生体において、エネルギーは主に肝臓で変換されている。しかし、肝硬変の患者などでは、エネルギーを肝臓で上手く変換できないため、エネルギーが筋肉で変換されることを期待して、分岐鎖アミノ酸が投与される。このような分岐鎖アミノ酸の混合物は実際に上市されており、肝硬変や肝ガンの患者に臨床で広く処方されている。特許文献1には分岐鎖アミノ酸を用いた肝障害患者の耐糖能異常治療のための医薬組成物が開示されている。
一方、エネルギーの変換が一番良いのは糖質である。糖質を摂取すると、血糖値が上昇し、この刺激を受けて、膵臓からインスリンが分泌され、インスリンの作用により、糖質は細胞(肝臓、筋肉、脂肪組織等)に取り込まれる。特に糖尿病の患者では、糖質の摂取が制限されるので、インスリンの分泌に影響しない糖質以外のエネルギー源により、エネルギーを補充することが有用と考えられる。非糖質性のエネルギー源を用いたエネルギー産生増強剤のさらなる開発が求められている。
本発明の課題は、糖質以外のエネルギー源によりエネルギー産生を調節可能に増強することができる、体内エネルギー産生増強剤を提供することである。
本発明者らは、インスリン分泌に影響しないアミノ酸自体をエネルギー源としたエネルギー産生に着目し、アミノ酸を利用した体内エネルギー産生増強剤の開発を試みた。本発明者らは、鋭意検討した結果、分岐鎖アミノ酸とは異なるアミノ酸、具体的にはアスパラギン、フェニルアラニン、アラニン、トリプトファン及びアルギニンが、即効性又は遅効性の非糖質性エネルギー産生増強効果を有することを見出し、また、トレオニン、ヒスチジン、セリン、リジン、グリシン、チロシン、アスパラギン酸、システイン、バリン、ロイシン、イソロイシン、プロリン、メチオニン、グルタミン、及びグルタミン酸が、それらアミノ酸自体をエネルギー源としたエネルギー産生が可能であり、即効性でも遅効性でもない非糖質性エネルギー産生増強効果を有することを見出し、本発明を完成させた。
具体的には本発明は、以下を包含する。
[1]アスパラギン、フェニルアラニン、アラニン、トリプトファン及びアルギニンからなる群から選択される1以上のアミノ酸を含有する非糖質性エネルギー産生増強剤。
[2]前記1以上のアミノ酸が、アスパラギン、フェニルアラニン及びアラニンからなる群から選択される、即効性の、前記[1]の非糖質性エネルギー産生増強剤。
[3]前記1以上のアミノ酸が、トリプトファン及びアルギニンからなる群から選択される、遅効性の、前記[1]の非糖質性エネルギー産生増強剤。
[4]前記1以上のアミノ酸が、アスパラギン、フェニルアラニン及びアラニンからなる群から選択される1以上のアミノ酸と、トリプトファン及びアルギニンからなる群から選択される1以上のアミノ酸とからなる、持効性の、前記[1]~[3]のいずれかに記載の非糖質性エネルギー産生増強剤。
[5]トレオニン、ヒスチジン、セリン、リジン、グリシン、チロシン、アスパラギン酸、システイン、バリン、ロイシン、イソロイシン、プロリン、メチオニン、グルタミン、及びグルタミン酸からなる群から選択される1以上のアミノ酸をさらに含む、前記[1]~[4]のいずれかに記載の非糖質性エネルギー産生増強剤。

本発明者らはまた、カルボキシル基を13C標識した各種のアミノ酸を投与した動物の13CO2の呼気排泄量をモニターすることにより、アラニン、フェニルアラニン、アスパラギン、アルギニン、トリプトファン、グリシン、プロリン、グルタミン酸、リジン等のアミノ酸自体をエネルギー源としてエネルギーを産生できることも見出した。具体的には本発明は、以下も包含する。
[a]アラニン、フェニルアラニン、アスパラギン、アルギニン及びトリプトファンからなる群の少なくともいずれか1種類又は2種類以上のアミノ酸を有効成分として含有する、エネルギーを糖分で補給することが困難な者のためのエネルギー産生剤。
[b]アミノ酸の1日当たりの摂取量が0.1~30 gである、前記[a]に記載のエネルギー産生剤。
[c]アミノ酸の含有量が0.2~100w/w %である、前記[a]~[b]のいずれかに記載のエネルギー産生剤。
[d]アミノ酸及び製剤化のための補助剤のみを含む、前記[a]~[c]のいずれかに記載のエネルギー産生剤。
[e]エネルギー産生に即効性があり、アミノ酸がアラニン、フェニルアラニン及びアスパラギンからなる群の少なくともいずれか1種類又は2種類以上のアミノ酸であることを特徴とする、前記[a]~[d]のいずれかに記載のエネルギー産生剤。
[f]エネルギー産生に遅効性があり、アミノ酸がアルギニン及びトリプトファンからなる群の少なくともいずれか1種類又は2種類以上のアミノ酸であることを特徴とする、前記[a]~[d]のいずれかに記載のエネルギー産生剤。
[g]エネルギー産生に持久性があり、さらにグリシン、プロリン、グルタミン酸及びリジンからなる群の少なくともいずれか1種類又は複数種類のアミノ酸を有効成分として含有することを特徴とする、前記[e]~[f]のいずれかに記載のエネルギー産生剤。
[h]エネルギーを糖分で補給することが困難な者のためのエネルギー産生剤を製造するための、アラニン、フェニルアラニン、アスパラギン、アルギニン及びトリプトファンからなる群の少なくともいずれか1種類又は2種類以上のアミノ酸の使用。
[i]エネルギー産生に即効性があり、アミノ酸がアラニン、フェニルアラニン及びアスパラギンからなる群の少なくともいずれか1種類又は2種類以上のアミノ酸であることを特徴とする、前記[h]に記載の使用。
[j]エネルギー産生に遅効性があり、アミノ酸がアルギニン及びトリプトファンからなる群の少なくともいずれか1種類又は2種類以上のアミノ酸であることを特徴とする、前記[h]に記載の使用。
本明細書は本願の優先権主張の基礎となる日本国特許出願2012-106160号の開示内容を包含する。
本発明に係る非糖質性エネルギー産生増強剤は、投与後の非糖質性エネルギー産生を、タイミングを調節しながら増強することができる。
