さらに、LXRはNF-κBの抑制を介して、NO合成酵素、シクロオキシゲナーゼ-2(COX-2)、インターロイキン-6(IL-6)のような炎症性メディエーターの発現調節において重要な役割を果たしていることが報告されている(非特許文献7)。動脈硬化病変において炎症が非常に重要であることはよく知られており、病変部位におけるマクロファージ炎症性メディエーターの発現による動脈硬化の悪化をLXRリガンド又はLXRアゴニストにより抑えられることが期待される(非特許文献6、8)。
また、LXRリガンド又はLXRアゴニストは肝臓、脂肪組織において糖代謝を制御し、糖尿病を改善することが期待される(非特許文献6、8)。近年、LXRアゴニストを糖尿病モデル動物に投与することにより、インスリン感受性や血糖値が改善されることが報告されている(非特許文献13、14)。また、アルツハイマー病、炎症性疾患、皮膚疾患治療薬としての可能性も指摘されている(非特許文献15)。

文献では、トリフルオロメチル源として、(トリフルオロメチル)トリメチルシランを用いているが、これに限定されるものでなく、トリエチル(トリフルオロメチル)シラン、トリイソプロピル(トリフルオロメチル)シラン、メチルジフェニル(トリフルオロメチル)シラン、ジメチル(ジフェニル)トリフルオロメチルシラン等を使用しても良い。また(ペンタフルオロエチル)トリメチルシランや(ヘプタフルオロプロピル)トリメチルシラン等のペルフルオロアルキルシラン類を用いれば、ペルフルオロアルキル化することも出来る。また、フッ素化合物として、テトラメチルアンモニウムフルオリドを用いているが、これに限定されるものでなく、テトラエチルアンモニウムフルオリド、テトラブチルアンモニウムフルオリド等のテトラアルキルアンモニウム塩や、フッ化リチウム、フッ化ナトリウム、フッ化カリウム、フッ化セシウム等の金属塩を用いても良い。溶媒はジメトキシエタンの他に、テトラヒドロフラン、トルエン、ジオキサン、N,N-ジメチルホルムアミド、N-メチルピロリドン、テトラメチルウレア、ジメチルスルホキシド、アセトニトリル、プロピオニトリル、アセトン、メチルエチルケトン等を単独又は組み合わせて使用することができる。エステルとしては、メタノール等の脂肪族アルコールとのエステル、ペンタフルオロフェノール等の芳香族アルコールとのエステル等が挙げられる。酸無水物としては、酢酸等の脂肪族カルボン酸との酸無水物、安息香酸等の芳香族カルボン酸との酸無水物等が挙げられる。
2-クロロニコチン酸クロライド(5.65g, 32.1mol)をテトラヒドロフラン(64mL)に溶解させ、氷冷下トリエチルアミン(5.59mL, 40.1mol)、メチルアミンのテトラヒドロフラン溶液(20.1mL, 40.1mol)を順に加え、室温にて3時間30分撹拌した。反応液を減圧濃縮し、再結晶(酢酸エチル/ヘキサン)を行い、表題化合物5.06g(収率92%)を淡褐色結晶として得た。
6-クロロ-N-メチルニコチンアミド(2.00g, 11.7mol)をテトラヒドロフラン(59mL)に溶解させ、氷冷下プロピルマグネシウムブロマイド(46.9mL, 46.9mol)を加え、室温にて4時間30分攪拌した。その後、氷冷下プロピルマグネシウムブロマイド(46.9mL, 46.9mol)を加え、さらに室温にて9時間攪拌した。反応液に氷冷下メタノール(140mL)、塩化アンモニウム(7.5g)を加え、室温にて30分間攪拌した。反応液に氷冷下DDQ(3.19g, 14.1mmol)を加え、室温にて2時間攪拌した。反応液にtert-ブチルメチルエーテルを加えセライト濾過を行い減圧濃縮した。酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(クロロホルム/メタノール)を用いて精製し、表題化合物1.88g(収率75%)を淡褐色結晶として得た。
6-クロロ-N-メチル-4-プロピルニコチンアミド(123mg, 0.578mol)をテトラヒドロフラン(2mL)に溶解させ、室温下1-ピペラジンエタノール(355μL, 2.89mol)を加え、100℃にて18時間攪拌した。反応液をシリカゲルカラムクロマトグラフィー(クロロホルム/メタノール)を用いて精製し、表題化合物177mg(収率>99%)を黄色結晶として得た。
6-[4-(2-ヒドロキシエチル)ピペラジン-1-イル]-N-メチル-4-プロピルニコチンアミド(477mg, 1.56mol)に無水酢酸(1.5mL)に溶解させ、氷冷下硫酸(50μL)を加え、室温にて10分攪拌した。反応液を減圧濃縮し、酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮し、表題化合物496mg(収率91%)を黄色油状物として得た。
6-[4-(2-アセトキシエチル)ピペラジン-1-イル]-N-メチル-4-プロピルニコチンアミド(496mg, 1.42mol)をアセトニトリル(7.0mL)に溶解させ、氷冷下N, N’-ジメチルアミノピリジン(35mg, 0.284mmol)、ジ-tert-ブチルジカーボネート(Boc2O)を加え、室温にて終夜攪拌した。反応液を酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮し、表題化合物754mgを粗生成物として得た。
メタノール(7.5mL)に氷冷下水素化ナトリウム(341mg, 7.10mmol)を加え、室温にて、6-[4-(2-アセトキシエチル)ピペラジン-1-イル]-4-プロピルニコチノイル(メチル)カルバミン酸 tert-ブチル(754mg)のメタノール(1.5mL)溶液を加え、室温にて1時間30分攪拌した。反応液を酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(クロロホルム/メタノール)を用いて精製し、表題化合物441mg(収率>99%)を黄色油状物として得た。
6-[4-(2-ヒドロキシエチル)ピペラジン-1-イル]-4-プロピルニコチン酸メチル(441mg, 1.43mmol)を真空ポンプ下乾燥し、エチレングリコールジメチルエーテル(7.2mL)に溶解させた。-78℃下、トリフルオロメチルトリメチルシラン (2.11mL, 14.3mmol)、テトラメチルアンモニウムフルオライド(1.20g, 12.9mmol)を順に加え、徐々に室温に上温し、終夜撹拌した。反応液に氷冷下、5%塩酸水溶液を加え、クロロホルムで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(クロロホルム/メタノール)を用いて精製し、表題化合物296mg(収率50%)を褐色結晶として得た。
1,1,1,3,3,3-ヘキサフルオロ-2-{6-[4-(2-ヒドロキシエチル)ピペラジン-1-イル]-4-プロピルピリジン-3-イル}プロパン-2-オール(208mg, 0.501mol)をジクロロメタン(2.5mL)に溶解させ、氷冷下ピリジン(121μL, 1.50mmol)、無水酢酸(142μL, 1.50mmol)を加え、室温にて3時間攪拌した。反応液にメタノールを加え、室温にて30分間攪拌した。反応液を減圧濃縮し、クロロホルムで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮し、表題化合物247mgを赤褐色油状物として得た。
酢酸 2-{4-[5-(1,1,1,3,3,3-ヘキサフルオロ-2-ヒドロキシプロパン-2-イル)-4-プロピルピリジン-2-イル]ピペラジン-1-イル}エチル(211mg, 0.461mol)をN,N-ジメチルホルムアミド(2.3mL)に溶解させ、室温下炭酸カリウム(191mg, 1.38mmol)を加えた。氷冷下塩化メトキシベンジル(125μL, 0.923mmol)を加え、室温にて終夜時間攪拌した。その後さらに室温下炭酸カリウム(96mg, 0.690mmol)を加え、氷冷下塩化メトキシベンジル(63μL, 0.462 mmol)を加え、室温にて8時間攪拌した。反応液に水を加え、クロロホルムで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮し、表題化合物479mgを赤褐色油状物として得た。
酢酸 2-(4-{5-[1,1,1,3,3,3-ヘキサフルオロ-2-(4-メトキシベンジルオキシ)プロパン-2-イル]-4-プロピルピリジン-2-イル}ピペラジン-1-イル)エチル(479mg)をメタノール(2.3mL)に溶解させ、炭酸カリウム(127mg, 0.722mmol)を加え、室温にて30分間撹拌した。反応液を減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(クロロホルム/メタノール)を用いて精製し、表題化合物117mg(収率47%)を黄色油状物として得た。
1-(4-ヒドロキシフェニル)エタノン(15.0g、110mmol)をアセトン(125mL)に溶解させ、炭酸カリウム(30.4g, 220mmol)、ヨウ化1-メチルエチル(16.5mL, 165mmol)を順に加え、70℃にて8時間撹拌した。反応液をろ過し、アセトンで洗浄し減圧濃縮した。得られた残渣に水、酢酸エチルを加え、酢酸エチルで抽出し、有機層を1N-水酸化ナトリウム水溶液、飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮し、1-[4-(1-メチルエトキシ)フェニル]エタノン 18.2g(収率93%)を白色結晶として得た。
1-[4-(1-メチルエトキシ)フェニル]エタノン(35.2g, 196mmol)をエタノール(200mL)、水(200mL)に溶解させ、シアン化ナトリウム(14.4g, 294mmol)、炭酸アンモ二ウム(226g, 9.41mol)を加え、70℃にて13時間撹拌した。反応液をろ過し水、ヘキサン/酢酸エチルで洗浄し、乾燥し、表題化合物35.1g (収率72%)を白色結晶として得た。
2-(4-{5-[1,1,1,3,3,3-ヘキサフルオロ-2-(4-メトキシベンジルオキシ)プロパン-2-イル]-4-プロピルピリジン-2-イル}ピペラジン-1-イル)エタノール(20mg, 0.0373mmol)、5-メチル-5-(4-(1-メチルエトキシ)フェニル)イミダゾリジン-2,4-ジオン(34.3mg, 0.138mmol)、トリフェニルホスフィン(34.5mg, 0.132mmol)を真空ポンプ化乾燥させた後、N,N-ジメチルホルムアミド(3mL)に溶解させ、氷冷下DEAD(51μL, 0.111mmol)を加え、室温にて1時間撹拌した。氷冷下、反応液に水(1.0mL)、2N塩酸水溶液(1.0mL)を加え、酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/アセトン)を用いて精製し、表題化合物30mg(収率99%)を黄色油状物として得た。
