Abstract
Die Erfindung stellt ein Verfahren zur Synthese von Carbonsäureamiden zur Verfügung, umfassend die Schritte: a) Bereitstellen eines Carbonsäureesters der Formel RCOOR' und Ammoniak (NH3), b) Mischen des Carbonsäureesters der Formel RCOOR' und des Ammoniaks (NE3), und c) Reagieren des Carbonsäureesters der Formel RCOOR' und des Ammoniaks (NH3) zu einem Carbonsäureamid der Formel RCONH2 und R'OH, dadurch gekennzeichnet, dass der Ammoniak in Schritt a) in flüssiger Form oder in überkritischer Form zugeführt wird. Die Erfindung stellt weiterhin eine Anlage zur Durchführung des erfindungsgemäßen Verfahrens zur Verfügung.
Description
Verfahren zur Synthese von Carbonsäu rcamiden
Die vorliegende Erfindung betrifft ein Verfahren zur Synthese von Carbons äureamiden.
Carbonsäureamide werden im Stand der Technik durch Umsetzung von Säurechloriden, -anhydriden und - e stern mit Ammoniak oder Aminen hergestellt. Die Carbonsäureamide wie Trichloressigsäureamid bzw. Tnfluoressigsäureamid sind wichtige Vorprodukte zur Herstellung agrochemischer Wirkstoffe.
Die Synthese von Trifluoressigsäureamid ausgehend vom Methylester wird beispielsweise in J. Agric. Food Chem. 2009, 57, 8303-8307 beschrieben. Trifluoressigsäuremethylester wird in Methanol vorgelegt und über Nacht mit einer methanolischen Ammoniak-Lösung umgesetzt. Das Lösungsmittel und überschüssiges Ammoniak werden am folgenden Tag abdestilliert, wobei das Produkt als weißer Feststoff erhalten wird. WO 03/080563 offenbart ein Verfahren zur Herstellung von Carbons äureamiden aus Carbonsäureestern und Aminen unter Bildung einer Lösung von Amid in Alkohol. Das flüssig gehaltene Carbons äureamid wird aus dem Reaktor ausgebracht, in kleinere Volumina aufgeteilt und diese unter Abkühlung in den festen Zustand überführt.
WO 2007/ 106100 A 1 offenbart ein Verfahren und eine Anlage zur Umsetzung von ß-Hydroxy- Carbonylverbindungen zu Produkten wie Acyrlsäure und Acrylamid.
Der Nachteil dieser bekannten Verfahren ist, dass diese kompliziert sind und durch die zeitliche Trennung der Schritte langwierig und teuer sind. Daher ist der bisher bekannte Weg zur Herstellung von Carbons äureamiden zeitintensiv und führt zu einer ungenügenden Raum- Zeit- Ausbeute, da es sich um reine batch- Verfahren handelt. Außerdem erfordern diese bekannten Verfahren den Einsatz von Lösungsmitteln mit allen Nachteilen hinsichtlich Kosten, mangelnder Effizienz und Gefahrenpotential.
Es bestand daher ein verstärktes Interesse an der Entwicklung eines neuen, einfachen, und preiswerten Verfahrens zur Herstellung von Carbonsäureamiden, insbesondere von Trichloressigsäureamid und Trifluoressigsäureamid, ausgehend von einfach zugänglichen Vorprodukten wie den Estern dieser Carbonsäureamide. Daher war es die Aufgabe der vorliegenden Erfindung, ein Verfahren zur Herstellung von Carbonsäureamiden bereit zu stellen, welches die Nachteile der bekannten batch- Verfahren vermeidet.
