Description
本発明は、体細胞から誘導される重層上皮組織形成能を有する細胞、及びその製造方法に関する。また、本発明は、該重層上皮組織形成能を有する細胞を利用した、皮膚、粘膜又は角膜組織再生用の細胞製剤、皮膚、粘膜又は角膜潰瘍の治療方法、皮膚、粘膜又は角膜疾患に対する被験物質の薬効判定方法、及び皮膚、粘膜又は角膜に対する外的要因のストレス判定方法に関する。更に、本発明は、体細胞から重層上皮細胞に誘導するための重層上皮細胞調製用組成物に関する。
重層上皮組織は、表皮、粘膜や角膜など、外界に触れる部位において身体内部を機械的障害、感染などの外的要因から保護すると同時に、血液、体液の漏出、蒸散を防ぐ、外皮の役割を担っている。重層上皮組織は基底層に存在する前駆細胞、ないしは幹細胞が自己複製、分化することによって重層し形成される。
外皮が障害されるもっとも代表的な病態として、熱傷、外傷、医原性損傷(腫瘍切除後等)、褥瘡などの外的要因や、糖尿病、末梢循環不全などの内的要因でもたらされる皮膚、潰瘍がある。外皮の障害を来すあらゆる要因が潰瘍の原因となりうる。また、口腔粘膜、肛門粘膜欠損など消化管外皮が障害される消化管潰瘍、角膜が障害される角膜潰瘍なども当該病態に含まれる。皮膚潰瘍は疼痛、感染、出血などの原因となるほか、広範な場合には、それのみで致死的である。粘膜潰瘍は、疼痛、感染、出血や消化管の穿孔の原因となりうる。角膜潰瘍は視力障害の原因となる。医学の発達や、生活習慣病の蔓延によって、褥瘡、糖尿病潰瘍、血行障害を基礎疾患とする難治性の皮膚潰瘍の罹患者は今後益々増加すると予測される。
上皮細胞は前駆細胞、幹細胞としての性質を有するため、上皮の潰瘍性病変は通常、潰瘍周囲に存在する上皮細胞の遊走により修復される。したがって限局した潰瘍性病変の場合には創部の清浄を保ち、軟膏や創傷被覆材で保護することによって瘢痕治癒をえることができる。しかし、創部が広範な場合や、感染や低栄養、高血糖、末梢循環障害など治癒を阻害する因子が存在する場合には、潰瘍は難治化する。従来このような場合には、身体他部位から、植皮術や皮弁形成術など外科的処置を介して皮膚を移植して治療が行われる。しかし、特に褥瘡や糖尿病性潰瘍、循環不全による潰瘍を有する患者の多くは全身状態が悪く、全身麻酔、外科的手術に伴う危険性が高くなる(非特許文献1、2参照)。また、重症熱傷のように創部が著しく広範な場合には、残存する自己の皮膚組織のみでは潰瘍部分の十分な縮小、閉鎖を得ることができず、感染や循環動態不全から死に至ることもある。さらに、消化管、特に重層上皮からなる口腔、舌、咽頭や肛門の粘膜欠損に対しては、皮弁形成術を要する。また、角膜潰瘍に対しては重篤な場合には、角膜移植を要することがある。
そこで、特に重症熱傷に対して近年、自己皮膚組織の一部を採取し、細胞培養を行い作成した自己表皮細胞シートを移植する治療が行われるようになった。しかし、本治療の適応となるような患者は、残存皮膚組織の絶対的な量が不足することから、採取、表皮細胞シートの作成に使用することができる皮膚組織量も不足し、皮膚移植なしでも患者の生存を得ることができる数週間の間に十分量の表皮細胞シートを作成することは困難なことが多々あった。
また、褥瘡や糖尿病性潰瘍、循環不全による潰瘍などの難治性潰瘍においては、従来行われている植皮術や皮弁形成術などの外科的処置によって必ずしも創部の閉鎖を得ることができないこともある。植皮術や皮弁形成術などの外科的処置は、移植する皮膚を採取する必要があることから、創部の閉鎖を得ることができない場合には、身体に対して負荷をかけるのみになってしまうため、より低侵襲で効率よく創部の上皮化を得ること手段が必要であった。
近年、Oct3/4、Klf4、c-Myc及びSox2をコードする各々の遺伝子を体細胞に導入することにより、体細胞を初期化して誘導多能性幹(iPS)細胞に誘導する技術が報告され、再生医薬の分野で革新的な技術が提供されている(特許文献1、非特許文献3-4参照)。しかしながら、iPS細胞の作製、iPS細胞から上皮細胞への誘導には長期間を有することから、本技術を重症熱傷患者に対して用いるには、更なる技術的課題を解決する必要がある。また、iPS作製過程で体細胞が多能性を獲得した状態から、再度上皮細胞へ誘導を行うことになるため、癌化の可能性など、多能性を有する状態を解する諸問題があるうえ、作製効率が悪いことから、比較的経過の遅い難治性皮膚潰瘍や、粘膜潰瘍、角膜潰瘍などの患者に対して用いるにも、更なる技術的課題を解決する必要がある。
一方、発生の過程で、ひとたび細胞系譜の定まった体細胞は、同一細胞系譜内で増殖、分化を継続する。対して、生理的には認められない細胞系譜の転換を直接転換とよび、皮膚線維芽細胞から筋細胞(非特許文献3)、神経細胞(非特許文献6参照)、心筋細胞(非特許文献7参照)、肝細胞(非特許文献8参照)、血液幹細胞(非特許文献9参照)などへの直接転換技術が報告されている。
このような従来技術を背景として、皮膚線維芽細胞、脂肪由来間葉系細胞をはじめとする体細胞から直接誘導でき、身体外皮として働きうる重層上皮組織形成能を有する細胞を開発し、広く皮膚潰瘍治療に使用できる細胞供給源の提供を実現させることが切望されているが、未だ重層上皮組織形成能を有する細胞が体細胞から直接転換された報告はない。

そこで、本発明は、上記従来技術の課題を解決するため、身体外皮として働きうる重層上皮組織形成能を有する細胞を体細胞から直接転換させることを目的の一つとする。より具体的には、本発明は、体の内外の遊離面を覆う外皮(皮膚、粘膜、角膜上皮)として働く重層上皮組織を形成する能力をもった細胞供給源を提供するための技術を確立することを目的とする。また、本発明は、かかる体細胞から誘導した重層上皮組織形成能を有する細胞の様々な用途を提供することを目的とする。
本発明者らは、前記課題を解決するために代表的な体細胞として皮膚線維芽細胞を、外皮として働きうる重層上皮組織を形成する能力を有する代表的な細胞として皮膚から採取されるケラチノサイトを選択し、各々の細胞が発現している遺伝子をDNAマイクロアレイ、microRNAマイクロアレイのデータにて調べ、ケラチノサイトに特異的、ないし相対的に強発現する遺伝子の導入によって、ケラチノサイトと同等の形態、特性を有する細胞を製造できることを見出した。実際、斯くして得られた誘導線維芽細胞は、ケラチノサイトの増殖に最適化された単層培養又はフィーダー培養にて増殖可能であり、ケラチノサイトの特異的マーカーを発現することを確認した。さらに、上記誘導線維芽細胞は、単層培養条件下で、自己複製能と終分化を示すことを確認した。さらに、誘導線維芽細胞を、皮膚線維芽細胞を含むコラーゲンゲルを足場として3次元培養した場合に、重層上皮組織を形成できることを確認した。本発明は、これらの知見に基づいて、更に検討を重ねることにより完成したものである。
即ち、本発明は、下記に掲げる態様の発明を提供する。
