본 발명은 강황 추출물; 및 마그네슘, 아미노산 또는 이들의 조합;을 유효성분으로 함유하는 관절 기능 향상용 조성물을 제공한다.
Description
본 발명은 관절 기능 향상용 조성물에 관한 것이다.
본 출원은 2015년 5월 14일에 한국 특허청에 제출된 한국 특허출원 제10-2015-0067697호와, 2016년 5월 3일에 한국 특허청에 제출된 한국 특허출원 제10-2016-0054871호 출원일의 이익을 주장하며, 그 내용 전부는 본 명세서에 포함된다.
관절은 2개 이상의 뼈가 서로 맞닿아 연결되어 있는 부분으로, 무릎, 팔꿈치, 허리, 어깨 등 인체의 도처에 존재한다. 관절은 두 뼈 사이에 윤활유 역할을 하는 활액이 존재하며, 관절은 뼈 사이에 근육과 인대로 연결되며, 이러한 근육과 인대의 견인력에 의해 구부러질 수 있다. 관절은 그 움직이는 방향 및 크기의 범위가 일정 범위로 정해져 있으므로, 이러한 범위 이상의 굽힘 또는 회전이 발생하거나, 일정 수준 이상의 굽힘 또는 회전이 반복적으로 발생하는 경우 관절에 장애가 발생한다.
관절염, 관절기능에 효과가 있다고 알려진 물질의 경우, 관절의 염증개선 및 이를 통한 통증완화 효과만 제공하고 있는 실정이나, 관절의 근육의 강화를 통하여 관절 기능을 향상시키는 물질은 알려진 바가 없었다.
본 발명은 관절의 염증 개선 및 통증 억제뿐만 아니라, 관절 근육의 기능 향상시키는 조성물을 제공하고자 한다.
본 발명은 강황 추출물; 및 마그네슘, 아미노산 또는 이들의 조합;을 유효성분으로 함유하는 관절 기능 향상용 조성물을 제공한다.
본 발명은 관절의 염증을 개선 및 관절의 통증을 완화시킬 뿐만 아니라 관절을 둘러싸고 있는 관절 근육의 강화시킴으로써, 관절 기능을 향상시키는 조성물을 제공하며, 구체적으로, 관절의 움직임 시 관절이 받는 무게부하를 감소시켜 주어 연골의 물리적 손상을 방지하고, 관절의 과도한 회전을 방지하여, 관절의 안정성 유지시킬 수 있다.
이하에서는, 본 발명에 따른 관절 기능 향상용 조성물을 구체적으로 설명한다.
본 명세서에 있어서, 강황 (Curcuma
longa
L. rhizome)은 생강과의 다년초 식물을 의미한다.
본 발명에 따른 조성물은 강황 추출물을 유효성분으로 함유함으로써, 관절의 염증을 개선 및 관절의 통증을 완화시킬 수 있다.
본 발명의 일 실시상태에 있어서, 상기 강황 추출물의 함량은 조성물 전체 중량에 대하여, 10 중량% 내지 80 중량%이다.
본 발명의 일 실시상태에 있어서, 상기 강황 추출물과 함께 마그네슘을 유효성분으로 함유하며, 이로 인하여 관절의 염증을 개선 및 관절의 통증을 완화시킬 수 있을 뿐 아니라, 관절 근육을 강화시킬 수 있다.
본 발명의 일 실시상태에 있어서, 상기 마그네슘의 함량은 조성물 전체 중량에 대하여, 2 중량% 내지 20 중량%이다.
본 발명의 일 실시상태에 있어서, 상기 강황 추출물과 함께 아미노산을 유효성분으로 함유하며, 이로 인하여 관절의 염증을 개선 및 관절의 통증을 완화시킬 수 있을 뿐 아니라, 관절 근육을 강화시킬 수 있다.
본 발명의 일 실시상태에 있어서, 상기 아미노산의 함량은 조성물 전체 중량에 대하여, 0.1 중량% 내지 10 중량%이다.
본 발명의 일 실시상태에 있어서, 상기 아미노산은 발린, 루신, 이소루신, 글루타민, 아르기닌, 메티오닌, 트레오닌, 라이신, 페닐알라닌 및 트립토판으로 이루어진 군에서 선택되는 어느 하나 이상이다.
본 발명의 일 실시상태에 있어서, 상기 아미노산은 발린, 루신, 이소루신, 글루타민 및 아르기닌으로 이루어진 군에서 선택되는 어느 하나 이상이다.
본 발명의 일 실시상태에 있어서, 상기 아미노산은 메티오닌, 트레오닌, 라이신, 페닐알라닌 및 트립토판으로 이루어진 군에서 선택되는 어느 하나 이상이다.