以下、本発明を詳細に説明する。
本発明は、アスパラギン、フェニルアラニン、アラニン、トリプトファン及びアルギニンからなる群から選択される1以上のアミノ酸を含有する非糖質性エネルギー産生増強剤を提供する。
本発明において「非糖質性エネルギー産生増強」とは、糖質ではないエネルギー源(ここでは特にアミノ酸)を用いた体内エネルギー産生(ATP産生)のレベルを増強することを意味する。本発明に係る非糖質性エネルギー産生増強剤は、有効成分として含まれるアミノ酸の種類に応じて、即効性、遅効性又は持効性の、非糖質性エネルギー産生増強効果を示す。
具体的には、アスパラギン、フェニルアラニン及びアラニンからなる群から選択される1以上のアミノ酸を含有する本発明に係る非糖質性エネルギー産生増強剤は、即効性である。本発明において「即効性」とは、投与後、そのアミノ酸をエネルギー源とした非糖質性エネルギー産生が行われる期間内において、エネルギー産生量のピークが投与後比較的短い時間で現れることを意味する。即効性の非糖質性エネルギー産生増強剤は、投与後できるだけ短い時間で非糖質性エネルギー産生を増強させる必要がある場合に好適に用いることができる。
一方、トリプトファン及びアルギニンからなる群から選択される1以上のアミノ酸を含有する本発明に係る非糖質性エネルギー産生増強剤は、遅効性である。本発明において「遅効性」とは、投与後、そのアミノ酸をエネルギー源とした非糖質性エネルギー産生が行われる期間内において、エネルギー産生量のピークが投与後比較的遅い時間に現れることを意味する。遅効性の非糖質性エネルギー産生増強剤は、投与後ある程度の時間経過後に非糖質性エネルギー産生を増強させる必要がある場合に好適に用いることができる。
さらに、本発明は、アスパラギン、フェニルアラニン及びアラニンからなる群から選択される1以上のアミノ酸(即効性アミノ酸)と、トリプトファン及びアルギニンからなる群から選択される1以上のアミノ酸(遅効性アミノ酸)とを含有する非糖質性エネルギー産生増強剤も提供する。このような即効性アミノ酸と遅効性アミノ酸の両方を含有する非糖質性エネルギー産生増強剤は、持効性である。本発明において「持効性」とは、投与後早期からより長い時間にわたってより高いエネルギー産生量が持続することを意味する。持効性の非糖質性エネルギー産生増強剤は、投与後早期からより長時間にわたって非糖質性エネルギー産生を増強させる必要がある場合に好適に用いることができる。
本発明に係る非糖質性エネルギー産生増強剤はまた、トレオニン、ヒスチジン、セリン、リジン、グリシン、チロシン、アスパラギン酸、システイン、バリン、ロイシン、イソロイシン、プロリン、メチオニン、グルタミン、及びグルタミン酸からなる群から選択される1以上のアミノ酸をさらに含んでもよい。これらのアミノ酸は、即効性でも遅効性でもなく、そのエネルギー産生量のピークが中間的な時点で現れる。このためこれらのアミノ酸は、上記のような即効性又は遅効性の非糖質性エネルギー産生増強剤にさらに添加することにより、非糖質性エネルギー産生を経時的により安定的に増強させることができる。
本発明に係る非糖質性エネルギー産生増強剤に用いられうるアラニンは、非必須アミノ酸の1つであり、D体とL体とが存在する。L-アラニンのCas No.は、56-41-7である。L-アラニンの別名として、(S)-2-アミノプロピオン酸、(2S)-2-アミノプロパン酸、α-アラニン、及びL-(+)-アラニンがある。アラニンの略号として、Ala又はAが用いられる。アラニンは、水や蟻酸に溶けやすく、エタノールには、ほとんど溶けない(第十六改正日本薬局方)。アラニンは、多種のタンパク質などの様々な素材や食品に含まれていることが知られている。アラニンは、タンパク質の構成アミノ酸の一種であり、脂肪の燃焼に関係することや、結合組織の材料であることが知られている。
本発明に係る非糖質性エネルギー産生増強剤に用いられうるフェニルアラニンは、必須アミノ酸の1つであり、D体とL体とが存在する。L-フェニルアラニンのCas No.は、63-91-2である。その別名として(S)-α-アミノベンゼンプロパン酸、(2S)-2-アミノ-3-フェニルプロパン酸、(S)-3-フェニル-2-アミノプロピオン酸、L-β-フェニルアラニン、L-(-)-フェニルアラニンがある。フェニルアラニンの略号として、Phe又はFが用いられる。フェニルアラニンは、蟻酸に溶けやすく、水にやや溶けにくく、希塩酸に溶け、エタノールにはほとんど溶けない(第十六改正日本薬局方)。フェニルアラニンは、多種のタンパク質などの様々な素材や食品に含まれていることが知られている。フェニルアラニンは、ドーパミンやノルエピネフリン、エピネフリンの原料であることが知られている。
本発明に係る非糖質性エネルギー産生増強剤に用いられうるアスパラギンは、非必須アミノ酸の1つであり、D体とL体とが存在する。L-アスパラギンのCas No.は、70-47-3である。その別名として(S)-α-アミノスクシンアミド酸、(S)-2-アミノ-3-カルバモイルプロパン酸、L-(-)-アスパラギンがある。アスパラギンの略号として、Asn又はNが用いられる。アスパラギンは、アスパラガス、肉類、大豆、玄米、レーズン、エビ、牛乳、ジャガイモなどの様々な素材や食品に含まれていることが知られている。
本発明に係る非糖質性エネルギー産生増強剤に用いられうるアルギニンは、非必須アミノ酸の1つであり、D体とL体とが存在する。L-アルギニンのCas No.は、74-79-3である。その別名として(S)-2-アミノ-5-グアニジノペンタン酸、(S)-2-アミノ-5-(アミジノアミノ)吉草酸、L-(+)-アルギニン、アルギUがある。アルギニンの略号として、Arg又はRが用いられる。アルギニンは、水や蟻酸に溶けやすく、希塩酸に溶け、エタノールにはほとんど溶けない(第十六改正日本薬局方)。アルギニンは、肉類等の多種のタンパク質などの様々な素材や食品に含まれていることが知られている。アルギニンは、尿素回路の中間体として生合成されるが、速やかに分解されるため子供では必須アミノ酸とされている。アルギニンは、成長ホルモンの分泌促進、免疫機能の向上、脂肪代謝の促進に関与することが知られている。