項1. 重層上皮組織形成能を有する細胞に相対的に強発現する少なくとも1種の遺伝子を体細胞に導入する工程を含む、重層上皮組織形成能を有する細胞の製造方法。

本発明によれば、ケラチノサイト、消化管上皮細胞、角膜上皮細胞と同等の特性を有する重層上皮組織形成能を有する細胞を提供できる。また、かかる重層上皮組織形成能を有する細胞は、熱傷、外傷、褥瘡、糖尿病、末梢循環不全などの皮膚潰瘍、消化管潰瘍、角膜潰瘍など外皮組織の潰瘍を伴う病態の治療に有効な医療手段又は細胞製剤を提供することができる。また、本発明によれば、皮膚潰瘍のみならず、表皮、消化管上皮、角膜上皮などの上皮に生じる広範な疾患について、患者の体細胞(患部の体細胞に限らない。例えば血液、脂肪組織間質細胞等)から重層上皮組織形成能を有する細胞を作成し、作成された細胞に対し様々な解析を行うことで疾患の病態解明又は治療に寄与できる。特にヒトの体細胞から作成した重層上皮組織形成能を有する細胞は、創薬や薬品開発の材料として、薬効確認の点においても適している。また、自己複製能と終分化の相反する特性を有する重層上皮組織形成能を有する細胞を用いることによって、癌など上皮系幹細胞を起源とする病態解明に寄与できる。
1.重層上皮組織形成能を有する細胞の製造方法、及び重層上皮組織形成能を有する細胞の用途
本発明において、「重層上皮組織形成能を有する細胞」とは、表皮、粘膜や角膜など、外界に触れる部位において身体内部を機械的障害、感染などの外的要因から保護する重層上皮の前駆細胞もしくは幹細胞として働く能力を備えている細胞(換言すれば、上皮幹細胞)のことを意味し、生体内の体細胞であるケラチノサイト、消化管上皮細胞、角膜上皮細胞も、本発明によって人工的に体細胞から誘導された細胞も含む。重層上皮組織形成能を有する細胞は、単層培養条件下において培養条件を調整することによって、自己複製能と終分化の双方の特性を呈する。又は、重層上皮組織形成能を有する細胞は、皮膚線維芽細胞などを含んだコラーゲンゲルを担体とした3次元培養においては重層上皮組織様の構造体を形成する能力をもつ。

本発明の重層上皮組織形成能を有する細胞の製造方法は、体細胞に、重層上皮組織形成能を有する細胞(例えば、ケラチノサイト、消化管上皮細胞又は角膜上皮細胞)に相対的に強く発現している少なくとも1種の遺伝子を導入する工程を含むことを特徴とする。以下、体細胞に導入する遺伝子を「導入遺伝子」と呼ぶ。導入遺伝子は、重層上皮組織形成能を有する細胞に相対的に強発現する遺伝子を複数種類導入してもよいし、重層上皮組織形成能を有する細胞に相対的に強発現する遺伝子に加えて、相対的に強発現しない遺伝子を組み合わせてもよい。
ここで、「導入遺伝子」は、タンパク質をコードする遺伝子のみならず、マイクロRNA等のnoncoding RNAも含む。また、「重層上皮組織形成能を有する細胞に相対的に強発現する遺伝子」とは、皮膚線維芽細胞等の間葉系細胞に比較して、重層上皮組織形成能を有する細胞において発現量が多いことがリアルタイムPCR法やマイクロアレイ等の遺伝子発現量の定量的評価法によって確認される遺伝子を指す。なお、「相対的に強発現しない遺伝子」とは、間葉系細胞に比較して、重層上皮組織形成能を有する細胞において発現量が少ない遺伝子を指す。
重層上皮組織形成能を有する細胞に相対的に強発現する遺伝子には、少なくとも次のタンパク質をコードする遺伝子を含む。TFAP2A遺伝子、TFAP2C遺伝子、GRHLファミリー遺伝子(GRHL1遺伝子、GRHL2遺伝子、及びGRHL3遺伝子等)、BNC1遺伝子、MYCファミリー遺伝子(c-Myc、N-Myc、及びL-Myc等)、GATA3遺伝子、OVOL1遺伝子、OVOL2遺伝子、ESRP1遺伝子、ESRP2遺伝子、TP63遺伝子、DNP63A遺伝子、MAPK13遺伝子、ARNTL2遺伝子、LASS3遺伝子、EHF遺伝子、ZNF165遺伝子、ZNF750遺伝子、ZBED2遺伝子、IRX4遺伝子、SOX7遺伝子、SOX9遺伝子、FOXQ1遺伝子、PPP1R13L遺伝子、KLF4遺伝子。また、重層上皮組織形成能を有する細胞に相対的に強発現する遺伝子には、少なくとも次のnoncoding RNAも含む。mir-182、mir-183、mir-96、mir-200b、mir-200a、mir-429、mir-200c、mir-141、mir-203、mir-205、mir-135b、mir-17、mir-18a、mir-19a、mir-19b-1、mir-20a、mir-92a-1、mir-367。
特に、本発明の重層上皮組織形成能を有する細胞の製造方法は、(1)TFAP2A遺伝子またはTFAP2C遺伝子、(2)GRHLファミリー遺伝子から選択された少なくとも一種の遺伝子、(3)BNC1遺伝子及び(4)MYCファミリー遺伝子から選択された少なくとも一種の遺伝子を体細胞に遺伝子導入する工程を含むことが好ましい。さらに、(1)~(4)に加えて、(5)重層上皮組織形成能を有する細胞に相対的に強発現する少なくとも1種の遺伝子を導入してもよい。
GRHLファミリー遺伝子としては、GRHL1遺伝子、GRHL2遺伝子、及びGRHL3遺伝子等が挙げられる。これらのGRHLファミリー遺伝子は、1種単独で使用してもよく、また2種以上を組み合わせて使用してもよい。Mycファミリー遺伝子としては、c-Myc、N-Myc、及びL-Myc等が挙げられる。これらのMycファミリー遺伝子は、1種単独で使用してもよく、また2種以上を組み合わせて使用してもよい。これらのMycファミリー遺伝子の中でも、本発明では、好ましくはc-Myc遺伝子又はL-Myc遺伝子、更に好ましくはc-Myc遺伝子が使用される。c-Myc遺伝子は、細胞の分化及び増殖に関与する転写制御因子として知られており(S. Adhikary, M. Elilers, Nat. Rav. Mol. Cell Biol., 6, pp635-645, 2005)、その塩基配列は公知である(表1参照)。
また、本発明の他の重層上皮組織形成能を有する細胞の製造方法は、導入遺伝子(直接転換因子)として、GATA3遺伝子、OVOL1遺伝子、OVOL2遺伝子、ESRP1遺伝子、TFAP2A遺伝子、ID1遺伝子、GRHL1遺伝子、GRHL2遺伝子、GRHL3遺伝子、TP63遺伝子、DNP63A遺伝子、MAPK13遺伝子、ARNTL2遺伝子、BNC1遺伝子、LASS3遺伝子、EHF遺伝子、ZNF165遺伝子、ZNF750遺伝子、ZBED2遺伝子、IRX2遺伝子、IRX4遺伝子、SOX7遺伝子、SOX9遺伝子、FOXQ1遺伝子、PPP1R13L遺伝子、KLF4遺伝子、c-MYC遺伝子の全部もしくは一部の遺伝子を、上記体細胞に導入することにより、体細胞を重層上皮組織形成能を有する細胞に誘導する。
