15. 芳香族化合物がサリチル酸、4-ヒドロキシ安息香酸、3-ヒドロキシ安息香酸、4-アミノ安息香酸、2-アミノ安息香酸、L-チロシン、又はフェノールであり、誘導体がムコン酸、ゲンチジン酸、マレイルピルビン酸、又はマレイン酸である、上記14記載の方法。
16. 炭素源としてグルコースのみを添加する、上記14又は15記載の方法。
微生物のホスホトランスフェラーゼ系酵素をコードする遺伝子の発現を抑制し、
該微生物のピルビン酸キナーゼをコードする遺伝子の発現を抑制し、そして
該微生物がコリスミ酸又はイソコリスミ酸から芳香族化合物を合成することを可能とする酵素をコードする1若しくは複数の遺伝子を微生物に導入する、
ことを含む、上記方法を提供する。
(1)微生物のホスホトランスフェラーゼ系酵素をコードする遺伝子の発現を抑制する、
(2)該微生物のピルビン酸キナーゼをコードする遺伝子の発現を抑制する、
(3)該微生物がコリスミ酸又はイソコリスミ酸から芳香族化合物を合成することを可能とする酵素をコードする1若しくは複数の遺伝子を微生物に導入する、
との3つのステップに分けて記載したが、この記載は、微生物の改変の順序を説明するものではなく、いかなる順序で実施しても、また同時に行っても良いことは理解されるであろう。
本実施例で使用する細胞株及びプラスミドを表1及び表2に記載する。遺伝子のクローニングのためには大腸菌 NovaBlue コンピテント細胞 (Novagen, Cambridge, MA) を使用した。尚、表中で供給元の記載がないものは、本発明者等が作製したものである。
ポリメラーゼ連鎖反応(PCR)はKOD FX Neo (TOYOBO, Osaka, Japan)及び表3に示すプライマー対を用いて実施した。それぞれの遺伝子を、Gibson Assembly (New England Biolabs, Ipswich, MA)を用いて各プラスミドとアセンブルした。pCFTdeltain(図2)は市販品を購入した(Invitrogen, San Diego, CA)。
5 mLの試験管でのサリチル酸の製造のためにはM9最小培地を用いた。M9最小培地は、1リットル当たり20 g グルコース、0.5 g NaCl、17.1 g Na2HPO4・12H2O、3 g KH2PO4、2 g NH4Cl、246 mg MgSO4・7H2O、14.7 mg CaCl2・2H2O、2.78 mg FeSO4・7H2O、10 mg 塩酸チアミン、40 mg L-チロシン及び40mg L-トリプトファンを含有する (ATCC31882はL-チロシン及びL-トリプトファン要求性を示す)。必要に応じてアンピシリン、カナマイシン、及び/又はクロラムフェニコールをそれぞれ100、50、及び15μg/mLの最終濃度まで添加した。CFT5株の培養においては、L-フェニルアラニンを100 mg/Lの最終濃度まで添加した。また、CFT3株及びCFT5株の前培養においては、増殖を促進するために培地に0.2% ピルビン酸ナトリウムを添加した。前培養培地を5 mL試験管中のM9培地に0.05の初期光学密度(OD600)で植菌した。試験管培養は、180 rpmで振とうしながら37℃で実施した。0.1 mM IPTGは最初に培地に添加した。
培養上清中のグルコース濃度は、Glucose CII test Wako (Wako, Kyoto, Japan)を使用して測定した。ピルビン酸ナトリウムを添加する場合、収率の評価のためにグルコース消費量の他にピルビン酸ナトリウムの消費量も考慮した([化合物生成量]-mol/[グルコース及びピルビン酸消費量]-mol%)。
コリスミ酸誘導体の産生のために大腸菌の形質転換を試みた。親株として、大腸菌のL-フェニルアラニン過剰産生株(ATCC31882)を使用した。
ATCC31882株を用い、そのサリチル酸合成能を高めるために、ICS及びIPLをコードする遺伝子を導入した。本実施例ではICSをコードする遺伝子としてmenF、entC及びpchAを使用し、IPLをコードする遺伝子としてpchBを使用した。
ATCC31882株の代わりに、実施例1で作製した変異株に、実施例2で作製したプラスミドを導入してサリチル酸の生成能を検討した。
CFT3株の変異に加えてpheA及びtyrAを欠損しているCFT5株にpZE12mIを導入して、menF及びpchBを発現できる株を作製した。本明細書において、これをCFT51株と記載する。
得られた細胞株を用い、大量培養によるサリチル酸の製造を行った。
バッチ培養は2.0リットルのジャーファーメンター(作業容積400 mL)で実施した。ジャーファーメンターでのサリチル酸の製造のために、0.4% 酵母抽出物を含むM9培地からなるM9Y培地を使用した。4mLの前培養培地を、400 mLの培地を入れたジャーファーメンターに入れた。培養中のpHを7.0に維持するために、NH4OHを自動的に添加するようにした。撹拌速度を自動制御し、37℃で酸素を供給することで、溶存酸素(DO)濃度を3.0 ppm以上に維持した。グルコースの初期濃度は40 g/Lとした。5時間培養した後に0.1 mM IPTGを培地に添加した。
CFT51株の親株であるCFT5株は、細胞内にコリスミ酸を蓄積することが想定された。従って本発明者等は、次にCFT5株に基づいて他のコリスミ酸誘導体の製造を試み、この株の多用途性を実証し、芳香族化合物製造のためのプラットフォームを構築することができた。