3-[2-(4-{5-[1,1,1,3,3,3-ヘキサフルオロ-2-(4-メトキシベンジルオキシ)プロパン-2-イル]-4-プロピルピリジン-2-イル}ピペラジン-1-イル)エチル]-5-[4-(1-メチルエトキシ)フェニル]-5-メチルイミダゾリジン-2,4-ジオン(10mg, 0.0131mmol)を酢酸エチル(2mL)に溶解させ、パラジウム炭素(1.0mg)を加え、水素添加し室温にて6時間撹拌した。反応液をセライトろ過し、減圧濃縮し、表題化合物7.3mg(収率86%)を黄色油状物として得た。
1-(6-メトキシピリジン-3-イル)エタノンを用いて実施例1-l)と同様に反応・処理し、表題化合物を白色結晶として得た。
5-(6-メトキシピリジン-3-イル)-5-メチルイミダゾリジン-2,4-ジオンを用いて実施例1と同様に反応・処理し、表題化合物を黄色油状物として得た。
2,3-ジヒドロベンゾフラン(10g, 83.2mmol)をジクロロメタン(400mL)に溶解させ、-10℃にてアセチルクロライド(11.8mL, 167mmol)、塩化アルミニウム(33.3g, 250mmol)を順に加え、-10℃にて30分間撹拌した。反応液に5%塩酸水溶液を加え、酢酸エチルで抽出し、有機層を飽和炭酸水素ナトリウム水溶液、飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮し、1-(2,3-ジヒドロベンゾフラン-5-イル)エタノン 13.4g(収率99%)を無色油状物として得た。
5-(2,3-ジヒドロベンゾフラン-5-イル)-5-メチルイミダゾリジン-2,4-ジオンを用いて実施例1と同様に反応・処理し、表題化合物を黄色油状物として得た。
3-ブロモ-4-フルオロ安息香酸(3.33g, 15.2mmol)をメタノール(30mL)に溶解させ、氷冷下アセチルクロライド(4.3mL, 60.8mmol)を加え、室温にて20時間撹拌した。反応液を減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)を用いて精製し、表題化合物3.2g (収率90%)を無色油状物として得た。
3-ブロモ-4-フルオロ安息香酸メチル(1.39g, 5.96mmol)をN,N-ジメチルホルムアミド(12mL)に溶解させ、1-ピペラジンエタノール(3.88g, 29.8mmol)、炭酸カリウム(2.47g, 17.9mmol)を加え、マイクロ波照射下100℃にて30分間撹拌した。反応液に室温下水を加え、酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/アセトン)を用いて精製し、表題化合物730mg(収率36%)を黄色油状物として得た。
3-ブロモ-4-[4-(2-ヒドロキシエチル)ピペラジン-1-イル]安息香酸メチル(100mg, 0.291mmol)を真空ポンプ下乾燥し、N,N-ジメチルホルムアミド(2.9mL)に溶解させた。氷冷下、水素化ナトリウム (15.2mg, 0.350mmol)、臭化ベンジル(38μL, 0.320mmol)を順に加え、室温にて15時間撹拌した。反応液に氷冷下水を加え、酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/アセトン)を用いて精製し、表題化合物43mg(収率34.3%)を黄色油状物として得た。
4-{4-[2-(ベンジルオキシ)エチル]ピペラジン-1-イル}-3-ブロモ安息香酸メチル(78mg, 0.180mmol)をN,N-ジメチルホルムアミド:水=3:1(1.8mL)に溶解させた。室温下、(Z)-プロペニルボロン酸(約10%のE体を含む。以下全て同じものを使用) (37mg, 0.433mmol)、テトラキストリフェニルホスフィンパラジウム(10mg, 0.00902mmol)、炭酸ナトリウム (76mg, 0.722mmol) を順に加え、マイクロ波照射下80℃にて1時間撹拌した。反応液を酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/アセトン)を用いて精製し、表題化合物61mg(収率86%)を黄色油状物として得た。
(Z)-4-{4-[2-(ベンジルオキシ)エチル]ピペラジン-1-イル}-3-(プロパ-1-エン-1-イル)安息香酸メチル(20mg, 0.0507mmol)をメタノール(1.0mL)に溶解させた。氷冷下、4N-水酸化ナトリウム水溶液(50μL)を加え、50℃にて終夜撹拌した。反応液に氷冷下4N-塩酸水溶液を加え、クロロホルムで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/アセトン)を用いて精製し、表題化合物13mg(収率65%)を黄色油状物として得た。
(Z)-4-{4-[2-(ベンジルオキシ)エチル]ピペラジン-1-イル}-3-(プロパ-1-エン-1-イル)安息香酸(13mg, 0.0331mmol)を酢酸エチル(3.3mL)に溶解させた。室温下、ペンタフルオロフェノール(7.3mg, 0.0397mmol)、N, N’-ジシクロヘキシルカルボジイミド(8.2mg, 0.0397mmol)を順に加え、室温にて14時間撹拌した。反応液に水を加え、酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/アセトン)を用いて精製し、表題化合物16mg(収率86%)を淡黄色結晶として得た。
(Z)-4-{4-[2-(ベンジルオキシ)エチル]ピペラジン-1-イル}-3-(プロパ-1-エン-1-イル)安息香酸ペルフルオロフェニル(60mg, 0.110mmol)を真空ポンプ下乾燥し、エチレングリコールジメチルエーテル(2.2mL)に溶解させた。-78℃下、トリフルオロメチルトリメチルシラン (162μL, 1.10mmol)、テトラメチルアンモニウムフルオライド(102mg, 1.10mmol)を順に加え、徐々に室温に上温し、12時間撹拌した。反応液に氷冷下水を加え、酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/アセトン)を用いて精製し、表題化合物40mg(収率73%)を橙色油状物として得た。
(Z)-2-(4-{4-[2-(ベンジルオキシ)エチル]ピペラジン-1-イル}-3-(プロパ-1-エン-1-イル)フェニル)-1,1,1,3,3,3-ヘキサフルオロプロパン-2-オール(40mg, 0.0796mmol)を真空ポンプ下乾燥し、ジクロロメタン(796μL)に溶解させた。氷冷下、ジイソプロピルエチルアミン (42μL, 0.239mmol)、クロロメチルメチルエーテル(6.6μL , 0.0876mmol)を順に加え、室温にて20時間撹拌した。さらに氷冷下、ジイソプロピルエチルアミン (84μL, 0.478mmol)、クロロメチルメチルエーテル(26μL, 0.350mmol)を順に加え、室温にて終夜撹拌した。反応液に氷冷下水を加え、酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/アセトン)を用いて精製し、表題化合物32mg(収率73%)を淡黄色油状物として得た。
(Z)-1-[2-(ベンジルオキシ)エチル]-4-{4-[1,1,1,3,3,3-ヘキサフルオロ-2-(メトキシメトキシ)プロパン-2-イル]-2-(プロパ-1-エン-1-イル)フェニル}ピペラジン(33mg, 0.0604mmol)をメタノール(4mL)に溶解させ、パラジウム炭素(3.3mg)を加え、水素添加し室温にて12時間撹拌した。反応液をセライトろ過し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/アセトン)を用いて精製し、表題化合物7.8mg(収率24%)を黄色油状物として得た。
2-(4-{4-[1,1,1,3,3,3-ヘキサフルオロ-2-(メトキシメトキシ)プロパン-2-イル]-2-プロピルフェニル}ピペラジン-1-イル)エタノール(7.8mg, 0.0170 mmol)、5-メチル-5-(4-(1-メチルエトキシ)フェニル)イミダゾリジン-2,4-ジオン(15.6mg, 0.0631mmol)、トリフェニルホスフィン(15.8mg, 0.0601mmol)を真空ポンプ下乾燥させた後、N,N-ジメチルホルムアミド(284μL)に溶解させ、氷冷下DEAD(23μL, 0.0167mmol)を加え、室温にて1時間撹拌した。氷冷下、反応液に水(1.0mL)、2N塩酸水溶液(1.0mL)を加え、酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/アセトン)を用いて精製し、表題化合物3.1mg(収率26%)を黄色油状物として得た。
3-[2-(4-{4-[1,1,1,3,3,3-ヘキサフルオロ-2-(メトキシメトキシ)プロパン-2-イル]-2-プロピルフェニル}ピペラジン-1-イル)エチル]-5-[4-(1-メチルエトキシ)フェニル]-5-メチルイミダゾリジン-2,4-ジオン(3.1mg, 0.00450mmol)をメタノール(1mL)に溶解させ、4N-塩酸水溶液(50μL)を加え、50℃にて1時間30分撹拌した。反応液を減圧濃縮し、得られた残渣に酢酸エチルを加え、氷冷下飽和炭酸水素ナトリウム水溶液を加えた。酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/アセトン)を用いて精製し、表題化合物3.0mg(収率99%)を黄色油状物として得た。
5-(ベンゾ[d][1,3]ジオキソール-5-イル)エタノンを用いて実施例1-l)と同様に反応・処理し、表題化合物を白色結晶として得た。
5-(ベンゾ[d][1,3]ジオキソール-5-イル)-5-メチルイミダゾリジン-2,4-ジオンを用いて実施例4と同様に反応・処理し、表題化合物を黄色油状物として得た。
3-ニトロ-4-フルオロ安息香酸(25.6g, 138mmol)をメタノール(300mL)に溶解させ、-78℃下塩化チオニル(18.4mL, 258mmol)を加え、-78℃にて10分間攪拌した後、室温にて終夜撹拌した。反応液を減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)を用いて精製し、表題化合物29.6g (収率>99%)を白色結晶として得た。
4-フルオロ-3-ニトロ安息香酸メチル(2.50g, 12.6mmol)のテトラヒドロフラン(75mL)溶液に、氷冷下にてピペラジン-1-カルボン酸 tert-ブチル(4.67g, 25.2mmol)、炭酸カリウム(4.34g, 25.2mmol)を順に加え、室温にて1時間攪拌した。反応液に氷冷下水を加えて、酢酸エチルで抽出した後、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(酢酸エチル)を用いて精製し、表題化合物4.83g (収率100%)を黄色結晶として得た。