本発明に係る非糖質性エネルギー産生増強剤に用いられうるトリプトファンは、必須アミノ酸の1つであり、D体とL体とが存在する。L-トリプトファンのCas No.は、73-22-3である。その別名として3-(1H-インドール-3-イル)-L-アラニン、(S)-3-(1H-インドール-3-イル)-2-アミノプロパン酸、(S)-α-アミノ-1H-インドール-3-プロパン酸、L-(+)-トリプトファン、エルトリップ、トリプタンがある。トリプトファンの略号として、Trp又はWが用いられる。トリプトファンは、蟻酸に溶けやすく、水に溶けにくく、希塩酸に溶け、エタノールには極めて溶けにくい(第十六改正日本薬局方)。トリプトファンは、多種のタンパク質などの様々な素材や食品に含まれていることが知られている。トリプトファンは、セロトニンやメラトニンの原料であり。鎮静作用や月経前不快気分障害に対する有効性があることが知られている。
本発明に係る非糖質性エネルギー産生増強剤において、アスパラギン、フェニルアラニン、アラニン、トリプトファン、及びアルギニンは、遊離塩基、一水和物等の水和物、又は塩の形態で配合されてもよい。塩は、有機酸(酢酸、酒石酸、脂肪酸など)、有機塩基、無機酸(塩酸、臭化水素酸、硝酸、硫酸、過塩素酸など)、又は無機塩基(カリウム、ナトリウム、亜鉛など)との間で形成された塩であってもよい。そして、アスパラギン、フェニルアラニン、アラニン、トリプトファン、及びアルギニンは、D体とL体のいずれであってもよい。
本発明に係る非糖質性エネルギー産生増強剤の製造に用いうる、アスパラギン、フェニルアラニン、アラニン、トリプトファン、又はアルギニンは、アスパラギン、フェニルアラニン、アラニン、トリプトファン、又はアルギニンを多く含む素材や食品などから、搾汁、濃縮、精製、結晶化、又は各種の溶媒を用いた抽出等によって得ることができる。抽出には、各種の溶媒として、水又は通常用いられる溶媒、例えば、アルコール類、炭化水素類、有機酸、有機塩基、無機酸、無機塩基、超臨界流体等を単独で又は複数を組み合わせて用いることが可能である。本発明に係る非糖質性エネルギー産生増強剤に用いるアミノ酸は、微生物で生産したものを用いてもよいし、化学的に合成されたものを用いてもよい。アスパラギン、フェニルアラニン、アラニン、トリプトファン、及びアルギニンは、市販品を入手することもできる。
本発明に係る非糖質性エネルギー産生増強剤は、アスパラギン、フェニルアラニン、アラニン、トリプトファン、及びアルギニン以外のアミノ酸をさらに含有するものであってもよいし、アスパラギン、フェニルアラニン、アラニン、トリプトファン、及びアルギニン以外のアミノ酸を含有しなくてもよい。例えば、これに該当するアミノ酸はトレオニン、ヒスチジン、セリン、リジン、グリシン、チロシン、アスパラギン酸、システイン、バリン、ロイシン、イソロイシン、プロリン、メチオニン、グルタミン、及びグルタミン酸である。しかしながら、本発明に係る非糖質性エネルギー産生増強剤は、非運動時のエネルギー補給を意図する場合などには、特に分岐鎖アミノ酸(バリン、ロイシン、及びイソロイシン)を含有しないことも好ましい。
本発明に係る非糖質性エネルギー産生増強剤に関して特定の成分(分岐鎖アミノ酸、糖質等)を「含有しない」とは、その成分を全く含有しないか、検出限界以下の量しか含有しないか、又はエネルギー産生に影響を及ぼさない程度の量でしか含有しないことを意味する。
本発明に係る非糖質性エネルギー産生増強剤に含まれる、アスパラギン、フェニルアラニン、アラニン、トリプトファン、及びアルギニンからなる群から選択される1種類又は2種類以上のアミノ酸の一日当たりの摂取量の合計は、年齢、症状、体重などによって異なるため、特に限定されないが、例えば、0.1~30 gの摂取量に設定することができ、好ましくは0.3~10 g又は0.3~3 gの摂取量、さらに好ましくは0.5~3 g又は0.5~1 gの摂取量に設定することができる。
本発明に係る非糖質性エネルギー産生増強剤における、アスパラギン、フェニルアラニン、アラニン、トリプトファン、及びアルギニンからなる群から選択される1種類又は2種類以上のアミノ酸の配合量の合計は、剤型、症状、体重などによって異なるため、特に限定されないが、例えば、0.2~100w/w%の配合量に設定することができ、好ましくは0.3~100w/w%の配合量、さらに好ましくは0.4~100w/w%の配合量に設定することができる。なおw/w%は重量/重量%である。
本発明に係る非糖質性エネルギー産生増強剤は、日用量あたり、アスパラギンを0.005~ 35g、通常は0.005~10g、好ましくは0.01~1 g含んでもよい。本発明に係る非糖質性エネルギー産生増強剤は、フェニルアラニンを0.005~7.7 g、通常は0.005~4.8 g、好ましくは0.01~1 g含んでもよい。本発明に係る非糖質性エネルギー産生増強剤は、日用量あたり、アラニンを0.005~8.5 g、通常は0.005~5.2 g、好ましくは0.01~1 g含んでもよい。本発明に係る非糖質性エネルギー産生増強剤は、日用量あたり、トリプトファンを0.005~2.1 g、通常は0.005~1.3 g、好ましくは0.01~1 g含んでもよい。本発明に係る非糖質性エネルギー産生増強剤は、日用量あたり、アルギニンを0.005~10.1 g、通常は0.005~5.9 g、好ましくは0.01~1 g含んでもよい。
