本発明にかかる化合物(I)は、後述する参考例1に具体的に記載されているように、3-オキソテトラヒドロフラン、2-フルオロ-5-メチルピリジンおよび4-ブロモ-2-フルオロ安息香酸を出発原料として合成することができる。
化合物(I)の塩は、通常の塩を製造する方法により得ることができる。具体的には、例えば、化合物(I)を溶媒に、必要に応じて加温して、懸濁または溶解させ、次いで、得られる懸濁液または溶液に、有機カルボン酸、有機スルホン酸および無機酸からなる群から選択される一つの酸を加え、室温下あるいは氷浴で冷却しながら数分から数日間撹拌することにより、または室温下あるいは氷浴で冷却しながら数分から数日間放置することにより、製造することができる。これらの製造方法により、化合物(I)の塩を、結晶または非晶質として得ることができる。ここで使用する溶媒としては、例えばエタノール、1-プロパノール、イソプロパノール等のアルコール系溶媒;アセトニトリル;アセトン、2-ブタノン等のケトン系溶媒;酢酸エチル等のエステル系溶媒;ヘキサン、ヘプタン等の飽和炭化水素系溶媒;t-ブチルメチルエーテル等のエーテル系溶媒または水を挙げることができる。これらの溶媒は単独で使用してもよく、2種以上を混合して使用してもよい。
化合物(I)の塩の結晶は、上述の化合物(I)の塩の製造方法により、または化合物(I)の塩を、溶媒中で加熱溶解し、攪拌下冷却して晶析することにより、製造することができる。

3-オキソテトラヒドロフラン(5.70g)をメタノール(150mL)に溶解し、ベンジル カルバゼート(10g)をこの溶液に加えた。混合物を室温にて12時間撹拌した。反応液を濃縮した。14.8gの残渣を粗生成物として得た。これ以上精製せずに次の反応に用いた。
ベンジル 2-[ジヒドロフラン-3(2H)-イリデン]ヒドラジンカルボキシレート(14.8g)を水(96mL)に懸濁した。この懸濁液に酢酸(42.1mL)を室温にて加えた。この混合物を室温にて1時間撹拌した。懸濁液が溶液になった。この溶液にシアノ水素化ホウ素ナトリウム(4.0g)を少しずつ加えた。この混合液を室温にて2時間撹拌した。反応液を0℃に冷却した。5N水酸化ナトリウム水溶液を加え中和した。この混合物をクロロホルムで抽出した。有機層を無水硫酸マグネシウムで乾燥した後、ろ過した。ろ液を減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(メタノール/酢酸エチル,5%)で精製した。標記化合物(13.9g)を得た。

(±)-ベンジル 2-(テトラヒドロフラン-3-イル)ヒドラジンカルボキシレート(11.5g)のMTBE(110mL)溶液へ、飽和炭酸水素ナトリウム水溶液(30mL)を加え、室温で10分間撹拌後、有機層を分離した。得られた有機層を飽和炭酸水素ナトリウム、飽和食塩水で順次洗浄し、無水硫酸マグネシウムで乾燥し、乾燥剤をろ過して除去した。ろ液を減圧下に濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(酢酸エチル/ヘキサン,25~50%)で精製し、目的画分を濃縮した。残渣へジエチルエーテル(30mL)とヘキサン(15mL)を加えた。析出した固体をろ取し、減圧下に乾燥して、純粋な(±)-ベンジル 2-(テトラヒドロフラン-3-イル)ヒドラジンカルボキシレート(6.17g)を得た。
この物を、エタノールに溶解し、ミリポアフィルターを通して、ろ過した。得られたろ液を2条件で光学分割した。
条件1:DAICEL社製CHIRALCEL(登録商標) OD-H(20mmφ×250mmL),20%IPA/ヘキサン,25mL/分。
条件2:DAICEL社製CHIRALPAK(登録商標) AD-H(20mmφ×250mmL),20%IPA/ヘキサン、24mL/分。
目的画分を濃縮して、短保持時間で(-)の旋光性を有する標記化合物(2.60g,>99%ee[OD-H,20%IPA/へキサン、保持時間:11.2分])および長保持時間で(+)の旋光性を有する標記化合物(2.59g,97.2%ee[OD-H,20%IPA/へキサン、保持時間:12.4分])を得た。
(-)-ベンジル 2-(テトラヒドロフラン-3-イル)ヒドラジンカルボキシレート(50g)をメタノール(500mL)に溶解し、ジ-t-ブチル ジカルボネート(92.4g)とパラジウム炭素(50%wet)(5g)を加え、水素雰囲気下、25℃で15psiで48時間撹拌した。反応液をろ過し、ろ液を減圧下濃縮した。得られた残渣をジイソプロピルエーテル(300mL)に溶解した。0℃に冷却した後、この溶液に塩酸/ジイソプロピルエーテル(500mL)加えた。この混合物を10℃で14時間撹拌した。析出した固体をろ取した。(-)-ベンジル 2-(テトラヒドロフラン-3-イル)ヒドラジンカルボキシレート(70g)から同様の操作を9回行い、(-)-ベンジル 2-(テトラヒドロフラン-3-イル)ヒドラジンカルボキシレート(50g)から同様の操作を1回行った。得られた固体をDCM/エタノール(10/1)(1L)で2時間トリチュレートした。析出した固体をろ取した。得られた固体を減圧下乾燥し、標記化合物(235g)を得た。

