医薬又は医療機器用途等において、環状オレフィン系樹脂を含む材料からなる成形体は、タンパク質等の生物由来の物質(生化学物質)を取り扱う保存容器や分析装置等に使用されている。
例えば、臨床検査等で免疫反応などの分子間相互作用を利用した測定(表面プラズモン共鳴(SPR)測定技術、水晶発振子マイクロバランス(QCM)測定技術、金のコロイド粒子から超微粒子までの機能化表面を使用した測定技術など)において、環状オレフィン系樹脂を含む材料からなる成形体からなるマイクロチップが使用される。
上記したマイクロチップは、典型的には一対のマイクロチップ基板が対向して接着された構造を有し、少なくとも1つの上記マイクロチップ基板の表面に微細な流路(例えば、幅10~数100μm、深さ10~数100μm程度)が形成されている。近年、軽量でありながらガラス基板に比べて破損しにくく、かつ、安価で、紫外線等の透過率が高い環状オレフィン系樹脂を含む材料から成形したマイクロチップ基板を用いたマイクロチップの開発が進められている。
すなわち、マイクロチップを利用した測定の高精度化には検出部の高感度化が必要であるが、検体の一部が検出部に到達するまでの流路(マイクロチャンネル)の表面にて吸着を起こすと測定精度が低下する不具合がある。この不具合を解消するために、マイクロチップに設けられた流路表面を生体物質が吸着し難い表面に改質する方法が求められている。
すなわち、特許文献1においては、環状オレフィン系樹脂を含む材料からなる成形体の表面に対して、1)成形体の表面をプラズマ放電処理する工程1、及び、2)工程1の後に、成形体の表面を強酸と接触させる工程2を含む表面処理が提案されている。
更には、工程2に引き続いて、3)当該表面を特定のフッ素含有シラン化合物と反応させる工程3を行うことが提案されている。
特許文献1に記載された工程1及び工程2による表面処理により、環状オレフィン系樹脂を含む材料からなる成形体の表面への生化学物質の吸着の抑制が成され、更に工程3の処理を引き続き行うことにより、陽イオン物質、例えば金属イオン等の上記成形体への非特異吸着によって生じる生化学分析の障害も回避することが可能となる。
特許文献1に記載されている工程1は、環状オレフィン系樹脂を含む材料からなる成形体に対してプラズマ放電処理する工程である。前記成形体の表面にプラズマ放電処理を行うことにより、成形体表面を活性化する。
すなわち、プラズマ放電により成形体表面にプラズマ粒子が照射されることになるが、その際、プラズマ中の高速粒子が成形体表面に衝突する。その結果基板表面にダメージが加わり、成形体表面状態が荒れることになる。
また、この目的を達成するためのプラズマ放電は真空中で行われる。そのため、プラズマ放電を実施するための真空雰囲気を形成する必要があり、表面処理工程のための設備がおおがかりとなる。よって例えば上記表面処理を施した環状オレフィン系樹脂を含む材料からなる成形体からなるマイクロチップの生産プロセスも複雑かつコストアップとなるので、当該マイクロチップの生産プロセスにおいては、真空中の処理を避けたい。
特許文献1に記載されている工程2は、酸処理により、環状オレフィン系樹脂を含む材料からなる成形体に対して、酸処理を施している。
酸処理は、人体のみならず機器類に対しても腐食する影響が考えられる上に、工業廃液の問題も生じる問題がある。さらに、多機能マイクロチップは、一つのマイクロチップ中に様々なセンサデバイスを組み込む。組み込んだデバイスを酸によって腐食させたくないため、この酸処理のプロセスを避けたい。
すなわち、当該流路表面は、プラズマ放電処理に起因する表面荒れ状態にある。
上記したように流路表面に表面荒れがあると、流路内を流す機能性分子の流れがスムーズでなくなり、流速等が時間的に不均一な流れとなる。よって、場合によっては、流速等の時間的不均一により、精度の高い測定が困難となる場合がある。
また、例えば環状オレフィン系樹脂を含む材料からなる成形体としてマイクロチップ基板を成形した場合においても、マイクロチップ内の流路表面の表面荒れを回避して、流路内を流す機能性分子の流れをスムーズにすることができる環状オレフィン樹脂を含む材料からなる成形体の表面の表面処理方法および該方法により生体機能分子の固定機能性が付与された環状オレフィン系樹脂を含む材料からなる成形体を提供することである。
本発明の環状オレフィン系樹脂を含む材料からなる成形体の表面処理方法において、従来の環状オレフィン系樹脂を含む材料からなる成形体の表面に対して成形体の表面をプラズマ放電処理する工程(工程1)に代えて、当該表面に対し真空紫外光を照射する工程(以下、工程A)を採用する。
真空紫外光としては、具体的には波長200nm以下の光(好ましくは波長180nm以下の光)が、上記表面に照射される。例えば、後述する実施の形態で示すように、真空紫外エキシマランプから放出される波長172nmに中心波長を有する単色光の光が上記表面に照射される。
