Abstract
Description
エクオール産生微生物を用い、大豆粉末又は豆乳を原料として発酵物を製造する際に、(1)エクオール産生微生物を用いて、ダイゼイン類の存在下、pH5.0以上、嫌気条件下で発酵することにより、マザースターターを調製する、(2)前記マザースターターを用いて、ダイゼイン類の存在下、pH5.0以上で発酵することにより、バルクスターターを調製する、(3)前記バルクスターターを用いて、大豆粉末又は豆乳を含む培地で発酵して発酵物を調製することによって、エクオール産生能が維持された微生物を生菌のままで含む発酵物の製造が可能になる。
エクオール産生微生物を用い、乳を原料として発酵物を製造する際に、マザースターターをダイゼイン類の存在下で嫌気条件下で発酵させることにより調製し、更にマザースターターを用いて乳原料を発酵させる際に、ダイゼイン類の存在下、pH4.6以上となる条件下で行うことによって、エクオール産生能が維持された微生物を生菌のままで含む発酵物の製造が可能になる。また、当該発酵物は、エクオール産生微生物のエクオール産生能が長期間に亘って保持され、保存安定性にも優れている。更に、マザースターターの調製から乳原料の発酵に亘って行われる全て発酵を、酵母エキスの存在下で実施することにより、エクオール産生微生物のエクオール産生能の維持、並びに保存安定性を一層向上させることができる。
エクオール産生微生物を生菌の状態で含む発酵製品に、アスコルビン酸及び/又はその誘導体を添加することによって、保存後でもエクオール産生微生物のエクオール産生能が喪失されることがない組成物の提供が可能になる。
項1. 以下の工程を含む、発酵物の製造方法:
(1)ダイゼイン配糖体、ダイゼイン、及びジヒドロダイゼインよりなる群から選択される少なくとも1種のダイゼイン類を含有する培地で、エクオール産生微生物を用いて、pH5.0以上を維持した状態で嫌気発酵を行うことにより、マザースターターを調製する工程、
(2)前記工程(1)で得られたマザースターターを用いて、前記ダイゼイン類を含有する培地で、pH5.0以上を維持した状態で発酵を行うことにより、バルクスターターを調製する工程、及び
(3)前記工程(2)で得られたバルクターターを用いて、大豆粉末及び/又は豆乳を含む培地で発酵を行うことにより、発酵物を得る工程。
項2. エクオール産生微生物が乳酸菌である、項1に記載の製造方法。
項3. エクオール産生微生物がラクトコッカス・ガルビエであるである、項1に記載の製造方法。
項4. 以下の工程を含む、発酵物の製造方法:
(I)ダイゼイン配糖体、ダイゼイン、及びジヒドロダイゼインよりなる群から選択される少なくとも1種のダイゼイン類を含有する培地で、エクオール産生微生物を用いて嫌気発酵することにより、マザースターターを調製する工程、及び
(II)前記工程(I)で得られたマザースターターを用いて、前記ダイゼイン類及び乳を含む培地で、pH4.6以上を維持した状態で発酵を行うことにより、発酵物を得る工程。
項5. ダイゼイン類として大豆胚軸抽出物が使用される、項4に記載の製造方法。
項6. 前記工程(I)及び(II)で使用される培地が、更に酵母エキスを含む、項4に記載の製造方法。

エクオール産生能が維持されたエクオール産生微生物を生菌の状態で含む飲食品を提供できれば、エクオールを産生できないヒト(エクオール非産生者)に対して、エクオール産生能を獲得させることができるので、本発明によってエクオール非生産者に対してエクオールに基づく有効生理活性を享受させることが可能になる。
本第1法の発酵物の製造方法は、マザースターター(以下、「マザースターター-1」と表記することもある)を調製する工程(1)、マザースターター-1を用いてバルクスターター(以下、「バルクスターター-1」と表記することもある)を調製する工程(2)、及びバルクスターター-1を用いて、大豆粉末又は豆乳を原料として発酵物を調製する工程(3)を含む。以下、本第1法の発酵物の製造方法について工程毎に詳述する。
本発明では、まず、エクオール産生微生物を、ダイゼイン類を含む培地(以下、「マザースターター培地-1」と表記することもある)で、pH5.0以上を維持した状態で嫌気発酵することにより、マザースターター(以下、「マザースターター-1」と表記することもある)を調製する(工程(1))。
