Abstract
본 발명은 말초혈액 단핵세포(peripheral blood mononuclear cell)를 유효성분으로 포함하는 자궁내막 이상에 의한 불임의 예방 또는 치료용 약제학적 조성물, 자궁내막 증식용 조성물 및 자궁내막 증식방법에 관한 것으로, 본 발명의 말초혈액 단핵세포는 자궁내막 상피세포의 비약적인 증식을 통해 얇은 자궁내막의 조직 두께를 증가시키며, 이로 인해 불임으로 이어지는 수정란의 착상 실패를 효과적으로 예방 또는 개선하는 데에 유용하게 이용될 수 있다.
Description
본 발명은 자궁내막 이상에 의한 불임의 예방 또는 치료용 조성물에 관한 것이다.
자궁내막은 포유류의 자궁 내벽을 이루는 층으로, 발정호르몬(에스트로겐)과 황체호르몬에 반응하고, 성주기(발정주기, 월경주기)에 따라 현저한 변화를 나타낸다. 이러한 자궁내막은 수정란이 착상하는 공간으로, 건강한 임신을 위해서는 자궁내막의 상태가 매우 중요하다. 즉, 자궁내막의 두께가 너무 얇은 경우, 또는 표면의 요철이 심하거나 유착되어 있는 경우에는 착상이 제대로 이루어지지 않아, 임신이 되지 않거나 초기유산이 될 수 있다. 이 중 자궁내막의 두께는 가장 중요한 요인으로, 임상적으로 건강한 임신을 위한 자궁내막의 두께는 최소 6mm로 알려져 있으며, 그보다 얇으면 수정란이 착상할 공간 자체가 없어지기 때문에 정상적인 임신이 이루어지지 않는다. 가장 높은 임신확률을 가지는 자궁내막의 두께는 8-14mm이다.
자궁내막이 얇아지는 데는 크게 두 가지 요인이 있다. 첫째는 인공유산이나 소파수술로 대표되는 기계적 손상으로, 자궁내막을 긁어내는 기계적 자극으로 인해 자궁내막을 얇게 만들거나 유착을 일으킬 수 있다. 둘째는 호르몬의 이상으로, 내막을 증식시키고 이를 유지시키는 호르몬 기능이 약해지면서 자궁내막이 얇아질 수 있다. 자궁내막은 호르몬의 영향을 받으면서 월경주기에 따라 변한다. 월경기에는 자궁내막이 탈락하고, 월경 이후 배란 전까지(증식기)는 에스트로겐의 영향으로 자궁내막이 증식하며, 배란기 이후(분비기)에는 프로게스테론의 영향으로 자궁내막선 상피세포의 분비기능 증가가 현저하게 나타난다. 중요한 것은 배란기 때의 자궁내막 두께로서, 자궁내막의 두께에 영향을 미치는 호르몬의 이상에 의해 배란기의 자궁내막이 6mm 이하로 얇아지면 임신의 확률이 크게 떨어지게 된다.
현재 불임 또는 반복적 착상 실패를 개선시키기 위한 방법으로는 자궁 조직으로 유입되는 혈류량을 증가시키는 치료법과, 호르몬 기능을 정상화시켜 자궁내막의 두께를 증가시키는 방법이 시도되고 있다. 그러나, 혈류개선의 경우 자궁에 탄력을 주어 자궁경화를 완화시켜 줄 뿐 자궁내막의 두께를 근본적으로 증가시키지는 못하며, 호르몬 요법은 장기간의 투여에 따른 부작용 및 고비용의 문제와 함께 그 효과도 유의적이지 않다는 문제가 있다.
한편, 말초혈액단핵세포(peripheral blood mononuclear cell, 말초혈액단핵구, PBMC)는 혈액 내 존재하는 구형 핵을 가진 세포를 의미한다. 이러한 말초혈액단핵세포에는 B 세포, T 세포, 대식세포(macrophage), 수지상 세포(dendritic cell), 자연살해세포(NK cell, natural killer cell)등의 면역세포들이 포함되어 있으며, 최근 이러한 면역세포들의 기능을 활용하여 질병에 대한 예방 및 치료를 위한 치료제로 이용하고자 하는 연구들이 이루어지고 있다.