更に、工程Aによって表面処理された成形体表面に対して自己組織化単分子膜を形成する工程Bを実施する。ここで、工程Aの処理が実行された環状オレフィン系樹脂からなる成形体表面の酸素と一端の官能基とが結合した自己組織化単分子膜の他端の官能基として、任意の官能基(例えば、カルボキシ基、アミノ基、メチル基など)を選択することにより、各官能基の機能に基づく機能性を環状オレフィン系樹脂からなる成形体表面に付与することが可能となる。例えば、工程Bとして、特許文献1と同様、先の工程で表面処理された成形体表面を特定のフッ素含有シラン化合物と反応させる工程3を採用することにより、成形体の表面への生化学物質の吸着の抑制を実現することが可能となる。
本発明者らが鋭意研究した結果、本発明にて採用した上記工程Aは、大きく2つの効果を奏することが確認された。
(i)従来のプラズマ放電処理と同様、真空紫外光の環状オレフィン系樹脂成形体表面への照射により、当該成形体表面が活性化する。具体的には、当該成形体表面におけるオレフィン環の開裂が発生する。
(ii)更には、上記活性化した成形体表面への酸素導入も、真空紫外光の照射処理により成し遂げられる。すなわち、従来技術における工程2の作用も同時に発生することが判明した。
(1)環状オレフィン樹脂を含む材料からなる成形体の表面の表面処理方法であって、環状オレフィン樹脂を含む材料からなる成形体表面に真空紫外光を照射し、当該照射表面に自己組織化単分子膜を形成する。
(2)上記(1)において、上記真空紫外光を上記表面の一部に選択的に照射し、この選択的照射表面に自己組織化単分子膜を形成する。
(3)上記(2)において、上記表面における照射領域が2箇所以上であって、この複数の照射領域に自己組織化単分子膜を形成する。
(4)上記(1)(2)(3)において、上記成型体は、少なくとも一方に微細な流路が形成されている第1の基板と第2の基板からなるマイクロチップ基板である。
(5)上記(1)(2)(3)(4)の成形体の表面処理を、当該成形体表面の生化学物質の非吸着化処理とする。
(6)上記(5)において、上記自己組織化単分子膜形成工程は、前記成形体の表面と下式[1]で表されるフッ素化合物を反応させる工程である。

(1)環状オレフィン樹脂を含む材料からなる成形体表面に真空紫外光を照射しているので、成形体表面の表面荒れを生じさせることなく、成形体表面に自己組織化単分子膜を形成することが可能となる。
したがって、例えば環状オレフィン系樹脂を含む材料からなる成形体としてマイクロチップ基板を成形し、当該マイクロチップ基板を貼り合わせて内部に機能性分子などの試験体を流す流路を含むマイクロチップを構成する場合、このマイクロチップ内の流路表面の表面荒れを生じさせることがなく、生化学物質の吸着の抑制するための表面処理を行うことが可能となる。このため、流路内を流す機能性分子の流れがスムーズとなり、機能性分子の流速等の特性を均一にすることができ、精度の高い測定が可能となる。
(2)環状オレフィン樹脂を含む材料からなる成形体表面に真空紫外光を照射することで、従来技術の工程2に相当する活性化した成形体表面への酸素導入もなされるので、従来のような酸素導入プロセスが不要となり、処理工程の簡略化が達成される。
(3)真空紫外光を照射する処理であるので、成型体表面の一部分に選択的に自己組織化単分子膜を形成させることができる。このため、例えばマイクロチップ基板において、試験体を流す流路部分等のみに自己組織化単分子膜を形成することができる。したがって、両マイクロチップ基板の接合等に不具合が発生することがない。
(4)本発明においては、工程Aによって表面処理された成形体表面に対して自己組織化単分子膜を形成する工程Bを実施しており、工程Bとして例えば上記成形体表面を特定のフッ素含有シラン化合物と反応させる工程を実施することにより、陽イオン物質、例えば金属イオン等の上記成形体への非特異吸着によって生じる生化学分析の障害も回避することが可能となる。
(5)本発明においては、工程Aによって表面処理された成形体表面に対して自己組織化単分子膜を形成する工程B’を実施しており、工程B’として例えば上記成形体表面を特定のアミノ基含有シラン化合物と反応させる工程を実施することにより、上記成形体表面に抗体などの生体機能分子の固定することが可能となる。環状オレフィン系樹脂は透明であるので、上記した成形体は、光透過型の測定や蛍光測定にも応用可能であり、広範なバイオセンシングへの応用を期待することができる。
[環状オレフィン系樹脂]
環状オレフィン系樹脂とは、主鎖が炭素-炭素結合からなり、主鎖の少なくとも一部に環状炭化水素構造を有する高分子化合物である。この環状炭化水素構造は、ノルボルネンやテトラシクロドデセンに代表されるような、環状炭化水素構造中に少なくとも一つのオレフィン性二重結合を有する化合物(環状オレフィン)を単量体として用いることで導入される。