次いで、前記工程(1)で得られたマザースターター-1を用いて、前記ダイゼイン類を含有する培地で(以下、「バルクスターター培地-1」と表記することもある)、pH5.0以上を維持した状態で発酵を行うことにより、バルクスターター-1(以下、「バルクスターター-1」と表記することもある)を調製する(工程(2))。
次いで、前記工程(2)で得られたバルクターター-1を用いて、大豆粉末及び/又は豆乳を含む培地を含む培地(以下、「本発酵用培地-1」と表記することもある)で発酵を行うことにより、発酵物を調製する(工程(3))。
本第2法の発酵物の製造方法は、マザースターター(以下、「マザースターター-2」と表記することもある)を調製する工程(I)と、マザースターター-2を用いて乳を原料として発酵を行う工程(II)を含む。以下、本発明の製造方法について工程毎に詳述する。
本発明では、まず、エクオール産生微生物を、ダイゼイン類を含む培地(以下、「マザースターター培地-2」と表記する)で嫌気発酵することにより、マザースターターを調製する(工程(I))。
次いで、前記工程(I)で得られたマザースターター-2を用いて、前記ダイゼイン類を含む培地(以下、「本発酵用培地-2」と表記する)を、pH4.6以上を維持した状態で発酵(以下、「本発酵」と表記することもある)させる(工程(II))。
更に、本発明は、上記第1法及び第2法で得られた発酵物を含む発酵製品を提供する。即ち、上記第1法及び第2法で得られた発酵物は、そのまま、或いは必要に応じて、ショ糖、スクラロース、ステビア等の甘味料;コーヒーエキス、紅茶エキス等のフレーバー剤;香料;大豆粉末、大豆片、果汁、果実片、野菜汁、野菜片等の植物由来成分;ナトリウム、カリウム、カルシウム、亜鉛、鉄等の金属;アスコルビン酸等のビタミン;ゲル化剤;グルコン酸等の安定化剤;水等の希釈剤等の添加成分を配合して発酵製品として提供される。とりわけ、上記第1法で得られた発酵物は、大豆粉末を含む水溶液と混合された発酵製品として好適に提供される。
エクオール産生微生物を生菌の状態で含む組成物において、アスコルビン酸、その誘導体、及びそれらの塩よりなる群から選択される少なくとも1種を添加することによって、低pH条件や好気保存条件下においてもエクオール産生微生物のエクオール産生能を安定に保持させることが可能になり、長期間保存しても該組成物においてエクオール産生微生物のエクオール産生能が喪失されるのを抑制することができる。そこで、本発明は、更に、生菌の状態のエクオール産生微生物(以下、単に「(A)成分」と表記することもある)、並びにアルコルビン酸、その誘導体、及びそれらの塩よりなる群から選択される少なくとも1種(以下、単に「(B)成分と表記することもある」を含む、エクオール産生微生物含有組成物(以下、単に「本組成物」と表記することもある)を提供する。
皮を剥いで半割に分割して脱皮大豆を蒸煮処理に供した。蒸煮処理は、脱皮大豆に対して、100℃の温度に達するまで蒸気を送り続け、100℃に到達した時点から140秒間100℃に保った。次に、蒸煮処理後の大豆をローラーミルのローラ間に通して、フレーク状にした。その後、フレーク状の大豆を80℃で熱風乾燥を行い、水分含量を3~6%程度になるまで乾燥させ、これをエアーグラインダーで粉砕することにより、大豆粉末を得た。エアーグラインダーでの粉砕は、粉砕粒子の径150μm以上が10%以下になるように行った。
1.発酵大豆食品の製造
エクオール産生能を有するラクトコッカス・ガルビエ(ラクトコッカス20-92株、FERM BP-10036号)を用いて、以下の条件で、マザースターターの調製、バルクスターターの調製、本発酵、及び容器への充填を行った。
大豆粉末溶液(乾燥重量で14%の大豆粉末を含む)80重量%、グルコース0.1重量%、L-アルギニン0.1重量%、精製水 残部を含む溶液(pH 7.48)100mlをオートクレーブ殺菌(121℃、15分間)し、マザースターター培地を調製した。このスターター培地に、ラクトコッカス20-92株の種菌を接種してGas Packにて37℃で96時間、嫌気培養を行うことにより、マザースターター(pH6.64)を得た。
