시각을 담당하는 우리의 눈은 생활에 필요한 대부분의 정보를 받아들이는 중 요한 감각기관이다. 눈의 앞 표면은 각막과 결막이 있고, 안구를 둘러싼 공막 안에 홍채와 모양체 , 수정체, 그리고 눈의 공모양을 유지시켜 주는 초자체, 뒤쪽의 망막 으로 이루어져 있다, 수정체와 유리체 및 안방수는 굴절매질에 해당된다. 질병으로 인한 눈의 시력장애 또는 시력 상실은 생활의 질을 떨어뜨리는 가장 큰 요인 중의 하나이다.
안구건조증 (Dry eye syndrome)은 건성안 (Dry eye) 또는 건조성 각결막염 (Keratoconjunct ivi t is sicca)라고 하며, 눈물의 결핍 또는 과다한 증발로 인하여 안구 표면 즉 각막과 결막이 건조해지는 질환으로, 눈물막의 장애로 알려져 있다. 국제 안구건조증 워크샵 ( Internat ional Dry Eye Workshop , DEWS) 및 미국의 눈 연 구소 (Nat ional Eye Inst i tute , NEI )에 의하면 안구건조증은 눈의 불편함, 눈물막 불안정성, 시력의 저하를 유발하는 눈물 및 눈 표면의 질환이다. 안구표면의 건조 는 눈물 막으로 보호받고 있던 각막 상피층에 손상 및 염증 반응에 영향을 미친다. 눈물 층의 증발량의 증가 또는 눈물샘에서 분비가 적어 눈물 층의 양이 부족해지면 눈물 막의 삼투압 증가, 눈물 부족으로 인한 눈 표면의 마찰력 증가로 인한 각막 상피층의 손상으로 인한 염증을 동반한다. 안구건조증은 수성 눈물-결핍성 안구건 조증 및 증발성 안구건조증으로 나눌 수 있다.
수성 눈물-결핍성 안구건조증은 눈물샘에서 분비되는 눈물의 부족 때문이다. 증발성 안구건조증은 눈물샘 분비는 정상적으로 기능을 하지만, 결막의 술잔세포
(Goblet cel l ) , 눈꺼풀의 메이봄샘의 분비능력이 저하되어 눈물에서 점성물질의 부 족으로 눈 표면으로부터의 과다한 수분 상실 때문이다. 이러한 원인은 안검 구조 또는 원동력 (dynamic) 에 영향을 주는 내재적 질병으로 인한 내인성과, 일부 외부 적 노출로 인해 눈 표면의 눈물 층이 빨리 파괴되는 외인성이 있다.
안구건조증을 야기하는 인자는 매우 다양하며, 예를 들면 논물 분비의 저하, 눈물의 과도한 증발, 눈물이 생성되는 기관의 염증, 쇼그렌 증후군 또는 스티브존 슨 증후군 등의 전신질환이 수반되는 경우, 스마트폰 또는 태블릿 PC등의 장기간 사용, 및 호르몬의 급격한 변화 등을 들 수 있다.
현재, 안구건조증의 치료를 위하여, 인공 누액 및 자각 증상을 경감할 목적 의 콘드로틴 술페이트, 글루타티온, 히알루론산, 피브로넥틴, 혈청 안약 등이 투여 되지만 아직까지 그 효과가 충분하지 않다. 따라서, 안구건조증 치료를 위한 효과 적인 치료제 개발의 필요성이 있어왔다.
한편, 이마티닙 ( Imat inib)은 백혈병 치료제인 글리백 (Gleevec)으로 잘 알려 져 있으며, 만성 골수성 백혈병 (CML) 치료제이고, 스위스의 노바티스 (Novart i s)사 가 개발하였다. 이마티닙은 만성 골수성 백혈병을 일으키는 주원인인 필라델피아 유전자 (Bcr 유전자와 Abl 유전자의 염색체 전위로 인해 만들어지는 암유전자)에 의 해 만들어지는 티로신 카이네이즈의 활성을 선택적으로 억제한다. 또한, 이마티닙 은 만성 골수성 백혈병 외에도 위암과 관련된 위장관 기저 종양 (GIST)에도 약효가 있다고 알려져 있다. 하지만, 안구건조증 또는 안구건조증과 관련된 질환의 치료 또는 예방효과에 대해서는 현재까지 알려진 바 없다.