環状オレフィンの具体例としては、シクロペンテン、シクロヘキセン、シクロオクテン;シクロペンタジエン、1,3-シクロヘキサジエン等の1環の環状オレフィン;
環状オレフィン系樹脂は、好ましくは、エチレンとノルボルネンの付加共重合体、又は、エチレンとテトラシクロドデセンの付加共重合体である。
環状オレフィン系樹脂の構造には、特に制限はなく、鎖状でも、分岐状でも、架橋状でもよいが、好ましくは直鎖状である。
本発明において、環状オレフィン系樹脂は、本発明の目的を阻害しない範囲の種類及び量の他の熱可塑性樹脂をブレンドした組成物として用いてもよい。環状オレフィン系樹脂と他の熱可塑性樹脂からなる樹脂組成物は、例えば、一軸押出機や二軸押出機等を用いて溶融混練することにより調製できる。
本発明の方法により生化学物質の非吸着化処理される成形体は、上記の環状オレフィン系樹脂を、公知の方法により成形することにより製造される。公知の成形方法としては、例えば、射出成形、射出圧縮成形、ガスアシスト法射出成形、押出成形、多層押出成形、回転成形、熱プレス成形、ブロー成形、発泡成形等の方法が挙げられる。
工程Aは環状オレフィン系樹脂成形体表面に真空紫外光を照射する工程である。当該表面への真空紫外光の照射により、当該表面は活性化される。
光源としては、例えば中心波長が172nmの真空紫外光を放出するエキシマランプを使用する。
放電空間Sにはキセノン、アルゴン、クリプトンなどの希ガスが封入される。容器11は石英ガラスからなる。内側管111の内周面には内側電極12が設けられ、外側管112の外周面には網状の外側電極13が設けられる。これら電極12,13は容器11と放電空間Sを介在されて配置していることになる。電極12,13は、リード線W11,W12を介して電源装置16が接続される。
電源装置16より高周波電圧が印加されると、電極12,13間に誘電体(111,112)を介在させた放電(いわゆる誘電体バリア放電)が形成され、キセノンガスの場合は中心波長172nmの真空紫外光が発生し、外部へ当該真空紫外光が放射される。
試料は環状オレフィン系樹脂としてTOPAS8007S-04(ポリプラスチックス株式会社製)を用いて成形した成形体であり、その形状は厚み10mm、縦100mm、横100mmの正方形の基板である。
図4、図5に示すように、XPS測定は、中心波長172nmの真空紫外光の照射前、30分照射後、60分照射後に実施した。図4、図5において、Aは照射なし、Bは30分照射、Cは60分照射の場合を示す。
図4に示すC1s測定では、真空紫外光の照射前に1本のepoxy由来と思われる構造のピークが得られた。また、真空紫外光を30分間および60分間、成形体表面に照射すると、真空紫外光の照射前に観察された炭素の1つのピークは、2つに分かれた。これは、環状オレフィン系樹脂成形体表面に真空紫外光を照射することにより、オレフィン環の開裂が発生したためであると考えられる。
また、真空紫外光を30分間および60分間、成形体表面に照射すると、真空紫外光の照射前に観察された酸素の1つのピークは大きく増大した。すなわち、大気中で真空紫外光を環状オレフィン系樹脂からなる成形体表面に照射すると、当該表面に酸素が導入されることが判明した。
次に工程Bについて説明する。
工程Bは、工程Aにより活性化した成形体表面と下記の式(1)で表されるフッ素化合物を反応させる工程である。本発明では、工程Aにより成形体表面に導入された酸素含有基と、式(1)で表されるフッ素化合物を反応させることにより、成形体表面に極性の小さなフッ素含有基を導入し、生化学物質の非吸着化を実現させるものである。

式(1)で表されるフッ素化合物のうち好ましいものとしては、例えば、3,3,3-トリフルオロプロピルトリクロロシラン、2,2,3,3,3-ペンタフルオロプロピルトリクロロシラン、1H,1H,2H,2H-パーフルオロデシルジメチルクロロシラン、1H,1H,2H,2H-パーフルオロデシルメチルジクロロシラン、1H,1H,2H,2H-パーフルオロデシルトリクロロシラン、1H,1H,2H,2H-パーフルオロオクチルジメチルクロロシラン、1H,1H,2H,2H-パーフルオロオクチルメチルジクロロシラン、1H,1H,2H,2H-パーフルオロオクチルトリクロロシラン、3,3,4,4,5,5,6,6,6-ノナフルオロヘキシルトリクロロシラン、3,3,4,4,5,5,6,6,6-ノナフルオロヘキシルメチルジクロロシラン等のフルオロアルキルトリハロシラン類;3,3,3-トリフルオロプロピルトリエトキシシラン、2,2,3,3,3-ペンタフルオロプロピルトリエトキシシラン、パーフルオロメチルトリエトキシシラン、1H,1H,2H,2H-パーフルオロデシルトリエトキシシラン、1H,1H,2H,2H-パーフルオロオクチルトリエトキシシラン、1H,1H,2H,2H-パーフルオロオクチルトリメトキシシラン等のフルオロアルキルトリアルコキシシラン類等が挙げられる。