なお、得られた標記化合物の絶対配置はX線結晶構造解析により(S)-体であると確認した。
THF(1.2L)にジイソプロピルアミン(92mL)を加え、窒素雰囲気下、-18℃に冷却した。この溶液に2.69M n-ブチルリチウム へキサン溶液(224mL)を滴下した。滴下終了後、この混合物を撹拌しながら20分かけて-5℃まで昇温した。反応液を-73℃に冷却した。この反応液に2-フルオロ-5-メチルピリジン(61g)のTHF溶液(240mL)を滴下した。反応混合物を-75℃にて3時間半撹拌した。この反応液にヨウ素(139g)のTHF溶液(24mL)を滴下した。反応混合物を-75℃にて1時間55分撹拌した。反応終了後、同温度で水(220mL)を反応液に加えた。混合物を5分間同温で撹拌した。反応液を室温に戻した後、水(1.2L)を加えた。この混合物に、チオ硫酸ナトリウム五水和物(136g)水溶液(300mL)と水(300mL)を加え、10分間撹拌した。この混合物をMTBE(1.2L)で抽出した。有機層を飽和食塩水(500mL)で洗浄した。合わせた水層をMTBE(1L)で抽出した。合わせた有機層を無水硫酸マグネシウムで乾燥した。乾燥剤をろ過して除き、ろ液を減圧下濃縮した。残渣にn-ヘプタンを加え、冷却した。析出した固体をろ取した。n-ヘプタンで洗浄した。ろ液を冷却し、析出した固体をろ取した。この操作を5回繰り返すことで、標記化合物(109.69g)を得た。

THF(1.2L)にジイソプロピルアミン(88mL)を加え、窒素雰囲気下、-18℃に冷却した。この溶液に2.69M n-ブチルリチウム へキサン溶液(215mL)を滴下した。滴下終了後、この混合物を撹拌しながら30分かけて-5℃まで昇温した。反応液を-72℃に冷却した。この反応液に2-フルオロ-3-ヨード-5-メチルピリジン(109.69g)のTHF溶液(240mL)を滴下した。反応混合物を-74℃にて1時間半撹拌した。この反応液にヨウ化メチル(36mL)のTHF溶液(160mL)を滴下した。反応混合物を-70℃~-74℃にて2時間撹拌した。反応終了後、同温度で水(200mL)を反応液に加えた。混合物を2分間同温で撹拌した。反応液を室温に戻した後、水(1.2L)を加えた。この混合液を3分間撹拌した。更に水(300mL)を加えた。この混合物をMTBE(1.2L)で抽出した。有機層を飽和食塩水(500mL)で洗浄した。合わせた水層をMTBE(1L)で抽出した。合わせた有機層を無水硫酸マグネシウムで乾燥した。乾燥剤をろ過して除き、ろ液を減圧下濃縮した。残渣にn-ヘプタン(100mL)を加え、冷却した。析出した固体をろ取した。n-ヘプタンで洗浄した。ろ液を冷却し、析出した固体をろ取した。この操作を2回繰り返すことで、標記化合物(86.9g)を得た。

2-フルオロ-4-ヨード-3,5-ジメチルピリジン(97.4g)のTHF(954mL)溶液に、20℃で、28%ナトリウムメトキシド メタノール溶液(185mL)を加えた。この混合物を55℃~65℃にて2時間撹拌した。反応液を冷却した後、MTBE(1L)と水(1L)を加えて分離した。有機層を飽和食塩水で洗浄した。合わせた水層をMTBE(500mL×2)で抽出した。合わせた有機層を無水硫酸マグネシウムで乾燥した。ろ過して乾燥剤を除き、ろ液を減圧下濃縮した。残渣にn-ヘプタン(50mL)を加え、0℃にて1時間撹拌した。析出した固体をろ取した。冷やしたn-ヘプタン(10mL)で固体を洗浄した。標記化合物(42.6g)を得た。ろ液を減圧下濃縮した。残渣にn-ヘプタン(5mL)を加え、0℃にて30分間撹拌した。析出した固体をろ取した。冷やしたn-ヘプタン(2mL)で固体を洗浄し、標記化合物(20.2g)を得た。ろ液を減圧下濃縮した。残渣にn-ヘプタン(5mL)を加え、0℃にて30分間撹拌した。析出した固体をろ取した。冷やしたn-ヘプタン(2mL)で固体を洗浄した。標記化合物(10.7g)を得た。合わせて標記化合物(73.5g)を得た。