4-[4-(メトキシカルボニル)-2-ニトロフェニル]ピペラジン-1-カルボン酸 tert-ブチル(200mg, 0.547mmol)の1,4-ジオキサン-水(2:1)(6.0mL)溶液に、氷冷下にて鉄粉末(76mg, 1.37mmol)、酢酸(1mL)を加え、室温にて終夜攪拌した。反応液に氷冷下水、飽和炭酸水素ナトリウム水溶液を加え、セライトろ過し、酢酸エチルで抽出した後、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/アセトン)を用いて精製し、表題化合物83mg(収率45%)を黄色油状物として得た。
4-[2-アミノ-4-(メトキシカルボニル)フェニル]ピペラジン-1-カルボン酸 tert-ブチル(231mg, 0.657mmol)のアセトニトリル(6.6mL)溶液に、氷冷下にてパラトルエンスルホン酸一水和物(375mg, 1.97mmol)を加えた後、亜硝酸ナトリウム(91mg, 1.31mmol)とヨウ化カリウム(273mg, 1.64mmol)の混合溶液を氷冷下加え、室温にて終夜攪拌した。反応液に氷冷下水、チオ硫酸ナトリウム水溶液を加え、酢酸エチルで抽出した後、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/アセトン)を用いて精製し、表題化合物127mg(収率43%)を白色結晶として得た。
4-(2-ヨード-4-(メトキシカルボニル)フェニル)ピペラジン-1-カルボン酸 tert-ブチル(127mg, 0.285mmol)をN,N-ジメチルホルムアミド:水 = 3:1(2.8mL)に溶解させた。室温下、(Z)-プロペニルボロン酸(59mg, 0.684mmol)、テトラキストリフェニルホスフィンパラジウム(16mg, 0.0143mmol)、炭酸ナトリウム (121mg, 1.14 mmol) を順に加え、マイクロ波照射下80℃にて1時間撹拌した。反応液を酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/アセトン)を用いて精製し、表題化合物96mg(収率94%)を黄色結晶として得た。
(Z)-4-[4-(メトキシカルボニル)-2-(プロパ-1-エン-1-イル)フェニル]ピペラジン-1-カルボン酸 tert-ブチル(96mg, 0.267mmol)をメタノール(3.0mL)に溶解させた。氷冷下、4N-水酸化ナトリウム水溶液(334μL)を加え、50℃にて終夜撹拌した。その後、さらに氷冷下、4N-水酸化ナトリウム水溶液(668μL)を加え、50℃にて終夜撹拌した。反応液に氷冷下4N-塩酸水溶液を加え、クロロホルムで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/アセトン)を用いて精製し、表題化合物108mg(収率>99%)を白色結晶として得た。
(Z)-4-[4-(tert-ブトキシカルボニル)ピペラジン-1-イル]-3-(プロパ-1-エン-1-イル)安息香酸(108mg, 0.267mmol)を酢酸エチル:アセトン = 4:1(5.7mL)に溶解させた。室温下、ペンタフルオロフェノール(59mg, 0.320mmol)、N, N’-ジシクロヘキシルカルボジイミド(66mg, 0.320mmol)を順に加え、室温にて10時間撹拌した。反応液に水を加え、酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/アセトン)を用いて精製し、表題化合物129mg(収率95%)を白色結晶として得た。
(Z)-4-{4-[(ペルフルオロフェノキシ)カルボニル]-2-(プロパ-1-エン-1-イル)フェニル}ピペラジン-1-カルボン酸 tert-ブチル(129mg, 0.252mmol)を真空ポンプ下乾燥し、エチレングリコールジメチルエーテル(5.0mL)に溶解させた。-78℃下、トリフルオロメチルトリメチルシラン (186μL, 1.26mmol)、テトラメチルアンモニウムフルオライド(118mg, 1.26mmol)を順に加え、徐々に室温に上温し、14時間撹拌した。反応液に氷冷下水を加え、酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/アセトン)を用いて精製し、表題化合物85mg(収率72%)を黄色結晶として得た。
(Z)-4-[4-(1,1,1,3,3,3-ヘキサフルオロ-2-ヒドロキシプロパン-2-イル)-2-(プロパ-1-エン-1-イル)フェニル]ピペラジン-1-カルボン酸 tert-ブチル(85mg, 0.181mmol)をN,N-ジメチルホルムアミド(1.8mL)に溶解させ、氷冷下にて水素化ナトリウム(9.5mg, 217mmol)、臭化ベンジル(23μL, 0.190mmol)を加え、室温にて終夜時間攪拌した。反応液に水を加え、酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/アセトン)を用いて精製し、表題化合物67mgを黄色油状物として得た。
(Z)-4-{4-[2-(ベンジルオキシ)-1,1,1,3,3,3-ヘキサフルオロプロパン-2-イル]-2-(プロパ-1-エン-1-イル)フェニル}ピペラジン-1-カルボン酸 tert-ブチル(67mg, 0.119mmol)をジクロロメタン(2.0mL)に溶解させた。氷冷下、トリフルオロ酢酸 (200μL)を加え、室温にて30分間撹拌した。反応液に氷冷下飽和炭酸水素ナトリウム水溶液を加え、クロロホルムで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(クロロホルム)を用いて精製し、表題化合物77mg(収率>99%)を白桃色油状物として得た。
(Z)-1-{4-[2-(ベンジルオキシ)-1,1,1,3,3,3-ヘキサフルオロプロパン-2-イル]-2-(プロパ-1-エン-1-イル)フェニル}ピペラジン(38mg, 0.0595mmol)をジクロロメタン(1.0mL)に溶解させた。氷冷下、N,N’-ジメチルアニリン (9.0μL, 0.0714mmol)、ブロモアセチルブロマイド (5.5μL, 0.0625mmol)を加え、氷冷下30分間撹拌した。その後、氷冷下、N,N’-ジメチルアニリン (9.0μL, 0.0714mmol)、ブロモアセチルブロマイド (5.5μL, 0.0625mmol)を加え、氷冷下30分間撹拌した。反応液を減圧濃縮し、得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/アセトン)を用いて精製し、表題化合物16mg(収率47%)を黄色油状物として得た。
5-ヒドロキシ-2-メチルピリジン(11.0g, 100mmol)をN,N-ジメチルホルムアミド(100mL)に溶解させ、氷冷下水素化ナトリウム(7.2g, 150mmol)、ヨウ化1-メチルエタン(12mL, 121mmol)を加え、室温にて終夜撹拌した。その後ヨウ化1-メチルエタン(4mL) を加え、60℃にて4時間撹拌した。反応液に水を加え、ジエチルエーテルで抽出し、有機層を飽和食塩水で洗浄し、硫酸ナトリウムで乾燥し減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)を用いて精製し、5-(1-メチルエトキシ)-2-メチルピリジン 12.7g (収率84%)を黄色油状物として得た。
5-メチル-5-[5-(1-メチルエトキシ)ピリジン-2-イル]イミダゾリジン-2,4-ジオン(7.7mg, 0.0309mmol)をN,N-ジメチルホルムアミド(2.8mL)に溶解させ、氷冷下炭酸カリウム(9.3mg, 0.0674 mmol)を加え、室温にて5分間撹拌した。その後、氷冷下で1-(4-{4-[2-(ベンジルオキシ)-1,1,1,3,3,3-ヘキサフルオロプロパン-2-イル]-2-(プロパ-1-エン-1-イル)フェニル}ピペラジン-1-イル)-2-ブロモエタノン(16mg, 0.0281mmol)を加え、室温にて終夜時間攪拌した。その後、60℃にて10時間攪拌した。氷冷下、反応液に水を加え、酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/アセトン)を用いて精製し、表題化合物17mg(収率80%)を黄色油状物として得た。
(Z)-3-[2-(4-{4-[2-(ベンジルオキシ)-1,1,1,3,3,3-ヘキサフルオロプロパン-2-イル]-2-(プロパ-1-エン-1-イル)フェニル}ピペラジン-1-イル)-2-オキソエチル]-5-[5-(1-メチルエトキシ)ピリジン-2-イル]-5-メチルイミダゾリジン-2,4-ジオン(17 mg, 0.0225mmol)をメタノール(2.0mL)に溶解させ、水酸化パラジウム(6.0mg)を加え、水素添加し室温にて12時間撹拌した。反応液をセライトろ過し、減圧濃縮し、得られた残渣をシリカゲルカラムクロマトグラフィー(クロロホルム)を用いて精製し、表題化合物10mg(収率67%)を淡黄色油状物として得た。
4-プロピルピリジン(10.0g, 82.5mmol)のトルエン(170mL)の溶液に、室温下、ナトリウムアミド(6.4g, 165mmol)を加え、140℃にて2日間攪拌した。氷冷下、反応液に6N-塩酸水溶液(30mL)を加え、酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル/1%トリエチルアミン)を用いて精製し、表題化合物5.70g (収率51%)を赤褐色固体として得た。
4-プロピルピリジン-2-アミン(5.70g, 41.8mmol)のエタノール(210mL)の溶液に、氷冷下、臭素のエタノール溶液(臭素2.38mL/エタノール50mL)を加え、氷冷下30分間攪拌した。氷冷下、反応液に炭酸水素ナトリウム水溶液を加え、減圧濃縮した。酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)を用いて精製し、表題化合物4.34g (収率48%)を褐色油状物として得た。
5-ブロモ-4-プロピルピリジン-2-アミン(5.67g, 26.3mmol)に10℃にて48%臭化水素(21mL)を加えた。氷冷下、臭素(4.0mL)を加え、-10℃にて亜硝酸ナトリウム(4.54g)の水溶液(4.5mL)を滴下した。-10℃にて30分間攪拌した後、1N-水酸化ナトリウム水溶液(10mL)加え、室温下2時間攪拌した。反応液に水を加えた。ジエチルエーテルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ジエチルエーテル)を用いて精製し、表題化合物6.96g (収率95%)を褐色油状物として得た。