工程Aを実施することにより活性化された成形体の表面と、式(1)で表されるフッ素化合物を反応させる方法は、反応が良好に進行する限り特に制限されない。式(1)で表されるフッ素化合物は、メタノール、エタノール等の有機溶媒により希釈されたフッ素化合物溶液として使用するのが好ましい。フッ素化合物溶液の濃度は0.01mMから10mMの範囲が好ましい。フッ素化合物溶液中のフッ素化合物の濃度が低すぎる場合、十分な生化学物質の非吸着化の効果が得られなかったり、反応に長時間を要したりする場合がある。
成形体表面とフッ素化合物との反応条件は、反応が良好に進行する限り特に制限されない。通常、成形体表面とフッ素化合物との反応は、0°Cから50°C、より好ましくは15から35°Cの温度において、15分から12時間、より好ましくは30分から2時間の条件で行われる。
試料としては、環状オレフィン系樹脂としてTOPAS8007S-04(ポリプラスチックス株式会社製)を用いて成形した成形体であって、その形状が厚み10mm、縦10mm、横10mmの正方形の基板を使用した。工程Aの処理としては、上記したようなエキシマランプを用いて、中心波長172nmの真空紫外光を15分間から60分間照射した。なお、試料表面における放射照度は10mW/cm2から20mW/cm2であった。
次に、工程Bとして、上記のように処理された試料をパーフルオロブチルトリクロロシランの濃度0.0001Mから0.005Mのエタノール溶液に室温で1時間浸漬する処理を実施した。
工程Bを経た試料の表面状態の調査としては、上記と同様、リガク社製XPS-7000型X線電子分光(XPS)装置を用いた当該表面のXPS測定を行った。
図6に工程Bによる処理後のプレートのO1sピークに対応する結合エネルギー領域のXPS測定結果を示し、図7に工程Bによる処理後のプレートのF1sピークに対応する結合エネルギー領域のXPS測定結果を示す。図6ではO-Si結合に対応するピークが観察され、図7ではCF2に対応するピークが観察されたことから、プレート表面(環状オレフィン系樹脂成形体表面)にフッ化炭素基が導入されたことが示された。
次に、工程Aおよび工程Bの処理を行った環状オレフィン系樹脂からなる成形体表面に対する生体物質の吸着性について調査した。
調査は、本発明の表面処理を行っていない環状オレフィン系樹脂からなる成形体、並びに、本発明の表面処理(工程Aの処理ならびにそれ引き続き行う工程Bの処理)を行った成形体プレートについて行った。
調査の具体的手順を図8に示す。
図8に示すように、成形体として板状体(プレート)の基板K1,K2を成形し、一方の基板K1にエキシマランプ10から真空紫外光を照射し(図8(a)参照)、フッ素化合物溶液(SAM溶液)に基板K1を浸漬して、フッ素化合物溶液とを反応させて、官能基としてフッ化炭素基を有する自己組織化単分子膜(以下、SAM膜とも言う)コーティングを行った(図8(b)参照)。
洗浄後の両基板K1,K2表面にハンディUVランプ(UVP社製UVGL-58 365nm長波長タイプ)をあて、440nmの蛍光を観察した(図8(d)参照)。
上記調査は、再現性を得るため、本発明の表面処理を施した基板K1および未処理の基板K2を、それぞれ3枚を別々に用意して行った。
未処理の基板K2と、本発明の表面処理を施した基板K1を観察した結果、未処理基板K2表面では3枚の基板全てで440nmの蛍光が観測されたことから、表面に蛍光標識化ウシ血清アルブミンの凝集体が存在することが確認された。
また、本発明の表面処理が施された基板K1表面は一枚も蛍光を示さない事から、蛍光標識化ウシ血清アルブミンが全く存在しないことがわかった。
以上の結果から、本発明の表面処理を施すことによりウシ血清アルブミンの吸着が抑制されることが確認された。
よって、特許文献1に記載されているような酸素導入プロセスが不要となり、処理工程の簡略化を達成することができる。
よって、例えば環状オレフィン系樹脂を含む材料からなる成形体としてマイクロチップ基板を成形し、当該マイクロチップ基板を貼り合わせて内部に機能性分子などの試験体を流す流路を含むマイクロチップを構成する場合、このマイクロチップ内の流路表面の表面荒れを回避することが可能となるので、流路内を流す機能性分子の流れがスムーズとなる。そのため、上記機能性分子の流速等の特性を均一にすることができ、精度の高い測定が可能となる。
ここで、自己組織化単分子膜他端の官能基をフッ化炭素基とし、上記成形体表面にフッ化炭素基を導入することにより、生化学物質の非特異吸着を防止できるとともに、成形体表面への陽イオン物質の非特異吸着を抑制することができる。