本発明に係る非糖質性エネルギー産生増強剤に関して、「日用量あたり、アスパラギンをX g含む」とは、本発明に係る非糖質性エネルギー産生増強剤に含まれるアスパラギンの量が、本発明に係る非糖質性エネルギー産生増強剤の1日当たりの投与量又は摂取量(日用量)に換算したときにX gとなる量であることを意味する。具体的には、例えば日用量が200 gである本発明に係る非糖質性エネルギー産生増強剤の場合には、その非糖質性エネルギー産生増強剤200 g中にX gのアスパラギンが含まれ、100 g中にはX gの1/2量のアスパラギンが含まれる。本発明において個々の非糖質性エネルギー産生増強剤の「日用量」は、健康上悪影響を生じない範囲で当業者が適宜定めることができる。本発明に係る非糖質性エネルギー産生増強剤に含まれうる他の成分(フェニルアラニン、アラニン、トリプトファン、及びアルギニン等)の含有量に関する記載も、上述のアスパラギンの含有量と同様に解釈される。
本発明に係る非糖質性エネルギー産生増強剤に用いられうる他のアミノ酸についても、例えば、前記アルギニンの情報を参考にし、0.005~35 g、通常は0.005~10 g、好ましくは0.01~1 g含んでもよく、その他の公知情報の日用量を参考に、日用量を設定してもよい。
本発明に係る非糖質性エネルギー産生増強剤はまた、投与又は摂取対象の体重1 kgあたり(kg・BW)、アスパラギン、フェニルアラニン、アラニン、トリプトファン及びアルギニンからなる群から選択される1以上のアミノ酸を、合計量で、典型的には、0.03~3 g/kg・BW、好ましくは0.05~2 g/kg・BW、さらに好ましくは0.08~1.5 g/kg・BW、例えば0.1~1 g/kg・BW、0.1~0.8 g/kg・BW、0.1~0.6 g/kg・BW、0.1~0.5 g/kg・BW、又は0.2~0.4 g/kg・BW含んでもよい。
本発明に係る非糖質性エネルギー産生増強剤は、上記のアミノ酸に加えて、製剤化補助剤をさらに含んでもよい。製剤化補助剤としては、例えば、賦形剤、結合剤、崩壊剤、滑沢剤、矯臭剤、溶解補助剤、懸濁剤、コーティング剤、担体(固体担体、水などの液体担体等)、保存剤、香料、甘味料、着色料、pH調整剤、安定化剤などの医薬品又は食品の製剤化の際に用いられる既知の添加剤が挙げられる。なお、これらは食品あるいは医薬品の製剤技術の分野において通常で用いうるものであればよい。本発明に係る非糖質性エネルギー産生増強剤はまた、適当量のビタミン、ミネラル、有機酸、ペプチド類などをさらに含有してもよい。
本発明に係る非糖質性エネルギー産生増強剤は、糖質を含有してもよいが、含有しないことがより好ましい。
本発明に係る非糖質性エネルギー産生増強剤は、固体状、ゲル状、若しくは液体状又はそれらの混合形態であってよい。本発明に係る非糖質性エネルギー産生増強剤は、錠剤(タブレット)、カプセル剤、顆粒剤、散剤、シロップ剤、水剤、懸濁剤、乳剤、ドリンク剤、ゼリー剤、ジェル剤等の任意の形態であってもよい。本発明に係る非糖質性エネルギー産生増強剤は、使用前に液体媒体に溶解させることを意図した乾燥物の形態であってもよい。本発明に係る非糖質性エネルギー産生増強剤は、非糖質性エネルギー産生の増強を意図する限り、医薬品として製造されるものであっても、食品(好ましくは、サプリメントや特定保健用食品等の機能性食品)として製造されるものであってもよい。
また、本発明に係る非糖質性エネルギー産生増強剤は、摂取時の衛生面や、輸送時、保管時の取り扱いやすさの観点から、1食あたりの単位量で包装されることも好ましい。したがって本発明は、1食あたりの単位包装形態からなる非糖質性エネルギー産生増強剤も提供する。「1食あたりの単位包装形態からなる非糖質性エネルギー産生増強剤」とは、1食あたりの単位量で包装された状態にある、非糖質性エネルギー産生増強剤を意味する。その具体例としては、例えば、パック、カップ又はボトル等の容器に充填された液剤やゼリー剤などが挙げられるが、これらに限定されるものではない。1食あたりの単位量は、典型的には、0.05~500 g、好ましくは0.1g~500g、さらに好ましくは1g~500g、例えば0.1g~100g(液体の場合、0.1mL~100mL)、50g~500g(液状の場合、50mL~500mL)、75g~300g(液状の場合、75mL~300mL)、100g~200g(液状の場合、100mL~200mL)でありうる。ここで、「1食あたりの単位量」は、1回に摂取することが想定される予め規定した量(1回摂取量)を意味する。「1回に摂取することが想定される予め規定した量」は、比較的短時間のうちにその全量の摂取を完了することが想定される量であればよく、例えば経口摂取の場合、一口で又は連続して摂取可能な量等に限定されない。「1食あたりの単位量」は、例えば本発明に係る非糖質性エネルギー産生増強剤の日用量であってもよいし、その日用量を小分けにした量(例えば日用量の1/2~1/10量)であってもよい。
本発明に係る1食あたりの単位包装形態からなる非糖質性エネルギー産生増強剤に含まれる、アスパラギン、フェニルアラニン、アラニン、トリプトファン及びアルギニンからなる群から選択される1以上のアミノ酸の合計量は、典型的には、0.1~10 g、好ましくは0.5~8 g、さらに好ましくは0.7~7 g、例えば、0.8~6 g、0.9~5 g、1~4 g、1.1~3 g、又は1.2~2 gであってよいが、これらに限定されるものではない。
本発明に係る非糖質性エネルギー産生増強剤は、通常は消化管経由で体内に取り込まれるものであることが好ましく、経口用又は経腸用であることがより好ましい。経口用又は経腸用とは、経口又は経腸での投与又は摂取のために調製されたものであることを意味する。本発明では、消化管経由で体内に取り込むことを指して「摂取」とも称する。本明細書では、「摂取」と「投与」を互換的に使用することがある。
