光学測定装置
 本発明は、セル中の試料液に光を照射し、それに対して該試料液から得られる透過光、散乱光、蛍光などを検出する光学測定装置に関する。
 液体クロマトグラフ(LC)の検出器として、カラムから溶出した試料液の吸光度(又は透過率)を測定する光学測定装置がしばしば用いられる。図9は、吸光測定のための光学測定装置の一例の概略構成図である(例えば特許文献1など参照)。
 光源であるLED71から出射した測定光は試料液が流通するフローセル72に照射される。測定光はフローセル72中の試料液を通過する際に、該試料液中の成分の種類や量に応じた吸収を受ける。そうした吸収を受けたあとの光が光検出器73に入射し、光検出器73はその光の光量に応じた検出信号を出力する。そして、図示しない信号処理部において、検出信号から試料液による吸光度を算出する。この構成では、フローセル72の長さLが試料液中の光路長となる。

 上述したように従来一般に、光学測定装置における試料液中の光路長の変更は、使用するフローセル自体を交換することで行われている。しかしながら、フローセルの交換は手間と時間が掛かる作業である。また、光路長を細かく複数段階に変更するためには、長さの相違するフローセルを複数用意しておく必要がある。
 これに対し、光源から出射した測定光がフローセルに入射する入射角を変更することによって、フローセル内での光の反射回数を調整し、それによって実質的な光路長を変更するようにした光学測定装置が提案されている(特許文献2など参照)。しかしながら、測定光の入射角を調整する機構は大掛かりになるため、それだけ光学測定装置が大きくなってしまう。また、光源又は入射光学系の位置や姿勢を変更することで測定光の入射角を精度良く変更するためには、高精度な機構部品が必要になりコストも高くなる。

 本発明は上記課題に鑑みて成されたものであり、その目的とするところは、試料液を収容するセルを交換することなく且つセルへの測定光の入射角を変更することなく、セル内の光路長を変更することによって、低濃度試料から高濃度試料まで幅広い濃度の試料に対する適切な測定を実施することができる光学測定装置を提供することにある。
 上記課題を解決するためになされた本発明に係る光学測定装置は、
 a)試料液が流通する又は試料液が収容される筒状の容器であって光学的に透明な試料セルと、
 b)前記試料セルに対し、その軸方向に又は該試料セルの軸に斜交する方向に測定光を照射する測定光照射部と、
 c)前記測定光照射部から前記試料セルに照射された測定光が該試料セル内を通過したあとの光又は該測定光に応じて試料液から放出された光を検出する検出部であって、光電変換を行う受光部と、その一部が前記試料セルの外面に接した状態で該試料セルと前記受光部との間に配置され、光を透過可能であってその屈折率が前記試料セルの壁面の材料の屈折率よりも大きい材料からなる接合部と、を含む検出部と、
 を備え、前記試料セルへの前記検出部の接触位置が変更可能であることを特徴としている。
 本発明に係る光学測定装置において、試料セルは例えばその全体が光学的に透明であって、且つ試料液の溶媒として一般に使用される有機溶媒及び試料セルの周囲の環境(例えば空気)よりも高い屈折率を有する材料から形成される。そうした材料としては例えば、合成石英、サファイア(酸化アルミニウム単結晶:Al2O3)、ダイヤモンドなどを利用することができる。
 測定光照射部は試料セルに対し、その軸に平行か又は該軸とのなす角度θが0<θ<90°であるように斜めに測定光を照射する。試料セルに入射した測定光は該セル内に充ちている試料液中を通過し、試料セル壁面の外面(試料セル壁面と周囲の環境との界面)で反射(好ましくは全反射)を繰り返しながら試料セル中を伝搬してゆく。つまり、試料液が充ちた試料セルは一種の光導波路として機能する。一般に、光導波路ではその外面に埃などが付着すると、そこから光が漏出して光の伝搬効率が低下することが知られており、これを避けるために光導波路の外面に埃などが付着しないような対策が採られる。