4-ブロモ-2-フルオロ安息香酸(CAS No.112704-79-7)(10g)のDCM(97mL)懸濁液へ、CDI(8.88g)を加え、室温にて3.5時間撹拌した。この溶液を「溶液1」とする。
別のフラスコ中、カリウム エチルマロネート(15.5g)のアセトニトリル(303mL)懸濁液へ、TEA(15.9mL)、次いで塩化マグネシウム(10.9g)を加え、室温で3時間10分間撹拌した。この反応混合物へ、先に調製した「溶液1」を25分間かけて滴下した後、反応混合物を室温で終夜撹拌した。反応液を減圧下に半量まで濃縮した。得られた残渣を酢酸エチル(500mL)で希釈し、氷冷下に5N塩酸(250mL)を加えた後、室温にて1時間撹拌した。有機層を分離した。有機層を飽和食塩水で洗浄し、無水硫酸マグネシウムで乾燥し、ろ過し、減圧下に濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(酢酸エチル/n-ヘプタン,5~20%)で精製し、標記化合物(12.8g)を得た。

エチル 3-(4-ブロモ-2-フルオロフェニル)-3-オキソプロパノエート(45g)のDMF-DMA(165mL)溶液を、50℃で2時間15分間撹拌した。反応液を減圧下に濃縮した。残渣にトルエン(200mL)を加え、再び減圧下濃縮した。残渣にエタノール(950mL)を加え50℃に加温した。その溶液へ(S)-(テトラヒドロフラン-3-イル)ヒドラジン塩酸塩(21.6g)の水溶液(60mL)を35分間かけて滴下で加えた。得られた反応混合物を、50℃で2時間10分間撹拌した。反応液を室温まで冷却した後、減圧下に半分量まで濃縮した。残渣へ水(200mL)を加え、減圧下にエタノールを留去した。得られた残渣に酢酸エチル(500mL)を加え、有機層を分離した。水層を酢酸エチル(100mL)で抽出した。合わせた有機層を飽和食塩水で洗浄し、無水硫酸マグネシウムで乾燥し、ろ過し、減圧下に濃縮した。残渣をシリカゲルカラムクロマトグラフィー(酢酸エチル/n-ヘプタン,10%~15%)で精製し、次いでショートパスNHシリカゲル(富士シリシア化学株式会社製のプロピルアミンコーティングシリカゲル)カラムクロマトグラフィー(酢酸エチル/n-ヘプタン,33%)で精製し、標記化合物(43.1g)を得た。

エチル 5-[2-フルオロ-4-(4,4,5,5-テトラメチル-1,3,2-ジオキサボロラン-2-イル)フェニル]-1-[(S)-テトラヒドロフラン-3-イル]-1H-ピラゾール-4-カルボキシレート(51.9g)の1,4-ジオキサン(500mL)溶液へ、水(170mL)、4-ヨード-2-メトキシ-3,5-ジメチルピリジン(35.6g)、Pd(PPh3)4(6.52g)および炭酸セシウム(110g)を加え、反応混合物を110℃で6時間反応した。反応液を室温に戻した後、有機層を分離した。有機層を減圧下濃縮した。得られた残渣へ、水層、酢酸エチル(700m)および水(100mL)を加え、有機層を分離した。水層を酢酸エチル(50mL)で再抽出した。あわせた有機層を、水および飽和食塩水で順次洗浄し、無水硫酸マグネシウム上で乾燥し、ろ過し、減圧下に濃縮した。残渣をNHシリカゲルカラムクロマトグラフィー(酢酸エチル/n-ヘプタン,5~14%)で精製した。ついで、再度NHシリカゲルカラムクロマトグラフィー(酢酸エチル/n-ヘプタン,2~10%)で精製し、標記化合物(43.5g)を得た。

エチル 5-[2-フルオロ-4-(2-メトキシ-3,5-ジメチルピリジン-4-イル)フェニル]-1-[(S)-テトラヒドロフラン-3-イル]-1H-ピラゾール-4-カルボキシレート(43.2g)のエタノール(574mL)溶液へ、室温で5N水酸化ナトリウム水溶液(79mL)を加え、反応混合物を60℃で2時間10分間撹拌した。反応液を室温まで冷却した後、減圧下に半量まで濃縮した。残渣へ水(300mL)を加え、減圧下にエタノールを留去した。得られた残渣にMTBE(130mL)を加え、水層を分配した。有機層を水(30mL)で抽出した。あわせた水層を,氷冷下に5N塩酸(78mL)にて酸性化後、酢酸エチルで2回抽出した。合わせた有機層を無水硫酸マグネシウムで乾燥し、ろ過し、減圧下に濃縮して標記化合物(39.0g)を得た。