Die Erfindung löst diese Aufgabe durch Bereitstellen eines Verfahrens zur Synthese von Carbonsäureamiden, insbesondere von Trichloressigsäureamid und Tri fluoressigsäureamid, ausgehend von den entsprechenden Carbonsäureestern und Ammoniak. Insbesondere löst die Erfindung diese Aufgabe durch Bereitstellen eines Verfahrens zur Synthese von Carbonsäureamiden, umfassend die Schritte:
a) Bereitstellen eines Carbonsäureesters der Formel RCOOR' und Ammoniak (NH3), b) Mischen des Carbonsäureesters der Formel RCOOR' und des Ammoniaks (NH3), und c) Reagieren des Carbonsäureesters der Formel RCOOR' und des Ammoniaks (NH3) zu einem Carbons äureamid der Formel RCONH2 und R'OH, wobei
R für verzweigte oder unverzweigte optional substituierte Alkylgruppen mit 1 bis 6 Kohlenstoffatomen, beispielsweise Methyl, Ethyl, n-Propyl, n-Butyl, Iso-Propyl, sec-Butyl, Iso- Butyl, tert-Butyl, n-Pentyl oder n-Hexyl, oder für substituiertes Phenyl, Naphthyl oder 5-6 gliedrige heterozyklische Ringe umfassend 1-3 Atome, die ausgewählt sind aus Stickstoff, Schwefel und Sauerstoff steht, und wobei
R' für verzweigte oder unverzweigte optional substituierte Alkylgruppen mit 1 bis 6 Kohlenstoffatomen, beispielsweise Methyl, Ethyl, n-Propyl, n-Butyl, Iso-Propyl, sec-Butyl, Iso- Butyl, tert-Butyl, n-Pentyl oder n-Hexyl steht, dadurc gekennzeichnet, dass der Ammoniak in Schritt a) in flüssiger Form oder in überkritischer Form zugeführt wird. in einer Ausführungsform des erfindungsgemäßen Verfahrens wird der Ammoniak in Schritt a) in flüssiger Form zugeführt.
In einer anderen Ausführungsform des erfindungsgemäßen Verfahrens wird der Ammoniak in Schritt a) in überkritischer Form zugeführt.
Im Rahmen der Erfindung bedeutet der Begriff„überkritische Form" den thermodynamischen Zustand des Ammoniaks, der sich durch Angleichen der Dichten von flüssiger und Gasphase kennzeichnet. Die Unterschiede zwischen beiden Aggregatzuständen hören an diesem Punkt auf zu existieren. Im Phasendiagramm stellt der Punkt das obere Ende der Dampfdruckkurve dar. Für Ammoniak beträgt der kritische Druck 113 bar und die kritische Temperatur 132,4°C. Oberhalb dieser Parameter liegt Ammoniak also in überkritischer Form vor.
Der technische Vorteil des erfindungsgemäßen Verfahrens ist es, dass eine sehr viel höhere Raum-Zeit- Ausbeute erzielt wird, als dies mit den oben beschriebenen bereits bekannten Verfahren möglich ist. Ein weiterer Vorteil dieses Verfahrens ist es, dass es ohne den Einsatz von Lösungsmitteln auskommt und daher effizienter und kostengünstiger betrieben werden kann.
Das erfindungsgemäße Verfahren kann als batch-V erfahren oder in einer kontinuierlichen Verfahrensweise durchgeführt werden. In einer bevorzugten Ausführungsform kann das Verfahren kontinuierlich durchgeführt werden. Eine kontinuierliche Verfahrensweise bedeutet hier, dass die Edukte kontinuierlich zugeführt werden und die entsprechenden Produkte kontinuierlich abgeführt werden. Die allgemeine Reaktionsgleichung, die dem erfindungsgemäßen Verfahren zugrunde liegt, sieht wie folgt aus:

Wo ein Rest R oder R' optional substituiert sein kann, können typische Substituenten umfassen: Halogen, CN, N02, OH. NH2, Ci-C8 Alkyl, Ci-C8 Alkoxy, C3-C8 Cycloalkyl, C2-C8 Alkenyl, C2-C8 Alkynyl, CO(Ci-C8 Alkyl), COH, SH. Ci-C8 Alkylthio, Ci-C8 Alkylsulphinyl, Ci-C8 Alkylsulphonyl, Alkylamin, Dialkylamin, C02H, C02(Ci-C8 Alkyl), 0(CO)Ci-C8 Alkyl, 0(CO)H, CON(Ci-C8 Alkyl)2, CONH(Ci-C8 Alkyl), NHCO(Ci-C8 Alkyl), NHCOH, N(Ci-C8 Alkyl)CO(Ci-C8 Alkyl) or NHCO(Ci-C8 Alkyl), wobei Alkyl, Alkoxy, Cycloalkyl, Alkenyl und Alkynyl optional substituiert sind mit einer oder mehreren Gruppen unabhängig ausgewählt aus Halogen, CN, NH2, N02, OH, C1-C4 Alkyl, C1-C4- Haloalkyl, C1-C4 Alkoxy und Ci-Ct Haloalkoxy. In einer weiterhin bevorzugten Ausführungsform ist R = CCI3 und R'= CH3 oder C2Hs. In einer weiterhin bevorzugten Aus führungs form ist R = CF3 und R'= CH3 oder C2Hs.
