Abstract
Description
しかしながら、特許文献2~4の前処理方法は、いずれも原料に対して20%前後のアルカリを用いているため薬剤費用がかかる上、酵素糖化前に必ず添加したアルカリの中和と洗浄が必要になるため、糖化工程まで含めた経済性と効率性に関する課題は解決されていない。さらに、特許文献2及び3は、前処理が実質的に粉砕とアルカリ蒸煮を組み合わせたものであることを唄っているが、その処理条件については、加熱温度と加熱時間、投入する原料やアルカリの数量などアルカリ蒸煮に関わる条件を示すにとどまり、粉砕に関わる装置の構成などについては提示されておらず、発明を実施するための形態は不明瞭である。
しかしながら、特許文献5は、そのためのフローチャートや押出機のスクリュー構成を記載しているが、処理方法をどのような条件により実施し、どれくらいの糖化性能が得られるかのバイオマスの糖化データの提示はなく、糖化工程まで含めたトータルの性能や効率性は不明である。
[1]スクリュー押出機を用いて連続的に水熱処理を行う、セルロース含有バイオマス原料から糖化用バイオマス組成物を低電力量で製造する前処理方法であって、スクリュー押出機の粉砕部で、セルロース含有バイオマス原料を最大粒径1000μm以下に粉砕し、かつ含水率を30~80%に調整し、次いで加熱部で、擂り潰し(擂潰)効果のある混錬粉砕を行いながら205~250℃の温度で0.1~10分間水熱処理を行い、続いて加熱部以降の冷却部で処理物を100℃以下に冷却し回収することを特徴とするバイオマスの処理方法。
[2]加熱部において、シールリングの直前にニーディングディスク及び/または逆ネジを1セット以上配置したエレメントを配置して、擂潰効果のある混錬粉砕を水熱処理と同時に行う[1]に記載のバイオマスの処理方法。

本発明の処理方法に用いるバイオマスとは、枯渇性資源(石油・石炭・天然ガスなどの化石燃料)を除く、生体高分子(核酸、タンパク質、多糖)やこれらの構成要素を起源とする産業資源を意味する。従って、セルロース含有バイオマスには、例えば木材などのハードバイオマスと、稲わら、麦わら、コーンコブ、キャッサバ、バガス、サトウキビ葉などのソフトバイオマスが挙げられる。前処理の容易性を考慮するとソフトバイオマスが好ましく、さらに全世界的な賦存量と収集コストを考慮すると、バガス、サトウキビ葉が特に好ましい。
本発明の処理方法に用いるスクリュー押出機は、単軸スクリュー押出機、多軸スクリュー押出機、特殊押出機のいずれでもよいが、より強いせん断をバイオマス材料に加えることができる多軸スクリュー押出機が好ましく、一般的で汎用性があることから二軸押出機がより好ましい。
スクリューのかみ合い型と非かみ合い型のいずれでもよいが、混練効果が大きく実用例も多いスクリューかみ合い型が好ましい。
スクリュー回転方向は、同方向回転型と異方向回転型のいずれでもよいが、自己清掃効果のある同方向回転型が好ましい。
原料をスクリュー押出機のシリンダーに安定して供給するためのホッパーとしては、例えば振動ホッパー、強制フィーダ付ホッパー、ホッパードライヤー、真空ホッパー、窒素置換ホッパーなど、原料がブリッジを起こさず、スクリューのフィード部に必要な供給圧力を生じさせることができる機能を持ち合わせていれば限定されないが、原料の安定供給の観点より、強制的に材料をシリンダーに押込むためのスクリューが内部に付いたホッパーが好ましい。
スクリュー押出機のシリンダー部は、シリンダーの中央部にあり原料を擂潰しながらヒーターで加熱して水熱処理をする加熱部と、加熱部の上流側に存在し、原料の粉砕と含水率を調整し、圧密化させて気密性を保持する粉砕部と、加熱部の下流側に存在し、冷却を行い、圧密化して気密性を保持する冷却部の3つの部分からなる。スクリュー押出機は、安定的にシールを保ち、原料バイオマスの糖化性能を改善する効果のある水熱処理を行うという観点より、粉砕部、加熱部及び冷却部を合わせた全体のL/Dは30~80が好ましく、40~80がより好ましく、50~80がさらに好ましい。粉砕部のL/Dは10~40が好ましく、10~30がより好ましく、15~25がさらに好ましい。また、加熱部のL/Dは10~65が好ましく、15~60がより好ましく、20~55がさらに好ましい。冷却部のL/Dは5~35が好ましく、5~20がより好ましく、5~10がさらに好ましい。なお、L/Dとは、ホッパー下のねじの切り始めから先端までのスクリューの長さ(L)と直径(D)との比率で表される有効長である。
