Abstract
Description
またα-シヌクレインに限らず、アルツハイマー病、プリオン病、ポリグルタミン病、ハンチントン病などの発症には、それぞれ、βアミロイドペプチド及びタウタンパク質、プリオンタンパク質、ポリグルタミンペプチド、ハンチンチンなどのアミロイドタンパク質の蓄積が深く関っている。これらのアミロイドタンパク質もまた、体内において可溶化形態で常在しているタンパク質であり、何らかの刺激をきっかけにコンフォメーションを変化させてオリゴマーを形成し、その後、不溶化形態のアミロイド線維となる。
従って本発明は、アミロイドタンパク質オリゴマーを特異的に認識可能なアプタマー及びこれを用いたアミロイドタンパク質オリゴマーの検出方法を提供することを目的とする。
[1] Gカルテット構造を有し、かつ、以下のポリヌクレオチドからなる群より選択された少なくとも1つであり、アミロイドタンパク質オリゴマーに対する結合性を有するアプタマー:
(1) 配列番号1~配列番号18のいずれかで示される塩基配列からなるポリヌクレオチド、
(2) 少なくとも2連続のグアノシンヌクレオチドを、少なくとも4つ含み、配列番号1~配列番号18のいずれかで示される塩基配列において1又は数個の塩基が欠失、置換、又は付加された塩基配列からなるポリヌクレオチド、並びに、
(3) 前記(1)又は(2)のポリヌクレオチドを構成単位として含む多量体であるポリヌクレオチド。
[2] 前記(3)のポリヌクレオチドが、上記(1)又は(2)のポリヌクレオチドと、各(1)又は(2)のポリヌクレオチド間を連結するリンカー配列とを有する[1]記載のアプタマー。

更に説明すれば、上述したようにアミロイドタンパク質は、モノマー型、オリゴマー型及び線維型にコンフォメーションを変更する。Gカルテット構造を有する特定の前記アプタマーは、オリゴマー型とモノマー型に対する結合能が異なることが本発明において見出された。このようなGカルテット型アプタマーを用いることにより、オリゴマー型のアミロイドタンパク質を特異的に識別することができる。
なお、本明細書における「アプタマー」とは、特定の分子に結合する核酸リガンドを表す。
また、本明細書において「~」を用いて示された数値範囲は、「~」の前後に記載される数値をそれぞれ最小値及び最大値として含む範囲を示す。
また、本発明において、組成物中の各成分の量について言及する場合、組成物中に各成分に該当する物質が複数存在する場合には、特に断らない限り、組成物中に存在する当該複数の物質の合計量を意味する。
以下、本発明について説明する。
本発明のアミロイドタンパク質結合アプタマーは、Gカルテット構造を有するポリヌクレオチドであって、以下のポリヌクレオチドからなる群より選択された少なくとも1つであり、アミロイドタンパク質オリゴマーに対する結合性を有するアプタマーである。
(1)配列番号1~配列番号18のいずれかで示される塩基配列からなるポリヌクレオチド、及び、
(2)少なくとも2連続のグアノシンヌクレオチドを、少なくとも4つ含み、配列番号1~18のいずれかで示される塩基配列において1又は数個の塩基が欠失、置換、又は付加された塩基配列からなるポリヌクレオチド、
(3) 少なくとも2量体以上の多量体の上記(1)又は(2)のポリヌクレオチドを有するポリヌクレオチド。
このような配列であれば、Gカルテット構造を有し、アミロイドタンパク質オリゴマーを特異的に認識することができる。

Gカルテット構造を有するアプタマーであることは、公知の測定手段によって確認することができる。例えば、CDスペクトルを用いて、所定の波形を確認することでGカルテット構造を有するアプタマーであると確認できる。例えば、アプタマーをTBS緩衝液(10m MTris-HCl、150mM NaCl、5mM KCl、pH7.4)に溶解して試料液を調製し、温度20℃、波長200nm~320nm、走査速度100nm/分、積算回数10回の条件下でスペクトル測定したときの条件によるCDスペクトルにおいて、240nm近傍の極小値と260nm近傍での極大値の出現により確認できる。
このようなアミロイドタンパク質結合アプタマーであれば、オリゴマー型アミロイドタンパク質を特異的に認識可能である。
ここでストリンジェントな条件とは、特異的なハイブリッドが形成され、非特異的なハイブリッドが形成されない条件をいう。典型的なストリンジェントな条件とは、例えば、カリウム濃度は約25mM~約50mM、及びマグネシウム濃度は約1.0mM~約5.0mM中において、ハイブリダイゼーションを行う条件が挙げられる。
また、得られたアプタマーに対して、オリゴマー型アミロイドタンパク質に対する結合能を指標に、アプタマーの配列を更に最適化して、他のアプタマーを得ることができる。最適化する方法としては、いわゆる、In silico maturation法を挙げることができる。in silico maturation法では、in vitroでのアプタマーの評価および順位づけの操作と、配列の組換えやシャッフリング及び種々の変異導入などのin silico(コンピュータ上)での進化模倣アルゴリズムに基づく新規配列の作製の操作から成る。この二つの操作を合わせて一世代と呼び、何世代か操作を繰り返すことで目的の機能をもつアプタマー配列を獲得する(例えば、Nucleic Acids Res., (2005) vol.33(12), pp.e108; Biochem Biophys Res Commun., (2006) vol. 347(1), pp.226-31; Biosens Bioelectron., (2010) vol.15;26(4), pp.1386-91参照)。
また、配列が既知のものについては、常法により化学合成してもよい。
例えば、ブロッティングアッセイの場合には、以下のように行う。
アミロイドタンパク質オリゴマーを0.5μg~1μg、ニトロセルロース膜に固定化して、ブロッティング用の膜を用意する。対象とするアプタマーに蛍光物質(例えば、FITC)を連結して、試料アプタマーを調製する。
上記ブロッティング用膜に対して、得られた試料アプタマーを50nM~5μMの濃度で含むTBS-T溶液(25mMトリス-HClpH7.4、0.15M NaCl、0.005M KCl、0.05%Tween20)を添加して、室温で1時間インキュベートする。その後、十分量のTBS-Tを用いて膜を洗浄する(2min×2回、10min×1回)。
洗浄後のニトロセルロース膜を、TBS-T で1000倍希釈した検出用酵素(例えば、HRP)で修飾した前記蛍光物質に対する抗体(例えば、抗FITC抗体)と共に、室温で1時間インキュベートする。その後、十分量のTBS-Tを用いて膜を洗浄する(2min×2回、10min×1回、5min×2回)。
次いで、前記検出用酵素に対する基質を添加して、化学発光を検出することにより、試料アプタマーとアミロイドタンパク質オリゴマーとの結合を評価する。
前記多量体アプタマーとしては、前記構成単位をタンデムに連結したものであってもよく、デンドリマー型であってもよい。タンデムに連結した場合には、連結の形態としては、構成単位が同方向に連結したもの(すなわち、(頭-尾)(頭-尾)型)及び逆向きに連結したもの(すなわち、(頭-尾)(尾-頭)型)を挙げることができる。デンドリマー型の場合には、コア部分としてエチレンジアミン、ヘキサメチレンジアミン等を挙げることができる。
リンカー配列としては、グアノシンヌクレオチド以外のヌクレオチド、チミジンヌクレオチド又はシトシンヌクレオチドが2つ以上連続することにより形成された配列など等を挙げることができる。リンカー配列の長さとしては、2~15とすることができ、3~10であることが結合能の向上の点で好ましい。
前記アミロイドタンパク質結合アプタマーは、既知の方法を用いて配列に従って化学合成すればよく、当業者であればアミロイドタンパク質結合アプタマーを得るための合成方法を適宜選択することができる。
本発明のアミロイドタンパク質オリゴマーの検出方法は、前記アミロイドタンパク質オリゴマー結合アプタマーと体液試料とを接触させること(接触工程)、及び、体液中のアミロイドタンパク質オリゴマー及び前記アミロイドタンパク質オリゴマー結合アプタマーの複合体を検出すること(検出工程)、を含むものである。
上記アミロイドタンパク質オリゴマー検出方法では、上述した特定配列を有し、アミロイドタンパク質を特異的に結合し得るアミロイドタンパク質オリゴマー結合アプタマーと、標的分子であるアミロイドタンパク質オリゴマーとを接触させて、これらから構成される複合体を検出するので、試料中のアミロイドタンパク質オリゴマーを特異的に検出することができる。
アミロイドタンパク質オリゴマー結合アプタマーと被検試料との接触前に、被検試料の調製工程を設けてもよい。このような調製工程としては、被検体からの採取、適切な希釈液を用いて検出に適した濃度に採取試料の希釈等を挙げることができる。
本発明のアミロイドタンパク質オリゴマー検出キットは、前記アミロイドタンパク質オリゴマー結合アプタマーを含むものである。
本キットに含まれるアミロイドタンパク質オリゴマー結合アプタマーは、上述したように、アミロイドタンパク質オリゴマーを特異的に検出することができるので、本キットを用いることにより、簡便にアミロイドタンパク質オリゴマーを検出することができる。
キットに含むことが可能な他の試薬としては、前記複合体を検出するために使用可能な試薬、陽性又は陰性対照サンプルなどを挙げることができる。
上記のように本発明の一態様は、(1)配列番号1~配列番号18のいずれかで示される塩基配列からなるポリヌクレオチド、(2)少なくとも2連続のグアノシンヌクレオチドを、少なくとも4つ含み、配列番号1~18のいずれかで示される塩基配列において1又は数個の塩基が欠失、置換、又は付加された塩基配列からなるポリヌクレオチド、及び(3)配列番号1~配列番号18のいずれかで示される塩基配列からなるポリヌクレオチドを構成単位として含む多量体ポリヌクレオチドを提供するものである。
これらのポリヌクレオチド又は多量体ポリヌクレオチドは、上述した用途の他にも、種々の用途に利用可能である。
特に、前記ポリヌクレオチド又は多量体ポリヌクレオチドは、アミロイドタンパク質オリゴマー結合アプタマーとして、アミロイドタンパク質オリゴマーに対する特異的な結合性を有するので、アミロイドタンパク質オリゴマーを標的とする種々の態様に使用可能である。
これらの治療又は予防の対象となる疾患としては、アミロイドタンパク質オリゴマーに関連する疾患を挙げることができる。このような疾患としては、例えば、パーキンソン病、プリオン病(狂牛病)、ポリグルタミン病、ハンチントン病、アルツハイマー型認知症、Lewy小体型認知症、パーキンソン病、多系統萎縮症、クロイツフェルト・ヤコブ病、ウシ海綿状脳症やスクレイピー等のプリオン病、ピック病や進行性核上性麻痺等のタウオパチー、ハンチントン病等のポリグルタミン病、筋萎縮性側索硬化症、II型糖尿病、全身性アミロイドーシスや透析アミロイドーシス等のアミロイドーシスなどを挙げることができる。
また、投与対象としては、アミロイドタンパク質オリゴマーの蓄積を生じうる生物であればよく、ヒト、サル等の霊長類の他、牛、馬、羊等の家畜動物を挙げることができる。
ここで「高周波数誘導加熱」とは、高周波の周波数、高周波電圧などの加熱条件で所定の温度にして、加熱することをいう。
前記アプタマー-導電性粒子結合体は、前記アミロイドタンパク質オリゴマー結合アプタマー部分を含むので、アプタマー部分を介してアミロイドタンパク質オリゴマーに特異的に結合可能であり、また、前記導電性粒子を含むので、高周波誘導加熱によって導電性粒子が発熱して熱を発生させることができる。このアプタマー-導電性粒子結合体を用いることにより、高周波交流磁場が照射された領域のみを局所加熱することができ、発生させる熱を調整することにより、アプタマーに結合した標的物質を破壊することができる。これにより、標的となるアミロイドタンパク質オリゴマーを破壊して、アミロイドタンパク質オリゴマーの蓄積を予防又は解消することができ、アミロイドタンパク質オリゴマーの蓄積による各種症状の軽減又は発症予防を行うことができる。
また、導電性粒子のかわりに、磁性粒子を使用してもよい。ここで、磁性粒子は、例えばFe2O3やFe3O4などの材料を含むことができる。
高周波誘導加熱は、例えば、所定の電気制御回路に信号発生器からの出力を増幅し、電磁場照射コイルに供給するためのインピーダンス整合回路を付加した装置により実現できる。
金ナノ粒子(直径1.4nm)30nmolに100μlのイソプロパノールを添加して混合物を得、該混合物をミリQで1/10に希釈して、全量1000μlとする(金ナノ粒子溶液)。次に、前記アミロイドタンパク質オリゴマー結合アプタマー3nmolを金ナノ粒子溶液に添加し、4℃で24時間インキュベートする。次いで、得られた混合溶液をについて、5mMのNaH2PO4、150mMのNaCl、pH6.5で平衡化したゲルろ過カラム(Suplex 7516/60)を用いてゲルろ過クロマトグラフィーを行う。次に、ゲルろ過クロマトグラフィーで得られた画分を脱塩し凍結乾燥を行うことにより、アプタマー・金粒子複合体を調製する。
<α-シヌクレインオリゴマー特異的アプタマーの作製>
(1) α-シヌクレイン試料の調製
(1-1)モノマーの調製
α-シヌクレイン遺伝子の入ったプラスミドを持つ大腸菌BL21(DE3)/pET-28a(+)α-シヌクレインを 終濃度30μg/mlのカナマイシンを含むLB培地で培養し、IPTGを添加することでα-シヌクレインの発現を誘導した。得られた湿菌体を懸濁後、破砕し、遠心分離(15,000g、4℃、20min)を行い、上清を回収した。得られた上清を沸騰した湯の中で20分間加熱した。加熱後、遠心分離 (15,000g, 4℃, 15min) を行い、熱によって変性・凝集したタンパク質を除去した。得られた上清を20mM Tris-HCl(pH8.0)で一晩透析した。透析後、微量超遠心(150,000g、4℃、20min)を行い、得られた上清を陰イオン交換カラム RESOURSE Q(GEヘルスケア社)6mlを用いてFPLCによる精製を行った。緩衝液には2種類のbufferA(20mM Tris-HCl、pH8.0)とbufferB(20mM Tris-HCl、pH8.0、1M NaCl)を用いた。段階的なグラジェントを設定し、流速、フラクション容量などはマニュアル、カラム容量にあわせて設定し精製を行った。
組み換え発現及び精製したα-シヌクレインを、MiliQで透析し、透析後のα-シヌクレインを4~6mgずつ分注し、凍結乾燥を行った。その後、MiliQを用いて30mg/mlの濃度になるように凍結乾燥後試料を復水し、30~60分間程度撹拌することで十分溶解させた後、再び凍結乾燥を行った。この試料を、PBS緩衝液を用いて30mg/mlの濃度になるように復水し、30~60分間程度撹拌することで十分溶解させた。得られた試料をSuperdex200 10/300 colunm(GEヘルスケア社)を用いてゲルろ過クロマトグラフィーを行うことで精製した。バッファーはPBS緩衝液を用い、流速0.5ml/minで分離を行った。分離後、オリゴマーが含まれていると考えられるフラクションを回収し、限外ろ過フィルター(MWCO100kDa)を用いて濃縮した。濃縮後、終濃度が0.02%になるようにアジ化ナトリウムを加え、4℃で保存した。得られた試料をα-シヌクレインオリゴマー試料とした。
PBS緩衝液中に1~2mg/mlになるように溶解させたα-シヌクレインモノマーを、37℃で振とうさせながらインキュベートし、線維形成を促進させた。α-シヌクレインの線維試料は、アミロイド線維に結合する蛍光色素 Thioflavin T(TfT)(シグマ社)の蛍光を指標に調製した。TfTの蛍光強度を測定しながらインキュベートし、TfTの蛍光が飽和したところでインキュベートを止めた。インキュベート試料を遠心(10,000g、20℃、10min)し、上清を回収した。
ペレットに400μlのPBS緩衝液を加え、ボルテックスしてよく撹拌した。その後、再度同じ条件で遠心を行い、同様にペレットを洗浄した。これを3回行い、最終的に得られたペレットを50μlのPBS緩衝液に懸濁した。これをα-シヌクレイン線維試料とした。回収した上清のタンパク質濃度を測ることで、線維試料のタンパク質濃度を算出し、α-シヌクレイン線維試料の濃度を算出した。
なお、線維試料は未精製のため、α-シヌクレインオリゴマーがわずかに混入していることが予想される。
(2-1)オリゴヌクレオチドの合成
下記(a)~(e)に示すランダムライブラリおよびプライマー(配列番号19~23)をそれぞれ設計し、各種オリゴヌクレオチドを合成した。ランダムライブラリおよびフォワードプライマーには5’末端にFITC配列(下記塩基配列中、「FITC」と表す。)を、リバースプライマーには3’末端にビオチン配列(下記塩基配列中、「Biotin」と表す。)を付加し、FITCまたはビオチンで修飾されたものをそれぞれ構築した。また、3’末端側プライマー領域の相補鎖については、5’末端にFITC修飾配列を有するものと未修飾のものをそれぞれ作製した。

得られたオリゴヌクレオチドを、ゲルシフトアッセイを用いてスクリーニングした。スクリーニングは、以下の(i)~(iv)の操作を繰り返し、全部で2回行った。
(i)α-シヌクレインオリゴマーに結合するssDNAの選択とssDNAライブラリの結合確認
5’末端にFITCが修飾されたDNAライブラリとプライマー領域の相補鎖DNAを等モル量ずつ混合し、TBS緩衝液 (10mM Tris-HCl, 150mM NaCl, 5mM KCl、 pH7.4)中でフォールディングさせた後、α-シヌクレインオリゴマーとDNAを混合し、高速振とう機で撹拌しながら室温で1時間インキュベートした。フォールディングは95℃で3分間加熱した後、30分間かけて25℃まで温度を下げることで行った。
その後、この混合溶液を3% アガロースゲルを用いて、TAE緩衝液中で電気泳動を行った。バリアブルイメージアナライザ Tyhoon8600(GEヘルスケア社、以下同じ)でFITCの蛍光を検出することにより、オリゴマーとDNAの結合を確認した。その後、ゲルをCBB染色し、タンパク質の位置を確認した。
上記(i)で用いたα-シヌクレインオリゴマーとDNAの混合溶液を泳動したゲルに対して、アルコールで洗浄し炙ったカッターとピンセットを用いて、タンパク質部分のゲルを切り取り、回収した。
このゲルからMERmaid kit(Q-biogene社)を用いて、α-シヌクレインオリゴマーに結合したDNAの抽出を行った。実験操作は通常のプロトコールに従って行った。最終的にDNAを1×TEバッファー(10mM Tris-HCl、1mM EDTA)に溶出させ、次のライブラリのテンプレートとした。
抽出されたssDNAをテンプレートとしてPCRを行った。FITC修飾5’プライマー(終濃度0.4μM)、ビオチン修飾3’プライマー(終濃度0.4μM)、dNTP(終濃度0.2mM)、5U/μlのTaq DNA polymerase、5μlのテンプレートDNAを含んだPCR反応液100μlを30本調製した。
サーマルサイクラーで、95℃で3分加熱した後、95℃で1分、52℃で1分、72℃で1分を1サイクルとし、30サイクル繰り返した。各PCR産物は、TAEバッファー中で3%アガロース21ゲルを用いて電気泳動を行い、テンプレートとなるDNAが増幅されていることを確認した。
アビジン固定化アガロース120μlを5倍量のColumnバッファー(30mM HEPES、500mM NaCl、5mM EDTA、pH7.0)で2回洗浄した。これに、PCR産物に、その1/10倍量の×50 TEバッファーおよび1/5倍量の5M NaClを添加し、この溶液を洗浄したアビジン固定化アガロースに加え、30分間インキュベートした。
インキュベート後、上清を取り除き、アガロースを5倍量のColumnバッファーで2回洗浄した後、アガロースの1.5倍量の0.15M NaOHを加えて10分間攪拌し、上清を回収した後、再びアガロースに同量の0.15M NaOHを加えて10分間攪拌し、ssDNAを溶出させ上清を回収した。ssDNAを含む上清を2M HClで中和し、エタノール沈殿によりssDNAを回収した。
得られたペレットを60μlのTEバッファーで溶解し、分光光度計を用いて260 nmの吸収を測定することで、DNA濃度を算出した。尚、操作はすべて室温で行った。
以下の(v)~(viii)の操作を繰り返し、アプタマーブロッティングを用いた競合スクリーニングを全部で3ラウンド行った。
(v)α-シヌクレインオリゴマーに結合するssDNAの選択
ニトロセルロース膜に調製した種々の量のα-シヌクレインモノマー、オリゴマー及び線維をそれぞれ固定化し、乾燥後、膜をTBS-T(25mMトリス-HClpH7.4、0.15M NaCl、0.005M KCl、0.05%Tween20)で調製した4% スキムミルクと室温で1時間インキュベートすることでブロッキングを行った。
その後、十分量のTBS-Tを用いて膜を洗浄した(2min×2回、10min×1回)。上記(i)と同様に5’末端にFITCが修飾されたDNAライブラリをフォールディングし、DNAと膜と室温で1時間インキュベートした後に、十分量のTBS-Tを用いて膜を洗浄した(2min×2回、10min×1回、5min×2回)。
オリゴマーを固定化した部分のニトロセルロース膜を切り抜き、200μlの8M 尿素および600μlのフェノールクロロホルムを添加し、3分間攪拌した後に、30分間室温で静置した。これにMilliQ100μlを加え、3分間攪拌し、遠心分離(12000rpm, 10~20℃)して得られた上清を新しいエッペンに移した。更に、上清と同量のクロロホルムを加え、3分間攪拌した後に、遠心分離(12000rpm, 10~20℃)し、上清を新しいエッペンに移し、この中に含まれるssDNA分子をエタノール沈殿により回収した後に得られたペレットを30μlのTEバッファー(10mM Tris、1mM EDTA)で溶解した。
上記(iii)と同様に操作した。
(viii)増幅したdsDNAの一本鎖化
上記(iv)と同様に操作した。
上述の4ラウンド目または5ラウンド目のスクリーニングにおいて抽出したDNAをテンプレートとし、未修飾のプライマーを用いてPCRを行った。得られたPCR産物を、3%アガロースゲルを用いてTAEバッファー中で電気泳動した後、目的の大きさのバンドを切り出し、MERmaid kitを用いてゲル精製を行った。
精製後、試料をTAクローニングに用い、pGEM-Tとライゲーションさせた。TAクローニングはpGEM-T Vector Systemを用いて行った。精製したssDNA溶液を3μl、5μlRapid Ligation Buffer、1μlのpGEM-T Vector(50ng)、1μlのT4 DNA Ligaseを加え、全量10μlとなるように調製した後、室温で1時間ライゲーションを行った。
目的のプラスミドを含む大腸菌DH5αを1mlのLB液体培地(Amp 100μg/ml)で培養した後、集菌し、アルカリ-SDS法によりプラスミド抽出を行った。抽出したプラスミドを用いてシーケンス解析を行った。
α-シヌクレインモノマー、オリゴマー、線維1μgをニトロセルロース膜に固定し、乾燥後、膜をTBS-T(25mMトリス-HClpH7.4、0.15M NaCl、0.005M KCl、0.05% Tween-20)で調製した4%スキムミルクと室温で1時間インキュベートすることでブロッキングを行った。その後、十分量のTBS-Tを用いて膜を洗浄した(2min×2回、10min×1回)。5’末端にFITCが修飾された得られた複数のオリゴヌクレオチドを(f.c. 100 nM)をフォールディングし、DNAと膜とを室温で1時間インキュベートした後に、十分量のTBS-T を用いて膜を洗浄した(2min×2回、10min×1回、5min×2回)。その後、TBS-T で1000倍希釈したHRP修飾抗FITC抗体と膜を室温で1時間インキュベートした後に、十分量のTBS-Tを用いて膜を洗浄した(2min×2回、10min×1回、5min×2回)。ECL Plus Western blotting detection reagents(GEへスルケア社)を膜に添加し、HRPの化学発光をTyphoon8600で検出することにより、DNAとタンパク質の結合を観察した。
この結果を図1に示す。

即ち、アプタマーをTBSバッファー(10mM Tris-HCl、150mM NaCl、5 mM KCl、pH 7.4)で20μMに調製し、フォールディングした。その後、円二色性分散計J-720を用いてCDスペクトルを測定した。測定は温度20℃、波長200~320nm、走査速度100nm/min、積算回数10回の条件下で行なった。
この結果、240nm近傍の極小値と260nm近傍の極大値の存在により、上述のα-シヌクレインオリゴマー結合アプタマーがGカルテット構造を有することがわかった。
上記で得られた8種類のアプタマーのうち、T-SO517及びT-SO606と、抗α-シヌクレインオリゴマー抗体A11(Life technologies社)との結合性の比較を行った。
同じニトロセルロース膜上にPQQを補酵素とするグルコース脱水素酵素(PQQGDH)(国際試薬社)、C反応性タンパク質(CRP)(Polyscience社)、ルシフェラーゼ、IgG抗体(シグマ社)を5pmolずつ、およびα-シヌクレインオリゴマーを0.5μg固定した膜を用いて、アプタマーブロッティング法により特異性の評価を行った。
この結果を図2に示す。なお、PQQGDH、CRP、ルシフェラーゼ、IgG抗体及びα-シヌクレインオリゴマーは、いずれもβ構造に富むタンパク質である。
上記で得られた8種のアプタマーのβアミロイドペプチドに対する結合性を、以下のようにして確認した。
(4-1)βアミロイドペプチドAβ1-40の調製
βアミロイドペプチドとして、Aβ1-40(Peptide Institute,Inc社)を使用した。
ヘキサフロロイソプロパノール(HFIP)で終濃度2.5mg/mlに溶解した100μLのAβ1-40溶液を、900μLのMilliQと混合し、室温で撹拌した後、遠心(14,000g,室温,15min)して上清を回収した。上清にArガスをバブリングした後、高速振とう機を用いて24時間撹拌した(800rpm,室温)。その後試料を遠心(14,000g,4℃,20min)し、回収した上清を限外ろ過フィルター(MWCO:10kDa)で濃縮した。これをAβ1-40オリゴマー試料とした。
なお、Aβ1-40モノマー試料は、上記にて調製したAβ1-40溶液を直ちに使用したものとした。
Aβ1-40線維試料は、以下のようにして調製した。まず、Aβ1-40を、DMSOに終濃度4.33mg/mlとなるように溶解して、ストック溶液を作製した。これをPBSで50μMに希釈し、9日間程度37℃で撹拌した。その後、14,000G、4℃、20分間遠心し、ペレットを回収した。ペレットに500μLのPBSを加え撹拌・遠心する操作を2回繰り返し、ペレットを洗浄した。各操作で得られた上清の280nmにおける吸光度を上記と同様にして測定することで、ペレットに残ったタンパク質量を算出した。その後、ペレットを、所望の濃度にPBSで復水し、線維試料として実験に用いた。
12.5pmolのAβ1-40モノマー・オリゴマー・線維を同一のニトロセルロース膜に固定化し、500nMの各アプタマーを用いてアプタマーブロッティングにより結合特異性を評価した。コントロールとしてA11を用い、同様の実験を行なった。
Aβ1-40オリゴマー試料を3μL、2μL、1μL、0.5μLずつ、また、α-Synオリゴマーを300ng、200ng、100ng、50ngずつ、それぞれニトロセルロース膜に固定化し、A11を用いてドットブロッティングを行なった。
α-Synオリゴマーを固定化した箇所のスポット強度を解析し、検量線を作製することでAβ1-40オリゴマー試料に含まれるオリゴマー濃度を概算した。その後、A11のスポット強度が同程度になるようにAβ1-40オリゴマーをニトロセルロース膜に固定化し、500nMの各アプタマーを用いてアプタマーブロッティングを行なった。コントロールとしてA11を用い、同様の実験を行った。
結果を図3に示す。
<ダイマー型アプタマーの結合性1>
実施例1で得られたアプタマー、T-SO504の配列を、5merのチミジン(t)ヌクレオチドで連結して、アプタマーダイマー(504W)を合成した。
また、比較対照として、T-SO504の塩基配列から5’末端側の3塩基及び3’ 末端側の8塩基を削除したポリヌクレオチド504M1(GGGTGGGCAAAGGG:配列番号24)、T-SO504の塩基配列5’末端側の7塩基及び3’末端側の8塩基を削除したポリヌクレオチド504M2(GGGCAAAGGG:配列番号25)も同様に合成した。
得られた各アプタマーの配列を表1に示す。表1中、小文字アルファベット(t又はa)は上記の付加的な配列を意味する。
この結果を図4に示す。
これに対して、T-SO504の塩基配列の一部を削除して二連続のグアノシンヌクレオチドが3つのみとなった504M1と、2つのみとなった504M2では、T-SO504の結合性を保持できなかった。
<ダイマー型アプタマーの結合性2>
(1)ダイマー型アプタマーの作製
実施例1で得られたアプタマーT-SO530の配列に対して、5mer又は10merのチミンリンカーを介して直列に並べたダイマー(以下5T dimer、10T dimerと表記)を設計し、合成した。各アプタマーに対して、FITCを5’末端に抗原として連結して修飾し、FITCとアプタマーの間には5merのチミンをリンカーとして挿入した。各アプタマー配列を表2に示す。
作製した5T dimer、10T dimer、及び元のアプタマーであるT-SO530のAβオリゴマーに対する結合能を評価した。
また、10T dimerと元のアプタマーであるT-SO530の濃度を、1nM、2.5nM、10nM、20nM、又は60nMとして、上記と同様にAβオリゴマーへの結合実験を行った。得られた結合シグナルに基づき、グラフパッドプリズム(GraphPad Prism;エムデーエフ社)を用いてカーブフィッティングを行い、結合解離定数(Kd)を算出した。
また、結合解離定数(Kd)については、T-SO530のKdは9.03±3.06nMであり、10T dimerのKdは1.44±0.68nMであった。ダイマー型のKd値は元のアプタマーに比べて6分の1程度に低下していたことから、ダイマー型のアプタマーでは、向上した結合親和性を有することがわかった。
上記で得られた5T dimerと10T dimerのβアミロイドペプチドに対する結合性を、以下のようにして確認した。
(3-1)βアミロイドモノマーの調製
βアミロイドペプチドとして、Aβ1-40(Peptide Institute,Inc社)を使用した。これをヘキサフロロイソプロパノール(HFIP)溶液に終濃度2.5mg/mlに溶解してストック溶液を調製した。このストック溶液を、PBSで所望の濃度に希釈して、直ちに用いたものをAβ1-40モノマーとした。280nmにおける吸光度を紫外可視分光解析システムDU 800(ベックマンコールター社)、又はNanoDrop 2000(Thermo Fisher Scientific社)により測定することにより、タンパク質濃度を確認し、βアミロイドモノマーとして実験に用いた。
Aβ1-40オリゴマーは、実施例1と同様にして調製した。280nmにおける吸光度を、モノマー試料と同様にして測定し、タンパク質濃度を確認して、βアミロイドオリゴマー試料として実験に用いた。
Aβ1-40を、DMSOに終濃度4.33mg/mlとなるように溶解して、ストック溶液を作製した。これをPBSで50μMに希釈し、9日間程度37℃で撹拌した。その後、14,000G、4℃、20分間遠心し、ペレットを回収した。ペレットに500μLのPBSを加え撹拌・遠心する操作を2回繰り返し、ペレットを洗浄した。各操作で得られた上清の280nmにおける吸光度を上記と同様にして測定することで、ペレットに残ったタンパク質量を算出した。その後、ペレットを、所望の濃度にPBSで復水し、線維試料として実験に用いた。
得られたAβ1-40のモノマー、オリゴマー、線維試料を0.5μgずつニトロセルロース膜に固定し、乾燥後、膜をTBS-T(0.05%Tween 20)で調製した4%スキムミルクと室温で1時間インキュベートすることでブロッキングを行って、Aβ1-40の各試料が固定化されたブロットアッセイ用のニトロセルロース膜を得た。このニトロセルロース膜に対して、各アプタマー200nMを用いた以外は、実施例1の(2-5)と同様にして、ブロットアッセイを行った。
結果を図6に示す。
図6に示されるように、5T dimerと10T dimerはいずれも、オリゴマーを固定した位置に強いスポットが確認できた。このことから、5T dimerと10T dimerはいずれも、Aβ1-40のオリゴマーに対して高い選択性を示すことがわかった。
(1) 改良型配列の作製
上記実施例1で得られた8種類のアプタマーの配列を親配列として、一点交叉及び2塩基の変異導入を行なうことで第一世代のアプタマー配列を24本作製した。
得られた第一世代の各アプタマー配列の3’側に、18merの相補鎖形成領域を付加したDNAを合成した。この相補鎖形成領域に相補的となる18merのDNAを合成し、その5’末端側にFITCを常法により結合した標識化配列を作製し、相補鎖形成領域を介して、第一世代のアプタマー配列にFITCを修飾した。得られた各FITC標識アプタマーをそれぞれTBSバッファーに添加して、FITC標識アプタマー溶液を調製した。
ウェルを洗浄後、HRP修飾抗FITC抗体(コスモバイオ社)をウェルに添加し、室温でインキュベートした。ウェルを洗浄後、HRPの発光基質を加え、化学発光により各アプタマーのオリゴマーへの結合を検出し、比較した。各プレート間のシグナル強度の標準化には、親配列であるT-SO508アプタマー配列の3’側に相補鎖形成を介してFITCを修飾したアプタマー(3’FC508)を用いた。
Aβオリゴマーに対する結合能に基づいて、第一世代のアプタマーのうち、最も結合能が高い6本の配列を選択した。