본 발명의 일 실시상태에 있어서, 상기 조성물은 관절 통증 억제용 또는 관절 근육 강화용이다. 상기 조성물은 관절의 염증을 개선 및 관절의 통증을 완화시킬 수 있을 뿐 아니라, 관절 근육을 강화시킬 수 있다.
본 발명의 일 실시상태에 있어서, 상기 조성물은 무릎 관절 기능 강화용이다. 본 발명에 따른 조성물은 관절 근육을 강화시킴으로써, 인체 중 가장 많은 하중을 부담하는 관절 중 하나인 무릎 관절 기능 강화에 특히 효과적이다.
본 발명에 따른 조성물은 피부 외용제일 수 있으나, 바람직하게는, 경구용 조성물이다.
본 발명의 일 실시상태에 있어서, 상기 조성물은 부형제 및 코팅제를 더 포함한다. 상기 부형제 및 코팅제는 결정셀룰로오스, 제이인산칼슘, 카르복시메틸셀룰로오스, 이산화규소, 스테아린산마그네슘 및 하이프로멜로스로 이루어진 군에서 선택된 어느 하나 이상일 수 있다.
본 발명의 일 실시상태에 있어서, 상기 조성물은 조성물의 제조에 통상적으로 사용하는 적절한 담체, 부형제 및 희석제를 더 포함할 수도 있다.
본 발명의 일 실시상태에 있어서, 상기 조성물은 섭취에 편리한 형태로 다양한 경구 제형을 가질 수 있으며, 예를 들어 정제, 캡슐제, 연질 캡슐제, 환제, 과립제, 드링크제 등의 제형을 가질 수 있다.
본 발명의 일 실시상태에 있어서, 상기 조성물은 건강식품 조성물이다.
본 발명의 일 실시상태에 있어서, 상기 조성물의 바람직한 섭취량은 신체의 건강 상태, 체중, 질병의 정도 등에 따라 다양하게 다르지만, 통상의 기술자에 의해 적절하게 선택될 수 있다.
이하에서는, 본 발명에 따른 관절 기능 향상 방법, 관절 근육 강화 방법, 관절 통증 억제 방법 및 무릎 관절 기능 강화 방법을 구체적으로 설명한다.
본 발명은 상기 관절 기능 향상용 조성물을 경구 투여하는 단계를 포함하는 관절 기능 향상 방법을 제공한다.
본 발명은 상기 관절 기능 향상용 조성물을 경구 투여하는 단계를 포함하는 관절 근육 강화 방법을 제공한다.
본 발명은 상기 관절 기능 향상용 조성물을 경구 투여하는 단계를 포함하는 관절 통증 억제 방법을 제공한다.

이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시 예는 오로지 본 발명을 예시하기 위한 것으로서, 본 발명의 범위가 이들 실시 예에 의해 제한되는 것으로 해석되지 않는 것은 해당 기술분야에서 통상의 지식을 가진 자에 있어서 자명할 것이다.
<제조예> 강황 추출물의 제조
Natural Remides PVT.LTD. (소재지: 인도, Plot No.5B Veerasandra Indl. Area, 19th K.M. Stone, Hosur Road, Electronic City Post, Bangalore-560 100)에서 입수한 강황을 증류수와 혼합하여 4회 여과하였으며, 여과된 추출물을 농축 및 분무 건조 (spray dry)하여, 강황 열수 추출물을 수득하였다. 수득된 강황 열수 추출물과 강황 유래 오일 (Curcuma oil)을 중량비 (강황 열수 추출물: 강황 유래 오일)로 99:1로 혼합하여 강황 추출물을 제조하였다.
<시험예 1> 정제 복용 시험
하기 표 1의 조성으로 비교예 1, 비교예 2 및 실시예 1의 조성물을 각각 제조하여, 이를 각각 700 mg의 정제 형태로 제조하였다.