2,5-ジブロモ-4-プロピルピリジン(100mg, 0.444mmol)のN,N-ジメチルホルムアミド(0.9mL)の溶液に、室温下ジニトリル亜鉛(57mg, 0.449mmol), テトラキストリフェニルホスフィンパラジウム(25mg, 0.0222mmol)を加え、マイクロ波照射下100℃にて20分間撹拌した。反応液に飽和炭酸水素ナトリウム水溶液を加え、ヘキサン/酢酸エチル(4/1)で抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)を用いて精製し、表題化合物60mg(収率60%)を無色結晶として得た。
5-ブロモ-4-プロピルピコリノニトリル(15mg, 0.0670mmol)のメタノール(1.3mL)の溶液に、氷冷下硫酸(180μL, 0.0670mmol)を加え、80℃にて終夜攪拌した。反応液に氷冷下1N-水酸化ナトリウム水溶液、飽和炭酸水素ナトリウム水溶液を加え、メタノールを減圧濃縮した。酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)を用いて精製し、表題化合物11mg(収率62%)を褐色結晶として得た。
5-ブロモ-4-プロピルピコリン酸メチル(100mg, 0.387mmol)のトルエン(1.94mL)の溶液に、1-tert-ブチトキシカルボニルピペラジン(84mg, 0.453mmol), トリス(ジベンジリデンアセトン)ジパラジウム(8.3mg, 0.00902mmol)、BINAP(11.2mg, 0.0180mmol)、炭酸セシウム(295mg, 0.906mmol)を加え、110℃にて14時間攪拌した。反応液に水を加え、クロロホルムで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(酢酸エチル)を用いて精製し、表題化合物70mg(収率49%)を淡黄色結晶として得た。
4-[6-(メトキシカルボニル)-4-プロピルピリジン-3-イル]ピペラジン-1-カルボン酸 tert-ブチル(111mg, 0.307mmol)のメタノール(3.0mL)の溶液に、氷冷下、4N-水酸化ナトリウム水溶液(383μL, 1.53mmol)を加え、室温にて終夜攪拌した。反応液に氷冷下4N-塩酸水溶液を加え、クロロホルムで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(クロロホルム)を用いて精製し、表題化合物134mg(収率>99%)を黄色結晶として得た。
5-[4-(tert-ブトキシカルボニル)ピペラジン-1-イル]-4-プロピルピコリン酸(134mg, 0.307mmol)を酢酸エチル(6.2mL)に溶液させた。室温下、ペンタフルオロフェノール(68mg, 0.368mmol)、N, N’-ジシクロヘキシルカルボジイミド(76mg, 0.368mmol)を順に加え、室温にて終夜撹拌した。反応液に氷冷下水を加え、酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)を用いて精製し、表題化合物189mg(収率>99%)を黄褐色結晶として得た。
4-{6-[(ペルフルオロフェノキシ)カルボニル]-4-プロピルピリジン-3-イル}ピペラジン-1-カルボン酸 tert-ブチル(189mg, 0.307mmol)を真空ポンプ化乾燥し、エチレングリコールジメチルエーテル(6.2mL)に溶解させた。-78℃下、トリフルオロメチルトリメチルシラン (227μL, 1.53mmol)、テトラメチルアンモニウムフルオライド(143mg, 1.53mmol)を順に加え、徐々に室温に上温し、終夜撹拌した。反応液に氷冷下水を加え、酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)を用いて精製し、表題化合物60mg(収率41%)を黄色油状物として得た。
4-[6-(1,1,1,3,3,3-ヘキサフルオロ-2-ヒドロキシプロパン-2-イル)-4-プロピルピリジン-3-イル]ピペラジン-1-カルボン酸 tert-ブチル(60mg, 0.126mmol)を真空ポンプ化乾燥し、N,N-ジメチルホルムアミド(2.5mL)に溶解させた。氷冷下、水素化ナトリウム (7.3mg, 0.152mmol)、臭化ベンジル(17μL, 0.139mmol)を順に加え、室温にて終夜撹拌した。反応液に氷冷下水を加え、酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/アセトン)を用いて精製し、表題化合物54mg(収率76%)を黄色油状物として得た。
4-{6-[2-(ベンジルオキシ)-1,1,1,3,3,3-ヘキサフルオロプロパン-2-イル]-4-プロピルピリジン-3-イル}ピペラジン-1-カルボン酸 tert-ブチル(32mg, 0.0563mmol)をジクロロメタン(10mL)に溶解させた。氷冷下、トリフルオロ酢酸 (43μL , 0.563mmol)を加え、室温にて30分間撹拌した。反応液に氷冷下飽和炭酸水素ナトリウム水溶液を加え、クロロホルムで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(クロロホルム)を用いて精製し、表題化合物27mg(収率>99%)を黄色油状物として得た。
1-{6-[2-(ベンジルオキシ)-1,1,1,3,3,3-ヘキサフルオロプロパン-2-イル]-4-プロピルピリジン-3-イル}ピペラジン(27mg, 0.0563mmol)をジクロロメタン(1.0mL)に溶解させた。氷冷下、N,N-ジメチルアニリン (15μL, 0.118mmol)、ブロモアセチルブロマイド (5.1μL, 0.0591mmol)を加え、氷冷下20分間撹拌した。反応液を減圧濃縮し、得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/アセトン)を用いて精製し、表題化合物25mg(収率78%)を黄色油状物として得た。
5-(2,3-ジヒドロベンゾフラン-5-イル)-5-メチルイミダゾリジン-2,4-ジオン(5.6mg, 0.0240mmol)をN,N-ジメチルホルムアミド(500μL)に溶解させ、氷冷下炭酸カリウム(7.5mg, 0.0545mmol)を加え、室温にて5分間撹拌した。その後、氷冷下1-(4-{6-[2-(ベンジルオキシ)-1,1,1,3,3,3-ヘキサフルオロプロパン-2-イル]-4-プロピルピリジン-3-イル}ピペラジン-1-イル)-2-ブロモエタノン(13mg, 0.0218mmol)を加え、室温にて18時間攪拌した。氷冷下、反応液に水を加え、酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/アセトン)を用いて精製し、表題化合物13mg(収率78%)を黄色油状物として得た。
3-[2-(4-{6-[2-(ベンジルオキシ)-1,1,1,3,3,3-ヘキサフルオロプロパン-2-イル]-4-プロピルピリジン-3-イル}ピペラジン-1-イル)-2-オキソエチル]-5-(2,3-ジヒドロベンゾフラン-5-イル)-5-メチルイミダゾリジン-2,4-ジオン(13 mg, 0.0170mmol)をメタノール(1.5mL)に溶解させ、パラジウム炭素(2.0mg)を加え、水素添加し室温にて2時間撹拌した。反応液をセライトろ過し、減圧濃縮し、得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/アセトン)を用いて精製し、表題化合物7.3mg(収率80%)を黄色油状物として得た。
1-tert-ブトキシカルボニルピペラジンの代わりに(R)-3-メチル-1-tert-ブトキシカルボニルピペラジンを用い、その他は実施例6-b)と同様に反応・処理し、表題化合物を黄色油状物として得た。

a-2-1-1) 2-(ベンジルアミノ)プロパン酸 (S)-メチルの製造:
L-アラニンメチルエステル塩酸塩(15.0g, 107mmol)をN, N’-ジメチルホルムアミド(150mL)に溶解させ、炭酸カリウム(32.5g, 235mmol)、臭化ベンジル(18.4g, 107mmol)を加え、80℃にて終夜攪拌した。その後、室温に戻し、反応溶液をセライトろ過し、酢酸エチルにて抽出し、水、飽和食塩水にて洗浄し、硫酸ナトリウムにて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)にて精製し、表題化合物(12.7g, 61%)を無色油状物として得た。
2-(ベンジルアミノ)プロパン酸 (S)-メチル(9.28g, 48.1mmol)のジクロロメタン(100mL)溶液にBoc-D-アラニン(10g, 52.9mmol)を加えた後、氷冷下、4-ジメチルアミノピリジン(587mg, 4.81mmol)、EDCI(11g, 57.7mmol)を加え、室温にて終夜攪拌した。反応溶液をクロロホルムにて抽出し、5%クエン酸水溶液、水、飽和炭酸水素ナトリウム水溶液にて洗浄し、硫酸ナトリウムにて乾燥後、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)にて精製し、表題化合物(9.34g, 53%)を無色油状物として得た。
2-{(R)-N-ベンジル-2-[(tert-ブトキシカルボニル)アミノ]プロパンアミド}プロパン酸 (S)-メチル(9.34g, 25.6mmol)のジクロロメタン(100mL)溶液に氷冷下、トリフルオロ酢酸(14.6g, 128mmol)を加え、室温にて終夜攪拌した。反応溶液を減圧濃縮した後、メタノール(40mL)を加え、氷冷下飽和炭酸水素ナトリウム水溶液(pH=8 になるまで加えた)を加えた後、80℃にて終夜攪拌した。反応溶液を減圧濃縮した後、酢酸エチルにて抽出し、飽和食塩水にて洗浄し、硫酸ナトリウムにて乾燥後、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(酢酸エチル)にて精製し、表題化合物(4.05g, 68%)を無色油状物として得た。
アルゴン雰囲気下、(3R,6S)-1-ベンジル-3,6-ジメチルピペラジン-2,5-ジオン(4.05g, 17.4mmol)のテトラヒドロフラン(70mL)溶液に氷冷下、リチウムアルミニウムハイドライド(993mg, 26.2mmol)を加え、室温にて10分間攪拌し、その後、80℃にて4時間攪拌した。その後、反応溶液に氷冷下、リチウムアルミニウムハイドライド(331mg, 8,72mmol)を加え、80℃にて1時間30分攪拌した。その後、反応溶液に氷冷下、テトラヒドロフラン、水を加え、室温にて終夜攪拌した。
反応溶液をセライトろ過し、酢酸エチル、水で洗浄し、硫酸ナトリウムにて乾燥後、減圧濃縮し、表題化合物(3.52g, 99%)を黄色油状物として得た。
(2R,5S)-1-ベンジル-2,5-ジメチルピペラジン(3.52g, 17.2mmol)のテトラヒドロフラン(50mL)溶液に氷冷下、トリメチルアミン(2.12g, 20.9mmol)、ジ-tert-ブチルカルボネート(4.56g, 20.9mmol)を加え、15分間攪拌した。その後、室温にて5時間30分攪拌した。反応溶液に水を加え、酢酸エチルにて抽出し、飽和食塩水にて洗浄し、硫酸ナトリウムにて乾燥後、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)にて精製し、表題化合物(3.16g, 60%)を淡黄色油状物として得た。