大豆粉末溶液(乾燥重量で14%の大豆粉末を含む)80重量%、グルコース0.1重量%、L-アルギニン0.1重量%、精製水 残部を含む溶液(pH 7.48)5Lをオートクレーブ殺菌(121℃、15分間)し、バルクスターター培地を調製した。このスターター培地に、上記で得られたマザースターターを1容量%となるように接種して、37℃で15時間、好気条件下で静置培養を行うことにより、バルクスターター(pH6.72)を得た。
大豆粉末溶液(乾燥重量で14%の大豆粉末を含む)50重量%、ニンジン汁(宮崎農協果汁)6重量%、かぼちゃペースト(長野サンヨーフーズ)6重量%、精製水 残部を含む溶液(pH 6.78)200Lを二重管式殺菌機にて95℃、30秒間殺菌し、本発酵用培地を調製した。殺菌後回収した本発酵用培地に、上記で得られたバルクスターター4Lを接種して、37℃で15時間、好気条件下で静置培養を行うことにより発酵大豆液(pH4.56)を得た。
大豆粉末溶液(乾燥重量で14%の大豆粉末を含む)65重量%、砂糖7.4重量%、ゲル化剤製剤FG2524(新田ゼラチン製)1.0重量%、グルコン酸0.8重量%、香料0.4重量%、L-アスコルビン酸0.1重量%、精製水 残部を含む溶液800Lを二重管式殺菌機にて95℃、30秒間殺菌し、副原料溶液を得た。この副原料溶液80重量部と上記で得られた発酵大豆液20重量部を37~40℃で混合した。混合液の温度を維持したまま衛生的にポリエチレンカップ(満注容量130ml、口径71mm)に100g充填し、アルミ蓋でシールした。充填物は5℃の冷蔵庫に入れ、冷却固化させ、ラクトコッカス・ガルビエ生菌を含む発酵大豆食品を得た。
上記で得られた容器入りの発酵大豆食品について10℃で3週間保存した。調製直後、1週間後、2週間後、及び3週間後に、発酵大豆食品のpHを測定し、更に発酵大豆食品に含まれるラクトコッカス・ガルビエの菌数、及び発酵大豆食品に含まれるラクトコッカス・ガルビエのエクオール産生能の有無を以下の方法で評価した。また、上記で調製したマザースターター、バルクスターター及び本発酵後の発酵物についても、ラクトコッカス・ガルビエの菌数、及びそれぞれに含まれるラクトコッカス・ガルビエのエクオール産生能について同様に測定した。
ラクトコッカス・ガルビエの生菌数の測定は、BCP加プレートカウント寒天培地(日水製薬株式会社製)を用いて37℃、72時間混釈培養を行い、生育したコロニー数を計測することにより求めた。
ダイゼインを10μg/mlの濃度で含む変法GAMブイヨン(日水製薬株式会社製)5mlに、発酵大豆食品を200μl添加して37℃で96時間、ガスパックにて嫌気培養を行った。次いで、得られた培養液をHPLCに供して、培養液中のエクオール量を定量した。HPLCによる分析は次の様にして実施した。先ず、培養液0.5 mlを酢酸エチル5.0 mlに加えて浸透抽出を行い、得られた抽出液を3000rpmにて10分間遠心し、上清を遠心エバポレーターにより溶媒を減圧乾固した。次いで得られた固形物を、1.0ml溶媒(移動相A/移動相B=50/50)にて再溶解してHPLCの検体に供した。HPLCの分析は、D7000シリーズ(日立製作所製)を使用し、カラムはCapcell Pack UGL205μm4.6Φ×250mm(資生堂製)を使用した。移動相は、移動相Aとして0.05%リン酸緩衝液(EDTA含有)と酢酸エチル-メタノール(1:10)を8:2で混合した溶液を使用し、移動相Bとして2%酢酸エチル含有メタノールした。グラジエント法で流速を1.0ml/分とした。検出はSPD-10AVP UV-VIS検出器を用い、検出波長は254nmと280nmとした。
結果を表1に示す。この結果から、マザースターター、バルクスターターのpHが5以上であれば、本発酵後の発酵物、発酵大豆食品のpHが4.6以下であってもエクオール産生能を保持したラクトコッカス・ガルビエを含む発酵大豆食品が製造できることが明らかとなった。なお、発酵大豆食品におけるラクトコッカス・ガルビエの菌数自体は、pHやエクオール産生能の有無にはさほど影響されず、長期間一定に保持されることも確認された。