特に生化学物質を含む試料の微量分析等に用いられるマイクロチップ用の基板の表面を本発明の方法により処理した場合は、試料中の生化学物質の基板表面への吸着が大きく抑えられることにより、分析精度の高い高性能なマイクロチップを得ることができる。
(1)工程Aを採用した環状オレフィン系樹脂を含む材料からなる成形体の表面処理方法によれば、従来技術同様、上記成形体の表面への生化学物質の吸着の抑制を実現することが可能となる。また、工程Aの採用により、プラズマ放電処理とは異なり、上記成形体表面の表面荒れが発生しない。よって、例えば環状オレフィン系樹脂を含む材料からなる成形体としてマイクロチップ基板を成形し、当該マイクロチップ基板を貼り合わせて内部に機能性分子などの試験体を流す流路を含むマイクロチップを構成する場合、このマイクロチップ内の流路表面の表面荒れを回避することが可能となるので、流路内を流す機能性分子の流れがスムーズとなる。そのため、上記機能性分子の流速等の特性を均一にすることができ、精度の高い測定が可能となる。
(3)本発明は、工程Aによって表面処理された成形体表面に対して自己組織化単分子膜を形成する工程Bを実施する。工程Bとして例えば上記成形体表面を特定のフッ素含有シラン化合物と反応させる工程を実施することにより、陽イオン物質、例えば金属イオン等の上記成形体への非特異吸着によって生じる生化学分析の障害も回避することが可能となる。
すなわち、自己組織化単分子膜を積層させて、巨視的にみて分子量の大きい自己組織化単分子膜を形成してもよい。当然ながら、上記したように規定した自己組織化単分子膜要素は3つ以上でもよい。
先に述べた実施の形態においては、工程Bの自己組織化単分子膜を形成するにあたり工程Aの処理が施された環状オレフィン系樹脂からなる成形体表面とフッ素化合物とを反応させた例を示したが、以下に示す実施の形態においては、自己組織化単分子膜を形成するにあたり工程Aの処理が施された環状オレフィン系樹脂からなる成形体表面とアミン末端、トシル末端、もしくはカルボキシ末端のシラン系化合物を反応させた例を示す。以下、このようなシラン系化合物を上記成形体表面と反応させる工程を工程B’と称する。
しかし、金基板は、光の透過性に乏しく、紫外線や可視光を用いた光透過型の計測に応用することは非常に難しい。また、金は蛍光を消光してしまう性質があるため、蛍光分析への応用ができない。
本センシング方法によれば、蛍光測定は可能であるもののポリプロピレン等の樹脂の光透過性は不十分であり、必ずしも光透過型の測定が可能というわけではない。
先に示した実施の形態と同様、工程Aは環状オレフィン系樹脂成形体表面に真空紫外光を照射する工程である。当該表面への真空紫外光の照射により、当該表面は活性化される。
光源としては、例えば中心波長が172nmの真空紫外光を放出するエキシマランプを使用する。
〔工程B’〕
本実施の形態においては、工程Aの処理が施された環状オレフィン系樹脂からなる成形体表面とフッ素化合物とを反応させる工程Bに代えて、シラン系化合物を上記成形体表面と反応させる工程B’を採用するものである。
すなわち、工程B’は、工程Aにより活性化した成形体表面と下記の式(2)で表されるアミン末端、トシル末端もしくはカルボキシ末端のシラン系化合物を反応させる工程である。本実施の形態では、工程Aにより成形体表面に導入された酸素含有基と、式(2)で表されるシラン系化合物を反応させることにより、成形体表面にシラン系化合物の分子膜を形成するものである。さらに工程B’にあと、アミドカプリング反応を利用して、当該成形体表面を標識化した抗体もしくはタンパク質等を固定可能な状態とするものである。
(NH2)Si(Rc)3・・・(2)
[Rcはアミノ基、トシル基、カルボキシル基から選択される基である。]
工程Aを実施することにより活性化された成形体の表面と、式(2)で表されるシラン系化合物を反応させる方法は、反応が良好に進行する限り特に制限されない。式(2)で表されるアミノ基含有シラン化合物は、メタノール、エタノール等の有機溶媒により希釈されたアミノ基含有シラン化合物溶液として使用するのが好ましい。アミノ基含有シラン化合物溶液の濃度は0.01mMから10mMの範囲が好ましい。アミノ基含有シラン化合物溶液中のアミノ基含有シラン化合物の濃度が低すぎる場合、十分な抗体等の固定可能化の効果が得られなかったり、反応に長時間を要したりする場合がある。
成形体表面とアミノ基含有シラン化合物との反応条件は、反応が良好に進行する限り特に制限されない。通常、成形体表面とアミノ基含有シラン化合物との反応は、18°Cから160°Cで行われ、反応時間は5分間から12時間、より好ましくは40分間から1時間の条件で行われる。
試料としては、環状オレフィン系樹脂としてTOPAS8007S-04(ポリプラスチックス株式会社製)を用いて成形した成形体であって、その形状が厚み10mm、縦10mm、横10mmの正方形の基板を使用した。