5-[2-フルオロ-4-(2-メトキシ-3,5-ジメチルピリジン-4-イル)フェニル]-1-[(S)-テトラヒドロフラン-3-イル]-1H-ピラゾール-4-カルボン酸(38.7g)のDMF(290mL)溶液へ、室温下にCDI(21.4g)を一度に加え、室温で95分間撹拌した。この反応混合物へ、28%アンモニア水(95mL)を加え、室温で35分間撹拌した。反応液へ再度28%アンモニア水(95mL)を加え、室温で90分間撹拌した。反応混合物を減圧下に濃縮した。得られた残渣へクロロホルム(250mL)と水(80mL)を加え、有機層を分離した。水層をクロロホルム(50mL)で再抽出した。合わせた有機層を、飽和塩化アンモニウム水溶液(60mL、3回)および飽和食塩水で順次洗浄し、無水硫酸マグネシウムで乾燥し、ろ過した。ろ液を、シリカパッド(NH-シリカゲル)に通過させた。ろ液を減圧下に濃縮して、標記化合物(37.2g)を得た。

5-[2-フルオロ-4-(2-メトキシ-3,5-ジメチルピリジン-4-イル)フェニル]-1-[(S)-テトラヒドロフラン-3-イル]-1H-ピラゾール-4-カルボキサミド(37.2g)のDMSO(186mL)溶液へ、室温下に、水酸化ナトリウム粉末(9.43g)を一度に加えた。反応混合物を、同温で50分間、次いで70℃で45分間撹拌した。水冷下に、反応液へ水(600mL)を滴下で加え、次いで氷酢酸(13.5mL)を滴下で加えた。析出した粉末をろ取した。ろ取した物を水およびMTBEで洗浄後、減圧下に乾燥して、標記化合物(34.0g)を得た。

なお、標記化合物は(-)の旋光性を示し、その光学純度は99%ee以上[AD-H, 100%エタノール、保持時間:9.7分]であった。
4-ブロモ-2-クロロ安息香酸(1g)をDCM(10mL)に懸濁した。その懸濁液に、CDI(960mg)を加え室温にて4時間撹拌した。この溶液を「溶液1」とする。別のフラスコに、窒素雰囲気下、カリウム エチルマロネート(1.1g)をアセトニトリル(20mL)に懸濁し、TEA(1.5mL)を加えた。この溶液を0℃に冷却し、塩化マグネシウム(805mg)を少しずつ加えた後、室温にて2時間撹拌した。この反応液を0℃に冷却し、先に調製した「溶液1」を滴下した。滴下終了後、室温にて17時間撹拌した。さらに50℃で9時間撹拌した。反応液を減圧下濃縮しDCMを除去した。得られた残渣を0℃に冷却し、酢酸エチル(50mL)と2N塩酸(20mL)を加え、室温にて1時間撹拌した。有機層を分離した。水層を酢酸エチルで抽出した。有機層を合わせ、無水硫酸マグネシウムで乾燥した。乾燥剤をろ去し、ろ液を減圧下濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(酢酸エチル/n-ヘプタン,0~10%)で精製し、標記化合物(1.2g)を得た。

エチル 3-(4-ブロモ-2-クロロフェニル)-3-オキソプロピオネート(2.00g)をDMF-DMA(6.96mL)に溶解し、室温にて1.5時間撹拌した。反応液を減圧下濃縮し、残渣をエタノール(40mL)に溶解した。その溶液に(±)-(テトラヒドロフラン-3-イル)ヒドラジン・塩酸塩(998mg)を加え、2時間加熱還流した。反応液を室温に冷却した後、減圧下濃縮した。残渣を酢酸エチルで抽出し、有機層をシリカゲルカラムクロマトグラフィー(酢酸エチル/n-ヘプタン,10~30%)で精製し、標記化合物(1.05g)を得た。

(±)-エチル 5-(4-ブロモ-2-クロロフェニル)-1-(テトラヒドロフラン-3-イル)-1H-ピラゾール-4-カルボキシレート(1.05g)および5N水酸化ナトリウム水溶液(1.58mL)の混合物をエタノール(20mL)と水(5mL)の混合溶媒中、60℃にて3時間撹拌した。反応液を室温に冷却した後、減圧下濃縮した。残渣に5N塩酸を加え、酢酸エチルで抽出した。有機層を無水硫酸マグネシウムで乾燥させ、乾燥剤をろ過した。ろ液を減圧下濃縮することで、標記化合物(1g)を得た。

(±)-5-(4-ブロモ-2-クロロフェニル)-1-(テトラヒドロフラン-3-イル)-1H-ピラゾール-4-カルボン酸(1g)をDCM(20mL)に溶解し、CDI(611mg)を加え、室温にて1時間撹拌した。反応液に2,4-ジメトキシベンジルアミン(0.809mL)を加え、室温にて2時間撹拌した。反応液に飽和炭酸水素ナトリウム水溶液を加え、DCMで抽出した。有機層を減圧下濃縮し、残渣をシリカゲルカラムクロマトグラフィー(酢酸エチル/n-ヘプタン,10~40%)で精製し、標記化合物(1.26g)を得た。