실제로, 최근 암 등의 질환을 치유하고자 자가 유래의 말초혈액 내 면역세포를 이용한 세포치료제 개발 연구가 활발히 진행되고 있으며, 이미 품목허가를 받은 면역세포치료제도 늘어나고 있는 추세이다. 하지만, 아직까지 자가 유래된 말초혈액단핵세포 자체를 이용하여, 착상 실패로 인한 불임을 치료하는 방법에 대해서는 알려진 바 없다.
이에, 본 발명자들은 임신적령기를 넘긴 만혼의 경향에 따라 불임인구가 점점 증가하고, 효율적인 불임치료의 수단으로서 유효한 자궁내막 증식 방법의 개발이 절실히 요구되고 있는 현 시점에서, 자가 유래된 말초혈액단핵세포를 이용하여, 불임치료의 수단으로서 효과적인 자궁내막 증식 방법을 개발하고자 하였다.
본 발명자들은 정상적인 수정란의 착상에 큰 영향을 미쳐 건강한 임신 여부를 좌우하는 자궁내막의 상태를 인위적으로 개선, 궁극적으로 자궁내막 이상에 기인한 불임의 치료법을 개발하기 위하여 예의 연구 노력하였다. 그 결과, 말초혈액 단핵세포를 이용할 경우, 자궁 조직 내 자궁내막 상피세포의 증식이 비약적으로 증가함으로써 자궁 내막의 두께 증가를 유도, 수정란의 착상 환경이 크게 개선된다는 사실을 발견함으로써, 본 발명을 완성하게 되었다.
따라서 본 발명의 목적은 자궁내막 이상에 의한 불임의 예방 또는 치료용 약제학적 조성물을 제공하는 데 있다.
본 발명의 다른 목적은 자궁내막 증식용 조성물을 제공하는 데 있다.
본 발명의 또 다른 목적은 자궁내막 증식방법을 제공하는 데 있다.
그러나 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 제한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 당업자에게 명확하게 이해될 수 있을 것이다.
상기 목적을 달성하기 위하여,
본 발명은 말초혈액단핵세포(peripheral blood mononuclear cell)를 유효성분으로 포함하는 자궁내막 이상에 의한 불임의 예방 또는 치료용 약제학적 조성물을 제공한다.
본 발명의 일 구현예로, 상기 말초혈액단핵세포는 자가 유래된 세포인 것을 특징으로 한다.
본 발명의 다른 구현예로, 상기 자궁내막 이상은 자궁내막의 두께가 7mm 이하인 것을 원인으로 하는 것을 특징으로 한다.
또한, 본 발명은 말초혈액 단핵세포(peripheral blood mononuclear cell)를 유효성분으로 포함하는 자궁내막 증식용 조성물을 제공한다.
또한, 본 발명은 말초혈액 단핵세포(peripheral blood mononuclear cell)를 대상체의 자궁 내에 투여하는 단계를 포함하는 자궁내막 증식방법을 제공한다.
본 발명의 일 구현예로, 상기 자궁내막 증식용 조성물 및 증식 방법에 이용되는 말초혈액단핵세포는 자가 유래된 세포인 것을 특징으로 한다.
또한, 본 발명은 상기 약제학적 조성물의 약제학적 유효량을 개체에 투여하는 단계를 포함하는 자궁내막 이상에 의한 불임의 예방 또는 치료방법을 제공한다.
아울러, 본 발명은 말초혈액 단색세포를 자궁내막 이상에 의한 불임의 예방 또는 치료용 조성물, 또는 자궁내막 증식용 조성물의 제조에 이용하는 용도를 제공한다.
본 발명의 특징 및 이점을 요약하면 다음과 같다:
(a) 본 발명은 말초혈액 단핵세포(peripheral blood mononuclear cell)를 유효성분으로 포함하는 자궁내막 이상에 의한 불임의 예방 또는 치료용 약제학적 조성물, 자궁내막 증식용 조성물 및 자궁내막 증식방법을 제공한다.
