Abstract

本发明公开了六氢β-酸双脂肪酸酯及其作为动物饲料添加剂的应用。本发明的如式1的六氢β-酸双脂肪酸酯(R1=异丙基,异丁基或2-丁基;R2=甲基—十七烷基中的任意一个烷基)对畜禽低毒或者无毒,适合作为饲料用生长促进剂,在养殖行业具有非常好的应用前景。

Description

六氢β-酸双脂肪酸酯及其作为动物饲料添加剂的应用 技术领域:
本发明属于畜禽饲料领域,具体涉及六氢β-酸双脂肪酸酯及其作为动物饲料添加剂的应用。
背景技术:
虽然一系列美国专利US8197863B2,US8012516B2,US20080213342A1,US20060269588,US20040219240,US7090873,US20060134248,US8012516,US20110311663,US7553504,US20130018106,US6251461等以及一系列欧洲专利EP2606742A1,EP1542543A1,EP2429311A2,EP1722635A2等都描述了酒花酸(Hops acids)及氢化β-酸类化合物作为替代饲用抗生素类生长促进剂的应用。
但是,首先这些专利所描述的都是应用酒花植物或酒花酸的提取物(α-酸及β-酸混合物等)作为饲料添加剂应用,而这些酒花提取物(混合物)其实已经在中国饲料添加剂品种目录上,只是作为食品用香料应用;其次,这些专利所依据的基本是体外试验结果和体外消化试验等,动物饲养试验结果很少;再次,这些天然产物及它们的酯等的安全性及稳定性,在满足作为动物饲料添加剂应用必需的要求方面没有资料支持。
发明内容:
本发明的第一个目的是提供具有高稳定性的、能够促进畜禽生长的,更适合作为饲料添加剂应用的六氢β-酸双脂肪酸酯。
本发明的六氢β-酸双脂肪酸酯,其结构式如式1所示:

R1=异丙基,异丁基或2-丁基;R2=甲基—十七烷基中的任意一个烷基。
优选,所述的R2=壬基、十一烷基或十七烷基。
上述六氢β-酸双脂肪酸酯在制备动物促生长饲料添加剂中的应用。
所述的动物为各个生长阶段的猪、鸡、鸭、鹅、肉牛、奶牛、羊、鱼、虾、狐、貂或貉等动物。
所述的动物促生长饲料为全价配合饲料,添加如式1所示的六氢β-酸双脂肪酸酯作为动物促生长饲料添加剂时的用量为全价配合饲料质量的0.1~200ppm。
六氢β-酸含有六氢蛇麻酮、六氢加蛇麻酮、六氢合蛇麻酮3个主要组分,属于现有技术中的物质。
六氢β-酸双脂肪酸酯包括六氢β-酸双乙酯到18碳的双硬脂酸酯。
本发明的如式1所示的六氢β-酸双脂肪酸酯对畜禽低毒或者无毒,具备饲用生长促进剂效力,且稳定性高,在养殖行业具有非常好的应用前景。
具体实施方式:
以下实施例是对本发明的进一步说明,而不是对本发明的限制。
实施例1:β-酸提取工艺
称取酒花浸膏β-acid浸膏1KG于5L容器中,加入1L无水乙醇,90gKOH(溶于100mL水中),随后室温搅拌均匀,搅拌下加入3L自来水,搅拌1小时后,静置30min,抽滤,除去油脂和不溶物。收集滤液,分液,收集下层水相。 搅拌下向水相中通入CO2,析出大量固体,加入1.2L 10%KOH水溶液。室温搅拌均匀,加入200mL正己烷,分液,除去有机相和中间的油层,收集水相。滤液用20%磷酸水溶液调节,正己烷(1L)提取。有机相真空浓缩后,得到β-acid粗产品约400g(含量90%左右)。
实施例2六氢β-酸氢化合成工艺

氢化釜中加入约360gβ-酸粗产品,随后加入95%乙醇至2.5L,加入36g 10%钯碳,开启搅拌,体系置换氮气三次,氢气三次,0.6~0.7MPa压力下室温反应12~15h,取样检测。反应完毕将反应液抽出,过滤除去钯碳,滤液真空浓缩掉3/4溶剂后,搅拌下滴加入3.5L自来水,搅拌1~2h左右,抽滤,收集固体得到六氢β-酸,50°干燥过夜,称重340g左右。
实施例3六氢β-酸二酯的合成