本発明において、体細胞に導入される遺伝子のうち、TFAP2A遺伝子、GRHL2遺伝子、BNC1遺伝子、c-MYC遺伝子は特に重要であるが、その他の遺伝子を組み合わせて遺伝子導入することによって、細胞増殖能の上昇、重層分化能の上昇、細胞遊走能の上昇をはかるなど、目的に応じた機能的特質を付与することができる。例えば、GATA3遺伝子、TFAP2A遺伝子、GRHL2遺伝子、TP63遺伝子、BNC1遺伝子、EHF遺伝子、ZNF165遺伝子及びc-MYC遺伝子を体細胞に遺伝子導入する工程を含む製造方法と、OVOL1遺伝子、TFAP2A遺伝子、GRHL2遺伝子、BNC1遺伝子、LASS3遺伝子、ZBED2遺伝子、SOX7遺伝子、SOX9遺伝子及びc-MYC遺伝子を体細胞に遺伝子導入する工程を含む製造方法とでは、前者は終分化能を高くすることができ、後者は増殖能を高くすることができる。
本発明で使用するいずれの遺伝子も、その塩基配列は公知である(表1)。なお、本明細書において、NCBIとは、米国立生物工学情報センター(National Center for Biotechnology Information)の略であり、表1のAccession No.もNCBIが提供するデータベースに登録されているものである。
これらの遺伝子の多くの由来は、ヒトを含む哺乳動物において共通して存在しており、任意の哺乳動物由来のものを使用できるが、導入する体細胞の由来に応じて適宜選択することが望ましい。例えば、体細胞としてヒト由来のものを使用する場合であれば、上記導入遺伝子はヒト由来であることが望ましい。また、上記導入遺伝子は、野生型遺伝子以外に、その遺伝子産物のアミノ酸配列における数個(例えば1~10個、好ましくは1~6個、更に好ましくは1~4個、より好ましくは1~3個、特に好ましくは1又は2個)のアミノ酸が置換、欠失、及び/又は挿入されており、且つ、野生型の遺伝子産物と同等の機能を有する変異遺伝子産物をコードしている変異遺伝子であってもよい。
本発明において、上記導入遺伝子は、公知の配列情報に基づいて、常法に従って調製することができる。例えば、哺乳動物由来の細胞からRNAを抽出し、常法に従ってクローニングすることにより、目的とする遺伝子のcDNAを調製することができる。
本発明において、重層上皮組織形成能を有する細胞に誘導される「体細胞」としては、その種類については特に制限されず、あらゆる組織又は部位由来のものが使用できる。本発明で使用される体細胞としては、例えば、皮膚、皮下脂肪、筋肉、胎盤、骨、軟骨、血液、角膜実質等の組織由来のものが挙げられ、より具体的には、皮膚線維芽細胞、皮下脂肪組織由来間質細胞(皮下脂肪細胞)、胚性線維芽細胞、脂肪細胞、筋肉細胞、骨芽細胞、軟骨細胞、循環血中の単核球、角膜実質内のケラトサイト等が例示される。これらの中でも、生体に対して侵襲が軽微であり、且つより効率的に重層上皮組織形成能を有する細胞を作製するという観点から、皮膚由来細胞、皮下脂肪由来細胞、もしくは血液由来細胞が好ましく、特に皮膚線維芽細胞及び皮下脂肪組織由来間質細胞、循環血液中の単核球が好ましい。このように、様々な細胞から材料を選択でき、とりわけ皮膚由来細胞や皮下脂肪由来細胞、循環血液中の単核球等の入手容易な細胞をも使用できることは、患者の負担を軽減し、細胞の安定な入手の点でも、臨床上の利点がある。また、上記体細胞として、市販品を使用してもよく、またES細胞や間葉系幹細胞等から分化させた体細胞を使用することもできる。
また、上記体細胞は、重層上皮組織形成能を有する細胞の使用目的に応じて、ヒト、マウス、ラット、ハムスター、ウサギ、ネコ、イヌ、ヒツジ、ブタ、ウシ、ヤギ、サル等の哺乳動物由来のものから適宜選択されるが、ヒトの治療、病態解明又は薬効確認等のヒトを対象とする目的で使用する場合にはヒト由来のものが好適である。また、ヒト由来の体細胞を使用する場合、胎児、幼児、小児、及び成人のいずれに由来するものであってもよい。重層上皮組織形成能を有する細胞をヒトの治療、病態解明又は薬効確認等の目的で使用する場合には、患者から採取した体細胞を使用することが望ましい。
上記導入遺伝子の体細胞への導入は、動物細胞のトランスフェクションにおいて通常使用される方法で行うことができる。具体的には、上記導入遺伝子を体細胞へ導入する方法として、ベクターを使用する方法;リン酸カルシウム法;リポフェクション法;エレクトロポレーション法;マイクロインジェクション法等が例示される。これらの中でも、導入効率の点から、ベクターを使用する方法が好ましい。ベクターを使用して上記導入遺伝子を体細胞に導入する場合には、ベクターとして、ウイルスベクター、非ウイルスベクター、人工ウイルス等を用いることができるが、アデノウイルス及びレトロウイルス、レンチウイルス等のウイルスベクターが、安全性の観点から好適に使用される。なお、ベクターを使用する場合であって、上記導入遺伝子が複数の場合は、各々別のベクターに組み込まれていてもよいし、1つのベクターに2種以上の導入遺伝子が組み込まれていてもよい。
斯くして上記導入遺伝子が導入された体細胞は、重層上皮組織形成能を有する細胞に誘導することができる。重層上皮組織形成能を有する細胞に誘導された細胞の選択は、ケラチノサイトの単離、増幅に適した培養条件下での増殖能の有無、および上皮細胞としての特性を有するか否かを指標として行うことができる。このような重層上皮組織形成能を有する細胞は、具体的には、ケラチノサイトを単離、増幅するのに適したフィーダー細胞(3T3-J2フィーダー細胞、3T3細胞、マウス胚線維芽細胞、ヒト皮膚線維芽細胞などに対して、マイトマイシンCや放射線処理を行い増殖能を失活させたもの)上や、ケラチノサイト無血清培地で培養することによって重層上皮組織形成能を有する細胞に比較して高い増殖能を呈することから、継代処理を継続することによって選択が可能である。フィーダー上でのケラチノサイトの分裂可能回数を比較的に向上させることができるRhoキナーゼ阻害剤(Y27632等)を加えることも有効である。また、上皮細胞に特異的な表面抗原(Epi-CAM等)を用いてフローサイトメトリーや磁気細胞分離装置を用いた細胞分離を行うことにより、重層上皮組織形成能を有する細胞の純度を高めることが可能である。また、体細胞に、予め上皮細胞マーカー遺伝子(CDH1, Epi-CAM等)のプロモーターに薬剤耐性遺伝子を結合して作ったレポーター遺伝子コンストラクトを導入しておいた場合には、上皮細胞の特性を獲得した細胞は薬剤存在下で生育可能になるので、薬剤存在下での生育を指標として、選択することもできる。
斯くして得られた重層上皮組織形成能を有する細胞は、フィーダー上でRhoキナーゼ阻害剤を加えた液体培地内で培養すると増殖可能であり、通常行われる10-20継代程度までは重層上皮組織形成能を維持したまま安定に増殖することができる。重層上皮組織形成能を有する細胞の培養には、動物細胞の培養に通常使用される培地を用いることができる。重層上皮組織形成能を有する細胞の培養に使用される好適な培地の一例として、無血清ケラチノサイト培地(Keratinocyte-SFM、Life technologies社)などが例示される。bFGFなど、培養条件下でのケラチノサイトの増殖を早めるサイトカインや各種の薬理活性物質を添加することも有用である。