 本発明に係る光学測定装置では、この光の漏出現象を積極的に利用し、試料セルの任意の位置から測定光を外部に取り出して検出する。即ち、試料セルの外面に検出部の接合部が接触していると、この接合部の屈折率は試料セル壁面の屈折率よりも大きいため、接触部位に達した光は試料セル壁面の外面で反射せずに接合部に侵入する。そして、接合部中を透過して受光部に到達する。受光部は到達した光の光量に応じた検出信号を出力する。
 例えば試料セルへの検出部の取付位置を測定光照射部から遠ざけるほど、試料セル壁面の外面における全反射の回数が相対的に多い測定光つまりは光路長が平均的に長い測定、又はそうした平均的に長い光路長を通った測定光に応じて試料液から放出される蛍光や散乱光が検出部に導入される。即ち、試料セルへの検出部の取付位置を試料セルの軸方向に変更することによって、試料液中の平均的な光路長を変更することができる。
 例えば、吸光度測定においては、試料液の濃度が高い場合には単位光路長当たりの吸光度が大きいから、試料セルへの検出部の取付位置を測定光照射部の近くにすることで平均的な光路長を短くし、検出信号が小さくなりすぎることを防止できる。逆に、試料液の濃度が低い場合には単位光路長当たりの吸光度が小さいから、試料セルへの検出部の取付位置を測定光照射部から遠ざけることで平均的な光路長を長くし、吸光による検出信号の減少度合いを大きくして感度を高めることができる。
 また例えば、蛍光測定やラマン散乱光測定においては、試料液の濃度が高い(分子数が多い)場合には単位光路長当たりの蛍光の放出や散乱が大きいから、試料セルへの検出部の取付位置を測定光照射部の近くにすることで平均的な光路長を短くし、検出信号が大きくなりすぎることを防止できる。逆に、試料液の濃度が低い(分子数が少ない)場合には単位光路長当たりの蛍光の放出や散乱が小さいから、試料セルへの検出部の取付位置を測定光照射部から遠ざけることで平均的な光路長を長くし、検出信号を大きくして感度を高めることができる。
 本発明に係る光学測定装置において、検出部は試料セルの外面の周方向の一部にのみ設けられてもよいが、その周方向全周に亘り設けられる構成としてもよい。
 例えば試料セルが円筒形状である場合、検出部はその中空部分に試料セルが挿通される円環形状体とすればよい。
 この構成によれば、測定光照射部から或る程度の角度を以て広がりながら試料セルに照射された測定光が試料セル内で様々な方向に全反射しつつ進行する光や、そうした光によって試料液から様々な方向に放出される蛍光や散乱光を、検出部で効率良く捉えることができる。
 また、本発明に係る光学測定装置において、接合部は、試料セルに接触する面から受光部に面する面に向かって屈折率が増加するものであるとするとよい。これにより、試料セルと接合部との界面、及び、接合部と受光部との界面のいずれにおいても、界面を挟んだ両側の屈折率の差を小さくすることができ、光の通過効率を高めることができる。
 なお、測定光照射部から照射される測定光は所定波長の単色光でも波長範囲の広い光でもよいが、後者が用いられる場合には、検出部において接合部と受光部との間に分光器を設け、該分光器で波長分散した光を受光部で波長毎に検出するか、又は該分光器で取り出した特定波長の光を受光部で検出するとよい。
 本発明に係る光学測定装置によれば、測定対象である試料液の濃度や分析目的などに応じて、試料セルへの検出部の取付位置を変更するだけで、試料液中の光路長を変更することが可能となる。検出部の取付位置の変更は測定者が容易に行うことができ、従来のフローセル交換のような面倒な作業が不要になるので、測定者の手間を軽減するとともに測定の効率化を図ることができる。また、様々な光路長の試料セルを用意しておく必要もなくなる。また、試料セルへの検出部の取付位置を変更するために、測定光の入射角を調整する機構のような大掛かりな機構は不要であるので、装置の小形化にも適している。さらにまた、検出部の取付位置を変更しても、測定光を試料セルに入射するための光路や試料セル内の光路自体は何ら変化しないので、面倒な光学系の再調整が必要になることもない。