第二世代のアプタマー配列に対して、上記と同様にして、Aβオリゴマーに対する結合能を検出し、比較した。第二世代のアプタマー配列の中で結合能の高かった4本の配列を選択した。最も高い結合能を示した第二世代のアプタマー配列は、親配列の結合能と比較して、8.69倍の化学発光を示した。
第三世代のアプタマー配列に対して、上記と同様にして、Aβオリゴマーに対する結合能を検出し、比較した。最も高い結合能を示した第三世代のアプタマー配列は、親配列の結合能と比較して、22.55倍の化学発光を示した。

上記(1)で得られたアプタマーのうち、2G16及び3G4と、親配列から得たアプタマー3’FC508について、結合能を評価した。
具体的には、各アプタマーの濃度を、1nM、5nM、1nM、25nM、50nM、100nM、又は200nMとして、上記(1)と同様にAβオリゴマーに対する結合能を確認した。得られた結合シグナルに基づき、グラフパッドプリズム(GraphPad Prism;エムデーエフ社)を用いてカーブフィッティングを行い、結合解離定数(Kd)を算出した。結果を図7に示す。図7中の各シンボルにおいて、菱形は3’FC508を示し、四角は2G16を示し、三角は3G4を示す。
その結果、3’FC508のKdは67.74nM、2G16のKdは67.10nM、及び3G4のKdは70.75nMであり、いずれのKdも70nM程度と、結合解離定数に相違は見られなかった。
上記(1)で得られたアプタマー、アプタマー3’FC508、2G16、3G4の各構造を、15質量%ポリアクリルアミドゲルを用いた電気泳動により解析した。15質量%Tris-グリシンゲル(WAKO)に、上記(1)で得られたFITC修飾された各アプタマー3’FC508、2G16、3G4をアプライし、TBEバッファー中で電気泳動を行なった。その後、FITCの蛍光を観察することで各アプタマーの泳動位置を確認した。
PAGE解析の結果を図8に示す。図8において各レーンは、左からラダー、アプタマー3’FC508、2G16、3G4を示す。
図8に示されるように、アプタマー2G16及び3G4はいずれも、50bp付近のバンドに加えて、1000bpと推測される泳動位置に複数のバンドが検出された。このことから、2G16及び3G4は、単量体の形態を取るほか、それぞれのアプタマー配列が十数個以上集まって形成された多量体の形態も取ることがわかった。
また、上述した条件下でのin silico maturation法により、多量体化し、且つ、結合するアプタマーが得られることがわかった。
上記で得られた2G16と3G4のβアミロイドペプチドに対する結合性を確認した。
βアミロイドモノマー、オリゴマー、線維の各試料は、実施例3(3)で得られたものを使用した。
2G16と3G4の各アプタマーを200nMの濃度で使用した以外は、実施例3(3)と同様にブロットアッセイを行った。結果を図9に示す。
図9に示されるように、2G16と3G4は、いずれもオリゴマー及び線維を固定した位置にスポットが確認でき、特に、オリゴマーを固定した位置に強いスポットが確認できた。これに対して、いずれのアプタマーについても、モノマーを固定した位置にはスポットが確認できなかった。このことは、2G16と3G4はいずれも、Aβ1-40のモノマーに対してオリゴマーと特異的に認識することがわかった。

Claims (7)
- Gカルテット構造を有し、且つ、以下のポリヌクレオチドからなる群より選択された少なくとも1つであり、アミロイドタンパク質オリゴマーに対する結合性を有するアプタマー:
(1) 配列番号1~配列番号18のいずれかで示される塩基配列からなるポリヌクレオチド、
(2) 少なくとも2連続のグアノシンヌクレオチドを、少なくとも4つ含み、配列番号1~配列番号18のいずれかで示される塩基配列において1又は数個の塩基が欠失、置換、又は付加された塩基配列からなるポリヌクレオチド、並びに、
(3)前記(1)又は(2)のポリヌクレオチドを構成単位として含む多量体であるポリヌクレオチド。 - 前記(3)のポリヌクレオチドが、上記(1)又は(2)のポリヌクレオチドと、各(1)又は(2)のポリヌクレオチド間を連結するリンカー配列とを有する請求項1記載のアプタマー。
- 請求項1又は請求項2記載のアプタマーを含むアミロイドタンパク質オリゴマー検出キット。
- 請求項1又は請求項2記載のアプタマーと被検試料とを接触させること、及び、
前記被検試料中のアミロイドタンパク質オリゴマー及び前記アプタマーの複合体を検出すること、
を含むアミロイドタンパク質オリゴマーの検出方法。 - 前記被検試料が、髄液、血清、血漿及びこれらの希釈物からなる群より選択された少なくとも1つである請求項4記載のアミロイドタンパク質オリゴマー検出方法。
- 配列番号1~配列番号18のいずれかで示される塩基配列からなるポリヌクレオチド。
- 配列番号1~配列番号18のいずれかで示される塩基配列からなるポリヌクレオチドを構成単位として含む多量体であるポリヌクレオチド。