(b) 본 발명의 말초혈액 단핵세포는 in vitro에서 자궁내막 상피세포를 비약적으로 증식시킬 뿐 아니라, in vivo에서도 마우스 자궁내막의 조직 두께를 현저히 증가시킨다.
(c) 또한, 본 발명의 말초혈액단핵세포는 1번의 발정 주기에서 자궁내막 증식 효과를 나타내므로, 과다한 자궁증식으로 인한 질환을 유발하지 않는다.
(d) 또한, 본 발명의 자가유래된 말초혈액단핵세포는 골수로부터 유래된 중간엽 줄기세포에 비해서도 자궁내막 증식 효과가 우수할 뿐 아니라, 자가유래된 세포이므로, 이종으로부터 유래된 세포 치료제 처리시 수반되는 자가면역반응의 부작용이 발생하지 않아, 면역억제제의 병용 투여가 필요치 않다. 아울러, 본 발명의 말초혈액단핵세포는 혈액을 공급원으로 하므로, 체내 소량으로 존재하는 골수로부터 유래한 세포에 비해, 공급원의 채취가 용이할 뿐 아니라, 많은 수의 세포를 손쉽게 확보하여 치료에 응용할 수 있다.
(e) 따라서, 본 발명의 자가유래 말초혈액단핵세포는 불임으로 이어지는 수정란의 착상 실패를 효과적으로 예방 또는 개선하는 데에 유용하게 이용될 수 있다.
도 1은 본 발명의 자궁내막 상피세포 및 말초혈액 단핵세포(PBMC)의 공배양 모식도를 나타낸 그림이다.

본 발명은 말초혈액 단핵세포(peripheral blood mononuclear cell)를 유효성분으로 포함하는 자궁내막 이상에 의한 불임의 예방 또는 치료용 약제학적 조성물을 제공한다.
또한, 본 발명은 상기 약제학적 조성물의 약제학적 유효량을 개체에 투여하는 단계를 포함하는 자궁내막 이상에 의한 불임의 예방 또는 치료방법을 제공한다.

본 명세서에서 용어 "자궁내막 이상에 의한 불임"은 수정란의 정상적인 착상을 위한 자궁내막의 공간적, 기능적 및 해부학적 특성이 갖추어지지 않아 불임에 이르는 질환 또는 병적 상태(pathologic condition)를 의미한다.
본 명세서에서 용어 "자궁내막 이상"은 자궁의 해부학적 형태의 이상 및 이에 따른 기능의 이상을 의미하며, 구체적으로는 자궁내막의 두께가 비정상적으로 얇은 상태를 의미한다. 보다 구체적으로는 자궁내막의 두께가 8mm 미만인 상태를 의미하며, 보다 더 구체적으로는 7mm 이하인 경우를 의미한다.
본 발명의 바람직한 구현예에 따르면, 본 발명에서 이용되는 말초혈액 단핵세포는 자가세포이다. 자가 말초혈액 단핵세포를 이용할 경우 불필요한 자가면역반응이 배제되어 염증 등의 부작용 없이 효율적으로 자궁내막의 구조적인 안정화를 꾀할 수 있다.
본 발명의 말초혈액 단핵세포는 직접 자궁강 내로 주입될 수도 있으며, 또는 조직에서 추출한 자궁내막 상피세포를 시험관에서 말초혈액 단핵세포와 공배양한 뒤 증식된 자궁내막 상피세포를 다시 자궁조직에 이식할 수도 있다.
본 발명은 말초혈액 단핵세포의 약제학적 유효량을 포함하는 불임의 예방 또는 치료용 약제학적 조성물로 제공될 수 있다.
