Definitions
- the present invention relates to an autosampler, and more particularly to an autosampler for injecting a sample into a sample concentrator using a trap column or an analyzer such as a liquid chromatograph equipped with such a sample concentrator.
- the present invention relates to a control technique of an autosampler for injecting a single component fractionated by a preparative LC (liquid chromatograph).
- Concentration with a trap column is often used because concentration can be performed using a vacuum evaporator, but additives can be removed by using a removal column in combination (see Patent Document 1).
- Dilution can also be performed when preparing the sample, but if the sample volume after dilution becomes too large, the processing becomes difficult, so online dilution is performed.
- online dilution a flow path similar to the flow path configuration in the sample concentrator as shown in FIGS. 1 and 4 is used as an example.
- the high pressure valve 2 is switched to the injection position in which the sample loop 4 is incorporated in the main flow path including the sample push section 5 and the trap column 8, and in FIG. 4, the high pressure valve 2 is switched to the syringe pump 3 and the sampling.
- the metering position including the needle 30 is switched to the metering position in which the sample loop 4 is incorporated.
- the high pressure valve 2 is switched from the idling state (the high pressure valve 2 is the same as the injection position in FIG. 1) to the sample weighing state in FIG.
- the sample is sucked and introduced into the sample loop 4.
- the high pressure valve 2 is switched to the injection state of FIG. 1, and the sample in the sample loop 4 is pushed out of the sample loop 4 by the mobile phase sent from the sample push unit 5.
- the sample is diluted by being mixed with a diluent (usually the same composition as the mobile phase) sent from the make-up unit 6 in the three-way joint 7 of the main flow path, and is guided to the trap column 8 and trapped. .
- a diluent usually the same composition as the mobile phase
- FIG. 3 shows an example of fractionation of two component peaks by the fractionation system.
- the peak of component B (retention time 5.8 minutes) overlaps with tailed component A (retention time 5.7 minutes).
- the vial capacity reaches the upper limit in 0.1 minutes.
- fraction 9 since fraction 9 has reached the maximum fraction volume of vial # 1 of 10 mL, it switches to fraction 10 and then rises to the peak of component B before reaching the maximum fraction volume of vial # 2 of 10 mL. Is detected and switched to the fraction 11, and when the falling edge of the peak of the component B is detected, it can be seen that the screen is switched to the fraction 12.
- fractions 9, 10, and 12 which are single components, are injected as samples into a concentration system using a trap column and concentrated.
- sample injection and analysis processing are programmed in a table called a batch table (sometimes referred to as a sequence table or a sample set).
- a batch table sometimes referred to as a sequence table or a sample set.
- One row of the batch table corresponds to one analysis, and each row includes the sample vial position, sample volume, and other analysis conditions.
- analysis conditions include an analysis initial parameter set and an injection pre-processing program that customizes the operation of the autosampler 1 (usually called a pre-processing program. If not customized, a standard pre-processing operation is programmed internally.
- a time program (sometimes referred to as an event or an event program), etc., and is usually stored in a parameter set called a device method.
- the preprocessing program may be specified by a batch table independently of the device method.
- the processing of one line of the batch table consists of downloading the sample vial position, sample volume, and other analysis conditions to the device (downloading), executing the preprocessing program (preprocessing execution), executing the time program and chromatogram It is performed in the order of recording start (analysis execution).
- injection means that, after introducing one sample into the sample loop 4 by the pretreatment program, the high-pressure valve 2 is switched to the injection position, and the sample loop 4 is connected to the main flow path.
- an event signal electrical signal, communication command, software command
- injection means that, after introducing one sample into the sample loop 4 by the pretreatment program, the high-pressure valve 2 is switched to the injection position, and the sample loop 4 is connected to the main flow path.
- an event signal electrical signal, communication command, software command
- the “injection” process of one sample is performed by the pre-processing program, the sample push unit 5 and the makeup unit 6 are controlled by the time program, and the sample is fixed to the trap column 8.
- an event signal is generated to start the transfer of the mobile phase and the diluent by the sample push unit 5 and the make-up unit 6 in the time program.
- the high-pressure valve 2 is switched after outputting and waiting for the liquid to stabilize.
- “loading” means that the high pressure valve 2 is not switched after the sample is introduced into the sample loop 4 by the preprocessing program.
- “loading” means that the high pressure valve 2 is not switched after the sample is introduced into the sample loop 4 by the preprocessing program.
- an event signal is output by the preprocessing program and the time program is started, but the sample push unit 5 and the makeup unit 6 do not need to operate. Therefore, the time program ends immediately.
- the time program is completed promptly, even the autosampler 1 that cannot output an event signal unless the high-pressure valve 2 is switched can perform processing equivalent to “loading” in the “injection” processing, but the detailed explanation here is omitted. To do.
- the device method for sample push up to the N-1th row and the device method for injection of the Nth row are prepared and combined.
- the sample push device method of fractions 9 and 10 one sample is “loaded” by the preprocessing program, and in the injection device method of fraction 12, one sample “ Injection "until the sample is fixed to the trap column 8.
- the processing content of the device method for injection is the same as the processing content of the device method of (a).
- the samples of fractions 9 and 10 are sent to the trap column 8 together with the sample of fraction 12.