(±)-5-(4-ブロモ-2-クロロフェニル)-N-(2,4-ジメトキシベンジル)-1-(テトラヒドロフラン-3-イル)-1H-ピラゾール-4-カルボキサミド(1.26g)をTHF(25mL)に溶解し、0℃にてKTB(597mg)を加え、少しずつ室温に昇温させながら12時間撹拌した。反応液を0℃に冷却し、水を加え、ろ過した。ろ取した物を別途保管した。ろ液を酢酸エチルで抽出し、有機層を減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(酢酸エチル/n-ヘプタン,10~70%)で精製した。得られたフラクションを先ほど得られた濾物と合わせて濃縮することで、標記化合物(488mg)を得た。

(±)-7-ブロモ-5-(2,4-ジメトキシベンジル)-1-(テトラヒドロフラン-3-イル)-1H-ピラゾロ[4,3-c]キノリン-4(5H)-オン(300mg)、ビス(ピナコラト)ジボロン(204mg)、Pd(dppf)Cl2・DCM錯体(13.6mg)および酢酸カリウム(182mg)の混合物を1,4-ジオキサン(15mL)とDMSO(1mL)の混合溶媒中にて、マイクロウェーブ反応装置を用い、130℃にて3時間反応させた。反応液を室温に冷却した後、減圧下濃縮した。残渣を酢酸エチルで抽出し、有機層を減圧下濃縮した。残渣をシリカゲルパッドに供し、酢酸エチルで溶出することで標記化合物(428mg)を粗生成物として得た。

2-メトキシ-ピリジン-4-イルアミン(15g)とNBS(47.3g)の混合物を酢酸(150mL)溶媒中、室温にて3時間撹拌した。反応液を減圧下濃縮し、残渣に0℃にて5N水酸化ナトリウム水溶液(200mL)を加え、ジエチルエーテルで抽出した。有機層をそのままシリカゲルパッド(酢酸エチル/n-ヘプタン,10%)で精製し、標記化合物(32.4g)を得た。

3,5-ジブロモ-2-メトキシピリジン-4-アミン(16g)、トリメチルボロキシン(19.8mL)、Pd(dppf)Cl2・DCM錯体(4.15g)および炭酸カリウム(23.5g)の混合物を、1,4-ジオキサン(320mL)および水(32mL)の混合溶媒中で12時間加熱還流した。反応液を室温に冷却した後、減圧下濃縮した。残渣に水と酢酸エチルを加え、セライトTMを用いてろ過した。ろ液を酢酸エチルで抽出し、有機層をシリカゲルパッド(NH-シリカゲル)に供し、酢酸エチルで溶出した。得られた溶液にNH-シリカゲル(30g)を加え、減圧下濃縮した。残渣をNHシリカゲルカラムクロマトグラフィー(酢酸エチル/n-ヘプタン,0~30%)で精製し、標記化合物(4.43g)を得た。

臭化銅(I)(12.1g)と亜硝酸t-ブチル(7.07mL)の混合物をアセトニトリル(80mL)溶媒中、70℃にて10分撹拌した。その反応液に同温度にて2-メトキシ-3,5-ジメチルピリジン-4-アミン(3.9g)のアセトニトリル(40mL)溶液を滴下し、70℃にて1時間撹拌した。反応液を室温に冷却した後、減圧下濃縮した。残渣に酢酸エチルおよび飽和炭酸水素ナトリウム水溶液を加え、室温にて30分撹拌した。反応液をセライトTMを用いてろ過し、ろ液を酢酸エチルで抽出した。有機層を減圧下濃縮し、残渣をNHシリカゲルカラムクロマトグラフィー(n-ヘプタン,100%、続いてNH-シリカゲルパッド,n-ヘプタン,100%、)で精製し、標記化合物(4.3g)を得た。

(±)-5-(2,4-ジメトキシベンジル)-1-(テトラヒドロフラン-3-イル)-7-(4,4,5,5-テトラメチル-1,3,2-ジオキサボロラン-2-イル)-1H-ピラゾロ[4,3-c]キノリン-4(5H)-オン(219mg)、4-ブロモ-2-メトキシ-3,5-ジメチルピリジン(134mg)、Pd(PPh3)4(23.8mg)および炭酸セシウム(403mg)の混合物を1,4-ジオキサン(8mL)および水(2mL)の混合溶媒中、マイクロウェーブ反応装置を用い130℃にて70分間反応させた。反応液を室温に冷却した後、そのままシリカゲルカラムクロマトグラフィー(酢酸エチル/n-ヘプタン,10%~90%)で精製した。得られたカップリング体をTFA(4mL)に溶解し、70℃にて2時間撹拌した。反応液を室温に冷却した後、減圧下濃縮した。残渣に飽和炭酸水素ナトリウム水溶液を加え、酢酸エチルで抽出した。有機層を減圧下濃縮し、残渣をシリカゲルカラムクロマトグラフィー(DCM,100%,続いて、酢酸エチル/n-ヘプタン,50%~100%)で精製することで標記化合物(78mg)を得た。

