細胞融合に用いられるミエローマ細胞としては、一般的にはマウスから得られた株化細胞、例えばP3-U1、NS-1、SP-2、653、X63、AP-1などを使用することができる。
また、本発明のモノクローナル抗体又はその抗体断片は、血管内皮リパーゼに対して特異的結合性を有するので、脂質代謝異常症、高脂血症、動脈硬化、アテローム性動脈硬化、高コレステロール血症、高トリグリセリド血症、糖尿病、肥満および/またはシンドロームXの診断にも応用可能である。
pShuttleベクター(クロンテック社製)に、PCR法、制限酵素を用いた手法により、C2タグを付加したヒヒELcDNA配列をクローニングした。このサブクローンベクターと、アデノウイルス骨格遺伝子を有するベクターを、PI-SceIおよびI-CeuI酵素により消化した(Adeno-x Accessory Kit, クロンテック社製)。消化断片のライゲーション反応を16℃、3時間行い(ligation high, 東洋紡社製)、ライゲーション産物を大腸菌株(ワンショットStbl3ケミカルコンピテントセル, インビトロジェン社製)にトランスフォームした。アンピシリン選択後、得られたクローンよりプラスミドを精製し(QIAprep spin Miniprep Kit, QIAGEN社製)、次いでPacI酵素(New England Biolabs社製)処理による大腸菌生育領域の切り出しを行った。以上により、アデノウイルスベクター作製のためのプラスミドDNAを得た。得られたプラスミドDNAをHEK293細胞(ATCCより入手)にLipofectamine2000(インビトロジェン社製)を用いてトランスフェクションし、10%ウシ血清含有DMEM培地中、37℃、5% CO2下で培養した。トランスフェクション後は5日おきに培地交換を行い、細胞変性効果(CPE)が確認されるまで培養を継続した。CPEの確認後、培養上清および細胞を回収した。回収した細胞は、ドライアイス・メタノール浴と温水浴を用い凍結融解を5回程度繰り返した後、3000rpmにて15分遠心して得られた上清を、細胞抽出液として回収した。この細胞抽出液と、培養上清を混合し、ウイルスベクター含有液とした。このウイルスベクター含有液をHEK293細胞に添加し、同様の操作を繰り返すことにより、ウイルスベクターの増幅を行った。増幅後、最終的に得られた細胞抽出液を、Benzonase(Merck-Novagen社製)で37℃、30分間処理後遠心し、得られた上清を、以下の密度勾配遠心によるウイルスベクター精製に用いた。1.5、1.35、1.25g/cm3の塩化セシウムを含むPBS(Phosphate buffered saline)を遠心管内に重層し、次いで上記上清を重層した。これを35000rpm, 16℃で1時間遠心し、目視によって得られたウイルスベクターを回収した。回収したウイルスベクターは、10%グリセロール含有PBSに対して透析を行い、精製アデノウイルスベクターとした。ウイルスベクターは一部をタイター測定(Adeno-X rapid titer kit, クロンテック社製)および自己増殖能獲得型出現判定に用い、異常のないもののみを以下の免疫に使用した。
C2タグが付されたヒヒELのアミノ酸配列は、配列番号2に記載した。
C2タグが付されたヒヒELをコードするDNAをクローニングしたpcDNA3.1(インビトロジェン社製)をFreeStyle HEK293F細胞にトランスフェクションし、37℃、8% CO2で3日間培養した。培養液を遠心し、細胞を回収して、20U/mLのヘパリンを含むPBSにて懸濁し、37℃で45分間インキュベーションした。その後、遠心により細胞を除去して得た上清をヒヒELヘパリン抽出液とした。
同様の方法で、C2タグが付された、カニクイザルEL、ヒト(1-157)マウスキメラEL、ヒト(1-53)マウスキメラEL、ヒト(61-111)マウスキメラEL、ウサギEL、マウスEL、ヒト(1-305)マウスキメラELおよびヒト(202-500)マウスキメラELのヘパリン抽出液を調製した。