シリンダーの粉砕部のスクリュー構成は、シールリングの前にニーディングディスク(送りニーディングディスク、中立ニーディングディスク、逆ニーディングディスク)または逆ネジを1セット以上配置したエレメント(以下、シールリングエレメントと略記する。)を少なくとも1つ以上配置することが好ましい。配置されたシールリングのせき止め効果で上流側の原料が圧縮された状態が形成され、シールリングの前にあるスクリューのせん断力が強化された状態となり、原料が効率的に粉砕と圧密化をされて加熱部で発生する蒸気圧力をシールする機能を持つ。原料の粉砕はシール機能のためだけでなく加熱部での水熱処理効率を向上させるためのものでもあり、そのための原料の最大粒径は1000μm以下とすることが好ましい。なお、最大粒径は、加熱部直前の粉砕部から抜き出したサンプルの顕微鏡観察により求められる。
また、粉砕部では水熱反応とシール性を最適に行うために原料の含水率を30~80質量%にすることが好ましく、30~75質量%にすることがより好ましく、35~70質量%にすることがさらに好ましい。含水率の調整は、投入前に別途行っても良いが、工程数を減らす観点から粉砕部の任意の場所に注液ラインを設置して高圧ポンプで供水することが好ましい。なお、含水率とは原料の有り姿全体の質量に対する含有する水分の質量の割合を示す。
シリンダーの加熱部のスクリュー構成は、シールリングエレメントを、少なくとも3セット以上配置することが好ましい。加熱部における複数シールリングの効果は、セルロース含有バイオマスがシールリングとシリンダーとの非常に狭いクリアランスの部分を原料が通過する際に生じる強い擂り潰し応力が、水熱処理と同時にかかることにより、バイオマス中のセルロースの糖化性を向上させることである。そのためのシールリングとシリンダーのクリアランスはシリンダー内径の0.5~10.0%が好ましく、1.0~8.0%がより好ましく、1.5~5.0%がさらに好ましい。なお、二軸押出機におけるシリンダー内径は、スクリュー一軸分を囲むシリンダーの内径を示す。加熱部の加熱は、シリンダーを加熱できるものであれば限定されないが温度制御性の観点から電気ヒーターが好ましい。原料温度は205~250℃の範囲にあることが好ましく、210~240℃がより好ましく、210~230℃がさらに好ましい。加熱部の通過時間は0.1~10分の範囲にあることが好ましく、0.2~9分がより好ましく、0.3~7.5分がさらに好ましい。加熱部の圧力は、1~20MPaの範囲にあることが好ましく、1.5~15MPaがより好ましく、2~12MPaがさらに好ましい。本発明においては、原料を水熱処理する際の条件が上記範囲であると、電力コストの観点から好適である。
シリンダーの冷却部は、加温部で加熱された原料を冷却するために冷却水冷ジャケット及び/または注液ラインを取り付けることが好ましい。冷却部での冷却は100℃以下に下げることが好ましく、80℃以下がより好ましく、70℃以下がさらに好ましい。これにより加熱部で生じた蒸気が水になり、処理されたバイオマスとともに下流への蒸気圧力をシールすることができる。さらに系内の蒸気圧力のより安定的なシールのために、冷却部の排出口に調圧バルブを取り付けることもできる。
本発明の処理方法では、原料であるセルロース含有バイオマスは粉砕処理なしで直接スクリュー押出機に供給することができるが、供給前に事前に粗粉砕して粒径調整を行うことが好ましい。粉砕手段は固体の物質を粉砕できる機能を備えているものであれば特に限定されない。例えば、装置の方式は乾式と湿式のいずれでもよく、装置の粉砕システムは回分式と連続式いずれでもよい。さらに、装置の粉砕力は、衝撃、圧縮、せん断、摩擦などのいかなるものをも用いることができる。