斯くして得られた重層上皮組織形成能を有する細胞は、in vivoで、皮膚線維芽細胞や脂肪組織由来間質細胞などの間葉系細胞を含む細胞外基質上、たとえば皮膚潰瘍の創部で重層上皮組織を形成することができる。さらに、in vitroで、皮膚線維芽細胞や脂肪組織由来間質細胞などの間葉系細胞を含むコラーゲンゲル上で培養し、気相液相界面を形成することにより重層扁平上皮様の三次元構造を有する上皮様組織を形成することができる。
このように、本発明で得られる重層上皮組織形成能を有する細胞は、増殖能を有しており、且つ生体内外で重層上皮組織の再生が可能であるので、熱傷、外傷、医原性損傷(腫瘍切除後等)、褥瘡、糖尿病、末梢循環不全でもたらされる皮膚潰瘍や、鼻腔、口腔、肛門周囲粘膜などの消化管潰瘍、角膜潰瘍の治療に有効であり、上皮組織再生用の細胞製剤(医薬組成物)として使用できる。上記重層上皮組織形成能を有する細胞は、そのまま単独で皮膚、消化管、角膜疾患部位に適用してもよい。
上記重層上皮組織形成能を有する細胞を上皮組織再生用の細胞製剤として調製する場合、上記重層上皮組織形成能を有する細胞と共に、必要に応じて、薬学的に許容される希釈用担体を含んでいてもよい。ここで、薬学的に許容される希釈用担体としては、例えば、生理食塩水、緩衝液等が例示される。更に、当該細胞製剤は、必要に応じて、薬理活性成分や重層上皮組織形成能を有する細胞の栄養源となる成分が含まれていてもよい。
上記重層上皮組織形成能を有する細胞は培養条件下で重層上皮様組織を形成した上皮細胞シートとして皮膚、消化管、角膜疾患部位に適用してもよい。
上記重層上皮組織形成能を有する細胞を上皮組織再生用の上皮細胞シートとして作製する場合、上記重層上皮組織形成能を有する細胞と共に、必要に応じて、薬理活性成分や重層上皮組織形成能を有する細胞の栄養源となる成分と併用してもよい。
上記重層上皮組織形成能を有する細胞は、皮膚線維芽細胞や脂肪組織由来間質細胞などの間葉系細胞を含むコラーゲンなどの細胞外基質を足場として、重層扁平上皮様の三次元構造を有する上皮様組織を作製したのちに皮膚、消化管、角膜疾患部位に適用してもよい。
上記重層上皮組織形成能を有する細胞を上皮組織再生用の三次元構造を有する上皮様組織として作製する場合、上記重層上皮組織形成能を有する細胞と共に、必要に応じて、薬理活性成分や重層上皮組織形成能を有する細胞の栄養源となる成分と併用してもよい。このように足場材料を使用することによって、より速やかに移植部位での重層上皮様構造が可能となり、上皮組織の再生を一層促進することが可能になる。
使用可能な足場材料としては、薬学的に許容される限り、特に制限されず、適用する軟骨組織の部位に応じて適宜選択されるが、例えば、ゲル状体の、生体親和性の高い(biocompatible)材料が挙げられる。使用可能な足場材料として、好ましくはコラーゲン、ファイブロネクチン、ヒアルロン酸及びこれらの複合体等が例示される。これらの足場材料は、1種単独で使用してもよく、2種以上を組み合わせて使用してもよい。
また、上記足場材料の形状についても特に制限されず、当該細胞製剤の適用対象となる上皮組織の損傷部位の形状に応じて適宜設計すればよい。
当該細胞製剤を上皮組織の疾患部位に適用する方法については、当該細胞製剤のタイプ、適用される上皮組織の部位等に応じて適宜設定されるが、例えば治療目的の皮膚潰瘍の部位に当該細胞製剤を直接散布する方法、植皮術に準じてシート、三次元構造体を縫合固定する方法などがあげられる。
また、上皮組織の疾患部位に適用される当該細胞製剤の投与量については、細胞製剤のタイプ、上皮組織の部位、症状の程度、患者の年齢や性別等に基づいて、上皮組織の再生に有効な量を適宜設定すればよい。
また、上記重層上皮組織形成能を有する細胞が投与されて、上記重層上皮組織形成能を有する細胞から形成された上皮組織を有する非ヒト哺乳動物は、上皮組織に対する被験物質の薬効を評価、分析するためのツールとして使用できる。即ち、上記重層上皮組織形成能を有する細胞から形成された上皮組織を有する非ヒト哺乳動物に被験物質を投与し、当該上皮組織に対する被験物質の薬効を判定、分析することによって、上皮組織に対する被験物質の薬効を評価、分析することができる。ここで、被験物質とは、上皮組織に対する薬効の評価、分析対象となる物質であり、具体的には、上皮疾患の治療薬の候補物質が挙げられる。非ヒト哺乳動物としては、マウス、ラット、ハムスター、ウサギ、ネコ、イヌ、ヒツジ、ブタ、ウシ、ヤギ、サル等が適宜選択される。
また、上皮上記重層上皮組織形成能を有する細胞から形成された上皮組織を有する非ヒト哺乳動物は、抗がん剤や放射線など、上皮組織に障害を及ぼす外的要因の上皮組織に対する影響を調べるモデルとして用いることができる。
また、上記重層上皮組織形成能を有する細胞および皮膚線維芽細胞を含むコラーゲンゲルなどの足場によって作成された三次元構造体は、上皮組織に対する被験物質の薬効を評価、分析するためのツールとして使用できる。即ち、上記重層上皮組織形成能を有する細胞および皮膚線維芽細胞を含むコラーゲンゲルなどの足場によって作成された三次元構造体に被験物質を投与し、当該上皮組織に対する被験物質の薬効を判定、分析することによって、上皮組織に対する被験物質の薬効を評価、分析することができる。ここで、被験物質とは、上皮組織に対する薬効の評価、分析対象となる物質であり、具体的には、上皮疾患の治療薬の候補物質が挙げられる。特に、末梢血循環単核球など、比較的侵襲が少なく採取可能な体細胞から誘導した重層上位形成能を有する細胞を用いることにより、多彩な遺伝的背景を有する多くのドナーに対する薬効を広く評価、分析することができる。
また、上記重層上皮組織形成能を有する細胞は、様々な上皮組織の病態を解明、分析するためのツールとして使用でき、更にはヒト体細胞から誘導した重層上皮組織形成能を有する細胞は、上皮疾患に対する創薬や薬品開発のためのツールとしても有用である。例えば、人体から取り出された体細胞に、導入遺伝子を導入して重層上皮組織形成能を有する細胞を製造し、さらにかかる細胞を培養して作製された上皮様組織に対し、被験物質を投与し、前記上皮様組織に対する被験物質の薬効を評価、分析したり、抗がん剤や放射線などのストレスを負荷し、前記上皮様組織に対するストレスを評価、分析したりすることができる。薬効又はストレスの評価、分析は、例えば、被験物質を投与した組織又はストレスを付加した組織と、投与又は付与していない組織とを比較して確認してもよい。
また、上記重層上皮組織形成能を有する細胞は、自己複製能と終分化という前駆細胞ないし幹細胞の特性を有することから、この特性を有さない皮膚線維芽細胞などの体細胞から重層上皮組織形成能を有する細胞への転換過程を解析することによって幹細胞の特性を明らかにする研究ツールとして用いることができる。
