な凍結薄切片を作製した。この切片上に、マトリックスとして9-amino acridineを噴霧し、質量分析顕微鏡による測定を行った。測定されたデータから質量電荷比(m/z)=465に出現するピークをタンデム質量分析によりコレステロール3-硫酸として同定し、このピーク強度分布を画像化した。続いて、測定後の切片をヘマトキシリン・エオジン(HE)染色した。
(コレステロール3-硫酸の検出2)
ヒト大腸癌のマウス肝転移モデルを用いて、癌組織中のコレステロール3-硫酸の分布を質量分析顕微鏡により検出した。より具体的には、GFP変異体であるvenus遺伝子を導入して蛍光標識したヒト大腸癌細胞株HCT116(HCT116/venus)を、NOGマウス(NOD/Shi-scid/IL-2RγKOマウス)の脾臓内に移入した。続いて、2週間後にNOGマウスから肝臓を摘出し、液体窒素で凍結した後、凍結切片を作製し、蛍光顕微鏡観察を行った。続いて、被検切片上に、マトリックスとして9-amino acridineを噴霧し、質量分析顕微鏡による測定を行った。測定されたデータからm/z=465に出現するピークをタンデム質量分析によりコレステロール3-硫酸として同定し、このピーク強度分布を画像化した。
(コレステロール3-硫酸の検出3)
ヒト非小細胞肺癌細胞の皮下移植モデルを用いて皮下腫瘍組織中のコレステロール3-硫酸及びアラキドン酸の分布を質量分析顕微鏡により検出した。より具体的には、ヒト非小細胞肺癌細胞株PC-9をヌードマウスの皮下に移植し、4週間後に皮下腫瘍を摘出し、HE染色及び質量分析顕微鏡による解析を行った。詳細には、被検切片上に、マトリックスとして9-amino acridineを噴霧し、質量分析顕微鏡による測定を行い、測定されたデータからm/z=465に出現するピークをタンデム質量分析によりコレステロール3-硫酸として同定し、このピーク強度分布を画像化した。続いて、測定後の切片をヘマトキシリン・エオジン(HE)染色した。
Claims (7)
- Dedicator of cytokinesis 2(DOCK2)によるRac活性化の制御剤を有効成分として含有する、免疫制御剤。
- DOCK2によるRac活性化の前記制御剤が、コレステロール3-硫酸又はその塩に対する特異的結合物質である、請求項1に記載の免疫制御剤。
- 前記特異的結合物質が、配列番号1に記載のアミノ酸配列からなるタンパク質、又は配列番号1に記載のアミノ酸配列において1若しくは数個のアミノ酸が欠失、置換若しくは付加されたアミノ酸配列からなり、かつコレステロール3-硫酸又はその塩に対する結合活性を有するタンパク質である、請求項3に記載の免疫制御剤。
- DOCK2によるRac活性化の前記制御剤が、配列番号1に記載のアミノ酸配列からなるタンパク質の発現ベクター、又は配列番号1に記載のアミノ酸配列において1若しくは数個のアミノ酸が欠失、置換若しくは付加されたアミノ酸配列からなり、かつコレステロール3-硫酸又はその塩に対する結合活性を有するタンパク質の発現ベクターである、請求項1に記載の免疫制御剤。
- DOCK2によるRac活性化の前記制御剤が、
配列番号2に記載のアミノ酸配列からなるタンパク質、又は配列番号2に記載のアミノ酸配列において1若しくは数個のアミノ酸が欠失、置換若しくは付加されたアミノ酸配列からなり、かつコレステロールの3位の炭素に硫酸基を付加する活性を有するタンパク質、
配列番号2に記載のアミノ酸配列からなるタンパク質の発現ベクター、又は配列番号2に記載のアミノ酸配列において1若しくは数個のアミノ酸が欠失、置換若しくは付加されたアミノ酸配列からなり、かつコレステロールの3位の炭素に硫酸基を付加する活性を有するタンパク質の発現ベクター、
配列番号2に記載のアミノ酸配列からなるタンパク質の発現を阻害するsiRNA、shRNA、miRNA、リボザイム又はアンチセンス核酸、又は
配列番号2に記載のアミノ酸配列からなるタンパク質の阻害剤

配列番号3に記載のアミノ酸配列からなるタンパク質の発現ベクター、又は配列番号3に記載のアミノ酸配列において1若しくは数個のアミノ酸が欠失、置換若しくは付加されたアミノ酸配列からなり、かつコレステロール-3硫酸の硫酸基を除去する活性を有するタンパク質の発現ベクター、
配列番号3に記載のアミノ酸配列からなるタンパク質の発現を阻害するsiRNA、shRNA、リボザイム又はアンチセンス核酸、又は