調製直後の発酵大豆食品及び3週間保存後の発酵大豆食品も大豆の風味よく異味異臭もなく、食品として好適であった。
マザースターターの調製において、グルコース添加量を0.1重量%から1.0重量%に変更すること以外は実施例1と同条件で、マザースターターの調製、バルクスターターの調製、及び本発酵を行い、発酵大豆食品(比較例1-1)を製造した。
1.発酵乳の製造
エクオール産生能を有するラクトコッカス・ガルビエ(ラクトコッカス20-92株、FERM BP-10036号)を用いて、以下の条件で、マザースターターの調製、バルクスターターの調製、本発酵、及び容器への充填を行った。
脱脂粉乳10重量%、生クリーム6.67重量%、酵母エキス(「SK酵母エキスHi-K」、日本製紙ケミカル株式会社)0.1重量%、大豆胚軸抽出物(「ソヤフラボンHG」、不二製油株式会社;大豆胚軸抽出物中のダイゼイン類の含有割合は約37重量%)0.025重量%、精製水 残部を含む溶液(pH 6.14)500mLをオートクレーブ殺菌(115℃、15分間)し、マザースターター培地を調製した。このスターター培地に、ラクトコッカス20-92株の種菌を接種してGas Packにて37℃で24時間、嫌気培養を行うことにより、マザースターターを得た。
脱脂粉乳10重量%、生クリーム6.67重量%、酵母エキス(「SK酵母エキスHi-K」、日本製紙ケミカル株式会社)0.1重量%、大豆胚軸抽出物(「ソヤフラボンHG」、不二製油株式会社)0.025重量%、精製水 残部を含む溶液10LをUHT(Ultra High Temperature)殺菌(140℃、4秒分間)し、バルクスターター培地を調製した。UHT殺菌後に回収したバルクスターター培地に、上記で得られたマザースターターを1v/v%となるように接種して、37℃で24時間、好気培養を行うことにより、バルクスターターを得た。
脱脂粉乳10重量%、生クリーム6.67重量%、酵母エキス(「SK酵母エキスHi-K」、日本製紙ケミカル株式会社)0.1重量%、大豆胚軸抽出物(「ソヤフラボンHG」、不二製油株式会社)0.025重量%、精製水 残部を含む溶液10LをUHT(Ultra High Temperature)殺菌(140℃、4秒分間)し、本発酵用培地を調製した。UHT殺菌後に回収した本発酵用培地に、上記で得られたバルクターターを1v/v%となるように接種して、37℃で24時間、好気培養を行うことにより、発酵乳を得た。
上記本発酵により得られた発酵乳を、滅菌済みの600mL容の容器に無菌的に入れ、更に所定量の乳酸を容器内に無菌的に添加して混合した後に、容器を密閉した。
上記で得られた容器入り発酵乳のそれぞれについて10℃で74週間保存した。製造直後、2週間後、6週間後、及び74週間後に、発酵乳のpHを測定し、更に発酵乳に含まれるラクトコッカス・ガルビエの菌数、及び発酵乳に含まれるラクトコッカス・ガルビエのエクオール産生能の有無を、実施例1と同様の方法で評価した。また、上記で調製したマザースターター及びバルクスターターについても、ラクトコッカス・ガルビエの菌数、及び発酵乳に含まれるラクトコッカス・ガルビエのエクオール産生能についても、同様に測定した。
製造直後の発酵乳(乳酸添加なし)及び74週間保存後の発酵乳(乳酸添加なし)はいずれも、酸味が少なくしかも飲み易く、食品として好適であった。
マザースターター培地、バルクスターター培地、及び本発酵用培地として、下記組成をベースとする培地に、表4に示す所定量の発酵促進剤を添加して調製したものを用いること以外は、上記実施例2と同条件で、マザースターターの調製、バルクスターターの調製、及び本発酵を行った。
マザースターターの調製において、好気培養すること以外は、実施例2と同条件で、マザースターターの調製、バルクスターターの調製、及び本発酵を行った。
1.エクオール産生微生物含有組成物の製造
変法GAM培地(日水製薬株式会社)5mLに、ラクトコッカス・ガルビエ(ラクトコッカス20-92株、FERM BP-10036号)の種菌を接種してGas Packにて37℃で24時間、嫌気培養を行った。次いで、得られた培養物2mLを変法GAM培地(日水製薬株式会社)50mLに接種してGas Packにて37℃で24時間、嫌気培養を行った。さらに得られた培養物8mLを変法GAM培地(日水製薬株式会社)200mLに接種してGas Packにて37℃で24時間、嫌気培養を行った。