 以下、本発明に係る光学測定装置の実施例について、添付図面を参照して説明する。
図1は第1実施例の吸光測定装置の概略構成図であり、(a)及び(b)はフローセルの中心軸を含む平面での断面図、(c)は該中心軸に直交する平面での断面図である。図2は本実施例の吸光測定装置におけるフローセル内の光路の説明図である。
 本実施例の吸光測定装置は、円筒直管状の流路を有するフローセル2と、該フローセルに対し測定光を照射する光照射部1と、フローセル2の外面に接触するように設けられる検出部3と、を備える。ここでは、フローセル2は空気中に置かれているものとする。
 光照射部1は例えば単一のLEDを光源とするものであり、図示しないLED駆動部から供給される駆動電流により駆動され、単色光とみなし得る狭い波長範囲の光を発する。光照射部1から発せられる光は所定の立体角を以て広がるが、その光軸はフローセル2の中心軸に対し0<θ<90°の範囲の所定の角度θとなるように、光照射部1とフローセル2との位置関係が定められている。
 フローセル2は、空気(周囲環境)の屈折率n1及び流路を流れる試料液の溶媒の屈折率n3よりも大きい屈折率n2を有する透明な材料から成る。本装置がLC用の検出器である場合には、試料液の溶媒としては、LCの移動相として一般に使用される様々な有機溶媒を想定すればよい。サファイア、合成石英、ダイアモンドなどが、フローセル2に好適な材料である。
 検出部3は、典型的にはフォトダイオードなどの半導体受光素子である受光部31と、フローセル2の外面に一面が接触され対向する面が受光部31の受光面に接触される接合部32と、を含み、フローセル2の外面の任意の位置に取り付け可能である。
 接合部32は、光学的に透明又は半透明であり、屈折率n4がフローセル2の材料の屈折率n2以上で且つ受光部31の受光面の材料の屈折率n5以下(ただしn2≦n5)である材料から成る。したがって、n2<n4<n5でもよいし、n2<n4=n5でもよいし、n2=n4<n5でもよいし、さらにn2=n4=n5でもよい。接合部32は、特にフローセル2の外面との密着性が高いことが望ましい。好ましい材料として例えば、樹脂製光ファイバ等に利用されている屈折率分散型ポリマー、LED用低硬度エラストマー・ゲル封止材(非特許文献1など参照)などのゲル状部材、シリコーンなどが挙げられる。また、プリズムを用いた光学測定などの際に使用されるマッチングオイルを利用することもできるし、ゼラチンなどを用いてもよい。
 いま、フローセル2の外面の図1(a)に示すような位置に検出部3が取り付けられている場合における測定動作を説明する。
 光照射部1から発せられた光(測定光)はフローセル2に斜めに入射し、フローセル2内の流路を流れる試料液中を通過する。フローセル2の管路の材料の屈折率n2は空気の屈折率n1よりも大きいため、フローセル2の管路と空気との界面(フローセル2の外面)で測定光は全反射し、再び試料液中を通過する。図1(a)の構成では、この1回だけ全反射した測定光が試料液中を通過し、先の界面とはちょうど反対側のフローセル2の管路と空気との界面に達する位置付近に検出部3が取り付けられている。上述したように、フローセル2の外面に接している接合部32の屈折率n4はフローセル2の管路の材料の屈折率n2以上であるため、測定光はこの界面では全反射することなく該界面を通り抜け、接合部32中を通過して受光部31に到達する。つまり、接合部32はフローセル2内から光を効率良く漏出させる機能を有する。
 測定光は主として試料液中を通過する際に、該試料液中に存在する成分によって吸収を受ける。図1(a)の構成では、試料液中を斜め方向に通過する光路長Pの約2倍の長さの経路(2×P)を経て測定光は受光部31に達する。したがって、受光部31は、この長さ2×Pの経路で吸収を受けた測定光の光量に応じた信号を出力する。