本発明に係る非糖質性エネルギー産生増強剤は、当業者に周知の他の様々な製剤技術や食品加工技術を用いて調製することができる。本発明に係る非糖質性エネルギー産生増強剤は、医薬品又は食品のいずれの製造にも使用することができる。
本発明に係る非糖質性エネルギー産生増強剤は、その有効成分であるアミノ酸自体をエネルギー源としたエネルギー産生を誘導することにより、体内の非糖質性エネルギー産生を増強することができる。
本発明では、本発明に係る非糖質性エネルギー産生増強剤に含まれうるアスパラギン、フェニルアラニン、及びアラニンについて、カルボン酸を13C標識した各種のアミノ酸を投与した動物の13CO2の呼気排泄量をモニターすることにより、そのアミノ酸自体をエネルギー源としてエネルギーを産生できること、及びそのエネルギー産生は即効性があることが見出された。同様に本発明では、本発明に係る非糖質性エネルギー産生増強剤に含まれうるトリプトファン及びアルギニンについて、それ自体をエネルギー源としてエネルギーを産生できること、及びそのエネルギー産生は遅効性があることが見出された。さらに本発明において、トレオニン、ヒスチジン、セリン、リジン、グリシン、チロシン、アスパラギン酸、システイン、バリン、ロイシン、イソロイシン、プロリン、メチオニン、グルタミン、及びグルタミン酸について、それ自体をエネルギー源としてエネルギーを産生できること、及びそのエネルギー産生がピークに達する早さは中程度であることが見出された。
本発明において、エネルギー源となるアミノ酸についてエネルギー産生の効率がよいとは、他のアミノ酸よりも1モル当たりのエネルギー産生量が高いことをいう。本発明において、エネルギー産生の効率並びに即効性及び遅効性は、具体的には、カルボン酸を13C標識したアミノ酸を非ヒト哺乳動物等に経口投与し、呼気中のΔ13CO2を経時的に測定し、アミノ酸の投与後から120分間の最高Δ13CO2排泄量(Cmax)、最高Δ13CO2排泄発現時間(Tmax)、及びΔ13CO2測定値-時間曲線の下面積(AUC120min)を算出することにより、評価することができる。このとき、20種の天然アミノ酸の平均値よりも高いAUC120min又はCmaxの値は、エネルギー産生の効率がよいことを示す。Tmax値が小さい場合、即効性のエネルギー産生であることを示し、Tmax値が大きい場合、遅効性のエネルギー産生であることを示す。さらに、Tmaxが平均的な値である場合、エネルギー産生がピークに達する早さが中程度であることを示す。
生体でのエネルギー産生においては、エネルギー源として糖質が優先的に利用され、次にタンパク質(アミノ酸)及び脂質が利用される。しかし、エネルギー産生量では、脂質が9kcal/g、糖質とタンパク質が4kcal/gとなり、脂質が糖質やタンパク質よりも高い。
一方、エネルギーの変換率が一番良いのは糖質である。糖質を摂取すると、消化・吸収の後に、血糖値が上昇する。血糖値の上昇の刺激を受けて、膵臓からインスリンが分泌され、インスリンの作用により、糖質は各種の細胞(筋肉、脂肪組織等)に取り込まれ、エネルギーを産生する。
しかし、インスリンの分泌が不十分な場合(例えば、I型糖尿病)や、インスリンに対して抵抗性が有る場合(例えば、II型糖尿病)には、細胞に必要な糖質が十分に取り込まれず、エネルギーを糖質で補給することが困難となる。また、このような者において、糖質が細胞に取り込まれず、血中に高濃度で存在し続けると、各種の合併症(血管肥厚、動脈硬化、血液の循環不良、腎症、神経障害、感染リスク上昇等)が誘発されるため、血糖値のコントロールが重要になる。そのために、このように糖質性エネルギー産生が困難な者に対しては、インスリン、インスリン分泌刺激剤、インスリン抵抗性改善剤、又は糖代謝抑制剤の投与や、食餌療法等の治療手段が用いられている。一方、脳細胞、心筋細胞、及び肝細胞への糖質の取り込みはインスリンに依存しないため、前述の治療によって、血中のインスリン濃度が過剰になると、これらの細胞ではエネルギー不足に陥る危険がある。
本発明に係る非糖質性エネルギー産生増強剤は、糖質以外の物質(アミノ酸)からエネルギーを補充することができるため、このように糖質性エネルギー産生が困難な者においてエネルギー産生を増強する目的に適している。
本発明は、本発明に係る非糖質性エネルギー産生増強剤を、非糖質性エネルギー産生の増強が望まれる対象に投与又は摂取させることにより、非糖質性エネルギー産生を増強させる方法にも関する。
本発明に係る非糖質性エネルギー産生増強剤は、例えば、非糖質性エネルギー産生の増強が望まれる対象に、特定保健用食品などの特別用途食品、栄養機能食品、栄養補助食品やサプリメントとして摂取させることにより、又は医薬品として直接投与することにより、エネルギー産生を増強することが期待される。
なお、本発明に係る非糖質性エネルギー産生増強剤を用いた特定保健用食品などの特別用途食品、栄養機能食品、栄養補助食品、サプリメント等は、一般食品と明確に区別しながら、非糖質性エネルギー産生を増強する目的で使用することができる。普段の生活や療養中はもとより、瞬発力を要する運動やスポーツ(重量挙げ、短距離走等)や持続力を要する運動やスポーツ(マラソン、長距離走、トライアスロン等)の前や途中に摂取することで、必要なエネルギーを有効に補給することができる。あるいは、糖質性エネルギー産生が困難な者に対してエネルギー補給用に摂取させることもできる。