【발명의 상세한 설명】
【기술적 과제】
본 발명자들은 이마티닙의 새로운 용도인 안구건조증 및 안구건조증과 관련 된 안과 질환 치료 효과를 발견하여 본 발명을 완성하였다. 이에 눈물막의 부족으 로 인한 안구 표면의 건조 상태에서, 각막상피의 손상과 이와 동반한 염증을 억제 하기 위하여, 이마티닙을 점안제로하여 안구건조증을 치료하고자 한다. 이에 본 발명자들은 이마티닙을 유효성분으로 포함하는 안구건조증 또는 안 구건조증과 관련된 안과 질환의 예방 또는 치료용 약학적 조성물을 개발하여 본 발
명을 완성하였다. 따라서, 본 발명의 목적은 이마티닙을 유효성분으로 포함하는 안구건조증 또 는 안구건조증과 관련된 안과 질환의 예방 또는 치료용 약학적 조성물을 제공하는 것이다. 본 발명의 또 다른 목적은, 이마티닙을 유효성분으로 포함하는 점안제를 제 공하는 것이다. 본 발명의 또 다른 목적은, 이마티닙을 필요로 하는 개체에 유효량으로 투여 하여 안구건조증 또는 안구건조증 연관 안과 질환을 치료하는 방법을 제공하는 것 이다. 본 발명의 또 다른 목적은, 이마티닙의 안구건조증 또는 안구건조증 연관 안 과 질환 치료제 제조를 위한 용도를 제공하는 것이다. ᅳ
【기술적 해결방법】
상기의 목적을 달성하기 위해, 본 발명은 이마티닙을 유효성분으로 포함하는 안구건조증 또는 안구건조증과 관련된 안과 질환의 예방 및 치료용 약학 조성물을 제공한다. 본 발명의 또 다른 목적을 달성하기 위해, 본 발명은 이마티닙을 유효성분으 로 포함하는 안구건조증 또는 안구건조증과 관련된 안과 질환의 예방 및 치료용 점 안제를 제공한다. 본 발명의 또 다른 목적을 달성하기 위해, 본 발명은 이마티닙을 필요로 하 는 개체에 유효량으로 투여하여 안구건조증 또는 안구건조증 연관 안과 질환을 치 료하는 방법을 제공한다. 본 발명의 또 다른 목적을 달성하기 위해, 본 발명은 이마티닙의 안구건조증 또는 안구건조증 연관 안과 질환 치료제 제조를 위한 용도를 제공한다.
이하 본 발명을 상세히 설명한다. 본 발명에서 사용돠는 "안구 건조증 (xerophthalmia)"은 건성안 (Dry eye) 또 는 건조성 각결막염 (Keratoconjunctivitis sicca)라고 하며 , 눈물이 부족하거나 눈 물이 지나치게 증발하거나 눈물 구성성분의 균형이 맞지 않아서, 안구 표면이 손상 되고 눈이 시리고 자극감, 이물감, 건조감 같은 자극증상을 느끼게 되는 눈의 질환 을 말하는 것이다. 본 발명에서 "이마티닙 (Imatinib)"은 백혈병 치료제 글리백 (Gleevec)으로 잘 알려져 있으며, 만성 골수성 백혈병 (CML) 치료제이고, 스위스의 노바티스 (Novartis)사가 개발하였다. 이마티닙은 필라델피아 유전자 (Bcr 유전자와 Abl 유전 자의 염색체 전위로 인해 만들어지는 암유전자)에 의해 만들어지는 티로신 카이네 이즈의 활성을 선택적으로 억제하며 구조는 하기 화학식 I과 같다.

본 발명에서 "각결막염' '은 세균, 바이러스, 알레르기, 또는 환경적 요인에 의해 각막과 결막에 발생하는 결막의 염증 질환으로서, 가장 일반적으로 발병되는 안질환이며, 보통은 잘 치유되나 경우에 따라서는 치명적이어서 조직손상에 의한 실명까지 초래할 수 있다. 각결막염은 크게 감염성 (infectious conjunctivitis) 및 비감염성 (non-infectious conjunct ivit is)으로 분류된다. 본 발명은 이마티닙을 유효성분으로 포함하는 안구건조증 또는 안구건조증
연관 안과 질환의 예방 또는 치료용 약학 조성물을 제공한다. 본 발명에서 안구 건조증 연관'안과 질환으로는 건성 각결막염, 건성으로 인 한 각막 궤양, 안검염 (눈꺼풀염증), 눈의 충혈, 각막신생혈관 생성 일 수 있다. 본 발명에서 안구건조증은 수성 눈물-결핍성 안구건조증 또는 증발성 안구건 조증인 것을 특징으로 한다.
