Description
本発明は飛行時間型質量分析装置に関する。
一般に飛行時間型質量分析装置(TOFMS)では、一定のエネルギーで以て加速したイオンが質量に応じた飛行速度を持つことに基づき、一定距離を飛行するのに要する時間を計測することで、その飛行時間からイオンの質量(厳密にはm/z値)を算出する。したがって、質量分解能を向上させるためには、飛行距離を伸ばすことが特に有効である。しかしながら、直線的に飛行距離を伸ばそうとすると装置が大形化することが避けられず実用的でない。
そこで、飛行距離を伸ばすために、多重周回飛行時間型質量分析装置と呼ばれる質量分析装置が開発されている(例えば特許文献1、2など参照)。多重周回飛行時間型質量分析装置では、2乃至4個(又はそれ以上の個数)の扇形電場を用いて8の字状や略円状などの形状の閉じた周回軌道を形成し、この周回軌道に沿ってイオンを多数回繰り返し周回させることで飛行距離を実効的に長くしている。こうした構成によれば、飛行距離は装置サイズの制約を受けず、周回数を増やすことで質量分解能を向上させることができる。
また、こうした多重周回飛行時間型質量分析装置では、リフレクトロン型TOFMSと同様に、扇形電場を形成する電極の曲率、形状などを工夫することにより、大きなエネルギーを持つイオンほど中心軌道よりも外側の、つまり距離の長い軌道を飛行させることで、イオンが持つエネルギーの拡がり(ばらつき)による飛行時間の拡がりを抑制することができる。これにより、イオンが加速される際の初期エネルギーのばらつきの影響を軽減し、質量分解能を一層向上させることができる。
しかしながら、従来の飛行時間型質量分析装置はイオンをm/z値に応じて分離するものであるため、例えば2価の窒素分子イオン(14N2
2+)と1価の窒素原子イオン(14N+)とを分離することはできない。
本発明は上記課題に鑑みて成されたものであり、その目的とするところは、高い質量分解能でもって異種のイオンを分離し、且つ、m/z値では分離できない異種のイオンも分離することで、従来よりも、子細な情報を収集することができる飛行時間型質量分析装置を提供することである。
上記課題を解決するために成された本発明は、イオンに所定の運動エネルギーを付与して飛行空間を飛行させることにより質量分析を行う飛行時間型質量分析装置において、
a)イオンの飛行経路の少なくとも一部に所定のガスを導入するガス導入手段と、
b)同一試料に対し、前記ガス導入手段によりガスが導入されない状態での質量分析とガスが導入された状態での質量分析とをそれぞれ実行し、各分析によりそれぞれ飛行時間スペクトルを取得する分析実行制御手段と、
c)前記分析実行制御手段の制御の下で得られた2つの飛行時間スペクトルに出現しているピークの位置、形状又は強度の少なくとも1つを比較することで、同一m/z値を持つ異種のイオンを識別するイオン識別手段と、
を備えることを特徴としている。

本発明に係る飛行時間型質量分析装置において、ガス導入手段によりガスが導入された領域をイオンが通過すると、イオンは所定の確率でガスと衝突し、持っている運動エネルギーの一部が失われて飛行速度が低下する。一般に、イオンとガスとの衝突確率はイオンの大きさに依存し、大きなサイズのイオンほどガスと衝突する回数が多くなり、運動エネルギーの損失は大きい。したがって、仮に或る異種のイオンのm/z値が同一であっても、その異種のイオンに、大きさや構造(形状)、分子クラス(脂質やペプチドなどの分子の分類)、価数などの相違があれば飛行時間に差に生じる。