C2タグが付された、カニクイザルEL、ヒト(1-157)マウスキメラEL、ヒト(1-53)マウスキメラEL、ヒト(61-111)マウスキメラEL、ウサギEL、マウスEL、ヒト(1-305)マウスキメラELおよびヒト(202-500)マウスキメラELのアミノ酸配列は、それぞれ配列番号2、3、4、5、6、7、96、97に記載した。
精製したヒヒEL発現アデノウイルスベクター2×109i.f.u.相当を、6週齢の雌マウス(A/J Jms Slc種、日本エスエルシーより入手)の静脈内、皮下、または後肢筋肉内に投与した。投与後は、7日毎に尾静脈より血液を採取し、これより調製した血清を用いて抗体価の測定を行った。また、初回のアデノウイルスベクター投与に加えて、追加免疫として同量のアデノウイルスベクターを静脈内、皮下、又は後肢筋肉内に、投与した。抗体価の上昇を認めたマウスについて、最終免疫として、尾静脈よりブースター免疫を実施した。
最終免疫の3日後に、ヒヒELに対する抗体価の上昇が認められた個体について開腹し、脾臓を摘出し、脾臓細胞を回収した。脾臓細胞とマウスミエローマ細胞(p3×63-Ag8.U1、東京腫瘤研究所)を50%のポリエチレングリコール4000を用いて融合させ、ヒポキサンチン、アミノプテリン、及びチミジンを含む培地で選択した。
細胞融合10日後に特異抗体産生細胞のスクリーニングを行った。スクリーニングに用いたELSIAは以下の通りである。384穴マイクロタイタープレート(ヌンク社製)の各ウェルに0.35μgの抗マウスIgG-Fc抗体(Jackson Immuno Research社製)を含むトリス緩衝液(50mM Tris-HCl、pH7.5)を35μl加えて4℃、16時間固定した。これらのウェルを90μlの洗浄液(0.01% Tween20を含む生理食塩水)で1回洗浄した後、ブロックエース(大日本製薬社製)を100μl加えて室温で2時間放置して、ブロッキングを行った(抗マウスIgG-Fc抗体固相化プレート)。各ウェルを90μlの洗浄液で1回洗浄した後、15μlのハイブリドーマ培養上清を加えて2時間放置した。これらのウェルを90μlの洗浄液で3回洗浄した後、15μlのヒヒELヘパリン抽出液を含むアッセイバッファー(4%ブロックエース、0.05% Tween20、150mM NaClを含む50mMトリス緩衝液pH7.4)を加えて、4℃で16時間放置した。これらのウェルを90μlの洗浄液で3回洗浄した後、15μlのビオチン標識抗C2タグ抗体(Fab)とHRP標識ストレプトアビジン(Thermo scientific社製)を含むアッセイバッファーを加え、室温で1時間放置した。これらのウェルを90μlの洗浄液で3回洗浄した後に、15μlのTMB+-Substrate-Chromogen(DAKO社製)を添加して室温で30分間発色させた後に、15μlの0.05Mの硫酸を添加し反応を停止し、450nmにおける吸光度を測定した。
スクリーニングの結果、ヒヒELと結合したハイブリドーマのクローンを11個(55A1、7D4、14A1、2D5、53A11、13B3、23H8、16B3、16E7、14E1および19E7)得た。55A1、7D4、14A1、2D5、53A11、13B3、23H8、16B3、16E7、14E1および19E7のクローンが産出する抗体を、それぞれ55A1、7D4、14A1、2D5、53A11、13B3、23H8、16B3、16E7、14E1および19E7抗体と命名した。マウスモノクローナル抗体アイソタイピングキット(BD Bioscience)を用いて、抗体のサブクラスを決定した。
抗体のサブクラスは、55A1抗体がIgG2a、7D4抗体がIgG1、14A1抗体がIgG1、2D5抗体がIgG2a、53A11抗体がIgG2a、13B3抗体がIgG2a、23H8抗体がIgG2a、16B3抗体がIgG2a、16E7抗体がIgG1、14E1抗体がIgG1、19E7抗体がIgG2aであった。
55A1、7D4、14A1、2D5、53A11、13B3、23H8、16B3、16E7、14E1および19E7の各抗体について、実施例2で調製したヒヒEL、カニクイザルEL、ヒト(1-157)マウスキメラEL、ヒト(1-53)マウスキメラEL、ヒト(61-111)マウスキメラEL、ウサギEL、マウスEL、ヒト(1-305)マウスキメラELまたはヒト(202-500)マウスキメラELに対するEL活性阻害能を測定した。具体的な方法を以下に示す。
