Definitions
- the present invention relates to an agent for improving and / or preventing cancerous anemia, and more particularly to an agent for improving or preventing cancerous anemia containing 5-aminolevulinic acid (ALA) or a derivative thereof or a salt thereof.
- ALA 5-aminolevulinic acid
- Cancer anemia is anemia peculiar to cancer patients, and is known as anemia that differs greatly from the commonly known iron deficiency anemia and renal anemia. About 80% of people affected with cancer have cancer anemia and anemia worsens as cancer progresses, and anemia progresses further due to anticancer drugs and radiation therapy. It has been known. Cancer anemia is a major problem because it significantly lowers the QOL of cancer patients.
- iron deficiency anemia is often ameliorated by the administration of iron, but iron has little effect on cancer anemia, and iron promotes cancer growth, Iron agents are rarely used for cancerous anemia.
- renal anemia is known to be caused by a decrease in the production of erythropoietin, a hematopoietic factor that promotes differentiation into erythrocytes, and erythropoietin and its derivatives, compounds that stimulate erythropoietin receptors, etc. have been developed and put to practical use. Yes. However, since there is no known example of a decrease in erythropoietin concentration in patients with normal cancer anemia, it is considered that renal anemia and cancer anemia are caused by completely different causes.
- An object of the present invention is to provide an anemia improvement and / or prevention agent useful for improvement and prevention of cancer anemia, which can improve QOL of cancer anemia patients without advancing cancer progression. There is.
- the present inventors scrutinized an enormous amount of data regarding existing studies and therapeutic agents for anemia in general, and further examined the differences between cancerous anemia and other types of anemia, and the causes of anemia in cancer patients. It was concluded that it was not hematopoietic factors or iron. Therefore, as a result of initiating screening of compounds that can improve and / or prevent cancerous anemia other than hematopoietic factors, and as a result of repeated investigations, ALA has a surprising effect of improving cancerous anemia. I found.
- ALA is widely used in photodynamic therapy (PDT) and photodynamic diagnosis (PDD).
- ALA is a common precursor of heme compounds, but in cancer cells, heme is not produced even when ALA is administered, and protoporphyrin IX (PPIX), a precursor of heme compounds, may accumulate.
- PPIX protoporphyrin IX
- PDD protoporphyrin IX
- PDT active oxygen is generated in the presence of oxygen. Since cancerous anemia occurs specifically in cancer patients, the relationship between ALA and PDT and PDD effects is recalled, but light irradiation is not required in the present invention.
- heme is not produced from ALA in cancer cells, but heme is an important component of erythrocytes, so in the case of hematopoietic cell cancer, adding ALA does not lead to hematopoiesis.
- cancer anemia is also seen in cancer patients other than hematopoietic cell cancer, and the fact that heme is not produced from ALA in cancerated hematopoietic cells is considered to cause cancer anemia directly. It is difficult.
- anemia is effective in preventing anemia in piglets (see Japanese Patent No. 4754731).
- Anemia in piglets occurs because hematopoiesis cannot catch up with rapid growth, and it has been reported that the addition of ALA is effective as one of the compounds necessary for hematopoiesis.
- growth does not necessarily increase, so it is difficult to think that ALA improves cancerous anemia with the same mechanism.

From this point onwards, oral intake of aminolevulinic acid phosphate and ferrous sodium citrate per day was started, and after about 20 months after the test on May 20, there was a significant recovery in all red blood cells, hemoglobin, and platelets. The platelet count recovered to normal. Of particular note is the white blood cell count. In leukemia treatment, suppression of the white blood cell count with Ritusakine usually reduces the white blood cell count but does not recover as it is, which often impairs immunity. The possibility of is increased.
- the agent for improving and / or preventing cancerous anemia of the present invention can be advantageously used in the medical field.