ln einer weiteren Ausführungsform des erfindungsgemäßen Verfahrens kann der Carbonsäuerester in Schritt a) ausgewählt sein aus der Gruppe bestehend aus Trifluoressigsäureester, Trichloressigsäurester, Trifluoressigsäure- ethylester und Trichloressigsäure- ethylester.
In einer bevorzugten Ausführungsform des erfindungsgemäßen Verfahrens kann das Verhältnis von Ammoniak (NH3) zu Carbonsäureester der Formel RCOOR' im Bereich von 100: 1 bis 1 : 1 liegen.
Überraschenderweise wurde gefunden, dass die Reaktionsgeschwindigkeit der zugrunde liegenden Reaktion des erfindungsgemäßen Verfahrens mit dem Anstieg des Verhältnisses von Ammoniak (NH3) zu Carbonsäureester der Formel RCOOR' zunimmt. Das bedeutet, es wurde ein eindeutiger Trend hin zur einer Beschleunigung der Reaktionsgeschwindigkeit zusammen mit dem Anstieg des Verhältnisses von Ammoniak (NH3) zu Carbonsäureester beobachtet.
Die zugrunde liegende chemische Reaktion des erfindungsgemäßen Verfahrens findet in der Regel schnell statt, wobei eine vollständige Umsetzung von 100% Edukt zu Produkt erzielt wird, sogar bei einer Temperatur von 0°C und in Gegenwart eines erhöhten Verhältnisses von Ammoniak (NH3) zu Carbonsäureester der Formel RCOOR'. Ein weiterer Gegenstand der vorliegenden Erfindung ist eine Anlage zur Durchführung des erfindungsgemäßen Verfahrens. Ein Beispiel einer solchen Anlage ist in Figur 1 gezeigt.
Insbesondere wird die der Erfindung zugrunde liegende Aufgabe weiterhin gelöst durch Bereitstellen einer Anlage zur Synthese von Carbonsäureamiden, umfassend die Komponenten: a) Vorlagenbehälter 1 für Ammoniak,
b) Vorlagenbehälter 2 für einen Carbonsäureester,
c) Kaskadenmischer 3,
d) Verweiler 4, und
e) Phasens ep arator 5. Das erfindungsgemäße Verfahren kann in dieser Anlage durchgeführt werden.
Die erfindungsgemäße Anlage kann weiterhin eine kontinuierliche Verfahrens weis e vorsehen.
Der Edukt-Fluss an Ammoniak (NH3) aus dem Vorlagenbehälter 1 wird als„Strom 1" bezeichnet
Der Edukt-Fluss an Carbonsäureester aus dem Vorlagenbehälter 2 wird als„Strom 2" bezeichnet.
Die erfindungsgemäße Anlage sieht folgende Mischreihenfolge vor: Strom 1 aus dem Vorlagenbehälter 1 (Ammoniak, flüssig oder in überkritischer Form) und Strom 2 aus dem Vorlagenbehälter 2 (z.B. Trifluoressigsäure-ethylester in THF) werden über Zuleitungen in den Kaskadenmischer 3 eingeleitet und dort vermischt.
Bevorzugte Kaskadenmischer 3 können solche von der Firma Ehrfeld Mikrotechnik GmbH sein, z.B. ein Kaskadenmischer 06 (Art.Nr. 0216-3).
Dieses Gemisch durchströmt daraufhin einen Verweiler 4. Die Strecken des Verweilers 4 können aus
V2A Edelstahl sein. Beispielsweise kann eine V2A-Edelstahl-Kapillare aus 1,0 m * 1/8" (I.D. 2.1 mm) und 5,0 m * 1/16" * 0,13 mm sein, wie es in Beispiel 1 gezeigt ist.