무릎 통증이 있거나, 무릎 기능에 이상이 있는 성인 남녀 120명을 6개의 군으로 나누어, 비교예 1의 정제를 20명의 성인남녀 비교예 1군에게, 비교예 2의 정제를 20명의 성인남녀 비교예 2군에게, 비교예 3의 정제를 20명의 성인남녀 비교예 3군에게, 비교예 4의 정제를 20명의 성인남녀 비교예 4군에게, 실시예 1의 정제를 성인남녀 실시예 1군에게, 실시예 2의 정제를 성인남녀 실시예 2군에게 제공하였으며, 비교예 1군 내지 4군, 실시예 1군 및 2군의 실험 대상자들은 제공받은 정제를 하루에 세 번, 총 2,100 mg를 복용시키는 시험을 28일 동안 진행하였다.
시험예 1의 정제 복용 시험에서, 실험 대상자들에게 VAS (visual analogue scale) 설문으로 일상생활에서 느끼는 무릎의 통증 정도를 상기 시험예 1의 개시 직전 및 상기 시험예 1 진행 28일 후에 각각 기록하게 하여, 이를 점수화하였다.
실험 대상자에게 사용한 VAS 설문은 10점 척도를 사용하여, 좌측의 시작점 (통증이 없는 상태)부터 오른쪽 끝 (극심한 통증 상태)으로 나타내었으며, 이는 하기와 같다.

VAS 설문의 측정 결과에 대하여, 비교예 1군 내지 4군, 실시예 1군 및 2군의 각 평균 점수를 하기 표 2에 나타내었다.

상기 표 1과 같이, 비교예 1군 내지 4군, 및 실시예 1군 및 2군의 시험 대상자들은 시험예 1의 개시 전 모두 극심한 통증 상태에 가까운 점수를 나타내었으나, 시험예 1의 진행 28일 후, 비교예 1군 내지 4군은 무릎 통증의 경미한 개선을 나타낸 반면, 실시예 1군 및 2군의 시험 대상자들은 통증이 없는 상태에 가까운 점수를 나타내어, 유의미한 무릎 통증 개선 효과를 확인할 수 있었다.
<시험예 3> 골관절염 지수의 측정
시험예 1의 정제 복용 시험에서, 실험 대상자들에게 골관절염 측정 지수 설문 (WOMAC, Western Ontario and McMaster Universities Osteoarthritis) 무릎 관절의 통증, 뻣뻣함, 무릎 관절의 기능을 조사하였으며, 상기 시험예 1의 개시 직전 및 상기 시험예 1 진행 28일 후에 각각 기록하게 하여, 이를 점수화하였다.
실험 대상자에게 사용한 골관절염 측정 지수 설문은 4점 척도를 사용하였으며, 그 구체적인 설문 내용은 하기와 같다.
골관절염 측정 지수 설문의 측정 결과에 대하여, 각 항목에 해당하는 점수를 합산하였으며 (0~96점), 이러한 합산 점수에 대한 비교예 1군 내지 4군, 실시예 1군 및 2군의 각 평균 점수를 하기 표 3에 나타내었다.

<시험예 4> 하지의 등속성 근력의 측정
시험예 1의 정제 복용 시험에서, 실험 대상자들에게 하지 근력 측정을 상기 시험예 1의 개시 직전 및 상기 시험예 1 진행 28일 후에 각각 실시하였다.
실험 대상자에게 실시한 하지 근력 측정은 실험 대상자의 양 다리에 측정 기구를 이용하여 각속도(speed selector) 를 60°/sec 에 두고, 시험 대상자에게 기구 연습을 3회 실시하였으며, 이후 측정 운동을 5회 실시하였다, 5회 실시된 측정 중 신전 (extension) 시 및 굴곡 (flexion) 시 측정된 최고 힘 값을 각각 기록하였으며, 신전 (extension) 시 및 굴곡 (flexion) 시 측정된 5회의 총 운동량을 각각 기록하였다.
하지 근력 측정에서 기록된 최고 힘값 및 총운동량에 대하여, 비교예 1군 내지 4군, 실시예 1군 및 2군의 각 평균값을 하기 표 4에 나타내었다.

시험예 1의 정제 복용 시험에서, 실험 대상자들에게 근지구력 측정을 상기 시험예 1의 개시 직전 및 상기 시험예 1 진행 28일 후에 각각 실시하였다.
실험 대상자에게 실시한 하지 근력 측정은 실험 대상자의 양 다리에 측정 기구를 이용하여 각속도(speed selector) 를 180°/sec 에 두고, 시험 대상자에게 기구로 측정 운동을 15 회 실시하였다, 15회 실시된 측정 중 신전 (extension) 시 및 굴곡 (flexion) 시 측정된 최고 힘 값을 각각 기록하였으며, 신전 (extension) 시 및 굴곡 (flexion) 시 측정된 5회의 총 운동량을 각각 기록하였다.
하지 근력 측정에서 기록된 최고 힘값 및 총운동량에 대하여, 비교예 1군 내지 4군, 실시예 1군 및 2군의 각 평균값을 하기 표 5에 나타내었다.