本発明の処理方法では、スクリュー押出機への供給前に事前に原料であるセルロース含有バイオマス含水率を調整することもできる。含水率を調整する方法は、調整前の原料の含水率に合わせて、加水または、脱水若しくは乾燥することが挙げられる。原料の含水率は、水熱反応とシール性を最適に行うために前述のように30~80質量%にすることが好ましい。
実施例及び比較例では5種類の装置条件のスクリュー押出機を用い、原料の粒径、水熱温度、水熱時間の条件を振ってセルロース含有バイオマスの処理を実施し、処理原料に対する押出機のモーターの消費電力量を比較した。さらに処理したサンプルの糖化率評価を行い、糖化工程まで含めた生成糖に対する消費電力量が低くトータルの性能、効率が優れた方法、条件を検討した。
L/D=77.0、スクリュー径32mmの二軸スクリュー押出機(商品名:(株)日本製鋼所製TEX30α)を、図1(A)に示す粉砕部6ブロック、加熱部14ブロック(シールリングエレメント6セット)、冷却部2ブロックのスクリュー構成にセットしたスクリュー押出機Aを実施例7~9、比較例2、3で用いた。
粉砕部のブロック数、加熱部のブロック数とシールリングエレメント数、冷却部のブロック数を表1のように変更したこと以外はスクリュー押出機Aと同様である、スクリュー押出機B~E(それぞれ図1(B)~(E)に示す)を、それぞれ表1に記載の実施例、比較例で用いた。
なお、表1にはL/D値、加熱部以外のブロック数やシールリングエレメント数、シールリング使用数、シールリングとスクリューとのクリアランスも併記した。また、スクリュー押出機Cでは、シリンダー径47mm(商品名:(株)日本製鋼所製TEX44α)を用いた。
原料のセルロース含有バイオマスにはバガスを用いた。
バガスは全く処理していないもの(含水率50%、セルロース含有率38%、以後、「未処理バガス」と略記する。)と、風乾したバガスを、スクリーン径3mmφのカッターミル(増幸産業株式会社製、MKCM-3、)で粉砕したもの(含水率10.0%、セルロース含有率42%、以後、「3mmバガス」と略記する。)を用いた。
バイオマス中のセルロース含有率、ヘミセルロース含有率、及びリグニンと灰分を合計した含有率は、NREL(米国・国立再生可能エネルギー研究所)の分析方法(Technical Report NREL/TP-510-42618)により求めた。
ガードカラム(昭和電工株式会社製、KS-G)と分離カラム(昭和電工株式会社製 KS-802)を接続し、カラム温度を75℃に設定した。純水を溶離液として0.5ml/分で供給し、分離成分は示差屈折率検出器を用いて定量しグルコース濃度を求め、下記式によりセルロース含有率を算出した。
酸緩衝液の調製:
酢酸30gを100mlメスフラスコに入れ、純水でメスアップし3M酢酸水溶液とした。酢酸ナトリウム41gを100mlメスフラスコに入れ、純水でメスアップし3M酢酸ナトリウム水溶液とした。3M酢酸ナトリウム水溶液に5M酢酸水溶液をpH=5.0になるまで加え、酢酸緩衝液とした。
メイセラーゼ(登録商標、明治製菓株式会社(現Meiji Seikaファルマ株式会社)製セルラーゼ)1.5gを純水98.5gに溶解させた。
酵素液のFPU活性(Filter Paper Assay for Saccharifying Cellulase)は、IUPAC(国際純正・応用化学連合)の分析方法(Pure & Appl.Chem.,Vol.59,No.2,pp.257-268,1987)より求めた結果、6FPU/gであった。
50mlの蓋つきガラス容器に回転子を入れ、セルロース量が0.5gになるように前処理組成物を秤量し、上記酢酸緩衝液0.6g、酵素液1.03g加え、さらに純水を加えて合計10gとした。40℃の恒温槽で撹拌しながら72時間(Hr)酵素糖化反応を行った。得られた糖化液を高速液体クロマトグラフィー分析してグルコースを定量し、糖化率及び糖利用率を以下の計算式により算出した。