实施例3.1六氢β-酸癸酸二酯的合成
取100g干燥的六氢β-酸(HPLC 98%)投入2L干燥的反应器中,氮气保护。加500ml正庚烷,1gDMAP。快速加137g癸酰氯,降温至0度。滴加72g三乙胺。加完后继续搅拌2h。点板,原料点消失。过滤除去三乙胺盐酸盐,减压脱溶剂得到黄色油状物。柱层析分离得到88g六氢β-酸癸酸二酯,收率约50%。
实施例3.2六氢β-酸月桂酸二酯的合成
取100g干燥的六氢β-酸(HPLC 98%)投入2L干燥的反应器中,氮气保护。加500ml正庚烷,1gDMAP。快速加157g十二酰氯,降温至0度。滴加72g三乙胺。加完后继续搅拌2h。点板,原料点消失。过滤除去三乙胺盐酸盐,减压脱溶剂得到黄色油状物。柱层析分离得到95g六氢β-酸月桂酸二酯,收率约50%。
实施例3.3六氢β-酸硬脂酸二酯的合成
取100g干燥的六氢β-酸(HPLC 98%)投入2L干燥的反应器中,氮气保护。加500ml正庚烷,1gDMAP。快速加218g硬脂酸酰氯,降温至0度。滴加72g三乙胺。加完后继续搅拌2h。点板,原料点消失。过滤除去三乙胺盐酸盐,减压脱溶剂得到黄色油状物。柱层析分离得到116g六氢β-酸硬脂酸二酯,收率约50%。
实施例4:不同种类六氢β-酸双脂肪酸酯在肉鸡料中的应用效果
800羽1日龄快大黄羽肉鸡按表1随机分成8个处理组,每组100羽,并分别在每组中添加六氢β-酸双脂肪酸酯、六氢β-酸单脂肪酸酯或六氢β-酸。试验期笼养,自由采食和自由饮水。试验期共30天,试验结果显示,不同种类的六氢β-酸双脂肪酸酯对试验鸡的增重和料肉比方面均有显著改善,且六氢β-酸双脂肪酸酯的效果优于相同剂量六氢β-酸或单酯的效果(表1)。

实施例5:不同剂量六氢β-酸月桂酸二酯在肉鸡料中的应用效果
500羽1日龄快大黄羽肉鸡按表2随机分成8个处理组,每组100羽,并分别在每组饲料中添加不同剂量的六氢β-酸月桂酸二酯。试验期笼养,自由采食和 自由饮水。试验期共30天,试验结果显示,不同剂量的六氢β-酸月桂酸二酯对试验鸡的增重和料肉比均有显著改善,且呈剂量效应(表2)。

实施例5六氢β-酸癸酸二酯在猪料中的应用效果
150头体重相近的70日龄杜大长三元杂瘦肉型小猪随机分为5组,每组3个重复,每个重复10头猪。按表3每组饲料中添加不同剂量的六氢β-酸癸酸二酯。试验期自由采食和饮水。试验期共28天,结果表明六氢β-酸癸酸二酯组在试验猪的增重和料肉比方面均有显著改善(表3)。

实施例6六氢β-酸癸酸二酯在肉鸭料中的应用效果
900羽1日龄樱桃谷肉鸭按表4随机分成3个试验组,每组4个重复,每个重复50羽,并分别在每组饲料中添加六氢β-酸癸酸二酯或六氢β-酸。试验期笼养,自由采食和自由饮水。试验期共28天。试验结果显示,六氢β-酸癸酸二酯 组在试验鸭的增重和料肉比方面均有显著提高,且效果优于相同剂量的六氢β-酸(表4)。

Claims (5)

  1. 六氢β-酸双脂肪酸酯,其结构式如式1所示:

    R1=异丙基,异丁基或2-丁基;R2=甲基—十七烷基中的任意一个烷基。
  2. 根据权利要求1所述的六氢β-酸双脂肪酸酯,其特征在于,所述的R2=壬基、十一烷基或十七烷基。
  3. 权利要求1所述的六氢β-酸双脂肪酸酯在制备动物促生长饲料添加剂中的应用。
  4. 根据权利要求3所述的应用,其特征在于,所述的动物为各个生长阶段的猪、鸡、鸭、鹅、肉牛、奶牛、羊、鱼、虾、狐、貂或貉。
  5. 根据权利要求4所述的应用,其特征在于,添加所述的六氢β-酸双脂肪酸酯作为动物促生长饲料添加剂时的用量为饲料质量的0.1~200ppm。