そこで、分析実行制御手段は、同一試料に対し、所定のガスが導入されない状態(一般に高真空雰囲気中)での質量分析と、所定のガスが導入された状態での質量分析とをそれぞれ実行し、飛行時間スペクトルを取得する。高真空雰囲気中での質量分析により得られた飛行時間スペクトルでは、異種イオンであっても同一m/z値を持てば1つのピークとして現れる。これに対し、ガスが導入された状態での質量分析により得られた飛行時間スペクトルでは、同一m/z値を持つものであっても異種のイオンであれば、即ち、イオンの大きさや構造などが相違すれば、飛行時間に差が生じるためにピークが2つに分離するか、明確なピーク分離まで至らなくてもピーク形状が変形するか、或いは、ピーク強度が変化する。イオン識別手段は2つの飛行時間スペクトル上の対応するピークの位置、形状又は強度を比較することで、同一m/z値を持つ異種のイオンが存在するか否かを判定する。
例えば、ガスが導入された状態における飛行時間スペクトル上で2つのピークに分離する場合、飛行時間が長いほうが大きなイオンであると判断することができる。また、ガスとの衝突によりイオンの損失がなければ(或いは無視できる程度であれば)、ガス導入時の飛行時間スペクトルに現れるピークはそれぞれのイオンの強度を表すから定量も可能である。
本発明に係る飛行時間型質量分析装置の好ましい一態様は、同一の飛行軌道を繰り返し飛行させる多重周回飛行時間型の質量分析装置である。
多重周回飛行時間型質量分析装置では、イオンはガスが導入された同一領域を繰り返し通過するため、飛行経路に導入されるガスの量が比較的少なく1回のイオンの通過では十分な飛行時間差が生じない場合でも、最終的に大きな飛行時間差を生じさせることができる。これにより、飛行時間スペクトル上でのピークの位置やピーク形状の変化などが顕著に現れ、異種のイオンが存在しているか否かの判断が容易に行え、その正確性も向上する。
一般に、運動エネルギーを持って飛行しているイオンがガスに衝突すると、衝突誘起解離によってイオンの開裂が生じ易い。そこで、本発明に係る飛行時間型質量分析装置では、できるだけ開裂が生じにくい条件の下でガスを導入した状態の質量分析を実行することが望ましい。そのために有効な方法の1つは、所定のガスとしてできるだけ軽いガスを用いることである。好適な一例として、最も軽い不活性ガスであるヘリウムを用いるとよい。
こうした軽いガスを用いるとイオンの開裂が生じにくいのみならず、ガスとの衝突の際にもイオンが飛行軌道を外れにくく、飛行途中でのイオンの消失を抑えるのにも有効である。もちろん、イオンの開裂を生じにくくするために、導入するガスの量を少なくする(つまりガス圧を低い状態とする)ようにしてもよい。但し、上述したようにガスの量が少ないとイオンの大きさに応じて飛行時間に差を生じさせる効果も小さくなるため、多重周回飛行時間型の構成にすることが望ましい。
さらにまた、イオンを飛行空間へ導入する際にイオンに与える初期的な運動エネルギーを小さくしても、衝突誘起解離によるイオンの開裂を回避するのに有効である。但し、初期運動エネルギーを小さくし過ぎると、途中で運動エネルギーを徐々に失ったイオンが検出器まで到達し得なくなるから、飛行経路の長さ(例えば多重周回飛行時間型質量分析装置の場合には周回数)とガス圧などに基づいて、或る程度以上の初期的な運動エネルギーをイオンに与えることが必要である。
本発明に係る飛行時間型質量分析装置によれば、通常の質量分析により高い質量分解能で試料中の成分由来のイオンのm/z値を測定することができるとともに、m/z値が同一(又はほぼ同一)でイオンの大きさ、構造、分子クラスなどが相違する異種のイオンが存在している場合に、少なくともそうした存在に関する情報を提供することができる。また、本発明に係る飛行時間型質量分析装置によれば、m/z値の相違に応じたイオンの分離・検出と、イオンの大きさ、構造、分子クラスなどに応じた同一のm/z値を持つイオンの分離・検出が同一の装置で且つ簡単な操作でもって行えるので、イオンの分子構造の解明などに有用な情報を効率よく収集することが可能となる。