水熱処理時の押出機のモーターの消費電力量として、処理原料(kg)に対する消費電力量(kWh)(以下、対原料モーター消費電力量と略記する。)、及び処理したサンプルを糖化して得られた生成糖(kg)に対する消費電力量(kWh)(以下、対糖モーター消費電力量と略記する。)を以下の計算式により算出した。
未処理バガスを質量フィーダ及びコンパクターを用いて、有り姿質量20.0kg/Hr、乾燥質量換算10.0kg/Hrの供給速度で、スクリュー回転数を200rpmとしたスクリュー押出機Cに投入し、粉砕部において注液ラインより5.0kg/Hrの供給速度で水を添加して、引き続き加熱部前の粉砕部での含水率が60質量%(固形分濃度 40質量%)となるよう調整し、水熱温度を210℃、圧力5MPaとして水熱処理を行った後、冷却部で原料を水冷ジャケット及び注液ラインから供給速度5kg/Hrの注水により70℃以下に冷却して、排出口からサンプルを回収した。この条件での水熱時間は2.5分であった。回収した処理サンプル100gに、サンプルの固形分に対して3倍量の水100gを添加して懸濁した後、遠心ろ過機(株式会社コクサン製、H-122、ろ布コットン)を用いて3000rpmで遠心ろ過し含水固形分を取得した。得られた含水固形分は前述の方法にてセルロース回収率及び糖化率を算出した。
各種条件を表2に記載のように変更したこと以外は実施例1と同様に操作した。なお、表2にはセルロース回収率、糖化率、糖利用率も併記した。
比較例1と比較例6は温度175℃、時間7.5分、比較例2と比較例7は温度190℃時間7.5分のそれぞれ同じ水熱条件を、異なるスクリュー構成の押出機で行った。
比較例1及び2の押出水熱は、加熱部にシールリングの直前にニーディングディスク及び/または逆ネジを1セット以上配置したエレメント(シールリングエレメント)を5セット配置した条件のスクリュー押出機B(図1(B))、及び6セット配置した条件のスクリュー押出機A(図1(A))により行った。それに対して、比較例6、7では加熱部にシールリングエレメントを配置しないスクリュー押出機E(図1(E))、D(図1(D))により行った。
水熱後のセルロース回収率は、いずれも90%を超えた値となったが、水熱温度が低い175℃の方が若干良い値となった。
このシールリングエレメントの効果は、セルロース含有バイオマスがシールリングとシリンダーとの非常に狭いクリアランスの部分を原料が通過する際に生じる強い擂り潰し応力が、水熱処理と同時にかかることにより、バイオマス中のセルロースの糖化性を向上させるためと推測される。
本明細書における実施例、比較例は、スクリュー押出で水熱処理ができたテスト例であり、それらの水熱直前の原料の最大粒径は、スクリュー押出機の加熱部より前の粉砕部から抜き出したサンプルを顕微鏡観察した結果いずれも1000μm以下まで粉砕されていることが分かった。
一方、テストはしたが実際には水熱処理できなかった条件がある。それはスクリュー押出機A及びスクリュー押出機Eのそれぞれの粉砕部にあるシールリングエレメントからシールリングを外した場合である。3mmバガスを供給して水熱処理を試みたが、粉砕部において圧力を保持できず蒸気が逆流して処理できなかった。この時の水熱前の粉砕部から回収した原料の最大粒径はいずれも1000μmを超え粉砕がしっかりされていなかった。
以上より粉砕部での粗粉砕は、押出水熱において糖化性能を向上させる効果と、水熱部で発生する蒸気の上流側のシールする効果が得られているものと推測され、そのレベルは最大粒径が1000μmを下回る方が好ましいことが確認された。
本明細書における実施例及び比較例は、スクリュー押出機で水熱処理ができたテスト例である。いずれのスクリュー押出機も、冷却部には水冷ジャケット及び注液ラインの冷却システム、排出口には調圧バルブを設置して用いた。各テストは、実施例7~9及び比較例1~7では水冷ジャケットにより冷却を行い、実施例1~6では水冷ジャケットと注液ラインから処理バイオマスへの注水による冷却を併用して、冷却部の温度を100℃以下にしたところ、いずれのテストも安定的に連続の水熱処理をすることができた。
