Definitions
- the present invention relates to a method for producing cells for treating cancer, and to an adjuvant for cancer, a kit for treating cancer, and a method for treating cancer comprising the cells prepared by the method.
- Brain tumors are very damaging to the brain and have very low survival rates. Surgical extraction is most effective as a conventional treatment for brain tumors, but surgery is often impossible depending on the type of brain tumor and the site of its occurrence.
- BBB brain-blood barrier
- chemotherapy using anticancer drugs requires a higher concentration of anticancer drugs than other cancers to treat brain tumors.
- gene therapy is a method of directly introducing a gene that inhibits the proliferation of cancer cells into cancer cells, mainly using a virus for the introduction of the gene, the virus itself does not have the ability to move to the cancer must be injected surgically. However, it is practically impossible to inject the virus individually into every minute tumor or cancer, which has limitations in targeting cancer cells.

- the vector after inserting a suicide gene into a retroviral vector to produce an expression vector, the vector is transduced into packaging cells, the transduced packaging cells are cultured, and filtered.
- Suicide genes can be introduced into mesenchymal stem cells by obtaining a retrovirus solution and using them to infect mesenchymal stem cells. Subsequently, mesenchymal stem cells continuously expressing suicide genes may be obtained using a selection marker included in the retroviral vector.
- HLA human leukocyte antigen
- the MSC / CD of the present invention may be obtained by first introducing and selecting a suicide gene into isolated stem cells and then proliferating and differentiating them under appropriate conditions in vitro, or by sufficiently proliferating the stem cells and then introducing a suicide gene thereto. This can be obtained by differentiating mesenchymal stem cells.
- MSC / CD is injected into a subject in need of treatment by the presence of a suicide gene, but is not limited thereto, for example, a period of 2-30 days, preferably 5-15 days, more preferably 5-10 days of infusion. It can kill itself afterwards to reduce side effects that can be induced by persistent action.
- F cells Frozen cells, F cells
- F cells refers to cells that have not undergone an additional culture step after freezing and thawing MSC / CD, which are mesenchymal stem cells into which cytosine dianamin (CD) is introduced, Define cells that are intended to be suspended in thawed state, or suspended and washed to be administered to a subject to be treated More specifically, F cells are introduced into MSC / by introducing cytosine deaminase (CD) into mesenchymal stem cells.
- CD cytosine deaminase

- the F cells of the present invention may have an advantage of being able to be immediately administered to a patient in need of treatment with enhancement of an anticancer effect.
- Storing cells in a frozen or frozen state in the present invention may include any means known in the art for allowing the cells to remain frozen, such as cryopreservation, freezing medium, and the like.
- CS10 2-10% DMSO medium, more specifically 2, 5, 10% DMSO medium, and the like can be used.
- F cells of the present invention show a similar in vitro colony forming ability as compared to I cells, and also effectively exhibits the viewer effect of conventional MSC / CD.
- F cells show an inhibitory effect on various tumors, including brain tumors and subcutaneous tumors, more than two times compared to I cells or controls in vivo, and are highly elevated when combined with existing anticancer drug, temozolomide (TMZ). It is characterized by having the effect of showing an anticancer effect.
- F cells have a significant anti-cancer effect and anticancer activity that cannot be predicted in vitro compared to other I cells due to the intrinsic activity of F cells given according to the manufacturing method, although there is no difference in colony forming ability. There is an advantage of showing the effect as a bunt.
- the F cells of the present invention can be differentiated from conventionally used I cells by being prepared through a preparation method that does not additionally perform a culture step after freezing.

It was confirmed that there is a synergistic effect even better than the combined effect of treatment with anticancer drugs.
- Temozolomide, carmustine, and irinotecan are all known as anticancer agents that act on proliferating cells, so if they affect the survival of not only U87 / GFP cells but also proliferating F cells, [F cells (MSC / CD) +5 -FC's anticancer effect may be reduced. Contrary to expectations, however, when [F cell (MSC / CD) + 5FC] is used in combination treatment with temozolomide in FIG. 9E, synergistic effects may be increased rather than administration of only anticancer agent alone or in combination.