図示しない真空ポンプにより真空排気される真空室6内には、イオン源1、周回飛行室4、検出器5が配設され、周回飛行室4の内部には、周回軌道2を形成するための複数の扇形電極対3が配置されている。周回飛行室4には、バルブ8の開放時にガス源7から所定圧で所定のガスが供給される。イオン源1、バルブ8、及び、扇形電極対3に所定の電圧を印加する電圧印加部9は制御部10により制御される。検出器5による検出信号はA/D変換器11で所定のサンプリング時間間隔でデジタルデータに変換され、そのデータはデータ処理部12で処理される。データ処理部12は本実施例に特徴的な機能ブロックとしてスペクトル記憶部13とスペクトル比較部14とを含み、処理結果は出力部15より出力される。ガス源7に用意される所定のガスとしては、後述する理由により、軽い不活性ガスが好ましく、ここではヘリウムガスを用いるものとする。
イオン源1では試料分子がイオン化され、生成された各種イオンは所定の初期エネルギーを付与されて飛行を開始する。なお、イオン源1は例えば3次元四重極型イオントラップなどのように、外部で生成された各種イオンを一時的に保持し、それらイオンに所定のタイミングで一斉にエネルギーを付与して飛行を開始させるものでもよい。
イオン源1を出発点として飛行を開始したイオンは、周回飛行室4に入って複数の扇形電極対3の間にそれぞれ形成される複数の扇形電場の作用により形成される周回軌道2に乗せられる。周回軌道2の形状は図1に記載のものに限らず、略楕円形状、8の字形状など、様々な形状が実現可能である。イオンは周回軌道2を1乃至複数回周回した後に周回軌道2から離脱され、周回飛行室4を出てその外側に設けられた検出器5に到達して検出される。各種イオンは同一の運動エネルギーが付与されて飛行を開始するので、m/z値が小さなイオンほど飛行速度が大きくなる。そのため、m/z値が小さなイオンが先に検出器5に到達し、m/z値が増加するに従い遅れて到達する。
バルブ8を閉鎖して周回飛行室4にヘリウムガスを導入しない状態では、従来知られている多重周回飛行時間型質量分析装置と同様の分析動作が実行される。即ち、周回軌道2上の周回数をnとし、周回軌道の1周の長さをLとし、さらに図1中に記載のように入射軌道の長さをLin、出射軌道の長さをLoutとしたとき、或るイオンがイオン源1を出発してから検出器5に到達するまでの飛行距離Ltolは、
Ltol=n・L+Lin+Lout
である。この飛行距離が長いほど、つまり周回周nが多いほど、質量分解能は向上する。

次に、本実施例の多重周回飛行時間型質量分析装置の特徴的な分析動作について、図2、図3を参照して説明する。
制御部10は、上記のようにバルブ8を閉鎖した状態で試料に対する1回目の質量分析を行い、データ処理部12において飛行時間スペクトルを取得する。いま、ここでは説明を簡単にするために、図2(a)に示すような単一ピークが飛行時間スペクトル上に得られる場合を考える。飛行時間はm/z値に一義的に換算可能であるから、図2(a)に示した飛行時間スペクトルからマススペクトルを求めると、マススペクトル上でも1つのピークが現れる。これは、質量分解能の誤差の範囲で同一とみなせるm/z値を持つイオンパケットによるピークである。従来の多重周回飛行時間型質量分析装置では、ここまでで分析は終了であり、後は得られたマススペクトルの解析処理を実行することになる。
これに対し、本実施例の多重周回飛行時間型質量分析装置では、上記1回目の質量分析で取得された飛行時間スペクトルをスペクトル記憶部13に格納し、引き続いて制御部10は、バルブ8を開放して周回飛行室4にヘリウムガスを導入して周回飛行室4内を所定ガス圧に保つ。その状態で1回目の分析時と同一の試料に対する2回目の質量分析を行い、データ処理部12において再び飛行時間スペクトルを取得する。周回飛行室4内にヘリウムガスを導入して所定ガス圧に維持する以外は、分析条件は1回目の分析と同じにする。
例えば2価の窒素分子イオン(14N2