また、いわゆる癌の多くは重層上皮組織にその起源を有するものであり(扁平上皮癌)、上記重層上皮組織形成能を有する細胞は癌研究のツールとして用いることができる。
2.導入遺伝子候補の決定手法
以下に、実施例等に基づいて本発明を詳細に説明するが、本発明はこれらによって限定されるものではない。

体細胞の代表的なものとして皮膚線維芽細胞を、重層上皮組織形成能を有する細胞の代表的なものとしてケラチノサイトを選択した。直接転換を惹起しうる候補因子の選択のため、培養ヒトケラチノサイトおよび培養ヒト皮膚線維芽細胞の発現遺伝子のマイクロアレイの結果を米国NCBIが提供するデータベース(Accession:GDS1505 ID: 1505)より得た(図1)。
参考文献1より表皮が発生する前後であるマウスの胎生11.5日と14.5日目のmRNA発現をマイクロアレイで評価したデータを得た(図2)。
さらに定量性の高いデータを得るために、同一ヒト皮膚検体から分離した初代培養線維芽細胞および初代培養ケラチノサイトについて、マイクロアレイ解析を行った。
初代培養線維芽細胞の分離、培養は、参考文献2に記載の方法で行った。具体的には採取した皮膚検体を0.25%トリプシン4℃でオーバーナイト処理し、表皮を剥がした後、ディッシュ上にエクスプラントして、少量の線維芽細胞増殖培地(FGM)を加え、皮膚検体からディッシュ上に線維芽細胞が遊走してくるのをまち、サブコンフルエントになった時点で継代した。
初代培養ケラチノサイトの分離、培養は、参考文献3に記載の方法で行った。具体的には、採取した皮膚検体を0.25%トリプシン4℃でオーバーナイト処理し、無血清ケラチノサイト培地(SFKGM)内で真皮を擦過し、真皮上に残存するケラチノサイトを回収した。SFKGM培地で培養を継続し、サブコンフルエントになった時点で継代した。
初代培養皮膚線維芽細胞、初代培養ケラチノサイトそれぞれ107個の細胞からtotal RNAを採取し、Gene ST1.0 microarray (Affimetrix社)を用いて、網羅的にmRNA発現を比較した(図3)。
初代培養線維芽細胞、初代培養ケラチノサイトの発現しているマイクロRNAの比較のために、初代培養線維芽細胞、初代培養ケラチノサイトを同一ドナーより採取した部位の異なる3皮膚検体より分離、培養し、107個の細胞からマイクロRNAを採取し、マイクロRNAマイクロアレイであるSurePrint G3 Human miRNA マイクロアレイ (Agilent Technologies社)を用いて、網羅的にマイクロRNA発現を比較した(図4)。
以上の解析より、少なくとも、TFAP2A遺伝子、TFAP2C遺伝子、GRHLファミリー遺伝子、BNC1遺伝子、MYCファミリー遺伝子、GATA3遺伝子、OVOL1遺伝子、OVOL2遺伝子、ESRP1遺伝子、ESRP2遺伝子、GRHL1遺伝子、GRHL2遺伝子、GRHL3遺伝子、TP63遺伝子、DNP63A遺伝子、MAPK13遺伝子、ARNTL2遺伝子、LASS3遺伝子、EHF遺伝子、ZNF165遺伝子、ZNF750遺伝子、ZBED2遺伝子、IRX4遺伝子、SOX7遺伝子、SOX9遺伝子、FOXQ1遺伝子、PPP1R13L遺伝子、KLF4遺伝子、hsa-mir-182、hsa-mir-183、hsa-mir-96、hsa-mir-200b、hsa-mir-200a、hsa-mir-429、hsa-mir-200c、hsa-mir-141、hsa-mir-203、hsa-mir-205、hsa-mir-135b、hsa-mir-17、hsa-mir-18a、hsa-mir-19a、hsa-mir-19b-1、hsa-mir-20a、hsa-mir-92a-1、hsa-mir-367が、ケラチノサイトで相対的に強発現する遺伝子であることが確認された。
3.[実施例1] 皮膚線維芽細胞から重層上皮組織形成能を有する細胞への誘導
上記解析結果を踏まえ、表2に記載の27種の導入遺伝子を選択し、体細胞に導入するためのレンチウイルスベクターを準備した。候補として選択した転写因子、マイクロRNAおよび強制発現、発現抑制のために用意したレンチウイルスベクターの仔細を表2に示す。PPP1R13L遺伝子については、2つのtranscript variantについて、それぞれベクターを準備した。
上記解析結果を踏まえ、表2に記載の27種の導入遺伝子を選択し、体細胞に導入するためのレンチウイルスベクターを準備した。候補として選択した転写因子、マイクロRNAおよび強制発現、発現抑制のために用意したレンチウイルスベクターの仔細を表2に示す。PPP1R13L遺伝子については、2つのtranscript variantについて、それぞれベクターを準備した。
レンチウイルスベクターは、適切なクローンが存在した場合にはNBRC(独立行政法人製品評価技術基盤機構 バイオテクノロジー分野)よりヒトGatewayエントリークローンを購入し、pLenti7.3 /V5-DEST Gateway Vector(Life Technologies社)へ、常法に従いサブクローニングした。その他のものはメーカー(System Biosciences社、Gene Copoeia社、Origene社)より購入した。
表1の直接転換候補因子のレンチウイルスベクターを皮膚線維芽細胞に遺伝子導入するためにウィルスを作製した。ウィルス作製には、ViraPower(登録商標) HiPerform(登録商標) Lentiviral Expression Systems(Life Technologies社)を用いた。ウイルスの作製には6wellプレートに各ウェル106個の293FT細胞を播種し、オーバーナイト後に、各ウェルに1mlの10%FBS加Opti-MEM Reduced Serum Medium, GlutaMAX(登録商標) Supplement(Life Technologies社)に培地交換したのち、600ngのコンストラクト、1.2μgのViraPower(登録商標) Lentiviral Packaging Mix、7.2μlのLipofectamine 2000を用いて遺伝子導入を行った。24時間後に2mlの293FT細胞増殖培地で培地交換の後、さらに48時間後にウイルス液を含んだ培地を回収した。ウイルス含有培地を遠心分離ののち上澄を回収し、4分の1量のPEG-it Virus Precipitation Solution(System Biosciences社)を加え4℃オーバーナイトで静置した。1500×Gで遠心分離を行い、上澄を廃棄し、20μlのPBSで沈殿をピペッティングし、ウイルス濃縮液を得た。以上の操作によって回収したウイルス液を1単位として皮膚線維芽細胞への遺伝子導入に用いた。