アルゴン雰囲気下、4-ベンジル-2,5-ジメチルピペラジン-1-カルボン酸 (2R,5S)-tert-ブチル(2.0g, 6.57mmol)をメタノール(60mL)に溶解させ、10%パラジウム炭素(670mg)、ギ酸(1.5g, 32.9mmol)を加えた。室温にて2時間攪拌した。その後、反応溶液をセライトろ過し、減圧濃縮した。得られた残渣にクロロホルム、飽和炭酸水素ナトリウム水溶液を加え、クロロホルムにて抽出し、硫酸ナトリウムにて乾燥し、減圧濃縮し、表題化合物(1.56g, >100%)を黄色油状物として得た。
L-アラニンメチルエステル塩酸塩の代わりにD-アラニンメチルエステル塩酸塩を用いて、実施例14-1-a-2-1-1)から同a-2-1-6)と同様に反応・処理し、表題化合物を無色油状物として得た。
a-3-1-1) racemic trans-1-ベンジル-2,5-ジメチルピペラジンの製造:
trans-2,5-ジメチルピペラジン (25g, 219mmol)をシクロヘキサン(250mL)-水(35mL)に溶解させ、室温にてtetra-ブチルアンモニウムクロライド (2.2g, 15.5mmol)、水酸化ナトリム水溶液(23mL)、ベンジルクロライド (26.5g, 208mmol)を加えた。40℃にて18時間撹拌した。その後、室温に戻し、反応液をシクロヘキサンで抽出した。有機層に37%塩酸水溶液を加え、攪拌した。抽出操作により得た水層に2-メチルテトラヒドロフラン、50%水酸化ナトリウム水溶液を加えて、2-メチルテトラヒドロフランにて抽出した。得られた有機層を無水硫酸ナトリウムで乾燥後、減圧濃縮し、表題化合物34g(収率76%) を無色油状物として得た。
racemic trans-1-ベンジル-2,5-ジメチルピペラジン (2.35g, 11.5mmol)をメタノール(14mL)に溶解させ、D-(-)-酒石酸 (3.45g, 23.0mmol)を加え、5℃にて20時間静置した。メタノールにてろ取し、減圧下乾燥し、表題化合物2.66g (収率44%) を白色結晶として得た。更に得られた白色結晶をメタノール(27mL)に溶解させ、再結晶を行い、表題化合物1.7g (収率28%, 99.9% ee) を白色結晶として得た。
(2S, 5R)- 1-ベンジル-2,5-ジメチルピペラジン-2-酒石酸塩(5.0g, 9.91mmol)を2-メチルテトラヒドロフラン(30mL)に溶解させ、氷冷下にて水酸化ナトリウム水溶液 (水酸化ナトリウム:2.62g, 65.4mmol、水:18mL)、二炭酸ジ-tert-ブチル(2.2g, 9.91mmol)の2-メチルテトラヒドロフラン溶液を加え、室温下にて終夜撹拌した。その後、反応液を2-メチルテトラヒドロフランにて抽出し、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥後、減圧濃縮し、表題化合物3.11g (収率>100%) を白色結晶として得た。
4-ベンジル-2,5-ジメチルピペラジン-1-カルボン酸 (2S, 5R)- tert-ブチル (3.11g, 10.2mmol)をメタノール(19mL)に溶解させ、アルゴン雰囲気下、パラジウム炭素(156mg)を加え、氷冷下ギ酸 (2.4g, 51.1mmol)を加えた。室温に戻し、終夜攪拌した。その後、反応液をセライトろ過し、ろ液を減圧濃縮した。得られた残渣に水、ヘキサンを加えて抽出し、水層に水酸化ナトリウム水溶液を加え、2-メチルテトラヒドロフランにて抽出し、無水硫酸ナトリウムにて乾燥後、減圧濃縮し、表題化合物2.21g(収率>100%) を無色油状物として得た。
D-(-)-酒石酸の代わりにL-(+)-酒石酸を用いて、実施例14-1-a-3-1-2)以下と同様に反応・処理し、表題化合物を無色油状物として得た。
ピペラジン-1-カルボン酸 tert-ブチルの代わりに2,5-ジメチルピペラジン-1-カルボン酸 (2R, 5S)-tert-ブチルを用いて、実施例6-b)と同様に反応・処理し、表題化合物を黄色油状物として得た。
(2R, 5S)-4-[4-(メトキシカルボニル)-2-ニトロフェニル]-2,5-ジメチルピペラジン-1-カルボン酸 tert-ブチルを用いて実施例6-c)と同様に反応・処理し、表題化合物を黄色油状物として得た。
(2R, 5S)- 4-[2-アミノ-4-(メトキシカルボニル)フェニル]-2,5-ジメチルピペラジン-1-カルボン酸 tert-ブチルを用いて実施例6-d)と同様に反応・処理し、表題化合物を黄色油状物として得た。
(2R, 5S)- 4-[2-ヨード-4-(メトキシカルボニル)フェニル]-2,5-ジメチルピペラジン-1-カルボン酸 tert-ブチルを用いて実施例6-e)と同様に反応・処理し、表題化合物を黄色油状物として得た。
(2R, 5S, Z)- 4-[4-(メトキシカルボニル)-2-(プロパ-1-エン-1-イル)フェニル]-2,5-ジメチルピペラジン-1-カルボン酸tert-ブチルを用いて実施例6-f)と同様に反応・処理し、表題化合物を黄色油状物として得た。
(2R, 5S, Z)- 4-[4-(tert-ブトキシカルボニル)-2,5-ジメチルピペラジン-1-イル]-3-(プロパ-1-エン-1-イル)安息香酸を用いて実施例6-g)と同様に反応・処理し、表題化合物7.3mg(収率80%)を黄色油状物として得た。
(2R, 5S, Z)- 2,5-ジメチル-4-{4-[(ペルフルオロフェノキシ)カルボニル]-2-(プロパ-1-エン-1-イル)フェニル}ピペラジン-1-カルボン酸 tert-ブチルを用いて実施例6-h)と同様に反応・処理し、表題化合物を黄色油状物として得た。
4-[4-(1,1,1,3,3,3-ヘキサフルオロ-2-ヒドロキシプロパン-2-イル)-2-((Z)- プロパ-1-エン-1-イル)フェニル]-2,5-ジメチルピペラジン-1-カルボン酸 (2R, 5S)-tert-ブチルを用いて実施例6-j)と同様に反応・処理し、表題化合物を黄色油状物として得た。
(2R, 5S, Z)- 2-[4-(2,5-ジメチルピペラジン-1-イル)-3-(プロパ-1-エン-1-イル)フェニル]-1,1,1,3,3,3-ヘキサフルオロプロパン-2-オールを用いて実施例12-i)と同様に反応・処理し、表題化合物を黄色油状物として得た。
(2R, 5S, Z)- 1-{4-[1,1,1,3,3,3-ヘキサフルオロ-2-(メトキシメトキシ)プロパン-2-イル]-2-(プロパ-1-エン-1-イル)フェニル}-2,5-ジメチルピペラジンを用いて実施例6-k)と同様に反応・処理し、表題化合物を黄色油状物として得た。
5-[4-(1-メチルエトキシ)フェニル]-5-メチルイミダゾリジン-2,4-ジオン(7.7mg, 0.0311mmol)をN,N-ジメチルホルムアミド(500μL)に溶解させ、氷冷下炭酸カリウム(8.6mg, 0.0622mmol)を加え、室温にて5分間撹拌した。その後、氷冷下、(2R,5S,Z)-2-ブロモ-1-(4-{4-[1,1,1,3,3,3-ヘキサフルオロ-2-(メトキシメトキシ)プロパン-2-イル]-2-(プロパ-1-エン-1-イル)フェニル}-2,5-ジメチルピペラジン-1-イル)エタノン(17mg, 0.0311mmol)を加え、室温にて18時間攪拌した。氷冷下、反応液に水を加え、酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣を酢酸エチル(1.0mL)に溶解させ、4N-塩酸-酢酸エチル溶液(1.0mg)を加え、室温にて1時間撹拌した。氷冷下、反応液に水、飽和炭酸水素ナトリウム水溶液を加え、酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)を用いて精製し、表題化合物17.4mg(収率83%)を黄色油状物として得た。
2,5-ジメチルピペラジン-1-カルボン酸 (2R, 5S)-tert-ブチルの代わりに2,5-ジメチルピペラジン-1-カルボン酸 (2S, 5R)-tert-ブチルを用いて、実施例14-1の(2R, 5S, Z)-3-(2-{4-[4-(1,1,1,3,3,3-ヘキサフルオロ-2-ヒドロキシプロパン-2-イル)-2-(プロパ-1-エン-1-イル)フェニル]-2,5-ジメチルピペラジン-1-イル}-2-オキソエチル)-5-[4-(1-メチルエトキシ)フェニル]-5-メチルイミダゾリジン-2,4-ジオンの製造と同様に反応・処理し、表題化合物を黄色油状物として得た。
(2R, 5S, Z)-3-(2-{4-[4-(1,1,1,3,3,3-ヘキサフルオロ-2-ヒドロキシプロパン-2-イル)-2-(プロパ-1-エン-1-イル)フェニル]-2,5-ジメチルピペラジン-1-イル}-2-オキソエチル)-5-[4-(1-メチルエトキシ)フェニル]-5-メチルイミダゾリジン-2,4-ジオン(実施例14-1)の代わりに(2S, 5R, Z)-3-(2-{4-[4-(1,1,1,3,3,3-ヘキサフルオロ-2-ヒドロキシプロパン-2-イル)-2-(プロパ-1-エン-1-イル)フェニル]-2,5-ジメチルピペラジン-1-イル}-2-オキソエチル)-5-[4-(1-メチルエトキシ)フェニル]-5-メチルイミダゾリジン-2,4-ジオン(実施例14-2)を用いて、実施例15-1の製造と同様に反応・処理し、表題化合物を黄色油状物として得た。

1-(4-ヒドロキシフェニル)エタノン(1.36g、10mmol)をアセトン(50mL)に溶解させ、ヨウ化テトラブチルアンモニウム(370mg, 1.0mmol)、炭酸カリウム(2.76g, 20mmol)、3-クロロ-2-メチル-1-プロペン(1.5mL, 15mmol)を順に加え、70℃にて終夜撹拌した。反応液をろ過し、アセトンで洗浄し減圧濃縮した。得られた残渣に水、酢酸エチルを加え、酢酸エチルで抽出し、有機層を1N-水酸化ナトリウム水溶液、飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮し、1-(4-(2-メチルアリルオキシ)フェニル)エタノン 1.71g(収率90%)を無色油状物として得た。
実施例24-1と同様に反応・処理し、表題化合物を黄色油状物として得た。
実施例25-1と同様に反応・処理し、表題化合物を黄色油状物として得た。
1-(3-ヒドロキシフェニル)エタノン(1.36g、10mmol)をアセトン(50mL)に溶解させ、炭酸カリウム(2.76g, 20mmol)、ヨウ化1-メチルエチル(1.5mL, 15mmol)を順に加え、70℃にて終夜撹拌した。反応液をろ過し、アセトンで洗浄し減圧濃縮した。得られた残渣に水、酢酸エチルを加え、酢酸エチルで抽出し、有機層を1N-水酸化ナトリウム水溶液、飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮し、1-(3-(1-メチルエトキシ)フェニル)エタノン 1.67g(収率94%)を黄色油状物として得た。