2+)と1価の窒素原子イオン(14N+)とは異なる種類のイオンであるが、同一のm/z値を持つ。そのため、上記1回目の分析で得られる飛行時間スペクトル上では1つのピークとなり、みかけ上、複数種のイオン由来のピークであることは分からない。これに対し、周回飛行室4内に適度のガス圧でヘリウムガスを導入した状態で実行された2回目の質量分析では、同一m/z値を持つイオンであっても、イオンの大きさが相違すれば飛行時間に差が生じる。
いま、図3(a)に示すように、同一m/z値を有し大きさの相違する2種のイオンが同時に、同一の運動エネルギーを付与されて飛行空間に導入された場合を考える。飛行空間内にガスが存在しない(つまり真空雰囲気)場合には、イオンの飛行速度はm/z値に依存するから飛行時間に差は生じず(図3(b)参照)、同時に検出器に到達する筈である。一方、飛行空間内にヘリウムガスが存在すると、イオンは飛行空間内でガスに衝突し、それによって徐々に運動エネルギーを失う。そのため、飛行速度が落ち、つまり減速する。大きなサイズのイオンほどガスとの衝突の機会は多いため、減速の程度も大きく、そのため、図3(c)に示すように、イオンの大きさに応じて飛行時間に差がつき、時間差をもって検出器に到達する。
イオンとガスとの衝突は、図4(a)に示すように、半径RAのイオンと半径RBのガスとの球体同士の衝突であるとみなすことができ、この場合、図4(b)に示すようにさらにモデル化して考えることができる。即ち、イオンは半径が無限に小さい点であり、この点が、半径がRA+RBである円形領域を通過したときがイオンとガスとの衝突であると考える。この円形領域の断面積が衝突断面積であり、π(RA+RB)2である。点であるイオンがこの領域を通過すると、イオンはガスとの相互作用(引力や斥力など)によりエネルギーの一部を失う。これに対し、イオンがこの領域の外側を通過する場合には、イオンはガスとの相互作用を受けず、エネルギーはそのまま維持される。衝突断面積はイオンから見たガスの見かけ上の大きさであると考えることができ、その衝突断面積は実際には、イオンの大きさのほか、イオンの分子構造(形状)、価数、イオンに付加されている官能基の種類、などにも依存する。
上述のようにm/z値が同一であっても大きさ(或いは衝突断面積に影響する上記のような各種要因)が相違する異種のイオンは異なる飛行時間を持つため、2回目の質量分析により取得される飛行時間スペクトル上には、図2(b)に示すように同一m/z値に由来する2本のピークが分離して現れる。時間的に先に現れる成分Aは遅れて現れる成分Bよりも例えばイオンの大きさが小さいものであると推測できる。そこで、スペクトル比較部14は、スペクトル記憶部13に保存されている1回目の質量分析で得られた飛行時間スペクトルと2回目の質量分析で得られた飛行時間スペクトルとを比較し、両者に現れているピークの位置、形状、強度などを比較する。この例では、1本のピークが2本のピークに分離されたことが直ぐに分かるので、少なくとも試料中に同一m/z値を持ち、イオンの大きさ、イオンの分子構造、価数、或いは分子クラスなどが相違する2種のイオンが存在すると判断でき、その結果が出力部15より出力される。
さらにスペクトル比較部14で分離されたピークの強度や時間差などを解析することにより、複数の成分の定量や分子構造に関する情報を得ることができる。この情報と通常の(つまり1回目の)質量分析で得られたマススペクトルとを利用することにより、試料中に含まれる各種物質の解析を、より子細に且つ精度よく行うことが可能である。
なお、飛行するイオンがガスに衝突した場合、条件によっては衝突誘起解離によりイオンが開裂してしまうことがある。開裂が生じると同一m/z値を持つ異なる種類のイオンの識別が困難になるから、2回目の質量分析では、できるだけ開裂が生じない条件とすることが好ましい。