본 발명에서 "수성 눈물-결핍성 안구건조증' '이란 눈물 자체 또는 눈물의 한 가지 성분이 부족하여 안구 표면이 건조하게 되고 이로 인해 증상이 유발되는 안구 건조증을 의미하며, "증발성 안구 건조증 "이란 눈물막이 과도하게 증발되는 현상으 로 인해 눈물막이 안구 표면에 층분한 윤활작용을 나타내지 못하여 안구 표면이 건 조하게 되고 이로 인해 불쾌감 및 자극 증상이 유발되는 질환이다. 본 발명의 이마티닙 함량은 조성물 전체 중량을 기준으로 0.005 중량 % ᅳ 5 중량 %인 것올 특징으로 한다. 즉, 본 발명의 조성물은 0.005 중량 % - 5 중량 %의 이 마티닙과 95 중량 - 99.995 중량 )의 담체를 흔합할 수 있다. 본 발명의 상기 조성물은 현탁액, 산제, 분말제, 과립제, 정제, 서방형 제 제, 주사제, 연고제, 점안제, 캡슐제, 콘텍트렌즈 세정제 또는 콘텍트렌즈 윤활제 형태임을 특징으로 한다. 상기 제제에는 약제학적으로 허용되는 담체, 회석제, 부형제, 또는 이들의 조합이 필요에 따라 포함될 수 있다. 이들 제제는 생물체내로 활성 성분의 투여를 용이하게 한다. 상기 담체 (carr i er )는 세포 또는 조직 내부로의 화합물의 부가를 용이하게 하는 화합물로 정의된다. 상기 담체는 제형에 따라 통상 사용되는 종류에서 특별한 제한 없이 사용할 수 있으나, 바람직하게는 전분, 젖당, 만니를, 카르복시메틸셀를 로오스, 콘스타치, 및 무기염 등의 고형 담체; 증류수 생리식염수, 포도당 수용 액, 에탄을 등의 알코올, 프로필렌글리콜, 및 폴리에틸렌글리콜 등의 액체담체; 및 각종 동식물유, 백색바샐린, 파라핀 및 왁스 등의 유성담체; 로 구성된 군에서 선 택된 적어도 하나 이상일 수 있다.
상기 부형제는, 예를 들어, 락토스, 수크로즈, 만니를, 또는 소르비를과 같 은 필러 옥수수 녹말, 밀 녹말, 쌀 녹말, 감자 녹말, 겔라틴, 검 트래거켄스, 메틸 셀를로우즈, 히드록시프로필메틸-셀를로우즈, 소듐 카르복시메틸 셀를로우즈, 및 / 또는 폴리비닐피를리돈 (PVP)와 같은 셀를루오즈계 물질 등일 수 있다. 상기 약학적 제제화는 공지의 방법으로 수행할 수 있고, 바람직하게는, 약제 학적으로 가능한 경구, 외용, 경피 경점막 제제의 형태로서, 분말제, 과립제, 액 제, 환제, 트로키 (troche) , 현탁제, 유제, 시럽제 정제, 산제, 경질 또는 연질의 캅셀제, 현탁제, 주사용 제제, 유화액, 비경구 투여용의 단위 투여형 또는 수회 투 여형 제제일 수 있고, 더욱 바람직하게는, 경구용 제제 또는 점안제 (eye drop)일 수 있다. 본 발명은 이마티닙을 유효성분으로 포함하는 안구건조증 또는 안구건조증과 관련된 안과 질환의 예방 또는 치료용 점안제를 제공한다. 본 발명의 점안제는 점증제를 추가로 포함할 수 있다. 본 발명에서 점증제는 당업계에서 일반적으로 사용되는 의약용 또는 식용의 점증제를 의미하며, 키토산, 히알루로닉산, 히드록시프로필메칠셀를로즈 (HPMC) , 히드록시프로필샐를로즈 (HPC) , 히드록시에칠셀를로즈 (HEC) , 폴리비닐피롤리돈 (PVP) , 카르복시메칠셀를로즈 (CMC) , 카보머 (carbomer ) , 글리세린 (glycer in), 폴리에칠렌옥사이드 (PE0), 히알루론산 (Hyaluroni c acid) 등이 있지만 이에 한정되지는 않는다. 본 발명의 점안제는 수용액을 포함하는 담체를 추가적으로 포함하며, 액상인 것을 특징으로 하며, 상기 수용액을 포함하는 담체는 증류수, 인산완층액, 균형염 용액 및 생리식염수로 이루어진 군에서 선택된 하나 이상의 약학적으로 허용되는 담체인 것을 특징으로 한다.