実施例26-1と同様に反応・処理し、表題化合物を黄色油状物として得た。

シクロプロピルフェニルチオエーテル( 5.0g, 33.3mmol) をテトラヒドロフラン (50mL) に溶解し、アルゴン雰囲気下0℃にてn-ブチルリチウム (25.1mL, 39.9mmol) を5分間にて滴下した。その後―78℃にてN-ヨウ化スクシンイミド(8.99g, 39.9mmol)のテトラヒドロフラン (100mL)溶液を滴下した。終夜攪拌し、室温まで徐々に昇温させた。反応液に飽和炭酸水素ナトリウム水溶液を加え、ヘキサンで抽出した。有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムで乾燥後、減圧濃縮し、シリカゲルカラムクロマトグラフィー (ヘキサン) で精製し、粗生成物5.64g ((1-ヨードシクロプロピル)(フェニル)スルファン:シクロプロピルフェニルチオエーテル=2.4:1) を得た。
次に、5-ヒドロキシ-2-メチルピリジン(1.82g, 16.7mmol) をトルエン (150mL) に溶解し、炭酸銀(9.19g, 33.3mmol)、(1-ヨードシクロプロピル)(フェニル)スルファン:シクロプロピルフェニルチオエーテル(=2.4:1)混合物 (5.64g, 16.7mmol(*1-ヨードシクロプロピル)(フェニル)スルファンの量で換算)) を加え、室温にて終夜攪拌した。その後、酢酸(200mL)を加え、10分間攪拌した。反応液をセライトろ過し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー (ヘキサン/酢酸エチル) で精製し、表題化合物1.88g (収率44%) を黄色油状物として得た。
2-メチル-5-[1-(フェニルチオ)シクロプロポキシ]ピリジン(2.04g, 7.93mmol) をクロロホルム (15mL) に溶解し、アルミナ(5.0g)、オクソン (3.79g, 6.18mmol) を加え、80℃にて1時間攪拌した。その後、さらにオクソン (1.36g, 2.22mmol) を加え、1時間攪拌した。反応液をセライトろ過し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー (ヘキサン/酢酸エチル) で精製し、表題化合物542mg(収率24%) を黄色油状物として得た。
2-メチル-5-[1-(フェニルスルホニル)シクロプロポキシ]ピリジン(540mg, 1.87mmol)をメタノール (5.5mL) に溶解し、亜リン酸ナトリウム(671mg, 5.598mmol)、氷冷下ナトリウムアマルガム(3.58g, 7.47mmol)を加え、同温にて30分間攪拌した後、室温にて3時間攪拌した。反応液に飽和炭酸水素ナトリウム水溶液を加え、ジエチルエーテルで抽出した。有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムで乾燥後、減圧濃縮した。得られた残渣を蒸留精製し、表題化合物240 mg (収率86%) を黄色油状物として得た。
5-シクロプロポキシ-2-メチルピリジンを用いて実施例6-l)と同様に反応・処理し、表題化合物を白色結晶として得た。

3-ブロモフェノール(1.68g, 9.77mmol)のN,N’-ジメチルホルムアミド (32mL)溶液に、氷冷下水素化ナトリウム(純度50%) (516mg, 10.7mmol)、0℃にて臭化アセトアルデヒドジエチルアセタール(1.76mL, 11.7mmol) を加え、120℃にて終夜撹拌した。室温にて反応液に水を加え、ジエチルエーテルで抽出した。有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムで乾燥後、減圧濃縮し、シリカゲルカラムクロマトグラフィー (ヘキサン) で精製し、表題化合物2.69g (収率>100%) を黄色油状物として得た。
1-ブロモ-3-(2,2-ジエトキシエトキシ)ベンゼン(2.3g, 8.35mmol)のトルエン(28mL)溶液に、PPA (5.0mL)、加熱還流下終夜攪拌した。室温にて反応液に水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムで乾燥後、減圧濃縮し、シリカゲルカラムクロマトグラフィー(ヘキサン) で精製し、表題化合物1.2g(収率68%、7-ブロモベンゾフランとの混合物)を黄色油状物として得た。
6-ブロモベンゾフランと7-ブロモベンゾフランの混合物(1.12g, 5.68mmol)のトルエン(19mL)溶液に、テトラキストリフェニルホスフィンパラジウム (650mg, 0.57mmol)、トリブチル(1-エトキシビニル)スズ (2.11mL, 6.25mmol)を加え100℃にて終夜攪拌した。室温にて反応液に水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムで乾燥後、減圧濃縮し、シリカゲルカラムクロマトグラフィー (ヘキサン) で精製し、表題化合物(280mg)を黄色結晶として得た。
1-(ベンゾフラン-6-イル)エタノンを用いて実施例1- l)と同様に反応・処理し、表題化合物を白色結晶として得た。
5-(ベンゾフラン-6-イル)-5-メチルイミダゾリジン-2,4-ジオンを用いて実施例1- l)と同様に反応・処理し、表題化合物を白色結晶として得た。
4-フルオロ-3-ニトロ安息香酸 (1.5g, 8.10mmol)をN,N-ジメチルホルムアミド(1.0mL)に溶解させ、塩化チオニル(16.6mL)を加え、2時間加熱還流した。その後、反応液を減圧濃縮し、粗生成物(3.57g)を得た。次にアルゴン雰囲気下、粗生成物(3.57g)のエチレングリコールジメチルエーテル(81mL)溶液に、氷冷下にてトリメチルシリルトリフルオロメタン(3.0mL. 20.26mmol)、テトラメチルアンモニウムフルオライド(1.89g. 20.26mmol)を加え、同温にて20分間攪拌した。その後、さらに室温にて12時間攪拌した。氷冷下にて反応液に水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムで乾燥後、減圧濃縮し、シリカゲルカラムクロマトグラフィー (ヘキサン) で精製し、表題化合物2.99g(収率>100%) を黄色油状物として得た。
1,1,1,3,3,3-ヘキサフルオロ-2-(4-フルオロ-3-ニトロフェニル)プロパン-2-オールとピペラジン-1-カルボン酸 tert-ブチルを用いて、実施例6-b)と同様に反応・処理し、表題化合物を黄色油状物として得た。
4-[4-(1,1,1,3,3,3-ヘキサフルオロ-2-ヒドロキシプロパン-2-イル)-2-ニトロフェニル]ピペラジン-1-カルボン酸 tert-ブチルエステルを用いて、実施例6-i)と同様に反応・処理し、表題化合物を黄色油状物として得た。

2,3-ヒドロキシ-5-ブロモピリジン (100mg, 0.53mmol)をN,N’-ジメチルホルムアミド (5mL)に溶解させ、アルゴン雰囲気中氷冷下にて水素化ナトリム (30mg, 0.63mmol)を加えた。5分後、同温にて1,2-ジブロモエタン(50μL, 0.58mmol) のN,N’-ジメチルホルムアミド(5mL)溶液を加え、110℃にて14時間撹拌した。反応液を室温に戻し、水を加えた後、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムで乾燥後、減圧濃縮し、分取薄層クロマトグラフィー (n-ヘキサン/ 酢酸エチル=2/1) で精製し、表題化合物20mg(収率17%) を白色固体として得た。
アルゴン雰囲気下、7-ブロモ-2,3-ジヒドロ-[1,4]ジオキシノ[2,3-b]ピリジン(65mg, 0.30mmol) をトルエン (1.5mL) に溶解し、テトラキス(トリフェニルホスフィン)パラジウム (35mg, 0.03mmol)、1-エトキシエテニルトリn-ブチルスズ(112μL, 0.33mmol) を加え、100℃にて一晩中撹拌した。反応液を室温に戻し、1N 塩酸を加えた。反応液をセライトろ過し、反応液に酢酸エチルを加えて抽出した。有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムで乾燥後、減圧濃縮し、分取薄層クロマトグラフィー (n-ヘキサン/ 酢酸エチル=1/1) で精製し、表題化合物37mg(収率69%) を白色固体として得た。
1-(2,3-ジヒドロ-[1,4]ジオキシノ[2,3-b]ピリジン-7-イル)エタノンを用いて実施例1-l)と同様に反応・処理し、表題化合物を白色結晶として得た。

1-tert-ブトキシカルボニルピペラジンの代わりに(S)-3-メチル-1-tert-ブトキシカルボニルピペラジンを用い、その他は実施例6-b)と同様に反応・処理した後、実施例12-b)からg)と同様に反応・処理し、表題化合物を黄色油状物として得た。

2-メチル-5-ヒドロキシピリジンを用いて実施例4-h)と同様に反応・処理し、表題化合物を無色油状物として得た。
5-メトキシメトキシ-2-メチルピリジン(3.0g, 19.6mmol)のテトラヒドロフラン (100mL) 溶液に、-78℃にてn-ブチルリチウム (18.4mL, 29.4mmol)を加え、-78℃にて40分間攪拌した。その後、イソプロポキシボラン酸エステル(6.8mL, 29.4mmol)を加え、-78℃にて45分間攪拌した。反応液に1N 塩酸水溶液を加えた後、昇温させた。反応液を酢酸エチルで抽出した。有機層を無水硫酸ナトリウムで乾燥後、減圧濃縮した。得られた残渣をジエチルエーテルで洗浄ろ過し、表題化合物2.08g (収率54%) を白色結晶として得た。
5-(メトキシメトキシ)-2-メチルピリジン-4-イルボロン酸(500mg, 2.54mmol)のテトラヒドロフラン (12.7mL) 溶液に、室温にて過酸化水素水溶液(純度30%) (2.9mL, 25.4mmol)を加え、室温にて4時間攪拌した。反応液に飽和過硫酸ナトリウム水溶液を加えた後、減圧濃縮した。得られた残渣をクロロホルム/メタノールで洗浄ろ過し、表題化合物440mg(収率>100%) を黄色アモルファスとして得た。
2-(5-(メトキシメトキシ)-2-メチルピリジン-4-イルオキシ)エタノール(900mg, 4.22mmol)を酢酸エチル(10mL)に溶解させ、4N 塩酸-酢酸エチル溶液(10mL)を加え、室温にて5時間攪拌した。反応終了後、氷冷下4N 水酸化ナトリウム水溶液を用いて反応溶液をpH=8 にした。反応溶液を減圧濃縮した。得られた残渣をクロロホルム/メタノールにて洗浄し、乾燥し、粗生成物として4-(2-ヒドロキシエトキシ)-6-メチルピリジン-3-オール(1.2g)を白色固形物として得た。