一般に、運動エネルギーを持ったイオンがガスに衝突して衝突誘起解離を起こす場合、重いガスであるほど衝突誘起解離を起こし易い。そこで、上記実施例では、これを避けるために軽い不活性ガスであるヘリウムガスを用いている。さらに、例えばキセノンのように重いガスを周回飛行室4内に導入した場合、イオンがガスに衝突すると開裂を生じなくても衝撃によりイオンの飛行軌道が大きく変化し、周回軌道2から外れるおそれが高くなる。これに対し、軽いガスを用いることにより、衝突したイオンが周回軌道2を外れることがなくなり、飛行途中でのイオンの損失を軽減できるという利点もある。
また、衝突誘起解離を起こしにくくするには、周回飛行室4内に導入するガスの量を少なくすることが考えられる。但し、上述したようなイオンの大きさに応じて飛行時間に差を生じさせるためには、或る程度の量のガスを周回飛行室4内に導入する必要がある。そこで、例えば、適切な、つまり大きさの相違するイオン由来のピークの位置や形状の変化が明確に現れ且つ開裂が問題にならないような、周回飛行室4内のガス圧を予め実験的に求めておき、周回飛行室4内の実際のガス圧が求めておいたガス圧になるようにガスの供給量を制御するとよい。
さらにまた、イオン源1から発したイオンに付与する初期運動エネルギーを小さくすることで、イオンがガスに衝突する際の衝突エネルギーを抑え、衝突誘起解離を起こしにくくすることも考えられる。但し、初期運動エネルギーを極端に小さくすると、飛行途中でのイオンの損失が多くなり、また飛行時間が全体的に長くなって分析時間が長引く。また、周回数を増やすと、途中で飛行できなくなるイオンが発生するおそれもある。したがって、この場合にも、予め実験的に適切な初期運動エネルギーを求めておくようにするとよい。
上記実施例では、周回軌道2全体にヘリウムガスを導入する構成としていたが、原理的には、イオンの飛行経路の一部にガスを導入する構成とすればよい。但し、できるだけ長い飛行経路にガスを導入することより、そのガスの量が少ない場合でも十分な減速効果を発揮させることができ、図2(b)に示したようなピークの位置や形状の変化を顕在化させることができる。
また、本発明に係る飛行時間型質量分析装置は、上記実施例のような多重周回飛行時間型のみならず、リニア型やリフレクトロン型など、様々な飛行経路を持つ飛行時間型質量分析装置に適用が可能である。但し、上記説明でも明らかなように、ガスが導入される飛行経路はできるだけ長いほうがよいから、その点で、多重周回飛行時間型の構成は好適である。なお、ここで言う「多重周回飛行時間型」とは閉じた軌道を繰り返し飛行するものではなく、直線状や曲線状の軌道を繰り返し往復するものも含むものとする。
また、それ以外の点についても、本発明の趣旨の範囲で適宜変形、修正、追加を行っても本願請求の範囲に包含されることは明らかである。
Claims (3)
- イオンに所定の運動エネルギーを付与して飛行空間を飛行させることにより質量分析を行う飛行時間型質量分析装置において、
a)イオンの飛行経路の少なくとも一部に所定のガスを導入するガス導入手段と、
b)同一試料に対し、前記ガス導入手段によりガスが導入されない状態での質量分析とガスが導入された状態での質量分析とをそれぞれ実行し、各分析によりそれぞれ飛行時間スペクトルを取得する分析実行制御手段と、
c)前記分析実行制御手段の制御の下で得られた2つの飛行時間スペクトルに出現しているピークの位置、形状又は強度の少なくとも1つを比較することで、同一m/z値を持つ異種のイオンを識別するイオン識別手段と、
を備えることを特徴とする飛行時間型質量分析装置。 - 請求項1に記載の飛行時間型質量分析装置であって、
同一の飛行軌道を繰り返し飛行させる多重周回飛行時間型であることを特徴とする飛行時間型質量分析装置。 - 請求項1又は2に記載の飛行時間型質量分析装置であって、
前記所定のガスはヘリウムであることを特徴とする飛行時間型質量分析装置。
