Abstract

본 발명은 새로운 혈청 내 다이옥신류의 생물학적 검출 방법, 및 이의 대사증후군 및 관련 증상에 대한 진단적 유용성에 관한 것으로서, 구체적으로 다이옥신 반응 요소(dioxin responsive element), 프로모터, 및 리포터 유전자가 작동가능하게 연결된 유전자 컨스트럭트를 포함하는 재조합 벡터로 형질전환된 세포와 혈청 전체를 사용함으로써, 루시퍼라제 활성을 측정하여 혈청 내 다이옥신 함량과 신체 변수들 간의 유의적인 상관성을 확인하였고, 혈청으로부터 다이옥신을 정제하기 위한 전처리 과정이 요구되는 기존의 방법으로부터 개선되어, 혈청 전체를 사용함에 따라 다수의 시료를 극소량으로도 쉽고 정확하게 분석할 수 있으므로, 다이옥신류 화합물에 의하여 발현이 조절되는 재조합 리포터 유전자를 포함하는 형질전환 세포주에 전체 혈청(total serum)을 처리하는 본 발명의 검출 시스템은 혈청 내 다이옥신의 생물학적 검출에 유용하게 이용할 수 있고, 이러한 방법을 사용하여 혈청 내 존재하는 다이옥신과 같은 POPs의 여부를 정확히 검출함으로써, 특정 POPs와 환자의 질병인자 간의 상관성을 연구하고, 이를 질병의 발병 예측 및 치료 여부를 판정하는데 유용하게 사용할 수 있다.
