  • Example II-XII were performed in the same way as example I but comprised variation of type and concentration of chelating agents as well as variations of extraction time. While parameters like protein content, fibrinogen-antigen content or fibrinopeptide-A content were not significantly influenced by these variations when being normalised to lmg fibrinogen, it was observed that the content of compounds of higher molecular weight than fibrinogen (HMW), determined by size exclusion chromatography, exceeded 10% when the concentration of the complexing agent was less than 3 mmol/l.

  • Said modification resulted in an unexpected increase of coagulation factor XIII activity in the final product of the present invention, i.e. up to 0.30 IU/mg fibrinogen (fibrinogen concentration 20-25 mg/ml ; determined by the Clauss method), as well as an increased yield .

  • the protein band at about 100 kD represent the dimer of the fibrinogen ⁇ -chain .
  • the faint band at about 30kD is caused by fibrinogen fragments.

Abstract

Cette invention concerne un procédé de purification de fibrinogène à partir d'une source contenant du fibrinogène par précipitation du fibrinogène par un agent de précipitation à partir d'une solution contenant du fibrinogène en présence d'un ou de plusieurs agents de chélation et l'élimination du surnageant d'une pâte de fibrinogène, ledit procédé étant caractérisé en ce que le fibrinogène est extrait de la pâte pour former une fraction liquide contenant le fibrinogène, et un résidu non dissous, qui est séparé du liquide.