 測定者が検出部3の取付位置を図1(b)に示すように変更した場合の測定動作は次のようになる。なお、図1(b)中に点線で示したのは、図1(a)中における検出部3の位置である。
 光照射部1から発せられた測定光がフローセル2に斜めに入射し、試料液中を通過して、フローセル2の管路と空気との界面で全反射する点は図1(a)の場合と同じである。ここでは、検出部3が光照射部1から遠い位置に設けられているため、試料液中を再び通過した測定光はフローセル2の管路と空気との界面で再度全反射される。こうした全反射を複数回繰り返し、検出部3が取り付けられている位置に測定光が達したときに、測定光は該界面で全反射せずにそこを通り抜け、接合部32中を通過して受光部31に到達する。図1(b)の構成では、試料液中を斜め方向に通過する光路長Pの約6倍の長さの経路(6×P)を経て測定光は受光部31に達する。したがって、受光部31は、この長さ6×Pの経路で吸収を受けた測定光の光量に応じた信号を出力する。
 図1の例では、光照射部1から発せられる光の光軸に沿った測定光のみを考えたが、上述したように、実際には、光照射部1から発せられる光は所定の立体角の範囲に広がる。そのため、図2に示すように、フローセル2の外面の或る位置に取り付けられた検出部3には、或る1種類の回数だけ全反射した光ではなく、様々な回数、全反射した測定光が到達し得る。しかしながら、どのような経路を経つつ全反射する光でも、フローセル2の中心軸方向に所定の範囲で検出部3を光照射部1に近付けるほど、検出部3に到達するまでの光路長は全体的に短くなる。即ち、フローセル2の中心軸方向に検出部3の取付位置を変更する(移動させる)ことによって、全反射を繰り返して検出部3に到達する測定光の平均的な光路長を調整することができる。したがって、例えば試料液の濃度が低く単位光路長当たりの吸光度が小さい場合には、測定者は、検出部3を光照射部1から離した位置に取り付けるようにして平均的な光路長を長くすることで、検出感度を高くすることができる。
 上述したように、検出部3における接合部32の材料は、その屈折率n4がフローセル2の材料の屈折率n2以上で受光部31の受光面の材料の屈折率n5と同じか或いはそれよりも小さければよい。その屈折率n4は一定(図3(b)参照)であってもよいが、フローセル2と接合部32との界面での光の損失をできるだけ抑え、より効率良くフローセル2から光を取り出すには、図3(c)に示すように、フローセル2との接触面32aから受光部31との接触面31aに向かうに従って屈折率n4が徐々に大きくなる、例えばn4がn2からn5まで緩やかに増加するようにするとよい。例えば接合部32を多層膜構造とすることで、こうした屈折率の勾配を容易に実現することができる。また、反射防止膜などに利用されるモスアイ構造を採用することで屈折率を徐々に変化させることもできる。なお、図3(c)では、屈折率の変化は直線的であるが、この変化は曲線的でも或いは階段状でもよい。
 上記実施例の吸光測定装置では、光照射部1からの光をフローセル2に対し斜めに入射するようにしていたが、こうした構成の利点の一つは、試料液をフローセル2に導入したり試料液をフローセル2から取り出したりするための流路を、フローセル2に対し直線的に接続することができる点である。こうした直線的な流路の接続によって、その接続部位における光の拡散を軽減することができ、光の利用効率が向上する。
 もちろん、図4に一例を示すように、光照射部1からの光をフローセル2の中心軸に平行な方向にフローセル2に入射するようにしてもよい。即ち、フローセル2の管路の外面で測定光が全反射するように、測定光をフローセル2に入射可能でありさえすれば、測定光の入射方向は特に問わない。
 また、図1、図4の例では、フローセル2は直管状であるが、フローセル2は直管状である必要はなく、湾曲形状や屈曲形状などでもよい。