工程Aの処理としては、上記したようなエキシマランプを用いて、中心波長172nmの真空紫外光を環状オレフィン系樹脂成形体である基板に30分間から60分間照射した。なお、試料表面における放射照度は10mW/cm2から20mW/cm2であった。
次に、工程B’として、上記のように処理された基板をアミノ基含有シラン化合物である3-アミノプロピルトリメトキシシラン(3-(Trimethoxysilyl)-1-propanamine:(CH3O)3Si(CH2)3NH2、東京化成工業株式会社製)の濃度2mMから10mMのエタノール溶液に室温で1時間から24時間浸漬する処理を実施した。浸漬後は、エタノールで十分に洗浄し、窒素で乾燥させた。
工程B’の処理が施された基板の表面は、アミノ末端の自己組織化単分子膜が形成される。このように処理された基板表面に生体機能物質を固定化するために、以下の手順で上記アミノ末端をスクシンイミド化した。
すなわち、上記のように処理された基板を、濃度50μg/mLから500μg/mLの
N-(3-ジメチルアミノプロピル)-N’-エチルカルボジイミド塩酸塩(EDAC:N-(3-Dimethylaminopropyl)-N’-ethylcarbodiimide hydrochloride,Sigma-Aldrich社製)と、濃度50μg/mLから500μg/mLのN-ヒドロキシスクシンイミド(NHS:N-Hydroxysuccinimide,Fluka社製)を含むトリス(tris(hydroxymethyl)aminomethane)緩衝溶液中に浸漬し、上記アミノ末端をスクシンイミド化した。
表面に形成された自己組織化単分子膜のアミノ末端をスクシンイミド化した基板は、トリス緩衝溶液とリン酸(Phosphate)緩衝溶液で十分に洗浄した。
上記の基板における、末端をスクシンイミド化した機能性自己組織化単分子膜表面に、濃度10ppmのマウスモノクローナル抗体IgG1(monoclonal Mouse-IgG1, Funakoshi株式会社製)を含むリン酸緩衝溶液を1mL滴下して、30分間、常温でスクシンイミドとマウスモノクローナル抗体IgG1とのアミドカプリング反応を維持した。
このアミドカプリング反応により、マウスモノクローナル抗体IgG1(以下、mouseIgG1という)とスクシンイミドが置換する反応が生じて、mouse IgG1 が基板の表面に固定化される。
mouse IgG1が表面に固定化された上記基板の反応性を調査するために、上記基板表面に濃度10ppmの抗マウスIgG1ヤギポリクオーナル抗体(anti-mouse IgG1, Goat-polyclonal, Funakoshi株式会社製、以下anti-mouse IgG1と称する)を含むリン酸緩衝溶液を1mL滴下して、30分間、常温で表面のmouse IgGとanti-mouse IgG1との抗原抗体反応を維持した。その後、基板表面をリン酸緩衝溶液で十分に洗浄した。
以下、(1)工程B’を経た時点、(2)スクシンイミド化工程を経た時点、(3)生体機能分子の固定工程を経た時点、(4)抗体抗原反応を経た時点それぞれにおける基板の表面状態の調査を行った。表面状態の調査としては、先の実施の形態のときと同様、リガク社製XPS-7000型X線電子分光(XPS)装置を用いた当該表面のXPS測定を行った。
なお、工程Aを経た時点での表面状態は、先の実施例のときと同様であったので詳細な説明は省略する。すなわち、図4、図5に示すXPS測定結果と同様、大気中で環状オレフィン系樹脂成形体表面に真空紫外光を照射することにより当該表面におけるオレフィン環の開裂がなされるとともに、当該表面への酸素導入がなされることが判明した。
図10に示すように環状オレフィン樹脂には含まれないN1sのピークが観測され、また図11に示すように環状オレフィン樹脂には含まれないSi2pのピークが観測された。
以上の結果により、工程Aによってオレフィン環の開裂され、かつ、酸素導入された基板表面の酸素と式(2)で表されるアミノ基含有シラン化合物とのシランカップリング反応により、基板表面にアミノ末端を有する機能性シランの自己組織化単分子膜が形成されていることが裏付けられた。
図10に示すN1sピーク強度は、アミノ末端のアルカンチオールのN1sのピーク強度とほぼ同じであった。このことから、表面へ固定化されたアミノ末端を有する機能性シランの結合量は、アルカンチオール単分子層と同程度の10-10mol/cm2であると考えられる。
図11に示すように、工程B’を経た時点と比較すると、スクシンイミド化工程を経た時点においてSi2pのピーク強度が減少しているのが観測される。これは、シラン化合物からなる自己組織化単分子膜の表面をスクシンイミド基が覆っているため、Si2pからの光電子が減衰したことを示唆している。