皮膚線維芽細胞への遺伝子導入にはTransdux(登録商標) (System Biosciences社)を用いた。導入の24時間前に48ウェルプレートに1ウェルあたり6000個の皮膚線維芽細胞を播種した。ウイルス液を含んだ300μlの 線維芽細胞増殖培地に培地交換し、800×Gで30分間の遠心をかけた。再度新しいウイルス液を含んだ300μlの 線維芽細胞増殖培地に培地交換し、800×Gで30分間の遠心をかけたのちPBSでウェル内を洗浄し、線維芽細胞増殖培地を300μl加えた。
遺伝子導入後の細胞は導入1日目、2日目、4日目に線維芽細胞増殖培地で培地交換を行い6日目に、マイトマイシン処理した6ウェルプレートに用意した3T3-J2フィーダー細胞上に線維芽細胞増殖培地を用いて継代した。継代後は2日目に培地をRho-kinase inhibitor であるY27632(Wako社)を含むケラチノサイトF培地に交換し、以降2日毎に培地交換を行った(図5、図6)。
フィーダーとして用いた3T3-J2フィーダー細胞は、J-TEC社より分譲された細胞株である。10% ウシ新生仔血清をDMEM培地に加えた3T3細胞培地を用いて、常法に従い維持の上、フィーダー細胞として用いる前日にマイトマイシンC 10 μg/mlの培地で1時間処理ののち、2.0×105 細胞/ウェルの濃度で継代してフィーダーとして用いた。
ケラチノサイトF培地は常法に従い調整した。本試験では、33.8mlのF12培地(Life technologies社)、11.2mlのhigh glucose DMEM (Life technologies社)を混合したものに2.5mlのFBSを加え、アデニン(Sigma社)24 μg/ml、コレラ毒素(Wako社)8.4 ng/ml、 インスリン 5 μg/ml、ハイドロコルチゾン(Sigma社) 0.4 μg/ml、ペニシリン(Sigma社)100 U/ml、ストレプトマイシン(Sigma社)100 μg/ml、EGF 10 ng/mlに調整した。さらに、本試験においては、3T3フィーダー細胞上で培地にRho-kinase inhibitor (Y27632)を加えることにより培養ケラチノサイトの寿命を延長できるという指摘(参考文献4)に従い、上記ケラチノサイトF培地をY-27632(Wako社 )10μMに調整して用いた。また、比較のために同一条件で初代培養ケラチノサイトを培養した(図7)。
実施例1の27種の遺伝子(GATA3遺伝子、OVOL1遺伝子、OVOL2遺伝子、ESRP1遺伝子、TFAP2A遺伝子、ID1遺伝子、GRHL1遺伝子、GRHL2遺伝子、GRHL3遺伝子、TP63遺伝子、DNP63A遺伝子、MAPK13遺伝子、ARNTL2遺伝子、BNC1遺伝子、LASS3遺伝子、EHF遺伝子、ZNF165遺伝子、ZNF750遺伝子、ZBED2遺伝子、IRX2遺伝子、IRX4遺伝子、SOX7遺伝子、SOX9遺伝子、FOXQ1遺伝子、PPP1R13L遺伝子、KLF4遺伝子及びc-MYC遺伝子)を導入した誘導線維芽細胞は、ケラチノサイト類似の増殖、形態のコロニーを呈する細胞を得た(図8)。図8において、コロニーは、周囲のフィーダーおよび誘導されていない細胞と明瞭な境界をもっており、コロニーの辺縁には平坦な細胞が位置し、中央にむかって重積しており、図7のケラチノサイトのコロニー類似の形態を示している。
当該細胞は、特別な細胞分離のない場合であっても、ケラチノサイトを選択的に増殖させると考えられる3T3-J2フィーダー細胞上の培養環境でケラチノサイト類似の増殖、形態を維持したまま継代、増殖が可能であった(図9)。図9は、図8の誘導線維芽細胞のコロニーを確認した後、継代した第2継代のコロニーの図である。図9から、実施例1の誘導線維芽細胞は、該培養条件下において、ケラチノサイト類似の状態に誘導されていない非誘導線維芽細胞に比べ、相対的に高い増殖能を示すことから、継代操作を行うことによって、誘導線維芽細胞の純度が上がっていくことが確認された。なお、図8の誘導線維芽細胞におけるケラチノサイト類似のコロニーを選択的に抽出し、継代することでより短時間で純度の高い誘導線維芽細胞を得ることが可能である。
また、汎上皮細胞マーカーであるEpi-CAMに対する磁気標識抗体を用いて磁気細胞分離を行い、よりケラチノサイトに類似した列序性の増殖、形態のコロニーを呈することを確認された(図10上図)。図10上図は、図8の誘導線維芽細胞のコロニーを確認した後、第2継代に際して抗Epi-CAM抗体による標識を用いて細胞分離を行った細胞の図である。さらに、同時に導入したKRT14-RFPレポーター(pRZ-hKeratin (Cat# SR900CS-1, System Biosciences社))によってKRT14を発現していることも確認された(図10下図)。磁気標識抗Epi-CAM抗体による磁気細胞分離には、CD326 (EpCAM) マイクロビーズ, ヒト(ミルテニーバイオテク社)を標識抗体として、マニュアル細胞分離装置、MACS細胞分離カラム(ミルテニーバイオテク社)を用いて行った。
さらに、抗Epi-CAM抗体を用いて細胞分離後、培養を継続し継代、増幅した誘導線維芽細胞を無血清ケラチノサイト培地で単層培養条件に継代し、8日間培養を継続し、無血清ケラチノサイト培地に誘導線維芽細胞を慣らした後、Ca2+濃度を1.8mMに調整した無血清ケラチノ培地に培地を交換した。経時間的に、細胞が平坦化し、拡大する様子が観察された(図11)。図11において、左列がCa2+添加なし、右列がCa2+を添加ありの状態であり、また、18時間及び24時間経過時については、KRT14-RFPレポーターによる赤色蛍光状態も併せて示している。左右の図から、Ca2+の添加により誘導線維芽細胞の終分化がはじまり、細胞体が平坦に拡大していることが確認できる。さらに、赤色蛍光状態の図から、併行してKRT14の発現が減少していくのが観察された。これらはケラチノサイトにCa2+を加えた際に認められる終分化能と同一であり、単層培養条件下での誘導皮膚線維芽細胞がケラチノサイト同様の終分化能を有していることをあらわしている。
抗Epi-CAM抗体を用いて細胞分離後、培養を継続し継代、増幅した誘導線維芽細胞を用いて、皮膚線維芽細胞を含むコラーゲンゲル上で細胞を培養し、液体気体界面に細胞をさらすことによって、上皮組織形成能を調べる3次元皮膚培養モデルによる上皮組織形成能の試験を行い、誘導線維芽細胞が該条件下で重層角化上皮組織形成能を有することを確認した(図12)。
三次元皮膚培養モデルの作製は、参考文献5、6の方法を用いて行った。具体的にはコラーゲン酸性溶液I-PC 5mg/mL(KOKEN社)3mlにHepes ph7.4, HaHCO3それぞれが終濃度10mMになるように加えた後、106個の線維芽細胞を含む10%FBS加DMEM培地 4mlを加え、撹拌したのち6cmディッシュにうつし、30分間 37℃インキュベーター内に静置する。ゲル化が開始した時点で、ディッシュを叩きゲルをディッシュ表面から浮かせる。以降ゲルの収縮が開始するため、2日おきに培地交換を行い7-10日目に皮膚線維芽細胞との3次元皮膚培養を開始した。
斯くして得られた、皮膚線維芽細胞を含有するコラーゲンゲルを滅菌ナイロンメッシュを用いて、アルミニウムで作製した支持台の上にのせ、さらに図12上図に示すように、コラーゲンゲルの上に、内径10mm、外径12mmのガラス筒をのせる。これらの操作によりガラス筒内側の空間が、ガラス筒およびコラーゲンゲルによってガラス筒外側と隔てられることになる。ガラス筒外側を10%FBS加DMEM培地と無血清ケラチノサイト培地を1対1で混合した培地で満たし、ガラス筒内側に、6ウェルのプレート1ウェルにサブコンフルエントになった誘導線維芽細胞を200μlのSFKGMで播種する。以降、内側の培地は無血清ケラチノサイト培地で毎日培地交換を、外側の培地をCa2+濃度1.8mMに調整した10%FBS加DMEM培地と無血清ケラチノサイト培地の1対1混合培地で外培地は2日に1回の培地交換を行い、4日目から内側培地は空に、外側培地の液面をコラーゲンゲルの高さに合わせて誘導線維芽細胞を液体気体界面の環境におき、14日目まで培養を継続した。
図12下図は、3次元培養後の誘導線維芽細胞およびコラーゲンゲルの断面写真であるが、コラーゲンゲルの上に、誘導線維芽細胞から重層する上皮様組織が形成され、さらに上層では角化層も形成されたことが確認された。
上述の方法に従い作製した誘導線維芽細胞のゲノムDNAに対してウイルスベクター由来の遺伝子に特異的なプライマーを用いてPCR反応を行ったところ、誘導線維芽細胞には、OVOL1遺伝子、TFAP2A遺伝子、GRHL2遺伝子、BNC1遺伝子、LASS3遺伝子、ZBED2遺伝子、SOX7遺伝子、SOX9遺伝子、c-MYC遺伝子の9種の遺伝子が挿入されていることが確認された。ウイルスベクター由来の挿入遺伝子の確認には、表3に示す特異的なプライマーを用いたPCR反応を行った。PCR反応はAccuPrime Pfx Supermix(Life technologies社)を用いて、1反応あたり11.5μlのAccuPrime Pfx SuperMix試薬に200nMの終濃度になるようForward primerおよびReverse primerを加え、10ngのTemplate DNAを加えた。その後、サーマルサイクラ―にて95℃5分間のホットスタートの後、95℃15秒、63℃30秒、68℃60秒のサイクルを35サイクル繰り返したPCR産物を、E-Gel(登録商標) EX Gel, 1%(INVITROGEN社)、E-Gel(登録商標)アガロースゲル電気泳動システム(INVITROGEN社)を用いて電気泳動し、ターゲット遺伝子の存在を確認した。
実施例1から理解されるように、重層上皮組織形成能を有する細胞の誘導には、GATA3遺伝子、OVOL1遺伝子、OVOL2遺伝子、ESRP1遺伝子、TFAP2A遺伝子、ID1遺伝子、GRHL1遺伝子、GRHL2遺伝子、GRHL3遺伝子、TP63遺伝子、DNP63A遺伝子、MAPK13遺伝子、ARNTL2遺伝子、BNC1遺伝子、LASS3遺伝子、EHF遺伝子、ZNF165遺伝子、ZNF750遺伝子、ZBED2遺伝子、IRX2遺伝子、IRX4遺伝子、SOX7遺伝子、SOX9遺伝子、FOXQ1遺伝子、PPP1R13L遺伝子、KLF4遺伝子、c-MYC遺伝子という27種の導入遺伝子のすべてが必要なわけではなく、少なくともOVOL1遺伝子、TFAP2A遺伝子、GRHL2遺伝子、BNC1遺伝子、LASS3遺伝子、ZBED2遺伝子、SOX7遺伝子、SOX9遺伝子、c-MYC遺伝子という9種の遺伝子を導入することにより、体細胞から重層上皮組織形成能を有する細胞に直接転換できる可能が高い。なお、実施例1からは、かかる9種の遺伝子すべてを導入することで重層上皮組織形成能を有する細胞に直接転換できることを意味するものであって、9種の遺伝子すべてが直接転換に必要不可欠であることを意味するものではない。後記実施例2も鑑みれば、9種の遺伝子の一部を導入するだけでも重層上皮組織形成能を有する細胞に直接転換でき得る。
4.[実施例2] 皮膚線維芽細胞から重層上皮組織形成能を有する細胞への誘導
表2に記載の27種の遺伝子から、IRX2遺伝子及びIRX4遺伝子の2種を除いた残りの25種の遺伝子を導入遺伝子として皮膚線維芽細胞に導入した。基本的な実験条件、実験方法は、実施例1と同様である。実施例1と同様に、25種の導入遺伝子を皮膚線維芽細胞に導入し、培養した誘導線維芽細胞を得た。図13は、上述の方法に従い作製した誘導線維芽細胞のコロニーを示す図であり、左が高密度の領域、右が低密度の領域を示す。図13から、実施例2の誘導線維芽細胞のコロニーは、周囲のフィーダーおよび誘導されていない細胞と明瞭な境界をもっており、コロニーの辺縁には平坦な細胞が位置し、中央にむかって重積しており、図7のケラチノサイトのコロニー類似の形態を示している。
表2に記載の27種の遺伝子から、IRX2遺伝子及びIRX4遺伝子の2種を除いた残りの25種の遺伝子を導入遺伝子として皮膚線維芽細胞に導入した。基本的な実験条件、実験方法は、実施例1と同様である。実施例1と同様に、25種の導入遺伝子を皮膚線維芽細胞に導入し、培養した誘導線維芽細胞を得た。図13は、上述の方法に従い作製した誘導線維芽細胞のコロニーを示す図であり、左が高密度の領域、右が低密度の領域を示す。図13から、実施例2の誘導線維芽細胞のコロニーは、周囲のフィーダーおよび誘導されていない細胞と明瞭な境界をもっており、コロニーの辺縁には平坦な細胞が位置し、中央にむかって重積しており、図7のケラチノサイトのコロニー類似の形態を示している。
実施例2の誘導線維芽細胞に対し、ゲノムDNAに対してウイルスベクター由来の遺伝子に特異的なプライマー(表3)を用いてPCR反応を行ったところ、GATA3遺伝子、TFAP2A遺伝子、GRHL2遺伝子、TP63遺伝子、BNC1遺伝子、EHF遺伝子、ZNF165遺伝子、c-MYC遺伝子の8種の遺伝子が挿入されていることが確認された。かかる8種の遺伝子が挿入された細胞は、ケラチノサイト類似の増殖、形態を呈するが、実施例1のOVOL1遺伝子、TFAP2A遺伝子、GRHL2遺伝子、BNC1遺伝子、LASS3遺伝子、ZBED2遺伝子、SOX7遺伝子、SOX9遺伝子、c-MYC遺伝子の9遺伝子が挿入された細胞に比較すると増殖能が高く、終分化能が低い形態を呈していた。
実施例2から理解されるように、重層上皮組織形成能を有する細胞の誘導には、GATA3遺伝子、OVOL1遺伝子、OVOL2遺伝子、ESRP1遺伝子、TFAP2A遺伝子、ID1遺伝子、GRHL1遺伝子、GRHL2遺伝子、GRHL3遺伝子、TP63遺伝子、DNP63A遺伝子、MAPK13遺伝子、ARNTL2遺伝子、BNC1遺伝子、LASS3遺伝子、EHF遺伝子、ZNF165遺伝子、ZNF750遺伝子、ZBED2遺伝子、SOX7遺伝子、SOX9遺伝子、FOXQ1遺伝子、PPP1R13L遺伝子、KLF4遺伝子、c-MYC遺伝子という25種の導入遺伝子のすべてが必要なわけではなく、少なくともGATA3遺伝子、TFAP2A遺伝子、GRHL2遺伝子、TP63遺伝子、BNC1遺伝子、EHF遺伝子、ZNF165遺伝子、c-MYC遺伝子という8種の遺伝子を導入することにより、体細胞から重層上皮組織形成能を有する細胞に直接転換できる可能性が高い。なお、実施例2からは、かかる8種の遺伝子すべてを導入することで重層上皮組織形成能を有する細胞に直接転換できることを意味するものであって、8種の遺伝子すべてが直接転換に必要不可欠であることを意味するものではない。