본 발명의 구체적인 실시예에서, 혈액으로부터 단핵구 세포를 분리하고(도 1 참조), 분리된 말초혈액단핵세포를 자궁내막 상피세포주인 SNU-685 및 SNU-1077과 공배양하여, 말초혈액단핵세포 처리에 의해, SNU-685 및 SNU-1077의 세포 수가 유의적으로 증가함을 확인하였다(도 2 내지 도 5 참조).
또한, 상기 분리된 말초혈액단핵세포를 마우스 자궁내막 조직에 직접 주입하여, 자궁 조직의 두께가 현저히 증가함을 확인하였으며(도 7 및 도 8 참조), 1번의 발정 주기에서만 자궁내막 증식 효과가 있음을 확인하여, 과다한 자궁내막 증식으로 인한 질환을 유발하지 않음을 확인하였다(도 9 참조).
아울러, 마우스 자궁내막에 말초혈액단핵세포와 골수 유래 사람 중간엽 줄기 세포를 각각 주입하여, 말초혈액단핵세포가 골수 유래 중간엽 줄기세포에 비해 자궁내막 증식 효과가 우수함을 확인하였다(도 10 및 도 11 참조).
따라서, 본 발명의 말초혈액단핵세포는 자궁 조직 내 자궁내막 상피세포의 증식을 비약적으로 증가시킴으로써 자궁 내막의 두께 증가를 유도, 수정란의 착상 환경을 크게 개선시켜, 자궁내막 이상에 의한 불임의 예방 및 치료용 조성물에 유용하게 이용될 수 있다.
본 명세서에서 용어 "약제학적 유효량"은 상술한 자궁내막 증식 활성을 달성하는 데 충분한 양을 의미한다.
본 발명의 조성물이 약제학적 조성물로 제조되는 경우, 본 발명의 약제학적 조성물은 약제학적으로 허용되는 담체를 포함한다. 본 발명의 약제학적 조성물에 포함되는 약제학적으로 허용되는 담체는 제제시에 통상적으로 이용되는 것으로서, 락토스, 덱스트로스, 수크로스, 솔비톨, 만니톨, 전분, 아카시아 고무, 인산 칼슘, 알기네이트, 젤라틴, 규산 칼슘, 미세결정성 셀룰로스, 폴리비닐피롤리돈, 셀룰로스, 물, 시럽, 메틸 셀룰로스, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 활석, 스테아르산 마그네슘 및 미네랄 오일 등을 포함하나, 이에 한정되는 것은 아니다. 본 발명의 약제학적 조성물은 상기 성분들 이외에 윤활제, 습윤제, 감미제, 향미제, 유화제, 현탁제, 보존제 등을 추가로 포함할 수 있다. 적합한 약제학적으로 허용되는 담체 및 제제는 Remington's Pharmaceutical Sciences (19th ed., 1995)에 상세히 기재되어 있다.
본 발명의 약제학적 조성물은 비경구 투여할 수 있으며, 예를 들어 피하 주입, 근육 주입, 경피 투여, 자궁강내 투여 등으로 투여할 수 있으며, 바람직하게는 복강경을 이용한 자궁강내 투여를 한다.
바람직하게는, 본 발명의 약제학적 조성물은 단회 투여 또는 반복 투여로 주입될 수 있으며, 보다 바람직하게는 반복 투여이다.
본 발명의 약제학적 조성물의 적합한 투여량은 제제화 방법, 투여방식, 환자의 연령, 체중, 성, 병적 상태, 음식, 투여 시간, 투여 경로, 배설 속도 및 반응 감응성과 같은 요인들에 의해 다양하게 처방될 수 있다. 본 발명의 약제학적 조성물의 일반적인 투여량은 성인 기준으로 1일 당 1X105 - 2X107 cells/kg(체중)이다.
본 발명의 약제학적 조성물은 당해 발명이 속하는 기술분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있는 방법에 따라, 약제학적으로 허용되는 담체 및/또는 부형제를 이용하여 제제화함으로써 단위 용량 형태로 제조되거나 또는 다용량 용기 내에 내입시켜 제조될 수 있다. 이때 제형은 오일 또는 수성매질중의 용액, 현탁액, 시럽제 또는 유화액 형태이거나 엑스제, 산제, 분말제, 과립제, 정제 또는 캅셀제 형태일 수도 있으며, 분산제 또는 안정화제를 추가적으로 포함할 수 있다.