即効性のエネルギー産生増強効果を示すアミノ酸、すなわちアスパラギン、フェニルアラニン、及びアラニンの少なくとも1つを含む本発明に係る非糖質性エネルギー産生増強剤を対象に投与又は摂取させることにより、即効性の非糖質性エネルギー補給が可能になる。遅効性のエネルギー産生増強効果を示すアミノ酸、すなわちトリプトファン及び/又はアルギニンを含む本発明に係る非糖質性エネルギー産生増強剤を対象に投与又は摂取させることにより、遅効性の非糖質性エネルギー補給が可能になる。アスパラギン、フェニルアラニン及びアラニンの少なくとも1つと、トリプトファン及び/又はアルギニンとを含む本発明に係る非糖質性エネルギー産生増強剤を対象に投与又は摂取させることにより、持効性の非糖質性エネルギー補給が可能になる。またアスパラギン、フェニルアラニン、アラニン、トリプトファン及びアルギニンからなる群から選択される1以上のアミノ酸に加えて、トレオニン、ヒスチジン、セリン、リジン、グリシン、チロシン、アスパラギン酸、システイン、バリン、ロイシン、イソロイシン、プロリン、メチオニン、グルタミン、及びグルタミン酸からなる群から選択される少なくとも1つを含有する非糖質性エネルギー産生増強剤を対象に投与又は摂取させることにより、より安定した非糖質性エネルギー補給が可能になる。エネルギー産生のピークに達する早さが中程度であるアミノ酸トレオニン、ヒスチジン、セリン、リジン、グリシン、チロシン、アスパラギン酸、システイン、バリン、ロイシン、イソロイシン、プロリン、メチオニン、グルタミン、グルタミン酸からなる群のうちの複数又は単独と、遅効性のエネルギー産生増強効果を示すアミノ酸トリプトファン及び/又はアスパラギンを組み合わせて、即効性ではないが持効性の非糖質性エネルギー補給が可能になる。
非糖質性エネルギー産生の増強が望まれる対象としては、糖質性エネルギー産生が困難な者の他、日常的に運動又は肉体労働(特に有酸素運動や持久力を必要とする、運動又は労働)を行っている対象、例えばアスリートが挙げられる。本発明において「アスリート」とは、プロ又はアマチュアのスポーツ競技者であって、自らの競技成績又は運動能力の向上を目標として競技(好ましくは有酸素運動や持久力を必要とする競技)を行っている人をいう。しかし本発明に係る非糖質性エネルギー産生増強剤は、糖質性エネルギー産生が困難な者に特に適しており、例えば分岐鎖アミノ酸を含有しない非糖質性エネルギー産生増強剤を用いる場合には、糖質性エネルギー産生が困難な者の非運動時のエネルギー補給にとりわけ適している。
即効性のエネルギー産生増強効果を示すアミノ酸、すなわちアスパラギン、フェニルアラニン、及びアラニンの少なくとも1つを含む本発明に係る非糖質性エネルギー産生増強剤は、即効性の非糖質性エネルギー産生が望まれる対象用に適している。遅効性のエネルギー産生増強効果を示すアミノ酸、すなわちトリプトファン及び/又はアルギニンを含む本発明に係る非糖質性エネルギー産生増強剤は、遅効性の非糖質性エネルギー産生が望まれる対象用に適している。アスパラギン、フェニルアラニン及びアラニンの少なくとも1つと、トリプトファン及び/又はアルギニンとを含む本発明に係る非糖質性エネルギー産生増強剤は、持効性の非糖質性エネルギー産生が望まれる対象用に適している。
本発明に係る方法では、非糖質性エネルギー産生の増強が望まれる対象に対して本発明に係る非糖質性エネルギー産生増強剤を、1回以上投与することが好ましい。本発明に係る非糖質性エネルギー産生増強剤は、日用量を1日に1回投与してもよいし、1日に日用量を数回に分割して投与してもよい。
なお、本発明に係る非糖質性エネルギー産生増強剤は、従来公知のエネルギー産生増強効果がある食品や医薬品の摂取と同時又はそれらの摂取の前後に併せて投与又は摂取してもよい。
本発明は、糖質性エネルギー産生が困難な者(エネルギーを糖分で補給することが困難な者)のための非糖質性エネルギー産生増強剤を製造するための、アスパラギン、フェニルアラニン、アラニン、トリプトファン及びアルギニンからなる群から選択される1以上のアミノ酸の使用(使用方法)も提供する。アスパラギン、フェニルアラニン、及びアラニンは即効性のエネルギー産生増強を目的とした剤への使用に適している。また、トリプトファン及びアルギニンは遅効性のエネルギー産生増強を目的とした剤への使用に適している。この方法において、即効性のエネルギー産生を示す前記アミノ酸と、遅効性のエネルギー産生を示す前記アミノ酸を組み合わせて用いてもよい。さらに、エネルギー産生のピークに達する早さが中程度であるトレオニン、ヒスチジン、セリン、リジン、グリシン、チロシン、アスパラギン酸、システイン、バリン、ロイシン、イソロイシン、プロリン、メチオニン、グルタミン、グルタミン酸をさらに組み合わせて用いてもよい。
本発明はさらに、13C標識された生体エネルギー源物質を非ヒト哺乳動物に経口投与し、呼気中の13C標識された二酸化炭素量を測定することにより、生体エネルギー源物質の体内エネルギー産生能を評価する方法も提供する。生体エネルギー源物質としては、タンパク質、アミノ酸、糖質、脂質等が挙げられる。呼気中の13C標識された二酸化炭素量の測定は、任意の公知方法を用いて行えばよいが、例えば、特開2006-267098及び特開2008-051787に記載の呼気採取方法を用いて行うことが好ましい。具体的には、図1及び下記実施例の記載を参照することができる。簡単に記載すると、デシケーター内の多数の小孔がある板状の中敷き上にラットを乗せ、ふたを閉めて固定し、デシケーターの呼気採取口から移送された呼気を直接呼気採取バッグに、一定速度(好ましくは、150ml/min)で継続的に採取し、採取した呼気中の13CO2率(例えば13CO2(‰))を測定する。なお‰=0.1%である。測定値から標準ガス、好ましくは混合ガス(O2:95%、CO2:5%)、の13CO2率を差し引いた値が、13C標識された生体エネルギー源物質がエネルギー源として代謝されて呼気中に排泄された二酸化炭素量に相当する。この呼気中に排泄された二酸化炭素量の経時的変化量に基づいて、生体エネルギー源物質の体内エネルギー産生能を評価することができる。
以下、本発明に関して、実施例を挙げて説明するが、本発明は、これにより限定されるものではない。
カルボキシル基を13C標識した20種類のアミノ酸について、特開2006-267098及び特開2008-051787に記載の呼気採取方法を用いて、経口投与後の13Cの呼気排泄量からそれぞれのエネルギー変換能(すなわち、エネルギー産生量)を評価した。具体的な方法を以下に示す。
