また、ハスの花部の採集時期としては、特に限定されるものではないが、開花時期である夏季、すなわち6~9月のいずれかに採集したものが用いられる。
上記の例示した低級アルコールの中でも、抽出効率および抽出後の溶媒留去の容易性の観点からメタノールまたはエタノールが好適に用いられる。
抽出材料を振盪下に浸漬する場合には、30分間~10時間程度行うのが適当であり、非振盪下に浸漬する場合には、1時間~20日間程度行うのが適当である。
なお、50℃より低い温度や室温で浸漬して抽出することも可能であるが、その場合には、上記の時間よりも長時間浸漬するのが好ましい。
抽出混合物から固形物を除去して得られる抽出液は、常法により減圧濃縮して抽出エキスおよび抽出物とし、減圧濃縮の際に回収される溶媒は、再度抽出に利用できる。
したがって、本願発明における抽出物とは、溶媒が完全に留去された抽出エキスおよび抽出エキスを減圧下に乾固するまで溶媒を留去した抽出物のいずれをも意味し、該抽出物と抽出エキスは同義である。
精製処理は、吸着クロマトグラフ法、イオン交換クロマトグラフ法、溶媒による分配抽出などを単独または組み合わせて採用することができる。
この際の担体、溶出溶媒などの精製条件は、各種クロマトグラフィーに対応して適宜選択することができる。
すなわち、得られたメタノール抽出エキスまたは減圧濃縮して得られた抽出物を1%酢酸水溶液に溶解し、ジエチルエーテルを用いて分配し、分離した水層にアンモニア水を添加してアルカリ性とした後、再度ジエチルエーテルで分配してジエチルエーテル可溶画分を得、該画分に希水酸化ナトリウム水溶液を加えて分配し、得られたジエチルエーテル可溶画分がアルカロイド含有画分として得られる。
各精製段階および各画分は、ドラーゲンドルフ試薬によるアルカロイドの検出方法により、アルカロイドの存在の有無をチェックすることが好ましい。
得られたMeOH抽出エキスを、AcOEtとH2Oで分配抽出後、H2O移行部をn-BuOHで分配抽出し、AcOEt移行部、n-BuOH移行部およびH2O移行部を得た。
次に、Et2O移行部をさらに10%NaOH水溶液により洗浄し、Et2O溶出部を高アルカロイド画分として得た。
また、ステロールの1種であるβ-シトステロールを得た。
その場合の使用量としては、例えば化粧品に用いられる公知の他の添加剤または賦形剤を含む製品中に、例えば0.001~5重量%の割合で用いることができる。

これらは、添加しようとする製品種別、形態に応じて常法的に行われる加工(例えば、粉砕、製粉、洗浄、加水分解、醗酵、精製、圧搾、抽出、分画、ろ過、乾燥、粉末化、造粒、溶解、滅菌、pH調整、脱臭、脱色などを任意に選択、組合わせた処理)を行い、各種の素材から任意に選択して供すれば良い。
原料とする植物(生薬)としては、当業者に公知生薬または動物由来の材料が用いられる。
海藻類としては、緑藻類、褐藻類、紅藻類、藍藻類、車軸藻類、黄色藻類などが挙げられる。
また、動物系原料由来の素材としては、牛、豚、鶏または魚類のコラーゲン加水(酸、アルカリ、酵素など)分解物や水溶性コラーゲンまたはアシル化コラーゲンなどのコラーゲン誘導体、牛または豚のエラスチンまたはエラスチン加水分解物(酸、アルカリ、酵素など)または水溶性エラスチン誘導体、ケラチン及びその分解物またはそれらの誘導体、シルク蛋白及びその分解物またはそれらの誘導体、牛乳、カゼイン及びその分解物またはそれらの誘導体、脱脂粉乳及びその分解物またはそれらの誘導体、ラクトフェリンまたはその分解物、鶏卵成分、魚肉分解物、核酸関連物質(リボ核酸、デオキシリボ核酸)が挙げられる。

高分解能質量分析(High resolution FAB-MSおよびEI-MS)ならびに質量分析(FAB-MSおよびEI-MS)は、JEOL JMS-SX 102およびJMS-GCMATE型質量分析装置(日本電子株式会社製)を用いて測定した。
実験で得られた数値は、平均値±標準誤差で表記し、対照群との平均値の有意差の検定にはDunnettの方法を使用し、p値が0.05以下のものを有意とみなした。
理研細胞バンクより分譲されたマウスメラノーマB16-メラノーマ-4A5細胞(RCB0557)を10%ウシ胎児血清(FCS)、100 ユニット/mLペニシリンおよび100 μg/mLストレプトマイシン含有DMEM(4500 mg/L グルコース)培地(Sigma-Aldrich社)で培養(5%CO2、37℃)した。

B16-メラノーマ-4A5細胞を、10%ウシ胎児血清(FCS)、100ユニット/mLペニシリンおよび100 μg/mLストレプトマイシン含有DMEM(4500 mg/L glucose)培地(Sigma-Aldrich社)で培養(5%CO2、37℃)した。

96ウェル マイクロプレート(住友ベークライト社製)に2.5mM L-DOPAリン酸緩衝溶液70 μL/ウェルを分注し、調整した試料溶液(DMSO溶液)20 μLを添加した。次に、終濃度46ユニット/mLチロシナーゼリン酸緩衝溶液120 μLを添加し、酵素反応(室温、5分)を開始した。また、ブランクにはリン酸緩衝溶液を用いた。生成されたメラニンの吸光度をマイクロプレートリーダー(model 550、BIO-RAD社製)にて測定した(測定波長:450 nm、参照波長:655 nm)。