본 발명의 다른 양태에 따르면, 본 발명은 말초혈액 단핵세포(peripheral blood mononuclear cell)를 유효성분으로 포함하는 자궁내막 증식용 조성물을 제공한다.
아울러, 본 발명은 말초혈액 단색세포를 자궁내막 이상에 의한 불임의 예방 또는 치료용 조성물, 또는 자궁내막 증식용 조성물의 제조에 이용하는 용도를 제공한다.
본 발명에서 이용되는 말초혈액 단핵세포에서는 이미 상술하였으므로, 과도한 중복을 피하기 위해 그 기재를 생략한다.
본 발명의 또 다른 양태에 따르면, 본 발명은 말초혈액 단핵세포(peripheral blood mononuclear cell)를 대상체의 자궁 내에 투여하는 단계를 포함하는 자궁내막 증식방법을 제공한다.
본 발명에서 이용되는 말초혈액 단핵세포에서는 이미 상술하였으므로, 과도한 중복을 피하기 위해 그 기재를 생략한다.
본 발명에 따르면, 말초혈액 단핵세포는 자궁내막 상피세포의 증식을 유도하므로, 자궁 내에 투여되었을 때 자궁 조직 내 상주세포(resident cell)의 증식을 촉진하여 자궁내막의 두께를 증가시킬 수 있다. 자궁 내 투여는 산부인과 및 외과적으로 통상적으로 사용되는 다양한 투여방법을 통해 수행될 수 있으며, 예를 들어 복강경(Laparoscopy)을 통해 수행될 수 있으나 이에 제한되는 것은 아니다.
상기 대상체는 자궁내막의 이상 또는 자궁내막의 두께가 7mm이하로 착상 실패에 의해 불임이 유발된 환자 또는 자궁내막 두께 증식이 필요한 환자가 될 수 있다. 상기 대상체는 인간인 것이 바람직하나, 이에 한정하지 않고 개, 쥐, 말, 소, 돼지, 양 등 자궁내막 이상으로 인해 불임이 발생할 수 있는 포유류는 어느 것이든 가능하다.
이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 하기 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
<
실시예 1> 말초혈액 단핵세포의 분리
실온에 보관한 DPBS(Dulbecco's phosphate buffered saline)를 5개의 팰콘 튜브에 25ml씩 넣어 준비하였다. 이후 1번 내지 5번 튜브에 말초혈액(발명자의 혈액을 임상의가 채취) 10m 씩을 각각 분주하였다. DPBS와 말초혈액이 담긴 각 튜브를 천천히 뒤집고 피콜(Ficoll, Ficoll-paque TM plus, GE Healthcare Lot. 10055780)을 12ml 씩 넣어주되, 바닥부터 천천히 넣어주어 각 튜브 내 피콜이 섞이지 않도록 하였다. 이후 실온(20℃)에서 2500rpm으로 30분간 원심분리를 하였다. 차가운 PBS를 팰콘 튜브에 35ml씩 넣어 준비한 뒤 5번 튜브의 맨 위 혈청 층을 석션(suction)하고, 그 아래의 단핵구(mononuclear)층을 1ml 피펫으로 수집하여 차가운 PBS가 담긴 팰콘 튜브에 각각 넣었다. 이후, 4℃에서 1700rpm으로 10분간 원심분리 후 상등액을 석션하고 침전물을 차가운 PBS에 풀어준 뒤 차가운 PBS를 가득 채우는 세척과정을 3회 반복하였다. 마지막으로 상등액을 석션하고 침전물을 α-MEM + 10% FBS + 100auf-anti+ⓟ 배양액(1ml)에 풀어준 후 배양액 4ml을 넣어 각각 총 5ml가 되도록 만들었으며, 이를 통해 말초혈액단핵세포를 분리하였다. 이후, 트리판 블루(상온 보관) 10㎕에 상기 배양액 10㎕를 섞은 다음 자동 세포 계수기로 상기 분리된 말초혈액단핵세포의 세포 수를 측정하였다.