Der Kaskadenmischer 3 kann von einem Thermostat / Kryostat 6 („Julabo") kontrolliert werden.
Nach dem Verweiler 4 wird das Gemisch in einen Phasenseparator 5 geleitet, um das überschüssige Ammoniak zu verdampfen und dieses somit kontinuierlich vom Produkt zu trennen.
Dem Phasenseparator 5 kann eine Probenentnahmestelle 7 nachgeschaltet sein, um Proben zu entnehmen. Die Probenentnahme kann hier ebenfalls kontinuierlich erfolgen.
Der Probenentnahmestelle 7 kann ein Ammoniakwäscher 8 nachgeschaltet sein, der zur Aufarbeitung/Neutralisation des eventuell vorhandenen Überschusses an Ammoniak dient.
Die vorliegende Erfindung einschließlich bevorzugter Ausführungsformen wird in Verbindung mit den nachfolgenden Zeichnungen und Beispielen erläutert, ohne hierauf beschränkt zu sein. Die Ausführungsformen können beliebig miteinander kombiniert werden, sofern sich nicht eindeutig das Gegenteil aus dem Kontext ergibt.

FIG. 1 zeigt schematisch eine Anlage zur Durchführung des erfindungsgemäßen Verfahrens. 1 = Vorlagenbehälter für Ammoniak, Strom 1 , flüssig oder in überkritischer Form 2 = Vorlagenbehälter für Carbonsäureester, Strom 2 (z.B. Trifluoressigsäure- ethylester in THF)
3 = Kaskadenmis eher, kann von einem Thermostat kontrolliert werden.

5 = Phasenseparator
6 = Thermostat / Kryostat („Julabo") 7 = Probenentnahmestelle
8 = Ammoniakwäscher
Tabelle 1 zeigt die Versuchsergebnisse aus Beispiel 1.
Beispiel 1 :
Die in Beispiel 1 durchgeführte Reaktion war wie folgt:
Reaktordetails
• Einstufige Reaktion mit Kaskadenmis ehern- Stücken (Ehrfeld Mikrotechnik GmbH) und Verweilerstrecken aus V2A;
• Strom 1 = Trifluoressigsäure-ethylester in THF (R'= C2H5)
• Strom 2 = Ammoniak, flüssig
• Mischreihenfolge: Strom 1 (Trifluoressigsäure- ethylester in TH F) und Strom 2 (Ammoniak, flüssig) werden in einem Kaskadenmis eher vermischt; diese Gemisch durchströmt daraufhin einen Verweiler.
• Verweiler hinter dem Mischer: Eine V2A- Kapillare aus 1,0 m * 1/8" (I.D. 2,1 mm) und 5,0 m * 1/16" * 0,13 mm.
• Druckregelkapillare hinter dem Magnetventil 1,0 m * 1/16" * 0,25 mm Der Aufbau der in Beispiel 1 verwendeten Anlage ist in Figur 1 dargestellt.
Ansatz Vorlage 1 :

Ans atz große der Eduktvorlage: Ausreichend für die Versuchspunkte plus genügend Volumen zum Pumpen spülen. Alle Eduktvorlagen und die Produktvorlagen müssen mit Argon überschleiert sein.
Den Reaktor nach den Versuchen mit THF spülen.
Probenahme: Erste Probennahme nach 5 Verweilzeiten, zweite Probennahme nach insgesamt 6 Verweilzeiten. Bei den konti- Versuchen für 1 min Probe ziehen, die Masse der gezogenen Probe auf +/- 0,01 g bestimmen.
Probeaufarbeitung:
Die Probeanalyse erfolgte mittels Gaschromatographie/HPLC.
Vergleichsversuche mit anderen Lösungsmitteln als Ammoniak zeigten, dass Ammoniak die schnellste Reaktionsgeschwindigkeit ermöglicht, gefolgt von Acetonitril, gefolgt von Ethanol.

Die Analytik basierte auf "offline" Gaschromatographie (interner Standard-Technik), wobei die Messungen direkt nach der Probenentnahme mit in situ„mid-IR" Spektroskopie (also im mittleren IR- B ereich) durchgeführt wurden. Für letzteres wurde ein "mid-IR" optischer Sensor für in situ ATR- (abgeschwächte Totalreflexion, "attenuated total reflectance") Messungen über ein T-Stück an dem Ausgang des Reaktors verbunden, der auf an einen Diamanten gekoppelte S ilberhalogenid-Fasern basierte.