- the preprocessing program performs “loading” of a fixed capacity of 1 sample and an indefinite capacity of 1 sample (capacity is specified in the batch table), and fraction 12
- injection of an indefinite volume of one sample (the volume is specified in the batch table) is performed by the preprocessing program, and the sample is fixed to the trap column 8.
- the processing content of the device method for injection is the same as the processing content of the device method of (a).
- Fractions 9, 10, and 12 are programmed as 1 row, 2 loading, and 1 injection, and the volumes of fractions 9, 10, and 12 are individually set by the autosampler pre-processing program included in the instrument method. Customize to “load” and “inject”.
- the apparatus method performs “loading” for each of the samples of fractions 9 and 10 in the preprocessing program, performs “injection” for the sample of fraction 12, and performs fixing until the trap column 8 is fixed.
- the processing content after the introduction into the sample loop 4 is the same as the processing content of the apparatus method of (a).
- the make-up unit 6 needs to send a diluting solution several times the actual sample injection amount.
- the “actual sample injection amount” introduced into the sample loop 4 and injected into the main channel is the total sample volume when no special pretreatment is required, and pretreatment such as mixing of additives is performed. In this case, it is a volume obtained by adding the volume of the additive to the total sample volume, and it is difficult to specify in advance. For this reason, normally, instead of “actual sample injection amount”, the “maximum sample injection amount” is defined from the volume of the sample loop 4 and the number of injections into the main flow path, the maximum fraction volume, and the maximum number of fraction vials. .
- liquid chromatographic analysis The main purpose of liquid chromatographic analysis is separation and analysis, and the retention time of the sample does not depend on the sample volume, and only the tailing factor that determines the elution range of the sample depends on the sample volume. For this reason, when using a specific sample, the analysis time in the liquid chromatograph analysis may be regarded as constant without depending on the sample volume.
- the device control of the liquid chromatograph analyzer can be programmed with the analysis time and time program (also called event or event program) in the instrument method, but it depends on the actual sample injection volume. Control such as operating the liquid pump is not possible.
- the pump is controlled by switching the instrument method prepared for each sample volume range and controlling the pump at the time calculated for the maximum sample injection volume. Is used.
- the maximum sample injection volume is 50 mL, the dilution rate is 5 times, and no additives are mixed, 250 mL of solvent (mobile phase + diluent) is required per sample, of which 225 mL of solvent is wasted. It will be consumed.
- fractionation volume of the sample concentrated by the trap column and the number of fractions depend largely on the concentration of the sample, the retention time, and the degree of separation from other components, and cannot be determined in general. The reason is as follows.
- the retention time can be changed by changing the mobile phase composition at the same flow rate.
- the number of fractions of a single component peak varies depending on the liquid feeding conditions.
- the fraction of single component peaks also varies from sample to sample.
- the fraction volume and number of fractions excluding interfering component peaks of the multi-component peak are also different from sample to sample. This is because the peak width and peak overlap differ depending on the sample.
- the sample volume is not known, and in some cases it evaporates or falls below the volume that can be aspirated with an autosampler (the solvent always remains at the bottom of the vial when aspirated) Therefore, it is difficult for the user to input the sample volume appropriately, and therefore troubles such as injecting bubbles also occur.
- the sample concentration method described above is an example, and the method for specifying the fraction volume and the number of fractions to be concentrated by the trap column and the method for controlling the liquid feed pump according to the actual sample injection amount are described in the present invention.

- Autosampler for liquid chromatographic analysis by automatically calculating the injectable sample volume of the specified fraction based on the fractionation conditions (flow rate), the fractionation result (vial position and fractional bandwidth), and the sample residual volume setting. Can automatically create a batch table used in
- FIG. 1 a method for detecting the sample volume with the detection probe 22 in the autosampler 1 is shown in FIG.
- the detection probe 22 uses the sampling needle 30 as a sensor for detecting the sample volume, and detects the sample liquid level based on changes in capacitance and dielectric constant.
- the autosampler 1 receives the output of the liquid level detection probe 22 integrated with the sampling needle 30 and performs amplification and conversion by a liquid level detection circuit 23 comprising an amplifier, a comparator, an AD converter, etc., and the physical properties of air and the sample solvent It has a liquid level detector that outputs changes (mainly changes in capacitance and dielectric constant).
- the height of the liquid surface detected by the detection probe 22 (detection sample volume), the height setting of the needle at the limit of sample suction (sample residual liquid amount setting), and the shape of the sample vial / test tube
- the sample volume that can be injected is determined by setting.
- the injectable volume calculation unit 56 of the control unit 50 of the autosampler 1 takes in the sample volume detected by the liquid level detection probe 22 and calculates the injectable sample volume for each vial 32.
- the injection / liquid feeding control unit 52 instructs the syringe pump 3 so that the syringe pump 3 sucks the sample of the vial 32 within the range of the injectable capacity calculated by the injectable capacity calculation unit 56 for each vial 32. It is configured to issue.
- the sample volume specified by the batch tables 17 and 21 is set as the upper limit, and injection calculated based on the detection signal of the detection probe 22 is possible. Aspirate the sample volume. Therefore, the maximum vial volume or the maximum fraction volume is designated as the sample volume of the batch table used in the liquid chromatograph analysis autosampler.

Abstract