<실시예 2> 자궁내막 상피세포주 및 말초혈액 단핵세포의 공배양
자궁내막 상피세포주인 SNU-685와 SNU-1077을 한국 세포주 은행에서 구매하여 6 웰플레이트(BD Falcon사)에 1.0 x 105 씩 씨딩(seeding)하여 24시간 동안 배양하였다. 그 후, 상기 <실시예 1>에서 분리된 말초혈액 단핵세포를 1.0 x 107 씩 0.4㎛ 포어 크기의 인써트(insert)에 넣어 공배양하였다. 사용 배지는 RPMI 1640 + 10% FBS + 페니실린/스트렙토마이신이며, 공 배양 24시간 후 말초혈액 단핵세포가 있는 인써트(insert)를 제거하고, 48시간 뒤, 세포 수를 측정하였다.
그 결과, 말초혈액 단핵세포와 자궁내막 상피세포주 SNU-685를 공배양 시, SNU-685 세포 수가 2배 이상 증가함을 확인하였다(도 2 및 도 3). 또한 자궁내막 상피세포주 SNU-1077 역시 말초혈액 단핵세포와의 공배양을 통해 유의적으로 세포수가 증가함을 관찰하였다(도 4 및 도 5).
이러한 결과를 통해, 말초혈액단핵세포가 자궁내막 상피세포의 증식 유도 효과가 있음을 알 수 있으며, 따라서, 복강경 등의 외과적 시술을 통해 자가 말초혈액 단핵 세포를 자궁 조직 내에 주입할 경우 자궁내막 조직이 효율적으로 재생되어 자궁내막이 얇은 환자가 수정란의 착상에 적당한 10mm 내외의 내막 두께를 유지하도록 할 수 있음을 예측할 수 있었다.
<실시예 3> in vivo에서 말초혈액단핵세포의 자궁내막 증식 효과 확인
본 발명의 말초혈액단핵세포가 자궁 조직 내 직접 주입할 경우, 자궁내막 조직이 효율적으로 증식되는지를 in vivo 상에서 확인하기 위하여, 마우스 자궁내막 조직에 말초혈액단핵세포를 직접 주입하여 그 효과를 확인하였다.
<3-1> 조직 두께 확인
마우스 또는 랫의 암컷의 발정 주기는 4-5일간으로, 발정전기(proestrus), 발정기(estrus), 발정 후기(metestrus), 및 발정 휴지기(diestrus)의 4단계로 나뉜다.
본 발명의 말초혈액단핵세포 처리에 의해 마우스 또는 랫 자궁내막 조직의 증식이 효율적으로 일어나는지 여부를 확인하기 위하여, 발정전기 상태의 12-15주령의 암컷 랫(rat)(구입처: 오리엔트바이오)을 마취한 다음, 등 쪽을 통해 자궁을 드러내고, 32 게이지(gage) 인슐린 주사기를 이용하여 상기 <실시예 1>에서 분리된 5X105의 말초혈액단핵세포(PBMC)를 자궁내막에 천천히 주입하였다(도 6). 이때, 분리된 말초혈액단핵세포는 세포 수를 계수하여, 5X105 cell/100ul PBS이 되도록 희석한 다음, 이를 하나의 vial로 분주하였으며, 실험시 1 vial(5X105 cell)또는 2 vial(1X106 cell)을 주입하였다. 이후, 랫의 몸에 자궁을 다시 넣고 봉합한 다음, 2일 후에 상기 랫에서 자궁을 적출하였다.
그 결과, 말초혈액단핵세포를 처리하지 않은 자궁 조직에 비해, 말초혈액단핵세포를 처리한 자궁 조직의 두께가 현저히 증가했음을 확인하였다(도 7).
<3-2> 조직 염색 결과
말초혈액단핵세포의 자궁내막 증식 효과를 확인하기 위하여, H&E 염색법 및 MT 염색법을 이용하여, 상기 적출된 자궁 조직의 염색을 수행하였다.