(±)-7-(2-メトキシ-3,5-ジメチルピリジン-4-イル)-1-(テトラヒドロフラン-3-イル)-1H-ピラゾロ[4,3-c]キノリン-4(5H)-オンのキラルカラム分析[AD-H(0.46cmφ×15cm)、移動層:100%エタノール]を行い、(+)-体を7.8分、(-)-体を9.7分確認し、光学分割可能であることを確認した。(±)-7-(2-メトキシ-3,5-ジメチルピリジン-4-イル)-1-(テトラヒドロフラン-3-イル)-1H-ピラゾロ[4,3-c]キノリン-4(5H)-オン(78mg)をエタノール(12mL)およびメタノール(12mL)の混合溶媒に溶解し、綿栓ろ過した。ろ液をキラルカラムクロマトグラフィー[DAICEL社製キラルカラム:AD-Hカラム,溶出溶媒:100%-エタノール,流速:10mL/分,溶出時間:80分/回,インジェクション:2mL/回,短保持時間:(+)-体,長保持時間:(-)-体]で光学分割することで、標記化合物(+)-体(26.4mg)および(-)-体(25.2mg)を得た。

(S)-7-(2-メトキシ-3,5-ジメチルピリジン-4-イル)-1-(テトラヒドロフラン-3-イル)-1H-ピラゾロ[4,3-c]キノリン-4(5H)-オン(23.53mg)に2-ブタノン(300μL)と95%硫酸(4μL)を加え、懸濁液を室温で一晩撹拌した。固形物をろ取して、減圧下室温で乾燥することにより、表題化合物(27.34mg)を白色固体として得た。

(S)-7-(2-メトキシ-3,5-ジメチルピリジン-4-イル)-1-(テトラヒドロフラン-3-イル)-1H-ピラゾロ[4,3-c]キノリン-4(5H)-オン(22.52mg)に2-ブタノン(300μL)とメタンスルホン酸(4.5μL)を加え、懸濁液を室温で一晩撹拌した。固形物をろ取して、減圧下室温で乾燥することにより、表題化合物(28.69mg)を白色固体として得た。

(S)-7-(2-メトキシ-3,5-ジメチルピリジン-4-イル)-1-(テトラヒドロフラン-3-イル)-1H-ピラゾロ[4,3-c]キノリン-4(5H)-オン・リン酸塩の合成
(S)-7-(2-メトキシ-3,5-ジメチルピリジン-4-イル)-1-(テトラヒドロフラン-3-イル)-1H-ピラゾロ[4,3-c]キノリン-4(5H)-オン(22.29mg)に2-ブタノン(300μL)とリン酸(4.6μL)を加え、懸濁液を室温で一晩撹拌した。さらにヘキサン(200μL)添加して撹拌後、固形物をろ取して、減圧下室温で乾燥することにより、表題化合物(23.09mg)を白色固体として得た。

(S)-7-(2-メトキシ-3,5-ジメチルピリジン-4-イル)-1-(テトラヒドロフラン-3-イル)-1H-ピラゾロ[4,3-c]キノリン-4(5H)-オン・L-酒石酸塩の合成
(S)-7-(2-メトキシ-3,5-ジメチルピリジン-4-イル)-1-(テトラヒドロフラン-3-イル)-1H-ピラゾロ[4,3-c]キノリン-4(5H)-オン(27.42mg)にアセトン(300μL)とL-酒石酸(20.18mg)を加え、懸濁液を室温で3日間撹拌した。固形物をろ取して、減圧下室温で乾燥することにより、表題化合物(34.89mg)を白色固体として得た。