4-(2-ヒドロキシエトキシ)-6-メチルピリジン-3-オール(714mg, 4.22mmol)、トリフェニルホスフィン(1.66g, 6.33mmol)を真空ポンプ化乾燥させた後、テトラヒドロフラン(42mL)に溶解させ、氷冷下アゾジカルボン酸ジ-tert-ブチル(DBAD)(1.46g, 6.33mmol)を加え、室温にて終夜撹拌した。氷冷下、反応液に水を加え、酢酸エチルで抽出し、有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを用いて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/アセトン、クロロホルム/メタノール)を用いて精製し、粗生成物として表題化合物1.74g(収率>100%)を白色固体として得た。
7-メチル-2,3-ジヒドロ-[1,4]ジオキシノ[2,3-c]ピリジンを用いて実施例6-l)と同様に反応・処理し、表題化合物を無色油状物として得た。

2,3-ジヒドロキシピリジン (2.22g, 20.0mmol)をN,N’-ジメチルホルムアミド(100mL)に溶解させ、室温にて炭酸カリウム (5.52g, 40.0mmol)を加えた。5分後、同温にて1,2-ジブロモエタン(2.6mL, 30.0mmol)を加え、90℃にて終夜撹拌した。反応液を室温に戻し、ろ過後、得られたろ液を減圧濃縮した。その後、得られた残渣に酢酸エチルを加えた。この懸濁液をろ取、酢酸エチルで洗浄し、減圧乾燥し、表題化合物157mg(収率5.7%) を無色油状物として得た。
2,3-ジヒドロ-[1,4]ジオキシノ[2,3-b]ピリジン(121mg, 790 μmol) をジクロロメタン (4.0mL)に溶解し、3-クロロ過安息香酸(234mg, 950μmol)を加え、室温にて1時間撹拌した。反応液を減圧濃縮し、得られた残渣をシリカゲルカラムクロマトグラフィー (クロロホルム/メタノール) で精製し、表題化合物102mg(収率84%) を白色固体として得た。
アルゴン雰囲気下、2,3-ジヒドロ-[1,4]ジオキシノ[2,3-b]ピリジン 5-酸化物 (102mg, 666μmol)をアセトニトリル(700μL)に溶解させ、室温にてトリエチルアミン (202mg, 2.00mmol)、トリメチルシリルニトリル (529mg, 5.33mmol)を加え、100℃にて終夜撹拌した。氷冷下にて水を加え、反応溶液を酢酸エチルで抽出し、飽和食塩水で洗浄し、無水硫酸ナトリウムにて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(クロロホルム/メタノール)にて精製し、表題化合物105mg(収率97%) を茶色固体として得た。
アルゴン雰囲気下、2,3-ジヒドロ-[1,4]ジオキシノ[2,3-b]ピリジン-6-カルボニトリル(16mg, 99μmol) をテトラヒドロフラン (500μL)に溶解し、氷冷下にてメチルリチウム(150μL(1.04M in THF溶液), 148μmol)を加え、同温にて5分間攪拌した。反応溶液に1M-硫酸(2.0mL)を加え、室温にて3時間撹拌した。氷冷下にて、反応液に飽和炭酸水素ナトリウム水溶液を加えた(pH=8を確認した)。反応溶液を酢酸エチルで抽出し、飽和食塩水にて洗浄し、無水硫酸ナトリウムにて乾燥し、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー (ヘキサン/酢酸エチル) で精製し、表題化合物8.3mg(収率47%) を黄色油状物として得た。
1-(2,3-ジヒドロ-[1,4]ジオキシノ[2,3-b]ピリジン-6-イル)エタノンを用いて実施例1-l)と同様に反応・処理し、表題化合物を白色結晶として得た。
(Z)-1-(4-{4-[2-(ベンジルオキシ)-1,1,1,3,3,3-ヘキサフルオロプロパン-2-イル]-2-(プロパ-1-エン-1-イル)フェニル}ピペラジン-1-イル)-2-ブロモエタノン及び5-(2,3-ジヒドロ-[1,4]ジオキシノ[2,3-b]ピリジン-6-イル)-5-メチルイミダゾリジン-2,4-ジオンを用いて実施例14-1と同様に反応・処理し、表題化合物を黄色油状物として得た。
a-1) 1-(4-(2,2-ジエトキシエトキシ)フェニル)エタノンの製造:
1-(4-ヒドロキシフェニル)エタノン(1.00g, 7.34mmol)のN,N’-ジメチルホルムアミド (24mL)溶液に、氷冷下水素化ナトリウム(純度50%) (380mg, 8.07mmol)、臭化アセトアルデヒドジエチルアセタール(1.30mL, 8.81mmol) を加え、120℃にて終夜撹拌した。氷冷下にて反応液に水を加え、ジエチルエーテルで抽出した。有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムで乾燥後、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー (ヘキサン/酢酸エチル) で精製し、表題化合物2.47g (収率>100%) を黄色油状物として得た。
1-(4-(2,2-ジエトキシエトキシ)フェニル)エタノン(2.46g, 7.34mmol)のトルエン(25mL)溶液に、PPA(2.50g)、加熱還流下にて1時間攪拌した。氷冷下にて反応液に水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムで乾燥後、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー (ヘキサン/酢酸エチル) で精製し、表題化合物44mg(収率3.7%) を白色固体として得た。
1-(ベンゾフラン-5-イル)エタノンを用いて実施例1-l)と同様に反応・処理し、表題化合物を白色結晶として得た。
実施例6において、5-[5-(1-メチルエトキシ)ピリジン-2-イル] 5-メチルイミダゾリジン-2,4-ジオンの代わりに5-(ベンゾフラン-5-イル)-5-メチルイミダゾリジン-2,4-ジオンを用いて同様に反応・処理し、表題化合物を黄色油状物として得た。
a-1) 2-(ヒドロキシメチル)-5-[(4-メトキシベンジル)オキシ]-4H-ピラン-4-オンの製造:
5-ヒドロキシ-2-ヒドロキシメチル-4H-4-ピラノン (5.0g, 35.2mmol)をN,N’-ジメチルホルムアミド(70mL)に溶解させ、アルゴン雰囲気中氷冷下にてtert-ブトキシカリウム (4.0g, 35.2mmol)を加えた。5分後、同温にて4-メトキシベンジルクロライド (6.06g, 38.7mmol)を加え、50℃にて21時間撹拌した。その後、反応液を減圧濃縮した。水、1N-水酸化ナトリウム水溶液を加えた後、酢酸エチルで抽出した。有機層を無水硫酸ナトリウムで乾燥後、減圧濃縮した。得られた残渣にジエチルエーテルを加えて再結晶にて精製し、表題化合物4.80g(収率52%) を茶色粉末状結晶として得た。
2-(ヒドロキシメチル)-5-[(4-メトキシベンジル)オキシ]-4H-ピラン-4-オン(1.2g, 4.56mmol)をエタノール(3.0mL)に溶解させ、アンモニア水溶液(9.0mL)を加え、密封し、100℃にて6時間撹拌した。この操作を4ロット分行い、まとめて引き続く処理を行った。反応液を減圧濃縮した。ジエチルエーテルを加えた後、反応溶液を氷冷した。析出した固体をろ取、ジエチルエーテルで洗浄後、減圧下乾燥し、表題化合物4.0g (収率84%) を茶色粉末状結晶として得た。
2-(ヒドロキシメチル)-5-[(4-メトキシベンジル)オキシ]ピリジン-4(1H)-オン(3.21g, 12.2mmol)をピリジン(60mL)に溶解させ、氷冷下にてアセチルクロライド(1.44g, 18.3mmol)を加え、60℃にて終夜撹拌した。その後、反応液を減圧濃縮した。氷冷下にて水を加えた後、室温にて30分間攪拌した。析出した固体をろ取、氷水、ジエチルエーテルで洗浄後、減圧下乾燥し、表題化合物3.56g (収率96%) を茶色粉末状結晶として得た。
酢酸 {5-[(4-メトキシベンジル)オキシ]-4-オキソ-1,4-ジヒドロピリジン-2-イル}メチル (3.56g, 11.7mmol)をジクロロメタン(60mL)に溶解させ、アルゴン雰囲気中氷冷下にてトリエチルアミン (3.55g, 35.1mmol)、トリフルオロメタンスルホン酸無水物 (6.60g, 23.4mmol)を加え、0℃にて15分間撹拌した。反応液に水、飽和炭酸水素ナトリウム水溶液を加えた後、酢酸エチルで抽出した。有機層を飽和食塩水にて洗浄し、無水硫酸ナトリウムで乾燥後、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)にて精製し、表題化合物4.84g(収率95%) を黄色油状物として得た。
酢酸 (5-[(4-メトキシベンジル)オキシ]-4-{[(トリフルオロメチル)スルホニル]オキシ}ピリジン-2-イル)メチル (1.2g, 2.78mmol)をアセトニトリル(9.3mL)に溶解させ、アルゴン雰囲気中氷冷下にてヨウ化銅 (53mg, 278μmol)、ジトリフェニルホスフィンパラジウム(II)ジクロライド (98mg, 139 μmol)、トリエチルアミン(8.0mL)、トリメチルシリルアセチレン(819mg, 8.34mmol)を加え、45℃にて終夜撹拌した。この操作を4ロット分行い、まとめて引き続く処理を行った。反応液に水を加えた後、セライトろ過を行った。得られたろ液を酢酸エチルで抽出し、有機層を飽和食塩水にて洗浄し、無水硫酸ナトリウムで乾燥後、減圧濃縮し、表題化合物の粗生成物 (5.38g)を黄色油状物として得た。
粗生成物の酢酸 (5-((4-メトキシベンジル)オキシ)-4-((トリメチルシリル)エチニル)ピリジン-2-イル)メチル (5.38g, 11.1mmol)をジクロロメタン (55mL)に溶解させ、室温にてトリメチルシラン (1.9mL, 35.1mmol)、トリフルオロ酢酸 (5.5mL)を加え、室温にて終夜撹拌した。反応液に氷冷下にて飽和炭酸水素ナトリウム水溶液を加え(pH=8 を確認)、反応溶液をセライトろ過した。得られたろ液を酢酸エチルで抽出し、有機層を飽和食塩水にて洗浄し、無水硫酸ナトリウムで乾燥後、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)にて精製し、表題化合物1.54g(収率53%) を黄色油状物として得た。
酢酸 (5-ヒドロキシ-4-((トリメチルシリル)エチニル)ピリジン-2-イル)メチル (1.47g, 5.59mmol)をピリジン(28mL)に溶解させ、室温にてヨウ化銅 (1.28g, 6.71mmol)を加え、加熱還流にて5時間撹拌した。反応液を室温に戻し、減圧濃縮後、反応溶液をセライトろ過した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)にて精製し、表題化合物345mg(収率32%) を橙色油状物として得た。