また、図10に示すように、工程B’を経た時点と比較すると、スクシンイミド化工程を経た時点においてN1sのピークが高エネルギー側へシフトしていることが観測された。これは、基板表面の末端を修飾していたアミノ基がスクシンイミド基へと構造が変わったためである。
図11に示すように、工程B’を経た時点、スクシンイミド化工程を経た時点と比較すると、生体機能分子(抗体)の固定工程を経た時点において、Si2pのピーク強度が更に減少しているのが観測される。これは、厚みのあるマウスモノクローナル抗体IgG1(mouseIgG1)が基板表面を覆い、Si2pからの光電子が更に減衰したことを示唆している。
また、図10に示すように、工程B’を経た時点、スクシンイミド化工程を経た時点と比較すると、生体機能分子(抗体)の固定工程を経た時点において、N1sのピークが更に高エネルギー側へシフトしていることが観測された。これは、基板表面がアミド結合を持つ化合物で覆われたことを示唆している。
図10に示すように、生体機能分子(抗体)の固定工程を経た時点と比較すると、抗体抗原反応を経た時点において、N1sのピーク強度が2倍に増加していることが観測された。これにより、1つのmouseIgG1に対して抗マウスIgG1ヤギポリクオーナル抗体(nti-mouse IgG1)が1つ結合していることが分かった。
また、図12、図13に示すように、生体機能分子(抗体)の固定工程を経た時点と抗体抗原反応を経た時点における基板表面状態を比較すると、mouse IgG1に対応するO1sピーク形状とanti-mouseIgG1に対応するO1sピーク形状が相違し、mouse IgG1に対応するC1sピーク形状とanti-mouse IgG1に対応するC1sピーク形状が相違することが観測された。しかしながら、両抗体の構造は非常に複雑であるため、各ピーク形状が抗体のどの部位由来のものであるかは同定することは難しい。なお、これらの結果から、mouse IgG1とanti-mouse IgG1とでは、カルボキシ基の量が相違することが示唆される。
よって、特許文献1に記載されているような酸素導入プロセスが不要となり、処理工程の簡略化を達成することができる。
よって、例えば環状オレフィン系樹脂を含む材料からなる成形体としてマイクロチップ基板を成形し、当該マイクロチップ基板を貼り合わせて内部に機能性分子などの試験体を流す流路を含むマイクロチップを構成する場合、このマイクロチップ内の流路表面の表面荒れを回避することが可能となるので、流路内を流す機能性分子の流れがスムーズとなる。そのため、上記機能性分子の流速等の特性を均一にすることができ、精度の高い測定が可能となる。
ここで、自己組織化単分子膜他端の官能基をアミノ基とし、上記成形体表面にアミノ基を導入し、当該表面上のアミノ末端をスクシンイミド化することにより、成形体表面に抗体などの生体機能分子の固定することができる。
なお、上記した成形体表面に生体機能分子の固定機能性を付与する例では、工程B’の処理を行うことにより、アミノ末端の自己組織化単分子膜を成形体表面に形成し、アミノ末端をスクシンイミド化して抗体を固定したがこれに限るものではない。例えば、成形体表面にカルボキシ末端あるいはトシル基末端の自己組織化単分子膜を形成してもよい。
また、抗体のみならず、無機物、例えば、貴金属の固定機能性を成形体表面に付与する場合は、成形体表面にチオール末端の自己組織化単分子膜を成形体表面に形成してもよい。また、アミンを含む有機分子の固定機能性を成形体表面に付与する場合は、アルデヒド基末端の自己組織化単分子膜を成形体表面に形成してもよい。
以下、本発明を適用した環状オレフィン系樹脂から成形したマイクロチップの製造工程例を示す。
図14にマイクロチップの構造例を示す。図14(a)はマイクロチップ30の上面図であり、図14(b)は、図14(a)におけるA-A断面図である。
図14(b)に示すように、上記マイクロチップ30は第1のマイクロチップ基板31と第2のマイクロチップ基板32とが積層されて接合された構造を有しており、上記した第1のマイクロチップ基板31および第2のマイクロチップ基板32はいずれも環状オレフィン系樹脂からなる成形体である。
図示は省略されているが、この2流路33の内部には、試薬が配置された反応領域などの各種機能を有する領域(検出部)が設けられている。この流路の流入口33aより検体を流し、当該検体と試薬との反応が検出部において観察・測定される。例えば、臨床検査等における免疫反応などの分子間相互作用を利用した測定が行われる。流入口33aより流路に供給した検体は、流出口33bより排出される。
上記したように、マイクロチップ30は第1および第2のマイクロチップ基板31,32を接合して形成されるものであり、SAM膜34は流路表面の少なくとも一部に選択的に形成することが望ましい。なぜならば流路表面以外の領域の一部は両マイクロチップ基板の接合面となるので、接合面にもSAM膜34が形成された場合、両マイクロチップ基板の接合に不具合が発生する可能性があるためである。