ハス各部位のメタノール抽出エキスのメラニン生成抑制作用
メラニンを生成するメラノサイトは表皮の基底層全体にわたって認められ、皮膚が日射を受けると、メラノサイトはより多くの色素を作り出し、皮膚は黄褐色または暗褐色になる。メラニンはメラノサイトやメラノーマなどといった特殊な細胞内で合成される色素性のポリマーであり、UVからの保護や色素、毒性のある化合物の吸収などに重要な役割を果たしている。
その結果を、以下の表2に示す。
ハス花部のメタノール抽出エキスおよび各画分のメラニン生成抑制作用
ハス花部の熱時(80 ℃)MeOH抽出エキスに有意なメラニン生成抑制作用が認められたことから、後記のスキーム1に示すようにハス花部のMeOH抽出エキスをAcOEt、n-BuOHおよびH2Oを用い分画した。得られた各画分についてB16-4A5細胞におけるメラニン生成抑制作用を検討した。
一方、ハス花部の室温下におけるMeOH抽出エキスおよびそのエキスを後記のスキーム2に示すように分画し得られたEt2O、H2O、高アルカロイド画分についても同様にメラニン生成抑制作用の検討を行った。
得られたMeOH抽出エキスのうち145.0 gを、AcOEtとH2Oで分配抽出後、H2O移行部をn-BuOHで分配抽出し、AcOEt移行部(31.2 g、3.1%)、n-BuOH移行部(41.0 g、4.1%)、H2O移行部(72.8 g、7.3%)を得た(スキーム1)。
但し、これら単離成分の収率はNelumbo nucifera花部からの単離収率である。
Fr. 5-6(200.1 mg)をHPLC [カラム:COSMOSIL 5C18-MS-II、溶離剤:MeOH:H2O =(85:15)]を用いて分離精製し、β-シトステロール(140.6 mg、0.0141%)を単離した。
Fr. 3-4(69.4 mg)をHPLC [カラム:COSMOSIL 5C18-MS-II、溶離剤:1% AcOH含有MeOH:H2O=(70:30)]を用いて分離精製し、N-メチルアシミロビン(6.6 mg、0.0007%)を単離した。
但し、これら単離成分の収率は、ハスの乾燥花部からの単離収率である。
より詳細には、タイ産ハス(Nelumbo nucifera)の乾燥花部(900.0 g)をMeOHで室温にて7時間抽出した。
ハス花部含有成分のメラニン生成抑制活性
活性の集約の認められたAcOEt,、n-BuOH移行部および高アルカロイド画分から単離した上記の6種のアルカロイドについて、B16-メラノーマ-4A5細胞におけるメラニン生成抑制作用を検討した結果を以下の表6および表7に示す。
さらに、これら6種のアルカロイド(1-6)について、マッシュルーム由来チロシナーゼ阻害活性についての検討を行った。
上記の実施例1においてアルカロイド含有画分0.01、0.1、1および5重量%をそれぞれ用いたこと以外は、実施例7と全く同様にして化粧水1kgをそれぞれ調製した。

乳液の調製
以下の処方:
上記の実施例12においてアルカロイド含有画分0.01、0.1、1および5重量%をそれぞれ用いたこと以外は、実施例12と全く同様にして乳液1kgをそれぞれ製造した。
上記の実施例17においてアルカロイド含有画分0.01、0.1、1および5重量%をそれぞれ用いたこと以外は、実施例17と全く同様にしてクリーム1kgをそれぞれ製造した。
上記の実施例22においてアルカロイド含有画分0.01、0.1、1および5重量%をそれぞれ用いたこと以外は、実施例22と全く同様にして日焼け止めクリーム1kgをそれぞれ製造した。
上記の実施例27においてアルカロイド含有画分0.01、0.1、1および5重量%をそれぞれ用いたこと以外は、実施例27と全く同様にしてファンデーション1kgをそれぞれ製造した。
上記の実施例32においてアルカロイド含有画分0.01、0.1、1および5重量%をそれぞれ用いたこと以外は、実施例32と全く同様にして全身用日焼け止めクリーム1kgをそれぞれ製造した。

- 有効成分が、以下の一般式(I):
R4、R5およびR6は水素原子またはメチル基であるか、あるいはR3およびR4ならびにR5およびR6は、互いに一緒になってそれぞれ結合手を表し、
2個のR7は水素原子を表わすか、または互いに一緒になってオキソを表し、
nは、0または1の整数であり、
nが0のとき、2個のR8は、それらが結合する炭素原子を含んで互いに一緒になって4-オキソ-シクロヘキサジエン環を形成し、

- 前記アルカロイドが、ハス科ハス属植物のハスの花部の低級アルコールで抽出して抽出液を得、この抽出液をさらに必要に応じて酢酸エチル/水およびn-ブタノール/水で分配処理して得られる有機溶媒可溶画分に含まれるか、あるいは
前記抽出液をジエチルエーテル/弱酸性水で分配処理して得られる水層を、アンモニア水で弱塩基性にしてジエチルエーテルで抽出し、該エーテル層を希NaOH水溶液で洗浄して得られるアルカロイド含有画分に含まれる請求項1~3のいずれか一つに記載の組成物。