(1)被検物質
カルボキシル基を13C標識した20種類のアミノ酸(L-トリプトファン、L-リジン、グリシン、L-システイン、L-セリン、L-アラニン、L-プロリン、L-アルギニン、L-ヒスチジン、L-トレオニン、L-バリン、L-グルタミン、L-アスパラギン、L-メチオニン、L-グルタミン酸、L-アスパラギン酸、L-イソロイシン、L-ロイシン、L-フェニルアラニン、L-チロシン)を被検物質として用いた。前記アミノ酸のうち、リジンは塩酸塩、アスパラギンは一水和物、他のアミノ酸は遊離塩基のものを用いた。
カルボキシル基を13C標識した20種類のアミノ酸(L-トリプトファン、L-リジン、グリシン、L-システイン、L-セリン、L-アラニン、L-プロリン、L-アルギニン、L-ヒスチジン、L-トレオニン、L-バリン、L-グルタミン、L-アスパラギン、L-メチオニン、L-グルタミン酸、L-アスパラギン酸、L-イソロイシン、L-ロイシン、L-フェニルアラニン、L-チロシン)を被検物質として用いた。前記アミノ酸のうち、リジンは塩酸塩、アスパラギンは一水和物、他のアミノ酸は遊離塩基のものを用いた。
(2)アミノ酸投与及び呼気の採取
7週齢のSD系雄性ラット(日本SLC株式会社)を群間の体重の平均値が等しくなるように群分けし(20群、各n=4)、16~20時間絶食した(但し、水は自由摂取とした)。アミノ酸投与群には、上記の各種13C標識アミノ酸(20種類)を1群に1種類ずつ、蒸留水に0.25mmol/2.5mlの濃度で懸濁又は溶解し、0.25mmol/kgの用量で経口投与した。13C標識アミノ酸の投与直後から、ラットを個体毎に、デシケーターに入れ、経時的に呼気を採取した。13C標識アミノ酸の投与から70分後までは5分毎に、13C標識アミノ酸の投与から70分後以降には90分後と120分後に、各時点の開始から1.5分間の呼気を採取した。
7週齢のSD系雄性ラット(日本SLC株式会社)を群間の体重の平均値が等しくなるように群分けし(20群、各n=4)、16~20時間絶食した(但し、水は自由摂取とした)。アミノ酸投与群には、上記の各種13C標識アミノ酸(20種類)を1群に1種類ずつ、蒸留水に0.25mmol/2.5mlの濃度で懸濁又は溶解し、0.25mmol/kgの用量で経口投与した。13C標識アミノ酸の投与直後から、ラットを個体毎に、デシケーターに入れ、経時的に呼気を採取した。13C標識アミノ酸の投与から70分後までは5分毎に、13C標識アミノ酸の投与から70分後以降には90分後と120分後に、各時点の開始から1.5分間の呼気を採取した。
呼気の採取は図1に示す評価システムを用いて次のとおり行った。まず容積2Lのデシケーター内に、数mm(ミリメートル)の多数の小孔がある板状の中敷きを設け、その上にラットを乗せ、ふたを閉めてから、呼気採取口又は空気採入口以外の部分から呼気が漏れないように、ふたを固定した。デシケーターの側面開口部(空気採入口)より、シリコンチューブを通し、チューブ開口部の一端をデシケーター内の最下部(動物の体躯以下かつ呼気採取口よりも下部に位置する)に固定して、呼気採取口とした。シリコンチューブには、空気採入口より口径の小さいものを用いた。呼気は、この呼気採取口から移送される。シリコンチューブの開口部の他端をUBiT・POCone専用呼気採取バッグ20に接続し、デシケーターとUBiT・POCone専用呼気採取バッグ20の間には、ペリスタポンプ(Master Flex L/S;Cole-Parmer Instrument Company)を接続して、デシケーター内に排泄された呼気を、一定速度の150ml/minで、それぞれ継続的に吸引して、UBiT・POCone専用呼気採取バッグ20(大塚製薬株式会社)に採取した。
(3)呼気の評価
呼気を採取したUBiT・POCone専用呼気採取バッグ20を、UBiT-IR300赤外分析装置(大塚電子株式会社)に接続したUBiT-IR300専用オートサンプラー UBiT-AS10(大塚製薬株式会社)へ接続し、混合ガス(O2:95%、CO2:5%)を対照として、呼気中のΔ13CO2(‰)(=呼気の13CO2(‰)値から混合ガスの13CO2(‰)値を差し引いた値)を測定した。個体毎に最高Δ13CO2排泄量(以降、Cmaxともいう)、最高Δ13CO2排泄発現時間(以降、Tmaxともいう)及び13Cアミノ酸投与後0~120分間のΔ13CO2測定値-時間曲線の下面積(台形面積法による。以降、AUC120minともいう)を算出した。各アミノ酸の平均値が20種類のアミノ酸全体のmean+SD(平均値+標準偏差)以上である場合を高い値を示したと判断した。また、各アミノ酸の平均値が20種類のアミノ酸全体のmean-SD(平均値-標準偏差)以下である場合を低い値を示したと判断した。各アミノ酸の平均値が20種類のアミノ酸全体のmean±SD(平均値±標準偏差)の範囲内にある場合を平均的な値であると判断した。
呼気を採取したUBiT・POCone専用呼気採取バッグ20を、UBiT-IR300赤外分析装置(大塚電子株式会社)に接続したUBiT-IR300専用オートサンプラー UBiT-AS10(大塚製薬株式会社)へ接続し、混合ガス(O2:95%、CO2:5%)を対照として、呼気中のΔ13CO2(‰)(=呼気の13CO2(‰)値から混合ガスの13CO2(‰)値を差し引いた値)を測定した。個体毎に最高Δ13CO2排泄量(以降、Cmaxともいう)、最高Δ13CO2排泄発現時間(以降、Tmaxともいう)及び13Cアミノ酸投与後0~120分間のΔ13CO2測定値-時間曲線の下面積(台形面積法による。以降、AUC120minともいう)を算出した。各アミノ酸の平均値が20種類のアミノ酸全体のmean+SD(平均値+標準偏差)以上である場合を高い値を示したと判断した。また、各アミノ酸の平均値が20種類のアミノ酸全体のmean-SD(平均値-標準偏差)以下である場合を低い値を示したと判断した。各アミノ酸の平均値が20種類のアミノ酸全体のmean±SD(平均値±標準偏差)の範囲内にある場合を平均的な値であると判断した。
