Description

一株短小芽孢杆菌及其保育仔猪用复合微生态制剂 技术领域
本发明涉及微生物技术领域,尤其是涉及一株短小芽孢杆菌,及其在有益于保育仔猪方面的应用。
背景技术
保育仔猪,简称断奶仔猪,此阶段的猪相当脆弱,母源抗体消失,新的免疫系统还没有建立,再加上生长速度快、应激因素多等原因,很容易发生各种疾病甚至出现大面积死亡。抗生素、化学药品虽然能在较短时间内解决出现的问题,但由此引发的药物抗性和药物残留问题,有可能会对仔猪后期生长产生影响。如何提高保育仔猪健康状况、成活率、整齐度、充分发挥生长后效性,是每个养殖户共同关心的问题。
微生态制剂具有调控动物肠道微生物平衡、增殖有益菌、抑制有害菌、降低肠道pH值、提高消化道酶活性、营养物质消化率和机体免疫力等作用,关于微生态制剂在仔猪养殖中的应用报道日益增多。CN 101703136B报道了植物乳杆菌活菌制剂在防治仔猪拉稀上的应用,仔猪拉稀有效率在90%以上,治愈率80%以上。CN 103255080B报道了凝结芽孢杆菌(Bacillus coagulans HM-09)在制备改善乳仔猪生产性能和腹泻的饲料中的应用,可以减少断奶仔猪腹泻的发生率,进而减少肠源性疾病的发生,提高饲料消化率,提高仔猪饲料利用率。CN 103205373B报道了一株用于乳仔猪养殖的丁酸梭菌,可在乳仔猪肠道内拮抗动物病原菌,维持和调节微生态平衡,增强机体免疫功能,并产生淀粉梅、维生素,促进动物对饲料的消化吸收和营养作用。
一般来说,微生态制剂起效慢,畜禽养殖过程中需要坚持使用才能收到很好的效果。因此,开发一种既能长效保健用、又能快速解决仔猪拉稀、下痢等问题的微生态制剂,对于实现保育仔猪平稳生产显得尤为必要。
发明内容
针对现有技术存在的上述问题,本申请人提供了一株短小芽孢杆菌及其保育仔猪用复合微生态制剂。本发明既能快速解决仔猪拉稀、下痢等问题,又能在 长期使用过程中提升保育仔猪的健康水平。
本发明的技术方案如下:
一株短小芽孢杆菌(Bacillus pumilus)ZS008,保藏于中国典型培养物保藏中心,保藏编号为CCTCC No.M2015451,保藏日期为2015年7月13日。
一种保育仔猪用复合微生态芽孢杆菌制剂,由以下菌种组合配制成为复合菌剂:短小芽孢杆菌CCTCC No.M2015451、凝结芽孢杆菌CCTCC No.M2015273、枯草芽孢杆菌CCTCC No.M 2011443、地衣芽孢杆菌CCTCC No.M 2012458。
所述复合微生态芽孢杆菌制剂的有效活菌数为1.4×108-7.0×1010cfu/g,各菌种的组成成分如下:

所述凝结芽孢杆菌为凝结芽孢杆菌(Bacillus coagulans)ZS007,保藏于中国典型培养物保藏中心,保藏编号为CCTCC NO:M 2015273。
所述的保育仔猪用复合微生态芽孢杆菌制剂的应用在于提升保育仔猪的生产性能。使用量为1.0~5.0g/头·天。
枯草芽孢杆菌CCTCC No.M 2011443、地衣芽孢杆菌CCTCC No.M 2012458的单菌粉购自泰州绿圣元生物科技有限公司,其产品规格均为5.0×1010cfu/g
本发明有益的技术效果在于:
(1)能快速解决仔猪拉稀、下痢等问题;
(2)长期使用后,可以提升保育仔猪的健康水平,从而达到提升保育仔猪生产能力的效果。
具体实施方式
1、短小芽孢杆菌的筛选
从江苏无锡方鼎鼎养猪场附近的土壤样品称取2g,加入98ml无菌生理盐水,80℃热处理10min后,在37℃摇床上120rpm条件下振荡20min,吸取上清液1ml加入LB培养基中稀释涂布进行菌株分离。通过对菌株进行形态学观察、生理生化实验、16S rDNA分析等手段有效而准确的确定菌种的属种。
最终得到编号为ZS008的菌种,鉴定为短小芽孢杆菌(Bacillus pumilus)。该菌种保藏于中国典型培养物保藏中心,保藏编号为CCTCC No.M2015451,保藏日期为2015年7月13日。
2、凝结芽孢杆菌的筛选
从江苏无锡马山某奶牛养殖场附近的土壤样品称取2g,加入98ml无菌生理盐水,80℃热处理10min后,在37℃摇床上120rpm条件下振荡20min,吸取上清液1ml加入LB培养基中稀释涂布进行菌株分离。通过对菌株进行形态学观察、生理生化实验、16S rDNA分析等手段有效而准确的确定菌种的属种。
最终得到编号为ZS-007的菌种,鉴定为凝结芽孢杆菌(Bacillus coagulans)。
ZS-007于2015年5月3日保藏于中国典型培养物保藏中心,保藏编号为CCTCC M 2015273。
3、复合微生态制剂的制备
复合微生态制剂由短小芽孢杆菌CCTCC No.M2015451、凝结芽孢杆菌CCTCC No.M 2015273、枯草芽孢杆菌CCTCC No.M 2011443、地衣芽孢杆菌CCTCC No.M 2012458复配而成。
其中枯草芽孢杆菌CCTCC No.M 2011443、地衣芽孢杆菌CCTCC No.M 2012458的单菌粉采购自泰州绿圣元生物科技有限公司,其产品规格均为5.0×1010cfu/g。
实施例1~3所配制的微生态制剂产品,各微生物含量如下表1:

单菌粉按照既定比例混合均匀后,添加无水葡萄糖作为载体,补充到总重量的100%(以芽孢杆菌作为有效成分,加入载体后使产品总重量满足最终活菌数达到4.0×108cfu/g,混合均匀后备用。
加工过程应保证操作环境的清洁,避免污染。控制环境湿度70%以下,各原料水分含量低于13%,该产品制成后常温下保存24个月,-18℃下保存期限可以达到60个月以上,保质期内活菌数稳定。
4、复合益生菌剂在保育仔猪上的应用效果
选择自繁自养同批的杜×长×大三元杂交断奶仔猪90头。依据胎次、体重、公母比例相同或相似的原则,将小猪分为A、B、C 3个组,其中A组为对照组,B试验组1,C组为试验组2,每组设2个重复,每个重复15头,随机分组,分头编号;各组基础日粮相同(正大全价料),营养水平一致。A组不添加任何酶制剂,B组基础日粮的基础上添加含有短小芽孢杆菌的复合微生态制剂(实施例1),C组同B组、但不含有短小芽孢杆菌;微生态制剂用量为的用量为1.0g/头·天。仔猪21日龄,平均体重6.5kg断奶,预实验7天,饲养试验30天。饲养地点南京某养殖合作社。
试验结果如表2~4所示。

仔猪发生拉稀现象后,按每头猪5g的份量使用微生态制剂产品进行治疗。产品用冷开水10mL搅拌后,抽入注射器,并用注射器沿仔猪口角灌服。微生态制剂产品中含有短小芽孢杆菌时,仔猪4-6hr后精神好转,24hr内拉稀终止,恢复食欲。微生态制剂产品中不含有短小芽孢杆菌时,仔猪12hr后精神好转;巩固治疗一次,2天左右拉稀终止,恢复食欲。

  1. 一株短小芽孢杆菌(Bacillus pumilus)ZS008,保藏于中国典型培养物保藏中心,保藏编号为CCTCC NO.M2015451,保藏日期为2015年7月13日。
  2. 一种保育仔猪用复合微生态芽孢杆菌制剂,其特征在于由以下菌种组合配制成为复合菌剂:短小芽孢杆菌CCTCC NO.M2015451、凝结芽孢杆菌CCTCC NO.M2015273、枯草芽孢杆菌CCTCC NO.M2011443、地衣芽孢杆菌CCTCC NO.M2012458。
  3. 根据权利要求2所述的保育仔猪用复合微生态芽孢杆菌制剂,其特征在于所述复合微生态芽孢杆菌制剂的有效活菌数为1.4×108-7.0×1010cfu/g,各菌种的组成成分如下:
