이에, 상기 콜린성 신경세포를 다양한 퇴행성 뇌신경계 질환의 치료제로서 사용하는 방법을 개발하려는 연구가 활발히 진행되고 있을 뿐만 아니라, 상기 치료에 사용되는 콜린성 신경세포를 줄기세포로부터 분화시킬 수 있는 방법을 개발하려는 연구 역시 활발하게 진행되고 있다. 지금까지 보고된 바에 의하면, 배아줄기세포에 bFGF, 레티논산(retinoic acid, RA) 및 SHH를 처리하여 콜린성 신경세포로 분화시킬 수 있다. 구체적으로, 배아줄기세포는 초기 신경외배엽 세포를 형성하고, 여기에 RA를 처리하면 효율적으로 배면화(posteriorize)되고, 여기에 SHH를 처리하여 콜린성 신경세포로 분화된다. 이외에도 쥐의 배아줄기세포를 쥐의 골수 기원 스트로멀 휘더 셀(stromal feeder cells)과 함께 공배양함으로써 콜린성 신경세포로 분화시킬 수 있음이 알려져 있고, 신경전구세포(neural progenitor cell)를 SHH 및 RA를 포함하는 배지에서 배양한 다음, 다시 bFGF 및 SHH를 포함하는 배지에서 배양하고, 이로부터 얻어진 배양물을 아스코빈산(ascorbic acid, AA) 및 BDNF를 포함하는 배지에서 배양하여 신경전구세포를 콜린성 신경세포로 분화시키는 방법(한국등록특허 제10-0683199호)이 알려져 있다.
그러나, 상기 기존의 방법에 의해 분화된 콜린성 신경세포에는 순수한 콜린성 신경세포 이외에도 다른 세포들이 함께 포함되어 있어, 신경퇴행성 질환의 치료에 직접적으로 사용할 수 없다는 문제점이 있으며, 아직까지는 이러한 문제점을 해결되지 않고 있는 실정이다.
본 발명자들은 줄기세포를 분화시켜서 순도가 높은 콜린성 신경세포를 생산할 수 있는 방법을 개발하기 위하여 예의 연구노력한 결과, 줄기세포를 직접적으로 콜린성 신경세포로 분화시키기 보다는 줄기세포로부터 신경전구세포를 형성한 다음, 상기 신경전구세포를 분화시키면서 콜린성 신경세포 외의 세포의 증식을 억제할 경우, 콜린성 신경세포를 높은 순도로 생산할 수 있음을 확인함으로써, 본 발명을 완성하였다.
본 발명의 하나의 목적은 줄기세포로부터 신경전구세포를 형성한 다음, 상기 신경전구세포를 콜린성 신경세포로 분화시키는 단계를 포함하는, 콜린성 신경세포의 생산방법을 제공하는 것이다.
본 발명의 다른 목적은 상기 방법으로 생산된 콜린성 신경세포를 제공하는 것이다.
본 발명에서 제공하는 콜린성 신경세포의 생산방법을 이용하면, 높은 순도로 콜린성 신경세포를 생산할 수 있고, 동일한 형질의 콜린성 신경세포를 보다 빠르게 생산할 수 있을 뿐만 아니라 신경전구세포를 사용하여 필요할 때에 신속하게 콜린성 신경세포를 분화시킬 수 있으므로, 알츠하이머 병과 같은 퇴행성 뇌신경계 질환의 효과적인 치료에 널리 활용될 수 있을 것이다.

본 발명자들은 줄기세포를 분화시켜서 순도가 높은 콜린성 신경세포를 생산할 수 있는 방법을 개발하기 위하여 다양한 연구를 수행하던 중, 신경전구세포를 경유하는 방법에 주목하게 되었다. 상기 신경전구세포는 다른 신경세포와는 달리 동결보존이 가능할 뿐만 아니라 분화된 세포의 순도를 조절하기 어려운 줄기세포와는 달리, 높은 순도의 콜린성 신경세포로 분화시키는 것이 가능함을 확인하였다. 이같은 방법을 사용하여 콜린성 신경세포를 생산하면, 줄기세포로부터 콜린성 신경세포를 분화시키는 종래의 방법과는 달리, 신경전구세포로부터 콜린성 신경세포를 분화시킬 수 있으므로, 콜린성 신경세포를 보다 신속하게 수득할 수 있다는 장점이 있다. 뿐만 아니라, 상기 신경전구세포를 생성하는 중간과정(수득한 배아유사세포 덩어리로부터 신경구세포를 수득하는 과정)에서 신경세포 분화유도물질로 알려진 소닉 헤지호그(sonic hedgehog, SHH)를 처리하면, 상기 신경전구세포로부터 분화된 콜린성 신경세포 외의 다른 분화부산물 세포의 비율이 급격하게 감소하므로, 종래의 방법에 비하여 콜린성 신경세포로 분화되는 효율이 증가하는 효과를 나타낼 수 있음을 확인하였는데, 이러한 효과는 공지된 방법을 사용하는 종래기술에서는 전혀 보고되지 않았고, 본 발명자에 의하여 최초로 확인되었다.
본 발명의 하나의 양태는 줄기세포로부터 신경전구세포를 형성한 다음, 상기 신경전구세포를 콜린성 신경세포로 분화시키는 단계를 포함하는 콜린성 신경세포의 생산방법을 제공한다.
본 발명의 용어 "줄기세포(stem cell)"란, 자기 복제 능력을 가지면서 두 개 이상의 새로운 세포로 분화하는 능력을 갖는 세포를 의미하며, 분화능에 따라 만능 줄기세포(totipotent stem cell), 전분화능 줄기세포(pluripotent stem cells), 다분화능 줄기세포(multpotent stem cells)로 분류할 수 있고, 줄기세포의 유래조직에 따라 중간엽 줄기세포, 배아줄기세포 및 역분화줄기세포로 분류할 수 있다.
본 발명에 있어서, 상기 줄기세포는 본 발명에서 제공하는 콜린성 신경세포로 분화될 수 있는 줄기세포로 해석될 수 있는데, 상기 줄기세포는 배아유사세포 덩어리, 로제트(Rosette), 신경구세포 및 신경전구세포를 거쳐 콜린성 신경세포로 분화될 수 있다. 따라서, 본 발명에서는, 상기 콜린성 신경세포로 분화가능하다면 중간엽 줄기세포, 배아줄기세포, 역분화줄기세포 등의 모든 종류의 줄기세포를 사용할 수 있는데, 구체적으로는 배아줄기세포, 역분화 줄기세포 또는 골수, 지방조직, 치아, 치아조직, 혈액, 제대혈, 간장, 피부, 위장관, 태반, 자궁, 태아로부터 유래한 중간엽 줄기세포가 될 수 있다.
본 발명의 구체적인 일 실시예에서는 콜린성 신경세포를 생산하기 위한 줄기세포로서 역분화줄기세포를 이용하였는데, 상기 역분화줄기세포는 피부 섬유아세포에 hOct3/4-p53sh, hSox-KLF4, hUl 및 EBNA1 유전자를 도입하여 수득하였다.
본 발명의 용어 "배아유사세포 덩어리(embryoid body)"는, 배상체라고도 하며, 세포분열 초기에 줄기세포들이 공모양으로 뭉쳐 형성된 세포덩어리를 의미한다. 상기 배아유사세포 덩어리는 통상적인 배아줄기세포와 유사한 수준의 전분화능을 나타내어, 골세포, 근육세포, 신경세포, 상피세포, 섬유세포 등의 다양한 생체조직으로 분화될 수 있다.
본 발명에 있어서, 상기 배아유사세포 덩어리는 줄기세포를 신경전구세포로 분화시키기 위한 중간매개체로서 사용될 수 있으며, 이는 줄기세포를 다양한 성분(예를 들어, 혈청, 비필수아미노산(NEAA), 항생제, LDN193189, SB431542 등)이 포함된 통상적인 줄기세포 배양용 배지(예를 들어, DMEM/F12 배지, KO-DMEM/F12 배지 등)에서 배양함으로써 형성될 수 있다. 상기 줄기세포로부터 배아유사세포 덩어리를 형성하기 위하여 배양하는 시간은 특별히 이에 제한되지 않으나, 구체적으로는 1 내지 10일이 될 수 있고, 보다 구체적으로는 2 내지 7일이 될 수 있으며, 가장 구체적으로는 4 내지 5일이 될 수 있다.
본 발명의 구체적인 일 실시예에서는 상기 수득한 역분화줄기세포를 4 내지 5일 동안 배양하여 배아유사세포 덩어리를 수득하였다.
본 발명의 용어 "신경전구세포(neural progenitor cell)"란, "뉴런전구세포(neuron precursor cell)"라고도 하는데, 넓은 범위에서는 신경세포로 분화할 수 있거나 또는 분화과정중에 존재하는 모든 세포를 의미한다. 상기 신경전구세포는 신경줄기세포 또는 그외의 다른 줄기세포가 분열되어 신경아세포(neuroblast)를 형성하고, 상기 형성된 신경아세포는 신경관 또는 신경세포가 형성될 부위로 이동한 다음, 이동한 부위에서 축삭과 수상돌기를 형성하도록 형태적 및 기능적으로 분화되어, 최종적으로 신경세포를 형성하는데, 상기 줄기세포로부터 분화가 완료되기 직전까지의 모든 세포가 넓은 범위의 신경전구세포에 해당하며, 보다 좁은 범위로는 분열이 종료된 신경아세포가 신경전구세포에 해당한다고 볼 수 있다.
in vitro에서 배아유사세포 덩어리로부터 상기 신경전구세포를 수득하기 위하여는, 통상적인 줄기세포배양용 배지에 소닉 헤지호그(sonic hedgehog, SHH) 등의 신경세포분화유도 성분을 가하여 제조한 신경분화유도용 배지(로제트 형성용 배지)에서 배아유사세포 덩어리를 배양하여 로제트(Rosette)를 수득한 다음, 상기 로제트를 회수하고 이를 동일한 신경분화유도용 배지에서 배양하여 신경구세포(neurosphere)를 수득하고, 상기 신경구세포를 신경전구세포 배양용 배지에서 배양함으로써, 신경전구세포를 수득할 수 있다.
또한, 본 발명에서 상기 신경분화유도용 배지는 배아유사세포 덩어리로부터 로제트를 형성할 수 있는 배지를 의미하는 것이며, 상기 신경분화유도용 배지는 배아유사세포 덩어리를 배양하여 로제트를 수득할 수 있는한 제한이 없으나, 구체적으로, N2, B27, bFGF(basic fibroblast growth factor) 및 SHH를 포함하는 배지, 더욱 구체적으로, N2, B27, bFGF 및 SHH를 포함하는 DMEM/F12 배지 또는 KO-DMEM/F12 배지일 수 있으나, 이에 제한되지 않는다.
또한, 본 발명에서 상기 "신경분화유도용 배지"는 "로제트 형성용 배지"와 혼용된다.
또한, 본 발명에서 상기 신경전구세포 배양용 배지는 신경구세포로부터 신경전구세포를 형성할 수 있는 배지를 의미하는 것이며, 신경구세포를 배양하여 신경전구세포를 수득할 수 있는한 제한이 없으나, 구체적으로 무혈청 배지일 수 있다.
이때, 상기 배아유사세포 덩어리부터 로제트를 형성하기 위하여 배양하는 시간은 특별히 이에 제한되지 않으나, 구체적으로는 4 내지 10일이 될 수 있고, 보다 구체적으로는 5 내지 9일이 될 수 있으며, 가장 구체적으로는 7일이 될 수 있다.
또한, 상기 로제트로부터 신경구세포를 형성하기 위하여 배양하는 시간은 특별히 이에 제한되지 않으나, 구체적으로는 4 내지 10일이 될 수 있고, 보다 구체적으로는 5 내지 9일이 될 수 있으며, 가장 구체적으로는 7일이 될 수 있다.
따라서, SHH 등의 신경세포분화유도 성분을 포함하는 신경분화유도 배지를 사용하여 상기 배아유사세포 덩어리로부터 신경구세포를 형성하기 위하여 배양하는 시간은 특별히 이에 제한되지 않으나, 구체적으로는 8 내지 20일이 될 수 있고, 보다 구체적으로는 10 내지 18일이 될 수 있으며, 가장 구체적으로는 14일이 될 수 있다.
배아유사세포 덩어리를 N2, B27, bFGF(basic fibroblast growth factor) 및 SHH를 포함하는 로제트 형성용 배지에서 배양하여 로제트를 수득한 다음, 상기 로제트를 배양하여 신경구세포를 수득하는 것인 방법.
또한, 상기 신경구세포로부터 신경전구세포를 형성하기 위하여 배양하는 시간은 특별히 이에 제한되지 않으나, 구체적으로는 1 내지 5일이 될 수 있고, 보다 구체적으로는 1 내지 3일이 될 수 있으며, 가장 구체적으로는 2일이 될 수 있다.
본 발명의 용어 "로제트(Rosette)"란, 배아유사세포 덩어리를 신경세포분화유도 성분을 포함하는 신경분화유도 배지에서 배양하여 생성되는 세포집합체를 의미하는데, 대체로 꽃모양의 형태로 세포가 결합된 형태를 나타낸다. 상기 로제트는 줄기세포에서 신경세포로 분화되는데 필요한 유전자가 발현되어 형태적인 변화가 함께 수반되는 중간단계의 세포라도 이해될 수 있는데, 상기 로제트로부터 보다 효과적으로 신경구세포를 형성하기 위하여는, 이들 로제트의 집적이 필요하므로, 상기 로제트의 형성이 확인되면, 로제트를 수집하여 높은 밀도를 갖도록 배양용기에 접종하고, 이를 배양하는 방법을 사용함이 바람직하다.
본 발명의 용어 "콜린성 신경세포"란, 화학전달물질로서 아세틸콜린을 분비하는 뉴런을 의미하며, 기저전뇌에 위치한 콜린성 신경세포를 BFCN(Basal forebrain cholinergic neurons)이라고 한다. 상기 콜린성 신경세포는 동물의 뇌신경계 질환의 유발에 직접적으로 관여하는 것으로 알려져 있는데, 알츠하이머 병, 파킨슨 병, 근위축성 측색경화증 등의 다양한 퇴행성 뇌질환의 발병시에 콜린성 신경세포의 손상이 수반된다고 알려져 있다. 특히, 알츠하이머 병의 주요 증상 중의 하나인 학습능력 장애는 콜린성 신경세포의 손상에 의한 것으로 예측되고 있다.
본 발명에 있어서, 상기 콜린성 신경세포는 신경전구세포를 콜린성 신경세포 분화용 배지에서 배양하여 생산할 수 있는데, 상기 콜린성 신경세포 분화용 배지는
신경전구세포를 콜린성 신경세포 분화시킬 수 있는 한 성분에 제한이 없으나, 구체적으로 Glutamax, B27, EGF, 헤파린, 아스코빈산, BDNF(brain-derived neurotrophic factor) 및 이들의 조합으로 구성된 군으로부터 선택되는 물질을 포함할 수 있다.
이때, 상기 신경전구세포로부터 콜린성 신경세포를 분화시키는데 소요되는 시간은 특별히 이에 제한되지 않으나, 구체적으로는 1 내지 10일이 될 수 있고, 보다 구체적으로는 3 내지 7일이 될 수 있으며, 가장 구체적으로는 5일이 될 수 있다.
아울러, 상기 콜린성 신경세포가 형성되면, 신경전구세포로부터 콜린성 신경세포 이외의 다른 종류의 신경세포가 생성될 수 있는데, 이러한 다른 종류의 신경세포의 생성을 억제하기 위하여, 분화억제물질(예를 들어, AraC 등)을 배지에 첨가하여, 콜린성 신경세포 이외의 다른 종류의 신경세포로의 추가적인 분화 및 증식을 억제할 수 있다. 이때, 상기 분화억제물질을 처리하는 시간은 특별히 이에 제한되지 않으나, 구체적으로는 1 내지 5일이 될 수 있고, 보다 구체적으로는 1 내지 3일이 될 수 있으며, 가장 구체적으로는 2일이 될 수 있다. 상기 AraC는 시타라빈(cytarabine)를 의미한다.
또한, 이미 생성된 콜린성 신경세포의 증식을 촉진시키기 위하여, 다양한 성장인자(예를 들어, bFGF 등)을 배지에 첨가하여 배양할 수 있는데, 이때 상기 성장인자를 배지에 첨가하여 배양하는 기간은 특별히 이에 제한되지 않으나, 구체적으로는 10 내지 30일이 될 수 있고, 보다 구체적으로는 15 내지 25일이 될 수 있으며, 가장 구체적으로는 20일이 될 수 있다.
따라서, 신경전구세포를 콜린성 신경세포 분화용 배지에 접종하고 배양하기 시작한 시점을 기준으로 구체적으로는 4 내지 15일, 보다 구체적으로는 6 내지 10일, 가장 구체적으로는 7일이 경과된 후에, 상기 성장인자를 배지에 첨가하여 배양함으로써, 콜린성 신경세포의 증식을 촉진시킬 수 있다.
본 발명의 방법을 이용하면, 높은 순도의 콜린성 신경세포를 생산할 수 있을 뿐만 아니라, 동결보존가능한 신경전구세포를 이용하여 동일한 형질의 콜린성 신경세포를 지속적으로 생산할 수 있으며, 신경전구세포로부터 콜린성 신경세포를 분화생산하는 과정만을 반복수행함으로써 콜린성 신경세포를 생산하는데 소요되는 시간을 단축시킬 수 있다는 장점이 있다. 본 발명에서 생산하는 콜린성 신경세포를 이용하여 알츠하이머 병을 치료할 경우에는, 환자의 생체내 조건에 부합되는 종류의 콜린성 신경세포를 사용하여야 하는데, 상기 알츠하이머 병을 치료하기 위하여는 콜린성 신경세포를 환자의 증세에 따라 상기 콜린성 신경세포를 일정 주기로 지속적으로 이식하는 과정이 요구될 수 있다. 이 경우, 최초 사용된 콜린성 신경세포와 이후에 사용된 콜린성 신경세포가 서로 다른 형질을 나타낼 경우에는, 상기 알츠하이머 병을 치료가능성이 급격히 저하될 수 있다는 문제점이 있는데, 상기 알츠하이머 병은 장기간의 치료시간이 필요하므로, 최초 사용된 콜린성 신경세포를 치료가 끝날때까지 유지시키는 것은 실질적으로 불가능한 것으로 알려져 있다. 그러나, 본 발명에서 제공하는 방법을 사용하면 콜린성 신경세포로 분화될 수 있는 전구세포를 보존하고 이를 사용하여 동일한 형질의 콜린성 신경세포를 알츠하이머 병이 발병된 환자의 치료가 완료될 때까지 지속적으로 생산할 수 있으므로, 알츠하이머 병의 치료성공율을 현저히 향상시킬 수 있다.
아울러, 동결보존가능한 신경전구세포로부터 콜린성 신경세포를 분화생산하는 과정만을 반복수행함으로써, 줄기세포로부터 콜린성 신경세포를 생산하는 종래기술에 비하여, 콜린성 신경세포를 생산하는데 소요되는 시간을 단축시킬 수 있다는 장점이 있다.
본 발명에서 제공하는 콜린성 신경세포의 생산방법의 구체적인 예로서, 본 발명의 콜린성 신경세포의 생산방법은,
(a) 줄기세포를 배양하여 배아유사세포 덩어리를 수득하는 단계;
(b) 상기 수득한 배아유사세포 덩어리를 SHH를 포함하는 배지에서 배양하여 신경구세포를 수득하는 단계;
(c) 상기 수득한 신경구세포를 배양하여 신경전구세포를 수득하는 단계; 및
(d) 상기 수득한 신경전구세포를 콜린성 신경세포 분화용 배지에서 배양하여 콜린성 신경세포를 생성하는 단계를 포함할 수 있으나, 이에 제한되지 않는다.
또한, 본 발명에서 구체적으로 상기 (b) 단계는 배아유사세포 덩어리를 SHH를 포함하는 배지에서 배양하여 로제트를 수득한 다음, 상기 로제트를 SHH를 포함하는 배지에서 배양하여 신경구세포를 수득하는 단계를 포함할 수 있으나, 이에 제한되지 않는다.
또한, 본 발명에서 구체적으로 상기 (d) 단계는,
(d1) 신경전구세포를 콜린성 신경세포 분화용 배지에서 배양하여 콜린성 신경세포를 생성하는 단계;
(d2) 상기 배지에 분화억제 물질을 처리하여, 콜린성 신경세포 이외의 다른 종류의 신경세포의 증식을 억제하는 단계; 및
(d3) 상기 배지에 성장인자를 처리하여, 콜린성 신경세포를 증식하는 단계를 포함할 수 있으나, 이에 제한되지 않는다.
상술한 바와 같이, (a) 단계의 줄기세포는 중간엽 줄기세포, 배아줄기세포 또는 역분화줄기세포가 될 수 있고; 상기 역분화줄기세포는 피부 섬유아세포에 hOct3/4-p53sh, hSox-KLF4, hUl 및 EBNA1 유전자를 도입하여 수득한 것이 될 수 있으며; (a) 단계의 줄기세포는 혈청, 비필수아미노산(NEAA), 항생제, LDN193189 및 SB431542를 포함하는 DMEM/F12 배지에서 배양하여 배아유사세포 덩어리를 형성할 수 있고; (b) 단계에서, 배아유사세포 덩어리에 SHH를 처리하면서 배양하여 로제트(Rosette)를 수득하고, 상기 수득한 로제트에 SHH를 처리하면서 배양하여 신경구세포를 수득할 수 있으며; (c) 단계에사 수득한 신경전구세포는 동결보존이 가능하여, 동일한 형질의 콜린성 신경세포를 반복재현하는데 사용될 수 있고; (d) 단계의 콜린성 신경세포 분화용 배지는 Glutamax, 항생제, B27, EGF 및 헤파린을 포함하는 KO-DMEM/F12 배지를 사용할 수 있고; (d) 단계의 콜린성 신경세포를 생성시킨 후에, AraC를 처리하여 콜린성 신경세포 이외의 세포증식을 억제하고, bFGF를 처리하면서 배양하여 생성된 콜린성 신경세포를 증식시키는 단계를 추가로 포함할 수 있다.
본 발명의 구체적인 일 실시예에 의하면, 피부 섬유아세포를 사용하여 역분화줄기세포를 수득하고, 상기 수득한 역분화줄기세포를 배양하여 배아유사세포 덩어리를 수득하였으며, 상기 수득한 배아유사세포 덩어리를 배양하여 로제트를 형성하고, 상기 형성된 로제트를 회수한 다음, 다시 배양하여 신경구세포를 수득하였다(도 2 및 3). 상기 수득한 신경구세포를 콜린성 신경세포 분화용 배지에서 배양하여 콜린성 신경세포를 생산하였다(도 4 및 5).
또한, 본 발명의 구체적인 일 실시예에서 상기 분화된 신경세포의 약 99.4 또는 100%가 콜린성 신경세포임을 확인하여, 동일한 형질의 콜린성 신경세포를 높은 순도로 생산할 수 있음을 알 수 있었다(도 6).
상술한 목적을 달성하기 위한 본 발명의 다른 하나의 양태는 상술한 콜린성 신경세포의 생산방법으로 제조된 콜린성 신경세포를 제공한다.
본 발명에서 제공하는 콜린성 신경세포는 종래의 방법으로 생산된 콜린성 신경세포에 비하여, 콜린성 신경세포를 제외한 다른 세포의 함량이 현저한 수준으로 낮아. 콜린성 신경세포의 순도가 현저하게 증가된 것이므로, 알츠하이머 병과 같은 콜린성 신경세포의 이식이 필요한 환자의 치료효율을 향상시킬 수 있다.
이하 본 발명을 실시예를 통하여 보다 상세하게 설명한다. 그러나 이들 실시예는 본 발명을 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.
실시예 1: 역분화줄기세포(iPSC)의 수득
정상인과 알츠하이머 환자로부터 수득한 각각의 피부 섬유아세포(skin fibroblast, AG06263, AG06264, AG07871, AG07926, coreill)를 배양하고, 배양된 세포에 neon™ transfection system(MPK10096, Invitrogen)을 이용하여 hOct3/4-p53sh, hSox-KLF4, hUl 및 EBNA1 유전자를 도입하여 형질전환체를 수득하였다. 상기 수득한 형질전환체를 80% 수준으로 포화배양한 다음, 마트리젤(matrigel,47743-720,corning)이 코팅된 배양용기에 접종하여 계대배양하여 역분화줄기세포를 수득하였다. 이때, 사용된 배지는 50ng/㎖ bFGF, 3uM CCHIR99021(13122-1,cayman), 0.5uM A83-01(sc-203791, santacruz), 소듐부티레이트 0.2mM 및 니코틴아미드 1mM을 포함하는 mTeSR™ 1 배지(05850, stem cell™ technologies)를 사용하였고, 계대 배양시 포화배양된 세포를 절단한 후, 콜라겐 분해효소(collagenase Ⅳ)를 처리하여 세포를 분리시켜서 수행하였다.
실시예 2: 콜린성 신경세포의 생산
상기 실시예 1에서 수득한 각각의 역분화줄기세포를 분화시켜서 콜린성 신경세포를 생산하였다.
실시예 2-1: 배아유사세포 덩어리(embryoid body)의 수득
상기 역분화줄기세포를 약 150um의 크기로 절단한 다음, 콜라겐 분해효소(collagenase Ⅳ)를 처리하여 세포를 분리시키고, 분리된 세포를 DMEM/F12 배지(11330, gibco)로 세척하였다. 세척된 역분화줄기세포를 배아유사세포 배양용 배지(80% DMEM/F12, 20% Serum replacement, 1% NEAA(11140-050, Gibco), 1% 페니실린/스트렙토마이신, 100nM LDN193189(S2618,selleckchem) 및 10uM SB431542(301836-41-9, ABcam))에 접종하고, 4-5일간 배양하여, 배아유사세포 덩어리를 수득하였다.
실시예 2-2: 로제트(Rosette) 및 신경구세포(neurosphere)의 수득
상기 실시예 2-1에서 수득한 배아유사세포 덩어리가 형성된 배양액을 정치시켜서 배아유사세포 덩어리를 침전시키고, 배양용 배지를 제거하여, 배아유사세포 덩어리를 회수하였다. 상기 회수된 배아유사세포 덩어리를 로제트 형성용배지(DMEM/F12, 1% N2(gib-17502-048, Gibco), 1% B27(17504044,Gibco), 20nM bFGF, 50ng/㎖ SHH(1845-sh-100/cf, R&D))와 함께 마트리겔이 코팅된 배양용기에 넣고, 7일 동안 배양하여 로제트를 수득하였다.
상기 수득한 로제트를 수집하고, 이를 로제트 형성용배지에 접종한 다음, 7일 동안 배양하여 신경구세포(neurosphere)를 수득하였다.
실시예 2-3: 신경전구세포(neuronal progenitor)의 수득
상기 실시예 2-2에서 수득한 신경구세포를 PBS로 세척하고, 아큐타제(accutase)를 처리하여 단일세포로 분리한 다음, FBS가 포함된 배지로 세척하였다. 세척된 단일세포를 stemdiff™ Neural Progenitor 배지(05833,stem cell™ technologies)와 함께 마트리젤이 코팅된 배양용기에서 2일 동안 배양하여 신경전구세포를 수득하였다. 수득한 신경전구세포에 트립신을 처리하여 신경전구세포를 단일세포로 분리한 다음, stemdiff™ Neural Progenitor 배지와 함께 마트리젤이 코팅된 배양용기에서 계대배양하였다(도 2).
도 2는 정상인과 알츠하이머 환자로부터 수득한 피부 섬유아세포로부터 유래된 배아유사세포 덩어리, 로제트, 신경구세포 및 신경전구세포의 형태를 나타내는 현미경사진으로서, 상단은 정상인으로부터 수득한 피부 섬유아세포(UND1-1)로부터 유래된 배아유사세포 덩어리, 로제트, 신경구세포 및 신경전구세포를 나타내고, 하단은 알츠하이머 환자로부터 수득한 피부 섬유아세포(SAD2-2)로부터 유래된 배아유사세포 덩어리, 로제트, 신경구세포 및 신경전구세포를 나타낸다. 도 2에서 보듯이, 정상인과 알츠하이머 환자의 피부 섬유아세포로부터 유래된 배아유사세포 덩어리, 로제트, 신경구세포 및 신경전구세포는 형태적으로 동일함을 확인하였다.
실시예 2-4: 콜린성 신경세포의 생산
상기 실시예 2-3에서 계대배양된 신경전구세포를, 콜린성 신경세포 분화용 배지(2mM Glutamax(35050-061, gibco), 1% 페니실린/스트렙토마이신, 1% B27, 20ng/㎖ EGF(GF144, milipore) 및 5㎍/㎖ 헤파린(h3393, SIGMA)을 포함하는 7:3비율의 KO-DMEM/F12 배지)와 함께, poly-D-리신/라미닌(#40210, #40232, Collaborative Research)이 코팅된 배양용기에서 5일 동안 배양하고, 2일 동안 배지에 2.4μM AraC(C1768, sigma)를 처리한 다음, 23일 동안 배지에 10ng/㎖ bFGF를 처리하여 배양함으로써, 신경전구세포를 콜린성 신경세포로 분화시켜서, 콜린성 신경세포를 생산하였다.
실시예 3: 콜린성 신경세포의 분화결과 검정
실시예 3-1: 줄기세포와 신경전구세포에서의 분화마커의 발현수준
상기 실시예 2의 방법으로 콜린성 신경세포를 분화시키는 과정에서, 역분화줄기세포와 신경전구세포를 각각 수득하고, 이들 각 세포에서 발현되는 신경전구세포 마커 단백질의 발현수준을 확인하고자 하였다.
상기 수득한 역분화줄기세포와 신경전구세포에 각각 trizol(TRI Reagent solution, ambion, am9738)을 처리한 다음, 각 세포의 총 RNA를 수득하였다. 상기 수득한 총 RNA를 대상으로 역전사 효소(M-MLV Reverse transcriptase, M1701, prormega)를 사용한 방법을 수행하여 cDNA를 합성하고, 상기 합성된 cDNA를 대상으로 SYBR Premix Ex Taq II(takara, RR820A)를 사용한 방법을 수행하여 줄기세포 마커인 SOX1과 SOX2; 신경전구세포 마커인 DCX, Musashi1, Nestin 및 Tuj1; 및 신경세포 마커인 NeuN의 발현수준을 비교하였다. 아울러, NeuN, Nestin 및 SOX2에 대한 면역형광염색을 수행하였다(도 3a 및 3b). 이때, 내부대조군으로 β-Ⅲ 튜불린을 사용하였다.
도 3a 및 3b에서 보듯이, 정상인 및 알츠하이머 환자의 피부 섬유아세포에서 수득한 역분화줄기세포에서는 모든 마커 단백질의 발현수준이 균일하게 발현됨을 확인하였다. 이에 반하여, 신경전구세포에서는 줄기세포 마커(SOX1, SOX2) 및 신경세포 마커(NeuN)의 발현수준은 낮은 수준을 나타내었고, 신경전구세포 마커(DCX, Musashi1, Nestin 및 Tuj1)는 현저하게 높은 수준을 나타냄을 확인하였다.
따라서, 상기 실시예 2에서 수득한 각각의 신경전구세포는 정상적으로 분화된 신경전구세포임을 알 수 있었다.
실시예 3-2: 콜린성 신경세포에서의 분화마커의 발현수준
상기 실시예 2의 방법을 통해 얻어진, 알츠하이머 환자로부터 유래된 콜린성 신경세포(BFCN_SAD2-2)를 대상으로, 신경세포 마커(Tuj1 및 MAP2)와 콜린성 신경세포 마커(VAChT 및 ChAT)에 대한 면역형광염색을 수행하여, 이들 마커의 발현수준을 비교하였다(도 4).
도 4에서 보듯이, 신경세포 마커인 Tuj1 및 MAP2는 약 63 내지 65%의 세포에서 발현되었고, 콜린성 신경세포 마커인 VAChT는 약 91.7%의 세포에서 발현됨을 확인하였으므로, 상기 알츠하이머 환자로부터 유래된 역분화 줄기세포로부터 분화된 세포가 대부분 콜린성 신경세포임을 알 수 있었다.
한편, 상기 실시예 2의 방법을 통해 얻어진, 알츠하이머 환자로부터 유래된 신경전구세포(NP_SAD2-2)와 콜린성 신경세포(BFCN_D25_SAD2-2)를 대상으로, 신경세포 마커(NeuN, Tuj1 및 MAP2)와 콜린성 신경세포 마커(VAChT 및 ChAT)의 상대적인 발현수준을 비교하였다(도 5).
도 5에서 보듯이, 신경전구세포(NP_SAD2-2)에서는 모든 신경세포 마커(NeuN, Tuj1 및 MAP2)와 콜린성 신경세포 마커(VAChT 및 ChAT)가 상대적으로 낮은 수준으로 발현되었으나, 콜린성 신경세포(BFCN_D25_SAD2-2)에서는 콜린성 신경세포 마커(VAChT 및 ChAT) 뿐만 아니라, 신경세포 마커(NeuN, Tuj1 및 MAP2)도 높은 수준으로 발현됨을 확인하였다.
따라서, 상기 실시예 2의 방법을 통해 최종적으로 분화된 세포는 콜린성 신경세포임을 다시한번 확인할 수 있었다.
또한, 상기 실시예 2의 방법을 통해 얻어진, 알츠하이머 환자로부터 유래된 콜린성 신경세포(BFCN_D25_SAD2-2)를 대상으로, 신경세포 마커(MAP2 및 Tuj1), 콜린성 신경세포 마커(VAChT 및 ChAT) 및 GABA성 신경세포(GABAergic neuron) 마커(GABA)의 상대적인 발현수준을 비교하였다(도 6).
도 6에서 보듯이, 신경세포 마커인 MPA2 또는 Tuj1으로 염색된 세포에서 대부분의 세포가 VAChT 또는 ChAT로는 염색이 되나, GABA성 신경세포 마커로는 거의 염색이 되지 않음을 확인하였다.
또한, 상기 면역염색 결과를 정량화한 결과, 신경세포 마커인 Tuj1-positive 세포에서 ChAT-positive 세포의 비율이 약 99.4 또는 100 %임을 확인하였다.
이를 통해, 본 발명에서 제공하는 콜린성 신경세포의 생산방법을 이용하면, 동일한 형질의 콜린성 신경세포를 매우 높은 순도로 생산할 수 있음을 알 수 있었다.
이상의 설명으로부터, 본 발명이 속하는 기술분야의 당업자는 본 발명이 그 기술적 사상이나 필수적 특징을 변경하지 않고서 다른 구체적인 형태로 실시될 수 있다는 것을 이해할 수 있을 것이다. 이와 관련하여, 이상에서 기술한 실시 예들은 모든 면에서 예시적인 것이며 한정적인 것이 아닌 것으로서 이해해야만 한다. 본 발명의 범위는 상기 상세한 설명보다는 후술하는 특허 청구범위의 의미 및 범위 그리고 그 등가 개념으로부터 도출되는 모든 변경 또는 변형된 형태가 본 발명의 범위에 포함되는 것으로 해석되어야 한다.