본 발명의 점안제 조성물은 또한 약제학적으로 허용 가능한 담체를 포함할 수 있다. 본 발명에서 용어, "약학적으로 허용가능한 담체" 란 생물체를 상당히 자 극하지 않고 투여 유효성분의 생물학적 활성 및 특성을 저해하지 않는 담체 또는 회석제를 말한다. 상기 약제학적으로 허용가능한 담체의 예로는 등장화제, 완충제, 안정제, ί)Η 조절제 및 용제 등이 있다. 상기 용제는 증류수, 인산완층액 (phosphate
buf fered sal ine) , 균형염 용액 (balanced sal t solut ion) , 생리식염수 (sal ine) 또 는 이들의 흔합물 등을 포함할 수 있다. 이때 사용되는 함량은 제조하고자 하는 점 안액의 총 용량에 필요한 양에 맞추어 사용량을 조절할 필요가 있다. 즉, 점안액의 경우에는 전체 점안액 중 상기 점안제 조성물 외에 용제로 사용량을 조절한다. 본 발명에 따른 점안제는 액제의 체형인 것이 바람직하다. 본 발명의 점안제는 안구건 즈증 또는 안구 건조증과 관련된 안구 질환의 치료 용도로 사용됨을 특징으로 한 다. 본 발명의 점안제 조성물은 주성분으로서의 이마티닙을, 0.05% - 0.5% 히알 루론산 또는 카르복시메틸셀를로오스에 혼합하거나, 보다 더 바람직하게는 0. 1%의 히알루론산 또는 카르복시메틸셀롤로오스에 혼합하여, 또는 인공눈물과 병용 투여 로 사용함으로써 안구건조증 또는 이와 관련된 안구 질환의 치료 효과를 극대화할 수 있다. 상기 중량비는 악제학적으로 볼 때, 상기 범위 미만으로 사용될 경우 점 탄성이 너무 낮아 습윤 및 윤활작용에 부적절한 문제가 발생하고, 상기 범위를 초 과하는 경우 과다한 고점탄성으로 침투성에 부적절한 문제가 발생하여 점안제로서 의 적합한 유동성을 확보하기 어려운 문제가 있다. 본 발명의 약학 조성물 또는 점안제는 국소 투여 또는 점안 투여되는 것을 특징으로 한다. 본 발명에서 용어, "투여 " 는 어떠한 적절한 방법으로 환자에게 본 발명의 약제학적 조성물을 도입하는 것을 의미하며, 본 발명의 조성물의 투여 경로는 목적 조직에 도달할 수 있는 한 경구 또는 비경구의 다양한 경로를 통하여 투여될 수 있 다. 바람직하게는 안구에 국소적으로 투여하는 것이다. 본 발명의 치료방법은 이마 티닙을 약학적 유효량으로 투여하는 것을 포함한다. 적합한 총 1일 사용량은 을바 른 의학적 판단범위 내에서 처치의에 의해 결정될 수 있다는 것은 당업자에게 자명 한 일이다. 특정 환자에 대한 구체적인 치료적 유효량은 달성하고자 하는 반웅의 종류와 정도, 경우에 따라 다른 제제가 사용되는지의 여부를 비롯한 구체적 조성 물, 환자의 연령, 체중, 일반 건강 상태, 성별 및 식이, 투여 시간, 투여 경로 및 조성물의 분비율, 치료 기간, 구체적 조성물과 함께 사용되거나 동시 사용되는 약 물을 비롯한 다양한 인자와 의약 분야에 잘 알려진 유사 인자에 따라 다르게 적용 하는 것이 바람직하다.