実施例1及び2によれば、最終的に体細胞から誘導された重層上皮組織形成能を有する細胞には、実施例1及び2のいずれにおいても、TFAP2A遺伝子、GRHL2遺伝子、BNC1遺伝子、c-MYC遺伝子の4遺伝子が共通して確認された。よって、TFAP2A遺伝子、GRHL2遺伝子、BNC1遺伝子、c-MYC遺伝子を導入することにより、体細胞から重層上皮組織形成能を有する細胞に直接転換できる可能が高い。また、TFAP2A遺伝子の代わりに、又はTFAP2A遺伝子と組み合わせて、同様の機能を有するTFAP2C遺伝子を導入してもよいし、GRHL2遺伝子の代わりに、又は組み合わせて、GRHLファミリー遺伝子を導入してもよいし、c-MYC遺伝子の代わりに、又は組み合わせて、MYCファミリー遺伝子を導入してもよい。さらにかかる4種の遺伝子に加えて、重層上皮組織形成能を有する細胞に相対的に強発現する少なくとも1種の遺伝子も導入してもよい。実施例1の誘導線維芽細胞と実施例2の誘導線維芽細胞とで、増殖能と、終分化能に違いが確認されているように、同時に導入する遺伝子の組み合わせによって、作製される細胞の特性、能力、例えば増殖能、終分化能などの調節を行うことが可能であり、使用目的によって導入遺伝子を加減することの有用性が確認された。

Claims (27)
- 重層上皮組織形成能を有する細胞に相対的に強発現する少なくとも1種の遺伝子を体細胞に導入する工程を含む、重層上皮組織形成能を有する細胞の製造方法。
- GATA3遺伝子、OVOL1遺伝子、OVOL2遺伝子、ESRP1遺伝子、TFAP2A遺伝子、ID1遺伝子、GRHL1遺伝子、GRHL2遺伝子、GRHL3遺伝子、TP63遺伝子、DNP63A遺伝子、MAPK13遺伝子、ARNTL2遺伝子、BNC1遺伝子、LASS3遺伝子、EHF遺伝子、ZNF165遺伝子、ZNF750遺伝子、ZBED2遺伝子、IRX2遺伝子、IRX4遺伝子、SOX7遺伝子、SOX9遺伝子、FOXQ1遺伝子、PPP1R13L遺伝子、KLF4遺伝子及びc-MYC遺伝子から選択された一つ以上の遺伝子を体細胞に遺伝子導入する工程を含む、重層上皮組織形成能を有する細胞の製造方法。
- (1)TFAP2A遺伝子またはTFAP2C遺伝子、
(2)GRHLファミリー遺伝子から選択された少なくとも一種の遺伝子、
(3)BNC1遺伝子及び
(4)MYCファミリー遺伝子から選択された少なくとも一種の遺伝子
を体細胞に遺伝子導入する工程を含む、重層上皮組織形成能を有する細胞の製造方法。 - (1)TFAP2A遺伝子またはTFAP2C遺伝子、
(2)GRHLファミリー遺伝子から選択された少なくとも一種の遺伝子、
(3)BNC1遺伝子、
(4)MYCファミリー遺伝子から選択された少なくとも一種の遺伝子及び
(5)重層上皮組織形成能を有する細胞に相対的に強発現する少なくとも1種の遺伝子
を体細胞に遺伝子導入する工程を含む、重層上皮組織形成能を有する細胞の製造方法。 - (1)TFAP2A遺伝子、(2)GRHL2遺伝子、(3)BNC1遺伝子、(4)c-MYC遺伝子を体細胞に遺伝子導入する工程を含む、重層上皮組織形成能を有する細胞の製造方法。
- GATA3遺伝子、TFAP2A遺伝子、GRHL2遺伝子、TP63遺伝子、BNC1遺伝子、EHF遺伝子、ZNF165遺伝子及びc-MYC遺伝子を体細胞に遺伝子導入する工程を含む、重層上皮組織形成能を有する細胞の製造方法。
- OVOL1遺伝子、TFAP2A遺伝子、GRHL2遺伝子、BNC1遺伝子、LASS3遺伝子、ZBED2遺伝子、SOX7遺伝子、SOX9遺伝子及びc-MYC遺伝子を体細胞に遺伝子導入する工程を含む、重層上皮組織形成能を有する細胞の製造方法。

- 前記体細胞が皮膚線維芽細胞である、請求項1乃至9の何れか1項に記載の重層上皮組織形成能を有する細胞の製造方法。
- 前記体細胞が脂肪組織由来間質細胞である、請求項1乃至9の何れか1項に記載の重層上皮組織形成能を有する細胞の製造方法。
- 前記体細胞が末梢循環血液中の単核球である、請求項1乃至9の何れか1項に記載の重層上皮組織形成能を有する細胞の製造方法。
- 請求項1乃至12の何れか1項に記載の製造方法により製造される重層上皮組織形成能を有する細胞。
- 請求項13に記載の重層上皮組織形成能を有する細胞を含む、細胞製剤。
- 足場材料を含む請求項14に記載の細胞製剤。
- 前記足場材料がコラーゲンである、請求項15に記載の細胞製剤。
- 前記細胞製剤は、重層上皮再生用、皮膚組織再生用、粘膜組織再生用又は角膜組織再生用である請求項14乃至16の何れか1項に記載の細胞製剤。
- 前記細胞製剤がシート状の上皮細胞シートである請求項14乃至17の何れか1項に記載の細胞製剤。
- 請求項13に記載の重層上皮組織形成能を有する細胞を非ヒト哺乳動物に投与して、前記哺乳動物の体内で前記重層上皮組織形成能を有する細胞から重層上皮組織を形成させることにより製造される、重層上皮組織を形成させた非ヒト哺乳動物。
- 請求項19に記載の非ヒト哺乳動物に被験物質を投与し、上皮組織に対する被験物質の薬効を判定する工程を含む、上皮組織に対する被験物質の薬効を判定する方法。
- 請求項19に記載の非ヒト哺乳動物に抗がん剤や放射線などのストレスを負荷し、上皮組織に対するストレスを判定する工程を含む、上皮組織に対する外的要因の影響を判定する方法。
- 請求項1乃至8の何れか1項に記載の体細胞に遺伝子導入される遺伝子を含む、重層上皮組織形成能を有する細胞調整用組成物。
- 前記遺伝子が、体細胞に導入可能な形態で含まれる、請求項22に記載の細胞調整用組成物。
- 請求項13に記載の重層上皮組織形成能を有する細胞から分化した上皮細胞組織。
- 動物から取り出された体細胞に、請求項1乃至項8の何れか1項に記載の遺伝子を遺伝子導入して製造された重層上皮組織形成能を有する細胞から分化した上皮細胞組織に対し、被験物質を投与し、上皮組織に対する被験物質の薬効を判定する工程を含む、上皮組織に対する被験物質の薬効の分析方法。
- 動物から取り出された体細胞に、請求項1乃至項8の何れか1項に記載の遺伝子を遺伝子導入して製造された重層上皮組織形成能を有する細胞から分化した上皮細胞組織に対し、抗がん剤や放射線などのストレスを負荷し、上皮組織に対するストレスを判定する工程を含む、上皮組織に対する外的要因の影響の分析方法。
- 請求項1乃至8の何れか1項に記載の遺伝子を体細胞に遺伝子導入するためのキット。