(4)結果
どのアミノ酸を投与した場合もカルボキシル基の13C標識に由来する13CO2が呼気中に排泄されたことが示されたことから、いずれのアミノ酸も、アミノ酸それ自体がエネルギーに変換されたこと、すなわちアミノ酸それ自体からエネルギーを産生できることが示された。
どのアミノ酸を投与した場合もカルボキシル基の13C標識に由来する13CO2が呼気中に排泄されたことが示されたことから、いずれのアミノ酸も、アミノ酸それ自体がエネルギーに変換されたこと、すなわちアミノ酸それ自体からエネルギーを産生できることが示された。
20種類のアミノ酸全体のTmaxは、31.9±13.0(平均値±標準偏差)分であった。各種の13Cアミノ酸の投与後に呼気に排泄された13CO2のTmaxは、15~70分に分布した。20種類のアミノ酸全体のmean-SD(18.9分)より短いTmaxを示したアミノ酸は、アラニン、フェニルアラニン、アスパラギン酸、及びアスパラギンであった。また、20種類のアミノ酸全体のmean+SD(44.8分)より長いTmaxを示したアミノ酸は、トリプトファンとアルギニンであった。また、20種類のアミノ酸全体のmean±SDの範囲内(18.9~44.8分)のTmaxを示したアミノ酸は、グリシン、プロリン、リジン等であった。
20種類のアミノ酸全体のCmaxは、173±137(平均値±標準偏差)‰であった。各種の13Cアミノ酸投与後に呼気に排泄された13CO2のCmaxは、40~496 ‰に分布した。20種類のアミノ酸全体のmean+SD(309 ‰)より高いCmaxを示したアミノ酸は、アラニン、フェニルアラニン、グルタミン酸、及びアスパラギンであった。また、20種類のアミノ酸全体のmean±SDの範囲内(36~309 ‰)のCmaxを示したアミノ酸は、グリシン、プロリン、グルタミン酸、リジン、アスパラギン酸、セリン、イソロイシン、バリン、ロイシン、チロシン、システイン、メチオニン、グルタミン、ヒスチジン、及びトレオニンであった。
20種類のアミノ酸全体のAUC120minは、11459±7498(平均値±標準偏差)‰・minであった。各種の13Cアミノ酸の投与後に呼気に排泄された13CO2のAUC120minは、3657~30348‰・minに分布した。20種類のアミノ酸全体のmean+SD(18957‰・min)より高いAUC120minを示したアミノ酸は、アラニン、グルタミン酸、及びアスパラギンであった。また、20種類のアミノ酸全体のmean±SDの範囲内(3961~18957‰・min)のAUC120minを示したアミノ酸は、グリシン、プロリン、グルタミン酸、リジン、アスパラギン酸、セリン、イソロイシン、バリン、ロイシン、チロシン、システイン、メチオニン、グルタミン、及びヒスチジンであった。
以上の結果から、そのアミノ酸自体をエネルギー源としたエネルギー産生の効率がよく、かつそのエネルギー産生のピークを投与後早期にもたらす即効性のアミノ酸として、アラニン、フェニルアラニン、及びアスパラギンを選抜した。また、そのアミノ酸自体をエネルギー源としたエネルギー産生が可能であり、かつそのエネルギー産生のピークを投与後比較的遅い時点でもたらす遅効性のアミノ酸として、トリプトファンとアルギニンを選抜した。これらの即効性と遅効性のアミノ酸を組み合わせることで、持効性のエネルギー産生をもたらすことができる。さらに、そのアミノ酸自体をエネルギー源としたエネルギー産生が可能であり、かつそのエネルギー産生のピークを投与後中間的な時点でもたらすアミノ酸として、トレオニン、ヒスチジン、セリン、リジン、グリシン、アスパラギン酸、システイン、バリン、ロイシン、イソロイシン、プロリン、グルタミン酸、メチオニン、グルタミン及びグルタミン酸を選抜することができた。
さらに、分岐鎖アミノ酸(バリン、ロイシン、イソロイシン)と比較しても、アスパラギン、フェニルアラニン、アラニンは、Cmaxが大きく、Tmaxは小さかった。すなわち、アスパラギン、フェニルアラニン、アラニンは分岐鎖アミノ酸(バリン、ロイシン、イソロイシン)よりも、エネルギー産生が即効性であることが示された。一方、トリプトファンとアルギニンは、分岐鎖アミノ酸(バリン、ロイシン、イソロイシン)と比較して、Cmaxが小さく、Tmaxは大きかった。すなわち、トリプトファンとアルギニンは、分岐鎖アミノ酸(バリン、ロイシン、イソロイシン)よりも、エネルギー産生が遅効性であることが示された。
図2に、アスパラギン、フェニルアラニン、アラニン、トリプトファン及びアルギニンの非糖質性エネルギー産生量の経時的変化をグラフで示した。
本発明に係る非糖質性エネルギー産生増強剤は、アミノ酸によりエネルギー産生を増強することを目的とした医薬品又は食品に利用できる。

- アスパラギン、フェニルアラニン、アラニン、トリプトファン及びアルギニンからなる群から選択される1以上のアミノ酸を含有する非糖質性エネルギー産生増強剤。
- 前記1以上のアミノ酸が、アスパラギン、フェニルアラニン及びアラニンからなる群から選択される、即効性の、請求項1に記載の非糖質性エネルギー産生増強剤。
- 前記1以上のアミノ酸が、トリプトファン及びアルギニンからなる群から選択される、遅効性の、請求項1に記載の非糖質性エネルギー産生増強剤。
- 前記1以上のアミノ酸が、アスパラギン、フェニルアラニン及びアラニンからなる群から選択される1以上のアミノ酸と、トリプトファン及びアルギニンからなる群から選択される1以上のアミノ酸とからなる、持効性の、請求項1~3のいずれか1項に記載の非糖質性エネルギー産生増強剤。