20mM トリス塩酸緩衝液(pH7.5)、0.5%ウシ血清アルブミン、4mM 塩化カルシウム、150mM 塩化ナトリウム、2mg/mLヒトHDL(Athens Research&Technology社製)で構成される反応溶液にEL抗体を添加した後に、EL酵素溶液を添加した(全量で10μl )。37℃で90分の反応後、EL酵素によってHDLから生成される遊離脂肪酸(NEFA)をNEFA C-テストワコー(和光純薬工業社製)で測定し、そのNEFA量を酵素活性指標とした。EL抗体を添加しない時の酵素活性をコントロール値とし、抗体の各濃度でのコントロール値に対する比活性を算出してプロットした(図1A~図1K)。図1A~図1Kの阻害曲線より、各抗体の50%阻害濃度(IC50値)を求めた。
ヒトHLをコードするDNAをクローニングしたpcDNA3.1(インビトロジェン社製)をFreeStyle HEK293F細胞にトランスフェクションし、37℃、8%CO2で2日間培養した。培養液を遠心し、細胞を回収して、20U/mLのヘパリンを含むPBSにて懸濁し、37℃で45分間インキュベーションした。その後、遠心により細胞を除去して得た上清をヒトHL酵素溶液とした。同様の操作により、ヒトLPL酵素溶液を調製した。20mM トリス塩酸緩衝液(pH7.5)、0.5%ウシ血清アルブミン、4mM 塩化カルシウム、150mM 塩化ナトリウム、0.5mg/mLヒトVLDL(INTRACEL社製)で構成される反応溶液にEL抗体溶液を添加した後、ヒトHL、またはヒトLPL酵素溶液を添加した(全量で10μl)。37℃で2時間反応後、HLもしくはLPL酵素によって、VLDLから生成される遊離脂肪酸(NEFA)をNEFA C-テストワコー(和光純薬工業社製)で測定し、そのNEFA量を酵素活性指標とした。EL抗体を添加しない時の酵素活性をコントロール値とし、コントロール値に対する比活性を算出した。比較のため、結果をカニクイザルELの阻害曲線と並べて記載した(図2A~図2K)。
その結果、55A1、7D4、14A1、2D5、53A11、13B3、23H8、16B3、16E7、14E1および19E7抗体全てが、HLとLPLの酵素活性を阻害しないことがわかった。
抗マウスIgG-Fc抗体固相化プレートに、15μlのEL抗体(1μg/mLから3倍希釈系列)を含むアッセイバッファー加えて2時間放置した。これらのウェルを90μlの洗浄液で3回洗浄した後、15μlのカニクイザルEL、ヒヒEL、ウサギEL、マウスEL、ヒト(1-157)マウスキメラEL、ヒト(1-305)マウスキメラELまたはヒト(202-500)マウスキメラELヘパリン抽出液を含むアッセイバッファーを加えて、4℃で16時間放置した。これらのウェルを90μlの洗浄液で3回洗浄した後、15μlのビオチン標識抗C2タグ抗体(Fab)とHRP標識ストレプトアビジンを含むアッセイバッファーを加え、室温で1時間放置した。これらのウェルを90μlの洗浄液で3回洗浄した後に、15μlのTMB+-Substrate-Chromogen(DAKO社製)を添加して室温で30分間発色させた後に、15μlの0.05Mの硫酸を添加し反応を停止し、450nmにおける吸光度を測定した。測定結果を、図3A~図3Kに示した。
55A1、7D4、14A1、2D5、53A11、13B3および23H8抗体は、カニクイザル、ヒヒ、ヒト(1-157)マウスキメラELに結合するが、マウスELには結合しないことが示された。また、13B3および23H8抗体はウサギELに結合するが、55A1、7D4、14A1、2D5および53A11抗体はウサギELに結合しないことが示された(図3A~図3G)。
一方、16B3、16E7、14E1および19E7抗体は、カニクイザルEL、ヒヒEL、ヒト(1-305)マウスキメラELおよびヒト(202-500)マウスキメラELに結合するが、マウスELおよびヒト(1-157)マウスキメラELに結合しないことが示された。また、16B3、14E1および19E7抗体は、ウサギELに結合するが、16E7抗体はウサギELに結合しないことが示された(図3H~図3K)。