(S)-7-(2-メトキシ-3,5-ジメチルピリジン-4-イル)-1-(テトラヒドロフラン-3-イル)-1H-ピラゾロ[4,3-c]キノリン-4(5H)-オン・マロン酸塩の合成
(S)-7-(2-メトキシ-3,5-ジメチルピリジン-4-イル)-1-(テトラヒドロフラン-3-イル)-1H-ピラゾロ[4,3-c]キノリン-4(5H)-オン(27.70mg)にアセトン(300μL)とマロン酸(26.65mg)を加え、懸濁液を室温で3日間撹拌した。固形物をろ取して、減圧下室温で乾燥することにより、表題化合物(30.49mg)を白色固体として得た。
粉末X線回折ピーク(2θ±0.2°):10.5°、16.8°、17.4°、17.8°、18.3°、18.9°、21.7°、22.8°、24.2°、25.2°、26.4°.
(S)-7-(2-メトキシ-3,5-ジメチルピリジン-4-イル)-1-(テトラヒドロフラン-3-イル)-1H-ピラゾロ[4,3-c]キノリン-4(5H)-オン・一マレイン酸塩の合成
(S)-7-(2-メトキシ-3,5-ジメチルピリジン-4-イル)-1-(テトラヒドロフラン-3-イル)-1H-ピラゾロ[4,3-c]キノリン-4(5H)-オン(500mg)に酢酸エチル(2mL)を加え25℃にて20分間撹拌した。マレイン酸(223.0mg)を酢酸エチル(7.5mL)にて完全溶解させた溶液を先の懸濁液に加え25℃にて7日間撹拌した。固体をろ取して、酢酸エチル(1mL)にて結晶洗浄し、40℃にて減圧乾燥することにより、表題化合物(366.9mg)を白色固体として得た。
得られた標記化合物は、実施例1と同一の粉末X線回折ピークを示した。
PDE9阻害活性試験
1)ヒトリコンビナントPDE9タンパク質の調製
GenBankデータベースに登録されているhsPDE9A1の塩基配列(Accession No.:AF048837)を基に、以下の配列(北海道システム・サイエンス社)をプライマーとして、また、Human hippocampus cDNAライブラリー(CLONTECH社)を鋳型DNAとして、Pfu50 DNA polymerase(Invitrogen社)を用いて、以下の条件のポリメラーゼチェーンリアクション(PCR)によりhsPDE9A1cDNAフラグメントを増幅した。
hPDE9-1プライマー:AGGATGGGATCCGGCTCCTCCA(配列番号1)
hPDE9A-3プライマー:CAGGCACAGTCTCCTTCACTG(配列番号2)
PCRの条件:[96℃、5分]×1サイクル、[(96℃、10秒)、(57℃、5秒)、(72℃、2分)]×30サイクル。
[3H]-cGMP(1mCi/mL)を0.5μCi/mLの濃度で含むバッファーD(40mmol/L Tris-HCl、pH7.4、10mmol/L MgCl2、1mM DTT、2μM cGMP)溶液100μLに、氷冷下にて評価化合物溶液(化合物をDMSOに溶解し、DMSO濃度が5%となるように希釈したもの)10μL及び上記で調製したPDE9酵素溶液をバッファーE(40mmol/L Tris-HCl pH7.4、10mmol/L MgCl2、1mM DTT、1mmol/L EGTA)にて希釈した溶液90μLを加えた。この混合溶液を30℃にて10分間インキュベートした後、沸騰水中で2分間加熱することにより、PDE9の酵素反応を停止させた。次に、室温に戻して、50μLの5’-Nucleotidase(BIOMOL社、10units/mL)を加え、30℃にて10分間インキュベートすることにより、先の反応で生成した[3H]-5’-GMPを[3H]-グアノシンに変換した。この反応液に、陰イオン交換樹脂(Bio-Rad AG1-X2 resin、mesh size 200-400、H2O:resin=2:1)を500μL添加し、10分間の放置後に遠心分離(2000rpm、10分)し、[3H]-グアノシンの存在する上清をLumaPlate(パーキンエルマー社)に移し、放射活性をTopCount NXTマイクロプレートシンチレーション・ルミネッセンスカウンター(パーキンエルマー社)で測定した。