酢酸フロ[2,3-c]ピリジン-5-イルメチル(722mg, 3.78mmol)をメタノール(19mL)に溶解させ、室温にて炭酸カリウム(1.04g, 7.56mmol)を加え、室温にて終夜撹拌した。反応液に氷冷下にて1N-塩酸水溶液を加え、クロロホルムで抽出し、有機層を無水硫酸ナトリウムで乾燥後、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)にて精製し、表題化合物521mg(収率92%) を黄色油状物として得た。
フロ[2,3-c]ピリジン-5-イルメタノール(521mg, 3.49mmol)をアセトン(17mL)に溶解させ、室温にて2,2,6,6-テトラメチルピペリジン 1-オキシル (27mg, 170 μmol)を加えた。その後、反応液に氷冷下にて1,3,5-トリクロロ-2,4,6-トリアジントリオン(892mg, 3.84mmol)を加え、同温にて5分間撹拌した。反応溶液を減圧濃縮し、氷冷下にて水、飽和炭酸水素ナトリウム水溶液を加え、酢酸エチルで抽出し、有機層を飽和食塩水にて洗浄し、無水硫酸ナトリウムで乾燥後、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)にて精製し、表題化合物488mg(収率95%) を黄色固体として得た。
アルゴン雰囲気下、フロ[2,3-c]ピリジン-5-カルバルデヒド (488mg, 3.31mmol)をテトラヒドロフラン(11mL)に溶解させ、氷冷下にてメチルマグネシウムブロマイド (5.5mL(1.0M in THF 溶液), 4.97mmol)を加えた。その後、室温にて1時間30分攪拌した。反応液に氷冷下にて1N-塩酸水溶液を加え、その後飽和炭酸水素ナトリウム水溶液を加えた。反応溶液を酢酸エチルで抽出し、有機層を飽和食塩水にて洗浄し、無水硫酸ナトリウムで乾燥後、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)にて精製し、表題化合物356mg(収率66%) を黄色固体として得た。
1-(フロ[2,3-c]ピリジン-5-イル)エタノール (356mg, 2.19mmol)をアセトン(11mL)に溶解させ、室温にて2,2,6,6-テトラメチルピペリジン 1-オキシル(34mg, 220μmol)を加えた。その後、反応液に氷冷下にて1,3,5-トリクロロ-2,4,6-トリアジントリオン(560mg, 2.41mmol)を加え、同温にて30分間撹拌した。反応溶液を減圧濃縮し、氷冷下にて水、飽和炭酸水素ナトリウム水溶液を加え、酢酸エチルで抽出し、有機層を飽和食塩水にて洗浄し、無水硫酸ナトリウムで乾燥後、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル)にて精製し、表題化合物119mg(収率34%)を黄色固体として得た。
1-(フロ[2,3-c]ピリジン-5-イル)エタノンを用いて実施例1-l)と同様に反応・処理し、表題化合物を白色結晶として得た。
実施例6において、5-[5-(1-メチルエトキシ)ピリジン-2-イル] 5-メチルイミダゾリジン-2,4-ジオンの代わりに5-(フロ[2,3-c]ピリジン-5-イル)-5-メチルイミダゾリジン-2,4-ジオンを用いて同様に反応・処理し、表題化合物を黄色油状物として得た。
実施例43-a)で得た1,1,1,3,3,3-ヘキサフルオロ-2-(4-フルオロ-3-ニトロフェニル)プロパン-2-オールとピペラジン-1-カルボン酸tert-ブチルの代わりに実施例14-1-a-2-2)で得た2,5-ジメチルピペラジン-1-カルボン酸 (2S, 5R)-tert-ブチルを用い、その他は実施例43-b)と同様に反応・処理し、表題化合物を黄色油状物として得た。
(2S,5R)-4-[4-(1,1,1,3,3,3-ヘキサフルオロ-2-ヒドロキシプロパン-2-イル)-2-ニトロフェニル]-2,5-ジメチルピペラジン-1-カルボン酸 tert-ブチルエステルを用いて、実施例43-c)と同様に反応・処理し、表題化合物を黄色油状物として得た。
(2S,5R)-4-{4-[2-(ベンジルオキシ)-1,1,1,3,3,3-ヘキサフルオロプロパン-2-イル]-2-ニトロフェニル}-2,5-ジメチルピペラジン-1-カルボン酸 tert-ブチルエステルを用いて、実施例43-d)と同様に反応・処理し、表題化合物を黄色油状物として得た。
(2S,5R)-4-{2-アミノ-4-[2-(ベンジルオキシ)-1,1,1,3,3,3-ヘキサフルオロプロパン-2-イル]フェニル}-2,5-ジメチルピペラジン-1-カルボン酸 tert-ブチルエステルを用いて、実施例43-e)と同様に反応・処理し、表題化合物を黄色油状物として得た。
(2S,5R)-4-{4-[2-(ベンジルオキシ)-1,1,1,3,3,3-ヘキサフルオロプロパン-2-イル]-2-ヨードフェニル}-2,5-ジメチルピペラジン-1-カルボン酸 tert-ブチルエステルを用いて、実施例43-f)と同様に反応・処理し、表題化合物を黄色油状物として得た。
(2S,5R)-4-{4-[2-(ベンジルオキシ)-1,1,1,3,3,3-ヘキサフルオロプロパン-2-イル]-2-(プロパ-1-エン-1-イル)フェニル}-2,5-ジメチルピペラジン-1-カルボン酸 tert-ブチルエステルを用いて、実施例43-g)と同様に反応・処理し、表題化合物を黄色油状物として得た。
(2S,5R)-1-{4-[2-(ベンジルオキシ)-1,1,1,3,3,3-ヘキサフルオロプロパン-2-イル]-2-(プロパ-1-エン-1-イル)フェニル}-2,5-ジメチルピペラジンを用いて、実施例43-h)と同様に反応・処理し、表題化合物を黄色油状物として得た。
1-((2S,5R)-4-{4-[2-(ベンジルオキシ)-1,1,1,3,3,3-ヘキサフルオロプロパン-2-イル]-2-(プロパ-1-エン-1-イル)フェニル}-2,5-ジメチルピペラジン-1-イル)-2-ブロモエタノンと5-(2,3-ジヒドロベンゾ[b][1,4]ジオキシン-6-イル)-5-メチルイミダゾリジン-2,4-ジオンを用いて実施例14-1と同様に反応・処理し、表題化合物を黄色油状物として得た。
<プラスミドの構築>
ヒトLXRα及びLXRβ cDNAのリガンド結合ドメイン(LBD)を哺乳類発現ベクターpBIND(Promega)の酵母GAL4転写因子DNA結合ドメイン(DBD)に隣接して挿入することによって発現構築物を調製し、それぞれpBIND-LXRα/GAL4及びpBIND-LXRβ/GAL4を作製した。GAL4応答性リポーター構築物、pG5lucは、Promega社から入手できる公知のベクターであり、プロモーターに隣接して位置する5コピーのGAL4応答エレメントとルシフェラーゼリポーター遺伝子を含む。
LXRα/GAL4又はLXRβ/GAL4ハイブリッド及びGAL4応答性リポーターベクターpG5luc安定発現CHOK-1細胞を、5%CO2の湿潤雰囲気下、37℃で、10%非働化処理ウシ胎児血清、100単位/mlペニシリンG及び100μg/ml硫酸ストレプトマイシンを含有するHAM-F12培地を入れた96ウェルプレートに20,000細胞/ウェルで播種した。24時間後、被検化合物を試験濃度範囲(0.01μM、0.1μM、1μM、10μM)にわたって溶解した培地を添加し、細胞とともに24時間インキュベーションした。ルシフェラーゼアッセイ基質としてBright-Glo(Promega)を用い、発光強度をルミノメーターLB960(Berthold Technologies)で測定することにより、試験化合物がLXRα又はβのLBDを介してルシフェラーゼ転写の活性化に及ぼす作用を測定した。比較化合物としてT0901317(国際公開WO2000/54759パンフレットの実施例12の化合物)を同時に評価した。ルシフェラーゼ活性の結果はT0901317の10μMにおける発光強度を100とした時の試験化合物の各濃度における活性値(%eff)として表1に示した。なお、ジメチルピペラジン構造を有する化合物の活性データは、(2R,5S)体と(2S,5R)体との混合物を用いた結果である。
表1に示す通り、本発明のカルビノール化合物は対照薬であるT0901317よりもLXRβに高い選択性を有するLXRアゴニストであることが実験的に確認された。
Claims (7)
- 次の一般式(I)
〔式中、V、Wは、それぞれ独立して、N又はC-R7を示し、
X、Yは、それぞれ独立して、CH2、CH(C1-8アルキル)、C(C1-8アルキル)2、C=O又はSO2を示し、
Zは、CH又はNを示し、
R1、R2、R7は、それぞれ独立して、水素原子、ハロゲン原子、置換基を有してもよいC1-8アルキル基又はC2-8アルケニル基を示し、
R3は、C1-8アルキル基を示し、
R4は、置換基を有してもよいC6-10アリール基又は置換基を有してもよい5-11員ヘテロ環基を示し、
R5は、水素原子又はC1-8アルキル基を示し、
R6は水素原子、C1-8アルコキシC1-8アルキル基又は置換基を有してもよいC6-10アリールC1-8アルキル基を示し、
Lはオキソ基又はスルホニル基で置換してもよいC1-8アルキル鎖を示し、
nは0~2のいずれかの整数を示す。〕
で表されるカルビノール化合物、若しくはその塩又はそれらの溶媒和物。 - 請求項1に記載のカルビノール化合物、若しくはその塩又はそれらの溶媒和物を有効成分とする医薬。
- アテローム性動脈硬化症、糖尿病に起因する動脈硬化症、脂質異常症、高コレステロール血症、脂質関連疾患、炎症性サイトカインにより引き起こされる疾患である炎症性疾患、皮膚疾患、糖尿病又はアルツハイマー病の予防及び/又は治療剤である請求項2記載の医薬。
- 請求項1に記載のカルビノール化合物、若しくはその塩又はそれらの溶媒和物を有効成分とするLXR調節剤。
- 請求項1に記載のカルビノール化合物、若しくはその塩又はそれらの溶媒和物、及び製薬上許容される担体からなる医薬組成物。
- 治療を必要としている患者に、請求項1に記載のカルビノール化合物、若しくはその塩又はそれらの溶媒和物の有効量を投与することを特徴とする、アテローム性動脈硬化症、糖尿病に起因する動脈硬化症、脂質異常症、高コレステロール血症、脂質関連疾患、炎症性サイトカインにより引き起こされる疾患である炎症性疾患、皮膚疾患、糖尿病又はアルツハイマー病の予防及び/又は治療方法。
- アテローム性動脈硬化症、糖尿病に起因する動脈硬化症、脂質異常症、高コレステロール血症、脂質関連疾患、炎症性サイトカインにより引き起こされる疾患である炎症性疾患、皮膚疾患、糖尿病又はアルツハイマー病の予防及び/又は治療のための製剤を製造するための、請求項1に記載のカルビノール化合物、若しくはその塩又はそれらの溶媒和物の使用。