実施例6と同様にサリチル酸の構造異性体である3-ヒドロキシ安息香酸(3HBA)の生成について検討した。コリスミ酸から3HBAを生成することを可能とするために、S. hygroscopicus 由来の3-ヒドロキシ安息香酸シンターゼ (EC 4.1.3.45)をコードする遺伝子(配列番号40、Hyg5)を導入した。
本発明の細胞株による4-アミノ安息香酸(PABA)の生成について検討した。コリスミ酸からPABAを製造するために、大腸菌由来のアミノデオキシコリスミ酸シンターゼ(EC 2.6.1.85)及び4-アミノ-4-デオキシコリスミ酸リアーゼ (EC 4.1.3.38) (PabA、PabB、及びPabC)を用いた。
本発明の細胞株による2-アミノ安息香酸(2ABA)の生成について検討した。コリスミ酸から2ABAを製造するために、大腸菌のL-トリプトファン生合成経路の一部を利用することができるため、大腸菌由来のアントラニル酸シンターゼ(EC 4.1.3.27)をコードする遺伝子(trpE及びtrpG)を導入した。
CFT5株を用い、L-チロシンを産生できる大腸菌株を作製した。コリスミ酸からL-チロシンを製造するために、大腸菌由来のコリスミ酸ムターゼ/プレフェン酸デヒドラターゼ(EC 5.4.99.5 及び EC 4.2.1.51)をコードする遺伝子(配列番号43、tyrAfbr)を導入した。
実施例10で得られたL-チロシン過剰産生株を用い、L-チロシンからチロシンフェノールリアーゼ(TPL) [EC 4.1.99.2]によって合成され得るフェノールを製造する株を作製した。
サリチル酸 1-モノオキシゲナーゼ (SMO) 及び カテコール 1,2-ジオキシゲナーゼ (CDO) によって触媒される反応によって、サリチル酸からムコン酸の生成が可能である(図7)。
本発明の細胞株を利用して、コリスミ酸から3-ヒドロキシ安息香酸、ゲンチジン酸、マレイルピルビン酸を介してマレイン酸を生成する株を作製した(図9)。
実施例1で作製したCFT5株に、3-ヒドロキシ安息香酸シンターゼをコードする遺伝子、3-ヒドロキシ安息香酸6-ヒドロキシラーゼをコードする遺伝子、ゲンチジン酸-1,2-ジオキシゲナーゼ、及びマレイルピルビン酸ヒドロラーゼをコードする遺伝子を導入した。
Claims (16)
- 糖代謝経路が改変された微生物の製造方法であって、
微生物のホスホトランスフェラーゼ系酵素をコードする遺伝子の発現を抑制し、
該微生物のピルビン酸キナーゼをコードする遺伝子の発現を抑制し、そして
該微生物がコリスミ酸又はイソコリスミ酸から芳香族化合物を合成することを可能とする酵素をコードする1若しくは複数の遺伝子を微生物に導入する、
ことを含む、上記方法。 - 更にコリスミ酸からL-フェニルアラニンを合成する酵素をコードする遺伝子の発現を抑制することを含む、請求項1記載の方法。
- 芳香族化合物がサリチル酸であり、イソコリスミ酸シンターゼをコードする遺伝子及びイソコリスミ酸ピルビン酸リアーゼをコードする遺伝子を導入する、請求項1又は2記載の方法。
- 芳香族化合物が4-ヒドロキシ安息香酸であり、コリスミ酸ピルビン酸リアーゼをコードする遺伝子を導入する、請求項1又は2記載の方法。
- 芳香族化合物が3-ヒドロキシ安息香酸であり、3-ヒドロキシ安息香酸シンターゼをコードする遺伝子を導入する、請求項1又は2記載の方法。
- 芳香族化合物が4-アミノ安息香酸であり、アミノデオキシコリスミ酸シンターゼをコードする遺伝子及び4-アミノ-4-デオキシコリスミ酸リアーゼをコードする遺伝子を導入する、請求項1又は2記載の方法。
- 芳香族化合物が2-アミノ安息香酸であり、アントラニル酸シンターゼをコードする遺伝子を導入する、請求項1又は2記載の方法。
- 芳香族化合物がL-チロシンであり、コリスミ酸ムターゼをコードする遺伝子及びプレフェン酸デヒドラターゼをコードする遺伝子を導入する、請求項1又は2記載の方法。
- 芳香族化合物がフェノールであり、チロシンフェノールリアーゼをコードする遺伝子を導入する、請求項1又は2記載の方法。
- 更にサリチル酸1-モノオキシゲナーゼをコードする遺伝子及びカテコール1,2-ジオキシゲナーゼをコードする遺伝子を導入する、請求項3記載の方法。
- 更に3-ヒドロキシ安息香酸からゲンチジン酸、マレイルピルビン酸、及び/又はマレイン酸を合成することを可能とする1種以上の遺伝子を導入する、請求項5記載の方法。
- 請求項1~11のいずれか1項記載の方法によって得られる、糖代謝経路が改変された微生物。
- 微生物が細菌類、酵母類、及び真菌類から選択される、請求項12記載の微生物。
- 請求項12又は13記載の微生物を培養し、培養液から芳香族化合物又はその誘導体を回収することを含む、芳香族化合物又はその誘導体の製造方法。
- 芳香族化合物がサリチル酸、4-ヒドロキシ安息香酸、3-ヒドロキシ安息香酸、4-アミノ安息香酸、2-アミノ安息香酸、L-チロシン、又はフェノールであり、誘導体がムコン酸、ゲンチジン酸、マレイルピルビン酸、又はマレイン酸である、請求項14記載の方法。
- 炭素源としてグルコースのみを添加する、請求項14又は15記載の方法。