Für eine quantitative Analyse wurde eine Multivarianten-Kalibrationsanalyse durchgeführt.
Um die Konzentration der Komponenten in jeder Probe gleichzeitig zu bestimmen, wurde eine partielle Methode der kleinsten Quadrate verwendet. Durch Kopplung des kontinuierlichen Reaktors mit Online-
IR-Messungen wurde der experimentelle und zeitliche Aufwand mimimiert. So konnten technische Probleme, wie Fehlfunktionen der Pumpen, direkt erkannt und behoben werden, und die daraus resultierenden wenig verlasslichen Daten konnten entsprechend aus der Analyse ausgeschlossen werden. So war die Verlässlichkeit und Genauigkeit der Daten nicht abhängig von dem Downstream-Prozess und der Probenentnahme. Die oben beschriebene Analyse in situ„mid-IR" Spektroskopie wurde auch erfolgreich mit anderen experimentellen Ansätzen durchgeführt, bei denen anstelle von Trichloressigsäure-ethylester ein anderers Edukt, Trifluoressigsäure- ethyiester, als Substrat verwendet wurde.
Claims
1. Verfahren zur Synthese von Carbons äureamiden, umfassend die Schritte: a) Bereitstellen eines Carbonsäureesters der Formel RCOOR' und Ammoniak (NH3), b) Mischen des Carbonsäureesters der Formel RCOOR' und des Ammoniaks (NH3), und c) Reagieren des Carbonsäureesters der Formel RCOOR' und des Ammoniaks (NH3) zu einem
Carbonsäureamid der Formel RCONH2 und R'OH, wobei
R für verzweigte oder unverzweigte optional substituierte Alkylgruppen mit 1 bis 6 Kohlenstoffatomen, beispielsweise Methyl, Ethyl, n-Propyl, n-Butyl, Iso-Propyl, sec-Butyl, Iso- Butyl, tert-Butyl, n-Pentyl oder n-Hexyl, oder für substituiertes Phenyl, Naphthyl oder 5-6 gliedrige heterozyklische Ringe umfassend 1-3 Atome, die ausgewählt sind aus Stickstoff, Schwefel und Sauerstoff steht, und wobei
R' für verzweigte oder unverzweigte optional substituierte Alkylgruppen mit 1 bis 6 Kohlenstoffatomen, beispielsweise Methyl, Ethyl, n-Propyl, n-Butyl, Iso-Propyl, sec-Butyl, Iso-
Butyl, tert-Butyl, n-Pentyl oder n-Hexyl steht, dadurc gekennzeichnet, dass der Ammoniak in Schritt a) in flüssiger Form oder in überkritischer Form zugeführt wird.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass das Verfahren als batch- Verfahren oder in einer kontinuierlich en Verfahrensweise durchgeführt wird.
3. Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass der Carbonsäuerester der Formel RCOOR' in Schritt a) ausgewählt ist aus der Gruppe bestehend aus Trifluoressigsäureester, Trichloressigsäurester, Trifluoressigsäure-ethylester und Trichloressigsäure-ethylester.
4. Verfahren nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass das Verhältnis von Ammoniak (NH3) zu Carbonsäureester der Formel RCOOR' im Bereich von 100:1 bis 1: 1 liegt.
5. Anlage zur Synthese von Carbonsäureamiden, umfassend die Komponenten: a) Vorlagenbehälter (1) für Ammoniak b) Vorlagenbehälter (2) für einen Carbonsäureester,
c) Kaskadenmischer (3), d) Verweiler (4), und e) Phasenseparator (5).
6. Anlage zur Synthese von Carbons äureamiden nach Anspruch 1, dadurch gekennzeichnet, dass Sie eine kontinuierliche Verfahrensweise vorsieht.
7. Verfahren nach einem der Ansprüche 1 bis 4, dadurch gekennzeichnet, dass das Verfahren in der Anlage nach Anspruch 5 oder 6 durchgeführt wird.