 また、上記実施例では、光源として発光の波長範囲が狭いLEDを用いていたが、図5に一例を示すように、例えば重水素ランプやそれ以外の発光波長の広い光源を使用した光照射部1を用い、そうした光を分光器4に導入して取り出した特定波長の単色光をフローセル2に照射するようにしてもよい。
 また、図6に一例を示すように、発光波長の広い光源を使用した光照射部1から出射した光をそのまま(つまり単色光化せずに)フローセル2に照射し、試料液中で吸収を受けた測定光をフローセル2から取り出したあとに分光器4に導入し、特定波長の光のみを取り出して受光部31に導入するようにしてもよい。また、この場合、受光部31としてフォトダイオードアレイを用い、分光器4で波長分散した光をフォトダイオードアレイの各受光素子で一斉に検出してもよい。これによって、特定の波長における吸光度だけでなく、吸光スペクトルを測定することができる。さらには、図7に一例を示すように、光照射側と光検出側との両方に分光器4A、4Bを設けてもよい。
 また、上記実施例では、検出部3はフローセル2の外面の周方向の一箇所にのみ設けられていたが、図8に一例を示すように、フローセル2の外面の周方向の全体に、つまりフローセル2を取り囲むような円環形状の検出部3を設ける構成としてもよい。この構成により、効率よくフローセル2から光を取り出して検出することができる。
 本発明に係る光学測定装置は吸光測定のみならず、蛍光測定やラマン散乱光測定にも有効である。何故なら、蛍光測定やラマン散乱光測定においても、検出信号は光路長依存性を有するからである。ただし、蛍光測定やラマン散乱光測定では、測定対象の光の波長は試料液に照射される光(上記各例における測定光)とは異なる波長であり、照射される光は不要光又は一種のノイズ光である。したがって、蛍光測定やラマン散乱光測定では、図6又は図7に示したような、検出側に分光器4、4Bを設けた構成として、測定対象である波長の光のみを取り出したり、所定波長範囲のスペクトルを測定したりするとよい。また、蛍光やラマン散乱光はあらゆる方向に放出されるから、特に図8に示すような構成を採ると、測定対象の光を効率良く集めることができる。
 また、上記実施例では、フローセル2に検出部3を一つのみ設けたが、複数の検出部3を一つのフローセル2に設け、その複数の検出部3で同時に検出を行うようにしてもよい。これにより、複数の異なる光路長に対応する感度の相違する測定を行うことができる。
 また、上記実施例はいずれも本発明の一例にすぎず、本発明の趣旨に沿った範囲でさらに適宜変形や修正、追加を行っても本願特許請求の範囲に包含されることは明らかである。例えば、上記実施例では試料セルはフローセルであるが、試料液が収容された、つまり試料液が流通せずに貯留される試料セルでも本発明を適用可能であることは明らかである。

Claims (3)

  1.  a)試料液が流通する又は試料液が収容される筒状の容器であって光学的に透明な試料セルと、
     b)前記試料セルに対し、その軸方向に又は該試料セルの軸に斜交する方向に測定光を照射する測定光照射部と、
     c)前記測定光照射部から前記試料セルに照射された測定光が該試料セル内を通過したあとの光又は該測定光に応じて試料液から放出された光を検出する検出部であって、光電変換を行う受光部と、その一部が前記試料セルの外面に接した状態で該試料セルと前記受光部との間に配置され、光を透過可能であってその屈折率が前記試料セルの壁面の材料の屈折率よりも大きい材料からなる接合部と、を含む検出部と、
     を備え、前記試料セルへの前記検出部の接触位置が変更可能であることを特徴とする光学測定装置。
  2.  請求項1に記載の光学測定装置であって、
     前記検出部は前記試料セルの外面の周方向全周に亘り設けられることを特徴とする光学測定装置。
  3.  請求項1又は2に記載の光学測定装置であって、
     前記接合部は、前記試料セルに接触する面から前記受光部に面する面に向かって屈折率が増加するものであることを特徴とする光学測定装置。