図16(a)は、本発明における工程Aを説明する図である。
工程Aを実施するにあたり、ワークステージ40上に第1のマイクロチップ基板31、第2のマイクロチップ基板32が設置される。その上方に図15に示すマスク43が配置され、さらにマスク43の上方に例えば真空紫外光を放出するエキシマランプ10を複数本収容したランプハウス41,42が設置される。
第1および第2のマイクロチップ基板31,32は、SAM膜形成領域に相当する真空紫外光の被照射領域Rがランプハウス41,42内のエキシマランプ10に対向するように、ワークステージ40上に設置される。
エキシマランプ10としては、例えば、図1に示す構造のものが使用され、エキシマランプの放電空間S(図1参照)には、キセノンガスが封入される。このようなエキシマランプからは、中心波長172nmの真空紫外光が放出される。
第1および第2のマイクロチップ基板31,32の被照射領域における中心波長172nmの真空紫外光の放射照度は、例えば20mW/cm2であり、照射時間は60分である。
工程Bは、工程Aにおける真空紫外光照射処理後の環状オレフィン系樹脂表面上にSAM膜34を形成する工程である。ここでは、例として、当該表面とフッ化化合物溶液とを反応させて、最表面上に官能基としてフッ化炭素基を有するSAM膜34を形成する場合を示す。
図16(b)において、中心波長172nmの真空紫外光が選択的に照射された第1および第2のマイクロチップ基板31,32は、フッ素化合物溶液45(パーフルオロブチルトリクロロシランの濃度0.005Mのエタノール溶液)によって満たされた槽44内に浸漬される。具体的には、室温で1時間浸漬される。このような工程Bを経ることにより、第1および第2のマイクロチップ基板31,32における真空紫外光の被照射領域に、最表面上にフッ化炭素基を有するSAM膜34が選択的に形成される。
なお、真空紫外光による活性化を利用して基板を接合する場合、工程Bにより生成したSAM膜に真空紫外光が照射されるとSAM膜自体の機能(図17に示す例では、生化学物質や陽イオン物質の流路表面への非特異吸着の抑制)に支障をきたす可能性も考えられる。よって、第1および第2のマイクロチップ基板31,32を接合するために両基板の接合面に真空紫外光を照射する場合、この真空紫外光がSAM膜形成領域に照射されないよう遮光することが好ましい。
図18に希ガス蛍光ランプの構成例を示す。同図(a)は管軸を含む平面で切った断面図を示し、(b)は(a)のA-A線断面図を示す。図18において、ランプ20は一対の電極22、23を有し、電極22、23は容器(発光管)21の外周面に配設され、電極22,23の外側には保護膜24が設けられる。容器21の内周面の光出射方向側に対して反対側の内面に紫外線反射膜25が設けられ(図18(b)参照)、その内周に低軟化点ガラス層26が設けられ、この低軟化点ガラス層26の内周面に、蛍光体層27が設けられる。
その他の構成は図1に示したものと同様であり、容器21内の放電空間Sに封入されるガス、蛍光体層27に用いられる蛍光体も同様である。
また、成形体表面における真空紫外光の照射領域が小さい場合は、真空紫外光を放出する光源として、放射光の波長領域内に真空紫外光波長を含む重水素ランプを使用することも可能である。

Claims (12)
- 環状オレフィン樹脂を含む材料からなる成形体表面に真空紫外光を照射し、
当該照射表面に自己組織化単分子膜を形成する
ことを特徴とする環状オレフィン樹脂を含む材料からなる成形体の表面の表面処理方法。 - 上記真空紫外光を上記表面の一部に選択的に照射し、この選択的照射表面に自己組織化単分子膜を形成する
ことを特徴とする請求項1に記載の環状オレフィン樹脂を含む材料からなる成形体の表面処理方法。 - 上記表面における照射領域が2箇所以上であって、この複数の照射領域に自己組織化単分子膜を形成する
ことを特徴とする請求項2に記載の環状オレフィン樹脂を含む材料からなる成形体の表面処理方法。 - 上記成型体は、少なくとも一方に微細な流路が形成されている第1の基板と第2の基板からなるマイクロチップ基板である
ことを特徴とする請求項1,2または請求項3に記載の環状オレフィン樹脂を含む材料からなる成形体の表面処理方法。 - 請求項1,2,3または請求項4の記載の成形体の表面処理が当該成形体表面の生化学物質の非吸着化処理である
ことを特徴とする環状オレフィン樹脂を含む材料からなる成形体の表面処理方法。 - 上記自己組織化単分子膜形成工程は、前記成形体の表面と下式(1)で表されるフッ素化合物を反応させる工程である
ことを特徴とする請求項5に記載の環状オレフィン樹脂を含む材料からなる成形体の表面処理方法。