上記で得られた各エクオール産生微生物含有組成物を、10℃で21日間保存を行った。保存期間中に、各エクオール産生微生物含有組成物について、pH、菌数及びエクオール産生能の評価を行った。菌数の測定方法は、上記実施例1と同様である。また、エクオール産生能は、上記実施例1と同様の方法でHPLCによる分析を行い、下記の式に従ってエクオール変換率を算出した。
1.エクオール産生微生物含有組成物の製造
実施例1で使用したマザースターター培地と同組成の培地5mlに、ラクトコッカス20-92株の種菌を接種してGas Packにて37℃で96時間、嫌気培養を行うことにより、マザースターターを得た。
エクオール産生微生物の菌数を測定した結果を表9に示し、エクオール産生能を測定した結果を表10に示す。保存開始時には、いずれの酸化防止剤を配合した場合であっても、エクオール産生能が認められた。しかしながら、保存7日後においてはアスコルビン酸を添加した発酵製品以外はすべてエクオール産生能が消失した。
Claims (19)
- 以下の工程を含む、発酵物の製造方法:
(1)ダイゼイン配糖体、ダイゼイン、及びジヒドロダイゼインよりなる群から選択される少なくとも1種のダイゼイン類を含有する培地で、エクオール産生微生物を用いて、pH5.0以上を維持した状態で嫌気発酵を行うことにより、マザースターターを調製する工程、
(2)前記工程(1)で得られたマザースターターを用いて、前記ダイゼイン類を含有する培地で、pH5.0以上を維持した状態で発酵を行うことにより、バルクスターターを調製する工程、及び
(3)前記工程(2)で得られたバルクターターを用いて、大豆粉末及び/又は豆乳を含む培地で発酵を行うことにより、発酵物を得る工程。 - エクオール産生微生物が乳酸菌である、請求項1に記載の製造方法。
- エクオール産生微生物がラクトコッカス・ガルビエであるである、請求項1に記載の製造方法。
- 以下の工程を含む、発酵物の製造方法:
(I)ダイゼイン配糖体、ダイゼイン、及びジヒドロダイゼインよりなる群から選択される少なくとも1種のダイゼイン類を含有する培地で、エクオール産生微生物を用いて嫌気発酵することにより、マザースターターを調製する工程、及び
(II)前記工程(I)で得られたマザースターターを用いて、前記ダイゼイン類及び乳を含む培地で、pH4.6以上を維持した状態で発酵を行うことにより、発酵物を得る工程。 - ダイゼイン類として大豆胚軸抽出物が使用される、請求項4に記載の製造方法。
- 前記工程(I)及び(II)で使用される培地が、更に酵母エキスを含む、請求項4に記載の製造方法。
- エクオール産生微生物が乳酸菌である、請求項4に記載の製造方法。
- エクオール産生微生物がラクトコッカス・ガルビエであるである、請求項4に記載の製造方法。
- 請求項1乃至3のいずれかに記載の製造方法により得られる発酵物を含有する、発酵製品。
- 請求項4乃至8のいずれかに記載の製造方法により得られる発酵物を含有する、発酵製品。
- pHが4.6以上である、請求項10に記載の発酵製品。
- (A)生菌の状態のエクオール産生微生物、並びに(B)アスコルビン酸、その誘導体、及びそれらの塩よりなる群から選択される少なくとも1種を含む、エクオール産生微生物含有組成物。
- 上記(B)成分が0.05~5重量%の配合割合で含まれる、請求項12に記載の組成物。
- pHが5.0以下である、請求項12に記載の組成物。
- 生菌の状態のエクオール産生微生物として、エクオール産生微生物を用いて発酵した発酵物を含む、請求項12に記載の組成物。
- 発酵大豆飲料又は発酵豆乳である、請求項12に記載の組成物。
- エクオール産生微生物が乳酸菌である、請求項12に記載の組成物。
- エクオール産生微生物がラクトコッカス・ガルビエであるである、請求項12に記載の組成物。
- 生菌の状態のエクオール産生微生物を含む組成物に、アスコルビン酸、その誘導体、及びそれらの塩よりなる群から選択される少なくとも1種を添加することを特徴とする、エクオール産生微生物のエクオール産性能の維持方法。