구체적으로, H&E 염색은 PBMC를 처리한 조직(A)과 처리하지 않은 조직(B)에서, 동일 선상의 조직을 채취하여, 조직 슬라이드를 준비하고, 각 조직 슬라이드를 수세 후, hematoxylin과 eosin을 이용하여 순차적으로 염색한 다음, 현미경으로 조직을 관찰하였다.
또한, MT(Masson's Trichorome) 염색은 PBMC를 처리한 조직(A)과 처리하지 않은 조직(B)에서, 동일 선상의 조직을 채취하여, 조직 슬라이드를 준비하고, 각 조직 슬라이드를 수세 후, Bouin solution, Biebrich scarlet-acid fuchsin 용액, P.T.A(phosphotugstic acid)-P.M.A(phosphomolybdic acid) 용액을 순서대로 처리한 다음, Aniline blue로 염색하여, 현미경으로 조직을 관찰하였다.
그 결과, 말초혈액단핵세포를 처리하지 않은 자궁 조직에 비해, 말초혈액단핵세포를 처리한 자궁 조직의 두께가 현저히 증가했음을 확인하였다(도 8).
<실시예 4> 발정 주기에 따른 자궁내막 증식 효과 확인
과다한 자궁증식은 자궁내막이 비정상적으로 과잉 증식되어 발생되는 자궁내막증식증 또는 자궁내막암을 유발할 수 있다.
이러한 관점에서, 본 발명의 말초혈액단핵세포가 자궁내막을 지속적으로 비정상적으로 과잉 증식시키는지의 여부를 확인하고자, 발정 주기에 따른 자궁내막 증식 효과를 확인하였다.
구체적으로, 발정전기 상태의 12-15주령의 암컷 랫(rat)(구입처: 오리엔트바이오)을 마취한 다음, 등 쪽을 통해 자궁을 드러내고, 32 게이지(gage) 인슐린 주사기를 이용하여 상기 <실시예 1>에서 분리된 5X105의 말초혈액단핵세포(PBMC)를 자궁내막에 천천히 주입하였다(도 6). 이때, 분리된 말초혈액단핵세포는 세포 수를 계수하여, 5X105 cell/100ul PBS이 되도록 희석한 다음, 이를 하나의 vial로 분주하였으며, 실험시 1 vial(5X105 cell) 또는 2 vial(1X106 cell)을 주입하였다. 이후, 랫의 몸에 자궁을 다시 넣고 봉합한 다음, 1번의 발정 주기가 끝난 6일 후에 상기 랫에서 자궁을 적출하였다.
그 결과, 상기 <실시예 3-1>에서 보는 바와 같이, 2일 후 적출된 자궁 조직에서는 PBMC 처리에 의해 자궁 조직의 두께가 현저히 증가함을 확인하였으나(도 7), 1번의 발정 주기가 끝난 6일 후 적출된 자궁 조직에서는 PBMC 처리 유무에 관계없이 자궁 두께에 차이가 없음을 확인하였다(도 9).
이를 통해, 본 발명의 말초혈액단핵세포는 1번의 발정 주기에서만 자궁내막 증식 효과가 있음을 알 수 있으며, 따라서, 과다한 자궁내막 증식으로 인한 질환을 유발하지 않음을 알 수 있다.
<실시예 5> PBMC 및 BM-MSC 주입에 따른 자궁내막 증식 효과 비교
<5-1> 조직 두께 확인
본 발명의 말초혈액단핵세포는 혈액으로부터 유래된 세포로, 골수로부터 유래된 세포와 비교하여, 자궁내막 증식 효과를 나타냄에 차이가 있는지 확인하기 위하여, 골수로부터 유래된 사람 중간엽 줄기 세포(bone-marrow mesenchymal stem cell, BM-MSC)를 이용하였다.