본 발명은 원래 사람의 치료에 관계되는 것이긴 하지만, 수의 목적으로 개 및 고양이 등과 같은 다른 포유동물을 대상으로 필요로 하는데 사용될 수 있다. 투약 단위는, 예를 들면 개별 투약량의 1, 2ᅳ 3 또는 4 배를 함유하거나 또 는 1/2, 1/3 또는 1/4 배를 함유할 수 있다. 개별 투약량은 바람직하게는 유효 약 물이 1 회 (1-2 방울, 한방울 에 투여되는 양을 함유하며, 이는 통상 1 일 투 여량의 전부, 1/2, 1/3 또는 1/4 배에 해당한다. 이마티닙의 유효용량은 농도 의존 적이나 바람직하게는 0.005 내지 5 중량 %이고, 더욱 바람직하게는 0.01 내지 1 중 량%이며, 하루 1 - 6 회 투여할 수 있다. . 본 발명의 실시예 1에서는 건조성 각결막염 모델에 이마티닙 점안제를 처리 하였을 때, 각막 상피 보호효과가 있다는 것을 확인하였다. 이 때 각막 손상의 확 인은 1 > 리사민그린 용액으로 각막 표면을 염색하여 손상된 부위를 확인하였다. 그 결과 대조군에 비하여 이마티닙을 투여한 군과 Cyclospor in A를 투여한 군에서 현 저하게 각막 손상이 억제된 것을 확인할 수 있었다 (도 2 참조) . 또한 농도의존적으 로 효과가 있음을 확인 하였다 (도 3참조) .
본 발명의 실시예 2에서는 건조성 각결막염에 이마티닙 점안제를 처리하였을 때, 각막 변성 보호효과가 있다는 것을 확인하였다. 이 때 각막 변성을 관찰하기 위해 F— act in에 대한 염색을 실시하였다. 그 결과 대조군에서는 각막상피세포가 각 질화되는 현상이 관찰되었지만, 이마티닙을 처리한 군에서는 각질화 현상이 발견되 지 않았다 (도 4 참조) .
본 발명의 실시예 3에서는 건조성 각결막염 모델에서 이마티닙 점안제의 특 정 염증 세포에 대한 염증 억제 효과를 확인하였다. LSP-1의 항원을 가진 염증세포 를 염색하여 관찰하였으며, 그 결과 대조군에 비하여 월등히 낮은 염증세포를 관찰 할 수 있었다 (도 6 참조) .
상기 실시예와 같이, 이마티닙을 유효성분으로 하는 조성물 또는 점안제는 각막 손상 및 변성을 억제하고, 염증을 억제하는 효과를 가지므로, 안구건조증 또 는 안구건조증과 관련된 질환을 효과적으로 예방 및 치료할 수 있다. 본 발명은 이마티닙을 필요로 하는 개체에 유효량으로 투여하여 안구건조증 또는 안구건조증 연관 안과 질환을 치료하는 방법을 제공한다.
또한, 본 발명은 이마티닙의 안구건조증 또는 안구건조증 연관 안과 질환 치 료제 제조를 위한 용도를 제공한다. 본 발명의 상기 "유효량"이란 환자에게 투여하였을 때, 암의 치료 및 예방 효과 또는 암 전이 억제효과를 나타내는 양을 말하며, 상기 "개체 "란 동물, 바람직 하게는 포유동물, 특히 인간을 포함하는 동물일 수 있으며, 동물에서 유래한 세포, 조직., 기관 등일 수도 있다. 상기 개체는 치료가 필요한 환자 (pat ient )일 수 있다. 【유리한 효과]
따라서 본 발명에서는 이마티님을 유효성분으로 하는 안구건조증 또는 안구 건조증과 관련된 안구 질환의 개선 또는 치료용 약학조성물과 점안제를 제공한다. 본 발명의 이마티닙은 효과적으로 각막상피를 보호하고 변성을 억제함으로써, 안구 건조증 또는 안구건조증과 관련된 안구 질환의 개선 또는 치료 용도로 유용하게 사 용될 수 있다.

단, 하기 실시예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실 시예에 한정되는 것은 아니다.