Claims (16)

  1. 줄기세포로부터 신경전구세포를 형성한 다음, 상기 신경전구세포를 콜린성 신경세포로 분화시키는 단계를 포함하는, 콜린성 신경세포의 생산방법.
  2. 제1항에 있어서,
    상기 콜린성 신경세포의 생산방법은
    (a) 줄기세포를 배양하여 배아유사세포 덩어리(embryoid body)를 수득하는 단계;
    (b) 상기 수득한 배아유사세포 덩어리를 SHH(Sonic Hedgehog)를 포함하는 배지에서 배양하여 신경구세포(neurosphere)를 수득하는 단계;
    (c) 상기 수득한 신경구세포를 배양하여 신경전구세포를 수득하는 단계; 및
    (d) 상기 수득한 신경전구세포를 콜린성 신경세포 분화용 배지에서 배양하여 콜린성 신경세포를 생성하는 단계를 포함하는 것인 방법.
  3. 제1항에 있어서,
    상기 줄기세포는 중간엽 줄기세포, 배아줄기세포 또는 역분화줄기세포인 것인 방법.
  4. 제3항에 있어서,
    상기 역분화줄기세포는 피부 섬유아세포에 hOct3/4-p53sh, hSox-KLF4, hUl 및 EBNA1 유전자를 도입하여 수득한 것인 방법.
  5. 제1항에 있어서,
    상기 줄기세포는 배아유사세포 덩어리, 로제트(Rosette), 신경구세포 및 신경전구세포를 거쳐 콜린성 신경세포로 분화되는 것인 방법.
  6. 제2항에 있어서,
    상기 (a) 단계는 줄기세포를 DMEM/F12 배지 또는 KO-DMEM/F12 배지에서 배양하는 것인 방법.
  7. 제2항에 있어서,
    상기 (b) 단계는 8 내지 20일 동안 수행되는 것인 방법.
  8. 제2항에 있어서,
    상기 (b) 단계에서 배아유사세포 덩어리를 N2, B27, bFGF(basic fibroblast growth factor) 및 SHH를 포함하는 로제트 형성용 배지에서 배양하여 로제트를 수득한 다음, 상기 로제트를 배양하여 신경구세포를 수득하는 것인 방법.
  9. 제8항에 있어서,
    상기 로제트 형성용 배지는 N2, B27, bFGF 및 SHH를 포함하는 DMEM/F12 배지 또는 KO-DMEM/F12 배지인 것인 방법.
  10. 제2항에 있어서,
    상기 (c) 단계에서 신경구세포를 무혈청 배지에서 배양하여 신경전구세포를 수득하는 것인 방법.
  11. 제2항에 있어서,
    상기 (d) 단계에 콜린성 신경세포 분화용 배지는 Glutamax, B27, EGF, 헤파린, 아스코빈산, BDNF(brain-derived neurotrophic factor) 및 이들의 조합으로 구성된 군으로부터 선택되는 물질을 포함하는 것인 방법.
  12. 제2항에 있어서,
    상기 (d) 단계는
    (d1) 신경전구세포를 콜린성 신경세포 분화용 배지에서 배양하여 콜린성 신경세포를 생성하는 단계;
    (d2) 상기 배지에 분화억제 물질을 처리하여, 콜린성 신경세포 이외의 다른 종류의 신경세포의 증식을 억제하는 단계; 및
    (d3) 상기 배지에 성장인자를 처리하여, 콜린성 신경세포를 증식하는 단계를 포함하는 것인 방법.
  13. 제12항에 있어서,
    상기 (d2) 단계는 1 내지 5일 동안 수행되고, 상기 분화억제 물질은 시타라빈(cytarabine, AraC)인 것인 방법.
  14. 제12항에 있어서,
    상기 (d3) 단계는 (d) 단계의 시작 시점으로부터 4 내지 15일이 경과된 후에 수행되고, 상기 성장인자는 bFGF 것인 방법.
  15. 제1항에 있어서,
    상기 콜린성 신경세포의 생산방법은
    (a') 줄기세포를 1 내지 10일 동안 배양하여 배아유사세포 덩어리를 수득하는 단계;
    (b') 상기 수득한 배아유사세포 덩어리에 SHH를 처리하면서 4 내지 10일 동안 배양하여 로제트를 수득하는 단계;
    (c') 상기 수득한 로제트에 SHH를 처리하면서 4 내지 10일 동안 배양하여 신경구세포를 수득하는 단계;
    (d') 상기 수득한 신경구세포를 1 내지 5일 동안 배양하여 신경전구세포를 수득하는 단계;
    (e') 상기 수득한 신경전구세포를 콜린성 신경세포 분화용 배지에서 3 내지 10일 동안 배양하여 콜린성 신경세포를 생성시키는 단계;
    (f') 상기 배지에 시타라빈을 처리하고 1 내지 5일 동안 배양하여, 콜린성 신경세포 이외의 다른 종류의 신경세포의 증식을 억제하는 단계; 및
    (g') 상기 배지에 bFGF를 처리하고 10 내지 30일 동안 배양하여, 콜린성 신경세포를 증식하는 단계를 포함하는 것인 방법.
  16. 제1항 내지 제15항 중 어느 한 항의 방법으로 제조된 콜린성 신경세포.