一方、テストはしたが実際には水熱処理できなかった条件がある。各スクリュー押出機において水冷ジャケット及び注液ラインのいずれの冷却システムも使用せずに処理を行ったところ、いずれのスクリュー押出機の場合においても、冷却部の温度は100℃を超えた高温状態のままで、サンプルが蒸気とともに断続的に噴出し安定的な処理ができなかった。以上の通り、冷却部では水冷ジャケット及び/または注液ラインの冷却システムを使用して温度を100℃以下にすることが安定運転に必要であることが確認された。
水熱温度による挙動を比較するため、実施例7~9及び比較例2、3は、スクリュー押出機Aを用いた水熱時間7.5分の処理において、また実施例1~3及び比較例4、5は、スクリュー押出機Cを用いた水熱時間2.5分の処理において、それぞれ水熱温度だけを変えたテストである。結果を表2、図2及び3に示す。
本発明のスクリュー押出機による水熱処理において、消費電力量と糖化率などのデータは、原料条件、装置条件、水熱条件の違いにより影響を受けて変動する。このため、図2、図3では、温度以外の条件が全て同一である実験例ごとに実線で結び、実線内の実験例では、温度の違いによる対原料消費電力量、対糖消費電力量の変化を評価した。また、実線間の実験例では、それ以外の条件の違いによる対原料消費電力量、対糖消費電力量の変化を評価した。
水熱と同時にスクリューにより擂り潰しや粉砕を含む混練を行う押出水熱のモーター消費電力は、処理時の原料の性状に依存する。このため上記の変曲点は、原料のバガスに含有する非セルロース成分であるヘミセルロースとリグニンの多くが200℃から210℃の温度で可溶化して、低粘度化して流動性が向上することにより引き起こされたものと推測される。
なお、「3mmバガス」と「未処理バガス」の違いによる影響は、他の条件が同一の場合、消費電力は未処理バガスは粉砕部での負荷がかかることにより増加し、糖化率は変動しない傾向となる。これに伴い、対原料消費電力、対糖消費電力とも、未処理バガスの方が若干高くなる傾向にある。
図2、3では3mmバガスを用いた実施例7~9及び比較例2、3の方が、未処理バガスを用いた実施例1~6及び比較例4、5より消費電力量が高い。従って、3mmバガスと未処理バガスの違いによる影響は小さく、他の条件の違いによる影響の方が大きいといえる。
実施例は4~6は、スクリュー押出機Cの2.5分の水熱処理で対糖消費電力量が低く効率的であった水熱温度210℃以上の実施例1~3に対して、それぞれ水熱時間を1.2分に短縮して行った水熱処理である。水熱時間の短縮は、原料の供給速度を増大してシリンダー内の加熱部の滞留時間を短くして行った結果、いずれも、供給速度増大により消費電力は増大したが、供給した原料に対する消費電力量は2.5分の水熱より低下した。
水熱後のセルロース回収率は、いずれも95%で、水熱時間2.5分に比べより高くなり前記と同じ傾向となった。
また、糖化率は、210℃(実施例4)は低下し、220℃(実施例5)、230℃(実施例6)は増大した。これは、各水熱処理では各温度により最適の水熱時間域があり、それより短い時間ではバイオマスへの前処理による作用が弱く糖化率が低下し、長い時間では不純物の変性や、糖の過分解物などの増大により糖化率が低下する傾向によるものと推測される。本処理条件における210℃の水熱では2.5分付近に、220℃、230℃の1.2分付近に最適時間条件があることが示唆された。
その結果、対糖消費電力量は、セルロース回収率と糖化率が反映された結果、いずれも水熱時間2.5分に比べて低くなり、220℃(実施例5)が2.7kWh/kgで最も低い値となった。

- スクリュー押出機を用いて連続的に水熱処理を行う、セルロース含有バイオマス原料から糖化用バイオマス組成物を低電力量で製造する前処理方法であって、スクリュー押出機の粉砕部で、セルロース含有バイオマス原料を最大粒径1000μm以下に粉砕し、かつ含水率を30~80%に調整し、次いで加熱部で、擂り潰し(擂潰)効果のある混錬粉砕を行いながら205~250℃の温度で0.1~10分間水熱処理を行い、続いて加熱部以降の冷却部で処理物を100℃以下に冷却し回収することを特徴とするバイオマスの処理方法。