<실시예 1>
건조성 각결막염 모델에서 이마티닙 점안제의 각막 상피 보호 효과 건조성 각막상피손상에 대한 실험을 위하여 건성안 (dry eye) 동물모델을 제 작하였다. 동물은 흰쥐 (sd rat, 8주령, 오리엔트바이오)를 사용하였고, 건조성 각 결막염의 유도는 1% 황산 아트로핀 (1% atropine sulfate, 알콘)에 0.1% 벤잘코니 움 클로라이드 (0.1% benzalkonium chloride, 시그마알드리치)가 포함된 점안액을 하루에 두 번씩 2주간 점안하여 유도하였다.
이마티닙 점안제의 제조는 균형염 용액 (balanced salt solution)으로 제조 하였으며 pH는 6.8 내지 7.4였다.
이마티닙 점안제 (lmg/nil)의 투여는 1% 황산아트로핀 (1% atropine sulfate) 과 0.1% 벤잘코니움 클로라이드 (0.1% benzalkonium chloride) 투여 7일 후 일주일 간 하루에 두 번 점안 투여하였다ᅳ 치료효과의 비교를 위하여 비교약물로는 0.05% 사이클로스포린 점안제 (레스타사스, 알러간)를 사용하였다.
건조성 각결막염 동물모델에서 각막상피 손상의 확인은 1% 리사민그린 (Lissamin green) 용액으로 각막 표면을 염색하여 손상된 부위를 확인하였다 (도 1 참조).
건조성 각결막염 (건성안)이 유도된 각막에서 건조로 인한 각막 상피세포의 상태를 리사민그린 염색을 시행하여 건조로 인한 각막상피세포의 손상이 있음을 확 인하였다 (도 1 참조). 리사민그린은 정상의 각막 또는 결막 상피세포를 가진 각막 에서는 염색되지 않고, 손상되었거나 각막상피가 없는 각막 부위에 염색이 되는 특 징을 가지며, 파란색으로 염색된다. 염색 후 염색된 면적은 전체 각막 면적에 대 해 백분율 (%)로 나타내었다. 대조군 (Vehicle, 약물 비처리군)에서는 11 士 1.26%로 각막표면에 염색이 되었으나, 사이클로스포린 A (0.05%)에서는 2.3 士 0.43%, 이마
티닙 (0.1%)에서는 2.6 士 0.39%로 각막표면의 리사민그린 염색이 현저하게 줄어든 것을 확인할 수 있었다 (도 2 참조). 또한 위와 같은 동물모델에서 이마티닙을 농 도별로 투여하였을 때, 농도에 의존적으로 효과가 보임을 확인하였다 (도 3 참조). 농도별 약효시험에서 대조군 (Vehicle, 약물 비처리군)에서는 12.8 土 1.98%로 각막 표면에 염색이 되었으나, 이마티닙을 투여한 군의 0.01% (O.lmg/ml)에서는 8.2 士 0.97%, 0.05% (0.5mg/ml)에서는 6.18 士 0.9%, 0.1% (lmg/ral)에서는 3.7 士 0.87, 0.5% (5mg/ml)에서는 3.5 士 1.1%, 1.0% (10mg/ml)에서는 3.1 士 1.2%로 각막표면 의 리사민그린 염색이 현저하게 줄어든 것을 확인할 수 있었다.
<실시예 2>
건조성 각 결막염에서 이마티닙 점안제의 각막 변성 보호 효과 건조성 각결막염 (Keratoconjunctivitis sicca)에 대한 실험을 위하여 실시 예 1에서와 '동일한 건성안 (dry eye) 동물모델을 제작하여 실험하였다. 동물은 흰 쥐 (sd rat)를 사용하였고, 건조성 각결막염의 유도는 1% 황산 아트로핀 (1% stropine sulfate)과 0.1% 벤잘코니움 클로라이드 (0.1% benzalkonium chloride)를 하루에 두 번씩 2주간 점안하여 유도하였다.
이마티닙 점안제 (0.1mg/ml)의 투여는 1% 황산 아트로핀 (1% atropine sulfate)과 0.1% 벤잘코니움 클로라이드 (0.1% benzalkonium chloride) 투여 후 일 주일부터 일주일간 하루에 두 번 점안 투여하였다. 비교약물로 0.05% 사이클로스포 린 A 점안제를 사용하였다.
건조성 각결막염 (건성안)에서 각막의 변성을 관찰하기 위하여 F-actin에 대 한 염색을 실시하였다. F-actin은 형광물질이 붙은 Phalloidin으로 염색하였다. 약 물을 투여하지 않은 각막 (vehicle)에서는 각막상피세포가 각질화되는 것이 관찰되 었고 (흰색 화살표머리), 각막 기질에서는 튜브형태로 염색된 세포 (흰색 화살표)가 관찰되었다. 그러나 사이클로스포린 A와 이마티닙 점안제를 투여한 각막에서는 Vehicle에서 관찰된 각막 변형이 관찰되지 않았고, 정상과 비교하여 차이가 없었다 (도 4 참조).
<실시예 3>
이마티닙 점안제의 건조성 각 결막염에서 특정 염증 세포의 침투억제 효과
실시예 1의 동물 모델과 같은 투여 방법으로 실험한 후, 각막에 침투한 특정 염증 세포에 대한 이마티닙의 염증 억제 효과를 확인하였다.
건조성 각결막염 (건성안)에서 각막내의 염증세포를 확인하기 위하여 안구를 넁동절삭하고, LSP-1의 항원을 가진 염증세포를 염색하여 관찰하였다 (도 5 참조) . LSP-1은 lymphocytes , neutrophi l s , macrophage의 염증에 관련된 세포가 가지고 있 으며, LSP-1 항체를 이용하여 면역 염색한 후 관찰이 가능하다. 염증 세포는 대조 군 (약물 무처리군)에서는 0.61 士 0.05, 이마티닙 점안제 (0. 1%)에서는 0.52 土 0.05로 관찰되었다 (도 6 참조) . 위의 염증세포 염색 실험 결과로 이마티닙 점안제 (0. 1%)가 건조성 각결막염에 대해 염증억제 효과가 있음을 확인하였다.
위의 결과로 비추어 볼 때, 본 발명의 이마티닙 점안제는 각결막염 및 안구 건조로 인한 건조성 각결막염의 염증 치료에 효과적으로 사용될 수 있다.
【산업상 이용가능성】
따라서 본 발명에서는 이마티닙을 유효성분으로 하는 안구건조증 또는 안구 건조증과 관련된 안구 질환의 개선 또는 치료용 약학조성물과 점안제를 제공한다. 본 발명의 이마티닙은 효과적으로 각막상피를 보호하고 변성을 억제함으로써, 안구 건조증 또는 안구건조증과 관련된 안구 질환의 개선 또는 치료 용도로 유용하게 사 용될 수 있어 산업상 이용가능성이 높다.

이마티닙 ( Imat inib)을 유효성분으로 포함하는 안구건조증 또는 안구건조증 연관 안과 질환 예방 및 치료용 약학 조성물.

게 1항에 있어서, 상기 안구건조증은 수성 눈물-결핍성 안구건조증 또는 증발 성 안구건조증인 것을 특징으로 하는 조성물.
【청구항 3]
제 1항에 있어서, 상기 안구건조증 연관 안과 질환은 건성 각결막염, 건성으 로 인한 각막 궤양, 안검염 (눈꺼풀염증), 눈의 층혈 및 각막신생혈관 생성으로 이 루어진 군에서 선택된 어느하나인 것임을 특징으로 하는 조성물.

제 1항에 있어서, 상가조성물은 각막의 변성을 억제하고, 염증 세포의 수를 감소시키는 것을 특징으로 하는 조성물.

제 1항에 있어서 , 상기 이마티닙 ( Imat inib)의 함량은 조성물 전체 중량을 기 준으로 0.005 내지 5 중량 %인 것을 특징으로 하는 조성물.

제 1항에 있어서, 상기 조성물은 국소 투여 또는 점안 투여되는 것을 특징으 로 하는 조성물.

거 18항에 있어서, 상기 수용액을 포함하는 담체는 증류수, 인산완층액, 균형 염용액 및 생리식염수로 이루어진 군에서 선택된 하나 이상의 약학적으로 허용되는 담체인 것을 특징으로 하는 점안제.
【청구항 10】
이마티닙을 필요로 하는 개체에 유효량으로 투여하여 안구건조증 또는 안구 건조증 연관 안과 질환을 치료하는 방법 .

이마티닙의 안구건조증 또는 안구건조증 연관 안과 질환 치료제 제조를 위한 용도.