구체적으로, 사람 중간엽 줄기세포(BM-MSC)(론자(lonza)에서 구입) 및 상기 <실시예 1>에서 분리한 말초혈액단핵세포를 세포 수를 계수하여, 1X106 cell/100ul PBS이 되도록 희석한 다음, 이를 하나의 vial로 분주하였다.
이후, 발정전기 상태의 7-8주령의 암컷 면역 결핍 마우스(구입처:오리엔트바이오)를 마취한 다음, 등 쪽을 통해 자궁을 드러내고, 32 게이지(gage) 인슐린 주사기를 이용하여 상기 준비된 PBMC 및 BM-MSC를 각각 1 vial씩 자궁내막에 천천히 주입하였다. 이후, 마우스에 자궁을 다시 넣고 봉합한 다음, 2일 후에, 상기 마우스에서 자궁을 적출하여 그 두께를 비교하였다.
그 결과, BM-MSC가 주입된 자궁 조직에 비해, PBMC가 주입된 자궁 조직에서 조직 두께가 더욱 효과적으로 증가했음을 확인하였다(도 10).
<5-2> 조직 염색 결과
PBMC 및 BM-MSC의 자궁내막 증식 효과를 비교하기 위하여, 상기 <실시예 3-2>에 기재된 MT 염색법과 동일한 방법으로, 상기 적출된 자궁 조직의 염색을 수행하였다.
그 결과, PBMC 및 BM-MSC를 처리한 자궁 조직의 경우, 처리하지 않은 자궁 조직에 비해 자궁 조직의 두께가 증가했음을 확인하였다. 또한, BM-MSC에 비해 PBMC 처리시 자궁 조직의 두께가 현저히 증가했음을 확인하였다(도 11).
전술한 본 발명의 설명은 예시를 위한 것이며, 본 발명이 속하는 기술분야의 통상의 지식을 가진 자는 본 발명의 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 쉽게 변형이 가능하다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적인 아닌 것으로 이해해야만 한다.
본 발명의 말초혈액 단핵세포는 자궁내막 상피세포의 비약적인 증식을 통해 얇은 자궁내막의 조직 두께를 증가시키며, 이로 인해 불임으로 이어지는 수정란의 착상 실패를 효과적으로 예방 또는 개선시킬 수 있으며, 이를 통해 자궁내막 이상에 의한 불임의 예방 또는 치료용 조성물 또는 자궁내막 증식용 조성물의 유효성분으로 유용하게 이용될 수 있다.
Claims (10)
- 말초혈액 단핵세포(peripheral blood mononuclear cell, PBMC)를 유효성분으로 포함하는 자궁내막 이상에 의한 불임의 예방 또는 치료용 약제학적 조성물.
- 제 1 항에 있어서, 상기 말초혈액 단핵세포는 자가세포인 것을 특징으로 하는 조성물.
- 제 1 항에 있어서, 상기 자궁내막 이상은 자궁내막의 두께가 7mm 이하인 것을 원인으로 하는 것을 특징으로 하는 조성물.
- 말초혈액 단핵세포(peripheral blood mononuclear cell)를 유효성분으로 포함하는 자궁내막 증식용 조성물.
- 제 4 항에 있어서, 상기 말초혈액 단핵세포는 자가세포인 것을 특징으로 하는 조성물.
- 말초혈액 단핵세포(peripheral blood mononuclear cell)를 대상체의 자궁 내에 투여하는 단계를 포함하는 자궁내막 증식방법.
- 제 6 항에 있어서, 상기 말초혈액 단핵세포는 자가세포인 것을 특징으로 하는 방법.
- 제 1항의 약제학적 조성물의 약제학적 유효량을 개체에 투여하는 단계를 포함하는 자궁내막 이상에 의한 불임의 예방 또는 치료방법.
- 말초혈액 단핵세포(peripheral blood mononuclear cell)를 자궁내막 이상에 의한 불임의 예방 또는 치료용 조성물의 제조에 이용하는 용도.
- 말초혈액 단핵세포(peripheral blood mononuclear cell)를 자궁내막 증식용 조성물의 제조에 이용하는 용도.