各EL抗体の結合親和性をBiacoreを用いて測定した。抗C2タグ抗体をSensor chip CM5(GE HealthCare社製)にアミンカップリング法により固定化し、HBS-P(GE HealthCare社製)で希釈したヒヒELヘパリン抽出液、カニクイザルEL、もしくは、ヒト(1-305)マウスキメラELヘパリン抽出液を添加し、Sensor ChipにELをトラップした。その後、HBS-Pで希釈したEL抗体を添加し、BIAevaluation softwareを用いて、Bivalent fittingにより結合親和性を算出した。各抗体のヒヒELに対する結合親和性を表1に示し、カニクイザルELに対する結合親和性を表2に示し、ヒト(1-305)マウスキメラELに対する結合親和性を表3に示した。
EL抗体の結合に重要なELのアミノ酸残基を決定するために、ヒヒELのアミノ酸残基に変異を加え、EL抗体の結合をELISAにて調べた。具体的には、Quikchange Site-Directed Mutagenesis Kit(アジレント・テクノロジー株式会社)を用いてヒヒELのcDNAに変異を加え、アラニンに置換した。ただし、ヒヒELとマウスELでアミノ酸残基が異なる場合は、マウスELのアミノ酸残基に置換した。変異を導入したヒヒELを発現するプラスミドDNAを用いて、実施例2と同様の方法でヘパリン抽出液を調製し、実施例8と同様のELISA法で抗体との結合を調べた。ヒヒELのアミノ酸残基の相対的な寄与率を評価するために、相対的な結合活性([A450が1.0となる抗体濃度_変異型EL]/[A450が1.0となる抗体濃度_野生型EL])を算出した。変異を導入したヒヒELの、55A1、7D4、14A1、2D5、53A11、13B3および23H8抗体に対する相対的な結合活性を図7A-Gに示した。また、変異を導入したヒヒELの、16B3、16E7、14E1および19E7に対する相対的な結合活性を図7H-Lに示した。
EL抗体について、常法を用いて可変領域の配列を決定し、図4A~図4Lに示した。
55A1、7D4、14A1、2D5、53A11、13B3および23H8抗体の重鎖および軽鎖のアミノ酸配列を遺伝子解析ソフトGENETYXを使用してアライメントした(図5Aから図5D)。
アライメントの結果、55A1、7D4、14A1および2D5抗体の重鎖および軽鎖の各CDRのアミノ酸配列が類似しており、残りの53A11、13B3および23H8抗体の重鎖のCDR1およびCDR2のアミノ酸配列が類似していることを見出した。
重鎖のCDR1は、NYGMNの5アミノ酸配列からなっていた。
重鎖のCDR2は、WINTYSGVPTY(AorT)(GorD)DFKGの17アミノ酸からなっていた。
重鎖のCDR3は、(RorF)(GorS)YYGR(RorH)YFD(VorY)の11アミノ酸配列からなっていた。
軽鎖のCDR1は、KASQSVDYD(VorG)DSYM(HorN)の15アミノ酸配列からなっていた。
軽鎖のCDR2は、AASNLXSの7アミノ酸配列からなっていた。Xは任意のアミノ酸を意味している。
軽鎖のCDR3は、(HorQ)Q(TorS)(IorTorN)(EorD)DPP(- orW)Tの9または10アミノ酸配列からなっていた。- はアミノ酸が欠失していることを意味している。
重鎖のCDR1は、(DorE)(YorN)T(MorI)Hの5アミノ酸配列からなっていた。
重鎖のCDR2は、XINPYYGGTX(YorN)NEKFKDの17アミノ酸からなっていた。Xは任意のアミノ酸配列を意味している。
重鎖のCDR3および軽鎖の各CDRには、アミノ酸配列の共通性が認められなかった。
16B3、16E7、14E1および19E7抗体の重鎖および軽鎖のアミノ酸配列を遺伝子解析ソフトGENETYXを使用してアライメントした(図6Aから図6D)。
アライメントの結果、16B3と16E7抗体の重鎖および軽鎖の各CDRのアミノ酸配列が類似していることを見出した。
重鎖のCDR1は、(GorS)YTMSの5アミノ酸配列からなっていた。