また、評価化合物のPDE9に対するIC50値は、種々の濃度における抑制率より求めた。参考例2に準じて合成した化合物(I)のPDE9に対するIC50値は、0.00943μMであった。
ICR系雄性マウス(日本チャールズリバー社)、Sprague-Dawley(SD)系雄性ラット(日本チャールズリバー社)、またはLong-Evans(LE)系雄性ラット((財)動物繁殖研究所)に評価化合物投与後、ペントバルビタール麻酔下に脳脊髄液を採取し、-20℃にて保管した。脳脊髄液中のcGMPの測定はcGMP EIAキット(GE Healthcare社)のAcetylation EIA Procedure、またはcGMP EIAキット(Cayman社)のNon-acetylation Procedureにしたがって行った。結果はvehicle投与群のcGMP量(A)に対しての評価化合物投与群のcGMP量(B)の増加量(C)とし、以下の式を用いて算出した。
cGMP増加量(C)=[(B)-(A)]/(A) x 100(%)
参考例1に準じて合成した化合物(I)のcGMP増加量はLE系ラットに10mg/kg投与したとき1時間後に274%であった。
SD系雄性ラット(日本チャールズリバー社)、またはLE系雄性ラット((財)動物繁殖研究所)に評価化合物投与後、ペントバルビタール麻酔下にマイクロウェーブ処置をおこない海馬を摘出し湿重量を測定後、液体窒素にて凍結し-80℃にて保管した。海馬中cGMPの測定では、湿重量をもとに5%(w/v)となるように0.5M過塩素酸/1mM EDTA溶液を添加し破砕した。破砕後、遠心分離(10000rpm、15分)をおこない、上清を採取した。採取した上清を2M炭酸水素カリウム溶液により中和し、遠心分離(13000rpm、10分)をおこなった。上清中のcGMP濃度をcGMP EIAキット(GE Healthcare社)のNon-acetylation EIA Procedureにしたがって行った。結果はvehicle投与群のcGMP量(A)に対しての評価化合物投与群のcGMP量(B)の増加量(C)とし、以下の式を用いて算出した。
cGMP増加量(C)=[(B)-(A)]/(A) x 100(%)
参考例1に準じて合成した化合物(I)のcGMP増加量はLE系ラットに10mg/kg投与したとき1時間後に58%であった。
化合物(I)(50mg)、実施例1の化合物(30mg)、実施例2の化合物(30mg)をそれぞれ等重量のラクトース一水和物とともにヒドロキシプロピルメチルセルロースカプセルに充填した。溶出試験器にはAgilent Technologies社製708‐DSに小型撹拌パドルと小型ベッセルを装着したものを用いた。それぞれの薬物充填カプセルを37℃に加温した50mLの絶食下ヒト小腸シミュレート液(fasted state simulated intestinal fluid、0.75mMのレシチンと3mMのタウロコール酸ナトリウムを含むpH6.5のリン酸緩衝液)に投入し、撹拌パドルを50rpmの速度で回転させて薬物を溶出させた。経時的に溶出液をサンプリングし、HPLCにより薬物濃度を測定した。このような絶食下ヒト小腸シミュレート液を使用した溶出試験は薬物の溶解性および吸収性の評価によく用いられている(たとえば、Takano et al., “Oral absorption of poorly water-soluble drugs: computer simulation of fraction absorbed in humans from a miniscale dissolution test”, Pharm Res., vol. 23,pp.1144-1156, 2006.)。結果を表1に、グラフを図12に示す。化合物(I)を用いた場合の化合物濃度に対して、実施例2の化合物を用いた場合は化合物濃度が約2倍、実施例1の化合物を用いた場合は化合物濃度が約4~5倍高い値を示した。
化合物(I)(100mg)、実施例1の化合物(130mg:一マレイン酸塩、フリー体換算で100mg)をそれぞれヒドロキシプロピルメチルセルロースカプセルに充填したものを投与検体とした。4頭のビーグル犬に対してカプセル剤を少量の水と共に投与後、0.5、1、2、4、6、8、24時間の時点で血液を採取した。遠心分離により得た血漿中の薬物濃度はLC-MS/MSにより測定した。一週間の休薬期間を置き、同一個体に対して化合物(I)および実施例1の化合物についてそれぞれクロスオーバーで投与を行い、血漿中の薬物濃度の推移を比較した。結果を表2に、グラフを図13に示す。BQLは定量限界以下を、NCは未計算を意味する。血漿中薬物濃度-時間曲線下面積(AUC)の値を表3に示す。化合物(I)を投与した場合の平均AUC値は1.88±0.95μg・h/mLであったのに対し、実施例1の化合物を投与した場合の平均AUC値は4.00±0.45μg・h/mLであり、実施例1の化合物の方が吸収率が高く、数値のばらつきも小さかった。
したがって、本発明の塩・結晶は医薬品の原料として好ましい溶出性および経口吸収性が示された。
Claims (7)
- 塩酸、臭化水素酸、硫酸、硝酸、リン酸、マロン酸、マレイン酸、酒石酸、メタンスルホン酸、ベンゼンスルホン酸およびトルエンスルホン酸からなる群から選択される一つの酸と、(S)-7-(2-メトキシ-3,5-ジメチルピリジン-4-イル)-1-(テトラヒドロフラン-3-イル)-1H-ピラゾロ[4,3-c]キノリン-4(5H)-オンと、からなる塩。
- (S)-7-(2-メトキシ-3,5-ジメチルピリジン-4-イル)-1-(テトラヒドロフラン-3-イル)-1H-ピラゾロ[4,3-c]キノリン-4(5H)-オン・一マレイン酸塩。
- (S)-7-(2-メトキシ-3,5-ジメチルピリジン-4-イル)-1-(テトラヒドロフラン-3-イル)-1H-ピラゾロ[4,3-c]キノリン-4(5H)-オン・一ベンゼンスルホン酸塩。
- 請求項1記載の塩の結晶。
- 粉末X線回折において、回折角度(2θ±0.2°)10.1°に回折ピークを有する、(S)-7-(2-メトキシ-3,5-ジメチルピリジン-4-イル)-1-(テトラヒドロフラン-3-イル)-1H-ピラゾロ[4,3-c]キノリン-4(5H)-オン・一マレイン酸塩の結晶。
- 粉末X線回折において、回折角度(2θ±0.2°)9.9°に回折ピークを有する、(S)-7-(2-メトキシ-3,5-ジメチルピリジン-4-イル)-1-(テトラヒドロフラン-3-イル)-1H-ピラゾロ[4,3-c]キノリン-4(5H)-オン・一ベンゼンスルホン酸塩の結晶。
- 請求項1記載の塩を有効成分として含有する医薬組成物。
